In the adult mouse brain, perineuronal net (PNN), a highly structured extracellular matrix, surrounds subsets of neurons. The AZGP1 gene encodes zinc-2-glycoprotein (ZAG) is a lipid-mobilizing factor. However, its expression and distribution in the adult brain have been controversial. Here, for the first time, we demonstrate that the secreted ZAG is localized to Wisteria floribunda agglutinin (WFA)-positive PNNs around parvalbumin (PV)-expressing interneurons in the hippocampus, cortex, and a number of other PNN-bearing neurons and co-localizes with aggrecan, one of the components of PNNs. Few ZAG-positive nets were seen in the area without WFA staining by chondroitinase ABC (ChABC) which degrades glycosaminoglycans (GAGs) from the chondroitin sulfate proteoglycans (CSPGs) in the PNN. Reanalysis of single-cell sequencing data revealed that ZAG mRNA was mainly expressed in oligodendrocyte lineages, specifically in olfactory sheathing cells. The ZAG receptor β3 adrenergic receptor (β3AR) is also selectively co-localized with PV interneurons and CA2 pyramidal neurons in the hippocampus. In addition, molecular docking provides valuable new insights on how GAGs interfere with ZAG and ZAG/β3AR complex. Finally, our results indicated that human recombinant ZAG could significantly inhibit serum derivation-induced cell apoptosis in HT22 cells. Our combined experimental and theoretical approach raises a unique hypothesis namely that ZAG may be a crucial functional attribute of PNNs in the brain to protect neuronal cell from apoptosis. Gully erosion, an integrated result of various social and environmental factors, is a severe problem for sustainable development and ecology security in southern China. Currently, the dominant driving forces on gully distribution are shown to vary at different spatial scales. However, few systematic studies have been performed on spatial scaling effects in identifying driving forces for gully erosion. In this study, we quantitatively identified the determinants of gully distribution and their relative importance at four different spatial scales (southern China, Jiangxi province, Ganxian county, and Tiancun township, respectively) based on the Boruta algorithm. The optimal performance of gully susceptibility mapping was investigated by comparing the performance of the multinomial logistic regression (MLR), logistic model tree (LMT), and random forest (RF). Across the four spatial scales, the total contributions of gully determinants were classified as lithology and soil (32.65%) > topography (22.40%) > human activities (22.31%) > climate (11.32%) > vegetation (11.31%). Among these factors, precipitation (7.82%), land use and land cover (6.16%), rainfall erosivity (10.15%), and elevation (11.59%) were shown to be the predominant factors for gully erosion at the individual scale of southern China, province, county, and township, respectively. In addition, contrary to climatic factors, the relative importance of soil properties and vegetation increased with the decrease of spatial scale. Moreover, the RF model outperformed MLR and LMT at all the investigated spatial scales. This study provided a reference for factor selection in gully susceptibility modeling and facilitated the development of gully erosion management strategies suitable for different spatial scales. Bird sex chromosomes play a unique role in sex-determination, and affect the sexual morphology and behavior of bird species. Core waterbirds, a major clade of birds, share the common characteristics of being sexually monomorphic and having lower levels of inter-sexual conflict, yet their sex chromosome evolution remains poorly understood. Here, by we analyse of a chromosome-level assembly of a female crested ibis (Nipponia nippon), a typical core waterbird. We identify neo-sex chromosomes resulting from fusion of microchromosomes with ancient sex chromosomes. These fusion events likely occurred following the divergence of Threskiornithidae and Ardeidae. The neo-W chromosome of the crested ibis exhibits the characteristics of slow degradation, which is reflected in its retention of abundant gametologous genes. Neo-W chromosome genes display an apparent ovary-biased gene expression, which is largely driven by genes that are retained on the crested ibis W chromosome but lost in other bird species. These results provide new insights into the evolutionary history and expression patterns for the sex chromosomes of bird species. The evolutionary trajectory of avian sex chromosomes may be more intricate than previously understood. A remarkable post-transitional modification of both histones and non-histone proteins is arginine methylation. Methylation of arginine residues is crucial for a wide range of cellular process, including signal transduction, DNA repair, gene expression, mRNA splicing, and protein interaction. Arginine methylation is modulated by arginine methyltransferases and demethylases, like protein arginine methyltransferase (PRMTs) and Jumonji C (JmjC) domain containing (JMJD) proteins. Symmetric dimethylarginine and asymmetric dimethylarginine, metabolic products of the PRMTs and JMJD proteins, can be changed by abnormal expression of these proteins. Many pathologies including cancer, inflammation and immune responses have been closely linked to aberrant arginine