The ESR International Forum at the ECR 2021 discussed effects of artificial intelligence on the future of radiology and the need for increased visibility of radiologists. The participating societies were invited to submit written reports detailing the current situation in their country or region. The European Society of Radiology (ESR) established the ESR International Forum in order to discuss hot topics in the profession of radiology with non-European radiological partner societies. At the ESR International Forum 2021, different strategies, initiatives and ideas were presented with regard to radiology community's response to the changes caused by the emerging AI technology. Detection of prostate-specific antigen (PSA) as a screening strategy for prostate cancer is limited by the inability of the PSA test to differentiate between malignant cancer and benign hyperplasia. Here, we report the use of a cancer-specific promoter, inhibition of differentiation-1 (Id1), to drive a dual-reporter system (Ad5/3-Id1-SEAP-Id1-mCherry) designed for detection of prostate cancer using a blood-based reporter-secreted embryonic alkaline phosphatase (SEAP) and tumor visualization using a fluorescent reporter protein, mCherry. In human prostate tumors, Id1 levels are correlated with increased Gleason grade and disease progression. To evaluate the performance of the dual-reporter system, a prostate cell panel with varying aggressive phenotypes was tested. Following infection with the Ad5/3-Id1-SEAP-Id1-mCherry vector, expression of the SEAP and mCherry reporters was shown to increase with increasing levels of cellular Id1. No correlation was observed between Id1 and PSA. To evaluate in vivo performance, flank tumors were grown in athymic male mice using three prostate cancer cell lines. Following intra-tumoral injection of the vector, tumors formed by cells with high Id1 had the greatest reporter expression. Interestingly, tumors with the lowest levels of Id1 and reporter expression produced the greatest amounts of PSA. These data support the use of Ad5/3-Id1-SEAP-Id1-mCherry as a predictor of prostate cancer malignancy and as a strategy for tumor localization. [ABSTRACT FROM AUTHOR]

In resource limited nations, cancer control is often a lower priority issue creating challenges for the prevention, early diagnosis, and treatment of cancer. Training and education are vital components of efforts to tackle this problem. A 3-day cancer control workshop was conducted at the Lagos State University Teaching Hospital (LASUTH), Nigeria, in 2013. The curriculum included didactic lectures, panel discussions, and interactive sessions on local cancer statistics, preventive strategies, cancer registries, screening and diagnostic options, and treatment approaches with limited resources (chemotherapy, radiotherapy, surgery, and palliative care) and several site-specific (breast, lung, cervical, prostate, and colon) topics. Pre-workshop and post-workshop questionnaires were completed by participants. Eighty-six percent of the 50 workshop participants completed at least one questionnaire. Participants were mainly nurses and physicians (89 % of responders), and 40 % reported >25 years of practice experience. The more common local needs identified were professional education (65 %) and increasing public cancer awareness (63 %). The greatest interest for future programs was on research collaborations (70 %). An immediate impact of the workshop was the commencement of monthly tumor board conferences and a review of the current cancer registry data. Capacity building is critical for the execution of effective cancer control strategies. Conducting collaborative workshops represents a cost-effective means of launching programs and energizing the medical community to pursue ongoing education and research addressing the anticipated cancer epidemic on the African continent. Desmoglein-2 (Dsg2) is a desmosomal cadherin that is aberrantly expressed in human skin carcinomas. In addition to its well-known role in mediating intercellular desmosomal adhesion, Dsg2 regulates mitogenic signaling that may promote cancer development and progression. However, the mechanisms by which Dsg2 activates these signaling pathways and the relative contribution of its signaling and adhesion functions in tumor progression are poorly understood. In this study we show that Dsg2 associates with caveolin-1 (Cav-1), the major protein of specialized membrane microdomains called caveolae, which functions in both membrane protein turnover and intracellular signaling. Sequence analysis revealed that Dsg2 contains a putative Cav-1-binding motif. A permeable competing peptide resembling the Cav-1 scaffolding domain bound to Dsg2, disrupted normal Dsg2 staining and interfered with the integrity of epithelial sheets in vitro. Additionally, we observed that Dsg2 is proteolytically processed; resulting in a 95-kDa ectodomain shed product and a 65-kDa membrane-spanning fragment, the latter of which localizes to lipid rafts along with full-length Dsg2. Disruption of lipid rafts shifted Dsg2 to the non-raft fractions, leading to the accumulation of these proteins. Interestingly, Dsg2 proteolytic products are elevated in vivo in skin tumors from transgenic mice overexpressing Dsg2. Collectively, these data are consistent with the possibility that accumulation of truncated Dsg2 protein interferes with desmosome assembly and/or maintenance to disrupt cell-cell adhesion. Furthermore, the