1. An advanced 3D lymphatic system for assaying human cutaneous lymphangiogenesis in a microfluidic platform.

Abstract

The human cutaneous lymphatic system strictly controls lymphatic functions by coordinating with skin cells. The lymphatic system plays important roles in removing cell waste, residual proteins, various antigens, and immune cells from tissues to maintain homeostasis and activate the immune system through the drainage of interstitial fluid1,2. The skin protects our body from external stimuli such as pathogens through the cutaneous lymphatic system3,4. Herein, to develop an in vitro human cutaneous lymphatic model, we present two 3D microfluidic platforms: a lymphangiogenesis model with a precollecting lymphatic vessel-like structure and an advanced lymphangiogenesis model with a functional cutaneous barrier and a precollecting lymphatic vessel-like structure. In addition, we rapidly analyzed prolymphangiogenic effects using methods that incorporate a high-speed image processing system and a deep learning-based vascular network analysis algorithm by 12 indices. Using these platforms, we evaluated the pro-lymphangiogenic effect of Lymphanax, a natural product derived from fresh ginseng. As a result, we demonstrated that Lymphanax induces robust lymphangiogenesis without any structural abnormalities. In conclusion, we suggest that these innovative platforms are useful for studying the interaction between the skin and lymphatic system as well as evaluating the prolymphangiogenic effects of drugs and cosmetics. To aid in this, researchers created a microfluidic chip model (a miniature device that simulates bodily functions) to examine lymphangiogenesis (the creation of new lymphatic vessels) in a regulated setting. This model lets scientists explore how a natural substance, Lymphanax, derived from the root of Panax Ginseng (a type of plant), impacts lymphangiogenesis. The study discovered that Lymphanax encourages the growth of new lymphatic vessels without inducing structural abnormalities, hinting at its potential medicinal value. This investigation paves the way for future study of the lymphatic system and potential treatment trials. Abstract

Recent policies have the potential to address the gaps between need for and access to mental health (MH) services among youth involved in the child welfare system (CWS). Yet, little is known about whether changes in the pattern of MH service use have occurred. This study sought to uncover whether the rates and predictors of MH service use among CWS-involved youth have changed over the past decade. This study relied on administrative data of CWS-involved youth in a large mid-Atlantic city of the United States. The sample consisted of two cohorts of youth who entered the CWS in 2003 and 2012, respectively. Logistic and negative binomial regression models were conducted to examine utilization and dosage of MH services in relation to individual (e.g., cohort, gender, age, and race/ethnicity) and case characteristics (e.g., placement type/instability, and juvenile justice system involvement). Both utilization and dosage of MH services were found to have increased between cohorts, even after controlling for individual and case characteristics. There were also promising trends toward more equitable access and utilization of MH services along age and racial lines. However, disparities by gender and placement type were still prevailing problems. Potential positive effects of the recent policies are likely reflected in our findings of increased MH service use between the two cohorts. 3. How do Maternal Nonstandard Work Schedules Affect Early Child Development? A Mediation Analysis.

Abstract

This study examined how the two dimensions of maternal nonstandard work schedules, namely working nonstandard hours and working weekend, influence early child development outcomes, with a focus on the mediating role of maternal involvement in children's education, parenting stress, and maternal health. Structural equation modelling, using data from mothers of young children (age 5–6) in Hong Kong, found that maternal weekend work was associated with lower levels of maternal involvement in children's education and suboptimal health status, and that these in turn were associated negatively with overall early child development. By contrast, no such negative mediation effects were detected for mothers working nonstandard hours. Instead, there was a direct, positive association between working nonstandard hours and early child development outcomes. This pattern of findings suggests that the hourly and weekly time frame of maternal nonstandard work schedules (i.e., working nonstandard hours vs. working weekend) may affect young children differently, with different pathways. Abstract

For the safe disposal of high-level radioactive waste, it is necessary to establish a numerical model that can simulate the phenomenon of buffer expansion due to groundwater inflow. The Barcelona basic model (BBM), one of many models that describe the swelling behavior of buffer, can represent the behavior of expansive soil but requires various hydro-mechanical input parameters. Conventional experiments to determine these parameters are time-consuming and complicated; therefore, this study proposes a method of determining the BBM parameters by comparing the results of swelling tests and numerical analysis. The relationships between these parameters were determined through an artificial neural network and multi-objective optimization was used to derive the Pareto optimal sets. Among various optimized solutions, single BBM parameters were derived from Pareto sets by considering additional conditions. 5. Development of an Extracellular Matrix Plate for Drug Screening Using Patient-Derived Tumor Organoids.

