Although damaged cells can be repaired, cells that are considered unlikely to be repaired are eliminated through apoptosis, a type of predicted cell death found in multicellular organisms. Apoptosis is a structured cell death involving alterations to the cell morphology and internal biochemical changes. This process involves the expansion and cracking of cells, changes in cell membranes, nuclear fragmentation, chromatin condensation, and chromosome cleavage, culminating in the damaged cells being eaten and processed by other cells. The ubiquitin–proteasome system (UPS) is a major cellular pathway that regulates the protein levels through proteasomal degradation. This review proposes that apoptotic proteins are regulated through the UPS and describes a unique direction for cancer treatment by controlling proteasomal degradation of apoptotic proteins, and small molecules targeted to enzymes associated with UPS. Over the last century, the demand for food resources has been continuously increasing with the rapid population growth. Therefore, it is critically important to adopt sustainable farming practices that can enhance crop production without the excessive use of fertilizers. In this regard, there is a growing interest in the use of nanomaterials for improving plant nutrition as an alternative to traditional chemical or mineral fertilizers. Using this technology, the efficiency of micro- and macro-nutrients in plants can increase. Various nanomaterials have been successfully applied in agricultural production, compared to conventional fertilizers. Among the major plant nutrients, phosphorus (P) is the least accessible since most farmlands are frequently P deficient. Hence, P use efficiency should be maximized to conserve the resource base and maintain agricultural productivity. This review summarizes the current research and the future possibilities of nanotechnology in the biofortification of plant nutrition, with a focus on P fertilizers. In addition, it covers the challenges, environmental impacts, and toxic effects that have been explored in the area of nanotechnology to improve crop production. This study proposes a hybrid control methodology that combines tuned liquid damper (TLD) and active mass damper (AMD) to suppress vibrations of multi-story building structure effectively. To this end, a dynamic model that can predict the vibration of multi-story building structure coupled with TLD and AMD is derived using the energy approach. The TLD that is mainly designed to suppress the main natural mode is developed with a multi-degree-of-freedom model to investigate the effect of the TLD on natural modes of structure numerically. The AMD is an active vibration controller that can suppress remaining resonant peaks, including ones resulting from the application of the TLD as well as other higher modes of interest. In this way, the proposed control can utilize the advantages of TLD and AMD simultaneously. We set up an experimental apparatus to verify the performance of the hybrid control methodology. The numerical and experimental results show that the proposed hybrid control method can successfully suppress the vibrations of multi-story building structure. Nanotechnology is poised to have a huge impact on food packaging applications. Bimetallic and trimetallic nanoparticles (NPs) are formed by combining two and three different metals and metal oxides, respectively. Compared with monometallic NPs, bimetallic and trimetallic NPs have attracted immense attention because of their diverse shape, size, high surface-to-volume ratios, chemical/physical stability, activity, and greater degree of selectivity, with respect to both technological and scientific view. Metal and metal oxide NPs proved to be highly potent antimicrobial and antioxidant properties, together with nanobiosensors for tracing and monitoring the condition of food. These nanostructured materials embedded with both biodegradable and non-degradable polymer to increase mechanical strength and barrier properties to extend the shelf life of various foods. In addition, polymer nanocomposites are treated with essential oils (EOs) to improve their performance in food packaging. In this review, we summarized most recent innovations in food packaging, merits, and demerits of biodegradable and non-degradable polymer used for food packaging. It also provides the different synthetic methods, importance of bimetallic and trimetallic NPs