In light of recent political events in the United States, and with the urging to consider Noddings' care ethics in early childhood education, this conceptual piece considers an often-overlooked question in political discourse: what are the rights of the child, and how might these be centered in discussions of pedagogy, curricula, and legislation? Through an examination of two influential documents, the United Nations' Declaration on the Rights of the Child in 1989 and the Parental Rights in Education Act passed in Florida in 2022, the authors analyze the positioning of children in relation to parents and teachers and discuss the affordances and limitations of this positioning. Evidence for seed transmission of phytoplasmas has grown in several pathosystems including coconut (Cocos nucifera). Bogia coconut syndrome (BCS) is a disease associated with the lethal yellowing syndrome associated with the presence of 'Candidatus Phytoplasma noviguineense' that affects coconut, betel nut (Areca catechu) and bananas (Musa spp.) in Papua New Guinea. Coconut and betel nut drupes were sampled from BCS-infected areas in Papua New Guinea, dissected, the extracted nucleic acid was used in polymerase chain reaction (PCR), and loop mediated isothermal amplification (LAMP) used to check for presence of phytoplasma DNA. In a second study, drupes of both plant species were collected from multiple field sites and grown in insect-proof cages. Leaf samples taken at 6 months were also tested with PCR and LAMP. The studies of dissected coconut drupes detected phytoplasma DNA in several tissues including the embryo. Drupes from betel nut tested negative. Among the seedlings, evidence of possible seed transmission was found in both plant species. The results demonstrate the presence of 'Ca. P. noviguineense' in coconut drupes and seedlings, and in seedlings of betel nut; factors that need to be considered in ongoing management and containment efforts. [ABSTRACT FROM AUTHOR]

Purpose: Development of valgus tibiotalar tilt is a significant complication after subtalar fusion for progressive collapsing foot deformity (PCFD) correction. However, its incidence and etiologic factors have not been extensively studied. The purpose of this study was to define the incidence of valgus tibiotalar tilt after subtalar fusion for PCFD reconstruction, and to determine predictors of this complication. Methods: This study included 59 patients who underwent PCFD reconstruction with subtalar fusion. Patients with tibiotalar tilt prior to surgery were excluded. On standard weightbearing radiographs, the talonavicular coverage angle, talo-1st metatarsal angle, calcaneal pitch, hindfoot moment arm (HMA), and medial distal tibial angle were measured. Weightbearing computed tomography (WBCT) was used to determine the presence of lateral bony impingement. A radiologist evaluated the superficial and deep deltoid ligaments using magnetic resonance imaging (MRI). Univariate regression analysis was used to identify the factors associated with development of postoperative valgus tibiotalar tilt, defined as tilt > 2 degrees. Results: Seventeen patients (28.8%) developed postoperative valgus tibiotalar tilt at a mean of 7.7 (range 2–31) months. Eight (47.1%) of these patients developed valgus tibiotalar tilt within 3 months. Univariate logistic regression demonstrated association between preoperative HMA and postoperative valgus tibiotalar tilt (odds ratio 1.06, P = 0.026), with a 6% increase in risk per millimeter of increased HMA. Deltoid ligament status and concomitant procedures on other joints did not correlate with postoperative valgus tilt. Conclusion: Our findings indicate that surgeons should be cognizant of patients with a greater degree of preoperative hindfoot valgus and their propensity to develop a valgus ankle deformity. Additionally, our relatively high incidence of valgus tibiotalar tilt suggests that weightbearing ankle radiographs should be included in the initial and subsequent follow-up of PCFD patients with hindfoot valgus treated with subtalar fusion. Vaccination constitutes the best long-term solution against Coronavirus Disease-2019; however, vaccine-derived immunity may not protect all groups equally, and the durability of protective antibodies may be short. We evaluate Spike-antibody responses following BNT162b2 or ChAdOx1-S vaccination amongst SARS-CoV2-naive adults across England and Wales enrolled in a prospective cohort study (Virus Watch). Here we show BNT162b2 recipients achieved higher peak antibody levels after two doses; however, both groups experience substantial antibody waning over time. In 8356 individuals submitting a sample ≥28 days after Dose 2, we observe significantly reduced Spike-antibody levels following two doses amongst individuals reporting conditions and therapies that cause immunosuppression. After