Prion disease is an infectious and fatal neurodegenerative disease. Human prion disease autopsy studies have revealed abnormal prion protein (PrPSc) deposits in the central nervous system and systemic organs. In deer, chronic wasting disease has also become a global problem, with PrPSc in saliva and feces. Therefore, understanding normal cellular prion proteins (PrPc) characteristics in human systemic organs is important since they could be a PrPSc source. This study used western blotting and immunohistochemistry to investigate endocrine and exocrine tissues, such as the human pituitary, adrenal, submandibular glands and the pancreas. All tissues had 30–40 kDa PrP signals, which is a slightly higher molecular weight than normal brain tissue. Most cytoplasmic PrP-positive adenohypophyseal cells were immunopositive for nuclear pituitary-specific positive transcription factor 1. The adrenal medulla and islet cells of the pancreas were PrP-positive and colocalized with chromogranin A. The duct epithelium in the submandibular gland and pancreas were immunopositive for PrP. This study reports the characteristic molecular properties and detailed tissue localization of PrPc in endocrine and exocrine tissues, which is important for infection control and diagnosis. Having glands that secrete milk to nourish neonatal offspring characterizes all mammals. We provide a brief overview of the development and anatomy of nipples and mammary glands in monotremes, marsupials, and marine mammals, and focus on the nipples and mammary glands in terrestrial eutherian species. We first classify eutherians into three groups: the altricial, precocial, and arboreal types based on their rearing system. We then summarize the physiology of lactation and the cell biology of nipples with specific focus on comparing these in the mouse, cow, and human, which represent the three different groups. Finally we propose that the nipple is an example of specialized epidermis. As specialized epidermis, it is dependent the underlying stroma for development and maintenance in adult life. Administration of 50 μg of bisphenol A (BPA) for the first 5 days after birth resulted in a decrease in the percentage of moving sperm, and an increase in the incidence of malformed sperm, in the epididymides of mice at 10 weeks of age, although no marked changes were found in the testicular histology between BPA-treated and vehicle-treated control mice. The deteriorating effects of 50 μg of BPA were ameliorated by the concurrent administration of 100 IU of retinol acetate (RA). Neonatal treatment with 0.5 μg of BPA for 5 days resulted in an increase in the incidence of malformed sperm, whereas the BPA effect became more severe in mice nursed by mothers fed a vitamin A-deficient (VAD) diet only a few days before and after parturition. On the other hand, neonatal treatment with 20 μg of estrogen for the first 5 days after birth resulted in an increase in the number of estrogen receptor α (ERα)-positive cells in the epithelium of the vas deferens, whereas only a few epithelial cells showed weak ERα-positive signals in the vehicle-treated control mice at 18 days after birth. This increase, however, was suppressed by the concurrent administration of RA. Although five daily treatments with 50 μg BPA led to no significant increase in the number of ERα-positive cells, it may have been due to the weak estrogenic activity of BPA, as discussed. These findings clearly showed that in mice, neonatal exposure to a relatively large dose of BPA causes damage to the motility and morphology of sperm, but the BPA effect is, to some extent, inhibited by a supplement of VA, and enhanced under a VAD condition. Male ddY mice were used to investigate the responses toward the odor of unfamiliar male mice. They were reared either in isolation from 3 weeks of age or in pairs. At 10 weeks of age, they went through a 4-day test period, in which the responses of the mice toward the odor of unfamiliar dominant and subordinate mice were investigated by differentiating the responses into the time to investigate the odor and the final decision of avoidance or preference. All the mice went through the encounter tests after this odor test in order to investigate the relationship between the responses toward the odor and the behavior toward the odor donor. The pair-reared dominants and the subordinates responded to the odor of the dominant and subordinate mice differentially. This indicated that they could discriminate the dominance status of unfamiliar mice by odor. Isolated mice did not respond to the odor of the dominant and subordinate mice differentially. Social experiences were concluded to be necessary to discriminate the dominance status of unfamiliar mice through odor. The length of the investigation time and the final decision of avoidance or preference did not correlate in pair-reared or isolated mice, and these 2 scales were concluded to be different. Male ddY mice were used to investigate the effect of a short period of isolation in adulthood on aggressive behavior. The relationship between the dominance status previous to isolation and the effect of isolation was investigated. The mice were kept in isolation for 3 weeks from 9 weeks of age, during which intruder tests were conducted once a week. They the went through an encounter test, in which the mice encountered dominant or subordinate mice in a neutral space. The number of the formerly-dominant isolated mice that attacked the intruder mice decreased at first and then increased. The latency to attack also lengthened at first and then shortened. Seven former-dominants continued to show aggression throughout the intruder The number of the formerly-subordinate isolated mice that attacked the intruder mice increased linearly. But 3 former-subordinates did not show aggression through the entire experiment. After 3 weeks isolation, the number of mice that showed aggression and the amount of aggression did not differ between the former-dominants and subordinatcs. Isolation housing was concluded to differentially affect the dominant and subordinate mice during the 3 weeks of isolation. It was also concluded to differentially affect the mice of absolute dominance and relative dominance differentially. Responses to the odor of familiar intruder male mice according to their dominance were investigated. Responses were classified into 2 aspects: the investigation of the odor and the decision-making regarding avoiding it or not. The results varied according to the dominance of the respondents and the odor donor, and also according to the context of previous encounter situations. The dominants that had attacked an intruder dominant mouse responded randomly to its odor, whereas the dominants that had fought with it tended to avoid the odor. The subordinates that had observed an intruder dominant mouse being attacked by its dominant cagemate preferred the passage with the