in relation to food packaging to enhance the shelf life of food. The Clp protease complex, which is the central protein degradation machinery in plastids, plays a role similar to proteasome. ClpC1 and ClpC2 comprise the chaperonic part of the Clp protease and share more than 90% nucleotide sequence similarity with each other. In a previous study, the co-suppression of Nicotiana benthamiana ClpC1 and ClpC2 (NbClpC1/C2) resulted in leaf chlorosis, growth retardation phenotype, disappearance of apical dominance and coarsely packed mesophyll cells. Quantitative real-time PCR analysis revealed that the co-suppression of NbClpC1/C2 upregulated the expression levels of defense-related genes including pathogenesis-related protein 1b (PR1b), glutathione-S-transferase, catalase, ascorbate peroxidase and systemic acquired resistance gene 8.2 (SAR8.2). NbClpC1/C2 co-suppressed leaves accelerated hypersensitive response by the infection with an incompatible pathogen, Pseudomonas syringae pv. tomato T1, but increased susceptibility via the infiltration of a compatible pathogen, P. syringae pv. tabaci. These findings indicate that disruption of the chaperones of ClpC1 and ClpC2 increases the growth of bacterial pathogens and therefore increases hypersensitive response and disease susceptibility. Key message: Co-suppression of chaperonicClpC1andClpC2inNicotiana benthamianasignificantly affect the development and exogenous application of gibberellin partially rescue the developmental defects. Over the past decade, the Clp protease complex has been identified as being implicated in plastid protein quality control in plant cells. CLPC1 and CLPC2 proteins form the chaperone subunits of the Clp protease complex and unfold protein substrates to thread them into the ClpP complex. Here, using the technique of virus-induced gene silencing (VIGS), we suppressed both Nicotiana benthamiana ClpC1 and ClpC2 (NbClpC1/C2) functioning as chaperone subunits in the protease complex. Co-suppression of NbClpC1/C2 caused chlorosis and retarded-growth phenotype with no seed formation and significantly reduced root length. We found that co-suppression of NbClpC1/C2 also affected stomata and trichome formation and vascular bundle differentiation and patterning. Analysis of phytohormones revealed significant alteration and imbalance of major hormones in the leaves of NbClpC1/C2 co-suppressed plant. We also found that application of gibberellin (GA3) partially rescued the developmental defects. Co-suppression of NbClpC1/C2 significantly affected the development of N. benthamiana and exogenous application of GA3 partially rescued the developmental defects. Ubiquitination is a reversible cellular process mediated by ubiquitin-conjugating enzymes, whereas deubiquitinating enzymes (DUBs) detach the covalently conjugated ubiquitin from target substrates to counter ubiquitination. DUBs play a crucial role in regulating various signal transduction pathways and biological processes including apoptosis, cell proliferation, DNA damage repair, metastasis, differentiation, etc. Since the transforming growth factor-β (TGF-β) signaling pathway participates in various cellular functions such as inflammation, metastasis and embryogenesis, aberrant regulation of TGF-β signaling induces abnormal cellular functions resulting in numerous diseases. This review focuses on DUBs regulating the TGF-β signaling pathway. Stem cells undergo partitioning through mitosis and separate into specific cells of each of the three embryonic germ layers: endoderm, mesoderm, and ectoderm. Pluripotency, reprogramming, and self-renewal are essential elements of embryonic stem cells (ESCs), and it is becoming evident that regulation of protein degradation mediated by the ubiquitin-proteasome system (UPS) is one of the key cellular mechanisms in ESCs. Although the framework of that mechanism may seem simple, it involves complicated proteolytic machinery. The UPS controls cell development, survival, differentiation, lineage commitment, migration, and homing processes. This review is centered on the connection between stem cell factors NANOG, OCT-3/4, SOX2, KLF4, C-MYC, LIN28, FAK, and telomerase and the UPS. Herein, we summarize recent findings and discuss potential UPS mechanisms involved in pluripotency, reprogramming, differentiation, and self-renewal. Interactions between the UPS and stem cell transcription factors can apply to various human diseases which can