adjusting for these, several common chronic conditions also appear to attenuate the antibody response. These findings suggest the need to continue prioritising vulnerable groups, who have been vaccinated earliest and have the most attenuated antibody responses, for future boosters. Vaccination can provide reliable and long-lasting protection against COVID-19, however the immune response to vaccination can vary between individuals and can decline over time, leading to differences in protective effects. Here the authors assess the immune response to COVID-19 vaccination across a large cohort of previously uninfected adults and demonstrate lower post-vaccination antibody levels amongst those with immune-suppressing conditions and medications, as well as those with several other more common chronic conditions. A range of studies globally demonstrate that the effectiveness of SARS-CoV-2 vaccines wane over time, but the total effect of anti-S antibody levels on risk of SARS-CoV-2 infection and whether this varies by vaccine type is not well understood. Here we show that anti-S levels peak three to four weeks following the second dose of vaccine and the geometric mean of the samples is nine fold higher for BNT162b2 than ChAdOx1. Increasing anti-S levels are associated with a reduced risk of SARS-CoV-2 infection (Hazard Ratio 0.85; 95%CIs: 0.79-0.92). We do not find evidence that this antibody relationship with risk of infection varies by second dose vaccine type (BNT162b2 vs. ChAdOx1). In keeping with our anti-S antibody data, we find that people vaccinated with ChAdOx1 had 1.64 times the odds (95% confidence interval 1.45-1.85) of a breakthrough infection compared to BNT162b2. We anticipate our findings to be useful in the estimation of the protective effect of anti-S levels on risk of infection due to Delta. Our findings provide evidence about the relationship between antibody levels and protection for different vaccines and will support decisions on optimising the timing of booster vaccinations and identifying individuals who should be prioritised for booster vaccination, including those who are older, clinically extremely vulnerable, or received ChAdOx1 as their primary course. Our finding that risk of infection by anti-S level does not interact with vaccine type, but that individuals vaccinated with ChAdOx1 were at higher risk of infection, provides additional support for the use of using anti-S levels for estimating vaccine efficacy. Ecological engineering for pest suppression aimed at promoting ecosystem services of biological control, involves a range of environmentally-benign approaches to conserve and promote arthropod natural enemies and suppress pest populations, and thus reduce the need for insecticide use. Major components in rice pest management involve providing vegetation that favors parasitoid overwintering during the fallow seasons, growing nectar-producing flowering plants on the rice bunds to enhance the biocontrol function, and planting trap plants around rice fields to minimize the initial populations of pests. Complementary tactics involve using sex pheromone traps and mass-releasing Trichogramma spp. parasitoids to reduce the densities of Lepidoptera pests, and synergistically culturing ducks or fish to reduce other planthoppers. After a decade of laboratory and field research accompanied by on-farm demonstrations, ecological engineering for rice pest management has shown growth in both the underlying body of theory and practical adoption, especially in China. Ecological engineering approaches have been listed as a China National Recommendation by the Ministry of Agriculture and Rural Affairs of the People's Republic of China (MARA) since 2014. We feel this is worth documenting, especially because only a portion of that work has been reported in English-language journals so would otherwise remain "invisible" to the international scientific community. This study is the first time to systematically review the research that has allowed this rapid development and uptake in China, highlighting priorities for future research that will enhance the prospects for ecological engineering in this and other agricultural systems internationally. The application of nitrogen (N) fertilizers to agricultural crops is a common practice globally and a crucial component in the current levels of productivity. Excessive N use, however, is costly and damaging to ecosystems. It is recognized that overuse of N fertilizer can promote pest herbivores by enhancing host plant nutritional quality, but less is known of the effects of N on natural enemies of pests via improved quality and availability of prey, and how these may cascade to indirect effects on pests. Here, we explored the effects of N fertilizer on a key egg parasitiod of Nilaparvata lugens (Stål) (Hemiptera: Delphacidae) (brown