methylation. Currently, the majority of the literature discusses the substrate specificity and function of arginine methylation in the pathogenesis and prognosis of cancers. Numerous investigations on the roles of arginine methylation in the central nervous system (CNS) have so far been conducted. In this review, we display the biochemistry of arginine methylation and provide an overview of the regulatory mechanism of arginine methyltransferases and demethylases. We also highlight physiological functions of arginine methylation in the CNS and the significance of arginine methylation in a variety of neurological diseases such as brain cancers, neurodegenerative diseases and neurodevelopmental disorders. Furthermore, we summarize PRMT inhibitors and molecular functions of arginine methylation. Finally, we pose important questions that require further research to comprehend the roles of arginine methylation in the CNS and discover more effective targets for the treatment of neurological diseases. Lack of sufficient cytotoxic T lymphocytes (CD8+ T cells) infiltration and dysfunctional state of CD8+ T cells are considered enormous obstacles to antitumor immunity. Herein, we construct a synergistic nanoplatform to promote CD8+ T cell infiltration in tumors while restoring T cell function by regulating methionine metabolism and activating the STING innate immune pathway. The CRISPR/Cas9 system down-regulates the methionine transporter SLC43A2 and restricts the methionine uptake by tumor cells, thereby relieving the methionine competition pressure of T cells; simultaneously, the released nutrition metal ions activate the cGAS/STING pathway. In this work, the described nanoplatform can enhance the effect of immunotherapy in preclinical cancer models in female mice, enhancing STING pathway mediated immunity and facilitating the development of amino acid metabolic intervention-based cancer therapy. Metabolic pressure, in particular the lack of methionine availability, on tumor-infiltrating CD8 + T cells is associated with T cell dysfunction. A classic problem in preservation is the microbes can grow in low-moisture foods. In this paper, the water sorption, and thermodynamic properties of glucose/WPI solid matrices were measured, while their molecular mobility was analyzed and associated with the microbial growth of D. Hansenii at various aw and 30 °C. Although the sorption isotherms, Tg, and relaxation processes of studied matrices were affected by aw and WPI, the microbial growth showed highly dependent on water mobility rather than aw. Hence, we introduced water usability (Uw), derived from the mobility difference between system-involved water and liquid pure water explicating from the classical thermodynamic viewpoint, to describe the dynamic changes of water mobility in glucose/WPI matrices. Despite to aw, the yeast growth rate was enhanced at high Uw matrices concomitantly with a rapid cell doubling time. Life science studies involving clustered regularly interspaced short palindromic repeat (CRISPR) editing generally apply the best-performing guide RNA (gRNA) for a gene of interest. Computational models are combined with massive experimental quantification on synthetic gRNA-target libraries to accurately predict gRNA activity and mutational patterns. However, the measurements are inconsistent between studies due to differences in the designs of the gRNA-target pair constructs, and there has not yet been an integrated investigation that concurrently focuses on multiple facets of gRNA capacity. In this study, we analyzed the DNA double-strand break (DSB)-induced repair outcomes and measured SpCas9/gRNA activities at both matched and mismatched locations using 926,476 gRNAs covering 19,111 protein-coding genes and 20,268 non-coding genes. We developed machine learning models to forecast the on-target cleavage efficiency (AIdit_ON), off-target cleavage specificity (AIdit_OFF), and mutational profiles (AIdit_DSB) of SpCas9/gRNA from a uniformly collected and processed dataset by deep sampling and massively quantifying gRNA capabilities in K562 cells. Each of these models exhibited superlative performance in predicting SpCas9/gRNA activities on independent datasets when benchmarked with previous models. A previous unknown parameter was also empirically determined regarding the "sweet spot" in the size of datasets used to establish an effective model to predict gRNA capabilities at a manageable experimental scale. In addition, we observed cell type-specific mutational profiles and were able to link nucleotidylexotransferase as the key factor driving these outcomes. Precise discrimination and eradication of cancer cells by immune cells independent of antigen recognition is promising for solid tumor therapeutics, yet remains a tremendous challenge. Inspired by neutrophils, here we design and construct a tumor discrimination nanodevice based on the differential histone H1 isoform expression. In this nanodevice, neutrophil membrane camouflage and glutathione (GSH)-unlocking effect on Fe-porphyrin metal−organic framework structure ensures selectivity to cancer cells. The released porcine pancreatic elastase (PPE) simulates neutrophils' action to induce histone H1 release-dependent selective cancer cell killing. Meanwhile, nuclear localization signal (NLS) peptide-tagged porphyrin (porphyrin-NLS) acts as in-situ singlet oxygen (1O2) generator to amplify histone H1 nucleo-cytoplasmic translocation by inducing DNA double-strand breaks (DSBs) under laser irradiation, further promoting elimination of cancer cells. The overexpressed histone H1 isoform in cancer cells improves selectivity of our nanodevice to cancer cells. In vivo studies demonstrate that our design can not only inhibit primary tumor growth, but also induce adaptive T-cell response-mediated abscopal effect to against distal tumors. Porcine neutrophil elastase (PPE) has been demonstrated to have a histone H1 isoform dependent anti-tumour effect. Surface characterization of metal nanoparticles is a critical need in nanocatalysis for in-depth understanding of the structure-function relationships. The surface structure of nanoparticles is often different from the subsurface, and it is challenging to separately characterize the surface and the subsurface. In this work, theoretical calculations and extended X-ray absorption fine structure (EXAFS) analysis illustrate that the surface atoms of noble metals (Pt and Pd) are oxidized in the air, while the subsurface atoms are not easily oxidized. Taking advantage of the oxidation properties, we suggest a stepwise reduction-oxidation approach to determine the surface atomic arrangement of noble metal nanoparticles, and confirm the rationality of this approach by identifying the surface structure of typical 2–3 nm Pt and Pd nanoparticles. The reduction-oxidation approach is applied to characterize the surface structure of model Pd-Sb bimetallic catalyst, which illustrates that the surface Pd is well isolated by Sb atoms with short bond distance at 2.70 Å, while there are still Pd-Pd bonds in the subsurface. Density functional theory (DFT) calculations and Pd L edge X-ray absorption near edge structure (XANES) indicate that the isolation of surface Pd significantly decreases the adsorption energies of Pd-hydrocarbon, which leads to the high propylene selectivity and turnover frequency Pd-Sb bimetallic catalyst for propane dehydrogenation. Natural killer (NK)/T-cell lymphoma-associated hemophagocytic lymphohistiocytosis (NK/T-LAHLH) is a rare and life-threatening disorder. Its clinical characteristics and appropriate treatment options are unclear. This study aimed to investigate the clinical characteristics and treatment options for this disease. We retrospectively analyzed the clinical data of 84 patients with NK/T-cell lymphoma and compared the characteristics, treatment, and survival between patients with and without HLH. Patients in the NK/T-LAHLH group were more likely to be younger age and have hepatosplenomegaly, B symptoms, neutropenia, anemia, thrombocytopenia, elevated lactate dehydrogenase levels, reduced serum albumin levels, bone marrow involvement, Ann Arbor stage III/IV, and International Prognostic Index score ≥ 3. After multivariate analysis, it was found that elevated lactate dehydrogenase and Ann Arbor stage III/IV were risk factors for HLH in patients with NK/T-cell lymphoma. After 2 weeks of therapy, 78.6% (11/14) patients who received the L-DEP/DEP regimen achieved an overall response rate of HLH, which was higher than that in 42.9% (9/21) patients who received the VP-16 + dexamethasone-based regimen. NK/T-LAHLH patients had poorer survival than non-HLH-NK/TCL patients. For NK/T-LAHLH, the L-DEP/DEP regimen may have a better response rate than the VP-16 + dexamethasone-based regimens. Arbutin, one of the main phenolic substances in pear fruits, has several health benefits. To optimize the extraction process of arbutin from pear fruitlets ('Huangli No. 1'), ultrasonic-assisted extraction (UAE) was investigated as an efficient method. Single-factor experiments combined with response surface methodology (RSM) were used to determine the optimum sonication parameters. The results of single factor experiments showed that the maximum extraction yield of arbutin was obtained with 60% ethanol, 1:10 solid-to-liquid ratio, 20 min ultrasound time, and 648 W ultrasound power. After optimization by RSM, the optimal conditions were 70% ethanol, 1:12 solid-to-liquid ratio, 20 min ultrasound time, and 576 W ultrasound power. Under these conditions, the extraction yield of arbutin was 5309.27 ± 15.87 µg/g by UAE, which was significantly higher than that by conventional extraction (CE; 1935.25 ± 44.13 µg/g). UAE generated more cell damage in pear fruitlets than CE. Additionally, arbutin from UAE had a higher antioxidant activity than arbutin from CE. DPPH free radical scavenging activity, FRAP reducing power, and ABTS free radical scavenging activity were 22.25%, 27.72%, and 17.35% higher in UAE than in CE, respectively. The substitution box (S-box) is an important nonlinear component in most symmetric cryptosystems and thus should have good properties. Its difference distribution table (DDT) and linear approximation table (LAT) affect the security of the cipher against differential and linear cryptanalysis. In most previous work, differential uniformity and linearity of an S-box are two primary cryptographic properties to impact the resistance against differential and linear attacks. In some cases, the branch number and fixed point are also be considered. However, other important cryptographic properties such as the frequency of