Visitors to Colter Bay Village in Grand Teton National Park were surveyed to elicit their evaluations of experimental outdoor lighting conditions. Luminaires capable of dimming and switching between two LED modules (white, blended red-white) were installed in street and parking areas. The blended red-white lamps consisted of 30 narrowband LED with a peak wavelength 623 nm and two 3000 K white LEDs. Similar “red” lamps were previously shown to reduce impacts to bats and insects. The white and red lamps were closely matched for luminance. Measured horizontal illuminance at survey locations had an interquartile range from 0.63 to 3.82 lx. The red lamps produced lower perceived brightness (VB2(λ)), even after reflection off asphalt, yet survey participants expressed higher ratings for visual comfort and safety under red lighting. Surveys conducted earlier in the evening, with higher levels of predicted solar and measured horizontal illuminance, rated higher on visual comfort and safety, though these correlations were not as strong as the effect of lamp color. Streetlight ratings and support for lighting that protected natural resources were not contingent upon age or gender. Survey participants assessed red lighting as more protective of the environment. Intentional actions exhibit control in a way that mere lucky successes do not. A longstanding tradition in action theory characterizes actional control in terms of the knowledge with which one acts when acting intentionally. Given that action theorists, no less than epistemologists, typically take for granted the orthodox thesis that knowledge is in the head (viz., realized exclusively by brainbound cognition), the idea that intentional action is controlled in virtue of knowledge is tantamount to the idea that the knowledge by which intentional actions exhibit control supervenes intracranially. We raise some challenges for this idea, and in doing show, we show how epistemic theories of actional control are naturally aligned moreso than has been appreciated with cognitive extension in the theory of mind. In our changing world, understanding plant community responses to global change drivers is critical for predicting future ecosystem composition and function. Plant functional traits promise to be a key predictive tool for many ecosystems, including grasslands; however, their use requires both complete plant community and functional trait data. Yet, representation of these data in global databases is sparse, particularly beyond a handful of most used traits and common species. Here we present the CoRRE Trait Data, spanning 17 traits (9 categorical, 8 continuous) anticipated to predict species' responses to global change for 4,079 vascular plant species across 173 plant families present in 390 grassland experiments from around the world. The dataset contains complete categorical trait records for all 4,079 plant species obtained from a comprehensive literature search, as well as nearly complete coverage (99.97%) of imputed continuous trait values for a subset of 2,927 plant species. A common objection to Sosa’s epistemology is that it countenances, in an objectionable way, unsafe knowledge. This objection, under closer inspection, turns out to be in far worse shape than Sosa’s critics have realised. Sosa and his defenders have offered two central response types to the idea that allowing unsafe knowledge is problematic: one response type adverts to the animal/reflective knowledge distinction that is characteristic of bi-level virtue epistemology. The other less-discussed response type appeals to the threat of dream scepticism, and in particular, to the idea that many of our everyday perceptual beliefs are unsafe through the nearness of the dream possibility. The latter dreaming response to the safety objection to Sosa’s virtue epistemology has largely flown under the radar in contemporary discussions of safety and knowledge. We think that, suitably articulated in view of research in the philosophy and science of dreaming, it has much more going for it than has been appreciated. This paper further develops, beyond what Sosa does himself, the dreaming argument in response to those who think safety (as traditionally understood) is a condition on knowledge and who object to Sosa’s account on the grounds that it fails this condition. The payoffs of further developing this argument will be not only a better understanding of the importance of insights about dreaming against safety as a condition on knowledge, but also some reason to think a weaker safety condition, one that is relativised to SSS (i.e., skill/shape/situation) conditions for competence exercise, gets better results all things considered as an anti-luck codicil on knowledge. Social difficulties during interactions with others are central to autism spectrum disorder (ASD). Understanding the links between these social difficulties and their underlying neural processes is a primary aim focused on improved diagnosis and treatment. In keeping with this goal, we have developed a multivariate classification method based on neural data acquired by functional near infrared spectroscopy, fNIRS, during live eye-to-eye contact with adults who were either typically developed (TD) or individuals with ASD. The ASD diagnosis was based on the gold-standard Autism Diagnostic Observation Schedule (ADOS) which also provides an index