In several technical combustion systems for lean, premixed combustion, mixture stratification plays an important role, such as in stationary and aero gas turbines. The current paper focuses on a detailed characterization of a dual-swirl gas turbine model combustor operated in a stratified regime. The influence of mixture stratification on flame stabilization and self-induced thermo-acoustic oscillations was studied using laser and optical diagnostics in combination with microphone probes to measure pressure oscillations in the plenums and combustion chamber. The overall flame-shape was imaged using OH* chemiluminescence. Laser Raman scattering was applied to study the thermo-chemical state of the flame, the flow-field was measured using Particle Image Velocimetry and pressure oscillations in the combustion chamber and in the air plenums were determined by using calibrated microphone probes. OH planar laser-induced fluorescence was used to determine fluctuations of the hot gas with high spatial resolution. Significant mixture stratification within the combustion chamber was found to only occur at or upstream of the flame root. Varying the stratification level did not significantly influence the mean flame shape and flow field, however, a clear influence on thermo-acoustic oscillations was observed. [ABSTRACT FROM AUTHOR]

Large Eddy Simulations (LES) of a swirl-stabilized natural gas-air flame in a laboratory gas turbine combustor is performed using six different LES combustion models to provide a head-to-head comparative study. More specifically, six finite rate chemistry models, including the thickened flame model, the partially stirred reactor model, the approximate deconvolution model and the stochastic fields model have been studied. The LES predictions are compared against experimental data including velocity, temperature and major species concentrations measured using Particle Image Velocimetry (PIV), OH Planar Laser-Induced Fluorescence (OH-PLIF), OH chemiluminescence imaging and one-dimensional laser Raman scattering. Based on previous results a skeletal methane-air reaction mechanism based on the well-known Smooke and Giovangigli mechanism was used in this work. Two computational grids of about 7 and 56 million cells, respectively, are used to quantify the influence of grid resolution. The overall flow and flame structures appear similar for all LES combustion models studied and agree well with experimental still and video images. Takeno flame index and chemical explosives mode analysis suggest that the flame is premixed and resides within the thin reaction zone. The LES results show good agreement with the experimental data for the axial velocity, temperature and major species, but differences due to the choice of LES combustion model are observed and discussed. Furthermore, the intrinsic flame structure and the flame dynamics are similarly predicted by all LES combustion models examined. Introduction: Late-stage ovarian cancer patient's survival depends on complete cytoreduction and chemotherapy. Complete cytoreduction is more often achieved in institutions with a case volume of >20 cases per year. The Integrated care program Ovar (IgV Ovar) was founded in 2005 and started recruiting in 2006 with 21 health insurances and six expert centers of ovarian cancer treatment as a quality initiative. Results of the pilot and outcomes of patients of three participating centers will be presented here. Methods: Data of 1038 patients with ovarian cancer were collected. Adjuvant patients ( n = 505) stage FIGO IIB-IV ( n = 307) were analyzed for cytoreduction and survival. FIGO IIIC patients were analyzed separately. Results: Median follow-up was 32.7 months. Progression-free survival (PFS) was 23.1 months and overall survival (OS) was 53.6 months for stage IIB-IV. Patients with FIGO IIIC were completely cytoreduced in 48 %. PFS was 21, 29 months if completely cytoreduced. OS was 47.4, 64.9 months if completely cytoreduced. Discussion: Although the IgV Ovar Rhineland proved to have some structural problems with recruitment and prospective data collection, cytoreduction rates and outcome of patients prove treatment of patients in expert centers is superior to the national and international mean. Therefore, a new quality initiative will be started to bring more awareness to women and to their gynecologists and general practitioners of just how important a good referral strategy is. Weak spherical blast waves in static air and their breakup of ethanol and Jet A-1 kerosene droplets were investigated. The blast waves were created by