Mpox virus (MPXV) can cause mpox in humans. Due to its quick and wide spread in the past two years, mpox has turned into a significant public health concern. Helicase E5 is a multi-domain protein; its primer synthesis and DNA unwinding activity are required for genome uncoating and DNA replication of MPXV. However, the in vitro DNA unwinding activity has never been demonstrated. Here, we report the structural and biochemical studies of MPXV E5, showing that the full-length protein adopts an auto-inhibited conformation. Truncation of the N-terminus can recover the in vitro unwinding activity of E5 towards the forked DNA. Further structural analysis reveals that MPXV E5 shares a conserved mechanism in DNA unwinding and primer synthesis with the homologous proteins. Vision-language models (VLM), such as Contrastive Language-Image Pretraining (CLIP), have demonstrated powerful capabilities in image classification under zero-shot settings. However, current zero-shot learning (ZSL) relies on manually tagged samples of known classes through supervised learning, resulting in a waste of labor costs and limitations on foreseeable classes in real-world applications. To address these challenges, we propose the mixup long-tail unsupervised (MLTU) approach for open-world ZSL problems. The proposed approach employs a novel long-tail mixup loss that integrated class-based re-weighting assignments with a given mixup factor for each mixed visual embedding. To mitigate the adverse impact over time, we adopt a noisy learning strategy to filter out samples that generated incorrect labels. We reproduce the unsupervised experiments of existing state-of-the-art long-tail and noisy learning approaches. Experimental results demonstrate that MLTU achieves significant improvements in classification compared to these proven existing approaches on public datasets. Moreover, it serves as a plug-and-play solution for amending previous assignments and enhancing unsupervised performance. With its high energy consumption and pollutant emissions, the iron and steel industry is a significant source of air pollution and carbon emissions in the Beijing–Tianjin–Hebei (BTH) region. To improve air quality and reduce greenhouse gas emissions, a series of policies involving ultra-low emission, synergistic reduction of pollution, and carbon application have been implemented in the region. This study has assessed air pollutant and CO2 emission patterns in the iron and steel industry of the region by employing co-control effects coordinate system, marginal abatement cost curve, and numerical modeling, along with the synergistic benefits of typical technologies. The results have demonstrated that: (1) the intensive production activities pertinent to iron and steel enterprises has contributed greatly to the emission in Tangshan and Handan, where the sintering process is the main source of SO2, NOx, PM2.5, and CO, accounting for 64.86%, 55.15%, 29.98%, and 46.43% of the total emissions, respectively. (2) Among the typical pollution control and reduction measures, industrial restructuring and adjustment of the energy-resource structure have led to the greatest effects on emission reduction. Technologies exhibiting great potential in emission reduction and high-cost efficiency such as Blast Furnace Top Gas Recovery Turbine Unit (TRT) need to be promoted. (3) In Tangshan city with the highest level of steel production, the iron and steel production activities contributed to the concentration of 30.51% of PM2.5, 50.67% of SO2, and 42.54% of NO2 during the non-heating period. During the heating period, pollutants pertinent to the combustion of fossil energy for heating have increased, while iron and steel induced emissions have decreased to 23.7%, 34.32%, and 29.13%, respectively. By 2030, it is speculated that the contribution of the iron and steel industry to air quality will be significantly decreased as result of successful implementation of ultra-low emission policies and typical synergistic reduction technologies. Large amount of ivory was excavated from Sanxingdui site which was waterlogged, severely degraded and in urgent need for conservation. There has been much effort for the conservation of waterlogged ivory by scientists. However, due to a lack of appropriate conservation material and the need to use non-destructive methods, no satisfactory results have been achieved previously. In this work, a novel formulated water-based Al–Si hybrid sol of size about 20 nm was prepared and introduced through a quasi-dynamic equilibrium method to waterlogged ivory tusk for the purpose of conservation. Good conservation performance could be achieved, since Al–Si sol gradually permeates into the interior of the ivory, distributes homogeneously and connects the loose components of ivory. Samples treated with appropriate amount of Al–Si sol displayed satisfactory compressive strength and porous intact structure. It was found that the fluidity of Al–Si sol had a significant influence on the conservation effect. Moreover, Al–Si sol not only consolidated HAP but also worked well on the soil embedded in unearthed ivory, which was beneficial to conserve ivory intactly. Slightly negatively charged Al–Si hybrid gel could interact with ivory matrix through multiple interactions including van der Waals force, electrostatic interaction, chemical and hydrogen bonding. This paper develops a process-level carbon emission calculation model for iron and steel enterprises through the carbon emission mechanism of the whole production process. The relationship between material, energy and carbon flows is considered and combined. The carbon emissions of enterprises are divided into industrial emissions and combustion emissions, and the indirect