association of Dsg2 with Cav-1 may provide a mechanism for regulating mitogenic signaling and modulating the cell-surface presentation of an important adhesion molecule, both of which could contribute to malignant transformation and tumor progression. Background:We investigated associations of known breast cancer risk factors with breast density, a well-established and very strong predictor of breast cancer risk.Methods:This nested case-control study included breast cancer-free women, 265 with high and 860 with low breast density. Women were required to be 40-80 years old and should have a body mass index (BMI) <35 at the time of the index mammogram. Information on covariates was obtained from annual questionnaires.Results:In the overall analysis, breast density was inversely associated with BMI at mammogram (P for trend<0.001), and parity (P for trend=0.02) and positively associated with alcohol consumption (ever vs never: odds ratio 2.0, 95% confidence interval 1.4-2.8). Alcohol consumption was positively associated with density, and the association was stronger in women with a family history of breast cancer (P<0.001) and in women with hormone replacement therapy (HRT) history (P<0.001). Parity was inversely associated with density in all subsets, except premenopausal women and women without a family history. The association of parity with density was stronger in women with HRT history (P<0.001).Conclusion:The associations of alcohol and parity with breast density appear to be in reverse direction, but stronger in women with a family history of breast cancer and women who ever used HRT. Results of the analysis of the stratospheric aircraft M-55 Geofizika measurements are presented. Estimates of the tropospheric air ratio near the thermal tropopause were conducted using the correlations between the water vapor and ozone values. The data of the measurements were used in the several aircraft campaigns held in the wintertime in the extratropical latitudes. The increased water vapor concentration was indicated during the flight on February 11, 2003 at the 12-km height which exceeds the averaged concentrations of water vapor in the extratropics by 1.5–2 times. The contribution of the tropospheric air near the thermal tropopause is quantitatively estimated. Traditional approaches to treating chronic neuropathic pain largely focus on manipulations directly altering neuronal activity or neuron-to-neuron communication. Recently, however, it has become clear that glial cells (including microglia and astroglia) play a significant role in pain expression in a variety of neuropathic pain models. Multiple aspects of the inflammatory response of glial cells, commonly observed in neuropathic pain conditions, have been implicated in pain expression. Thus, glial cell inflammation has emerged as a potential therapeutic target in neuropathic pain. Our laboratory has been exploring the use of an anti-inflammatory cytokine, interleukin-10 (IL-10), to control glial inflammatory activation thereby controlling neuropathic pain. IL-10 protein delivery is limited by a short half-life and an inability to cross into the central nervous system from the periphery, making a centrally delivered gene therapy approach attractive. We have recently characterized a non-viral gene therapy approach using two injections of naked DNA to achieve long-term (>3 months) control of neuropathic pain in a peripheral nerve injury model. Friction-stir processing (FSP) was used to modify surface microstructures, to enhance the bending of thick-plate 6061-T6 and 7050-T7451 aluminum alloys. Plates were bent at room temperature into a V-shaped die, to various angles. Bending performance in the friction-stir-processed plates was significantly better than that ill the base plates, where processing caused localized softening of the pretensile surface of the plate. A finite-element model of the plate-bending process was developed, to predict the bending limits of both the unprocessed base plates and of the friction-stir-processed plates. For the friction-stir-processed plates, the model employed a mesh divided into two or more zones; one zone was for unprocessed base material and other zones were for the processed material or for material that was affected by the heat of processing. The model used both the von Mises and the Latham and Cockroft criteria to predict bending limits. The bending-limit predictions were reasonably accurate, provided the gradient in true stress-strain behavior through the plate thickness was well characterized. [ABSTRACT FROM AUTHOR]

High-resolution array comparative genomic hybridisation (aCGH) analysis of DNA copy number aberrations (CNAs) was performed on breast carcinomas in premenopausal women from Western New York (WNY) and from Gomel, Belarus, an area exposed to fallout from the 1986 Chernobyl nuclear accident. Genomic DNA was isolated from 47 frozen tumour specimens from 42 patients and hybridised to arrays spotted with more than 3000 BAC clones. In all, 20 samples were from WNY and 27 were from Belarus. In total, 34 samples were primary tumours and 13 were lymph node metastases, including five matched pairs from Gomel. The average number of total CNAs per sample was 76 (range 35–134). We identified 152 CNAs (92 gains and 60 losses) occurring in more than 10% of the samples. The most common amplifications included gains at 8q13.2 (49%), at 1p21.1 (36%), and at 8q24.21 (36%). The most common deletions were at 1p36.22 (26%), at 