The valorization of lignin-derived feedstocks by catalytic means enables their defunctionalization and upgrading to valuable products. However, the development of productive, safe, and low-waste processes remains challenging. This paper explores the industrial potential of a chemoenzymatic reaction performing the decarboxylation of bio-based phenolic acids in wet cyclopentyl methyl ether (CPME) by immobilized phenolic acid decarboxylase from Bacillus subtilis, followed by a base-catalyzed acylation. Key-to-success is the continuous control of water activity, which fluctuates along the reaction progress, particularly at high substrate loadings (triggered by different hydrophilicities of substrate and product). A combination of experimentation, thermodynamic equilibrium calculations, and MD simulations revealed the change in water activity which guided the integration of water reservoirs and allowed process intensification of the previously limiting enzymatic step. With this, the highly concentrated sequential two-step cascade (400 g·L–1) achieves full conversions and affords products in less than 3 h. The chemical step is versatile, accepting different acyl donors, leading to a range of industrially sound products. Importantly, the finding that water activity changes in intensified processes is an academic insight that might explain other deactivations of enzymes when used in non-conventional media. Lignin-derived phenolic acids can be upgraded to styrene derivatives through chemoenzymatic processes, however, the scale-up of such processes remains challenging. Development of efficient enzymatic reaction cascades necessitates individual and careful handling of reaction conditions, which may differ from one enzymatic step to another one. Compartmentalization is a key solution that can be either done under static or flow conditions. Multi-enzymatic cascade reactions enable the (re)design of synthetic routes via efficient multi-step biotransformations. Interestingly, enzymatic cascades can profit both from spatial separation (e. g. by compartmentalization) and from co-localization of the involved enzymes (e. g. by co-immobilization). Baeyer-Villiger monooxygenases (BVMOs) are valuable enzymes for specific oxyfunctionalization chemistry. They catalyze the oxidation of ketones to esters, but are also capable of oxidizing other chemical functions, namely aldehydes and heteroatoms such as sulfur, nitrogen, selenium and boron. The oxidation specificity and enantioselectivity of a newly characterized BVMO (BVMO4) from a strain of Dietzia towards sulfide- and aldehyde substrates have been studied. BVMO4 could react with sulfides containing an aromatic group. The presence of a substituent on the aromatic group was tolerated when they were in the meta- and para position and the oxidations yielded predominantly the ( R)-sulfoxides. Similarly, BVMO4 displayed a higher activity for aldehydes containing a phenyl group, but long aliphatic aldehydes, namely octanal and decanal, were also accepted as substrate by this enzyme. The major oxidation products of the aldehyde substrates were the respective carboxylic acids in contrast to formate ester that was obtained in most of the previous reports. The Baeyer-Villiger oxidation of the substrate 2-phenylpropionaldehyde was studied in further detail and the corresponding acid product was obtained with good regio- and enantioselectivity. Cofactor-dependent enzymes catalyze a broad range of synthetically useful transformations. However, the cofactor requirement also poses economic and practical challenges for the application of these biocatalysts. For three decades, considerable research effort has been devoted to the development of reliable in situ regeneration methods for the most commonly employed cofactors, particularly NADH and NADPH. Today, researchers can choose from a plethora of options, and oxidoreductases are routinely employed even on industrial scale. Nevertheless, more efficient cofactor regeneration methods are still being developed, with the aim of achieving better atom economy, simpler reaction setups, and higher productivities. Besides, cofactor dependence has been recognized as an opportunity to confer novel reactivity upon enzymes by engineering their cofactors, and to couple (redox) biotransformations in multi-enzyme cascade systems. These novel concepts will help to further establish cofactor-dependent biotransformations as an attractive option for the synthesis of biologically active compounds, chiral building blocks, and bio-based platform molecules. If an adequate biocatalyst is identified for a specific reaction, immobilization is one possibility to further improve its properties. The immobilization allows easy recycling, improves the enzyme performance, and it often enhances the stability of the enzyme. In this work, the immobilization of the benzoylformate decarboxylase (BFD) variant, BFD A460I-F464I, from Pseudomonas putida was accomplished on spherical silica. Silicagel is characterized by its high mechanical stability, which allows its application in different reactor types without restrictions. The covalently bound enzyme was characterized in terms of its activity, stability, and kinetics for the formation of chiral 2-hydroxypropiophenone (2-HPP) from benzaldehyde and acetaldehyde. Moreover, temperature as well as pressure dependency of immobilized BFD A460I-F464I activity and enantioselectivity were analyzed. The used wide-pore silicagel shows a good accessibility of the immobilized enzyme. The activity of the immobilized BFD A460I-F464I variant was determined to be 70% related to the activity of the free enzyme. Thereby, the enantioselectivity of the enzyme was not influenced by the immobilization. In addition, a pressure-induced change in stereoselectivity was found both for the free and for the immobilized enzyme. With increasing pressure, the enantiomeric excess ( ee) of ( R)-2-HPP can be increased from 44% (0.1 MPa) to 76% (200 MPa) for the free enzyme and from 43% (0.1 MPa) to 66% (200 MPa) for the immobilized enzyme. Selection of the suitable material handling equipment (MHE) is a very difficult task for the manufacturing companies because of the considerable capital investment required. There are many tangible and intangible factors for choosing the suitable MHE. Multiple criteria decision making (MCDM) has been found to be a useful approach to analyze these conflicting factors. The evaluation of MHE alternatives within the frame of various subjective criteria and the weights of the criteria are usually expressed in linguistic terms. This makes fuzzy logic a more natural approach to this kind of problems. This paper proposes a combined MCDM methodology for evaluation and selection of MHE types for a company in the steel construction industry in Istanbul, Turkey. Fuzzy analytic network process (FANP) is utilized for assigning weights of the criteria for MHE selection and fuzzy technique for order preference by similarity to ideal solution (TOPSIS) is used to determine the most proper system alternative using the criteria weights attained by FANP. The selection is based on the compatibility between MHE and production characteristics. Objective is to select the most efficient MHE considering also the cost efficiency. The selection of the appropriate machine tools for a manufacturing company is one of the important points to achieving high competitiveness in the market. Besides, an appropriate choice of machine tools is very important as it helps to realize full production quickly. Today’s market offers many more choices for machine tool alternatives. There are also many factors one should consider as part of the appropriate machine tool selection process, including productivity, flexibility, compatibility, safety, cost, etc. Consequently evaluation procedures involve several objectives and it is often necessary to compromise among possibly conflicting tangible and intangible factors. For these reasons, multiple criteria decision making (MCDM) has been found to be a useful approach to solve this kind of problem. Most of the MCDM models are basically mathematical and ignore qualitative and often subjective considerations. The use of fuzzy set theory allows incorporating qualitative and partially known information into the decision model. This paper describes a fuzzy technique for order preference by similarity to ideal solution (TOPSIS) based methodology for evaluation and selection of vertical CNC machining centers for a manufacturing company in Istanbul, Turkey. Biocatalysis in organic media enables higher concentrations of mostly hydrophobic reagents compared to traditional aqueous reaction systems.