differential uniformity (resp. linearity) and the number of Bad Input and Bad Output (BIBO) patterns in DDT (resp. LAT) are often ignored. These properties substantially affect lightweight cryptography based on substitution bit permutation networks (SbPN) such as PRESENT, GIFT and RECTANGLE. This paper introduces a new method to search for S-boxes satisfying all above criteria simultaneously. In our strategy, we transform the process of searching for S-boxes under certain constraints on cryptographic properties into a satisfiability (SAT) problem. As applications, we use our new approach to search out 4-bit and 5-bit S-boxes with the same or better cryptographic properties compared with the S-boxes from well-known ciphers. Finally, we also utilize our method to verify a conjecture proposed by Boura et al. in the case of all 3-bit and 4-bit S-boxes. Sewage sludge, a common by-product of wastewater treatment plants, is one important repository of antibiotic resistance genes (ARGs). The growing demands of sewage sludge reclamation, such as land application, increase the possibility of introducing ARGs into the environment and even the further dissemination of antibiotic resistance. Previous studies have paid much attention to the removal efficiencies of conventional pollutants such as heavy metals and pathogenic microorganisms during the sludge treatment processes. However, the effects on the abundance and diversity of ARGs got great concerns only recently. This paper mainly focuses on the enrichment and transmission modes of ARGs in sludge and the effects of representative sludge treatment technologies on ARG distributions in sludge. It seems that most physical and chemical techniques such as microwave, alkali treatment, and coagulation are ineffective in ARG reduction. The impacts of biological sludge treatment technologies on ARGs are varied, probably because of the diverse microbial community structures, operational parameters, and even environmental factors such as rainwater. Therefore, the sensitivities of potential hosts of specific ARG to the sludge treatments should determine the abundance of ARG before and after treatment. In addition, a reasonable combination of different sludge process techniques is usually a better choice than the single one for ARGs' removal due to its better ability to efficiently damage the embedded cells and directly degrade the released ARGs. Norovirus (NoV) is a major cause of sporadic cases and outbreaks of acute gastroenteritis (AGE), thereby imposing threat to health globally. It is unclear how quantitation of wastewater NoV reflects the incidence of human AGE infections; therefore, we conducted this systematic review and meta-analysis of published NoV wastewater surveillance studies. A literature search was performed, and all studies on NoV wastewater surveillance were identified. Quantitative results were evaluated. The results showed that the overall detection rate of NoV in wastewater was 82.10% (95% confidence interval [CI]: 74.22–89.92%); NoV concentration was statistically significant in terms of season (P < 0.001), with higher concentration in spring and winter. There were positive correlations between NoV GII concentration in wastewater and GII AGE cases (rs = 0.51, 95% CI: 0.18–0.74, I2 = 0%), total AGE cases (rs = 0.40, 95% CI: 0.15–0.61, I2 = 23%) and NoV outbreaks (rs = 0.47, 95% CI: 0.30–0.62, I2 = 0%). Results of cross-correlation analysis of partial data indicated that variations in GII concentration were consistent with or ahead of those in the number of AGE cases. The diversity of NoV genotypes in wastewater was elucidated, and the dominant strains in wastewater showed a consistent temporal distribution with those responsible for human AGE. Our study demonstrated the potential association of NoV detected in wastewater with AGE infections, and further studies are needed to confirm this conclusion. [ABSTRACT FROM AUTHOR]

The soluble dietary fiber from Asparagus officinalis (ASDF) was successively prepared using enzymolysis combined with spray-drying technology. High-performance liquid chromatography analysis showed that ASDF contained two polysaccharide fractions with the average molecular weight of 2.77 × 105 and 6.44 × 103 Da, and was composed of mannose, rhamnose, galacturonic acid, glucose, galactose, and arabinose with a molecular ratio of 19.93:1.02:1.94:32.17:1.00:1.91, respectively. ASDF showed potential in vitro antioxidant activities. The oral administration of ASDF significantly reduced the levels of total cholesterol, triglyceride, and low-density lipoprotein cholesterol in HD-induced mice serum. Furthermore, 16S rRNA gene sequencing analysis showed that ASDF significantly affected the composition of intestinal microbiota, especially reducing the Firmicutes/Bacteroidotetes ratio and the relative abundances of Desulfobacterota, Proteobacteria, Actinobacteriota and increasing that of Muribaculaceae, Bacteroides, and Alloprevotella. These results demonstrated that the intake of ASDF could regulate intestinal microbiota and serum lipid levels in hyperlipidemic conditions. [ABSTRACT FROM AUTHOR]

