Experiments were conducted to identify different ratios of Bacillus sp. SJ-10 and Lactobacillus plantarum KCCM 11322 mixtures at a concentration of 1 × 108 CFU/g diet; the effects on growth and cellular and humoral immune responses and the characteristics of disease protection in olive flounder (Paralichthys olivaceus). Flounder were divided into six groups and fed control diet D-1 (without Bacillus sp. SJ-10 and L. plantarum KCCM 11322), positive control diets D-2 (Bacillus sp. SJ-10 at 1 × 108 CFU/g feed) and D-3 (L. plantarum KCCM 11322 at 1 × 108 CFU/g feed); or treatment diets D-4 (3:1 Bacillus sp. SJ-10 and L. plantarum KCCM 11322 at 0.75 + 0.25 × 108 CFU/g feed), D-5 (1:1 Bacillus sp. SJ-10 and L. plantarum KCCM 11322 at 0.50 + 0.50 × 108 CFU/g feed), or D-6 (1:3 Bacillus sp. SJ-10 and L. plantarum KCCM 11322 at 0.25 + 0.75 × 108 CFU/g feed) for 8 weeks. Group D-4 demonstrated better growth and feed utilization (P < 0.05) compared with the controls and positive controls. Similar modulation was also observed in respiratory burst for all treatments and in the expression levels of TNF-α, IL-1β, IL-6, and IL-10 in different organs in D-4. D-4 and D-5 increased respiratory burst, superoxide dismutase, lysozyme, and myeloperoxidase activities compared with the controls, and only D-4 increased microvilli length. When challenged with 1 × 108 CFU/mL Streptococcus iniae, the fish in the D-4 and D-5 groups survived up to 14 days, whereas the fish in the other groups reached 100% mortality at 11.50 days. Collectively, a ratio-specific Bacillus sp. SJ-10 and L. plantarum KCCM 11322 mixture (3:1) was associated with elevated growth, innate immunity, and streptococcosis resistance (3:1 and 1:1) compared with the control and single probiotic diets. In recent years, considerable and growing attention has been given to the application of host-associated microorganisms as a more suitable source of probiotics in aquaculture sector. Herein, we isolated and screened the olive flounder gut microbiota for beneficial bacterial strains that might serve as potential probiotics in a low fishmeal extruded aquafeed. Among the ten identified isolates, Bacillus amyloliquefaciens SK4079 and B. subtilis SK4082 were screened out based on their heat-resistant ability as well as enzymatic and non-hemolytic activities. Although both strains were well able to utilize carboxymethyl cellulose (CMC), xylan, and soybean meal (SBM) as a single carbon source in the minimal nutrient M9 medium, B. subtilis exhibited significantly higher cellulase, xylanase, and protease activities than B. amyloliquefaciens. The two selected strains were well able to degrade the undesirable anti-nutritional component of the SBM, which would limit its utilization as protein source in aquafeed industry. Significantly higher biofilm formation capacity and notably stronger adhesive interactions with the flounder's skin mucus were detected in B. subtilis than B. amyloliquefaciens. Immobilization of the spores from the selected strains, in a SBM complex carrier, remarkably enhances their thermal resistance at 120 °C for 5 min and different drying conditions. It was also interesting to learn that the B. subtilis spores could survive and remain viable after being sprayed onto extruded low-fish meal feed pellets for as long as 6 months. The purpose of this study was to evaluate Bacillus sp. SJ-10, isolated from traditional Korean seafood, as a probiotic. Strain SJ-10 was demonstrated to be safe, on the basis of in vitro tests confirming the absence of cytotoxicity, hemolysis, and genes with toxigenic potential, and was susceptible to antibiotics. It met the probiotic prerequisites of a spore count that remained almost constant, acid and bile tolerance under simulated gastrointestinal conditions, and significant adhesion to Caco-2 cells. Moreover, SJ-10 demonstrated beneficial properties as a probiotic: broad-spectrum antimicrobial activity, hydroxyl radical, antioxidant activity, production of functional enzymes such as β-galactosidase and phytase, and selective growth via β-glucan fermentation. The fish-feeding trial demonstrated that olive flounder fed diets containing SJ-10 alone or in combination with β-glucan exhibited significantly higher growth performance and pathogenic disease resistance compared with those fed diets containing β-glucan