Key message: Fine-mapping of a locus on chromosome 1 of flax identified an S-lectin receptor-like kinase (SRLK) as the most likely candidate for a major Fusarium wilt resistance gene. Fusarium wilt, caused by the soil-borne fungal pathogen Fusarium oxysporum f. sp. lini, is a devastating disease in flax. Genetic resistance can counteract this disease and limit its spread. To map major genes for Fusarium wilt resistance, a recombinant inbred line population of more than 700 individuals derived from a cross between resistant cultivar ‘Bison’ and susceptible cultivar ‘Novelty’ was phenotyped in Fusarium wilt nurseries at two sites for two and three years, respectively. The population was genotyped with 4487 single nucleotide polymorphism (SNP) markers. Twenty-four QTLs were identified with IciMapping, 18 quantitative trait nucleotides with 3VmrMLM and 108 linkage disequilibrium blocks with RTM-GWAS. All models identified a major QTL on chromosome 1 that explained 20–48% of the genetic variance for Fusarium wilt resistance. The locus was estimated to span ~ 867 Kb but included a ~ 400 Kb unresolved region. Whole-genome sequencing of ‘CDC Bethune’, ‘Bison’ and ‘Novelty’ produced ~ 450 Kb continuous sequences of the locus. Annotation revealed 110 genes, of which six were considered candidate genes. Fine-mapping with 12 SNPs and 15 Kompetitive allele-specific PCR (KASP) markers narrowed down the interval to ~ 69 Kb, which comprised the candidate genes Lus10025882 and Lus10025891. The latter, a G-type S-lectin receptor-like kinase (SRLK) is the most likely resistance gene because it is the only polymorphic one. In addition, Fusarium wilt resistance genes previously isolated in tomato and Arabidopsis belonged to the SRLK class. Few natural antisense (as) RNAs have been reported as yet in the unicellular protozoan Leishmania. Here, we describe that Leishmania produces natural asRNAs complementary to all ribosomal RNA (rRNA) species. Interestingly, we show that drug-induced apoptosis-like programmed cell death triggers fragmentation of asRNA complementary to the large subunit gamma (LSU-γ) rRNA, one of the six 28S rRNA processed fragments in Leishmania. Heat and oxidative stress also induce fragmentation of asrRNA, but to a lesser extent. Extensive asrRNA cleavage correlates with rRNA breakdown and translation inhibition. Indeed, overexpression of asLSU-γ rRNA accelerates rRNA degradation upon induction of apoptosis. In addition, we provide mechanistic insight into the regulation of apoptosis-induced asrRNA fragmentation by a 67 kDa ATP-dependent RNA helicase of the DEAD-box subfamily. This helicase binds both sense (s)LSU-γ and asLSU-γ rRNAs, and appears to have a key role in protecting rRNA from degradation by preventing asrRNA cleavage and thus cell death. Remarkably, the asrRNA fragmentation process operates not only in trypanosomatid protozoa but also in mammals. Our findings uncover a novel mechanism of regulation involving asrRNA fragmentation and rRNA breakdown, that is triggered by apoptosis and conditions of reduced translation under stress, and seems to be evolutionary conserved. Preharvest sprouting (PHS) in spring wheat ( Triticum aestivum L.) causes significant economic losses due to a reduction in grain functionality, grain yield and viability of seed for planting. Genetic resistance to PHS reduces these losses. Development of PHS resistant cultivars is complicated by the effects of genotype, environment, kernel diseases and spike morphological factors. RL4137 has consistently exhibited high levels of resistance to PHS over years and environments. The mean PHS scores of Canada Western Red Spring (CWRS) wheat cultivars with RL4137 in their ancestry are lower than that of CWRS wheat cultivars without. RL4137 has two mechanisms for PHS resistance, one associated with kernel color and the other not associated with kernel color. RL4137 was the source of PHS resistance in white wheats HY361, AC Vista, Snowbird, Kanata, and Snowstar, all of which had significantly lower PHS scores than the white-seeded check, Genesis. Known DNA markers relating to PHS were used to compare haplotypes with and without RL4137 in the ancestry. Coefficients of parentage also demonstrated the relationship. Because cultivars that have RL4137 in their ancestry were grown on about 77% of the spring wheat area for 2003–2007, RL4137 continues to contribute to protecting market grade from preharvest sprouting. Hard white wheat ( Triticum aestivum L.) is a value-added product because of its processing advantages