In the past, diatoms were considered the primary food source for copepods in both wild and aquaculture settings. However, recent studies have found that diatoms have defense mechanisms against predators, making them unsuitable as copepod feed. This study assessed the impact of addition of 100 μg L−1 of silicate to an inorganic nutrition formula containing 700 μg L−1 nitrogen, 100 μg L−1 phosphorus, and 100 μg L−1 iron. Our objective was to assess the impact of increased diatom abundance on copepod production. The experiment was carried out in 1000 L outdoor tanks over a period of 20 days, with adult Pseudodiaptomus annandalei copepods, introduced into each tank on the second day at an initial density of 10 ind. L−1. The results indicated that adding silicate reduced the prevalence of Chlorophyta, replaced by a higher proportion of Dinophyta, and eventually dominated by diatoms. While silicate had a positive effect on diatom culture, it had a negative effect on copepod production. We report an innovative and facile approach to fabricating an ultrasensitive plasmonic paper substrate for surface-enhanced Raman spectroscopy (SERS). The approach exploits the self-assembling capability of poly(styrene-b-2-vinyl pyridine) block copolymers to form a thin film at the air-liquid interface within the single microdroplet scale for the first time and the subsequent in situ growth of silver nanoparticles (AgNPs). The concentration of the block copolymer was found to play an essential role in stabilizing the droplets during the mass transfer phase and formation of silver nanoparticles, thus influencing the SERS signals. SEM analysis of the morphology of the plasmonic paper substrates revealed the formation of spherical AgNPs evenly distributed across the surface of the formed copolymer film with a size distribution of 47.5 nm. The resultant enhancement factor was calculated to be 1.2 × 107, and the detection limit of rhodamine 6G was as low as 48.9 pM. The nanohybridized plasmonic paper was successfully applied to detect two emerging pollutants—sildenafil and flibanserin—with LODs as low as 1.48 nM and 3.45 nM, respectively. Thus, this study offers new prospects for designing an affordable and readily available, yet highly sensitive, paper-based SERS substrate with the potential for development as a lab-on-a-chip device. Copper is an essential trace element for the human body. The epidemiological evidence for the association of dietary intake of copper with the risk of Parkinson's disease (PD) is limited. We conducted an evaluation of the cross-sectional data gathered from the National Health and Nutrition Examination Surveys spanning from 2007 to 2018, which comprised a total of 17,948 participants. To discern the distinct characteristics of the participants, we performed a univariate analysis and utilized a 1:2 ratio propensity score matching method to minimize the effects of selection bias. We employed weighted univariate as well as three multivariate logistic regression models both prior to and following matching, with the aim of examining the association between dietary copper intake and PD risk. Finally, we used the restricted cubic spline (RCS) methodology in order to investigate possible non-linear relationships. Furthermore, subgroup analysis was undertaken to elicit further understanding concerning the association between copper intake and PD. A negative correlation resulted between dietary copper intake and PD risk in both univariate and multivariate logistic regression models, prior to and following matching. Our findings demonstrate that there is a nonlinear, dose-dependent relationship between copper intake and PD, according to our RCS analysis. In subgroup analysis, copper intake was identified as an important protective factor for individuals who were non-Hispanic White, unmarried, and had completed higher education. Electrochemical dechlorination reaction (EDR) is a promising, environmentally friendly, and economically profitable technology for treating chlorinated organic pollutants. For efficient environmental protection, electrocatalysts with high stability and low cost are of extremely significance to the development of EDR technology. Carbon-based materials have aroused broad interest as electrocatalysts for many electrochemical reactions due to their characteristics including large specific surface area, controllable structure, good conductivity, and chemical stability. For EDR, the carbon-based materials also show many unique superiorities, like strong adsorption capacity to chlorinated organic compounds (COCs), excellent catalytic activity and stability, and environmental compatibility. This review starts with a detailed summary on the mechanisms of electrochemical dechlorination (direct and indirect electron transfer pathway) and factors affecting the effectiveness of EDR. Then the paper comprehensively overviews the current progresses of carbon-based materials for EDR of COCs, following their two major application scenarios, i.e., directly as electrocatalysts and as advanced supports for other catalysts. Moreover, the formation of different active sites in carbon-based electrocatalysts and their EDR activities are analyzed. Finally, the current challenges and perspectives in this field are discussed. Mutations of NKX2-5 largely contribute to congenital heart diseases (CHDs), especially atrial septal defect (ASD). We identified a novel heterozygous splicing mutation c.335-1G > A in NKX2-5 gene in an ASD family via whole exome sequencing (WES) and linkage analysis. Utilizing the human induced pluripotent stem cell (hiPSC)-derived cardiomyocytes (hiPSC-CMs) as a disease model, we showed that haploinsufficiency of NKX2-5 contributed to aberrant orchestration of apoptosis and proliferation in ASD patient-derived hiPSC-CMs. RNA-seq profiling and dual-luciferase reporter assay revealed that NKX2-5 acts upstream of PYK2 via miR-19a and miR-19b (miR-19a/b) to regulate cardiomyocyte apoptosis. Meanwhile, miR-19a/b are also downstream mediators of NKX2-5 during cardiomyocyte proliferation. The novel splicing mutation c.335-1G > A in NKX2-5 and its potential pathogenic roles in ASD were demonstrated. Highland birds evolve multiple adaptive abilities to cope with the harsh environments; however, how they adapt to the high-altitude habitats via the gut microbiota remains understudied. Here we integrated evidences