Nowadays, ciphers have been widely used in high-end platforms, resource-constrained, and side-channel attacks vulnerable environments. This motivates various S-boxes aimed at providing a good trade-off between security and efficiency. For small S-boxes, the most natural approach of constructing such S-boxes is a comprehensive search in the space of permutations, which inevitably becomes more challenging when the size grows. For large S-boxes (e.g., 8-bit), previous works concentrated on creations from finite fields or smaller ones (e.g., 4-bit). This paper proposes a new algorithm with a layered structure to search for 8-bit SKINNY-like S-boxes. We compare our new S-box with the original 8-bit SKINNY S-box by analyzing its security properties. Besides, due to our searching algorithm's rules and constraints, SKINNY-like S-boxes have other features of lightweight implementation, low multiplicative complexity, low AND depth, and an effective inverse. Eventually, the searching algorithm outputs 224000 8-bit SKINNY-like S-boxes. It is more and more important to consider the dependence structure among multiple testings, especially for the genome-wide association studies (GWAS). The existing procedures, such as local index of significance (LIS) and pooled local index of significance (PLIS), were proposed to test hidden Markov model (HMM)-dependent hypotheses under the framework of compound decision theory, which was successfully applied to GWAS. However, the etiology of complex diseases is not only with respect to the genetic effects, but also the environmental factors. Failure to account for the covariates in multiple testing can produce misleading bias of the association of interest, or suffer from loss of testing efficiency. In this paper, we develop a covariate-adjusted multiple testing procedure, called covariate-adjusted local index of significance (CALIS), to account for the effects of environmental factors via a factorial hidden Markov model. The theoretical results show that our procedure can control the false discovery rate (FDR) at the nominal level and has the smallest false non-discovery rate (FNR) among all valid FDR procedures. Conductive and degradable nanofibrous scaffolds have great potential in promoting cell growth, proliferation, and differentiation under an external electric field. Although the issue of inferior electrical conductivity in body fluids still exists, polyaniline (PANI)-based degradable nanofibers can promote cell adhesion, growth, and proliferation. To investigate whether the effect is caused by the PANI morphology, we selected three inorganic acids as dopants in the process of PANI in situ oxidative polymerization: hydrochloric acid, sulfuric acid, and perchloric acid. The obtained polyaniline/polylactic acid (PANI/PLA) composite nanofibers were characterized via SEM, FTIR, and XPS analysis, and we confirmed that the PLA nanofibers were successfully coated by PANI without any change to the porous structure of the PLA nanofibers. The in vitro mechanical properties and degradability indicated that the oxidation of acid dopants should be considered and that it was likely to have a higher oxidation degradation effect on PLA nanofibers. The contact angle test demonstrated that PANI/PLA composite nanofibers with different surface morphologies have good wettability, implying that they meet the requirements of bone tissue engineering scaffolds. The surface roughness and cell viability demonstrated that different PANI morphologies on the surface can promote cell proliferation. The higher the surface roughness of the PANI, the better the biocompatibility. Consequently, the regulated surface morphology of PANI/PLA composite nanofibers via different acids doping has positive effect on biocompatibility in tissue engineering. [ABSTRACT FROM AUTHOR]

Pear (Pyrus spp.) belongs to the genus Pyrus, in the family Rosaceae. Some varieties of pear fruit exhibit bulged surface, which seriously affects the quality and commodity value of the pear fruit. In this study, we performed anatomical, physiological, and transcriptomic analysis to explore the mechanism of paclobutrazol (PBZ) on the bulged surface of pear fruit. The vascular bundles of flesh were more evenly distributed, and the fruit cells were more compactly arranged and smaller in size treated with PBZ. However, the auxin (IAA) content of flesh was decreased in the treated group. Furthermore, the GO and KEGG analysis of differentially expressed genes (DEGs) showed that auxin, phenylpropanoid metabolic pathways, and transcriptional factor genes were significantly enriched on the relieved bulged surface of pear fruit. And it was analyzed that some genes contained auxin responded cis-elements from the selected DEGs in the promoter region. We conclude that PBZ plays a negative role in cell division, cell elongation, and vascular bundle development on the bulged surface of pear fruit through the involvement of auxin-related genes. OVATE family proteins (OFPs) are the plant-specific transcription factors, and have significant functions in regulating plant growth, development and resistance. The OFP genes have been investigated in several plants, but they still lack a systematic analysis of OFP genes in Chinese pear and some other five Rosaceae genomes. Here, 28 PbrOFPs were identified within Chinese pear and compared them with those of other five Rosaceae genomes. Evolutionary tree revealed that all OFP genes from six Rosaceae genomes were divided into eight groups. Seventeen conserved microsynteny