Background: Polycystic ovary syndrome (PCOS) is a gynecological endocrine disorder characterized by ovulatory disorders, hyperandrogenemia, and polycystic changes in the ovaries. FDX1 is a ferredoxin-reducing protein on human mitochondria that plays an important role in steroid anabolism. Liraglutide, a glucagon-like peptide-1 receptor agonist (GLP-1RA), has recently emerged as a potential therapeutic agent for PCOS. Recent studies have suggested that FDX1 may be associated with the development of PCOS. This study aims to explore the pivotal role of FDX1 in the amelioration of PCOS through liraglutide intervention. Materials and methods: A PCOS rat model was induced via subcutaneous DHEA injections. Following successful model establishment, the rats were treated with liraglutide combined with metformin, or with each drug individually, over a six-week period. After 6 weeks of treatment, we assessed changes in body weight, fasting blood glucose, sex hormone levels, estrous cycle regularity, ovarian morphology, FDX1 expression in ovarian tissue, and ovarian ROS levels. Results: PCOS rats exhibited significant increases in body weight and fasting blood glucose levels, disrupted estrous cycles, and polycystic ovarian morphology. FDX1 expression was notably reduced in the ovarian tissues of PCOS rats. Treatment with liraglutide, both alone and in combination with metformin, led to improvements in body weight, fasting blood glucose, sex hormone balance, estrous cycle regularity, ovarian morphology, and ovarian ROS levels. Notably, FDX1 expression was significantly restored in all treatment groups, with the most substantial increase observed in the liraglutide-treated group. Conclusion: This study suggests that FDX1 could serve as a potential biomarker for elucidating the underlying mechanisms of liraglutide's therapeutic effects in PCOS management. Thermoelastic damping (TED) is a fundamental dissipation mechanism that inevitably exists in high-performance micro/nano-electro-mechanical systems. In this paper, the damping characteristics in semiconductor microbeam are studied under coupled carrier-thermo-elastic theory. Using conventional LR model and considering the effect of carrier, the governing equations for TED in semiconductor beam were obtained and the analytical solution for temperature and carrier density was obtained using complex frequency method. The expression for inverse quality factor was given by solving the vibration equation. The effects of cantilever beam size, material parameters, temperature, and doping concentration on TED were studied, and the results showed that the carrier has significant influence on the dissipation performance of semiconductor microstructures. Background: Branched-chain amino acids (BCAAs), including leucine, isoleucine, and valine, are essential amino acids for mammals. Maternal BCAAs during pregnancy have been associated with newborn development. Meanwhile, BCAAs have been tightly linked with insulin resistance and diabetes in recent years. Diabetes in pregnancy is a common metabolic disorder. The current study aims to assess the circulating BCAA levels in pregnant women with diabetes and their relationship with neonatal development. Methods: The serum concentrations of BCAAs and their corresponding branched-chain α-keto acids (BCKAs) catabolites in 33 pregnant women with normal glucose tolerance, 16 pregnant women with type 2 diabetes before pregnancy (PDGM), and 15 pregnant women with gestational diabetes mellitus (GDM) were determined using a liquid chromatography system coupled to a mass spectrometer. The data were tested for normal distribution and homogeneity of variance before statistical analysis. Correlations were computed with the Pearson correlation coefficient. Results: The maternal serum BCAAs and BCKAs levels during late pregnancy were higher in women with PGDM than those in healthy women. Meanwhile, the circulating BCAAs and BCKAs showed no significant changes in women with GDM compared with those in healthy pregnant women. Furthermore, the circulating BCAA and BCKA levels in women with PGDM were positively correlated with the weight of the newborn. The circulating leucine level in women with GDM was positively correlated with the weight of the newborn. BCAA and BCKA levels in healthy pregnant women showed no correlation with newborn weight. Conclusions: The serum BCAAs in pregnant women with diabetes, which was elevated in PGDM but not GDM, were positively correlated with newborn weight. Female germline cells in flowering plants differentiate from somatic cells to produce specialized reproductive organs, called ovules, embedded deep inside the flowers. We investigated the molecular basis of this distinctive developmental program by performing single-cell RNA sequencing (scRNA-seq) of 16,872 single cells of Arabidopsis thaliana ovule primordia at three developmental time points during female germline differentiation. This allowed us to identify the characteristic expression patterns of the main cell types, including the female germline and its surrounding nucellus. We then reconstructed the continuous trajectory of female germline differentiation and observed dynamic waves of gene expression along the developmental trajectory. A focused analysis revealed transcriptional cascades and identified key transcriptional factors that showed distinct expression patterns along the germline differentiation trajectory. Our study provides a valuable reference dataset of the transcriptional process during female germline differentiation at