from comparative analysis of gut microbiota to explore the adaptive mechanism of black-necked crane, a typical highland bird in the Qinghai-Tibet Plateau. Firstly, the gut microbiota diversity and function was compared among seven crane species (one high-altitude species and six low-altitude species), and then among three populations of contrasting altitudes for the black-necked crane. Microbiota community diversity in black-necked crane was significantly lower than its low-altitude relatives, but higher microbiota functional diversity was observed in black-necked crane, suggesting that unique bacteria are developed and acquired due to the selection pressure of high-altitude environments. The functional microbial genes differed significantly between the low- and high-altitude black-necked cranes, indicating that altitude significantly impacted microbial communities' composition and structure. Adaptive changes in microbiota diversity and function are observed in response to high-altitude environments. These findings provide us a new insight into the adaptation mechanism to the high-altitude environment for birds via the gut microbiota. As a kind of sensing and imaging fluorescent probe with the merit of low toxicity, good stability, and environment-friendly, silicon nanoparticles (SiNPs) are currently attracting extensive research. In this work, we obtained mitoxantrone-SiNPs (MXT-SiNPs) with green emission by one-pot synthesis under mild temperature condition. The antenna based on pyridoxal phosphate (PLP) was designed for light-harvesting to enhance the luminescence of MXT-SiNPs and to establish a novel sensing strategy for alkaline phosphatase (ALP). PLP transfers the absorbed photon energy to MXT-SiNPs by forming Schiff base. When PLP is dephosphorized by ALP, the released free hydroxyl group reacts with aldehyde group to form internal hemiacetal, which leads to the failure of Schiff base formation. Based on the relationship between antenna formation ability and PLP hydrolysis degree, the activity of ALP can be measured. A good linear relationship was obtained from 0.2 to 3.0 U/L, with a limit of detection of 0.06 U/L. Furthermore, the sensing platform was successfully used to detect ALP in human serum with recovery of 97.6–106.2%. The rational design of antenna elements for fluorescent nanomaterials can not only provide a new pathway to manipulate the luminescence, but also provide a new direction for fluorescence sensing strategy. Immunostimulatory therapies based on pattern recognition receptors (PRRs) have emerged as an effective approach in the fight against cancer, with the ability to recruit tumor-specific lymphocytes in a low-immunogenicity tumor environment. The agonist cyclic dinucleotides (CDNs) of the stimulator of interferon gene (STING) are a group of very promising anticancer molecules that increase tumor immunogenicity by activating innate immunity. However, the tumor immune efficacy of CDNs is limited by several factors, including relatively narrow cytokine production, inefficient delivery to STING, and rapid clearance. In addition, a single adjuvant molecule is unable to elicit a broad cytokine response and thus cannot further amplify the anticancer effect. To address this problem, two or more agonist molecules are often used together to synergistically enhance immune efficacy. In this work, we found that a combination of the STING agonist CDGSF and the Toll-like receptor 7/8 (TLR7/8) agonist 522 produced a broader cytokine response. Subsequently, we developed multicomponent nanovaccines (MCNVs) consisting of a PC7A polymer as a nanocarrier encapsulating the antigen OVA and adjuvant molecules. These MCNVs activate bone marrow-derived dendritic cells (BMDCs) to produce multiple proinflammatory factors that promote antigen cross-presentation to stimulate specific antitumor T-cell responses. In in vivo experiments, we observed that MCNVs triggered a strong T-cell response in tumor-infiltrating lymphocytes, resulting in significant tumor regression and, notably, a 100% survival rate in mice through 25 days without other partnering therapies. Cell aging is the result of deteriorating competence in maintaining cellular homeostasis and quality control. Certain cell types are able to rejuvenate through asymmetric cell division by excluding aging factors, including damaged cellular compartments and extrachromosomal rDNA circles, from entering the daughter cell. Recent findings from the budding yeast S. cerevisiae have shown that gametogenesis represents another type of cellular rejuvenation. Gametes, whether produced by an old or a young mother cell, are granted a renewed replicative lifespan through the formation of a fifth nuclear compartment that sequesters the harmful senescence factors accumulated by the mother. Here, we describe the importance and mechanism of cellular remodeling at the nuclear envelope mediated by ESCRT-III and the LEM-domain proteins, with a focus on nuclear pore biogenesis and chromatin interaction during gamete rejuvenation. Neutral aqueous rechargeable Co3O4//Zn batteries with high-output voltage and outstanding cycling stability have yielded new insights into wearable energy-storage devices. To meet the increasing demand for a means of powering wearable and portable devices, the development of a high-performance fiber-shaped Co//Zn battery would be highly desirable. However, the intrinsically poor conductivity of Co3O4 significantly restricts the application of these high-capacity and high-rate aqueous rechargeable battery. Encouragingly, density functional theory (DFT) calculations demonstrate that the substitution of Zn for Co3+ leads to an insulator-metal transition in the Zn-doped Co3O4 (Zn-Co3O4). In this study, we used metallic Zn-Co3O4 nanowire arrays (NWAs) as a novel binder-free cathode to successfully fabricate an all-solid-state fiber-shaped aqueous rechargeable (AFAR) Co//Zn battery. The resulting fiber-shaped Co//Zn battery takes advantage of the enhanced conductivity, increased capacity, and improved rate capability of Zn-Co3O4 NWAs to yield a remarkable capacity of 1.25 mAh·cm−2 at a current density of 0.5 mA·cm−2, extraordinary rate capability (60.8% capacity retention at a high current density of 20 mA·cm−2) and an admirable energy density of 772.6 mWh·cm−3. Thus, the successful construction of Zn-Co3O4 NWAs provides valuable insights into the design of