17p13.2 (26%), and at 8p23.3 (23%). Belarussian tumours had more amplifications and fewer deletions than WNY breast cancers. HER2/neu negativity and younger age were also associated with a higher number of gains and fewer losses. In the five paired samples, we observed more discordant than concordant DNA changes. Unsupervised hierarchical cluster analysis revealed two distinct groups of tumours: one comprised predominantly of Belarussian carcinomas and the other largely consisting of WNY cases. In total, 50 CNAs occurred significantly more commonly in one cohort vs the other, and these included some candidate signature amplifications in the breast cancers in women exposed to significant radiation. In conclusion, our high-density aCGH study has revealed a large number of genetic aberrations in individual premenopausal breast cancer specimens, some of which had not been reported before. The effect of friction stir processing on the superplastic behavior of extruded Al-4Mg-1Zr was examined at 350 °C to 600 °C and at initial strain rates of 1 x 10-3 to 1 s-1. A combination of a fine grain size of 1.5 µm and high-angle grain boundaries in the friction stir-processed (FSP) alloy led to considerably enhanced superplastic ductility, much-reduced flow stress, and a shift to a higher optimum strain rate and lower optimum temperature. The as-extruded alloy exhibited the highest superplastic ductility of 1015 pct at 580 °C and an initial strain rate of 1 x 10-2s-1, whereas a maximum elongation of 1280 pct was obtained at 525 °C and an initial strain rate of 1 x 10-1s-1 for the FSP alloy. The FSP alloy exhibited enhanced superplastic deformation kinetics compared to that predicted by the constitutive relationship for superplasticity in fine-grained aluminum alloys. We give a brief description of the Clark Lake Multifrequency Radioheliograph which is presently used for solar research in the frequency range 20-125 MHz. We present two-dimensional images of the Sun's corona at several frequencies within this range, obtained with time resolution as good as 1.28 s. The shape of the Sun's outer corona as observed at meter-decameter wavelengths appears to change in time periods as short as a few seconds. [ABSTRACT FROM AUTHOR]

The Teepee Tee array of the Clark Lake Radio Observatory has been used to compare the flux of the Sun with that of the sidereal sources Tau A and Vir A at several frequencies in the range 109.0-19.0 MHz. Only the two central banks of the E-W arm of the array were used as elements of a phase switched interferometer so that the Sun could be observed as a point source and compared directly to the sidereal sources. The Sun was still partially resolved however, and appropriate corrections for this effect were made. The observations were taken at times when the Sun and either Tau A or Vir A were at the same declination. We have therefore been able to derive the values for the solar flux, without having to resort to a gain vs zenith distance correction. The observations, combined with those available in the literature, allow us to derive an accurate meter and decameter wavelength spectrum of the quiet Sun. [ABSTRACT FROM AUTHOR]

Forming limits in conventional sheet metal forming are given by strain levels obtainable prior to the onset of a localized neck or tear in the sheet. While the external appearance of such a neck is not observed in superplastic metals until strains become quite large, the formation of internal cavities could dictate the tolerable levels of strain in formed components. In this paper, these useful strain limits for a superplastic 7475 Al alloy have been explored. The approach used was to establish the influence of strain state (uniaxial, plane strain, and balanced biaxial) on the inception and growth characteristics of cavities and to correlate the extent of cavitation with material properties. Based on these data, it was then possible to establish strain states for which little or no loss in properties was observed, and thereby to define forming limits for superplastic forming this material. These results, coupled with comparisons against strains developed in actual parts as well as analytically predicted strains, show that a wide range of structural parts can be superplastically formed within the constraints of the recommended forming limits. It has been shown that the application of hydrostatic gas pressures during superplastic deformation of fine grained 7475 Al can entirely prevent the intergranular cavitation normally encountered at atmospheric pressure. A critical ratio of hydrostatic pressure to flow stress may be defined for each superplastic forming condition above which virtually no cavitation occurs. In superplastic deformation conditions where intergranular cavitation plays a significant part in final tensile rupture, the superplastic ductility may be improved by the application of hydrostatic pressures. Similarly, detrimental effects of large superplastic strains on service properties may be reduced or eliminated by the application of suitable hydrostatic pressures during superplastic forming. A method has previously been described whereby a fine and stable grain size may be achieved in conventional, heat-treatable aluminum alloy sheet by thermomechanical processing. The present work has examined the final recrystallization stage more closely. In particular, the effects of heating rate on recrystallized grain size have been determined and explained. It has been shown that heating rates greater than about 5 K . s should be employed in the final recrystallization stage in order to obtain maximum benefit from the fine grain processing technique. The coarser recrystallized grain sizes obtained with slower heating rates are mainly due to early activation of the most highly favored nucleation sites. Thermal recovery of the matrix defect structure below the recrystallization temperature is an additional, though less significant, effect. The influence of the degree of cold work and the volume fraction of insoluble particles on recrystallized grain size is discussed in relation to the heating rate. [ABSTRACT FROM AUTHOR]