planthopper, BPH). Application of N to rice plants significantly prolonged the development time of Anagrus flaveolus (Hymenoptera: Mymaridae), increased wing size and decreased fecundity of adult females. Importantly, N application to rice plants significantly decreased the per capita parasitism of BPH eggs by A. flaveolus. Utilizing planthopper prey on N-treated rice plants led to reduced searching efficiency of A. flaveolus. Ultimately, the strength of biological control exerted by A. flaveolus was negatively affected by nitrogen application under field conditions. Parasitoids were able to discriminate between BPH infested rice plants with different levels of nitrogen using visual plant cues. We conclude that N fertilizer use can have profound effects on natural enemy efficiency which potentially increase the dependence on insecticides, another potentially hazardous input. Nitrogen fertilizers are widely used to maximize crop productivity but are also known to benefit pest insects. Here we show that nitrogen not only increased the egg size of the key pest, brown planthopper (BPH), Nilaparvata lugens Stål, but also benefitted the predatory mirid bug, Cyrtorhinus lividipennis Reuter (Hemiptera: Miridae). Predator nymph survival and development rate, female adult weight, longevity and fecundity were all enhanced. Importantly, however, high nitrogen levels also extended the predator handling time (Th), reducing the number of prey eggs attacked per predator and signalling a weakening of top-down control. We conclude that nitrogen fertilizer can exacerbate pest impact by natural enemy-mediated effects in addition to previously reported direct effects on herbivores. Our results highlighted the need to moderate the use of nitrogen fertilizers to maintain effective biological control of these rice pests to reduce dependence on insecticides, another potentially hazardous input. Purpose: Patients with stable isolated injuries of the ankle syndesmosis can be treated conservatively, while unstable injuries require surgical stabilisation. Although evaluating syndesmotic injuries using ankle arthroscopy is becoming more popular, differentiating between stable and unstable syndesmoses remains a topic of on-going debate in the current literature. The purpose of this study was to quantify the degree of displacement of the ankle syndesmosis using arthroscopic measurements. The hypothesis was that ankle arthroscopy by measuring multiplanar fibular motion can determine syndesmotic instability.Methods: Arthroscopic assessment of the ankle syndesmosis was performed on 22 fresh above knee cadaveric specimens, first with all syndesmotic and ankle ligaments intact and subsequently with sequential sectioning of the anterior inferior tibiofibular ligament, the interosseous ligament, the posterior inferior tibiofibular ligament, and deltoid ligaments. In all scenarios, four loading conditions were considered under 100N of direct force: (1) unstressed, (2) a lateral hook test, (3) anterior to posterior (AP) translation test, and (4) posterior to anterior (PA) translation test. Anterior and posterior coronal plane tibiofibular translation, as well as AP and PA sagittal plane translation, were arthroscopically measured.Results: As additional ligaments of the syndesmosis were transected, all arthroscopic multiplanar translation measurements increased (p values ranging from p < 0.001 to p = 0.007). The following equation of multiplanar fibular motion relative to the tibia measured in millimeters: 0.76*AP sagittal translation + 0.82*PA sagittal translation + 1.17*anterior third coronal plane translation-0.20*posterior third coronal plane translation, referred to as the Arthroscopic Syndesmotic Assessment tool, was generated from our data. According to our results, an Arthroscopic Syndesmotic Assessment value equal or greater than 3.1 mm indicated an unstable syndesmosis.Conclusions: This tool provides a more reliable opportunity in determining the presence of syndesmotic instability and can help providers decide whether syndesmosis injuries should be treated conservatively or operatively stabilized. This study shows the effectiveness of deliberately selecting for Coptera haywardi individuals to increase a population's capacity to discriminate against parasitised hosts. In the 'selected colony' (F1–F4), females were selected based on their ability to discriminate parasitised fruit fly pupae, determined by their host searching, foraging and oviposition behaviour. Female parasitoids of successive generations of the selected colony (F1–F4) showed an increasing discriminatory ability, including reduced host searching and foraging time. The last selected generation, i.e. F4 showed an increase in fecundity compared to the standard colony. In F4 individuals from the selected colony, antennae length increased but the hind tibia size did not, compared to individuals from the control colony. Flight ability and survival remained unchanged across all generations. This selection process could be an effective method of recuperating the discriminatory capacity of a C. haywardi colony under mass rearing conditions. [ABSTRACT FROM AUTHOR]

