Introduction:The success of obesity therapy is dependent on the genetic background of the patient. Circadian Locomotor Output Cycles Kaput (CLOCK), one of the transcription factors from the positive limb of the molecular clock, is involved in metabolic alterations.Objective:To investigate whether five candidate polymorphisms from CLOCK were associated with anthropometric, metabolic measures and weight loss in response to a behavioural weight reduction programme based on the Mediterranean diet.Methods:Five hundred overweight/obese subjects, aged 20–65 years, who attended outpatient clinics specializing in obesity, were studied. Anthropometric, biochemical and dietary intake variables were analysed. Effectiveness of the programme and weight loss progression during 28 weeks of treatment was assessed.Results:Four of five CLOCK SNPs selected were significantly associated with obesity variables (P<0.05). The genetic variation in the rs1801260 CLOCK was associated with obesity at baseline and also affected weight loss. Patients with the variant allele (G) lost significantly less weight i(P=0.008) compared with wild type. Repeated measures analysis showed that weight loss over time was significantly different between rs1801260 CLOCK variations (P=0.038). Carriers of the G allele displayed greater difficulty in losing weight than non-carriers. In this particular polymorphism, the frequency of short-time sleepers (6 h per day) was greater in minor allele carriers than in non-carriers (59% vs 41%; P<0.05). CLOCK polymorphisms were also associated with significant differences in total plasma cholesterol at the completion of dietary treatment (P<0.05).Conclusions:We have replicated previous studies showing a relationship between CLOCK gene polymorphisms and obesity. Detailed reviews of high-resolution acoustic studies in the continental slope of the Gulf of Cadiz has revealed the following gas-related features: acoustic turbidity and blanking, bright spots, ancient and modern pockmarks, high-amplitude diffractions, acoustic plumes and turbidity in the water column, and BSRs. The origin of the gas is believed to be biogenic and thermogenic. The BSR-like acoustic anomalies occur intermittently in some areas of the upper slope and tend to occur in the volcanoes/diapirs. The pressure–temperature conditions deduced for the location of those acoustic anomalies do not correspond to the conditions of stability of gas hydrates. It is suggested that these volcanoes/diapirs intrusions may locally induce anomalously higher pore pressure conditions on the immediately surrounding sediments, affecting the stability field of the gas hydrates. Multibeam bathymetric and ultra high-resolution seismic data reveal that the distal course of the Equatorial Atlantic Mid-Ocean Channel (EAMOC) extends further east and south than was previously known, and is controlled by the presence of morphologic highs related to the Fernando de Noronha Fracture Zone. Distal course of the EAMOC is buried by sediments, and does not have bathymetric expression on the seafloor. The channel fill consists of three seismic sequences, suggesting that the recent geological evolution of the channel is composed of successive phases of decreasing sedimentary activity that finally resulted in its complete burial. Tectonic and volcanic activity related to the Fernando de Noronha Fracture Zone and Ridge, together with the effect of strong pulses of the Antarctic bottom water current during the upper Pliocene are suggested to have contributed to the progressive burial and the final abandonment of the EAMOC. Five depositional bodies occur within the Quaternary deposits of the northwestern Alboran Sea: Guadalmedina-Guadalhorce prodelta, shelf-edge wedges, progradational packages, Guadiaro channel-levee complex, and debris flow deposits. The sedimentary structure reflects two styles of margin growth characterized: 1) by an essentially sediment-starved outer, shelf and upper slope and by divergent slope seismic facies; 2) by a prograding sediment outer shelf, and parallel slope seismic facies. Eustatic oscillations, sediment supply, and tectonic tilting have controlled the type of growth pattern, and the occurrence of the depositional bodies. Laboratory analysis of core samples from the western Alboran Sea slope reveal a large variability in texture and geotechnical properties. Stability analysis suggests that the sediment is stable under static gravitational loading but potentially unstable under seismic loading. Slope failures may occur if horizontal ground accelerations greater than 0.16 g are seismically induced. The, Alboran Sea is an active region, on which earthquakes inducing accelerations big enough to exceed the shear strength of the soft soil may occur.