high-capacity and high-rate cathode materials for aqueous rechargeable high-voltage batteries. Images created from measurements made by wireline microresistivity imaging tools have longitudinal gaps when the well circumference exceeds the total width of the pad-mounted electrode arrays. The gap size depends on the tool design and borehole size, and the null data in these gaps negatively affect the quantitative evaluation of reservoirs. Images with linear and texture features obtained from microresistivity image logs have distinct dual fabric features because of logging principles and various geological phenomena. Linear image features usually include phenomena such as fractures, bedding, and unconformities. Contrarily, texture-based image features usually indicate phenomena such as vugs and rock matrices. According to the characteristics of this fabric-based binary image structure and guided by the practice of geological interpretation, an adaptive inpainting method for the blank gaps in microresistivity image logs is proposed. For images with linear features, a sinusoidal tracking inpainting algorithm based on an evaluation of the validity and continuity of pixel sets is used. Contrarily, the most similar target transplantation algorithm is applied to texture-based images. The results obtained for measured electrical imaging data showed that the full borehole image obtained by the proposed method, whether it was a linear structural image reflecting fracture and bedding or texture-based image reflecting the matrix and pore of rock, had substantially good inpainting quality with enhanced visual connectivity. The proposed method was effective for inpainting electrical image logs with large gaps and high angle fractures with high heterogeneity. Moreover, ladder and block artifacts were rare, and the inpainting marks were not obvious. An electrochemical immunosensor based on ferrocene (Fc)-functionalized nanocomposites was fabricated as an efficient electroactive signal probe to amplify electrochemical signals for Salmonella typhimurium detection. The electrochemical signal amplification probe was constructed by encapsulating ferrocene into S. typhimurium–specific antimicrobial peptides Magainin I (MI)-Cu3(PO4)2 organic-inorganic nanocomposites (Fc@MI) through a one-step process. Magnetic beads (MBs) coupled with antibody were used as capture ingredient for target magnetic separation, and Fc@MI nanoparticles were used as signal labels in the immunoassays. The sandwich of MBs-target-Fc@MI assay was performed using a screen-printed carbon electrode as transducer surface. The immunosensor platform presents a low limit of detection (LOD) of 3 CFU·mL−1 and a linear range from 10 to 107 CFU·mL−1, with good specificity and precision, and was successfully applied for S. typhimurium detection in milk. Multiple ultrashort laser pulses are widely used in optical spectroscopy, optoelectronic manipulation, optical imaging and optical signal processing etc. The laser pulse multiplication, so far, is solely realized by using the optical setups or devices to modify the output laser pulse from the optical gain medium. The employment of these external techniques is because the gain medium itself is incapable of modifying or multiplying the generated laser pulse. Herein, with single femtosecond laser pulse excitation, we achieve the double-pulsed stimulated emission with pulse duration of around 40 ps and pulse interval of around 70 ps from metal-halide perovskite multiple quantum wells. These unique stimulated emissions originate from one fast vertical and the other slow lateral high-efficiency carrier funneling from low-dimensional to high-dimensional quantum wells. Furthermore, such gain medium surprisingly possesses nearly Auger-free stimulated emission. These insights enable us a fresh approach to multiple the ultrashort laser pulse by gain medium. Laser pulse multiplication is desired in many applications but has been challenging to realize by gain medium. Here Guo et al. achieve double-pulsed stimulated emission in quasi-2D metal-halide perovskites due to the two-channel carrier funneling effect in their multiple-quantum-wells structure. Although a massive research has been devoted on the exploration of noble metal–based nanozyme, less progress has been made in the investigation of palladium (Pd) nanozyme and the interaction between ions and Pd nanozyme. In this study, a new type of Pd nanozyme was prepared by a facile one-pot approach by using carboxylated chitosan as the stabilizer. Owing to the synergistic effect of carboxylated chitosan stabilized Pd nanoparticles (CC-PdNPs) can effectively catalyze the H2O2-mediated oxidation of 3,3′,5,5′-tetramethylbenzidine sulfate (TMB) accompanied by a blue color change (oxidized TMB), indicating the peroxidase-like activity of CC-PdNPs. Furthermore, the Michaelis-Menten constants and catalytic stability of CC-PdNPs render them suitable for environmental analysis and bio-detection. Here, we found that while introducing the iodine ions (I−) into the reaction medium, the peroxidase-like activity of CC-PdNPs has been rapidly and effectively inhibited through the formation of Pd-I bond; thus, the active sites of PdNPs can be blocked by I−. Based on this specific inhibition by I−, a facile colorimetric assay has been performed for the detection of I− with an extremely low limit of detection (0.19 nM). Furthermore, the practicality of the proposed sensor also has been demonstrated in tap water, and the satisfactory recoveries were obtained. The AAA + ATPase R2TP complex facilitates assembly of a number of ribonucleoprotein particles (RNPs). Although the architecture of R2TP is known, its molecular basis for acting upon multiple RNPs remains unknown. In yeast, the core subunit of the box C/D small nucleolar RNPs, Nop58p, is the target for R2TP function. In the recently observed U3 box C/D snoRNP as part of the 90 S small subunit processome, the unfolded regions of Nop58p are observed to form extensive interactions, suggesting a possible role of R2TP in stabilizing the unfolded region of Nop58p prior to its assembly. Here, we analyze the interaction between R2TP and a Maltose Binding Protein (MBP)-fused Nop58p by biophysical and yeast genetics methods. We present evidence that R2TP interacts largely with the unfolded termini of Nop58p. Infectious complications are common and sometimes life threatening