A thermomechanical process for grain refinement in precipitation hardening aluminum alloys is reported. The process includes severe overaging, deformation, and recrystallization steps. Microstructural studies by optical and transmission electron microscopy of grain refinement in 7075 aluminum have revealed that precipitates formed during the overaging step create preferential nucleation sites for recrystallizing grains. The relationship between precipitate density following severe overaging and recrystallized grain density has been investigated; the results show that the localized deformation zones associated with particles larger than about 0.75 μ m can act at preferential nucleation sites for recrystallizing grains. The density of particles capable of producing nucleation sites for new grains is approximately ten times greater than the density of recrystallized grains. A close relationship between dislocation cell size after the deformation step and recrystallized grain density has also been established. Both quantities saturate for rolling reductions larger than approximately 85 pct. The grain size produced in 2.5 mm thick sheet by the optimum processing schedule is approximately 10 μm in longitudinal and long transverse directions and 6 μm in the short transverse direction. Operative creep mechanisms in laboratory melts of Ti-5Zr-0.5Si and Ti-5Al-5Zr-0.5Si have been investigated as a function of microstructure, creep stress, and temperature. From creep rate data and transmission electron microscopy results, it has been shown that an important creep strengthening mechanism at 811 K in Si bearing Ti alloys is clustering of solute atoms on dislocations. All of the alloys investigated showed anomalously high apparent activation energies and areas for creep, and a high exponent ( n) in the Dorn equation. In addition, the effect of heat treatment was investigated and it is shown that the highest creep strength was obtained by using a heat treatment which retained the maximum amount of silicon in solution. This is consistent with the proposed creep strengthening mechanism. An investigation of the creep behavior of several other Si containing alloys including two commercial alloys, Ti-11 and IMI-685 indicated similar results. [ABSTRACT FROM AUTHOR]

We investigated the ability of different doses and durations of exposure to the chemotherapeutic drugs 1-beta-D-arabinofuranosylcytosine (Ara-C), mitoxantrone (MTN), and paclitaxel (taxol, TXL) to induce internucleosomal DNA fragmentation and apoptosis in human acute myeloid leukemia (AML) HL-60 cells in suspension culture. At clinically achievable concentrations, all three drugs have been shown to induce apoptosis in HL-60 cells. An improved method was developed for the isolation of pure genomic DNA and the detection of drug-induced internucleosomal DNA fragmentation in < 1.0 microgram of DNA sample by agarose gel electrophoresis. Morphologic evidence for apoptosis was determined by light microscopy following Wright staining, and cell viability was assessed by trypan blue dye exclusion. Internucleosomal DNA fragmentation was observed following exposure to 1.0 microM Ara-C for 4 h, which increased with 10 and 50 microM Ara-C. Incubation with 100 microM Ara-C produced internucleosomal DNA fragmentation starting at 3 h, which increased with longer periods of exposure to Ara-C. Utilizing a schedule of 1-h exposure followed by 3-h suspension in drug-free medium, 0.25 microM MTN was found to initiate DNA fragmentation, which increased with exposure to 1.0 and 5.0 microM MTN. However, identical treatment with higher concentrations of MTN resulted in random DNA degradation. Alternatively, continuous exposure to 1.0 microM MTN for 3 h was necessary to initiate internucleosomal DNA fragmentation. This increased with exposure intervals of up to 6 h. Exposure to TXL concentrations as low as 0.01 microM for 24 h caused internucleosomal DNA fragmentation, which increased with dose escalation (0.05, 0.1, 0.5, and 1.0 microM) of TXL. Although continuous exposure to 1.0 microM TXL for a period as short as 8 h produced internucleosomal DNA fragmentation, this increased significantly with longer exposure intervals. In general there appears to be a threshold concentration and duration of exposure below which none of these three drugs activates endonucleolytic internucleosomal DNA fragmentation and apoptosis. This threshold is lower for the DNA-interactive drugs MTN and Ara-C but higher for the non-DNA-interactive drug TXL. Higher doses or prolonged treatments with the drugs produce random DNA fragmentation associated with necrotic cell death. These in vitro results may further improve our understanding of the antileukemic cytotoxic effects of these drugs, which may enable a more rational design of drug regimens for optimal treatment of AML.
