A resumption of climate warming in maritime Antarctica, arising from continued greenhouse gas emissions to the atmosphere, is predicted to lead to further expansions of plant populations across the region, with consequent increases in nutrient inputs to soils. Here, we test the main and interactive effects of warming, applied with open top chambers (OTCs), and nutrient amendment with tryptic soy broth (TSB), an artificial growth substrate, on bacterial community composition and diversity using Illumina sequencing of 16S rRNA genes in soil from a field experiment in the southern maritime Antarctic. Substantial effects of TSB application on bacterial communities were identified after 49 months, including reduced diversity, altered phylogenetic community assembly processes, increased Proteobacteria-to-Acidobacteria ratios and significant divergence in community composition, notably increases in the relative abundances of the gram-positive genera Arthrobacter, Paeniglutamicibacter and Planococcus. Contrary to previous observations from other maritime Antarctic field warming experiments, we recorded no effects of warming with OTCs, or interactive effects of OTCs and TSB application, on bacterial community composition or diversity. Based on these findings, we conclude that further warming of the maritime Antarctic is unlikely to influence soil bacterial community composition or diversity directly, but that increased nutrient inputs arising from enhanced plant growth across the region may affect the composition of soil bacterial communities, with possible effects on ecosystem productivity. Background and aims: Biochar addition to soil is a carbon capture and storage option with potential to mitigate rising atmospheric CO2 concentrations, yet the consequences for soil organisms and linked ecosystem processes are inconsistent or unknown. We tested biochar impact on soil biodiversity, ecosystem functions, and their interactions, in temperate agricultural soils. Methods: We performed a 27-month factorial experiment to determine effects of biochar, soil texture, and crop species treatments on microbial biomass (PFLA), soil invertebrate density, crop biomass and ecosystem CO2 flux in plant-soil mesocosms. Results: Overall soil microbial biomass, microarthropod abundance and crop biomass were unaffected by biochar, although there was an increase in fungal-bacterial ratio and a positive relationship between the 16:1ω5 fatty acid marker of AMF mass and collembolan density in the biochar-treated mesocosms. Ecosystem CO2 fluxes were unaffected by biochar, but soil carbon content of biochar-treated mesocosms was significantly lower, signifying a possible movement/loss of biochar or priming effect. Conclusions: Compared to soil texture and crop type, biochar had minimal impact on soil biota, crop production and carbon cycling. Future research should examine subtler effects of biochar on biotic regulation of ecosystem production and if the apparent robustness to biochar weakens over greater time spans or in combination with other ecological perturbations. Soils store about four times as much carbon as plant biomass, and soil microbial respiration releases about 60 petagrams of carbon per year to the atmosphere as carbon dioxide. Short-term experiments have shown that soil microbial respiration increases exponentially with temperature. This information has been incorporated into soil carbon and Earth-system models, which suggest that warming-induced increases in carbon dioxide release from soils represent an important positive feedback loop that could influence twenty-first-century climate change. The magnitude of this feedback remains uncertain, however, not least because the response of soil microbial communities to changing temperatures has the potential to either decrease or increase warming-induced carbon losses substantially. Here we collect soils from different ecosystems along a climate gradient from the Arctic to the Amazon and investigate how microbial community-level responses control the temperature sensitivity of soil respiration. We find that the microbial community-level response more often enhances than reduces the mid- to long-term (90 days) temperature sensitivity of respiration. Furthermore, the strongest enhancing responses were observed in soils with high carbon-to-nitrogen ratios and in soils from cold climatic regions. After 90 days, microbial community responses increased the temperature sensitivity of respiration in high-latitude soils by a factor of 1.4 compared to the instantaneous temperature response. This suggests that the substantial carbon stores in Arctic and boreal soils could be more vulnerable to climate warming than currently predicted. [ABSTRACT FROM AUTHOR]

