The effects of long-term selenium supplementation on the selenium body status were investigated in humans and rats. Selenium was determined in human muscle biopsies and monitor materials and in rat tissues by neutron activation analysis. The results showed that the body selenium load is raised by additional supply of selenomethionine or selenomethionine-containing yeast but not proportionally to the intake. The surplus selenium can serve as an endogenous source to maintain the selenoprotein levels during insufficient supply. Highly significant correlations between the muscle selenium concentrations and those in blood, blood fractions, hair and nails indicate that the selenium status can be assessed by analysis of these monitor materials. Due to the essential functions of selenium-containing enzymes and the relationships between changes in the selenium status and diseases, the determination of the element and its compounds is of great interest. Radiotracer studies with 75Se have been valuable tools in selenium research. NAA and ICP-MS allow both total element and stable isotope measurements. ICP-MS in conjunction with chromatographic separation techniques and gel electrophoretic procedures coupled with scanning methods such as XRF, PIXE and laser ablation ICP-MS have been used in the determination of the selenium compounds. In this survey the application of these methods in selenium research is discussed with the help of examples on the regulation of the selenium metabolism and the detection and investigation of novel selenium-containing proteins. Investigations have been carried out on rats to obtain information about the selenium-containing proteins present in the microsomal fraction and especially in the endoplasmatic reticulum (ER) of the kidney. For the determination of the selenium levels instrumental neutron activation analysis via 75Se was used. After labeling of rats in vivo with 75Se-selenite and separation of the proteins in the renal homogenate and cell compartments by electrophoretic methods, the 75Se-containing proteins were detected by autoradiography. In this way, six selenium-containing proteins with molecular masses of 15, 16, 20, 23-25, 40-42 and 58-60 were found in the endoplasmatic reticulum. Nuclear methods have been applied in the investigation of selenium-containing proteins in rat tissues. Selenium was determined in tissues, cells, and cellular compartments by instrumental neutron activation analysis viaSe orSe. For tracer studies, the selenium compounds were labeled in vivo by administeringSe with a high specific activity to rats. Quantitative determination of very small amounts of the element in protein fractions was achieved by measurement of the tracer after replenishment of selenium-depleted animals with the labeled element. The application of the nuclear methods in the detection, characterization, and identification of new selenium-containing proteins is shown with the help of some examples. [ABSTRACT FROM AUTHOR]

In several studies on rats, the metabolism of selenium was investigated. The quantitative determination of the element was carried out by instrumental neutron activation analysis. For in vivo tracer experiments,Se-labeled selenium compounds were used. In addition to these methods, procedures for the measurement of the selenoenzyme glutathione peroxidase, and for the investigation of other selenoproteins, were applied. In this way, information on the specific pools and sites of action of the element, on biologically important selenoproteins and the regulation of the selenium metabolism, was obtained. Sex-specific differences with regard to the intensity of transferrin bands were observed in a noninbred adult mouse population after separation of the serum proteins by polyacrylamide gel electrophoresis. Amongst the female animals, an additional protein fraction was found just above the position of the transferrin bands. By means of a tracer method, usingFe-labelling, it could be shown that the additional fraction is not a part of the transferrin bands. [ABSTRACT FROM AUTHOR]

The glutathione peroxidase activity in the plasma of rats on the 20th day of pregnancy was found to be 50% lower than in nulliparous control animals. During lactation, the level rose again but was still significantly different from that of the control rats on the 20th day post partum. The erythrocyte enzyme activity remained unchanged. The serum selenium content of rats was measured by means of neutron activation analysis. It was found to drop between the 12th day of pregnancy and term and to return to its original level within 2 days after delivery. Hysterectomy on the 10th day of gestation prevented this decrease. Nursing had no influence on the element level. The findings suggest that placental secretions might be involved in the control of the serum selenium content of rats during pregnancy.