Mobile health, or 'mHealth', is the application of mobile devices, their components and related technologies to healthcare. It is already improving patients' access to treatment and advice. Now, in combination with internet-connected diagnostic devices, it offers novel ways to diagnose, track and control infectious diseases and to improve the efficiency of the health system. Here we examine the promise of these technologies and discuss the challenges in realizing their potential to increase patients' access to testing, aid in their treatment and improve the capability of public health authorities to monitor outbreaks, implement response strategies and assess the impact of interventions across the world. Combining mobile phone technologies with infectious disease diagnostics can increase patients' access to testing and treatment and provide public health authorities with new ways to monitor and control outbreaks of infectious diseases. Intensive agriculture, which is associated with heavy inputs of synthetic insecticides, has serious ecological impacts, leading to loss of vital ecosystem services including insect-mediated pest suppression. In recent years, efforts have been made towards obtaining safer options to chemical insecticides for sustainable pest management. Habitat manipulation is a part of conservation biological control which aims at providing floral resources, alternative prey and shelter to predators and parasitoids to enhance and sustain natural pest suppression. The use of plant extracts as botanical insecticides is also an important provisioning ecosystem service. Selection of plant species for habitat manipulation has focused mainly on plants with suitable floral qualities to support natural enemies. To increase the benefits, habitat manipulation plants that can provide multiple ecosystem services in addition to floral resources would be an ideal. In this review, we focus on the potential of achieving the dual ecosystem services of bioinsecticidal source plants in addition to the provision of floral resources from selected plant species. Our literature search found 283 plants species from 44 plant families that have been involved in habitat manipulation studies. Fifteen of these plant families have species that have been exploited for their insecticidal properties. Three families, Apiaceae, Asteraceae and Lamiaceae, have the largest number of species that have been used for both habitat manipulation and botanical insecticides. Of the four most popular habitat manipulation plants, alyssum Lobularia maritime (L.) Desv. (Brassicaceae), buck wheat Fagopyrum esculentum Moench (Polygonaceae), coriander Coriandrum sativum L. (Apiaceae) and phacelia Phacelia tanacetifolia Benth. (Boraginaceae), buckwheat and coriander have been used for insecticidal purposes whilst no records exist of phacelia and alyssum as botanical insecticide species. There is great potential for identifying plant species that can support natural enemies as well as providing potent plant extracts as botanical insecticides by selecting species from the Apiaceae, Asteraceae and Lamiaceae families. Purpose: Radiography has a low sensitivity for the detection of fractures related to the talonavicular and calcaneocuboid articulations, also known as Chopart fractures. The purpose of this study is to determine the sensitivity of radiographs for detecting additional foot and ankle fractures related to Chopart fracture using CT or MRI as the reference standard.Method: We performed an IRB-approved, retrospective review of radiology reports between 2010 and 2014. Inclusion criteria were (1) diagnosis of a Chopart fracture and (2) at least one radiograph and subsequent cross-sectional imaging (CT or MR). CT or MRI was considered the diagnostic reference standard. Results were stratified by the energy of trauma and by type of radiograph performed (weight-bearing (WB) versus non-WB).Results: One hundred eight patients met the inclusion criteria. The calcaneocuboid articulation was the most commonly involved type of Chopart fracture, seen in 75% of cases (81/108). Chopart fractures were detected on the initial radiographs in 67.6% of cases (73/108). Additional fractures of the ankle and midfoot were diagnosed in 34.2% of cases (37/108), with 56.7% (21/37) of these cases having at least one additional fracture seen on CT or MRI that was not seen on the initial radiographs, with fractures of the midfoot most often missed. In 56.7% (17/30) patients whose radiographs detected Chopart fractures, at least one additional fracture was