Non-small cell lung cancer (NSCLC) is an aggressive and rapidly expanding lung cancer. Abnormal upregulation or knockdown of PDIA6 expression can predict poor prognosis in various cancers. This study aimed to investigate the biological function of PDIA6 in NSCLC. SOX2 and PDIA6 expression in NSCLC tissues and regulatory relationship between them were analyzed using bioinformatics. GSEA was performed on the enrichment pathway of PDIA6. qRT-PCR was utilized to examine expression of SOX2 and PDIA6 in NSCLC tissues and cells, and dual-luciferase reporter assay and ChIP experiments were performed to validate their regulatory relationship. CCK-8 experiment was conducted to assess cell viability, western blot was to examine levels of stem cell markers and proteins related to aerobic glycolysis pathway in cells. Cell sphere formation assay was used to evaluate efficiency of cell sphere formation. Reagent kits were used to measure glycolysis levels and glycolysis products. High expression of PDIA6 in NSCLC was linked to aerobic glycolysis. Knockdown of PDIA6 reduced cell viability, expression of stem cell surface markers, and cell sphere formation efficiency in NSCLC. Overexpression of PDIA6 could enhance cell viability and promote aerobic glycolysis, but the addition of 2-DG could reverse this result. Bioinformatics predicted the existence of upstream transcription factor SOX2 for PDIA6, and SOX2 was significantly upregulated in NSCLC, and they had a binding relationship. Further experiments revealed that PDIA6 overexpression restored repressive effect of knocking down SOX2 on aerobic glycolysis and cell stemness. This work revealed that the SOX2/PDIA6 axis mediated aerobic glycolysis to promote NSCLC cell stemness, providing new therapeutic strategies for NSCLC. [ABSTRACT FROM AUTHOR]

Purpose: Numerous studies had reported the diagnostic value of alkaline phosphatase (ALP) and its bone-specific isoforms (BAP) in the metastases of breast cancer (BC). The purpose of this meta-analysis was to summarize the diagnostic value of serum ALP and BAP in metastatic BC, especially focused on bone metastases. Methods: We searched comprehensively in the PubMed, Cochrane Library, and EMBASE for studies to explore the diagnostic accuracy of serum ALP/BAP level for metastatic BC. Qualities of including studies were assessed and pooled sensitivity, specificity, and summary receiver operating characteristic curve were calculated. Publication bias was assessed and meta-regression was conducted. Results: We finally included 25 studies with a total of 12,155 BC patients (1681 metastatic cases and 10,474 controls). According to the QUADAS-2 tool to assessment the methodological quality, most of the included studies were judged as high risk of patient selection bias. High serum levels of ALP/BAP in bone metastatic BC patients could be found compared with non-metastatic BC patients. The pooled sensitivity and specificity of ALP for BC bone metastases were 0.62 and 0.86, and the area under the curve (AUC) was 0.80. The pooled sensitivity and specificity of ALP for all site metastases (mainly bone and liver) were 0.56 and 0.91, and the AUC was 0.90. The pooled sensitivity and specificity of BAP for BC bone metastases were 0.66 and 0.92, and the AUC was 0.89. Conclusion: Although not promising, serum ALP and BAP could bring useful information for the early detection of BC metastases especially for the bone metastases. [ABSTRACT FROM AUTHOR]

To explore impact of Ataxia telangiectasia mutated (ATM) kinase on immunoreactions in lymphocyte-predominant breast cancer (LPBC), particularly its role in triple negative breast cancer (TNBC), 194 cases of LPBC were identified with pertinent clinical information retrieved. The expressions of ATM, activated ATM (P-ATM), Fas ligand (FASL), tumor necrosis factor-related apoptosis-induced ligand (TRAIL), major histocompatibility complex class I chain-related protein A (MICA), CD8, and Forkhead box P3 (FOXP3) were assessed by immunohistochemically. We found that ATM expressed on tumor cells was correlated with upregulated expression of P-ATM and MICA (P < 0.05), down-regulated expression of FASL and TRAIL (P < 0.01), and decreased Ki-67 tumor labeling (P < 0.05). However, within the TNBC group, only a negative correlation with FASL expression was found (P = 0.001). ATM and MICA expressions were significantly down -regulated in TNBC (P < 0.01) compared to non-TNBC, while TRAIL was significantly upregulated (P < 0.01). Tregs were increased in TNBC (P < 0.05), with CD8 + TILs decreased (P < 0.01). Ki-67 index was higher in TNBC than in non-TNBC (P < 0.01). ATM may play an important role in immunoreaction of LPBC, probably through upregulation of MICA and down-regulation of FASL and TRAIL. The down-regulated ATM expression in TNBC might be responsible for impaired tumor immunoactivity, rapid tumor growth, and aggressive clinical course. [ABSTRACT FROM AUTHOR]

