Additive manufacturing (AM) of biomedical materials provides enormous opportunities to fabricate multifunctional and structurally designed frameworks for tissue engineering, such as dental implants and bone substitutes. Despite several advantages of the binder jet 3D printing technology over other AM methods, for example, no limitations in materials selection, high materials recycling efficiency, no thermal stress development, no need to support materials, and the possibility of fabrication of printing functionally graded materials, the fabrication of biomedical-grade titanium alloys with high-density, fine microstructure, and low pickup of impurities is still challenging. This work presents the effects of powder particle size and 3D printing conditions on the microstructural features and mechanical properties of Ti-6Al-4V alloy. The formation of large and inter-aggregate pores during binder jetting is demonstrated and discussed. Design and selection of particle size distribution with a mean diameter of ~20 μm and large span and positive skewness are proposed to minimize binder-induced powder aggregation and fabricate green parts with a density of 65±1% PFD (pore-free density). Dilatometric studies under a partial pressure of argon (0.1 bar) determine that sintering just above the α/β transus temperature (~980 °C) provides a high strain rate to remove pores, but high-temperature sintering (≥1250 °C) is required to attain 97% PFD. The successful fabrication of high-density Ti-6Al-4V parts (≥96% PFD) with the microstructure comparable to metal injection molding (MIM) titanium parts (≈100 μm α grains + β lattes) is demonstrated. The tensile strength and elongation values fall in the range of 880±50 MPa and 6±2% (depending on the processing condition), which is comparable with metal injection molded parts and superior to the laser powder bed fusion technology concerning ductility. The content of carbon (<0.02 wt.%) and nitrogen (0.01 wt.%) also falls in the standard region of metal injection molding. However, oxygen pickup during sintering moderately increases the oxygen content (for 30–50%) over the standard level. The concentration of interstitials entrapped in the metal is comparable to that of parts manufactured by the powder bed fusion process, but the mechanical properties are better matched with the commercial titanium alloy. The fabrication of the titanium alloy as per the ASTM F2885 standard provides an excellent opportunity for the binder jetting process to develop custom-made biomaterials. [ABSTRACT FROM AUTHOR]

Key message: Structural variants (SV) of 23 barley inbreds, detected by the best combination of SV callers based on short-read sequencing, were associated with genome-wide and gene-specific gene expression and, thus, were evaluated to predict agronomic traits. In human genetics, several studies have shown that phenotypic variation is more likely to be caused by structural variants (SV) than by single nucleotide variants. However, accurate while cost-efficient discovery of SV in complex genomes remains challenging. The objectives of our study were to (i) facilitate SV discovery studies by benchmarking SV callers and their combinations with respect to their sensitivity and precision to detect SV in the barley genome, (ii) characterize the occurrence and distribution of SV clusters in the genomes of 23 barley inbreds that are the parents of a unique resource for mapping quantitative traits, the double round robin population, (iii) quantify the association of SV clusters with transcript abundance, and (iv) evaluate the use of SV clusters for the prediction of phenotypic traits. In our computer simulations based on a sequencing coverage of 25x, a sensitivity > 70% and precision > 95% was observed for all combinations of SV types and SV length categories if the best combination of SV callers was used. We observed a significant (P < 0.05) association of gene-associated SV clusters with global gene-specific gene expression. Furthermore, about 9% of all SV clusters that were within 5 kb of a gene were significantly (P < 0.05) associated with the gene expression of the corresponding gene. The prediction ability of SV clusters was higher compared to that of single-nucleotide polymorphisms from an array across the seven studied phenotypic traits. These findings suggest the usefulness of exploiting SV information when fine mapping and cloning the causal genes underlying quantitative traits as well as the high potential of using SV clusters for the prediction of phenotypes in diverse germplasm sets. O-GlcNAc modification plays important roles in metabolic regulation of cellular status. Two homologs of O-GlcNAc transferase, SECRET AGENT (SEC) and SPINDLY (SPY), which have O-GlcNAc and O-fucosyl transferase activities, respectively, are essential in Arabidopsis but have largely unknown cellular targets. Here we show that AtACINUS is O-GlcNAcylated and O-fucosylated and mediates regulation of transcription, alternative splicing (AS), and developmental transitions. Knocking-out both AtACINUS and its distant paralog AtPININ causes severe growth defects including dwarfism, delayed seed germination and flowering, and abscisic acid (ABA) hypersensitivity. Transcriptomic and protein-DNA/RNA interaction analyses demonstrate that AtACINUS represses transcription of the flowering repressor FLC and mediates AS of ABH1 and HAB1, two negative regulators of ABA signaling. Proteomic analyses show AtACINUS's O-GlcNAcylation, O-fucosylation, and association with splicing factors, chromatin remodelers, and transcriptional regulators. Some AtACINUS/AtPININ-dependent AS events are altered in the sec and spy mutants, demonstrating a function of O-glycosylation in regulating alternative RNA splicing. AtACINUS is an Arabidopsis homolog of a mammalian splicing regulator and previously found to be O-GlcNAcyated. A central problem in speciation is the origin and mechanisms of reproductive barriers that block gene flow between sympatric populations. Wind-pollinated plant species that flower in synchrony with one another rely on post-pollination interactions to maintain reproductive isolation. In some locations in Mexico, sympatric populations of domesticated maize and annual teosinte grow in intimate associate and flower synchronously, but rarely produce hybrids. This trait is typically conferred by a single haplotype, Teosinte crossing barrier1-s. Here, we show that the Teosinte crossing barrier1-s haplotype contains a pistil-expressed, potential speciation gene, encoding a pectin methylesterase homolog. The modification of the pollen tube cell wall by the pistil, then, is likely a key mechanism for pollen rejection in Zea and may represent a general mechanism for reproductive isolation in grasses. Purpose: Rapamycin, an immunosuppressive in transplant surgery, has an additional antiproliferative effect. The aim of this study was to investigate the potential protective effects of rapamycin on postoperative adhesion development. Methods: Ten rats per group underwent midline incision laparotomy and adhesion induction including bowel sutures. Therapy groups received daily intraperitoneal rapamycin injections (1.5 mg/kg body weight) for 3 weeks postoperatively. Controls were rats without any postoperative treatment, rats receiving the rapamycin solvent or a hyaluronic acid-carboxymethylcellulose membrane (Seprafilm). Results: Postoperative rapamycin application led to enhanced adhesion development and there was a higher rate of wound infections. In addition, Seprafilm did not reduce adhesions, in subgroups there were even more. Conclusions: Rapamycin is not recommendable for perioperative immunosuppression, it enhances adhesion development and leads to a higher rate of wound infections.
