This study presents an efficient chemiluminescence (CL) probe based on perovskite nanocrystals (NCs) for detection of L–cysteine (L–Cys). It consists of nickel–doped CsPbBr3 NCs embedded in the mesoporous SiO2 matrix as CL reagent and cerium (IV) as an oxidant in aqueous environment. The probe was designed for the highly selective determination of L–Cys based on its remarkable enhancing effect on the CL intensity. The colloidal nanocomposite of nickel–doped CsPbBr3 NCs@SiO2 with photoluminescence quantum yield of 58% was fabricated by ligand–assisted re–precipitation method and characterized by using UV–Vis absorption, FT–IR, X–ray diffraction, and transmission electron microscopy. The sensor was utilized to determine L–Cys in the linear concentration range of 20–300 nM with a detection limit of 12.8 nM. Direct chemical oxidation of Ni–doped CsPbBr3 NCs@SiO2 by Ce(IV) was the single cause of the formation of the excited-state NCs and subsequent production of CL. The developed probe provides outstanding selectivity towards L–Cys over structurally related compounds. Two-pore domain K+ (K2P) channel activity was previously thought to be controlled primarily via a selectivity filter (SF) gate. However, recent crystal structures of TASK-1 and TASK-2 revealed a lower gate at the cytoplasmic pore entrance. Here, we report functional evidence of such a lower gate in the K2P channel K2P17.1 (TALK-2, TASK-4). We identified compounds (drugs and lipids) and mutations that opened the lower gate allowing the fast modification of pore cysteine residues. Surprisingly, stimuli that directly target the SF gate (i.e., pHe., Rb+ permeation, membrane depolarization) also opened the cytoplasmic gate. Reciprocally, opening of the lower gate reduced the electric work to open the SF via voltage driven ion binding. Therefore, it appears that the SF is so rigidly locked into the TALK-2 protein structure that changes in ion occupancy can pry open a distant lower gate and, vice versa, opening of the lower gate concurrently promote SF gate opening. This concept might extent to other K+ channels that contain two gates (e.g., voltage-gated K+ channels) for which such a positive gate coupling has been suggested, but so far not directly demonstrated. Late-stage specific and selective diversifications of peptides and proteins performed at target residues under ambient conditions are recognized to be the most facile route to various and abundant conjugates. Herein, we report an orthogonal modification of cysteine residues using alkyl thianthreium salts, which proceeds with excellent chemoselectivity and compatibility under mild conditions, introducing a diverse array of functional structures. Crucially, multifaceted bioconjugation is achieved through clickable handles to incorporate structurally diverse functional molecules. This "two steps, one pot" bioconjugation method is successfully applied to label bovine serum albumin. Therefore, our technique is a versatile and powerful tool for late-stage orthogonal bioconjugation. A novel accelerator, cerium cysteine (Ce-Cys) was synthesised by double replacement reaction. The complex was characterised by FT-IR analysis, thermal analysis, EDTA titration and elemental analysis, respectively, to determine that the molecular formula of the complex was Ce (Cys)3Cl3·3H2O. Cure characteristics, crosslink density, mechanical properties and thermo-oxidative ageing of NR compounds were investigated. The research revealed that the vulcanisates containing Ce-Cys manifested high scorch time, outstanding mechanical properties, and better thermo-oxidative ageing properties. Furthermore, based on kinetic calculations, it was concluded that as the amount of Ce-Cys increased, the reactivity of the crosslink precursor increased while the activation energy decreased. Antioxidants agents play an essential role in the food industry for improving the oxidative stability of food products. In the last years, the search for new natural antioxidants has increased due to the potential high toxicity of chemical additives. Therefore, the synthesis and evaluation of the antioxidant activity in peptides is a field of current research. In this study, we performed a Quantitative Structure Activity Relationship analysis (QSAR) of cysteine-containing 19 dipeptides and 19 tripeptides. The main objective is to bring information on the relationship between the structure of peptides and their antioxidant activity. For this purpose, 1D and 2D molecular descriptors were calculated using the PaDEL software, which provides information about the structure, shape, size, charge, polarity, solubility and other aspects of the compounds. Different QSAR model for di- and tripeptides were developed. The statistic parameters for di-peptides model (R2train = 0.947 and R2test = 0.804) and for tripeptide models (R2train = 0.923 and R2test = 0.847) indicate that the generated models have high predictive capacity. Then, the influence of the cysteine position was analyzed predicting the antioxidant activity for new di- and tripeptides, and comparing them with glutathione. In dipeptides, excepting SC, TC and VC, the activity increases when cysteine is at the N-terminal position. Pictet-Spengler reaction is one of the main ways to synthesize spiroindolones, which widely exists in natural products and pharmaceutical agents. We accidentally discovered bovine serum albumin (BSA) had catalytic activity for Pictet-Spengler reaction previously. After a series of exploratory experiments around the catalytic process, it was found that some amino acids played a key role. It is worth noting that, Pictet-Spengler reaction catalyzed by amino acids has been rarely described. Herein, we report the cysteine-catalyzed Pictet-Spengler reaction between tryptamine and isatin under metal-free and convenient conditions. The catalytic performance of amino acid was optimized through screening reaction conditions such as time, temperature, solvent, molar ratio and catalyst amount and a series of spiroindolones can be obtained in moderate to excellent yields. Purpose: The Cr(VI) removal capacity of green synthesized nano-zero-valent iron (GnZVI) using tea polyphenols (TPs) remains limited. To improve their application in contaminated soil and groundwater, the GnZVI was modified. This is necessary for in-situ remediation of heavy metal-contaminated soil and groundwater. Materials and methods: The GnZVI-based carbon-composite (L&GnZVI@BC) was successfully constructed via the synergistic effect of L-cysteine modification and biochar support. The Cr(VI) removal capacity and transportability of L&GnZVI@BC were investigated in soil–water system by batch and column experiments. Results and discussion: Comparison with two materials of GnZVI separately modified by L-cysteine (L-GnZVI) or supported by biochar (GnZVI@BC)), GnZVI composite with a combination modification of L-cysteine and biochar (L&GnZVI@BC, L-cysteine/biochar/Fe = 0.1/0.1/1) showed a much higher Cr(VI) removal capacity in soil and groundwater. The synergistic effect of the reduction of L-cysteine functional groups and the dispersibility of biochar support can enhance the transportability of L&GnZVI@BC in water-saturated sand media for more Cr(VI) adsorption at neutral pH; while that improved the soluble Fe(II) released from composite for the higher reduction of Cr(VI) into