over red wheat; however, white wheat tends to be more susceptible to pre-harvest sprouting (PHS). To identify quantitative trait loci (QTLs) associated with PHS tolerance, we developed a doubled haploid (DH) mapping population from the cross AC Domain (red seeded) × White-RL4137 (white seeded). A genetic map was constructed using microsatellite markers located on chromosome groups 3, 4, 5 and 6. A population of 174 DH lines was characterized for important aspects of PHS including sprouting index, germination index, Hagberg falling number and seed coat colour. A total of 11 QTLs were identified on group 3 chromosomes and on chromosome 5D. Seven QTLs associated with the PHS traits were found to be co-incident with seed coat colour on chromosomes 3A, 3B and 3D. The 5D PHS QTL was notable because it is independent of seed coat colour. Development of high-yielding wheat varieties with good end-use quality has always been a major concern for wheat breeders. To genetically dissect quantitative trait loci (QTLs) for yield-related traits such as grain yield, plant height, maturity, lodging, test weight and thousand-grain weight, and for quality traits such as grain and flour protein content, gluten strength as evaluated by mixograph and SDS sedimentation volume, an F1-derived doubled haploid (DH) population of 185 individuals was developed from a cross between a Canadian wheat variety “AC Karma” and a breeding line 87E03-S2B1. A genetic map was constructed based on 167 marker loci, consisting of 160 microsatellite loci, three HMW glutenin subunit loci: Glu-A1, Glu-B1 and Glu-D1, and four STS-PCR markers. Data for investigated traits were collected from three to four environments in Manitoba, Canada. QTL analyses were performed using composite interval mapping. A total of 50 QTLs were detected, 24 for agronomic traits and 26 for quality-related traits. Many QTLs for correlated traits were mapped in the same genomic regions forming QTL clusters. The largest QTL clusters, consisting of up to nine QTLs, were found on chromosomes 1D and 4D. HMW glutenin subunits at Glu-1 loci had the largest effect on breadmaking quality; however, other genomic regions also contributed genetically to breadmaking quality. Resistance based on slow-rusting genes has proven to be a useful strategy to develop wheat cultivars with durable resistance to rust diseases in wheat. However this type of resistance is often difficult to incorporate into a single genetic background due to the polygenic and additive nature of the genes involved. Therefore, markers, both molecular and phenotypic, are useful tools to facilitate the use of this type of resistance in wheat breeding programs. We have used field assays to score for both leaf and yellow rust in an Avocet- YrA × Attila population that segregates for several slow-rusting leaf and yellow rust resistance genes. This population was analyzed with the AFLP technique and the slow-rusting resistance locus Lr46/Yr29 was identified. A common set of AFLP and SSR markers linked to the Lr46/Yr29 locus was identified and validated in other recombinant inbred families developed from single chromosome recombinant populations that segregated for Lr46. These populations segregated for leaf tip necrosis (LTN) in the field, a trait that had previously been associated with Lr34/Yr18. We show that LTN is also pleiotropic or closely linked to the Lr46/Yr29 locus and suggest that a new Ltn gene designation should be given to this locus, in addition to the one that already exists for Lr34/Yr18. Coincidentally, members of a small gene family encoding β-1 proteasome subunits located on group 1L and 7S chromosomes implicated in plant defense were linked to the Lr34/Yr18 and Lr46/Yr29 loci. Molecular markers based on DNA sequence variations of the coding and/or promoter regions of the wheat (Triticum aestivum L.) HMW glutenin genes located at the Glu-1 loci were developed. Markers characteristic of alleles Glu-A1-1a (encoding Ax1 subunit) and Glu-A1-1c (encoding Ax2* subunit) at the Glu-A1 locus, alleles Glu-B1ak (encoding Bx7* subunit) and Glu-B1al for overexpressed Bx7 subunit at the Glu-B1 locus and alleles Glu-D1-1a (encoding Dx2 subunit) and Glu-D1-1d (encoding Dx5 subunit) at the Glu-D1 locus were tested using genomic DNA of haploid leaf tissue. A method for simultaneously extracting DNA from 96 haploid leaf tissue pieces is described. Two of the developed markers were dominant and two were co-dominant. A F1-derived population segregating for all HMW glutenin genes was used to test the