in patients with immune thrombocytopenia (ITP), mainly due to the immune-suppressive therapy. Recent evidence suggests a potential role of platelets in the inflammation process. In this clinical study, we further investigated the role of thrombocytopenia on infections in patients with primary ITP. We retrospectively evaluated data from the recently published large randomized clinical trial of a cohort of 195 patients with primary ITP, who were randomized for prednisone or high-dose dexamethasone. From 158 patients (81%), data on platelet count and infections within the first month of treatment were collected. In this period, 24% of the ITP patients had an infection. Patients with infection had significant lower platelet counts during the first month of treatment leading to a significant lower therapy response at 1 month and a significant longer hospital stay (14.0 versus 9.8 days). Additionally, Cox regression analysis showed that an increase in platelet count of 20 × 109/L led to a reduction of 52% in infections in the next week, showing low platelet count is a significant risk factor for infection. Platelet transfusion led to an increase in platelet count in ITP patients without infection, but not in patients with infection. In conclusion, infections are common in patients with primary ITP leading to significant worse response rates and a longer hospital stay. Purpose: Sclerostin is an osteocyte-derived protein that has a potent inhibitory effect on osteoblast activity. The osteocyte apoptosis induced by various causes of osteonecrosis of the femoral head (ONFH) plays a key role in the promotion of femoral head collapse. But the effect of altering sclerostin level on the collapse of ONFH has not been studied. Our aim was to assess the role of sclerostin level in the collapse of ONFH.Methods: Between May 2016 and November 2016, 236 subjects were enrolled in the present study. The patients were classified according to the Association Research Circulation Osseous (ARCO) classification. The clinical bone histomorphology, the expression position, and level of sclerostin as well as the plasma sclerostin level were evaluated.Results: The sclerostin level was significantly lower in the non-traumatic ONFH group than those in the healthy control group (P = 0.002). The sclerostin level was negatively associated with ARCO stages (r = - 0.239, P = 0.009) and significantly lower in the postcollapse group (P = 0.025).Conclusions: The reduced expression of sclerostin may play a key role in the collapse process of ONFH and be predictive of the disease progression of ONFH. This paper presents an integrated investigation of the ground stability of a deep gateroad with a 1 km burial depth based on a field test, case studies, and numerical modelling. In situ stress measurements and mechanical properties tests were first conducted in the test site. Then, the deformation behavior, stress and yield zone distributions, as well as the bolts load of the gateroad, were simulated using FLAC3D software. The model results demonstrated that the soft rock properties and high in situ stress were the main factors for the deep gateroad instability, and the shear slip failure induced by the high stress was the primary failure model for the deep rock mass. In addition, the unsuitable support patterns, especially the relatively short bolts/cables with low pre-tensions, the lack of high-strengthen secondary supports and the unsupported floor strata, also contributed to the gateroad instability. Subsequently, a new combined supporting strategy, incorporating longer bolts/cables, yielding ring supports, and grouting measures, was proposed for the deep gateroad, and its validity was verified via field monitoring. The conversion of solar energy into chemical fuels can potentially address many of the energy and environment related challenges we face today. In this study, we have demonstrated a photochemical diode artificial photosynthesis system that can enable efficient, unassisted overall pure water splitting without using any sacrificial reagent. By precisely controlling charge carrier flow at the nanoscale, the wafer-level photochemical diode arrays exhibited solar-to-hydrogen efficiency ~3.3% in neutral (pH ~ 7.0) overall water splitting reaction. In part of the visible spectrum (400-485 nm), the energy conversion efficiency and apparent quantum yield reaches ~8.75% and ~20%, respectively, which are the highest values ever reported for one-step visible-light driven photocatalytic overall pure water splitting. The effective manipulation and control of charge carrier flow in nanostructured photocatalysts provides critical insight in achieving high efficiency artificial photosynthesis, including the efficient and selective reduction of CO2 to hydrocarbon fuels. The mechanical properties of rock under unloading condition have significant differences with those under conventional loading condition. To investigate the failure characteristics of schist under complex stress state, uniaxial and trixial compression and unloading confining pressure tests have been carried out on samples in the paper. The results show that the rock samples failed in coupling mode of shear and tension under uniaxial compression, and failed in shear mode under conventional trixial compression. However, the failure modes of the samples can be classified as shear, multiple fracturing, and multiple extension, and the strength of schist is closely related with the failure mode under unloading confining condition. For the unloading condition, the peak strengths of schist samples with shear failure mode are close to those under conventional trixial compression, but the peak strengths with multiple fracturing or multiple extension mode are much higher than those under trixial compression. The internal structure characteristics and stress paths were found to have major influences on the failure modes of schist. Dicyandiamide (DICY) is a common ligand that exhibits low toxicity but can irritate the skin and eyes and cause methemoglobinemia on long-term exposure. Crystalline Cu-dicyandiamide (Cu-DICY) was obtained via facile synthesis and its molecular structure and theoretical Raman spectra were simulated by using density functional theory (DFT). The results suggested that the Cu2+ coordinates with two H2O molecules and two different DICY molecules (an imino DICY and an amino DICY). The