Cr(III) at acidic pH. Particularly, L&GnZVI@BC favored more Cr(III) generation during transport in porous media at lower pH when applied in the treatment of Cr(VI) contamination. Conclusion: This research highlights that the modification of both L-cysteine and biochar was beneficial to sufficient transport and efficient remediation in Cr(VI)-contaminated soil and groundwater environments at different pH ranges. Background: Inflammation serves as our body's immune response to combat infections, pathogens, viruses, and external stimuli. Inflammation can be classified into two types: acute inflammation and chronic inflammation. Non-steroidal anti-inflammatory medications (NSAIDs) are used to treat both acute and chronic inflammatory disorders. However, these treatments have various side effects such as reduced healing efficiency, peptic ulcers, gastrointestinal toxicities, etc. Method: This review assesses the potential of anti-inflammatory peptides (AIPs) derived from various natural sources, such as algae, fungi, plants, animals, and marine organisms. Focusing on peptides rich in cysteines and methionine, sulphur-containing amino acids known for their role in suppression of inflammation. Result: Due to their varied biological activity, ability to penetrate cells, and low cytotoxicity, bioactive peptides have garnered interest as possible therapeutic agents. The utilisation of AIPs has shown great potential in the treatment of disorders associated with inflammation. AIPs can be obtained from diverse natural sources such as algae, fungi, plants, and animals. Cysteine and methionine are sulphur-containing amino acids that aid in the elimination of free radicals, hence assisting in the treatment of inflammatory diseases. Conclusion: This review specifically examines several sources of AIPs including peptides that contain numerous cysteines and methionine. Cysteine (Cys) plays an indispensable role as an antioxidant in the maintenance of bioredox homeostasis. We have constructed an efficient fluorescent probe Mito-Cys based on the binding of indole and naphthol. The acrylic ester group serves as a recognition switch for specific detection of Cys, which undergoes Michael addition and intramolecular cyclization reactions, thereby ensuring the chemical kinetics priority of Cys compared to other biothiols. The probe has good water solubility, large Stokes shift (137 nm), with a detection limit of 21.81 nM. In addition, cell imaging experiments have shown that the probe has excellent mitochondrial targeting ability (R = 0.902). The probe can distinguish between Cys, homocysteine (Hcy) and glutathione (GSH), and can detect Cys specifically and quickly (100 s) to ensure accurate quantitative analysis of Cys changes in cells. More importantly, the probe confirms that ferroptosis inducing factors trigger thiol starvation in mitochondria, which helps to gain a deeper understanding of the physiological and pathological functions related to Cys and ferroptosis. A digital image colorimetry method utilizing a smartphone as the detection tool was developed for the determination of acetylcysteine and cysteine in effervescent tablets. The method is based on a simple nitroprusside test specific for free −SH groups in aminothiols, which results in orange-red color sample solutions for colorimetric detection. The analytical response of the colorimetric detection decreased as the amino acid concentration increased. The data of the digital images taken were processed to RGB (red–green–blue) color scales using the Color Detector application. Today's technology allows faster and easier analysis with the use of easily accessible and portable smartphones in colorimetric detection systems. The developed method has good linearity in a linear working range of 5.0–25 mg/L (41.27–206.34 µM) and 4.94–47.30 mg/kg (30.27–289.85 µM) for cysteine and acetylcysteine, respectively. The limit of detection and quantification values for cysteine and acetylcysteine were calculated as 1.0 and 3.4 mg/L (8.25–28.06 µM) and 1.8 and 6.1 mg/kg (11.03–37.38 µM), respectively. The applicability and accuracy of the developed method were tested on drug samples. Sugar esters display surface-active properties, wetting, emulsifying, and other physicochemical phenomena following their amphipathic nature and recognize distinct biological activity. The development of nutritional pharmaceuticals and other applications remains of great interest. Herein, three novel homologous series of several N-mono-fatty acyl amino acid glucosyl esters were synthesized, and their physicochemical properties and biological activities were evaluated. The design and preparation of these esters were chemically performed via the reaction of glucose with different fatty acyl amino acids as renewable starting materials, with the suggestion that they would acquire functional characteristics superior and competitive to certain conventional surfactants. The synthesized products are characterized using FTIR, 1H-NMR, and 13C-NMR spectroscopy. Further, their physicochemical properties, such as HLB, CMC, Γmax, γCMC, and Amin, were determined. Additionally, their antimicrobial and anticancer efficiency were assessed. The results indicate that the esters' molecular structure, including the acyl chain length and the type of amino acid, significantly influences their properties. The measured HLB ranged from 8.84 to 12.27, suggesting their use as oil/water emulsifiers, wetting, and cleansing agents. All esters demonstrate promising surface-active characteristics, with moderate to high foam production with good stability. Notably, compounds 6-O-(N-dodecanoyl, tetradecanoyl cysteine)-glucopyranose (34, 35), respectively and 6-O-(N-12-hydroxy-9-octadecenoyl cysteine)-glucopyranose (38) display superior foamability. Wetting efficiency increased with decreasing the chain length of the acyl group. The storage results reveal that increasing the fatty acyl hydrophobe length enhances the derived emulsion's stability for up to 63 days. Particularly, including cysteine in these glucosyl esters improves wetting, foaming, and emulsifying potentialities. Furthermore, the esters exhibit antibacterial activity against several tested Gram-positive and Gram-negative bacteria and fungi. On the other hand, they show significant antiproliferative effects on some liver tumor cell lines. For instance, compounds 6-O-(N-12-hydroxy-9-octadecenoylglycine)-glucopyranose (28), 6-O-(N-dodecanoyl, hexadecanoyl, 9-octadecenoyl and 12-hydroxy-9-octadecenoylvaline)- glucopyranose (29, 31, 32 and 33), respectively in addition to the dodecanoyl, hexadecanoyl, 9-octadecenoyl and 12-hydroxy-9-octadecenoyl cysteine glucopyranose (34, 36, 37 and 38), respectively significantly inhibit the examined cancer cells. Plasma total cysteine (tCys) is strongly associated with fat mass in humans. Mesna lowers plasma tCys in a dose-dependent manner, but it is not known whether it interferes with metabolism of other amino acids or protein. In this Phase-1 study, we show that a single dose of mesna administered at 400, 800, 1200 or 1600 mg to 6–7 individuals per dose only slightly affects amino acid profiles, with increases in plasma valine across dose levels. This study presents a novel approach to modifying copolymers with active azide end-groups through click chemistry, specifically targeting