Acidithiobacillus caldus plays an important role in commercial bioleaching. To understand how NaCl stress adaptation occurs in A. caldus, we grew A. caldus strain SM-1 in media containing high NaCl concentrations. SM-1 grew at concentrations of up to 1.0-mol L NaCl, but growth was severely inhibited at higher concentrations. Proteomic analysis showed that SM-1 used multiple strategies to respond to NaCl stress. In addition to several heat-shock proteins, enzymes involved in proline biosynthesis increased under NaCl stress. In addition, two DNA-binding proteins and a third protein of unknown function (Atc_1291), which was subsequently identified as a putative single-stranded DNA-binding protein, were up-regulated in the presence of NaCl stress. These DNA-binding proteins might play a role in response to osmotic stress. Atc_1291 was cloned and expressed in Escherichia coli. Surprisingly, we found that E. coli BL21/pET28a-atc_1291 grew to higher cell densities than E. coli BL21/pET28a, regardless of NaCl stress. Homologs to Atc_1291 were identified in several groups of Proteobacteria. The role of Atc_1291 in enhancing cell growth needs further investigation. [ABSTRACT FROM AUTHOR]

Purpose: The human gene PTPN11, which encodes the non-receptor protein tyrosine phosphatase of Src homology phosphotyrosine phosphatase 2 (Shp2), has been previously well interpreted as a proto-oncogene in a variety of malignancies. However, the tumor suppressor role of Shp2 has also been reported. The present study was conducted to investigate the role of Shp2 expression and its associated clinical manifestations in hepatocellular carcinoma (HCC). Methods: A tissue microarray of 333 pairs of HCC and self-matched adjacent non-tumor tissues was constructed, and the expression of Shp2 was determined by immunohistochemistry. The results were also conformed by Western blotting and quantitative PCR of 31 self-paired fresh HCC specimens. The associations of Shp2 expression with 25 clinicopathologic features were analyzed. Overall survival analysis and multivariate analysis were performed. Results: Significantly decreased Shp2 expression in tumor tissues (T) compared with adjacent non-tumor tissues (NT) could be detected, and the positive rate was 66.1 and 96.7%, respectively. We combined the T and NT Shp2 immunoreactivity by a variable of the decrease in Shp2 expression (ΔShp2) and divided cases into 2 groups: T < NT and T ≥ NT. Survival analysis showed both low Shp2 expression and T < NT group were significantly associated with short overall survival. Multivariate analysis showed ΔShp2 was an independent prognostic marker ( P = 0.033; HR: 0.527; 95% CI: 0.293-0.950). Conclusion: Shp2 is a tumor suppressor, and the decrease in Shp2 expression was a new prognostic marker in HCC. Hexavalent chromate reductase was characterized and was found to be localized in the cytoplasmic fraction of a chromium-resistant bacterium Pannonibacter phragmitetus LSSE-09. The Cr(VI) reductase activity of cell-free extract (S) was significantly improved by external electron donors, such as NADH, glucose, acetate, formate, citrate, pyruvate, and lactate. The reductase activity was optimal at pH 7.0 with NADH as the electron donor. The aerobic and anaerobic Cr(VI)-reduction enhanced by 0.1 mM NADH were respectively 3.5 and 3.4 times as high as that without adding NADH. The Cr(VI) reductase activity was inhibited by Mn, Cd, Fe, and Hg, whereas Cu enhanced the chromate reductase activity by 29% aerobically and 33% anaerobically. The aerobic and anaerobic specific Michaelis-Menten constant K of S fraction was estimated to be 64.95 and 47.65 μmol L, respectively. The soluble S fractions showed similar activity to S and could reduce 39.7% and 53.4% of Cr(VI) after 1 h of incubation aerobically and anaerobically while the periplasmic contents showed no obvious reduction activity, suggesting an effective enzymatic mechanism of Cr(VI) reduction in the cytoplasmic fractions of the bacterium. Previous study on refolding of sulfur oxygenase reductase (SOR) inclusion bodies from recombinant Escherichia coil showed that iron was critical to the activity of the SOR from Acidianus ambivaiens. In this study, enzymatic assays showed that 2,2'-Dipyridyl, Tiron and 8-hydroxyquinoline, which are specific for chelating ferrous or ferric ions, strongly inhibited the activity of SOR from A. tengchongensis, suggesting that iron atom is essential for SOR activity. Alignment of several functionally identified SORs and SOR-like sequences from genome database revealed a conserved, putative iron binding motif, H86-X3-H90-Xn-E114-Xn-E129 (numbering according to the Acidianus tengchongensis SOR sequence). Three mutants of SOR were generated by site-directed mutagenesis of H86, H90 and E129 into phenylalanine or alanine residue in this study. Circular dichroism spectrum determination indicated that there was no change of the secondary structures of mutant SORs, H86F, H90F and E129A, but all mutants were completely inactive. Through determination of iron contents we found that SOR mutants of H86F, H90F and E129A completely or partially lost iron, while mutants of C31S, C101S, and C104S (generated in a previous study) did not. This result indicated that H86, H90 and E129 but not C31, C101, and C104 were involved in binding to iron atom. Based on this and previous studies, it is proposed that the concerved motifs, C31-Xn-C101-X2-C104 and H86-X3-H90-X23-E114-X14-(E/D)129, are respectively for sulfur and molecular oxygen binding and activation.