be treated by generating more efficient iPSCs. Such complexes may permit the design of novel therapeutics and the establishment of biomarkers that may be used in diagnosis and prognosis development. Guava ( Psidium guajava L.) is a highly perishable fruit crop comparable to mango owing to its high medicinal value and intense aroma. The presence of high genetic variability limits the chances of further expansion of guava improvement using biotechnological interventions. Conventional methods of guava improvement encountered with restricted achievement in progress of disease resistant varieties because of existing high genetic variability in the germplasm. There is a considerable demand for the establishment of successful and efficient regeneration protocols via somatic embryogenesis. Plants regenerated through somatic embryogenesis could be more useful than plants obtained through organogenesis because, in most cases, somatic embryos are of single-cell origin, and have a low frequency of chimeras and a high number of regenerations. This review is a snapshot of the recent status of somatic embryogenesis as a basis for expanding genetic improvement in guava for quality traits and future perspectives using advanced technologies. Background: Porphyra tenera (Kjellman, 1897) is the most common eatable red seaweed in Asia. In the present study, P. tenera volatile oil (PTVO) was extracted from dried P. tenera sheets that were used as food by the microwave hydrodistillation procedure, after which the characterization of its chemical constituents was done by gas chromatography and mass spectroscopy and its antioxidant potential was evaluated by a number of in vitro biochemical assays such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging, nitric oxide (NO) scavenging, superoxide radical scavenging, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging, hydroxyl radical scavenging and reducing power assay and inhibition of lipid peroxidation. Results: A total of 30 volatile compounds comprising about 99.4% of the total volume were identified, of which trans-beta-ionone (20.9%), hexadecanoic acid (9.2%) and 2,6-nonadienal (8.7%) were present in higher quantities. PTVO exhibited strong free radical scavenging activity by DPPH scavenging (44.62%), NO scavenging (28.45%) and superoxide scavenging (54.27%) at 500 µg/mL. Similarly, it displayed strong ABTS radical scavenging (IC value of 177.83 µg/mL), hydroxyl radical scavenging (IC value of 109.70 µg/mL), and moderate lipid peroxidation inhibition activity (IC value of 231.80 µg/mL) and reducing power (IC value of 126.58 µg/mL). PTVO exhibited strong antioxidant potential in a concentration dependent manner and the results were comparable with the BHT and α-tocopherol, taken as the reference standard compounds (positive controls). Conclusions: Taken together, PTVO with potential bioactive chemical compounds and strong antioxidant activity could be utilized in the cosmetic industries for making antioxidant rich anti-aging and sun-screen lotion and in the food sector industries as food additives and preservatives. All organs consisting of single cells are consistently maintaining homeostasis in response to stimuli such as free oxygen, DNA damage, inflammation, and microorganisms. The cell cycle of all mammalian cells is regulated by protein expression in the right phase to respond to proliferation and apoptosis signals. Post-translational modifications (PTMs) of proteins by several protein-editing enzymes are associated with cell cycle regulation by their enzymatic functions. Ubiquitination, one of the PTMs, is also strongly related to cell cycle regulation by protein degradation or signal transduction. The importance of deubiquitinating enzymes (DUBs), which have a reversible function for ubiquitination, has recently suggested that the function of DUBs is also important for determining the fate of proteins during cell cycle processing. This article reviews and summarizes the diverse roles of DUBs, including DNA damage, cell cycle processing, and regulation of histone proteins, and also suggests the possibility for therapeutic targets. Key message: We report the significant increase of the content of free amino acids in Nicotiana benthamiana by the co-suppression of the ClpC1 and ClpC2 genes, which are translated to be the chaperonic part in the Clp protease at plastids. Abstract: Clp protease with ClpC1 and ClpC2 proteins as the