the incorporation of cysteine amino acid. The synthesis commenced with the reaction between cysteine amino acid and propargyl amine, yielding the alkyne-terminated amino acid Boc-Cys(Bnz)-PA compound. Through the click chemistry methodology, the copolymer with active azide ends was modified with alkyne-terminated Cys-PA. Thermal characterization of the copolymers was performed via differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). DSC measurements revealed an increase in glass transition temperature (Tg) from 65 °C to 83 °C upon the modification with cysteine, indicating enhanced thermal stability. However, TGA data suggested a decrease in thermal stability as evidenced by lower initial degradation temperatures for the modified copolymer compared to the unmodified counterpart.The degradation activation energies of the copolymers were estimated using the Flynn-Wall-Ozawa method across various conversion ranges, demonstrating higher thermal degradation activation energy for the modified copolymer compared to the unmodified one. Dielectric behavior analysis conducted at room temperature over a frequency range from 100 Hz to 20 kHz revealed a decreasing trend in dielectric constant and dielectric loss values with increasing frequency for all copolymer structures. The alternating current (AC) conductivities at 30 kHz were calculated as 1.84 × 10−7 S cm−1, and 4.75 × 10−7 S cm−1 for P(VBCl-co-VBN3 0.77) and (P(VBCl-co-(VBN3-click-Cys)0.77), respectively, indicating a frequency-dependent conductivity enhancement with increasing frequency. Overall, the results suggest that these polymeric materials, containing amino acid structures, hold promise as dielectric materials for low-frequency and low-voltage devices such as sensors, owing to their tunable thermal and dielectric properties achieved through click chemistry modification. In the system L-cysteine-H3PO3–H2O, a new salt with a dimeric cation, (L-CysH⋯L-Cys)(H2PO3), has been obtained. The compound exhibits incongruent solubility and is synthesized using an equimolar ratio of L-cysteine and phosphorous acid. It crystallizes in the orthorhombic space group P212121. The dimeric (L-CysH⋯L-Cys) cation is formed due to a strong O–H⋯O hydrogen bond with an O⋯O distance of 2.498(2) Å. Phosphite anions in the structure are linked to infinite chains. Water softening is a treatment process required to manage calcium and magnesium cations in water streams. Moreover, detection of such cations in water samples using simple portable techniques is required for monitoring and inspection of water quality. Nanocomposites can provide solutions for such multiple challenges while showing high performance and cost-effectiveness. In this work, the Cu-Fe layered double hydroxides (LDH)/cysteine-based electrodes were synthesized and are used as active materials for the electrochemical detection of calcium ions. The electrode materials were characterized using XRD, FTIR, and SEM. The synthesized electrode showed a limit of detection and a limit of quantification of 0.21 µM and 0.73 µM, respectively. The concentration range of detection was 5–30 µM. The intermolecular interactions of Ca ions with cysteine-free LDH and cysteine-LDH were investigated by Monte Carlo and molecular dynamics simulations. Monte Carlo simulations revealed that the (001) surface is favored for Ca(II) adsorption within the LDH structure with an energy of adsorption equals to 372.46 kcal/mol. This work paves the road towards developing cost-effective disposable electrode based on portable electrochemical detection of Ca ions for on-site water softening applications. In the ratiometric fluorescent (RF) strategy, the selection of fluorophores and their respective ratios helps to create visual quantitative detection of target analytes. This study presents a framework for optimizing ratiometric probes, employing both two-component and three-component RF designs. For this purpose, in a two-component ratiometric nanoprobe designed for detecting methyl parathion (MP), an organophosphate pesticide, yellow-emissive thioglycolic acid-capped CdTe quantum dots (Y-QDs) (analyte-responsive), and blue-emissive carbon dots (CDs) (internal reference) were utilized. Mathematical polynomial equations modeled the emission profiles of CDs and Y-QDs in the absence of MP, as well as the emission colors of Y-QDs in the presence of MP separately. In other two-/three-component examples, the detection of dopamine hydrochloride (DA) was investigated using an RF design based on blue-emissive carbon dots (B-CDs) (internal reference) and N-acetyl L-cysteine functionalized CdTe quantum dots with red/green emission colors (R-QDs/G-QDs) (analyte-responsive). The colors of binary/ternary mixtures in the absence and presence of MP/DA were predicted using fitted equations and additive color theory. Finally, the Euclidean distance method in the normalized CIE XYZ color space calculated the distance between predicted colors, with the maximum distance defining the real-optimal concentration of fluorophores. This strategy offers a more efficient and precise method for determining optimal probe concentrations compared to a trial-and-error approach. Alkylation of 9-methylthio-nido-carborane under the action of the corresponding iodo derivatives of alanine and homoalanine furnished new (nido-carboran-9-yl) derivatives of (R)-cysteine and (S)-methionine. The synthesized chiral nido-carborane derivatives contain asymmetric carbon and sulfur atoms, as well as a chiral plane. The developed approach to the synthesis of sulfur-containing B(9)-substituted nido-carborane derivatives can be used in the preparation of bioactive compounds, including potential boron delivery agents for boron neutron capture therapy of oncological diseases. In the initial stages of Alopecia Areata (AA), the predominance of hair breakage or exclamation mark hairs serves as vital indicators of disease activity. These signs are non-invasive and are commonly employed in dermatoscopic examinations. Despite their clinical salience, the underlying etiology precipitating this hair breakage remains largely uncharted territory. Our exhaustive review of the existing literature points to a pivotal role for cysteine—a key amino acid central to hair growth—in these mechanisms. This review will probe and deliberate upon the implications of aberrant cysteine metabolism in the pathogenesis of AA. It will examine the potential intersections of cysteine metabolism with autophagy, ferroptosis, immunity, and psychiatric manifestations associated with AA. Cysteine metabolism occurs across cellular compartments to support diverse biological functions and prevent the induction of ferroptosis. Though the disruption of cytosolic cysteine metabolism is implicated in this form of cell death, it is unknown whether the substantial cysteine metabolism resident within the mitochondria is similarly pertinent to ferroptosis. Here, we show that despite the rapid depletion of intracellular cysteine upon loss of extracellular cystine, cysteine-dependent synthesis of Fe-S clusters persists in the mitochondria of lung cancer cells. This promotes a retention of respiratory function and a maintenance of the mitochondrial redox state. Under these limiting conditions, we find that glutathione catabolism by CHAC1 supports the mitochondrial cysteine pool to sustain