stability constants of Cu-DICY were calculated, and the electrochemical properties were studied. Two electrochemical redox processes occur in Cu-DICY in an aqueous solution: a reversible reaction with a formal potential of 0.2 V vs. MSE and an irreversible reaction between –0.4 and –1.2 V vs. MSE. The standard rate constant k0 for the reversible reaction was estimated to be 7.6×10–3 cm/s. In addition, based on the reversible reaction of Cu-DICY, square wave voltammetry was used to rapidly determine the concentration of Cu(II) and the detection limit was 66.7 μg/L, which satisfies the detection limit requirements for copper in tap water (2 mg/L) as provided by the World Health Organization. This paper presents an integrated approach for field test and numerical modelling to investigate the relationship between gateroad stability and yield pillar size. The test site is located at Yuncheng city, Shanxi Province, China. Field tests indicated that when the yield pillar width was 17 m, the total convergence of the roof, yield pillar rib and virgin coal rib were 882, 587 and 352 mm, respectively, and severe roof sagging and yield rib spalling occurred during the panel retreat. A meticulously validated numerical model, incorporating a double-yield model for the gob materials and calibrated parameters, was developed to investigate the stress changes and yield zone distribution across the yield pillar with different sizes. The results of the simulation indicate that a yield pillar 17 m wide puts the gateroad in a high-stress environment; conversely, a yield pillar 8 m wide is subjected to a relatively low load and puts the gateroad in a good stress environment. Consequently, the rational yield pillar width was estimated at 8 m, and a support strategy was proposed. Field measurement data demonstrate that the newly designed pillar size and support pattern can efficiently ensure gateroad stability. The proposed numerical simulation procedure and calibrated method could be a viable alternative approach to yield pillar design. Herein, two classic animal behavior despair tests-the forced swim test and tail suspension test were used to evaluate the antidepressant-like activity of saringosterol from Sargassum fusiforme in mice. Saringosterol was found to significantly shorten immobility time in the forced swim test and tail suspension test at doses of 10, 20, and 30 mg/kg in mice. The measurement of locomotor activity indicated that saringosterol had no central nervous system-stimulating effects. In addition, it was found that the saringosterol significantly increased noradrenaline, serotonin (5-HT), and the metabolite 5-hydroxyindoleacetic acid in the mouse brain, suggesting that the antidepressant-like activity may be mediated through these neurotransmitters. Density functional theory is used to investigate geometric structures and mechanisms for hydroxylation and epoxidation from propene for four non-heme iron complexes, [Fe(L)(TMC)(O)], which are the inverted isomers of [Fe(O)(TMC)(L)] (L = acetonitrile (AN), monoanionic trifluoroacetate (TF), azide (N3), thiolate (SR)). The Fe(IV)O unit is found to be sterically less hindered in [Fe(L)(TMC)(O)] than that in [Fe(O)(TMC)(L)]. Becke, three-parameter, Lee-Yang-Parr (B3LYP) calculations show that hydroxylation and epoxidation proceed via a two-state-reactivity on competing triplet and quintet spin surfaces; and the reactions have been invariably mediated by the S = 2 state. The reaction pathways computed reveal that 2-AN is the most reactive in the four [Fe(L)(TMC)(O)] complexes; along the reaction pathway, the axial ligand moves away from the iron center, and thus, the energy of the LUMO decreases. The anionic axial ligand, which is more electron releasing than neutral AN, shows a strong overlap of iron orbitals. Thus, the anionic ligand does not move away from the iron center. The H-abstraction is affected by the tunneling contribution, the more electron donation power of the axial ligand, the more effect of the tunneling contribution. Adding the tunneling correction, the relative reactivity of the hydroxylation follows the trend: 2-AN > 2-SR ≈ 2-N3 > 2-TF. However, for the epoxidation, the reactivity is in the following order of 2-AN > 2-TF > 2-N3 > 2-SR. In this study, we examined a series of 2′,4′,6′-trihydroxychalcone derivatives for their inhibitory activity as inhibitors of CDC25B and PTP1B. The pharmacological results showed that all of the tested compounds significantly inhibited CDC25B and PTP1B phosphatase in vitro. Among them, three compounds 2, 6, and 7 had the best inhibition activity, with inhibition rates against CDC25B were 99.56, 99.68, and 99.63 %, respectively, and with inhibition rates against PTP1B were 98.99, 99.37, and 98.08 %, respectively, which is similar to reference drugs NaVO and Oleanolic acid, respectively. Cytotoxic activity assays showed compounds 2, 6, and 7 are potent against HeLa and HCT116. Moreover, compound 6 delayed the potent antitumor inhibitory activity in a colo205 xenograft model in vivo. The insertion of CO2 into the (PSiP)palladium-allyl bond has been investigated using DFT. Three possible modes of CO2 insertion into (PSiP)Pd-allyl bond have been calculated, that is, direct 1.2-insertion mode, metallo-ene mode, and SE2 mode. The metallo-ene mode is the most favorable via the six-membered ring transition state. The results of calculations are consistent with the regioselectivity observed experimentally. The steric and electronic effects of different phosphine substituents have been evaluated by ONIOM and energy decomposition analysis (EDA) methods. For the phosphine substituents CP(i-Pr)2 and PPh2, the contribution of electronic effect is greater than that of steric effect for the CO2 insertion into (PSiP)Pd-allyl bond; while for the phosphine substituent PMe2, the contribution of steric effect is slightly greater than that of electronic effect. The controllability and the observability of fuzzy dynamical system are studied. The uncertainty of the fuzzy dynamical system is due to the initial condition. The available information on the uncertainty can be described via a membership function. In this paper, we propose a systematic approach to investigate the controllability and the observability of fuzzy dynamical system via the use of the membership function. The concepts of the controllability and the observability in the fuzzy sense are