The Sumida river estuary of Tokyo bay is often affected by fecal contamination from combined sewer overflows (CSOs). This study monitored the surface water quality from the upstream of the Sumida river to the estuary in October 2017, June 2018, and July 2018 after three long-duration rainfall events. Several types of sewage markers, including fecal bacteria and two types of bacteriophages as microbial markers, and five pharmaceuticals and personal care products (PPCPs) as chemical markers were used to evaluate fecal contamination. CSO discharge was estimated separately from pumping stations and overflow chambers. The dominant contribution from overflow chambers was estimated to be as high as 86 – 91% of total discharge volume indicating their significance in controlling CSO pollution. High concentrations of sewage marker were observed in a wide area due to CSO discharge of more than 30 h in all 3 events. Escherichia coli was found to be as high as 4.00 – 4.57 log10 (CFU/100 mL). Meanwhile, caffeine showed the highest concentration of 2105 ng/L among PPCPs. It was found to be a useful indicator of recent contamination that captured a unique spatial distribution tendency. On the other hand, crotamiton, a conservative PPCP, was found to be highly diluted and might not be appropriate for tracking pollutants under heavy rainfall events. The effect of CSO discharge pattern and tidal change on the distribution of sewage markers, including dispersion degree and pollutants travel time, was described. The presence of pathogenic viruses in drinking water is a major public health concern. Although viability RT-qPCR methods were developed to quantify infectious viruses, they may not always reflect viral infectivity, therefore leading to false-positive results. In this study, sodium deoxycholate (SD) pre-treatment was used to improve the efficiency of viability RT-qPCR methods with respect to exclusive quantification of infectious viruses. The ability of SD pre-treatment to enhance the penetration of three viability markers, namely, ethidium monoazide (EMA, 100 µM), propidium monoazide (PMA, 100 µM), and cis-dichlorodiammineplatinum (CDDP, 1000 µM), into heat-treated (90 °C for 1 min) Aichi virus at various concentrations (0.01-0.5%) was evaluated. The optimal SD concentration was found to be 0.1% for all markers. EMA/PMA/CDDP-RT-qPCR with 0.1% SD pre-treatment was significantly more effective than without SD pre-treatment in determining AiV inactivation after heat (50, 60, 70, 80, or 90 °C for 1 min) or chlorine treatment (1 mgCl2/L for 1, 2, 5, or 10 min). Among the viability RT-qPCR methods tested, CDDP-RT-qPCR with SD pre-treatment (SD-CDDP-RT-qPCR) was the most effective in reflecting viral infectivity. Performance testing of SD-CDDP-RT-qPCR in concentrated drinking water samples did not reveal any significant effects of SD-CDDP treatment. Ammonia is a precursor to trichloramine, which causes an undesirable chlorinous odor. Granular activated carbon (GAC) filtration is used to biologically oxidize ammonia during drinking water purification; however, little information is available regarding the abundance and diversity of ammonia-oxidizing archaea (AOA) and bacteria (AOB) associated with GAC. In addition, their sources and fates in water purification process remain unknown. In this study, six GAC samples were collected from five full-scale drinking water purification plants in Tokyo during summer and winter, and the abundance and community structure of AOA and AOB associated with GAC were studied in these two seasons. In summer, archaeal and bacterial amoA genes on GACs were present at 3.7 × 10-3.9 × 10 gene copies/g-dry and 4.5 × 10-4.2 × 10 gene copies/g-dry, respectively. In winter, archaeal amoA genes remained at the same level, while bacterial amoA genes decreased significantly for all GACs. No differences were observed in the community diversity of AOA and AOB from summer to winter. Phylogenetic analysis revealed high AOA diversity in group I.1a and group I.1b in raw water. Terminal-restriction fragment length polymorphism analysis of processed water samples revealed that AOA diversity decreased dramatically to only two OTUs in group I.1a after ozonation, which were identical to those detected on GAC. It suggests that ozonation plays an important role in determining AOA diversity on GAC. Further study on the cell-specific activity of AOA and AOB is necessary to understand their contributions to in situ nitrification performance. [ABSTRACT FROM AUTHOR]