of symptom severity. Using a nested cross-validation method, a support vector machine (SVM) was trained to discriminate between ASD and TD groups based on the neural responses during eye-to-eye contact. ADOS scores were not applied in the classification training. To test the hypothesis that SVM identifies neural activity patterns related to one of the neural mechanisms underlying the behavioral symptoms of ASD, we determined the correlation coefficient between the SVM scores and the individual ADOS scores. Consistent with the hypothesis, the correlation between observed and predicted ADOS scores was 0.72 (p < 0.002). Findings suggest that multivariate classification methods combined with the live interaction paradigm of eye-to-eye contact provide a promising approach to link neural processes and social difficulties in individuals with ASD. The expansion of many wetland species is a function of both clonal propagation and sexual reproduction. The production of ramets through clonal propagation enables plants to move and occupy space near parent ramets, while seeds produced by sexual reproduction enable species to disperse and colonize open or disturbed sites both near and far from parents. The balance between clonal propagation and sexual reproduction is known to vary with plant density but few studies have focused on reproductive allocation with density changes in response to global climate change. Schoenoplectus americanus is a widespread clonal wetland species in North America and a dominant species in Chesapeake Bay brackish tidal wetlands. Long-term experiments on responses of S. americanus to global change provided the opportunity to compare the two modes of propagation under different treatments. Seed production increased with increasing shoot density, supporting the hypothesis that factors causing increased clonal reproduction (e.g., higher shoot density) stimulate sexual reproduction and dispersal of genets. The increase in allocation to sexual reproduction was mainly the result of an increase in the number of ramets that flowered and not an increase in the number of seeds per reproductive shoot, or the ratio between the number of flowers produced per inflorescence and the number of flowers that developed into seeds. Seed production increased in response to increasing temperatures and decreased or did not change in response to increased CO2 or nitrogen. Results from this comparative study demonstrate that plant responses to global change treatments affect resource allocation and can alter the ability of species to produce seeds. Low temperature stress is the primary factor determining the latitudinal limits of tropical plants. As the climate warms, tropical species are migrating poleward, displacing native species and modifying ecosystem structure and function. Changes are particularly evident along latitudinal gradients with the highest velocity of change occurring in wetlands. In coastal wetlands, saltmarshes dominate at latitudes above 30°, whereas mangroves occur mostly in the tropics because most species are intolerant of freezing temperatures, but others, like Avicennia germinans (black mangrove), do tolerate freezing temperatures. In response to a warmer climate and fewer killing freezes, mangroves are currently expanding into saltmarshes. However, the speed of the transition from saltmarsh to mangrove can also be modified by extreme events and nutrient subsidies. In a fertilization experiment along the Atlantic coast of North America, we found that nitrogen addition altered plant traits in Avicennia, which increased their resistance to freezing temperatures. This trait shift resulted in negligible freeze effects during a January 2018 extreme freeze event compared to unfertilized plants, which lost more than 80% of the leaves and more than 40% of the wood in their canopies. The freeze-killed litter from unfertilized plants provided a nutrient pulse that influenced recovery, growth and mangrove cover for three years following the freeze. Nutrient enrichment and recovery from the freeze effects led to increased growth and structural complexity of the mangrove canopy, which further enhanced freeze tolerance, shrub growth form and the ability of Avicennia to displace the saltmarsh in the temperate–tropical ecotone. Intentional action is, in some sense, non-accidental, and one common way action theorists have attempted to explain this is with reference to control. The idea, in short, is that intentional action implicates control, and control precludes accidentality. But in virtue of what, exactly, would exercising control over an action suffice to make it non-accidental in whatever sense is required for the action to be intentional? One interesting and prima facie plausible idea that we wish to explore in this paper is that control is non-accidental in virtue of requiring knowledge—either knowledge-that or knowledge-how (e.g., Beddor and Pavese 2021; cf., Setiya 2008; 2012 and Habgood-Coote 2018). We review in detail some key recent work defending such knowledge-centric theories of control, and we show that none of these accounts holds water. Microbes carry out many critical biogeochemical transformations in the biosphere such as greenhouse gas production and consumption. Characterizing how microbial communities vary through space and time may hold insights into understanding ecosystem function, particularly