presented via the use of the membership function. The necessary and sufficient conditions for the controllability and the observability of the linear time-variant and time-invariant uncertain system are derived. Danhong injection, a Chinese Materia Medica standardized product extracted from Radix Salviae miltiorrhizae and Flos Carthami tinctorii, is used extensively for the treatment of cerebrovascular diseases such as acutely cerebral infarction in clinic. In this study, we further investigated the mechanisms of Danhong injection on cerebral ischemia/reperfusion (I/R) damage relating to Nrf2/ARE signalling pathway in vivo and in vitro. For in vivo experiment, cerebral I/R injury was induced through middle cerebral artery occlusion. Rats were randomly divided into five groups: sham-operated group, I/R injury group, 6 mg/kg edaravone injection (positive control drug) group, 0.9 ml/kg Danhong injection (DHI-L) group, 1.8 ml/kg Danhong injection (DHI-H) group. The neurological score, cerebral infarction and brain edema were assessed while levels of superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA) in brain tissue were also evaluated. Transcription levels of nuclear factor erythroid 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), NAD(P)H: quinone oxidoreductase 1 (NQO1) were analysed by real-time polymerase chain reaction. For in vitro experiment, mouse Neuro-2A cells were wounded with HO then cell viability and mRNA transcriptions levels of Nrf2, HO-1, NQO1 were detected. Protein expression level of Nrf2 was assayed by western blotting. The results showed that Danhong injection could ameliorate neurological score, cerebral infarction and brain edema. Also it can increase levels of SOD, GSH and decrease of MDA and upregulate expressions of Nrf2, HO-1, NQO1 in ischemic brain tissue in vivo. What's more, it increased the mRNA transcription of Nrf2 and HO-1 and upregulated protein expression of Nrf2 in vitro. Purpose: We investigated the disagreement between the positive cell-free fetal DNA test for trisomy 13 and the standard cytogenetic diagnosis of one case. Methods: Cell-free fetal DNA testing was performed by massively parallel sequencing. We used conventional cytogenetic analysis to confirm the commercial cell-free fetal DNA testing. Additionally, postnatal fluorescent in situ hybridization (FISH) testing was performed on placental tissues. Results: The cell-free fetal DNA testing result was positive for trisomy 13. G-banded analysis of amniotic fluid was normal, 46, XY. FISH testing of tissues from four quadrants of the placenta demonstrated mosaicism for trisomy 13. Conclusions: A positive cell-free fetal DNA testing result may not be representative of the fetal karyotype because of placental mosaicism. Summarize the value of the change of different parts in treadmill exercise test (TET) ECG to coronary heart disease (CHD) diagnosis. Four hundred and forty-five cases have been included in this investigation, which stayed in our hospital from January of 2006 to March of 2011 and underwent TET and coronary arteriography (CAG). The change of different parts in TET ECG in these patients had been retrospective summarized to determine its diagnosis value to CHD. (1) In the 445 cases of TET testers, 200 cases showed positive in TET with 150 cases of positive CAG, and 50 cases of negative CAG; 245 cases showed negative in TET with 206 cases of negative CAG, and 39 cases of positive CAG. The diagnosis sensitivity of CHD was 79.36 % (150/189), the specificity was 80.47 % (206/256), positive prediction value was 75.00 % (150/200), negative prediction value was 84.08 % (206/245), and false-positive rate was 25.00 % (50/200) with prediction accuracy of 80.00 % (356/445). (2) In the 200 cases with positive TET: 51 cases were in the limb lead group; 73 cases were in the chest lead group; and 76 cases were in the limb lead + chest lead group. There were 150 cases showing positive in CAG: 22 cases were in the limb lead group; 58 cases were in the chest lead group; and 70 cases were in the limb lead + chest lead group. The positive diagnosis rate of ST change in the chest lead was obviously higher than that of simple limb lead group ( P < 0.05). (3) People with healthy coronary artery will have decreased amplitude of R wave while patients with coronary stenosis have elevated amplitude of R wave. (4) As for the T wave, the positive CAG had no statistical significance between normal T wave group and TET positive group ( P > 0.05); CAG results had statistical significance between normal T wave group and TET negative group ( P < 0.05). (5) Positive CAG results had no statistical significance between U-wave inversion group and TET positive group ( P > 0.05); positive CAG results has statistical significance when TET negative group compared with U-wave inversion group or TET positive group ( P < 0.05). TET is a relatively idea invasive diagnosis method for coronary disease, which can be utilized to evaluate the stage of CHD when integrating with the change of TET ECG. [ABSTRACT FROM AUTHOR]

Purpose: To explore the prevalence and clinical features, especially the reproductive function, of 45,X mosaicism patients in northeast China. Methods: GTG-banding was performed on a series of 2,250 patients from our genetic counseling clinic. Each of these patients underwent a physical examination and was interviewed about their medical history and reproductive problems. Literature on 45,X mosaicism was accessed using PubMed and reviewed. Results: The prevalence of 45,X mosaicism in northeast China is 0.36 % (8/2250), and the mosaic karyotype of our study accounted for 61.54 % (8/13) of Turner syndrome cases. This is comparable with studies from Asia, Europe, South America and other regions. The affected patients showed genital abnormalities, abnormal pregnancy or infertility. Conclusion: 45,X mosaicism is commonly seen in the genetic counseling clinic. Extensive cytogenetic assessment may improve the detection rate in patients with congenital dysplasia, or history of abnormal pregnancy or infertility. Purpose: To compare the frequency of chromosomal heteromorphisms in reproductive failure and fertile control individuals in Northeast China, and investigate the impact on reproductive failure Methods: 1751 males and 1424 couples with reproductive failure ( n = 4599) and 777 fertile control individuals in Northeast China were