the function of the Fe-S proteins critical to oxidative metabolism. We find that disrupting Fe-S cluster synthesis under cysteine restriction protects against the induction of ferroptosis, suggesting that the preservation of mitochondrial function is antagonistic to survival under starved conditions. Overall, our findings implicate mitochondrial cysteine metabolism in the induction of ferroptosis and reveal a mechanism of mitochondrial resilience in response to nutrient stress. The relevance of mitochondrial cysteine metabolism to ferroptosis is unknown. Thiol-bearing microcrystalline cellulose was demonstrated for the first time as a substrate for colorimetric detection of urinary cysteine based on the aggregation of silver nanoparticles (AgNPs) on the cellulose surface. The cellulose was functionalized with 3-mercaptopropyl trimethoxysilane and doped with Ag(NH3)2+ (Ag-MCC). The obtained Ag-MCC was used to extract cysteine from samples, given the strong affinity of the thiol group of cysteine toward silver species. After treating the material with NaBH4 solution, AgNPs were produced on the material surface and the aggregation of AgNPs was induced by cysteine. The material color changed from yellow to orange and purple with increasing cysteine concentration. The color intensities were observed using a smartphone and ImageJ software. Under the optimized conditions, the linear working range for cysteine determination was in the range of 0 – 25 µM with the limit of detection (LOD) and limit of quantification (LOQ) of 0.25 and 1.0 µM, respectively. The method was further applied to detect cysteine in human urine samples, and the recovery was found in the range of 81.2 to 110%. This cellulose showed potential as a green material for sensing applications with easy surface modification. Ditylenchus destructor is a migratory plant-parasitic nematode that severely harms many agriculturally important crops. The control of this pest is difficult, thus efficient strategies for its management in agricultural production are urgently required. Cathepsin L-like cysteine protease (CPL) is one important protease that has been shown to participate in various physiological and pathological processes. Here we decided to characterize the CPL gene (Dd-cpl-1) from D. destructor. Analysis of Dd-cpl-1 gene showed that Dd-cpl-1 gene contains a signal peptide, an I29 inhibitor domain with ERFNIN and GNFD motifs, and a peptidase C1 domain with four conserved active residues, showing evolutionary conservation with other nematode CPLs. RT-qPCR revealed that Dd-cpl-1 gene displayed high expression in third-stage juveniles (J3s) and female adults. In situ hybridization analysis demonstrated that Dd-cpl-1 was expressed in the digestive system and reproductive organs. Silencing Dd-cpl-1 in 1-cell stage eggs of D. destructor by RNAi resulted in a severely delay in development or even in abortive morphogenesis during embryogenesis. The RNAi-mediated silencing of Dd-cpl-1 in J2s and J3s resulted in a developmental arrest phenotype in J3 stage. In addition, silencing Dd-cpl-1 gene expression in female adults led to a 57.43% decrease in egg production. Finally, Dd-cpl-1 RNAi-treated nematodes showed a significant reduction in host colonization and infection. Utilizing monoalgal species for wastewater treatment is facing tremendous challenges owing to changing wastewater complexity in terms of physico-chemical characteristic, nutrient and metal concentration. The environmental conditions are also fluctuating therefore, the formation of robust system is of utmost importance for concomitant sustainable wastewater treatment and bioenergy production. In the present study, the tolerance and adaptability potential of algal consortia-1 (Chlorococcum humicola and Tetradesmus sp.) and consortia-2 (Chlorococcum humicola, Scenedesmus vacuolatus and Tetradesmus sp.) treated with municipal wastewater were examined under natural environmental conditions. The results exhibited that consortia-2 was more competent in recovering nitrate-nitrogen (82.92%), phosphorus (70.47%), and heavy metals (31–73.70%) from municipal wastewater (100%) than consortia-1. The results further depicted that total chlorophyll, carbohydrate, and protein content decreased significantly in wastewater-treated consortia-1 as compared to consortia-2. However, lipid content was increased by 4.01 and 1.17 folds in algal consortia-1 and consortia-2 compared to their respective controls. Moreover, absorption peak at 1740.6 cm−1 reflected higher biofuel-producing potential of consortia-1 as compared to consortia-2 as confirmed through FTIR spectroscopy. The results also revealed that consortia-2 showed the highest photosynthetic performance which was evident from the increment in the active photosystem-II reaction center (1.724 ± 0.068), quantum efficiency (0.633 ± 0.038), and performance index (3.752 ± 0.356). Further, a significant increase in photosynthetic parameters was observed in selected consortia at lag phase, while a noteworthy decline was observed at exponential and stationary phases in consortia-1 than consortia-2. The results also showed the maximum enhancement in ascorbic acid (2.43 folds), proline (3.34 folds), and cysteine (1.29 folds) in consortia-2, while SOD (1.75 folds), catalase (2.64 folds), and GR (1.19 folds) activity in consortia-1. Therefore, it can be concluded that due to remarkable flexibility and photosynthetic performance, consortia-2 could serve as a potential candidate for sustainable nutrient resource recovery and wastewater treatment, while consortia-1 for bio-fuel production in a natural environment. The present study deals with the kinetics of improved poly(3-hydroxybutyrate) (PHB) production by an L-cysteine HCl-depressed mutant of Bacillus licheniformis. Production of biodegradable polymers is to eliminate use of materials derived from petrochemicals and also because of their environmental impact. For the current study, mutant strain (NA-21) & wild-type (IIB-isl19) were used for PHB production. Submerged culture with two-stage fermentation technique was used for PHB production. Results indicated that PHB production was improved with 300 mM of –HNO2. The superior mutant strain (NA-21) resulted in 2-fold more PHB as compared to the wild-type (IIB-isl9). It was selected, and resistance against L-cysteine HCl was developed. At 4 ppm concentration of L-cysteine HCl, PHB production by mutant strain (NA-cys4) was higher than its wild counterpart by 5.7-fold. Kinetic study of parameters including specific growth rate (µ h− 1), growth (Yx/s,Ys/x), product yield coefficients (Yp/s,Yp/x), volumetric rate constants (Qp, Qs, Qx) and specific rate constants (qp, qs, qx), were also accomplished. Moreover, Yp/x, Qp and qp = µ × Yp/x were found to be very significant as 1.254 ± 0.06 (g/g biomass), 0.134 ± 0.01 (g/l/h) and 0.168 ± 0.01 (g/g/h), respectively. The effect of fatty acids on PHB production highlighted the improvement in PHB production by 1.94-fold. The highest PHB production during the study was 16.35 ± 3.12 g/l which highlighted its significance (p ≤ 0.05) and impact on the overall process. The variation in PBH yield between wild-type and mutant B. licheniformis is possibly because of induced DNA interstrand thus making unstable thymidine-thymidine dymers. Background: Odontogenic cysts/tumor can cause