laser-induced air breakdowns at ambient temperature and pressure. In the first part of this study, they were visualized with schlieren imaging, and their trajectories were tracked with high temporal resolution. The laser pulse energy was varied to create blast waves of different strengths. Their initial energies were determined by the application of a numerical and a semi-empirical blast wave model. In the second part, monodisperse ethanol and kerosene droplet chains were injected. Their interaction with the blast waves was visualized by the application of shadowgraph imaging. The perpendicular distance of the breakdown origin toward the droplet chains was varied to study the effect on the fuel droplets as a function of the distance. Droplets within a few millimeters around the breakdown origin were disintegrated into two to three secondary droplets. The blast-induced flow velocities on the post-shock side and the corresponding Weber numbers were calculated from the data of a non-dimensional numerical simulation, and a close look was taken at the breakup process of the droplets. The analysis showed that the aerodynamic force of the blast-induced flow was sufficient to deform the droplets into disk-like shapes, but diminished too fast to accomplish breakup. Tomographic particle image velocimetry (tomographic-PIV) is a recently developed measurement technique used to acquire volumetric velocity field data in liquid and gaseous flows. The technique relies on line-of-sight reconstruction of the rays between a 3D particle distribution and a multi-camera imaging system. In a turbulent flame, however, index-of-refraction variations resulting from local heat-release may inhibit reconstruction and thereby render the technique infeasible. The objective of this study was to test the efficacy of tomographic-PIV in a turbulent flame. An additional goal was to determine the feasibility of acquiring usable tomographic-PIV measurements in a turbulent flame at multi-kHz acquisition rates with current-generation laser and camera technology. To this end, a setup consisting of four complementary metal oxide semiconductor cameras and a dual-cavity Nd:YAG laser was implemented to test the technique in a lifted turbulent jet flame. While the cameras were capable of kHz-rate image acquisition, the laser operated at a pulse repetition rate of only 10 Hz. However, use of this laser allowed exploration of the required pulse energy and thus power for a kHz-rate system. The imaged region was 29 × 28 × 2.7 mm in size. The tomographic reconstruction of the 3D particle distributions was accomplished using the multiplicative algebraic reconstruction technique. The results indicate that volumetric velocimetry via tomographic-PIV is feasible with pulse energies of 25 mJ, which is within the capability of current-generation kHz-rate diode-pumped solid-state lasers. A planar solid oxide fuel cell (SOFC) operated with hydrogen at T = 1,123 K was equipped with an optically transparent anode flow field to apply species concentration measurements by 1D laser Raman scattering. The flow channels had a cross section of 3 mm × 4 mm and a length of 40 mm. The beam from a pulsed high-power frequency-doubled Nd:YAG laser ( λ = 532 nm) was directed through one channel and the Raman-scattered light from different molecular species was imaged onto an intensified CCD camera. The main goal of the study was an assessment of the potential of this experimental configuration for a quantitative determination of local gas concentrations. The paper describes the configuration of the optically accessible SOFC, the laser system and optical setup for 1D Raman spectroscopy as well as the challenges associated with the measurements. The auto-ignition of a pulsed methane jet issuing into a laminar coflow of hot exhaust products of a lean premixed hydrogen/air flat flame was examined using high-speed laser and optical measurement techniques with frame rates of 5 kHz or more. OH* chemiluminescence was used to determine the downstream location of the first auto-ignition kernel as well as the stabilization height of the steady-state lifted jet flame. OH planar laser-induced fluorescence (PLIF) was used to determine further details of the auto-ignition with a higher spatial resolution. Simultaneous imaging of broadband luminosity from a viewing angle perpendicular to the OH* chemiluminescence was applied, to three-dimensionally reconstruct the ignition kernel location in space and to determine whether the first occurrence of the kernel was within or beyond the PLIF laser sheet. The development and expansion of the jet was characterized by high-speed Schlieren imaging. Statistics have been compiled for both the ignition time as well as the downstream location of the first auto-ignition kernel and the stabilization height of the steady-state lifted jet flame. From the PLIF images it was found that auto-ignition tended to occur at the interface between bulges of the inflowing jet and the coflow. For steady-state conditions, auto-ignition kernels were observed frequently below the flame base, emphasizing that the lifted jet flame is stabilized by auto-ignition. [ABSTRACT FROM AUTHOR]