validity of the markers and their usefulness in doubled haploid breeding programs. SDS-PAGE analysis of seed storage protein was performed on seeds from the doubled haploid lines. A total of 299 lines were tested with the DNA markers on the haploid tissue and validated by protein analysis of the corresponding DH seeds. PCR markers and SDS-PAGE analysis showed between 2 and 8.5% discrepancies depending on the marker. Applications of DNA markers for gene-assisted-selection of haploid tissue and use in breeding programs are discussed. Advantages and disadvantages of dominant and co-dominant markers are outlined. Branch sampling of branch diameter and fruit crop on 22 species of Barbadian trees and shrubs provided sufficient data to build regressions between plant size and fruit crop weight. Orchard plants bear much more fruit than wild, feral or garden plants of similar size, but this difference disappears in multiple regression of fruit crop weight (F in g, fresh mass) on branch or stem diameter (D in cm) and individual fruit weight (W in g): F=22D W. This explains 89% of the variation in F and successfully predicts crop weight for wild tropical and temperate trees and shrubs, but underestimated the crops on commercial, temperate, fruit trees by an order of magnitude. Comparisons of crop weight for feral, wild, and garden plants (F) using a simple regression F=47D show that crop weight is a minor load relative to branch weight for larger branches. Although fruit crops represent a declining proportion of total plant weight as plants become larger, the crops become larger relative to leaf and twig weight and in this sense, reproductive investment increases in larger plants. Finally, our equations, combined with the self-thinning rule, suggest that stands of large species of fruit plants produce more fruit per unit of land area than stands of small ones. We identified sources of mapping inaccuracy during the construction of RFLP linkage maps from one F2 population and two F1 microspore-derived populations from the same cross of oilseed Brassica napus. The genetic maps were compared using a total of 145 RFLP marker loci including 82 loci common to all three populations. In the process, we identified a series of mapping events that could lead to ambigous conclusions. Superimposed restriction fragments could be mistaken as a single dominant restriction fragment in a F2 population and, when analyzed as such, would yield inaccurate linkage information. Residual heterozygosity in parental lines resulted in complicated allelic assignment and yielded subsequent difficulties in linkage determination. Loose and spurious linkages occurred during mapping and were identified by comparing maps derived from different populations. LOD scores and χ2 test of independence were compared for their capacity to detect loose linkages or generate spurious ones. Extreme segregation distortions towards the same parental allele also contributed to an additional source of spurious linkage. Small but significant segregation distortions resulted in reduced estimates of the recombination fraction. The use of the same ‘probe× enzyme’ combinations in doubled haploid populations allowed the identification of the correct allele assignment as well as loose and spurious linkages. A translocation between two homoeologous linkage groups was observed. The consequences of such a chromosomal event as a source of error in mapping applications are discussed. [ABSTRACT FROM AUTHOR]

RFLP segregation analyses were performed on a F population and two F microspore-derived populations from the same cross between a microspore culture-responsive parent ('Topas') and a non-responsive parent ('Westar'). A total of 145 loci were detected with 87 cDNA clones. Eighty-two markers were common across all three populations. A total of 66 markers was assembled into 18 linkage groups and 16 markers remained unlinked. Segregation distortions were significant for 29% of the markers in the F population and 23% and 31% in microspore-derived populations M3 and M5, respectively. An equivalent number of markers showed biased segregation towards each parental allele in the F population while more markers showed a significant deviation from the expected Mendelian ratio towards the responsive parent in both microspore-derived populations. Different subsets of markers showed segregation distortions in the three populations indicating that the selective pressures leading to microsporederived plants are different from those acting during selfing of the F.