as it responds to ongoing climate change. However, it remains unclear to what extent variability in the composition of microbial communities exerts long-term control over biogeochemistry independently of the physiochemical and macroorganismal context in which microbes reside. In this synthesis, we reviewed literature about the versatility and adaptability of microbial communities, and analogous research in medicine, agriculture, and bioremediation. We synthesized data from microbial diversity experiments to determine thresholds at which loss of microbial richness impairs function and compared it to actual microbial richness in nature. The evidence suggests that, in environments such as surface soils, sediments, rivers, lakes, oceans, and the atmosphere, which are open to microbial inoculum and which dominate biogeochemical processes in the biosphere, microbial function equilibrates to environmental conditions over a much shorter interval (days to years) than the time scale on which anthropogenic climate change influences ecosystems (decades to centuries). We conclude that the degree of microbial control over ecosystem processes has been overstated because the correlation of taxonomic information with ecosystem function has obscured the understanding of causality in natural ecosystems. We recommend experiments in which microbial communities are manipulated in ways that allow us to disentangle the influence of microbial community structure from confounding environmental covariates. Tidal marsh plant species commonly zonate along environmental gradients such as elevation, but it is not always clear to what extent plant distribution is driven by abiotic factors vs. biotic interactions. Yet, the distinction has importance for how plant communities will respond to future change such as higher sea level, particularly given the distinct flooding tolerances and contributions to elevation gain of different species. We used observations from a 33-year experiment to determine co-occurrence patterns for the sedge, Schoenoplectus americanus, and two C4 grasses, Spartina patens and Distichlis spicata, to infer functional group interactions. Then, we conducted a functional group removal experiment to directly assess the interaction between sedge and grasses throughout the range in which they cooccur. The observational record suggested negative interactions between sedge and grasses across sedge- and grass-dominated plots, though the relationship weakened in years with greater flooding stress. The removal experiment revealed mutual release effects, indicating competition was the predominant interaction, and here, too, competition tended to weaken, though nonsignificantly, in more flooded, lower elevation zones. Whereas zonation patterns in undisturbed portions of marsh suggest that the sedge will dominate this marsh as flooding stress increases with sea level rise, we propose that grasses may exhibit a competition release effect and contribute to biomass and elevation gain even in sedge-dominated communities as sea level continues to rise. Even as abiotic stresses drive changes in the relative contributions of sedges and grasses, competition among them moderates fluctuations in total plant biomass production through time. Structural analogies connect Williamson’s (Knowledge and its limits, Oxford University Press, Oxford, 2000; Acting on knowledge. In: Carter JA, Gordon EC, Jarvis B (eds) Knowledge first: approaches in epistemology and mind, Oxford University Press, Oxford, pp 163–181, 2017) epistemology and action theory: for example, action is the direction-of-fit mirror image of knowledge, and knowledge stands to belief as action stands to intention. These structural analogies, for Williamson, are meant to illuminate more generally how ‘mirrors’ reversing direction of fit should be understood as connecting the spectrum of our cognitive and practically oriented mental states. This paper has two central aims, one negative and the other positive. The negative aim is to highlight some intractable problems with Williamson’s preferred analogical picture, which links the cognitive and the practical through the nexus of direction-of-fit mirroring. The positive aim of the paper is to propose a better alternative. In particular, we show that an achievement-theoretic proposal captures what is in common across the range of attitudes that exhibit the kind of structure that knowledge-belief, action-desire/intention do, while at the same time avoiding the problems shown to face Williamson’s proposed picture. Moreover, we draw attention to several key theoretical benefits of embracing our proposed achievement-theoretic picture, including some of the key benefits of the knowledge-first programme that Williamson’s own analogies were designed to secure. YouTube has been implicated in the transformation of users into extremists and conspiracy theorists. The alleged mechanism for this radicalizing process is YouTube's recommender system, which is optimized to amplify and promote clips that users are likely to watch through to the end. YouTube optimizes for watch-through for economic reasons: people who watch a video through to the end are likely to then watch the next recommended video as well, which means that more advertisements can be served to them. This is a seemingly innocuous design choice, but it has a troubling side-effect. Critics