Silver nanoparticles (SNPs) are among the most commercialized nanoparticles worldwide. Often SNP are used because of their antibacterial properties. Besides that they possess unique optic and catalytic features, making them highly interesting for the creation of novel and advanced functional materials. Despite its widespread use only little data exist in terms of possible adverse effects of SNP on human health. Conventional synthesis routes usually yield products of varying quality and property. It thus may become puzzling to compare biological data from different studies due to the great variety in sizes, coatings or shapes of the particles applied. Here, we applied a novel synthesis approach to obtain SNP of well-defined colloidal and structural properties. Being stabilized by a covalently linked small peptide, these particles are nicely homogenous, with narrow size distribution, and form monodisperse suspensions in aqueous solutions. We applied these peptide-coated SNP in two different sizes of 20 or 40 nm (Ag20Pep and Ag40Pep) and analyzed responses of THP-1-derived human macrophages while being exposed against these particles. Gold nanoparticles of similar size and coating (Au20Pep) were used for comparison. The cytotoxicity of particles was assessed by WST-1 and LDH assays, and the uptake into the cells was confirmed via transmission electron microscopy. In summary, our data demonstrate that this novel type of SNP is well suited to serve as model system for nanoparticles to be tested in toxicological studies in vitro. [ABSTRACT FROM AUTHOR]

The absorption of OH chemiluminescence and laser-induced fluorescence (LIF) in the exhaust gas of confined premixed laminar CH/air flames at atmospheric pressure was investigated. One flame was used as source and a second as absorber. OH LIF was excited in the ν″=0→ ν′=1 band of the A-X electronic system around ≈283 nm and spectrally resolved detected in the (0,0) and (1,1) vibrational bands around 305-320 nm. For OH chemiluminescence, spectrally resolved detection was performed in the wavelength range 280-340 nm. For an absorption path of 54 mm and at T≈2000 K, signal trapping on the order of 10-40% was observed. Signal trapping was most pronounced in the (0,0) band, as expected from the thermal population distribution of OH in the electronic ground state. The spectral distribution of the signals and the wavelength dependence of the signal trapping are addressed in this paper. Mixture fraction measurements in a jet-in-cross flow configuration at high pressures (15 bar) and temperatures (above 1000 K) were performed using planar laser induced fluorescence of nitric oxide (NO-PLIF) as trace species. The goal was the evaluation of this tracer LIF technique for the characterization of the mixing of fuel and hot exhaust gas in the mixing channel. The fuel (natural gas (NG) or H/N/NG mixture) along with the tracer were injected into the crossflow of the exhaust gas and PLIF measurements were performed in different planes. In order to relate the measured NO-LIF signal to fuel concentration and mixture fraction, effects of pressure, temperature and species concentration were taken into account. Numerical calculations and spectroscopic simulations that mimic the experimental conditions were performed to identify excitation schemes that give optimum correlations between the NO-LIF signal and the mixture fraction. The measured NO-PLIF images were transformed into mixture fraction plots using the computed correlations. The paper reports on the experimental challenges encountered during the measurements and the steps taken to overcome those difficulties. The Arctic cryosphere is a critically important component of the earth system, affecting the energy balance, atmospheric and ocean circulation, freshwater storage, sea level, the storage, and release of large quantities of greenhouse gases, economy, infrastructure, health, and indigenous and non-indigenous livelihoods, culture and identity. Currently, components