alone, indicating that SJ-10 diets exerted a beneficial effect as an antibiotic replacer in terms of growth performance and disease resistance in olive flounder. We evaluated the effects of diets incorporating the red algae Pyropia yezoensis, prepared by several different extraction methods, on the growth of juvenile Japanese flounder Paralichthys olivaceus. We assessed growth performance, as well as the levels of amino acids, fatty acids, insulin-like growth factor I (IGF-I) and interleukins (ILs). Four experimental diets were developed based on different methods of processing P. yezoensis. A commercial feed, laver powder (P), high-pressure heat extraction of laver (HPHE) and acid hydrolysis extraction of laver were used as the experimental diets. Three experimental replicates were established for each diet (40 fish/group, body weight 123.7 ± 1.1 g), and the fish were fed for 6 weeks. We found no significant differences in weight gain, specific growth rate or feeding efficiency among the groups ( P > 0.05); however, the fish fed HPHE had the greatest growth performance. Fish fed the laver extracts exhibited the highest protein efficiency ratio compared with the control and P groups. The experimental groups fed P. yezoensis extracts had significantly higher levels of IGF-I ( P < 0.05) than those of the control group. High levels of IL-2 were found in the P and HPHE groups, IL-12 in the HPHE group, and IL-6 in all experimental groups. Therefore, these results suggest that a P. yezoensis extract improved the growth performance and immunity of Japanese flounder. In particular, the high-pressure heating process was a useful extraction method for preparing a P. yezoensis extract, which had beneficial effects as a dietary supplement in Japanese flounder. promising solution to solve the low phosphorus availability of most plant protein sources and make those ingredients more useful and functional in feeds for fish is reported. Three experimental diets supplemented with 0, 3, and 6% meju (designated as Meju 0, Meju 3, and Meju 6) were formulated. After 8 weeks of a feeding trial, growth performance, feed utilization, and survival were not different among fish groups fed the test diets. Phosphorus retention in fish fed Meju 6 diet was significantly higher than that of fish fed the control diet. Increased antioxidant activity in meju diets was observed with increasing polyphenol and flavonoids contents. Liver superoxide dismutase activity was significantly increased by the supplementation of meju. Microbial fermentation of soybean, which also can be simply adapted for soybean meal, is a promising method for increasing phosphorus availability in soybean meal, antioxidant activities in diets, and nonspecific immune response of olive flounder. This study aimed to assess the effectiveness of low-fish meal diets (LFMD), incorporating animal (tankage meal, tuna byproduct meal and poultry byproduct meal) and plant (soy protein concentrate and wheat gluten meal) protein sources as a fish meal (FM) replacement in juvenile olive flounder Paralichthys olivaceus. A control diet (FM70) was prepared to contain 70% of sardine and anchovy FM (1:1, v:v). Six experimental diets were formulated reducing FM levels in FM70 diet down to 35, 30, 25, 20, 15 and 10% (designated as FM35, FM30, FM25, FM20, FM15 and FM10, respectively) with increasing proportions of animal and plant protein sources. Fish (initial mean body weight: 22.3 ± 0.01 g) were fed each experimental diet for 12 weeks. At the end of the feeding trial, growth performance, survival and biological indices were not affected by dietary FM levels. Feed utilization was significantly decreased in FM15 and FM10 groups than in FM70 group. The gene expression of tumor necrosis factor-α (TNF-α) was significantly upregulated in FM10 group compared to FM70 group. In muscle fatty acid profiles, a reduction in palmitoleic, eicosapentaenoic and docosahexaenoic acids was observed, while oleic and linoleic acid levels increased with the decrease in dietary FM levels. These results indicate that the combined use of animal and plant protein sources would be a good option as a FM substitute for juvenile olive flounder. Considered with the decrease in feed utilization and upregulation of TNF-α gene expression, the dietary FM levels could be reduced only down to 20%.