severe bone destruction, which affects maxillofacial function and aesthetics. Meanwhile, metabolic reprogramming is an important hallmark of diseases. Changes in metabolic flow affect all aspects of disease, especially bone-related diseases. At present, the researches on pathogenesis of odontogenic cysts/tumor are mainly focused on the level of gene regulation, but the effects of metabolic alterations on odontogenic cysts/tumor have still underexplored. Materials and methods: Imaging analysis was used to evaluate the lesion size of different odontogenic lesions. Tartrate resistant acid phosphatase (TRAP) and immunohistochemistry (IHC) assays were utilized to detect the differences in bone destruction activity in odontogenic cysts and tumors. Furthermore, metabolomics and weighted gene co-expression network analysis (WGCNA) were conducted for the metabolomic features and key metabolite screening, respectively. The effect of ferroptosis inhibition on bone destruction was confirmed by IHC, immunofluorescence, and malondialdehyde colorimetric assay. Results: The bone destruction activity of ameloblastoma (AM) was the strongest and the weakest in odontogenic cysts (OC). High-throughput targeted metabolomics was used to map the metabolomic profiles of OC, odontogenic keratocyst (OKC) and AM. WGCNA and differential analysis identified L-cysteine in OKC and AM. Cystathionine γ-lyase (CTH) was further screened by Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis. The functions of L-cysteine were further validated. Finally, we confirmed that CTH affected destructive activities by regulating the sensitivity of epithelial cells to ferroptosis. Conclusion: High-throughput targeted metabolomics performed on diseased tissue confirmed the unique alteration of metabolic profiles in OKC and AM. Cysteine string protein (CSP) is a member of the DnaJ/Hsp40 family of molecular chaperones. CSP is enriched in neurons, where it mainly localises to synaptic vesicles. Mutations in CSP-encoding genes in flies, worms, mice and humans result in neuronal dysfunction, neurodegeneration and reduced lifespan. Most attention has therefore focused on CSP's neuronal functions, although CSP is also expressed in non-neuronal cells. Here, we used genome editing to fluorescently tag the Caenorhabditis elegans CSP orthologue, dnj-14, to identify which tissues preferentially express CSP and hence may contribute to the observed mutant phenotypes. Replacement of dnj-14 with wrmScarlet caused a strong chemotaxis defect, as seen with other dnj-14 null mutants. In contrast, inserting the reporter in-frame to create a DNJ-14-wrmScarlet fusion protein had no effect on chemotaxis, indicating that C-terminal tagging does not impair DNJ-14 function. WrmScarlet fluorescence appeared most obvious in the intestine, head/pharynx, spermathecae and vulva/uterus in the reporter strains, suggesting that DNJ-14 is preferentially expressed in these tissues. Crossing the DNJ-14-wrmScarlet strain with GFP marker strains confirmed the intestinal and pharyngeal expression, but only a partial overlap with neuronal GFP was observed. DNJ-14-wrmScarlet fluorescence in the intestine was increased in response to starvation, which may be relevant to mammalian CSPα's role in microautophagy. DNJ-14's enrichment in worm reproductive tissues (spermathecae and vulva/uterus) parallels the testis-specific expression of CSPβ and CSPγ isoforms in mammals. E. coli exposure to ciprofloxacin disturbs cysteine homeostasis; an increase in the intracellular concentration of cysteine is dangerous due to its ability to enhance ROS generation. Unlike wild-type bacteria, in which the cysteine content did not exceed the control level, cells of the gshA mutant lacking glutathione are characterized by increased concentration of intracellular cysteine in proportion to the concentrations of the antibiotic, despite the intensive export of cysteine into the medium. To conveniently monitor bioactive cysteine (Cys) and Fe2+ in practice, a kind of poly-β-cyclodextrin strengthen praseodymium oxide (Pr6O11) porous oxidase mimic (p-β-CD@Pr6O11) was constructed by virtue of the strong coordination between nano Pr6O11 and poly-β-cyclodextrin substrate. After its microstructure and physicochemical property were characterized in detail, it was noted that porous p-β-CD@Pr6O11 exhibited excellent enzyme-like catalytic activity to accelerate the oxidation of 3,3′,5,5,′-tetramethylbanzidine (TMB) and 2,2′-azinobis (3-ethylbenzo-thiazoline-6-sulfonic acid) ammonium salt (ABTS) with significant color-enhancement effect in the air. Based on the signal amplification, trace Cys could exclusively deteriorate the UV-vis absorbance at 653 nm of p-β-CD@Pr6O11-TMB and Fe2+ alter the one at 729 nm of p-β-CD@Pr6O11-ABTS with visual color changes. Introduction: Osteoporosis is a global health issue. Bisphosphonates that are commonly used to treat osteoporosis suppress both bone resorption and subsequent bone formation. Inhibition of cathepsin K, a cysteine proteinase secreted by osteoclasts, was reported to suppress bone resorption while preserving or increasing bone formation. Analyses of the different effects of antiresorptive reagents such as bisphosphonates and cysteine proteinase inhibitors will contribute to the understanding of the mechanisms underlying bone remodeling. Materials and Methods: Our team has developed an in vitro system in which bone remodeling can be temporally observed at the cellular level by 2-photon microscopy. We used this system in the present study to examine the effects of the cysteine proteinase inhibitor E-64 and those of zoledronic acid on bone remodeling. Results: In the control group, the amount of the reduction and the increase in the matrix were correlated in each region of interest, indicating the topological and quantitative coordination of bone resorption and formation. Parameters for osteoblasts, osteoclasts, and matrix resorption/formation were also correlated. E-64 disrupted the correlation between resorption and formation by potentially inhibiting the emergence of spherical osteoblasts, which are speculated to be reversal cells in the resorption sites. The protein S-nitrosylation (SNO) is a significant post-translational modification that affects the stability, activity, cellular localization, and function of proteins. Therefore, highly accurate prediction of SNO sites aids in grasping biological function mechanisms. In this document, we have constructed a predictor, named PPSNO, forecasting protein SNO sites using stacked integrated learning. PPSNO integrates multiple machine learning techniques into an ensemble model, enhancing its predictive accuracy. First, we established benchmark datasets by collecting SNO sites from various sources, including literature, databases, and other predictors. Second, various techniques for feature extraction are applied to derive characteristics from protein sequences, which are subsequently amalgamated into the PPSNO predictor for training. Five-fold cross-validation experiments show that PPSNO outperformed existing predictors, such as PSNO, PreSNO, pCysMod, DeepNitro, RecSNO, and Mul-SNO. The PPSNO predictor achieved an impressive accuracy of 92.8%, an area under the curve (AUC) of 96.1%, a Matthews correlation coefficient (MCC) of 81.3%, an F1-score of 85.6%, an SN of 79.3%, an SP of 97.7%, and