of the Arctic cryosphere are subjected to dramatic change due to global warming. The need to document, understand, project, and respond to changes in the cryosphere and their consequences stimulated a comprehensive international assessment called 'SWIPA': Snow, Water, Ice, Permafrost in the Arctic. Some of the extensive key SWIPA chapters have been summarized and made more widely available to a global audience with multi-disciplinary interests in this Special Report of Ambio. In this article, an overview is provided of this Special Report in the context of the more detailed and wider scope of the SWIPA Report. Accelerated changes in major components of the Arctic cryosphere are documented. Evidence of feedback mechanisms between the cryosphere and other parts of the climate system are identified as contributing factors to enhanced Arctic warming while the growing importance of Arctic land-based ice as a contributor to global sea-level rise is quantified. Cryospheric changes will result in multifaceted and cascading effects for people within and beyond the Arctic presenting both challenges and opportunities. [ABSTRACT FROM AUTHOR]

The relation between flow field and flame structure of a turbulent swirl flame is investigated using simultaneous particle image velocimetry (PIV) and planar laser-induced fluorescence of OH (OH-PLIF). The measurements are performed in one axial and three transverse sections through the combustion chamber of a gas turbine model combustor, which is operated with methane and air under atmospheric pressure. Analysis of the velocity fields using proper orthogonal decomposition (POD) shows that the dominant unsteady flow structure is a so-called precessing vortex core (PVC). In each of the four sections, the PVC is represented by a characteristic pair of POD eigenmodes, and the phase angle of the precession can be determined for each instantaneous velocity field from its projection on this pair. Phase-conditioned averages of velocity field and OH distribution are thereby obtained and reveal a pronounced effect of the PVC in the form of convection-enhanced mixing. The increased mixing causes a rapid ignition of the fresh gas, and the swirling motion of the PVC leads to an enlarged flame surface due to flame roll-up. A three-dimensional representation shows that the PVC is accompanied by a co-precessing vortex in the outer shear layer, which, however, has no direct impact on the flame. As an alternative to phase averaging, a low-order representation of the phase-resolved dynamics is calculated based on the first pair of POD modes. It is found that small-scale structures are represented more accurately in the phase averages, whereas the low-order model has a considerable smoothing effect and therefore provides less detailed information. Computational fluid dynamics (CFD) modelers require high-quality experimental data sets for validation of their numerical tools. Preferred features for numerical simulations of a sooting, turbulent test case flame are simplicity (no pilot flame), well-defined boundary conditions, and sufficient soot production. This paper proposes a non-premixed CH/air turbulent jet flame to fill this role and presents an extensive database for soot model validation. The sooting turbulent jet flame has a total visible flame length of approximately 400 mm and a fuel-jet Reynolds number of 10,000. The flame has a measured lift-off height of 26 mm which acts as a sensitive marker for CFD model validation, while this novel compiled experimental database of soot properties, temperature and velocity maps are useful for the validation of kinetic soot models and numerical flame simulations. Due to the relatively simple burner design which produces a flame with sufficient soot concentration while meeting modelers' needs with respect to boundary conditions and flame specifications as well as the present lack of a sooting 'standard flame', this flame is suggested as a new reference turbulent sooting flame. The flame characterization presented here involved a variety of optical diagnostics including quantitative 2D laser-induced incandescence (2D-LII), shifted-vibrational coherent anti-Stokes Raman spectroscopy (SV-CARS), and particle image velocimetry (PIV). Producing an accurate and comprehensive characterization of a transient sooting flame was challenging and required optimization of these diagnostics. In this respect, we present the first simultaneous, instantaneous PIV, and LII measurements in a heavily sooting flame environment. Simultaneous soot and flow field measurements can provide