SIVA-1 plays a critical role in the induction of apoptosis in a number of different cell lines and participates in the mechanism of cisplatin (DDP)-mediated antitumor effects. However, the involvement of SIVA-1 in cisplatin resistance in gastric carcinoma has not been revealed. To explore the effect of SIVA-1 on DDP resistance, a recombinant pGV358-GFP-SIVA-1 lentiviral vector was constructed and transfected into human cisplatin-resistant MKN45/DDP gastric cancer cells. Subsequently, stable SIVA-1 overexpression was established in MKN45/DDP cells, which resulted in increased DDP sensitivity in MKN45/DDP cells in vitro. Flow cytometry demonstrated that SIVA-1 overexpression increased the percentage of apoptotic cells compared to that in the control. The colony formation assay clearly revealed that cell growth and proliferation were significantly suppressed following SIVA-1 overexpression. In addition, overexpression of SIVA-1 inhibited the migratory and invasive potential of MKN45/DDP cells in vitro. Western blot analysis indicated that SIVA-1 increased the expression levels of p53, p73, and p14ARF, whereas it reduced Bcl-2, MDM2, and Bcl-xL expression. In short, SIVA-1 upregulated the protein expression of p53, p73, and p14ARF and decreased that of Bcl-2, MDM2, and Bcl-xL in vitro and subsequently reversed cisplatin resistance in gastric cancer cells, suggesting that SIVA-1 serves as a valuable potential target for attenuating chemotherapy resistance. Yolk-shell nanostructures have recently attracted tremendous research interests in various areas because of their unique structural merits. Currently, there is an urgent need for developing porous shells with multifunctional features to enhance their performance in various applications. Herein, advanced yolk-shell nanospheres have been facilely prepared by encapsulating silver nanoparticles with porous Janus polymer shells that consist of a hypercrosslinked polystyrene (xPS) outer layer and a tethered poly(acrylic acid) (PAA) brush lining. The xPS outer layer possesses well-developed porosity as mass diffusion pathways. More importantly, the tethered PAA brushes with customized carboxyl groups exhibit great affinity toward specific species (e.g., cationic dyes), leading to their enrichment from the bulk solution into the interior cavity. The unusual combination of highly porous outer layers with customizable inbuilt polymer linings in the porous Janus shells endows them with great promise for synergistically catalytic degradation of cationic dyes. The permeation flux of pure graphene oxide (GO) membrane is low because of its narrow and variable interlayer spacing. To overcome this drawback, a multilayer membrane with sandwich structure was assembled alternately by GO and oxidized carbon nanotubes (OCNTs) on a polyacrylonitrile substrate via a layer-by-layer self-assembly technology. The role of polydimethyl diallyl ammonium chloride (PDDA) was to connect GO and OCNTs. The PDDA/GO/OCNTs multilayers (Mn) consisting of 3, 5, 7 and 9 bilayers were deposited. The performance of nanofiltration membranes can be optimized by adjusting the number of sandwich layers. As the assembled layers increased, obvious fluctuation and fold forms of membrane increased, which was conducive to increase separation performance of composite membrane. Rejection rate for Alphaurine A and water flux of M7 were 94.42% and 28.53 L m−2 h−1, respectively, at 3 bar. Meanwhile, M7 had higher flux compared with the reported literature in which rejection also reached up to 90%. Rejection rate of M3, M5, M7 and M9 for Alphaurine A remained 50.45, 87.7, 94.5 and 97.1%, respectively, after the 12-h filtration test. Thus, we thought that composite membrane with sandwich structure via layer-by-layer self-assembly technology of GO and OCNTs under the effect of PDDA had excellent stability and high flux. To improve the interfacial properties of carbon fibers/epoxy composites, we introduced a gradient interphase reinforced by graphene sheets between carbon fibers and matrix with a liquid phase deposition strategy. Interlaminar shear strength and flexural strength of the composites are both improved. The interfacial reinforcing mechanisms are explored by analyzing the structure of interfacial phase with linear scanning system of scanning electron microscope and atomic force microscope. Results indicate that carbon element shows a graded dispersion in the