This study investigated and characterised organic matter present in sediment particles deposited in infiltration facilities using an excitation-emission matrix method combined with parallel factor analysis (EEM-PARAFAC). The organic fluorophore identified was correlated with sediment bound metals. The PARAFAC analysis identified three major components. The fluorophore in each of the three components appeared in different locations with different spectral shapes. The maximum fluorescence intensity ( F ) observed for each fluorescent component was correlated with seven heavy metals (Cr, Mn, Co, Ni, Cu, Zn and Pb). F of component 1 displayed a negative relationship with all the metals (correlation coefficient = −0.28 to −0.72), and F of component 3 showed a positive relationship (0.20 to 0.62), and among them, Cu, Ni and Zn had higher correlation. Our results demonstrate that a PARAFAC approach can help to further elucidate organic matter species, thereby allowing a better understanding of the mobility of elemental species in the deposited sediment. Scenario analysis of rainwater harvesting and utilization (RWHU) was performed considering various non-potable water uses in different building types over a year. Six building types were identified in the study area using GIS data: residential houses, offices, commercial buildings, restaurants, public buildings, and 'others'. Rainwater storage capacity was considered as 30 mm rainwater depth. Water demand for each building type was calculated as the sum of the individual water uses for toilet flushing, air conditioning, garden irrigation, and cleaning defined in this study as 'miscellaneous usages'. To incorporate water quality considerations, rainwater with suspended solids level of less than 2 mg l was used as the quality criterion. The RWHU scenario was compared with other storage and water use scenarios. This study quantified the rainwater availability throughout a year and its seasonal variation and consumption in each building type. We constructed a 60-mer oligonucleotide microarray on the basis of benzene monooxygenase gene diversity to develop a new technology for simultaneous detection of the functional gene diversity in environmental samples. The diversity of the monooxygenase genes associated with benzene degradation was characterized. A new polymerase chain reaction (PCR) primer set was designed using conserved regions of benzene monooxygenase gene (BO12 primer) and used for PCR-clone library analysis along with a previously designed RDEG primer which targeted the different types of benzene monooxygenase gene. We obtained 20 types of amino acid sequences with the BO12 primer and 40 with the RDEG primer. Phylogenetic analysis of the sequences obtained suggested the large diversity of the benzene monooxygenase genes. A total of 87 60-mer probes specific for each operational taxonomical unit were designed and spotted on a microarray. When genomic DNAs of single strains were used in microarray hybridization assays, corresponding sequences were successfully detected by the microarray without any false-negative signals. Hybridization with soil DNA samples showed that the microarray was able to detect sequences that were not detected in clone libraries. It is important to evaluate the removal of enteric viruses by drinking water treatment processes so that viral infection risk can be assessed and managed. However, evaluating the removal of enteric viruses by full-scale treatment processes can be challenging due to the low numbers of viruses and the presence of substances in the water samples that inhibit detection. In this study, we evaluated the removal of pepper mild mottle virus (PMMoV) by microfiltration (MF) and slow sand filtration (SSF) at two full-scale drinking water treatment plants in Japan, quantifying virus concentrations with real-time polymerase chain reaction (qPCR). The removal of PMMoV by MF ranged from 0.0 to >0.9 log10, although concentrations were below the detection limit for half of the treated water samples. SSF removed PMMoV by up to 2.8 log10; however, the removal efficiency decreased to 0.0–1.0 log10 under cold water temperatures. Process control showed that nucleic acid extraction and qPCR efficiency were inhibited in nearly 40% of water samples. Dilution, DAX-8, and ferrihydrite treatments for purification were effective in mitigating these inhibitory effects. [ABSTRACT FROM AUTHOR]

This study is aimed to investigate biological oxidation of arsenic [As(III)] in sand component of arsenic–iron removal plant (AIRP). Water and sand samples were collected from the municipal and household AIRP units (MAIRP and HAIRPs) at Manikganj district, Bangladesh. These samples were analyzed using arsenic speciation cartridge with inductively coupled plasma-mass spectrometry for arsenic and iron detection and phylogenetic analysis based on aoxB gene sequencing. It was observed that arsenic concentration in the raw water, before and after sand filtration, ranged from 13.0–81.0 to 4.0–21.0 μg/L, respectively, which majorly contained reduced arsenic [As(III)] in the inlet and oxidized arsenic [As(V)] in the outlet. The results of our batch experiments showed that indeed sand unit of MAIRP oxidized As(III) with 36 g/h estimated biological oxidation potential which was considered enough to oxidize all the As(III) loading (6.3 g/h) in the influent. Additionally, bacterial arsenite oxidase gene (aoxB) was detected in the sand sample and has 80% sequence similarity with Polymorphum gilvum, an alphaproteobacteria on the phylogenetic tree. This study, therefore, revealed that AIRP sand units have enough potential of biological activity to ensure overall arsenic removal through As(III) oxidation.