of YouTube have alleged that the recommender system tends to recommend extremist content and conspiracy theories, as such videos are especially likely to capture and keep users' attention. To date, the problem of radicalization via the YouTube recommender system has been a matter of speculation. The current study represents the first systematic, pre-registered attempt to establish whether and to what extent the recommender system tends to promote such content. We begin by contextualizing our study in the framework of technological seduction. Next, we explain our methodology. After that, we present our results, which are consistent with the radicalization hypothesis. One notable line of argument for epistemic relativism appeals to considerations to do with non-neutrality: in certain dialectical contexts—take for instance the famous dispute between Galileo and Cardinal Bellarmine concerning geocentrism—it seems as though a lack of suitably neutral epistemic standards that either side could appeal to in order to (non-question-beggingly) resolve their first-order dispute is itself—as Rorty (Philosophy and the mirror of nature, Princeton University Press, Princeton, 1979) influentially thought—evidence for epistemic relativism. In this essay, my aim is first to present a more charitable reformulation of this line of reasoning, one that is framed not merely in terms of the availability of epistemic norms that are suitably neutral between interlocutors, but in terms of the availability of what I call Archimedean metanorms. Once this more charitable line of argument is developed, I show how, even though it avoids problems that face 'non-neutrality' versions of the argument, it nonetheless runs into various other problems that appear ultimately intractable and, further, that the strategy in question gives us no decisive reason to draw the relativist's conclusion rather than the Pyrrhonian sceptic's. A vexing problem in contemporary epistemology—one with origins in Plato's Meno—concerns the value of knowledge, and in particular, whether and how the value of knowledge exceeds the value of mere (unknown) true opinion. The recent literature is deeply divided on the matter of how best to address the problem. One point, however, remains unquestioned: that if a solution is to be found, it will be at the personal level, the level at which states of subjects or agents, as such, appear. We take exception to this orthodoxy, or at least to its unquestioned status. We argue that subpersonal states play a significant—arguably, primary—role in much epistemically relevant cognition and thus constitute a domain in which we might reasonably expect to locate the "missing source" of epistemic value, beyond the value attached to mere true belief. This paper is about two topics: metaepistemological absolutism and the epistemic principles governing perceptual warrant. Our aim is to highlight—by taking the debate between dogmatists and conservativists about perceptual warrant as a case study—a surprising and hitherto unnoticed problem with metaepistemological absolutism, at least as it has been influentially defended by Paul Boghossian (Fear of knowledge: against relativism and constructivism, Oxford University Press, Oxford, 2006a) as the principal metaepistemological contrast point to relativism. What we find is that the metaepistemological commitments at play on both sides of this dogmatism/conservativism debate do not line up with epistemic relativism nor do they line up with absolutism, at least as Boghossian articulates this position. What this case study reveals is the need in metaepistemological option space for the recognition of a weaker and less tendentious form of absolutism, what we call "environment relativism". On this view, epistemic principles are knowable, objective, and they can serve as the basis of particular epistemic evaluations, but their validity is relative to the wider global environment in which they are applied. Moore and Russell thought that perceptual knowledge of the external world is based on abductive inference from information about our experience. Sosa maintains that this ‘indirect realist’ strategy has no prospects of working. Vogel disagrees and thinks it can and does work perfectly well, and his reasoning (and variations on that reasoning) seem initially promising, moreso than other approaches. My aim, however, will be to adjudicate this dispute in favor of Sosa’s pessimistic answer, and in doing so, to better uncover the important role abductive inference does have in a wider theory of perceptual knowledge, even if it doesn’t feature in any promising vindication of (anti-skeptical) indirect realism. According to one prominent view of exercising abilities (e.g., Millar in Acta Anal 24(4):223–236, 2009), a subject, S, counts as exercising an ability to ϕ if and only if S successfully ϕ s. Such an 'exercise-success' thesis looks initially very plausible for abilities, perhaps even obviously or analytically true. In this paper, however, I will be defending the position that one can in fact exercise an ability to do one thing by doing some entirely distinct thing, and in doing so I'll highlight various reasons (epistemological, metaphysical and linguistic) that favor the alternative approach I develop over views that hold that the exercise of an ability is a success notion in the sense Millar maintains. In