single-cell resolution, shedding light on the mechanisms underlying germline cell fate determination. Quantitative real-time PCR (qRT-PCR) is a convenient tool for gene expression analysis. However, reliability of its result is influenced largely by the selection of reference genes. Pineapple molecular breeding is limited due to lack of understanding of sexual organs' development. Similarly, only a few reference genes have identified for qRT-PCR analysis in pineapple stamen and ovule. In this study, we initially identified 20 candidate reference genes using transcriptome data, and determined their expression stabilities in 36 ovule and stamen developmental samples using qRT-PCR. Three algorithms, GeNorm, NormFinder, and BestKeeper were used to determine the superiority of those candidate genes. Our results revealed that the use of combined RPS4 and RPL23 during ovule development, CCR and RPS4 during stamen development were sufficient for reliable normalization. These recommended reference genes were further verified by evaluating the temporal expression abundance of the meiosis-specific protein-encoding genes AcASY1 and AcASY3 in all experimental samples. Our results complement previous pineapple normalization study by providing appropriate reference genes during the entire reproductive period, and will beneficial for future studies on the pineapple stamen and ovule development. Nitrate is the preferred nitrogen source of higher plants and an essential nutrient for plant growth and development. Nitrate transporters (NRTs) play vital roles in the nitrate uptake and transportation. However, the NRT gene family in pineapple is still unexplored. In this study, we performed a genome-wide analysis of the pineapple genome and identified 48 NRT genes (AcNRTs) distributed unevenly across 9 chromosomes and 2 scaffolds. Phylogenetic analysis showed that these genes can be divided into three groups, namely, AcNRT1/PTR, AcNRT2 and AcNRT3/NAR1 with 44, 3 and 1 members, respectively. AcNRTs within the same phylogenetic group share similar gene structure and domain composition. In addition, syntenic and phylogenetic analyses identified 34 Arabidopsis NRT genes with 31 pineapple NRT genes as orthologs. By investigating the expression profiles of these genes in various tissues, we showed that the expression pattern of some AcNRTs genes is tissue-specific. Furthermore, we examined the expression of the AcNRT2s under nitrate starvation and found that AcNRT2.1 and AcNRT2.2 both have the strongest response in roots suggesting that AcNRTs may play a broad role in the pineapple in response to nitrate deficiency. Metastatic outcomes depend on the interactions of metastatic cells with a specific organ microenvironment. Our previous studies have shown that triple-negative breast cancer (TNBC) MDA-MB-231 cells passaged in astrocyte-conditioned medium (ACM) show proclivity to form brain metastases, but the underlying mechanism is unknown. The combination of microarray analysis, qPCR, and ELISA assay were carried out to demonstrate the ACM-induced expression of angiopoietin-like 4 (ANGPTL4) in TNBC cells. A stable ANGPTL4-knockdown MDA-MB-231 cell line was generated by ANGPTL4 short-hairpin RNA (shRNA) and inoculated into mice via left ventricular injection to evaluate the role of ANGPTL4 in brain metastasis formation. The approaches of siRNA, neutralizing antibodies, inhibitors, and immunoprecipitation were used to demonstrate the involved signaling molecules. We first found that ACM-conditioned TNBC cells upregulated the expression of ANGPTL4, a secreted glycoprotein whose effect on tumor progression is known to be tumor microenvironment- and tumor-type dependent. Knockdown of ANGPTL4 in TNBC MDA-MB-231 cells with shRNA decreased ACM-induced tumor cell metastatic growth in the brain and attributed to survival in a mouse model. Furthermore, we identified that astrocytes produced transforming growth factor-beta 2 (TGF-β2), which in part is responsible for upregulation of ANGPTL4 expression in TNBC through induction of SMAD signaling. Moreover, we identified that tumor cells communicate with astrocytes, where tumor cell-derived interleukin-1 beta (IL-1β) and tumor necrosis factor alpha (TNF-α) increased the expression of TGF-β2 in astrocytes. Collectively, these findings indicate that the invading TNBC cells interact with astrocytes in the brain microenvironment that facilitates brain metastases of TNBC cells through a TGF-β2/ANGPTL4 axis. The migration and distribution of contaminants in heterogeneous aquifers are mostly investigated through numerical simulations. They are rarely examined through experimental simulations because of sampling difficulties. In this study, we used the light transmission method (LTM) to explore the migration and distribution of hexavalent chromium [Cr(VI)] in homogeneous sandy aquifers with or without a low-permeability bottom and lens (heterogeneous aquifer). Experimental phenomena were observed via images obtained with a high-power light source. The migration distances of pollution plume were also monitored. Under heterogeneous conditions, low-permeability aquifer materials considerably inhibited the migration of Cr(VI) and altered the Cr(VI) migration rate of horizontal and vertical plumes spread to a certain extent. Common horizontal and vertical migration curve equations in all three conditions were also obtained through distance-time curve fitting. The difference between high- and low-concentration zones was accurately revealed, and the experimental phenomenon was further explained.