regions were detected in Chinese pear genome, suggested that these PbrOFP genes might be considered to have originated from the large-scale duplication events., indicating these PbrOFP genes might contain specialized regulatory mechanisms in these tissues, such as flower, ovary and fruit. Remarkably, two PbrOFP genes (Pbr010426.1 and Pbr010427.1) were up-regulated under Venturia nashicola treatment, and five PbrOFP genes were up-regulated under PEG treatment, suggesting that these genes might play crucial roles in defence to environmental stresses. Our data presented a systematic analysis and might aid in the selection of appropriate PbrOFPs for further functional studies in Chinese pear, especially in response to the mechanism of biotic and abiotic stresses. Key message: Genomic editing with CRISPR/Cas9 system can simultaneously modify multiple copies of theBnaFAD2 gene to develop novel variations in fatty acids profiles in polyploidy rapeseed. Fatty acid composition affects edible and processing quality of vegetable oil and has been one of the primary targets for genetic modification in oilseed crops including rapeseed (Brassica napus). Fatty acid desaturase 2 gene, FAD2, is a key player that affects three major fatty acids, namely oleic, linoleic and linolenic acid, in oilseed plants. Previously, we showed that there are four copies of BnaFAD2 in allotetraploid rapeseed. In this study, we further established spatiotemporal expression pattern of each copy of BnaFAD2 using published RNA-seq data. Genomic editing technology based on CRISPR/Cas9 system was used to mutate all the copies of BnaFAD2 to create novel allelic variations in oleic acid and other fatty acid levels. A number of mutants at two targeting sites were identified, and the phenotypic variation in the mutants was systematically evaluated. The oleic acid content in the seed of the mutants increased significantly with the highest exceeding 80% compared with wild type of 66.43%, while linoleic and linolenic acid contents decreased accordingly. Mutations on BnaFAD2.A5 caused more dramatic changes of fatty acid profile than the mutations on BnaFAD2.C5 alleles that were identified with gene editing technique for the first time. Moreover, combining different mutated alleles of BnaFAD2 can even broaden the variation more dramatically. It was found that effects of different mutation types at BnaFAD2 alleles on oleic levels varied, indicating a possibility to manipulate fatty acid levels by precise mutation at specific region of a gene. SpoIIID is a small, sequence-specific DNA-binding protein which can direct many genes' transcription and has an effect on spore formation in Bacillus subtilis. We investigated the role of SpoIIID in mother cell lysis in Bacillus thuringiensis. A β-galactosidase assay based on the promoter fusions with lacZ indicated that the sigK gene was positively regulated by SpoIIID and σK negatively regulated the expression of sigE. The spoIIID mutant strain exhibited no mother cell lysis in Schaeffer's sporulation medium (SSM) but did in ½ Luria-Bertani (LB) medium. cwlC is an essential hydrolase gene for mother cell lysis. Moreover, the expression of a PcwlC-lacZ fusion in spoIIID mutant was proved to be higher in ½ LB medium than in SSM. HD (ΔspoIIID)(ΔcwlC) mutant was obtained by knocking out the cwlC gene in HD(ΔspoIIID) and displayed no mother cell lysis in both SSM and ½ LB mediums. The deletion of spoIIID decreased the crystal protein production in HD73. The expression of Porf1cry8E and P5014 promoter fusions with lacZ gene in the acrystalliferous HD−(ΔspoIIID) mutant showed similar activity to that in the acrystalliferous HD73− strain before T7 and slightly higher than that in the acrystalliferous HD73− after T7. Sodium dodecyl sulfate polyacrylamide gel electrophoresis showed that Cry1Ac production in HD−(ΔspoIIID) directed by the Porf1cry8E and P5014 promoters was at a similar level as that in HD73 wild strain. Altogether, these results suggested that the spoIIID mutant with Porf1cry8E or P5014 promoters could be an alternative delivery system for cry gene expression with no mature spore formation and medium-dependent mother cell lysis. Integral attack is one of the most powerful tools in the field of symmetric ciphers. In order to reduce the time complexity of original integral one, Wang et al. firstly proposed a statistical integral distinguisher at FSE’16. However, they don’t consider the cases that there are several integral properties on output and multiple structures of data should be used at the same time. In terms of such cases, we put forward a new statistical integral distinguisher, which enables us to reduce the data complexity comparing to the traditional integral ones under multiple structures. As illustrations, we use it into the known-key distinguishers on AES-like ciphers including AES and the permutations of Whirlpool, PHOTON and Grøstl-256 hash functions based on the Gilbert’s work at ASIACRYPT’14. These new distinguishers are the best ones comparing with previous ones under known-key setting. Moreover, we propose a secret-key distinguisher on 5-round AES under chosen-ciphertext mode. Its data, time and memory complexities are 2114.32 chosen ciphertexts, 2110 encryptions and 233.32 blocks. We present a facile and versatile method for introducing