intruder's odor. The odor of a subordinate mouse was neither avoided nor preferred by either the dominants or subordinates. It was suggested that mice distinguished the dominance of the odor donor regardless of the context of the previous encounter situation, but they responded differently according to it and also according to their own dominance status. In previous studies, isolated mice are found to be extremely more aggressive than group-reared mice. In most of those studies, however, the pairings of the group-reared mice were randomly determined, without attention to individuals' ranks in their home cages. In this study, the effect of isolation was analyzed by comparing isolated mice with dominants of group-reared mice. In experiment 1 the group-reared mice were randomly chosen as in previous studies. Isolated mice were significantly more aggressive, but the pairings of the group-reared mice were found to be all dominant vs. subordinate. In experiment 2, the group-reared dominants encountered unfamiliar dominants in an unfamiliar place. Dominant mice were found to be less aggressive than the isolated mice. This result was inconsistent with the 'territorial dominant hypothesis' on the hyperaggressiveness of the isolated mice. Sequential analysis of behavior clarified that the isolated mice had a sequential pattern quite different from the group-reared mice, and provided an illustrative figure for the 'escalation hypothesis'. The lack of aggression in the group-reared mice, however, left a question about the sequence similarity of aggressive behavior in isolaed and group-reared mice. [ABSTRACT FROM AUTHOR]

The hypothesis of dominant/territorial aggression on isolation-induced aggression was examined by comparing the behavior of isolated mice with that of familiar dominant mice. Familiar dominant mice showed as much aggression in an already known neutral space as the isolated mice. The behavioral sequences of aggression in the 2 groups showed both differences and similarities between each other. There was more variance in the aggressive behavior of the dominant mice. The differences were considered to be due to the effect of isolated housing, and the similarities were considered to be the change due to the occurrence of aggression. The temporal and spatial processing of pain perception in human was traced by magnetoencephalography (MEG). We applied a painful CO2 laser beam to the forearm of 11 normal subjects, and estimated the activated areas using a single equivalent current dipole (ECD) at each time point, and a brain electric source analysis (BESA) as a spatio-temporal multiple source analysis method. The four-source model was found to be the most appropriate; sources 1 and 2 at the secondary sensory cortex (SII) contralateral and ipsilateral to the stimulation, and sources 3 and 4 at the anterior medial temporal area (probably the amygdalar nuclei or hippocampal formation) contralateral and ipsilateral to the stimulation, respectively. Activities in all 4 areas were temporally overlapped. Activity in the primary sensory cortex (SI) contralateral to the stimulated site was not identified. Activity in the cingulate cortex was also not clearly identified. These results are probably due to one or more of the following factors; (1) the cingulate cortex is too deep, (2) the ECDs generated in the cingulate cortex are mainly oriented radially, and (3) the ECDs generated in bilateral hemispheres interfere with each other. No significant or consistent magnetic fields were recorded after 500 msec following the stimulation, probably due to the complicated spatial and temporal overlapping of activities in multiple areas. We investigated the activation of posterior parietal cortex (PPC) to somatosensory stimulation in humans to determine its fundamental role as a somatosensory associated area using magnetoencephalography (MEG). We studied somatosensory evoked magnetic fields (SEF) after stimulation of median nerve, posterior tibial nerve and lip, and analyzed them by the single dipole model and also by the multidipole model using brain electric source analysis (BESA) system. In single source model analysis, the dipole at the peak latency of short-latency components following each site stimulation were located in the corresponding receptive fields in the primary somatosensory cortex (SI) contralateral to the stimulation. The dipole at the peak latency of the middle latency components were located in bilateral upper bank of Sylvian fissure (SII). By contrast, in the five-dipole model of BESA, the equivalent current dipoles (ECDs) of the middle-latency SEF after stimulation of median nerve and posterior tibial nerve were identified in the contralateral SI and in the bilateral SII and PPC, while all activities of middle-latency SEF after lip stimulation appeared to be restricted in the contralateral SI and bilateral SII. Around 80 msec in latency, the ECD location in PPC after median nerve stimulation was, on the average, 2.4 cm posterior, 2.9 cm medial and 2.6 cm superior to the hand area in SI. The ECD in PPC after posterior tibial nerve stimulation was also located posterior to the foot area in SI, but it was close to the SI area of foot, their distance being approximately 1.3 cm. ECD in PPC was almost equally demonstrated in each hemisphere. These findings suggested that the somatosensory associated cortex in PPC represented somatotopic organization in parallel with ‘homunculus’ in SI, but the hand area was much wider than the foot area. We investigated the movement-related cortical fields (MRCFs) recorded by magnetoencephalography (MEG) to identify the motor and sensory brain activities at the instant of the unilateral finger movement using six normal subjects. We focused our investigation on the source analysis of the events tightly linked to movement onset, and we used brain electric source analysis (BESA) to model the sources generating MRCFs during the interval from 200 ms before to 150 ms after the movement onset. Four sources provided satisfactory solutions for MRCF activities in this interval. Sources 1 and 2, which were located in the pre-central regions in the hemisphere contralateral and ipsilateral to the moved finger, respectively, generated the readiness fields (RF), but source 1 was predominant just before movement onset. The motor field (MF), the peak of which was just after movement onset, was mainly generated by source 1. Sources 3 and 4 were located in the post-central regions in the hemisphere contralateral and ipsilateral to the moved finger, respectively. The first motor evoked field (MEF-I), the peak of which was about 80 ms after the movement, was mainly generated by source 3, but with the participation of sources 1, 2 and 4. The results indicated that the activities of both pre -and post-central regions in bilateral hemispheres were related to voluntary movements, although the predominant areas varied over time.