missed; 30% of them demonstrated intra-articular extension and 56.7% were considered displaced. High-energy trauma was related to higher incidence of additional fractures. There was no significant difference in the sensitivity of radiographs to detect additional fractures between high versus low-energy trauma (p = 0.3) and WB versus non-WB radiographs (p = 0.5). Most patients were treated nonoperatively (56.5%, 61/108), with surgical intervention more frequent in patients with a high energy of trauma (51.7% versus 33.3%, p = 0.05).Conclusion: In the setting of a Chopart fracture, CT or MRI can add significant value in the detection of additional ankle or midfoot fractures, irrespective of the energy of trauma. Triggering of the toes is rare, with isolated cases reported in the literature involving predominantly the flexor hallucis longus (FHL) tendon, a condition known as "hallux saltans" (HS). We report the dynamic sonographic findings of a 42-year-old female with a 2-month history of ankle pain and triggering of right hallux, consistent with HS. Sonography demonstrated tenosynovitis and focal thickening of FHL at the level of hindfoot, with a fibrous band adjacent to the tendon. Dynamic ultrasound showed triggering and snapping of FHL tendon during hallux flexion and extension, respectively. Conservation biological control avoids the need for mass releases of costly agents and the risks associated with introducing exotic agents by promoting existing natural enemies. This is done by alleviating insecticide-induced mortality and by manipulating the habitat to provide resources such as nectar, but there is a dearth of information on the relative and interactive effects of these two approaches. Here we used a large-scale factorial experiment with plots comprised of entire fields to test the effects of, and interactions between, withholding insecticides and planting borders of sesame (Sesamum indicum) on natural enemies and pests over 2 years. We used yellow sticky traps, sweeping netting and sentinel bait plants to monitor natural enemies and pests in the canopy and basal zones of the rice crop. Numbers of rice planthopper egg parasitoids and lepidopterous egg parasitoids in the rice canopy, as well as planthopper parasitism rates, were significantly greater in plots that were unsprayed and bordered by sesame, and scarcest in sprayed crops without sesame. Spraying of sesame-bordered crops gave parasitoid numbers similar to sprayed crops without sesame. Spiders in the canopy were significantly reduced in numbers by spraying, but there was no main effect of sesame borders. This study demonstrates that withholding insecticides and sowing nectar plant borders each have measurable as well as additive benefits on in-crop densities of ecosystem service providers responsible for predating and parasitising pests but the identity of the natural enemy determines the impact of these management practices. Patterns of sexual partnership formation and dissolution are key drivers of sexually transmitted infection transmission. Sexual behavior survey participants may be unable or unwilling to report accurate details about their sexual partners, limiting the potential to capture information on sexual mixing and timing of partnerships. We examined how questions were interpreted, including recall strategies and judgments made in selecting responses, to inform development of a module on recent sexual partnerships in Britain's third National Survey of Sexual Attitudes and Lifestyles ("Natsal-3"). Face-to-face cognitive interviews were conducted with 14 men and 18 women aged 18-74 years, during development work for Natsal-3. People with multiple recent partners were purposively sampled and questions were presented as a computer-assisted self-interview. Participants were generally agreeable to answering questions about their sexual partners and practices. Interpretation of questions designed to measure concurrent (overlapping) partnerships was broadly consistent with the epidemiological concept of concurrency. Partners' ages, genders, ethnicity, and participants' perceptions of whether partner(s) had had concurrent partnerships were reported without offense. Recall problems and lack of knowledge were reported by some participants (of all ages), especially about former, casual, and/or new partnerships, and some reported guessing partners' ages and dates of sex. Generally, participants were able to answer questions about their sexual partners accurately, even when repeated for multiple partners. Cognitive interviews provided insight into the participants' understanding of, ability to answer, and willingness to answer questions. A stratified probability sample survey of the British general population, aged 16 to 44 years, was conducted from 1999 to 2001 ( N = 11,161) using face-to-face interviewing and computer-assisted