We have measured total soil organic carbon (SOC), dissolved organic carbon (DOC), and microbial lipid contents (as indices of microbial biomass and community structure), and their distributions to 60 cm depth in soils from replicated medium-term (2003-2008) experimental arable plots subject to different tillage regimes in Scotland. The treatments were zero tillage (ZT), minimum tillage (MT; cultivation to 7 cm), the conventional tillage (CT) practice of ploughing to 20 cm, and deep ploughing (DP) to 40 cm depth. In the 0-30 cm depth range, SOC content (corrected for bulk density differences between tillage treatments) was greatest under ZT and MT, but over 0-60 cm depth the SOC contents of these treatments were similar to the CT and DP treatments. DOC concentrations declined with increasing depth in ZT and MT above 20 cm, but there were no significant differences with depth in the CT and DP treatments. Beneath 20 cm, there was little change in DOC concentration with depth for all treatments, although for the MT treatment, there was less DOC beneath the depth of cultivation. The total microbial biomass decreased with increasing depth over the 0-60 cm range in the ZT and MT treatments, whereas it decreased with depth only below 30-40 cm in the CT and DP treatments. The microbial biomass was significantly different only between 0-5 cm in the ZT, CT and DP treatments, but not for other depths between all treatments. The bacterial biomass was greater in the ZT treatment than in MT, CT and DP near the soil surface, but not significantly different over the whole profile (0-60 cm). The fungal biomass decreased with depth in the ZT and MT treatments over the whole 0-60 cm depth range, whereas it decreased with depth only below 20 cm in the CT and DP treatments. The agronomic and pulping performance of transgenic trees with altered lignin has been evaluated in duplicated, long-term field trials. Poplars expressing cinnamyl alcohol dehydrogenase (CAD) or caffeate/5-hydroxyferulate O-methyltransferase (COMT) antisense transgenes were grown for four years at two sites, in France and England. The trees remained healthy throughout the trial. Growth indicators and interactions with insects were normal. No changes in soil microbial communities were detected beneath the transgenic trees. The expected modifications to lignin were maintained in the transgenics over four years, at both sites. Kraft pulping of tree trunks showed that the reduced-CAD lines had improved characteristics, allowing easier delignification, using smaller amounts of chemicals, while yielding more high-quality pulp. This work highlights the potential of engineering wood quality for more environmentally benign papermaking without interfering with tree growth or fitness. [ABSTRACT FROM AUTHOR]

Abiotic and biotic factors control ecosystem biodiversity, but their relative contributions remain unclear. The ultraoligotrophic ecosystem of the Antarctic Dry Valleys, a simple yet highly heterogeneous ecosystem, is a natural laboratory well-suited for resolving the abiotic and biotic controls of community structure. We undertook a multidisciplinary investigation to capture ecologically relevant biotic and abiotic attributes of more than 500 sites in the Dry Valleys, encompassing observed landscape heterogeneities across more than 200 km2. Using richness of autotrophic and heterotrophic taxa as a proxy for functional complexity, we linked measured variables in a parsimonious yet comprehensive structural equation model that explained significant variations in biological complexity and identified landscape-scale and fine-scale abiotic factors as the primary drivers of diversity. However, the inclusion of linkages among functional groups was essential for constructing the best-fitting model. Our findings support the notion that biotic interactions make crucial contributions even in an extremely simple ecosystem. A letter to the editor is presented in response to correspondence on biotechnological potential and the environmental risks of releasing transgenic trees that was published in the past issues.

The diversity and phylogenetic relationships of fungi obtained from Antarctic soils were analysed using molecular techniques. Direct extraction of soil community DNA from two locations, Fossil Bluff (FB) and Jane Col (JC), was supplemented with isolation studies. Nucleic acids from both the community DNA and the colony extracts were PCR amplified using primers specific for the 18S rRNA gene (18S rDNA). Amplicons were separated in denaturant gels (DGGE) or following endonuclease digestion (ARDRA). Clones presenting unique ARDRA banding patterns and unique DGGE bands were sequenced. Comparison of the experimental sequences from the different techniques employed with those held online resulted in the repeated recovery of a limited range of related organisms indicating low species diversity of microfungi in these soils. A total of 102 fungal sequences were obtained from FB (37 sequences) and JC (65 sequences) that together were distributed among the Basidiomycota (48 sequences), Ascomycota (48 sequences) and Zygomycota (6 sequences). Sequences of the latter were only recovered from the JC soils. Phylogenetic comparisons of the experimental sequences with those held online have shown high rRNA gene relatedness with those obtained from other, less extreme, environments. [ABSTRACT FROM AUTHOR]