chaperonic part degrades denatured or improperly folded protein in plastids. Nicotiana benthamiana ClpC1 and ClpC2 genes ( NbClpC1 and NbClpC2: NbClpC1/C2) share 93 % similarities; therefore, co-suppression of the NbClpC1/C2 was possible using a single virus-induced silencing vector. Co-suppression of NbClpC1/ C2 resulted in a pleiotropic phenotype including disappearance of apical dominance and formation of chlorotic leaves. NbClpC1/C2 co-suppressed leaves accumulated 11.9-fold more free amino acids than the GFP-silenced leaves. The co-suppression of NbClpC1/C2 did not change the expression levels of some selected genes in the biosynthetic pathways for the free amino acids, but reduced the total protein amounts to 32.5 %, indicating that co-suppression affected the incorporation of free amino acids in proteins during translation. The loosely packed mesophyll cells and abnormal vascular bundles in the leaves suggested structural problems associated with translocation of free amino acids to sink tissues. Genetically encoded fluorescent proteins are extensively utilized for labeling and imaging proteins, organelles, cell tissues, and whole organisms. In this study, we explored the feasibility of mRFP1 and its variants for measuring intracellular temperature. A linear relationship was observed between the temperature and fluorescence intensity of mRFP1 and its variants. Temperature sensitivities of E. coli expressing mRFP1, mRFP-P63A and mRFP-P63A[(4 R)-FP] were −1.27%, −1.26% and −0.77%/°C, respectively. Key message: We report the production of taxadiene by transformation of N. benthamiana with a taxadiene synthase gene. The production was significantly increased by an elicitor treatment or metabolic pathway shunting. Abstract: Paclitaxel (Taxol) was first isolated from the bark of the pacific yew tree as an anticancer agent and has been used extensively to treat various types of cancer. Taxadiene, the first committed product of paclitaxel synthesis is cyclized from geranylgeranyl diphosphate (GGPP), and further complex hydroxylation and acylation processes of the unique taxane core skeleton produce paclitaxel. To accomplish de novo production of taxadiene, we transformed Nicotiana benthamiana with a taxadiene synthase ( TS) gene. The introduced TS gene under the transcriptional control of the CaMV 35S promoter was constitutively expressed in N. benthamiana, and the de novo production of taxadiene was confirmed by mass spectroscopy profiling. Transformed N. benthamiana homozygous lines produced 11-27 μg taxadiene/g of dry weight. The highest taxadiene production line TSS-8 was further treated with an elicitor, methyl jasmonate, and metabolic pathway shunting by suppression of the phytoene synthase gene expression which resulted in accumulation of increased taxadiene accumulation by 1.4- or 1.9-fold, respectively. In summary, we report that the production of taxadiene in N. benthamiana was possible by the ectopic expression of the TS gene, and higher accumulation of taxadiene could be achieved by elicitor treatment or metabolic pathway shunting of the terpenoid pathway. Tomatoes ( Solanum lycopersicum L.) are known as a super food because they contain many compounds beneficial to human health. To elucidate the role of specific wavelength of light on the accumulation of free amino acids and -aminobutyric acid (GABA) in postharvest tomato fruits, mature green tomato fruits were treated with darkness, or blue or red light by using light emitting diode (LED) for seven days. All free amino acids, except glutamic and aspartic acids, increased more in fruits treated with blue light than in those treated with either darkness or red light. The GABA, a non-protein amino acid and having health beneficial effects, increased in the blue light treatment with the maximum content being observed on the third day. Taken together, these findings indicate that irradiation with blue light has an effect in increasing the GABA contents in postharvest tomato fruits. [ABSTRACT FROM AUTHOR]