self-interviewing. We used these data to estimate the population prevalence of masturbation, and to identify sociodemographic, sexual behavioral, and attitudinal factors associated with reporting this behavior. Seventy-three percent of men and 36.8% of women reported masturbating in the 4 weeks prior to interview (95% confidence interval 71.5%–74.4% and 35.4%–38.2%, respectively). A number of sociodemographic and behavioral factors were associated with reporting masturbation. Among both men and women, reporting masturbation increased with higher levels of education and social class and was more common among those reporting sexual function problems. For women, masturbation was more likely among those who reported more frequent vaginal sex in the last four weeks, a greater repertoire of sexual activity (such as reporting oral and anal sex), and more sexual partners in the last year. In contrast, the prevalence of masturbation was lower among men reporting more frequent vaginal sex. Both men and women reporting same-sex partner(s) were significantly more likely to report masturbation. De novo dominant mutations in the GFAP gene have recently been associated with nearly all cases of Alexander disease, a rare but devastating neurological disorder. These heterozygous mutations must occur very early in development and be present in nearly all cells in order to be detected by the sequencing methods used. To investigate whether the mutations may have arisen in the parental germ lines, we determined the parental chromosome bearing the mutations for 28 independent Alexander disease cases. These cases included 17 different missense mutations and one insertion mutation. To enable assignment of the chromosomal origin of the mutations, six new single nucleotide polymorphisms in the GFAP gene were identified, bringing the known total to 26. In 24 of the 28 cases analyzed, the paternal chromosome carried the GFAP mutation ( P<0.001), suggesting that they predominantly arose in the parental germ line, with most occurring during spermatogenesis. No effect of paternal age was observed. There has been considerable debate about the magnitude of the male to female germ line mutation rate; our ratio of 6:1 is consistent with indirect estimates based on the rate of evolution of the sex chromosome relative to the autosomic chromosomes. Alexander disease, a rare fatal disorder of the central nervous system, causes progressive loss of motor and mental function. Until recently it was of unknown etiology, almost all cases were sporadic, and there was no effective treatment. It was most common in an infantile form, somewhat less so in a juvenile form, and was rarely seen in an adult-onset form. A number of investigators have now shown that almost all cases of Alexander disease have a dominant mutation in one allele of the gene for glial fibrillary acidic protein (GFAP) that causes replacement of one amino acid for another. Only in very rare cases of the adult-onset form is the mutation present in either parent. Thus, in almost all cases, the mutation arises as a spontaneous event, possibly in the germ cell of one parent. [ABSTRACT FROM AUTHOR]

Our study represents a rural case management agency's use of its data to aid in its transition from being funded by federal grant funds to self-sustainment. We wanted to learn if some characteristics of those children served could predict the resources needed to serve them. We analyzed data on 90 children and adolescents living in a rural community. Using multiple regression analysis we found that CAFAS total and CBCL total problem scores predicted Medicaid reimbursements but not flexible funds spending or case management hours. History of psychiatric hospitalization, however, predicted flexible funds spending and case management hours. Using one-way ANOVA and post hoc analysis we found that the staff members' assessments of each child and family's level of service needs were reflected in differences in mean CAFAS total scores, Medicaid reimbursements and case management hours, but not CBCL total problem scores or flexible funds spending. The compound [Cu2 II(D1)(H2O)2](ClO4)4 .2H2O [D1=binucleating ligand with tris(2-pyridylmethyl)amine (TMPA) moieties linked in the 5-pyridyl position by a -CH2CH2- bridge] mediated efficient oxidative cleavage of pBR322 plasmid DNA under reducing conditions. A mononuclear analogue, [Cu(TMPA)(H2O)](ClO4)2, was less effective at linearizing supercoiled (Form I) plasmid DNA as compared to the binuclear complex. A new method for quenching the copper-dependent reactions has been developed to avoid plasmid scission by the binuclear complex and the standard gel loading buffer. EDTA was not sufficient for retarding copper reaction, but diethyldithiocarbamic acid was capable of inhibiting all reactivity. Investigation of oxidative cleavage of double-helical