Chapter 3 of Judgment and Agency, Sosa (Judgment and Agency, 2015) explicates the concept of a fully apt performance. In the course of doing so, he draws from illustrative examples of practical performances and applies lessons drawn to the case of cognitive performances, and in particular, to the cognitive performance of judging. Sosa's examples in the practical sphere are rich and instructive. But there is, I will argue, an interesting disanalogy between the practical and cognitive examples he relies on. Ultimately, I think the source of the disanalogy is a problematic picture of the cognitive performance of guessing and its connection to knowledge and defeat. Once this critical line of argument is advanced, an alternative picture of guessing, qua cognitive performance, is articulated, one which avoids the problems discussed, and yet remains compatible with Sosa's broader framework. Blue carbon (C) ecosystems are among the most effective C sinks of the biosphere, but methane (CH4) emissions can offset their climate cooling effect. Drivers of CH4 emissions from blue C ecosystems and effects of global change are poorly understood. Here we test for the effects of sea level rise (SLR) and its interactions with elevated atmospheric CO2, eutrophication, and plant community composition on CH4 emissions from an estuarine tidal wetland. Changes in CH4 emissions with SLR are primarily mediated by shifts in plant community composition and associated plant traits that determine both the direction and magnitude of SLR effects on CH4 emissions. We furthermore show strong stimulation of CH4 emissions by elevated atmospheric CO2, whereas effects of eutrophication are not significant. Overall, our findings demonstrate a high sensitivity of CH4 emissions to global change with important implications for modeling greenhouse-gas dynamics of blue C ecosystems. Coastal systems have enormous carbon-sequestering potential, but any positive climate effects can be countered by methane emissions. A bi-level account of trust is developed and defended, one with relevance in ethics as well as epistemology. The proposed account of trust—on which trusting is modelled within a virtue-theoretic framework as a performance-type with an aim—distinguishes between two distinct levels of trust, apt and convictive, that take us beyond previous assessments of its nature, value, and relationship to risk assessment. While Sosa (A virtue epistemology: apt belief and reflective knowledge, volume I, Oxford University Press, Oxford, 2009; Judgment and agency, Oxford University Press, Oxford, 2015; Epistemology, University Press, Princeton, 2017), in particular, has shown how a performance normativity model may be fruitfully applied to belief, my objective is to apply this kind of model in a novel and principled way to trust. Intellectual autonomy has long been identified as an epistemic virtue, one that has been championed influentially by (among others) Kant, Hume and Emerson. Manifesting intellectual autonomy, at least, in a virtuous way, does not require that we form our beliefs in cognitive isolation. Rather, as Roberts and Wood (Intellectual virtues: an essay in regulative epistemology, OUP Oxford, Oxford, pp. 259–260, 2007) note, intellectually virtuous autonomy involves reliance and outsourcing (e.g., on other individuals, technology, medicine, etc.) to an appropriate extent, while at the same time maintaining intellectual self-direction. In this essay, I want to investigate the ramifications for intellectual autonomy of a particular kind of epistemic dependence: cognitive enhancement. Cognitive enhancements (as opposed to therapeutic cognitive improvements) involve the use of technology and medicine to improve cognitive capacities in healthy individuals, through mechanisms ranging from smart drugs to brain-computer interfaces. With reference to case studies in bioethics, as well as the philosophy of mind and cognitive science, it is shown that epistemic dependence, in this extreme form, poses a prima facie threat to the retention of intellectual autonomy, specifically, by threatening to undermine our intellectual self-direction. My aim will be to show why certain kinds of cognitive enhancements are subject to this objection from self-direction, while others are not. Once this is established, we'll see that even some extreme kinds of cognitive enhancement might be not merely compatible with, but constitutive of, virtuous intellectual autonomy. The relationship between knowledge, virtue and action is a topic of central interest in mainstream epistemology, and increasingly so over the past decade.