an average precision (AP) of 92.2%. We also employed ROC curves, PR curves, and radar plots to show the superior performance of PPSNO. Our study shows that fused protein sequence features and two-layer stacked ensemble models can improve the accuracy of predicting SNO sites, which can aid in comprehending cellular processes and disease mechanisms. Background : Cystinosis is a rare autosomal recessive lysosomal disorder that mainly affects the kidney and eye. Early treatment with cysteamine significantly improves the prognosis. However, early diagnosis of cystinosis, especially the juvenile nephropathic form, remains challenging because typical symptoms only become apparent in adulthood. We herein describe a 13-year-old girl who presented with proteinuria only but was diagnosed with juvenile nephropathic cystinosis based on multinucleated podocytes in her kidney biopsy specimen. We also studied the nephropathology of another case to determine the features of the multinucleated podocytes. Case diagnosis: A previously healthy 13-year-old girl presented to our hospital because proteinuria had been detected in her school urine screening. She had been noted to have proteinuria on her school urine screening when she was 11 years of age but there was no consultation with her physician at that time. She was asymptomatic and had no other abnormalities on examination other than a relatively high urinary β-2 microglobulin level. Her kidney biopsy showed 15 multinucleated podocytes in 34 glomeruli, and the mean number of nuclei per multinucleated podocyte was 4.4. Ophthalmological examination showed cystine crystals in her cornea. Her white blood cell cystine level was high, and she was diagnosed with juvenile nephropathic cystinosis. She started oral cysteamine treatment and showed almost no progression of the disease after 2 years. In another patient with juvenile nephropathic cystinosis, there were 25 multinucleated podocytes in 63 glomeruli, and the mean number of nuclei per multinucleated podocyte was 2.9. Conclusion: Early diagnosis is crucial to improve the prognosis of patients with cystinosis. Gold(III) complexes are widely studied as antitumor agents and show good results. The interaction with biologically active thiols (thiomalate, cysteine, glutathione (GSH) and human serum albumin) of a number of gold(III) complexes with N-containing polydentate ligands in aqueous solution with pH 7.4 and 0.2 M NaCl was studied. Complexes with 1,10-phenanthroline and 2,2′-bipyridyl, Au(phen)(OH)2+ and Au(bipy)(OH)2+, react fast with an excess of any of these thiols and in less than a few seconds transform into gold(I) bis-thiolate complexes. For complexes with deprotonated ethylenediamine and diethylenetriamine, Au(en)(en-H)2+ and Au(dien-H)(Cl,OH)+, at a significant excess of GSH, a relatively long-lived gold(III) complex AuIII(GSH)iLj is formed. At t = 37 °C, it transforms into the gold(I) bis-thiolate complex Au(GSH)2 by 90% in 4 h. However, for other thiols, the rate of decomposition of similar complexes is about 10 times higher. The click chemistry reactions and its use in living organisms have been widely developed; however, few studies describe the safety of the reagents and products. Furan–maleimide represent one important pair of reagents for the click Diels–Alder reaction, and were chosen here to perform the minimalist click reaction with cysteine to reveal their preliminary safety features by cytotoxicity tests. The functional groups of amino acids allow them to be chemically modified, mainly for the synthesis of monomers and polymers. Here, cysteine was used in the click reaction, since it displays an SH group in its side chain, useful to perform thiol–ene reactions. Its amine group easily reacts with anhydrides, allowing it to couple with the furan derivative "furan–maleic anhydride adduct". The mixture of the cysteine with the adduct in solution gives rise to the double simultaneous thiol–ene/amine maleimide click reactions. The thiol group and the furan double bond provide the reactive functional groups for the polymerization. The synthesis of the polymer based on cysteine–Diels–Alder adduct confirmed the condensation nature of the reaction. Preliminary safety and biocompatibility tests of this new polymer and its starting reagents were carried out, since these parameters represent one of the most important features for operating and scaling up the production. L-cysteine is an amino acid with relevance to the pharmaceutical, food, feed, and cosmetic industry. The environmental and societal impact of its chemical production has led to the development of more sustainable fermentative L-cysteine production processes with engineered E. coli based on glucose and thiosulfate as sulphur source. Still, most of the published processes show low yields. For the identification of further metabolic engineering targets, engineered E. coli cells were withdrawn from a fed-batch production process, followed by in vivo metabolic control analysis (MCA) based on the data of short-term perturbation experiments, metabolomics (LC–MS), and thermodynamic flux analysis (TFA). In vivo MCA indicated that the activities of the L-cysteine synthases of the cells withdrawn from the production process might be limiting, and we hypothesised that the L-cysteine precursor O-acetylserine (OAS) might be exported from the cells faster than it took to transform OAS into L-cysteine. By increasing the expression of the L-cysteine synthases, either sulfocysteine synthase or L-cysteine synthase, which transform OAS into L-cysteine, an improvement of up to 70% in specific L-cysteine productivity and up to 47% in the final L-cysteine concentration was achieved in standardised fed-batch processes thereby increasing the yield on glucose by more than 85 to 9.2% (w/w). The high-protein diet (HPD) has emerged as a potent dietary approach to curb obesity. Peroxisome, a highly malleable organelle, adapts to nutritional changes to maintain homeostasis by remodeling its structure, composition, and quantity. However, the impact of HPD on peroxisomes and the underlying mechanism remains elusive. Using Drosophila melanogaster as a model system, we discovered that HPD specifically increases peroxisome levels within the adipose tissues. This HPD-induced peroxisome elevation is attributed to cysteine and methionine by triggering the expression of CG33474, a fly homolog of mammalian PEX11G. Both the overexpression of Drosophila CG33474 and human PEX11G result in increased peroxisome size. In addition, cysteine and methionine diets both reduce lipid contents, a process that depends on the presence of CG33474. Furthermore, CG33474 stimulates the breakdown of neutral lipids in a cell-autonomous manner. Moreover, the expression of CG33474 triggered by cysteine and methionine requires TOR signaling. Finally, we found that CG33474 promotes inter-organelle contacts between peroxisomes and lipid droplets (LDs), which might be a potential mechanism for CG33474-induced fat loss. In summary, our findings demonstrate that CG33474/PEX11G may serve as an essential molecular bridge linking HPD to peroxisome dynamics and lipid metabolism. Inhibition of Helicobacter pylori urease is an effective method in the treatment of several gastrointestinal diseases in humans. This bacterium plays an important role in the pathogenesis