oligonucleotides by [Cu2 II(D1)(H2O)2](ClO4)4 confirmed the enhanced reactivity of the binuclear over the mononuclear complex and provided mechanistic insights into the nature of the reaction. Cleavage of DNA required both the binuclear complex and a reductant and likely proceeded through an O2-derived intermediate that does not include a diffusible hydroxyl radical. The greater efficiency of the binuclear complex relative to the mononuclear analogue is consistent with their relative abilities to activate dioxygen. Caspase-3 deficiency can limit the efficiency of pro-apoptotic anticancer treatments. Irofulven (hydroxymethylacylfulvene, HMAF, MGI 114, NSC 683863) is an antitumor drug, currently in a Phase III and multiple Phase II trials, which can differentiate between tumor and normal cells in apoptosis induction. This study investigated whether apoptosis induced by irofulven requires caspase-3. Irofulven action was compared in breast cancer cells differing in caspase-3 status: deficient MCF-7 cells and proficient MDA-MB-231 cells and in normal human mammary epithelial cells, HMEC. Irofulven induces significant, concentration and time-dependent apoptotic DNA fragmentation in breast cancer cell lines, regardless of caspase-3 status. After 12, 24 and 48 h incubation at 1 μM irofulven (∼ 3 × GI50), fragmented DNA comprised 3.7, 14.1 and 34.6% and 8.4, 12.6 and 20.3% of total DNA in MCF-7 and MDA-MB-231 cells, respectively. Cell viability (trypan blue exclusion) remained largely unaffected during the first 24 h but decreased markedly after 48 h, indicating secondary necrosis. Net losses in cell numbers were apparent at 48 h. Normal HMEC cells were refractory to 1 μM drug with only ∼3–9% fragmented DNA after 12–48 h, although apoptosis was observed at drug levels >3 μM. The broad-spectrum caspase inhibitor Z-VAD-fmk inhibited irofulven-induced apoptosis of all cell lines at 20 μM with nearly complete abrogation of apoptosis at 100 μM. Irofulven treatment resulted in marginal caspase-3 processing in MDA-MB-231 and HMEC cells. Alexander disease is a rare disorder of the central nervous system of unknown etiology. Infants with Alexander disease develop a leukoencephalopathy with macrocephaly, seizures and psychomotor retardation, leading to death usually within the first decade; patients with juvenile or adult forms typically experience ataxia, bulbar signs and spasticity, and a more slowly progressive course. The pathological hallmark of all forms of Alexander disease is the presence of Rosenthal fibers, cytoplasmic inclusions in astrocytes that contain the intermediate filament protein GFAP in association with small heat-shock proteins. We previously found that overexpression of human GFAP in astrocytes of transgenic mice is fatal and accompanied by the presence of inclusion bodies indistinguishable from human Rosenthal fibers. These results suggested that a primary alteration in GFAP may be responsible for Alexander disease. Sequence analysis of DNA samples from patients representing different Alexander disease phenotypes revealed that most cases are associated with non-conservative mutations in the coding region of GFAP. Childhood ataxia with diffuse central nervous system hypomyelination syndrome (CACH) is a recently described leukodystrophy of unknown etiology. To characterize the neuropathological features and gain insight as to the pathogenesis of this disorder, we studied cerebral tissue from six patients with the CACH syndrome. Evaluation of toluidine blue-stained, semithin sections of white matter from CACH patients disclosed unusual cells with “foamy” cytoplasm, small round nuclei and fine chromatin. Electron microscopy (EM) revealed cells in the white matter with abundant cytoplasm containing many mitochondria and loosely clustered, membranous structures, but lacking the lysosomal structures seen in macrophages. Further analysis of tissue sections with antibodies and special stains demonstrated that the abnormal cells with abundant cytoplasm labeled with oligodendroglial markers, but did not react with macrophage or astrocytic markers. Double immunolabeling with macrophage and oligodendroglial markers clearly distinguished macrophages from the “foamy” oligodendroglial cells (FODCs). Proteolipid protein (PLP) mRNA in situ hybridization demonstrated PLP mRNA transcripts in a high proportion of oligodendrocytes in CACH patients compared to control patients, and PLP mRNA transcript signal in cells, morphologically consistent with FODCs. Normal and pathological brain control tissues did not contain FODCs. Adrenal glands from eight patients with the cerebro-hepatorenal syndrome, a disease in which there are no morphologically demonstrable peroxisomes, were studied histologically; one of the eight was also examined ultrastructurally. Seven of the eight demonstrated striated adrenocortical