interface region and a gradient interface layer with the modulus decreasing from fibers and matrix is found to be built. To verify the effect of gradient interphase on the interfacial properties of composites, the mixture of carbon fiber/graphene/epoxy is sonicated before curing to disperse graphene sheets in matrix homogeneously. As a result, gradient interphase structures are disappeared and interfacial performance of composites is found to be weakened. Objective: To explore the effects of insulin-like growth factor-1 (IGF-1) on migration, proliferation and differentiation of mesenchymal stem cells (MSCs). Methods: MSCs were obtained from Sprague-Dawley rats by a combination of gradient centrifugation and cell culture techniques and treated with IGF-1 at concentrations of 5-20 ng/ml. Proliferation of MSCs was determined as the mean doubling time. Expression of CXC chemokine receptor 4 (CXCR4) and migration property were determined by flow cytometry and transwell migration essay, respectively. mRNA expression of GATA-4 and collagen II was determined by reverse transcription-polymerase chain reaction (RT-PCR). Results: The mean doubling time of MSC proliferation was decreased, and the expression of CXCR4 on MSCs and migration of MSCs were increased by IGF-1, all in a dose-dependent manner, while the optimal concentration of IGF-1 on proliferation and migration was different. IGF-1 did not affect the expression of GATA-4 or collagen II mRNA. Conclusions: IGF-1 dose-dependently stimulated the proliferation of MSCs, upregulated the expression of CXCR4, and accelerated migration. There was no apparent differentiation of MSCs to cardiomyocytes or chondrocytes after culturing with IGF-1 alone. [ABSTRACT FROM AUTHOR]

Liposarcoma is the most common primary retroperitoneal malignant neoplasm. However, preoperative diagnosis is a common problem due to lack of characteristic clinical presentations. It has been assumed that MRI, based on its high soft-tissue resolution, could discover and discern different subtypes of this tumor. Moreover, there has been little in the literature to compare the MRI features with pathological appearances of the retroperitoneal liposarcomas. We retrospectively analyzed 19 cases of retroperitoneal liposarcoma (11 males and 8 females, aged 41–79 years) proved surgically and histologically. All patients underwent MRI examination before and after the administration of contrast agent. The MRI features and postoperative pathological appearances were studied correlatively. Nine cases were located in the anterior pararenal space, four cases in the posterior pararenal space and six cases in the perirenal space. Among all cases, ten cases were present in the right retroperitoneum and nine cases in the left. The average diameter of the tumors was 14.7 cm (range from 7.5 cm to 26 cm). The MR signal intensity of liposarcoma was heterogeneous and varied greatly, depending on the components of the tumor and the different histological patterns. There are five subtypes of retroperitoneal liposarcoma. Myxoid liposarcoma ( n = 7) exhibited low signal intensity on T1W image and high signal intensity on T2W image. On histologic analysis, myxoid liposarcoma consists of a myxoid matrix as the predominant component and small amounts of mature fat. Enhanced solid tissues and thickened septa of myxoid liposarcoma were seen on post-contrast image. Well-differentiated liposarcoma ( n = 5) presented in high signal intensity on T1W images, intermediate signal intensity on T2W images, drop-out signal intensity on fat-suppressed MR images; The enhanced tenuous septa and solid tissues were seen on post-contrast image. Round-cell liposarcoma ( n = 2) and pleomorphic liposarcoma ( n = 2) exhibited soft-tissue tumor signal intensity without characteristic fat signal. One case of round-cell liposarcoma appeared with intratumoral hemorrhage and invaded the inferior vena cava. Dedifferentiated liposarcoma ( n = 3) exhibited small amounts of fatty components with a clear demarcation between fat and nonadipose solid tissue on MR images. One case of dedifferentiated liposarcoma invaded the kidney. MRI can clearly demonstrate shape, margin, internal components and surrounding tissues. Different subtypes of retroperitoneal liposarcoma exhibited varying MRI features, depending on tumor histological components. MRI should be an ideal method for diagnosing retroperitoneal liposarcoma.