enrolled. Chromosome karyotype analysis was performed on peripheral blood lymphocytes with standard G-banding. Additionally, C-banding was performed with heterochromatin heteromorphisms, and NORs-banding with satellites/stalks variations. Multiplex polymerase chain reaction (PCR) adopted for the amplification using nine specific sequence tagged sites (STS) were used to detect Y-chromosome microdeletions with Y chromosome variations (Yqh±). At the same time, 38 heteromorphic probands' family members were recalled for performing karyotype analysis and to be surveyed for their detailed reproductive history. Results: The frequency of chromosomal heteromorphisms in reproductive failure patients (2.74 %, 126/4599) was of no statistically significant difference as compared with fertile control individuals (2.06 %, 16/777) ( P > 0.05). Eight cases of Y variation (Yqh±) probands with Y-chromosomal microdeletions were detected among 44 reproductive failure patients and 6 fertile control men. In the 38 recalled families, the probands of fathers or mothers, even some of their brothers or sisters, had the same heteromorphic karyotypes as probands' despite that they didn't have any adverse reproductive history. Conclusions: There was no statistically significant difference in frequency of chromosomal heteromorphisms between reproductive failure and fertile control individuals in Northeast China. Males with Y variations (Yqh±) should be ordered Y-chromosomal microdeletions detection. Through the analysis of 38 recalled families, we can also conclude that chromosomal heteromorphisms were not the impact factors for reproductive failure. [ABSTRACT FROM AUTHOR]

A prerequisite for determination of chromosome dynamics in live cells is development of a method for staining or marking the chromosome of interest. We describe here a unique chromosome-tracking system that differentially marks two large chromosome segments from homologs in the budding yeast Saccharomyces cerevisiae. Using yeast genetics and the special features at the repetitive ribosomal RNA (rRNA) gene cluster, we incorporated arrays of the tet operator and the lac operator into each repeat of the two rDNA homologs by homologous recombination. Expression of tet repressor-fused green fluorescent protein and lac repressor-fused red fluorescent protein in engineered cells led to the differential labeling of rDNA homologs. Using live-cell three-dimensional fluorescence microscopy, we showed that homologs undergo contraction and expansion cycles in an actin-dependent manner during meiosis and that chromosome mobility appears to be correlated with nuclear positioning. Our observations further revealed that, in contrast to mitosis, in meiosis the yeast nucleolus, the site of rRNA processing, was disassembled upon anaphase onset, suggesting a differential regulation of the rDNA array during meiotic chromosome segregation. Because rRNA genes are highly conserved, a similar chromosome-engineering approach may be adaptable in other eukaryotes for functional assays of chromosome organization in live cells. Purpose: Matrix metalloproteinases (MMPs) play important roles in pathogenesis and development of cancer. Recently, lots of studies showed that there were associations between polymorphisms in the promoter regions of MMPs and risk of colorectal cancer; however, the results remained inconclusive. To clarify these associations, we conducted a meta-analysis. Methods: We conducted a computerized literature search in the database of PubMed, Embase, ISI Web of Knowledge, and Medline (from January 2000 to July 2010). Overall and subgroup analysis based on the ethnicity of study population was carried out. Odds ratio (OR) and 95% confidence interval (95%CI) were used to evaluate these associations. Statistical analysis was performed with software Review Manager (version 5.0). Results: There were 12 studies involving five polymorphic sites in four MMP genes. For MMP1 (−1607), 2G polymorphism increased the risk of colorectal cancer under dominant and recessive models (dominant, OR = 1.23, 95%CI 1.01-1.49; recessive, OR = 1.52, 95%CI 1.30-1.77). For MMP3 (−1612), 6A/6A genotype increased this risk under the recessive model (OR = 1.33, 95%CI 1.04-1.70); however, this association was lost under the dominant model. For MMP2 −1306 C>T, MMP3 −1171 5A>6A, and MMP9 −1562 C>T, there was no association between these polymorphisms and the risk of colorectal cancer under the dominant and recessive models. Conclusions: Polymorphisms of MMP1 (−1607) and MMP3 (−1612) increased the risk of colorectal cancer; these two polymorphic sites could be used as markers for early diagnosis of colorectal cancer. However, for MMP2 (−1306), MMP3 (−1171), and MMP9 (−1562), further studies with large sample size should be carried out. [ABSTRACT FROM AUTHOR]

The Arabidopsis gene FRO6(AtFRO6) encodes ferric chelate reductase and highly expressed in green tissues of plants. We have expressed the gene AtFRO6 under the control of a 35S promoter in transgenic tobacco plants. High-level expression of AtFRO6 in transgenic plants was confirmed by northern blot analysis. Ferric reductase activity in leaves of transgenic plants grown under iron-sufficient or iron-deficient conditions is 2.13 and 1.26 fold higher than in control plants respectively. The enhanced ferric reductase activity led to increased concentrations of ferrous iron and chlorophyll, and reduced the iron deficiency chlorosis in the transgenic plants, compared to the control plants. In roots, the concentration of ferrous iron and ferric reductase activity were not significantly different in the transgenic plants compared to the control plants. These results suggest that FRO6 functions as a ferric chelate reductase for iron uptake by leaf cells, and overexpression of AtFRO6 in transgenic plants can reduce iron deficiency chlorosis. Bifunctional spherical polyelectrolyte brushes (SPBs) with tunable thermo- and pH-sensitivity are synthesized by combining thermo-controlled emulsion polymerization and photo-emulsion polymerization. They consist of a spherical polystyrene core and a shell of mixed brushes of poly( N-isopropylacrylamide) (PNIPAM) and poly(acrylic acid) (PAA) whose composition can be easily modulated by the dose of monomers. The kinetics of SPB synthesis as well as their size change with temperature and pH is determined by dynamic light scattering (DLS). Three