cells in the inner portion of the adrenal cortex. Ultrastructural examination confirmed that the striated cells contained the lammellae and lamellar-lipid profiles of very long chain fatty acids-cholesterol esters that are characteristic of adreno-leukodystrophy. Physostigmine was given intravenously to a total dose of 3 mg to 13 subjects; a placebo of 0.25 N saline was given intravenously to 10 other subjects; both groups received 1 mg of methscopolamine bromide subcutaneously preceding the intravenous infusions. A 'physostigmine syndrome' consisting of decreased speech, slowed thoughts, mild sedation, expressionless faces, nausea, and decreased spontaneous activity was evident following doses of 1.5 to 2.0 mg of physostigmine. The capacity of short-term memory (STM) as measured by digit span tasks was significantly less for the subjects who received physostigmine than for the subjects who received placebo. No difference was observed between the two groups on tasks of consolidation from STM to long-term memory (LTM). Subiects who received physostigmine did not significantly differ from subjects who received placebo in their mood. The ATP-binding DNA aptamer is often used as a model system for developing new aptamer-based biosensor methods. This aptamer follows a structure-switching binding mechanism and is unusual in that it binds two copies of its ligand. We have used isothermal titration calorimetry methods to study the binding of ATP, ADP, AMP and adenosine to the ATP-binding aptamer. Using both individual and global fitting methods, we show that this aptamer follows a positive cooperative binding mechanism. We have determined the binding affinity and thermodynamics for both ligand-binding sites. By separating the ligand-binding sites by an additional four base pairs, we engineered a variant of this aptamer that binds two adenosine ligands in an independent manner. Together with NMR and thermal stability experiments, these data indicate that the ATP-binding DNA aptamer follows a population-shift binding mechanism that is the source of the positive binding cooperativity by the aptamer. Mulching with organic materials is a management practice with long history for weed suppression, soil water conservation and erosion control. Its potential impact on crop pests is less well explored. Here we report its utility for reducing crop damage by the serious pest, sweetpotato weevil (Cylas formicarius). Laboratory bioassays measured the response of adult female weevils to sweetpotato storage roots beneath mulches of fresh or dried plant materials. Weevils were significant repelled by fresh basil, catnip, basil lime and dry eucalyptus, cypress, lucerne and sugarcane. A subsequent field study found that mulches of dry cypress, eucalyptus and lucerne reduced movement of weevils from a release point to reach sweetpotato plants and lowered level of damage to storage roots. Results demonstrate that mulching with organic materials merits further testing as part of the integrated management of sweetpotato weevil, particularly to protect developing storage roots during dry periods when soil cracking can facilitate access by pests. Diamondback moth (Plutella xylostella) is a globally significant pest of Brassicaceae crops that has attracted enormous research investment. It is typical of many agricultural pests, with insecticides remaining the most common method of control, despite frequent cases of resistance in pest populations and the potential for other management options such as natural enemies to provide suppression. Here we review scope to make better use of neglected natural enemy taxa and integrate recent work on landscape ecology to identify opportunities for more effective pest suppression. Our main findings are as follows: (1) relatively neglected taxa of natural enemies, especially predators and entomopathogens, are now attracting growing levels of research interest, although parasitoids remain most frequently used and researched; (2) knowledge of the spatio-temporal dynamics of populations at the landscape scale have advanced rapidly in the last decade; (3) ecological insights open new possibilities for exploiting spatial heterogeneity at scales larger than individual fields and even farms that influence pests and their natural enemies; (4) there is evidence for landscapes that selectively favor particular guilds and this knowledge could be developed to favor targeted natural enemies over pests in focal crops; and (5) landscape-scale effects can even over-ride field-scale management practices. The significance of these advances is that future management of diamondback moth and similar pests will benefit from a move away from reliance on the use of particular species of biological control agents, especially exotic parasitoids, and strategies that depend on use of broad-spectrum insecticides.