As a major stilbene phytoalexin, resveratrol is produced or elicited in several plant species as a part of defense systems protecting plants against diseases. Resveratrol can be present in both the trans- and cis-isomeric forms, and only the trans-form increases the life expectancy and lowers the risk of cardiovascular diseases as the most bioactive form. In addition to the usages for diet and industry, peanut plant ( Arachis hypogaea) and peanuts are getting higher attention due to their containment of resveratrol in the kernels and other parts of peanut plant, such as leaves, roots, and peanut shell. Recently, natural resveratrol derived from peanuts has also become a promising nutraceutical agent, promoting human health. Resveratrol has also been detected in peanut products including peanut butters, roasted peanuts, and boiled peanuts. Although, smaller and immature peanuts contain higher levels of resveratrol than mature peanuts, resveratrol in peanuts can also be preserved by cooking or manufacturing processes. Moreover, the amount of resveratrol in peanut plants and peanuts has been found to increase by external stimuli including microbial infection, wounding, UV light irradiation, ultrasonication, yeast extract treatment and by plant stress hormones. In addition, molecular level analysis has confirmed that four resveratrol synthase (RS) genes (RS1, RS2, RS3 and RS4) which catalyze synthesis of resveratrol have been identified in peanuts, and up-regulation of the genes is positively correlated to the increased contents of resveratrol. In this review, we summarize the natural biosynthesis of resveratrol in peanuts and peanut plants, as well as the occurrence of this natural phytoalexin in various peanut products. A brief knowledge on the biosynthetic pathway of resveratrol synthesis has been described. This review also deals on highlighting the effect of various external stimuli (biotic and abiotic stresses) in order to achieve the maximum induction and/or elicitation of resveratrol in peanuts and peanut plants. It has become apparent that ubiquitination plays a critical role in cell survival and cell death. In addition, deubiquitinating enzymes (DUBs) have been determined to be highly important regulators of these processes. Cells can be subjected to various stresses and respond in a variety of different ways ranging from activation of survival pathways to the promotion of cell death, which eventually eliminates damaged cells. The regulatory mechanisms of apoptosis depend on the balanced action between ubiquitination and deubiquitination systems. There is a growing recognition that DUBs play essential roles in regulating several binding partners to modulate the process of apoptosis. Thus, the interplay between the timing of DUB activity and the specificity of ubiquitin attachment and removal from its substrates during apoptosis is important to ensure cellular homeostasis. Numerous sequence variants of wheat ( Triticum aestivum L.) manganese superoxide dismutase (MnSOD) genes have been found. Quantitative real-time PCR was used to measure the expression levels of three MnSOD genes distinguished by a variable amino acid, and three genes distinguished by sequence variation in the 3′ untranslated region (3′ UTR), in wheat plants grown at 20°C and cold-acclimated for 1–4 weeks at 2°C. The amino acid variants did not differ significantly in expression levels, however, differential expression of genes differing in the 3′ UTR was observed. Diploid wheat-related species also carried sequence variants of MnSOD, with differing levels of expression, suggesting diversification of the MnSOD gene family occurred prior to the polyploidization events of hexaploid wheat. HAUSP (herpes virus-associated ubiquitin specific protease, known as ubiquitin specific protease 7), one of DUBs, regulates the dynamics of the p53 and Mdm2 network in response to DNA damage by deubiquitinating both p53 and its E3 ubiquitin ligase, Mdm2. Its concerted action increases the level of functional p53 by preventing proteasome-dependent degradation of p53. However, the protein substrates that are targeted by HAUSP to mediate DNA damage responses in the context of the HAUSP-p53-Mdm2 complex are not fully identified. Here, we identified nucleolin as a new substrate for HAUSP by proteomic analysis. Nucleolin has two HAUSP binding sites in its N- and C-terminal regions, and the mutation of HAUSP interacting peptides on nucleolin disrupts their interaction and it leads to the increased level of nucleolin ubiquitination. In addition, HAUSP regulates the stability of nucleolin by removing ubiquitin from nucleolin. Nucleolin exists as a component of the HAUSP-p53-Mdm2 complex, and both Mdm2 and p53 are required for the interaction between HAUSP and nucleolin. Importantly, the irradiation increases the HAUSP-nucleolin interaction, leading to