alginate lyases (A, B, and C) from an alginate-degrading marine bacterium strain HZJ216 isolated from brown seaweed in the Yellow Sea of China and identified preliminarily as Pseudomonas fluorescens are purified, and their biochemical properties are described. Molecular masses of the three enzymes are determined by SDS-PAGE to be 60.25, 36, and 23 kDa with isoelectric points of 4, 4.36, and 4.59, respectively. Investigations of these enzymes at different pH and temperatures show that they are most active at pH 7.0 and 35 °C. Alginate lyases A and B are stable in the pH range of 5.0-9.0, while alginate lyase C is stable in the pH range of 5.0-7.0. Among the metal ions tested, additions of Na, K, and Mg ions can enhance the enzyme activities while Fe, Fe, Ba, and Zn ions show inhibitory effects. The substrate specificity results demonstrate that alginate lyase C has the specificity for G block while alginate lyases A and B have the activities for both M and G blocks. The chloroplast is one of the most important organelles in plants. Proteomic investigations of chloroplasts have been undertaken for many herb plant species, but to date no such investigation has been reported for woody plant chloroplasts. In the present study we initiated a systematic proteomic study of Populus chloroplasts using a shotgun proteomic method. After isolation of chloroplasts and tryptic digestion of the proteins, the protein fragments were separated via HPLC using an SCX column, and the peptides were analyzed by LC-MS/MS; 119 proteins were successfully identified. Based on annotation information in the UniProtKB/Swiss-Prot database, these proteins were identified as being localized in the chloroplast thylakoid membrane, chloroplast stroma, chloroplast thylakoid lumen, and plastoglobules. Over 50% of all identified proteins were confirmed as chloroplast thylakoid proteins, and 85 are encoded by the chloroplast genome with the remaining proteins encoded by the nuclear genome. Based on functional annotation, these proteins were classified into four functional categories, including photosynthesis, redox regulation and stress, primary and secondary metabolism, transport and signaling. The current study demonstrates vanadium plays the role of antitumor, and its antitumor effect is dosage-dependent. N-acetyl-galactosamine-transferase 2 (polypeptide: N-acetyl-α-galactosaminyl-transferases 2, ppGalNAc-T2) is a member of ppGalNAcTs (polypeptide: N-acetyl-α-galactosaminyl-transferases) family, which proves to play a vital role in the tumor emergence and development process. In this study, we focused on ppGalNAc-T2 and vanadium and aimed to determine whether ppGalNAc-T2 is correlated with vanadium's antitumor effect. We discovered that ppGalNAc-T2 changed with the variation of HL-60 cell growth induced by vanadium at mRNA level. Peanut agglutinin (PNA) is an exogenous lectin. PpGalNacT2 can be indirectly recognized by PNA. By means of flow cytometry and immunofluorescent staining, we found the deviation of PNA binding increased significantly at high concentration vanadium. Then we docked one of the possible compound substances of vanadium onto the body, VO (molecular formula OV, partial vanadate tetramer) and ppGalNAcT2, and simulated them via molecular dynamics, which showed that VO may inhibit the activity of the enzyme by stemming conformational changes of a key loop of ppGalNAcT2. To sum up, our results suggested that ppGalNacT2 participated in vanadium induced HL-60 cell differentiation, which might be able to provide a new mechanism of vanadium's antitumor effect. In this study, we presented the construction of a library of expression clones for the herpes simplex virus type 1 (HSV-1) proteome and subcellular localization map of HSV-1 proteins in living cells using yellow fluorescent protein (YFP) fusion proteins. As a result, 21 proteins showed cytoplasmic or subcytoplasmic localization, 16 proteins showed nuclear or subnuclear localization, and others were present both in the nucleus and cytoplasm. Interestingly, most capsid proteins showed enriched or exclusive localization in the nucleus, and most of the envelope proteins showed cytoplasmic localization, suggesting that subcellular localization of the proteins correlated with their functions during virus replication. These results present a subcellular localization map of HSV-1 proteins in living cells, which provide useful information to further characterize the functions of these proteins. Chain reactions on a surface offer an important route to linear nanopatterning. We recently reported cooperative reactions on a silicon surface in which the reaction of one halogen atom with a silicon atom of a silicon dimer induced the halogenation of its neighbouring silicon atom through surface-mediated charge transfer. The reaction was unable to propagate further but here we describe how, by chemically bridging the gaps between the rows of these silicon dimers, this mechanism is able to form extended chains. The agents for chain growth are CH3Cl molecules that dissociatively attach CH3 groups and chlorine atoms to silicon atoms from different dimers. By means of charge transfer through the surface, this gives rise to dangling bonds adjacent to the CH3 groups and chlorine atoms (in effect, 'free radicals') that dissociate further incoming CH3Cl molecules, thereby providing the growing points for chains of indefinite length. Using the complex variable method and a new conformal mapping, the fracture behavior of multiple cracks emanating from a circular hole in piezoelectric materials is considered under remotely uniform in-plane electric and anti-plane mechanical loadings. The analytic solutions of the field intensity factors and the energy release rate are presented by taking the effect of dielectric permittivity into consideration. Known results can be derived as special cases from the general solutions. The results are illustrated with plots, showing that the piezoelectric material containing three radial cracks spaced equally at 120° apart originating from a circular hole is the easiest to fail for all cases of cracks originating from a circular hole under in-plane electric and anti-plane mechanical loadings. Moreover, if there exist multiple cracks ( n ≥ 3) originating from a circular hole in piezoelectric materials, an increase in the number of cracks can enhance the reliability of these materials.