various non-precious metal nanoparticles (NPs) in small nanotubes, such as single-walled carbon nanotubes (SWNTs), including 3d-metals (V, Mn, Fe and Co), 4d-metals (Mo), and 5d-metals (W). This is realized by oxidizing encapsulated cycloalkene metal carbonyl complexes below their sublimation temperatures. This novel technique is significant because it avoids the diffusion and deposition of metal species on the outer walls of nanotubes, which has been challenging to achieve using the conventional filling methods. High-resolution transmission electron microscopy (HRTEM), high angle annular dark field scanning transmission electron microscopy (HAADF-STEM), energy-dispersive X-ray spectroscopy (EDX), Raman, and X-ray photoelectron spectroscopy (XPS) analyses revealed high filling efficiencies (> 95% SWNTs filled with metal NPs). This method also provides a unique approach to fabricate highly dispersed and uniform SWNT-metal nanoparticle encapsulates with lower valence states, which are often not stable in the bulk. Integral attack, as a powerful technique in the cryptanalysis field, has been widely utilized to evaluate the security of block ciphers. Integral distinguisher is based on balanced property on output with probability one. To obtain a distinguisher covering more rounds, an attacker will usually increase the data complexity by iterating through all values of more bits of plaintexts under the firm limitation that the data complexity should be less than the whole plaintext space. In order to release the limitation and reduce the data complexity, Wang et al. proposed a statistical integral distinguisher at FSE'16. In this paper, we exploit the statistical integral distinguisher to attack the IDEA and CAST-256 block ciphers. As a result, we manage to mount a key recovery attack on 29-round CAST-256 with 2 chosen plaintexts, 2 encryptions and 2 bytes of memory. By making a trade-off between the time complexity and data complexity, the attack can be achieved by 2 chosen plaintexts, 2 encryptions and 2 bytes of memory. As far as we know, these are the best attacks on CAST-256 in the single-key model without weak-key assumption so far. What's more, we find an integral distinguisher of IDEA block cipher, which is the longest integral distinguisher known to now. By taking advantage of this distinguisher, we achieve a key recovery attack on 4.5-round IDEA with 2 known plaintexts, 2 encryptions and 2 bytes of memory respectively. Purpose: Returning straw to soil improved soil carbon sequestration capacity and increase soil organic matter. However, in different soil depth, especially in subsoil, there were few studies on the effects of straw decomposition on soil carbon sequestration and the properties of humic substances. Therefore, an in-situ incubation study, with six different straw rates and three different soil depths, was carried out to explore the effects of straw decomposition on soil organic carbon and humic substance composition at different soil depths. Materials and methods: The experiment was composed of six straw rates: 0, 0.44, 0.88, 1.32, 2.64, and 5.28% of soil dry mass. The maize straw was proportionately mixed with soil and put into nylon bags. Then, the nylon bags were buried in soil at three depths (15, 30, and 45 cm) and the straw decomposition trial lasted for 17 consecutive months in-situ. Soil samples were collected after completion of the field trial. Humic substances were quantitatively and qualitatively analyzed using the modification method of humus composition and the methods specified by the International Humus Association. Fourier transform infrared spectroscopy and fluorescence spectroscopy were used in this study. Results and discussion: Results indicated that CO concentration increased with increase in soil depth. Compared with the 'zero' straw control, soil organic carbon contents in the treatments amended with 1.32, 2.64, and 5.28% maize straw increased significantly, and most accumulations were at 30-45 cm depths. FTIR and fluorescence emission spectra analyses indicated that the addition of straw enhanced the aliphatic structure and decreased the aromaticity of humic acid (HA), that was to say that HA molecular structure approaches to the development of simplification and younger. The maximum change in HA molecular structure was under the 5.28% treatment in the 30-45 cm depth. Conclusions: Returning maize straw to the subsoil layers is more conducive to the accumulation of soil organic carbon and improvement of the quality and activity of HA and the organic carbon in the subsoil can be renewed. The multidimensional linear cryptanalysis and the multidimensional zero-correlation linear cryptanalysis have been widely used in the attacks on block ciphers. In the multidimensional linear cryptanalysis with $$\chi ^2$$ -method and the multidimensional zero-correlation linear cryptanalysis, the statistics used to distinguish the right key and wrong keys are calculated from the probability distribution of multidimensional (zero-correlation) linear approximations. In this paper, we show that the statistics can be computed directly from the empirical correlations of multidimensional (zero-correlation) linear approximations for random plaintext set. In this way, the computation cost of the probability distribution can be removed. In the