nucleolin stabilization significantly. The use of fungi as biological nanofactories is an innovative and promising research field of nanoscience. It offers the potential to yield sustainable and cost-effective nanostructures with fine-tuned features. This study reports the trichogenic synthesis of silver oxide/copper oxide (Ag2O/CuO) nanoalloys, using the culture filtrate of the endophytic fungus Trichoderma virens. The filtrate of T. virens mediated the synthesis of the Ag2O/CuO nanoalloys, which was visually verified by the formation of dark bluish-greenish colloidal suspension upon reacting with AgNO3 and CuSO4·5H2O (1:1 v/v). X-ray diffraction revealed the crystalline nature of the trichogenic Ag2O/CuO nanoalloys, showing the characteristic diffraction peaks of cubic Ag2O (Pn-3/201) and tenorite monoclinic CuO (C2/c/15). X-ray photoelectron spectroscopy revealed spectra corresponding to the core levels of Ag 3d, Cu 2p, and O 1 s in the Ag2O/CuO nanoalloys. Fourier-transform infrared spectroscopy showed nitro and ester stretching vibrations in addition to characteristic interatomic metal–metal vibrations near 500 cm−1. Dynamic light scattering revealed that the Ag2O/CuO nanoalloys had an average particle size distribution of 124 nm, while zeta potential measurement was + 23.8 mV. Field-emission scanning electron microscopy (FESEM) revealed a mixture of hemispherical and plate-like Ag2O/CuO particles arranged in crystalline clusters. High-resolution transmission electron microscopy revealed the formation of quasi-spherical, circular, cuboid, and triangular Ag2O NPs along with the sharp-edged nanoplates of CuO particles. Energy-dispersive X-ray analysis showed prominent emission peaks of Ag (12.41%), Cu (17.68%), and O (69.91%). The antibacterial efficacy of the Ag2O/CuO nanoalloys was evaluated against six Gram-negative and three Gram-positive bacterial strains. The Ag2O/CuO nanoalloys exhibited significant antibacterial activity against streptomycin-sensitive Xanthomonas citri pv. citri (Xcc) (17.85 ± 0.735 mm), streptomycin-resistant Xcc (17.77 ± 0.720 mm), Clavibacter michiganensis subsp. michiganensis (17.44 ± 3.492 mm), and Escherichia coli (16.23 ± 2.468 mm), whereas the effectiveness of the Ag2O/CuO nanoalloys was moderate against Salmonella enterica serovar Typhimurium (12.74 ± 1.302 mm), Staphylococcus aureus (12.56 ± 0.667 mm), streptomycin-sensitive Pectobacterium carotovorum subsp. carotovorum (Pcc) (11.77 ± 0.431 mm), streptomycin-resistant Pcc (11.63 ± 0.741 mm), and C. michiganensis subsp. capsici (11.62 ± 0.958 mm). The bacterial cells exposed to the Ag2O/CuO nanoalloys exhibited notable morphological changes, such as cell membrane disruption, structural damage, compromised membrane integrity, shrinkage, and swelling. Field-emission scanning electron microscopy (FESEM) revealed a mixture of hemispherical and plate-like Ag2O/CuO particles arranged in crystalline clusters. High-resolution transmission electron microscopy revealed the formation of quasi-spherical, circular, cuboid, and triangular Ag2O NPs along with the sharp-edged nanoplates of CuO particles. Energy-dispersive X-ray analysis showed prominent emission peaks of Ag (12.41%), Cu (17.68%), and O (69.91%). The antibacterial efficacy of the Ag2O/CuO nanoalloys was evaluated against six Gram-negative and three Gram-positive bacterial strains. The Ag2O/CuO nanoalloys exhibited significant antibacterial activity against streptomycin-sensitive Xanthomonas citri pv. citri (Xcc) (17.85 ± 0.735 mm), streptomycin-resistant Xcc (17.77 ± 0.720 mm), Clavibacter michiganensis subsp. michiganensis (17.44 ± 3.492 mm), and Escherichia coli (16.23 ± 2.468 mm), whereas the effectiveness of the Ag2O/CuO nanoalloys was moderate against Salmonella enterica serovar Typhimurium (12.74 ± 1.302 mm), Staphylococcus aureus (12.56 ± 0.667 mm), streptomycin-sensitive Pectobacterium carotovorum subsp. carotovorum (Pcc) (11.77 ± 0.431 mm), streptomycin-resistant Pcc (11.63 ± 0.741 mm), and C. michiganensis subsp. capsici (11.62 ± 0.958 mm). The bacterial cells exposed to the Ag2O/CuO nanoalloys exhibited notable morphological changes, such as cell membrane disruption, structural damage, compromised membrane integrity, shrinkage, and swelling. This research lays the groundwork for developing and using trichogenic Ag2O/CuO nanoalloys as highly effective nano-antimicrobial agents for a diverse array of applications. [ABSTRACT FROM AUTHOR]