of gastritis and peptic ulceration. Considering the presence of cysteine and N-arylacetamide derivatives in potent urease inhibitors, here, we designed hybrid derivatives of these pharmacophores. Therefore, cysteine-N-arylacetamide derivatives 5a-l were synthesized through simple nucleophilic reactions with good yield. In vitro urease inhibitory activity assay of these compounds demonstrated that all newly synthesized compounds exhibited high inhibitory activity (IC50 values = 0.35–5.83 μM) when compared with standard drugs (thiourea: IC50 = 21.1 ± 0.11 μM and hydroxyurea: IC50 = 100.0 ± 0.01 μM). Representatively, compound 5e with IC50 = 0.35 μM was 60 times more potent than strong urease inhibitor thiourea. Enzyme kinetic study of this compound revealed that compound 5e is a competitive urease inhibitor. Moreover, a docking study of compound 5e was performed to explore crucial interactions at the urease active site. This study revealed that compound 5e is capable to inhibit urease by interactions with two crucial residues at the active site: Ni and CME592. Furthermore, a molecular dynamics study confirmed the stability of the 5e-urease complex and Ni chelating properties of this compound. The obtained fish protein hydrolysates (FPHs) are rich in peptides and amino acids, but bitterness and aroma defects impede further utilization of FPHs. The present study adopted Maillard reaction to improve FPHs' flavor and illustrated the role of cysteine in this system. We investigated the impact of cysteine (0, 0.25%, 0.5%, 0.75%, and 1%) on the browning intensity, free amino acids (FAAs), molecular weight distribution, structure of MRPs, volatile compounds changes and organoleptic characteristics of xylose–glycine–FPHs Maillard reaction systems. Results showed that the addition of cysteine lowered the browning degree of Maillard reaction products (MRPs) by inhibiting the cross-linking of small peptides and reducing the production of melanin. GC–MS and GC–IMS analysis indicated that cysteine inhibited the formation of furans and nitrogen-containing compounds and facilitated the formation of sulfur-containing compounds contributing to the meaty flavor. Sensory analysis revealed that 0.25–0.75% range of cysteine increased the meaty, caramel, umami, mouthfulness and salty notes, and caused a decrease in bitter taste of the MRPs as confirmed by GC–MS. A highly significant correlation between the organoleptic characteristics and physicochemical indicators of MRPs was found by Mantel test. The research focuses on utilizing gamma irradiation to synthesize polyacrylic acid-co-polyacrylamide p(AAm-co-AAc) hydrogels. The effect of synthetic parameters on physicochemical features of p(AAm-co-AAc) hydrogls were examined, including acrylic acid (AAc): acrylamide (AAm) weight ratios, monomer concentration, and gamma irradiation dosage (kGy). At the optimum synthetic conditions (30 kGy and 75% AAc), different chemical modifications are explored to incorporate sulfonate, hydroxyl, carboxyl, cysteine, thiol, and amine functional groups within the bare hydrogel (Cpd 0) structure. Fourier-transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM) analyses confirmed the success development of functionalized hydrogels (namely Cpd 1 to 6) with three-dimensional porous structures. These modified hydrogels include Cpd 1, a sulfonated hydrogel through a sulfonation reaction; Cpd 2, modified via NaOH hydrolysis; Cpd 3, modified using thionyl chloride; Cpd 4, incorporating cysteine modification through reaction with cysteine; Cpd 5, with 4-(Dimethylamino) benzaldehyde; and Cpd 6, modified with 3,4-Dimethylbenzoic acid.The effect of hydrogel composition and surface functionalities on the swelling capacity and interactions with scale-forming/heavy metal ions (e.g., Ba2+, Sr2+, and Cu2+) was investigated in saline water solution (NaCl = 1000 mg/L). Batch adsorption studies reveal that all modified hydrogels exhibited higher removal efficiency for the three metal ions than unmodified p(AAm-co-AAc) hydrogel, validating the key role of surface functionalities in tailoring hydrogel affinity for metal ions adsorption. Amongst these, NaOH-treated hydrogel (Cpd 2) outperformed all other modified ones in the removal of Cu2+, Ba2+, and Sr2+ ions, with maximum capacities of 13.67, 36.4, and 27.31 mg/g, respectively. Based on adsorption isotherm and kinetic modeling, the adsorption process of the three metal ions onto all modified hydrogels better obeyed Freundlich isotherm and pseudo-first-order kinetic models. Thermodynamic studies also indicated that the adsorption behavior of Sr2+ ions can exhibit both exothermic and endothermic characteristics, depending on the nature of hydrogel surface chemistry. Conversely, the adsorption process of Cu2+ and Ba2+ ions onto all modified hydrogels is endothermic, suggesting favorable chemical adsorption mechanisms. These findings reveal that the specific adsorption performance of hydrogel is dependent on the type of modification and the targeted heavy metal ions. Based on the nature of hydrogel surface functionality, surface modifications can change the charge density, hydrophilicity, and overall chemical environment of the hydrogel, offering a versatile approach to optimize the adsorption affinity/selectivity of hydrogel's in removing scale-forming/heavy metals from water solutions. Human rhinovirus picornain 3C is a high-value commercial cysteine protease, which is widely used to remove affinity tags and fusion proteins during the purification of the target proteins. A variant of rhinovirus A28 picornain 3C produced in this study is not annotated in the NCBI databases, shares 79% sequence identity in the PDB, and was not previously used in the protein engineering. A protocol was developed for the isolation and purification of the protein to use it in structural studies. The initial crystallization conditions were found. The determination and analysis of the structure of rhinovirus A28 picornain 3C will provide new possibilities for performing basic research on the evolution of proteolytic enzymes and for the design of the optimal variant of this protease. Single-chain monellin (SCM) is an engineered protein that links the two chains of monellin, a naturally sweet-tasting protein. This protein is an attractive candidate for use as a sugar replacement in food and beverages and has numerous other applications. Therefore, generating SCM mutants with improved stability is an active area of research to broaden the range of its potential applications. In this study, we focused on the Cys41 residue of SCM, which is a single cysteine residue present at a structurally important position. This residue is often substituted with Ser. However, this substitution may destabilize SCM because Cys41 is buried in the hydrophobic core of the protein. Therefore, we designed mutants that substituted Ala, Val, and Leu for this residue, namely C41A, C41V, and C41L. We characterized these three mutants, SCM C41S, and wild type (WT). Differential scanning fluorimetric analysis revealed that substituting Cys41 with Ala or Val increased the thermal stability of SCM, while substitution with Ser or Leu decreased its stability. Determination of the crystal structures of SCM C41A and C41V mutants revealed that the overall structures and main chain structures around the 41st residue of both mutants were almost identical to the WT. On the other hand, the orientations of the amino acid side chains near the 41st residue differed among the SCM variants. Taken together, our results indicate that substituting Cys41 with Ala or Val increases the stability of SCM and provide insight into the structural basis of this improvement. [ABSTRACT FROM AUTHOR]