[1] These topics have also occupied an important place in Eastern thought, though Eastern and Western scholarship on this topic - and, more generally, scholarship in epistemology - has traditionally occurred in isolation from one another. According to Mi and Ryan, skilful reflection is best understood as a "master virtue" in the sense that skilful reflection shapes and corrects the other epistemic and intellectual virtues. In "Situationism, Virtue Epistemology, and Self-Determination Theory", Iizuka ([20]) brings a new twist to the "epistemic situationist" critique of virtue epistemology (e.g., Alfano [1]). Iizuka's key move is to resist this empirically-driven challenge to virtue epistemology, by drawing attention to additional empirical results that indicate that virtuous motivations are causally efficacious in ways that favour the virtue epistemologist over the situationist. Between the land and ocean, diverse coastal ecosystems transform, store, and transport material. Across these interfaces, the dynamic exchange of energy and matter is driven by hydrological and hydrodynamic processes such as river and groundwater discharge, tides, waves, and storms. These dynamics regulate ecosystem functions and Earth’s climate, yet global models lack representation of coastal processes and related feedbacks, impeding their predictions of coastal and global responses to change. Here, we assess existing coastal monitoring networks and regional models, existing challenges in these efforts, and recommend a path towards development of global models that more robustly reflect the coastal interface. A novel solution is offered for how emotional experiences can function as sources of immediate prima facie justification for evaluative beliefs, and in such a way that suffices to halt a justificatory regress. Key to this solution is the recognition of two distinct kinds of emotional skill (what I call generative emotional skill and doxastic emotional skill) and how these must be working in tandem when emotional experience plays such a justificatory role. The paper has two main parts, the first negative and the second positive. The negative part criticises the epistemic credentials of Epistemic Perceptualism (e.g., Tappolet, in: Clotilde (ed) Perceptual illusions. The second part of the paper develops an account of emotional skill and uses this account to frame a revisionary form of Epistemic Perceptualism that succeeds where the traditional views could not. During litter decomposition, three major fates of litter carbon (C) are possible: emission as carbon dioxide (CO2) into the atmosphere, leaching of dissolved organic carbon (DOC), and translocation and transformation into soil organic carbon (SOC). Soil moisture, one of the key drivers of litter decomposition, is predicted to change in the future due to shifts in precipitation patterns. We explored the effects of low, medium and high rainfall intensities on the partitioning of litter carbon fates in a 6-month long laboratory experiment. We tracked carbon in 13C-labeled tulip poplar litter in a laboratory mesocosms by measuring respiration rates, dissolved organic carbon in the leachate, and soil organic carbon at the end of the experiment. Mesocosms with the same three rainfall intensities but without leaf litter were also set up. Leaching of labile carbon caused priming, but the effect was stronger in the low intensity treatment. Transport of litter-derived carbon also differed: in high intensity treatment there was more total carbon in the surface soil and more litter-derived carbon in in the deep soil layers. The cumulative CO2 efflux was not significantly different. Our results highlight that extreme rainfall events, as projected by most climate models, may lead to altered carbon cycling in temperate forest soils. [ABSTRACT FROM AUTHOR]

In a series of works Sosa (in: Knowledge in perspective, Cambridge University Press, Cambridge, 1991; A virtue epistemology: apt belief and reflective knowledge, Oxford University Press, Oxford, 2007; Reflective knowledge: apt belief and reflective knowledge, Oxford University Press, Oxford, 2009; 'How Competence Matters in Epistemology', Philos Perspect 24(1):465–475, 2010; Knowing full well, Princeton University Press, Princeton, 2011; Judgment and agency, Oxford University Press, Oxford, 2015; Epistemology, Princeton University Press, Princeton, 2017) has defended the view that there are two kinds or 'grades' of knowledge, animal and reflective. One of the most persistent critics of Sosa's attempts to bifurcate knowledge is Kornblith (in: Greco (ed) Ernest sosa and his critics, Wiley, Hoboken, 2004; 'Sosa in Perspective', Philos Stud 144(1):127–136, 2009; On reflection, Oxford University Press, Oxford, 2012). Our aim in this paper is to outline and evaluate Kornblith's criticisms. We will argue that, while they raise a range of difficult (exegetical and substantive) questions about Sosa's 'bi-level' epistemology, Sosa has the resources to adequately respond to all of them. This article addresses a stated need within the food justice movement scholarship to increase the attention paid to the political socialization of hired farm hands in industrial agriculture. In Canada, tackling the problem of farm worker equity has particular social and political contours related to the Canadian horticultural industry's reliance on a state-managed migrant agricultural labour program designed to fill the sector's labour market demands. As Canada's Seasonal Agricultural Worker Program (SAWP) produces relations of 'unfree labour', engaging migrant farm workers in social movement initiatives can be particularly challenging. Critical educational interventions designed to encourage migrant farm workers' contribution to contemporary social movements in Canada must therefore confront the socio-cultural obstacles that constrict migrant farm workers' opportunities to participate as full members of their communities. In this article, I argue that social justice oriented approaches to community-based arts can provide a means for increasing the social movement contributions of farm workers employed through managed labour