emissions of purchased intermediate products and electricity purchased from the grid are also considered. Carbon emission targets and corresponding emission reduction strategies are formulated at the enterprise and process levels. For example, consider an iron and steel enterprise. The different types of carbon emissions are accounted for, with their reduction potential analysed based on the carbon material flow analysis method. The results show that the carbon emission of this enterprise is 1930.87 kgCO2/t (CS), and the combustion emission caused by energy flow is the main contributor to the enterprise's carbon emission, accounting for 57.02% of the total emission. The carbon emission during iron-making accounts for 69.06% of the entire process and is critical in any carbon emission reduction of the enterprise. Among them, process emissions from the blast furnace process account for 81.79% of industrial emissions of the whole process, which is 356.51 kgCO2/t (CS), and is the main challenge of low carbon transformation in this extensive process. This study highlights that increasing the integrated steel-making scrap ratio and electric furnace steel production can break through the existing emission reduction limits. A 65.02% carbon emission reduction can be achieved, and using green electricity can reduce emissions by 24.15%. Reasonably determining the amount of purchased coke and paying attention to the high-value recycling of byproduct gas resources in the plant are essential to achieve low-carbon economic development of steel. Neuroblastoma is the most common paediatric solid tumour and prognosis remains poor for high-risk cases despite the use of multimodal treatment. Analysis of public drug sensitivity data showed neuroblastoma lines to be sensitive to indisulam, a molecular glue that selectively targets RNA splicing factor RBM39 for proteosomal degradation via DCAF15-E3-ubiquitin ligase. In neuroblastoma models, indisulam induces rapid loss of RBM39, accumulation of splicing errors and growth inhibition in a DCAF15-dependent manner. Integrative analysis of RNAseq and proteomics data highlight a distinct disruption to cell cycle and metabolism. Metabolic profiling demonstrates metabolome perturbations and mitochondrial dysfunction resulting from indisulam. Complete tumour regression without relapse was observed in both xenograft and the Th-MYCN transgenic model of neuroblastoma after indisulam treatment, with RBM39 loss, RNA splicing and metabolic changes confirmed in vivo. Our data show that dual-targeting of metabolism and RNA splicing with anticancer indisulam is a promising therapeutic approach for high-risk neuroblastoma. The prognosis of high-risk neuroblastoma is poor despite the availability of multimodal treatment. Based on a novel algorithm, known as the upper-layer-solution-aware (USA), a new algorithm, in which the penalty method is introduced into the empirical risk, is studied for training feed-forward neural networks in this paper, named as USA with penalty. Both theoretical analysis and numerical results show that it can control the magnitude of weights of the networks. Moreover, the deterministic theoretical analysis of the new algorithm is proved. The monotonicity of the empirical risk with penalty term is guaranteed in the training procedure. The weak and strong convergence results indicate that the gradient of the total error function with respect to weights tends to zero, and the weight sequence goes to a fixed point when the iterations approach positive infinity. This paper proposes a framework based on harmonic mean normalized Laplace-Beltrami spectral descriptor. A series of experiments show that the harmonic mean normalization has better performance for non-rigid 3D retrieval, and it is robust to holes, local scaling, noise and sampling. To better distinguish shapes with fine or rough details, weighting method and fusion method are also employed. Weighting method reduces the negative impact of high-frequency information, and fusion method combines multi-level spectral information in both low and high frequencies. Our approach has better performance than other state-of-the-art methods on both retrieval accuracy and time consumption for stretched non-rigid 3D shapes. [ABSTRACT FROM AUTHOR]

Ectonucleoside triphosphate diphosphohydrolase 3 (ENTPD3), a plasma membrane-bound metabolic enzyme, converts extracellular nucleotides into nucleosides. ENTPD3 is involved in various pathophysiological processes, including cellular adhesion, metabolism, activation, and migration. However, its specific function in the uterus remains unclear. This study aimed to investigate the expression pattern and localization of Entpd3 in the mouse uterus throughout the estrous cycle using immunohistochemistry (IHC), quantitative real-time PCR, and western blot analysis. The effect of sex steroid hormones on Entpd3 expression was also examined in ovariectomized (OVX) mice treated with 17β-estradiol (E2)/progesterone (P4) and estrogen receptor antagonist (Fulvestrant)/progesterone receptor antagonist (Mifepristone). Results demonstrated that elevated levels of Entpd3 mRNA and protein were noted during estrus and metestrus, with a decline in diestrus and proestrus. IHC revealed abundant ENTPD3 in the cytoplasm of glandular and luminal epithelial cells during estrus and metestrus. Additionally, treatment with E2 or P4 in OVX mice downregulated the expression of Entpd3 in the mouse uterus, which was rescued by Fulvestrant or Mifepristone. This study demonstrated that the expression of Entpd3 in the mouse uterus varied dynamically throughout the estrous cycle and was steroid-dependent, suggesting a potential role for Entpd3 in female mice’s reproductive function.