The synthesis of gold nanoparticles (AuNPs) by bottom-up methods, such as redox reactions using amino acids and gold salts, has turned out to be a novel method for obtaining nanoparticles due to the reducing properties of these biomolecules and the ability to give the nanoparticle peculiar physicochemical characteristics for its biological application, thus derived from the known structure and amino acids functional groups. In this sense, this work shows the characterization using UV-Vis, DLS, FTIR, XPS, and HRTEM techniques of AuNPs synthesized using sodium borohydride (NaBH4) as a reducing compound and L-cysteine methyl ester hydrochloride (cysteine precursor) (HSCH2CH (NH2) COOCH3 • HCl) as a stabilizing agent. The above elucidates the reaction mechanisms for the formation of the nanoparticle through these reactions, as well as the stabilizing action and possible reducing potential of cysteine. Likewise, the resulting Cis@AuNP compounds were subjected to a preliminary biological evaluation using cell viability toxicity tests. The Cis@AuNPs showed high colloidal stability in a pH range of 3 to 11, where the L-cysteine methyl ester hydrochloride functional groups strongly influenced the hydrodynamic diameter and zeta potential behavior. Cytotoxicity assays in mouse leukocytes demonstrated the safety of these nanoparticles. Abstract—Low molecular-weight thiols as glutathione and cysteine are an important part of the cell's redox regulation system. Previously, we have shown that inactivation of ADP-heptose synthesis in Escherichiacoli with a gmhA deletion induces the oxidative stress. It is accompanied by rearrangement of thiol homeostasis and increased sensitivity to antibiotics. In our study, we found that restriction of cysteine metabolism (∆cysB and ∆cysE) and inhibition of glutathione synthesis (∆gshAB) lead to a decrease in the sensitivity of the ∆gmhA mutant to antibiotics but not to its expected increase. At the same time, blocking of the export of cysteine (∆eamA) or increasing import (Ptet-tcyP) into cells of the oxidized form of cysteine–cystine leads to an even greater increase in the sensitivity of gmhA-deleted cells to antibiotics. In addition, there is no correlation between the cytotoxic effect of antibiotics and the level of reactive oxygen species (ROS), the total pool of thiols, or the viability of the initial cell population. However, a correlation between the sensitivity to antibiotics and the level of oxidized glutathione in cells was found in our study. Apparently, a decrease in the content of low-molecular-weight thiols saves NADPH equivalents and limits the processes of protein redox modification. This leads to increasing of resistance of the ∆gmhA strain to antibiotics. An increase in low-molecular-weight thiols levels requires a greater expenditure of cell resources, leads to an increase in oxidized glutathione and induces to greater increase in sensitivity of the ∆gmhA strain to antibiotics. The robustness of NMR coherence transfer in proximity of a paramagnetic center depends on the relaxation properties of the nuclei involved. In the case of Iron-Sulfur Proteins, different pulse schemes or different parameter sets often provide complementary results. Tailored versions of HCACO and CACO experiments significantly increase the number of observed Cα/C' connectivities in highly paramagnetic systems, by recovering many resonances that were lost due to paramagnetic relaxation. Optimized 13C direct detected experiments can significantly extend the available assignments, improving the overall knowledge of these systems. The different relaxation properties of Cα and C' nuclei are exploited in CACO vs COCA experiments and the complementarity of the two experiments is used to obtain structural information. The two [Fe2S2]+ clusters containing NEET protein CISD3 and the one [Fe4S4]2+ cluster containing HiPIP protein PioC have been taken as model systems. We show that tailored experiments contribute to decrease the blind sphere around the cluster, to extend resonance assignment of cluster bound cysteine residues and to retrieve details on the topology of the iron-bound ligand residues. Acetaminophen (APAP) belong among the most used analgesics and antipyretics. It is structurally derived from p-aminophenol (PAP), a potent inducer of kidney toxicity. Both compounds can be metabolized to oxidation products and conjugated with glutathione. The glutathione-conjugates can be cleaved to provide cysteine conjugates considered as generally nontoxic. The aim of the present report was to synthesize and to purify both APAP- and PAP-cysteine conjugates and, as the first study at all, to evaluate their biological effects in human kidney HK-2 cells in comparison to parent compounds. HK-2 cells were treated with tested compounds (0–1000 µM) for up to 24 h. Cell viability, glutathione levels, ROS production and mitochondrial function were determined. After 24 h, we found that both APAP- and PAP-cysteine conjugates (1 mM) were capable to induce harmful cellular damage observed as a decrease of glutathione levels to 10% and 0%, respectively, compared to control cells. In addition, we detected the disappearance of mitochondrial membrane potential in these cells. In the case of PAP-cysteine, the extent of cellular impairment was comparable to that induced by PAP at similar doses. On the other hand, 1 mM APAP-cysteine induced even larger damage of HK-2 cells compared to 1 mM APAP after 6 or 24 h. We conclude that cysteine conjugates with aminophenol are potent inducers of oxidative stress causing significant injury in kidney cells. Two non-metallicolous and metallicolous populations of harmel plants were compared regarding the role of proline, cysteine, reducing sugars, hydrogen peroxide (H2O2), glutathione, thiol compounds, organic acids, total free amino acids, and lipid peroxidation in detoxification and tolerance of silver stress (0, 1, 2.5, 5, 10 ppm Ag). The results of the present research state that the effects of Ag were increased total free amino acids, glutathione, organic acids, proline, reducing sugars, thiol compounds, and cysteine, so the accumulation of these compounds was higher in metallicolous populations than non-metallicolous. On the other hand, non-metallicolous populations showed higher content of lipid peroxidation and H2O2 than metallicolous populations under Ag stresses. Also, the accumulation of phytochelatins (PC) was observed with increasing Ag concentration, which shows that compared to glutathione, non-protein thiols have a higher concentration. The number of organic acids (malic acids, fumaric, oxalic, and citric) except acetic acid increased in the leaves of harmel in both populations. According to the results of this research, the harmel metallophilic population has a crucial role in the tolerance and detoxification of Ag stress, so the antioxidant responses of the plant against Ag stress in the non-metallicolous population were lower than the metallicolous population.