migration schema such as Canada's SAWP. Mcl-1 is a member of the Bcl-2 family of proteins that promotes cell survival by preventing induction of apoptosis in many cancers. High expression of Mcl-1 causes tumorigenesis and resistance to anticancer therapies highlighting the potential of Mcl-1 inhibitors as anticancer drugs. Here, we describe AZD5991, a rationally designed macrocyclic molecule with high selectivity and affinity for Mcl-1 currently in clinical development. Our studies demonstrate that AZD5991 binds directly to Mcl-1 and induces rapid apoptosis in cancer cells, most notably myeloma and acute myeloid leukemia, by activating the Bak-dependent mitochondrial apoptotic pathway. AZD5991 shows potent antitumor activity in vivo with complete tumor regression in several models of multiple myeloma and acute myeloid leukemia after a single tolerated dose as monotherapy or in combination with bortezomib or venetoclax. Based on these promising data, a Phase I clinical trial has been launched for evaluation of AZD5991 in patients with hematological malignancies (NCT03218683). [ABSTRACT FROM AUTHOR]

This article examines how the dormitory labour system as it is employed in the agricultural streams of Canada’s Temporary Foreign Worker Program (TFWP) affects workers’ everyday sociality. In the article, I demonstrate how the physical compression of home and work into a singular geographic site shapes workers’ identities and everyday relationships. Drawing on findings gathered from interviews with migrant farm workers from Mexico and Guatemala working in Southern Ontario, I explore how the requirement to warehouse temporary foreign workers directly on employer property collides with workers’ ability to establish an autonomous and dignified life in Canada. In particular, I demonstrate how the TFWP agricultural dormitory system produces inter-generational dynamics that intensify worker self-discipline and generates gender dynamics that support the development of a hyper-productive transnational workforce. An account of meta-epistemic defeaters—distinct from traditional (first-order) epistemic defeaters—is motivated and defended, drawing from case studies involving epistemic error-theory (e.g., Olson 2011, Reasons for belief; cf., Streumer 2012, J Philos 110:1-25) and epistemic relativism (e.g., MacFarlane 2005, Oxford Stud Epistemol 1:197-233; 2011; 2014, Assessment sensitivity: Relative truth and its applications). Purpose: Rest-pause (4-s unloaded rest between repetitions) training effects on one repetition maximum (1 RM), lifting volume, and neural activation via electromyography (EMG) are currently vague in the literature and can benefit strength and conditioning professionals for resistance training programme design. Therefore, this study compared 1 RM, neural activation via (EMG), and volume differences between rest-pause vs. traditional resistance training.Methods: Trained males (N = 20) were randomly assigned to either a rest-pause or a traditional training group. Pre- and post-1 RM testing was recorded. Training sessions were completed twice a week for 4 weeks and consisted of four sets of bench press to volitional fatigue at 80% of pre-test 1 RM with a 2-min rest between sets. Total volume completed was recorded on each training day. Neural activation of the pectoralis major was measured on the first and last training days.Results: A two-way repeated-measures ANOVA indicated both groups significantly increased their 1 RMs following the 4-week training protocol (p < .05). However, no significant differences were found in 1 RM and neural activation between the two groups (p > .05). An independent samples t test indicated that total volume lifted was significantly higher for the rest-pause group (56,778 vs. 38,315 lbs; p < .05) throughout the protocol and independently during weeks 2, 3, and 4.Conclusions: While strength and neural activation changes did not differ between groups, both increased 1 RMs and the rest-pause group achieved greater increases in volume than the traditional group. Purpose: Implantation of pre-endothelialized stents could enhance cellular recovery of a damaged vessel wall provided attached cells remain viable, functional and are present in sufficient numbers after deployment. The purpose of this study was to evaluate the feasibility of grooved stainless steel (SS) stents as a primary endothelial cell (EC) carrier with potentially enhanced EC protection upon stent deployment.Materials and Methods: Attachment and behavior of enzymatically harvested human adult venous ECs seeded onto gelatin-coated vascular stents were evaluated in an in vitro setting. Smooth and grooved SS stents and smooth nitinol stents were studied.Results: All cells expressed EC markers vWF and CD31. Using rotational seeding for a period of 16-24 h, ECs attached firmly to the stents with sufficient coverage to form a confluent EC monolayer. The grooved SS wire design was found to enable attachment of three times the number of cells compared to smooth wires. This also resulted in an increased number of cells remaining on the stent after deployment and after pulsatile flow of 180 ml/min for 24 h, which did not result in additional EC detachment.Conclusions: The grooved stent provides a potential percutaneous means to deliver sufficient numbers of viable and functional cells to a vessel segment during vascular intervention.