situation where FFT technique can be applied to calculate the correlations, our proposed computing method for the statistics can decrease the time complexity of multidimensional (zero-correlation) linear cryptanalysis. As an illustration, the Feistel network with bijective round functions consisting of the modular additions or XORs with subkeys and CAST-256 have been attacked with our revised multidimensional zero-correlation linear cryptanalysis. Our attacks on such kind of Feistel networks are the best according to the number of rounds and we improved the previous multidimensional zero-correlation attack on CAST-256 from 28 to 29 rounds. Compared with the best attack on 29-round CAST-256 with multiple zero-correlation linear cryptanalysis method, our attack leads to the same complexity but without any assumption of independence. Improving the nitrogen use efficiency (NUE) may considerably increase maize yield and decrease the use of nitrogen (N) fertilizer. But the genetic basis of NUE in maize is still poorly understood. In this study, an association panel of 139 maize inbred lines genotyped with 50,790 single nucleotide polymorphism (SNP) was used to dissect the genetic basis of NUE-related traits by genome-wide association study (GWAS). NUE, N uptake efficiency (NupE), N utilization efficiency (NutE), grain N concentration (GNC), stover N concentration (SNC) and N harvest index (NHI) were estimated under two N levels. GWAS was performed using a fixed and random model circulating probability unification (FarmCUP) method. In total, 27 and 23 significant SNP-traits association signals were identified under normal and low N levels. In addition, 10 significant association signals were detected based on the traits relative value (normal N supply / low N supply) of two N levels. Further, 60 candidate genes were predicted for these traits base on linkage disequilibrium (LD) and low nitrogen transcriptome analysis of significant SNP regions. Among the candidate genes identified in this study, 66.7% involved in nitrogen compound metabolic process. Zm00001d025831 and Zm00001d004633 encoded ammonium transporter1 and transmembrane amino acid transporter family protein, respectively, may be important candidate genes for NUE. The markers identified in this study maybe has important significance and could be useful in molecular marker assisted selection in breeding of high-NUE maize varieties, and the candidate genes could deepen the understanding of the genetic basis of NUE. Kernel size and weight are important agronomic traits, as well as crucial traits that influence grain yield in maize. In the present study, 150 F7 recombinant inbred lines derived from a cross 178×K12 were evaluated for kernel length (KL), kernel width (KW), kernel thickness (KT), and 100-kernel weight (HKW) across seven environments. Natural variations in KL, KW, KT, and HKW were observed in the population. A set of quantitative trait loci (QTLs) for the kernel-related traits were identified by inclusive composite interval mapping method. For the four kernel traits from seven environments and the best linear unbiased prediction data, a total of 52 QTLs were detected, which distributed on all chromosomes except chromosome 6. The LOD values ranged from 2.52 to 8.91, the additive effect from − 2.22 to 1.37, and the range of individually explaining phenotypic variation was from 5.8 to 23.49%. Amongst these QTLs, most were detected only in one or two environments. Three stable QTLs, qKL4-1 at bin 4.07/4.08, qKW4-2 at bin 4.06 and qKT2-1 at bin 2.02/2.03, were identified across at least three environments. Besides, several overlapping QTLs associated with multiple traits were identified. For example, qKW3-1 for KW and qHKW3-1 for HKW were located in the same marker interval at Bin 3.01/3.02. These stable QTLs and overlapping QTLs found in this study will contribute to the understanding of genetic components of grain yield and provide the foundation for molecular marker-assisted breeding in maize. In vitiligo, melanocytes are particularly vulnerable to oxidative stress owing to the pro-oxidant state generated during melanin synthesis and to the genetic antioxidant defects. Autophagy is a controlled self-digestion process which can protect cells against oxidative damage. However, the exact role of autophagy in vitiligo melanocytes in response to oxidative stress and the mechanism involved are still not clear. To determine the implications of autophagy for melanocyte survival in response to oxidative stress, we first detected the autophagic flux in normal melanocytes exposure to H2O2, and found that autophagy was significantly enhanced in normal melanocytes, for protecting cells against H2O2-induced oxidative damage. Nevertheless, vitiligo melanocytes exhibited dysregulated autophagy and hypersensitivity to H2O2-induced oxidative injury. In addition, we confirmed that the impairment of Nrf2-p62 pathway is responsible for the defects of autophagy in vitiligo melanocytes. Noteworthily, upregulation of the Nrf2-p62 pathway or p62 reduced H2O2-induced oxidative damage of vitiligo melanocytes. Therefore, our data demonstrated that dysregulated autophagy owing to the impairment of Nrf2-p62 pathway increase the sensitivity of vitiligo melanocytes to oxidative stress, thus promote the development of vitiligo.