Abstract

With the advances in organoid culture, patient-derived organoids are utilized in diverse fields to broaden our understanding of conventional 2-dimensional (2D) culture methods and animal models. Patient-derived organoids have found new applications, such as screening for patient-matching drugs, immune checkpoint drugs, and mutation-target drugs, in the field of drug screening. However, conventional dome-shaped Matrigel drop-based screening methods using 24-, 48-, and 96-well plates are not effective for carrying out large-scale drug screening using organoids. Here, we present a newly developed 96-well plate-based method for the effective screening of patient-derived tumor organoids embedded in Matrigel. The new screening plate has a central hole with a diameter of 3 or 5 mm to provide a definite space for placing Matrigel in a cylindrical shape. Compared to the conventional dome-shaped Matrigel where the Matrigel drop is located arbitrarily, a cylinder-shaped Matrigel position in confined central wells allowed for faster and cost-effective tumor organoid screening. Importantly, the cylinder-shaped Matrigel ensures better consistency in high-throughput image-based analysis, which is used worldwide. Our results demonstrate the possibility of replacing the conventional 24-, 48-, and 96-well plates with the newly developed plates for effective tumor organoid screening. [ABSTRACT FROM AUTHOR]

Abstract

While there a number of measures to assess work-family conflict already exist, there are no well-validated measures exist for clinical use with Chinese parents. This study sought to validate a Chinese version of the work and family conflict scale (WAFCS), a brief 10-item scale developed for clinical and research use with parents. Relying on a sample of 447 Chinese parents in Hong Kong, this study examined the psychometric properties of the Chinese version of the WAFCS. Results showed that a two-factor model, similar to that of the original scale, fit the data well. The scale had concurrent and discriminant validity, while the scale's measurement invariance across gender and its internal consistency were also supported. Such robust psychometric properties suggest that the WAFCS is a promising tool that can be applied in a variety of research and clinical settings to examine work-family conflict experienced by Chinese parents. 7. Do Mental Health Services Influence Child Welfare Involvement among Juvenile Justice System Involved Youth.

Abstract

There is limited understanding of mental health service utilization among youth in the juvenile justice system (JJS). Using administrative data, the current study examined service system trajectories of two cohorts of youth who initially entered the JJS in 2003 (N = 10,170) and 2012 (N = 5,787). We tracked mental health (MH) service utilization and dosage and child welfare system (CWS) involvement for 3 years and found that utilization and dosage of MH services increased between both cohorts. Notably, MH service use positively associated with CWS involvement and concurrent (dual) involvement in the CWS and JJS associated with decreased MH dosage if youth remained at home versus being placed in out-of-home care. Lastly, African American children received less services than their Caucasian peers. Future efforts should focus on preparing MH providers to detect youth who are dually involved and tailor services to enhance collaboration and information sharing across systems. Recent efforts have been devoted to understanding the conditions by which research evidence use (REU) is facilitated from the perspective of system leaders in the context of implementing evidence-based child mental health interventions. However, we have limited understanding of the extent to which outer contextual factors influence REU. Outer contextual factors for 37 counties in California were gathered from public records in 2008; and child welfare, juvenile justice, and mental health system leaders' perceptions of their REU were measured via a web-based survey from 2010 to 2012. Results showed that leaders with higher educational attainment and in counties with lower expenditures on inpatient mental health services were significantly associated with higher REU. Positive relationships between gathering research evidence and racial minority concentration and poverty at the county level were also detected. Results underscore the need to identify the organizational and socio-political factors by which mental health services and resources meet client demands that influence REU, and to recruit and retain providers with a graduate degree to negotiate work demands and interpret research evidence. Abstract

The etiology of poor mental health outcomes among foster care alumni of color remains largely unknown. We examined the influence of a subset of adverse childhood experiences (ACEs), placement instability, and childhood disability in uniquely predicting mental health outcomes among racially diverse foster care alumni from a private foster care agency. Multiple group path modeling was used to examine direct and indirect relationships between the individual level risk factors, mediators (perceived agency helpfulness), and past-year psychiatric disorder. Results showed that while the overall model was moderated by race/ethnicity, no significant group differences occurred in which ACEs increased the likelihood of psychiatric diagnoses. However, there were racial/ethnic differences in whether perceived agency helpfulness mediated the relationship between a subset of ACEs and diagnosis with a psychiatric disorder. Implications for future research to inform the development and implementation of effective services and supports that can promote positive developmental outcomes among racially diverse alumni are discussed. Abstract

Little is known about how the challenges faced by caregivers influence the variation in social, emotional, and behavioral (SEB) outcomes of youth placed in kinship versus non-relative foster care. This study examined SEB symptoms among youth in kinship and non-relative foster care settings, hypothesizing that changes in caregiver depression would modify children's change in behavior over time. Child Behavior Checklist (CBCL) assessments of 199 children placed with kinship and non-relative foster care providers in a Mid-Atlantic city were conducted at time of placement and 6-12 months post-placement. Linear regression estimated CBCL change scores for youth across placement type and caregiver depression trajectories. Kinship caregivers were more likely to become depressed or remained depressed than non-relative foster caregivers. Youth in kinship care always exhibited better change in SEB outcomes than youth in non-relative foster care, but these positive outcomes were principally observed among families where caregivers demonstrated a reduction in depression over time or were never depressed. Adjusted change scores for non-relative foster care youth were always negative, with the most negative scores among youth whose caregivers became depressed over time. Caregiver well-being may modify the influence of placement setting on SEB outcomes for youth placed into out-of-home care. Findings lend to policy relevance for child welfare systems that seek kinship settings as a panacea to the challenges faced by youth, without allocating resources to address caregiver needs. In this paper, we present the architecture of a sensor node for automatic stall management. Automatic stall management is emerging application fields of sensor network and our proposed sensor node acts as a key role in the applications. Our proposed sensor node consists of 8-bit microprocessor, temperature sensor and humidity sensor to acquire temperature and humidity data in real world. We design and implement our sensor node to according to the requirements of stall management environment. Through our experiments, we show that deviation of temperature and humidity of our sensor node is 5°C and 23.2% respectively, and the proposed sensor node is reliable in real applications.