new insights into the interaction between a turbulent vortex and flame chemistry, especially since soot structures in turbulent flames are known to be small and often treated in a statistical manner. Background: We assess the prognostic value of chemotherapy-induced leukopenia and sensory neuropathy in the CALYPSO trial patients treated with carboplatin-paclitaxel (CP) or carboplatin-liposomal doxorubicin (CPLD).Methods: We performed a landmark analysis at first month after randomisation to correlate leukopenia (nadir white blood cell <4.0 × 10(9) per litre or severe infection) during cycle 1 of chemotherapy with progression-free survival (PFS). Using time-dependent proportional-hazards models, we also investigated the association between neuropathy and PFS.Results: Of 608 patients with nadir blood and did not receive growth factors, 72% (CP=70%, CPLD=73%) had leukopenia. Leukopenia was prognostic for PFS in those receiving CP (adjusted hazard ratio (aHR) 0.66, P=0.01). Carboplatin-liposomal doxorubicin was more effective than CP in patients without leukopenia (aHR 0.51, P=0.001), but not those experiencing leukopenia (aHR 0.93, P=0.54; interaction P=0.008).Of 949 patients, 32% (CP=62%, CPLD=28%) reported neuropathy during landmark. Neuropathy was prognostic for PFS in the CP group only (aHR 0.77, P=0.02). Carboplatin-liposomal doxorubicin appeared to be more effective than CP among patients without neuropathy (aHR 0.70, P<0.0001), but not those with neuropathy (aHR 0.96, P=0.81; interaction P=0.15).Conclusion: First-cycle leukopenia and neuropathy were prognostic for patients treated with CP. Efficacy of CP treatment was similar to CPLD in patients who developed leukopenia. These findings support further research to understand the mechanisms of treatment-related toxicity. We describe a newly developed combustion diagnostic for the simultaneous planar imaging of soot structure and velocity fields in a highly sooting, lifted turbulent jet flame at 3000 frames per second, or two orders of magnitude faster than 'conventional' laser imaging systems. This diagnostic uses short pulse duration (8 ns), frequency-doubled, diode-pumped solid state (DPSS) lasers to excite laser-induced incandescence (LII) at 3 kHz, which is then imaged onto a high framerate CMOS camera. A second (dual-cavity) DPSS laser and CMOS camera form the basis of a particle image velocity (PIV) system used to acquire 2-component velocity field in the flame. The LII response curve (measured in a laminar propane diffusion flame) is presented and the combined diagnostics then applied in a heavily sooting lifted turbulent jet flame. Dynamic processes in gas turbine (GT) combustors play a key role in flame stabilization and extinction, combustion instabilities and pollutant formation, and present a challenge for experimental as well as numerical investigations. These phenomena were investigated in two gas turbine model combustors for premixed and partially premixed CH/air swirl flames at atmospheric pressure. Optical access through large quartz windows enabled the application of laser Raman scattering, planar laser-induced fluorescence (PLIF) of OH, particle image velocimetry (PIV) at repetition rates up to 10 kHz and the simultaneous application of OH PLIF and PIV at a repetition rate of 5 kHz. Effects of unmixedness and reaction progress in lean premixed GT flames were revealed and quantified by Raman scattering. In a thermo-acoustically unstable flame, the cyclic variation in mixture fraction and its role for the feedback mechanism of the instability are addressed. In a partially premixed oscillating swirl flame, the cyclic variations of the heat release and the flow field were characterized by chemiluminescence imaging and PIV, respectively. Using phase-correlated Raman scattering measurements, significant phase-dependent variations of the mixture fraction and fuel distributions were revealed. The flame structures and the shape of the reaction zones were visualized by planar imaging of OH distribution. The simultaneous OH PLIF/PIV high-speed measurements revealed the time history of the flow field-flame interaction and demonstrated the development of a local flame extinction event. Anionic copolymerization and Williamson reaction of PS- co-PD ( d-isoprene (I) or butadiene (B)) prepolymers (bearing hydroxyl or benzyl bromide end groups) and ethylene oxide or mono-methyl poly(ethylene glycol) (PEGs) were used to prepare a series of PS- co-PD- b-PEO amphiphilic copolymers. Investigations on the association and self-assembly of copolymers in dilute organic and in mixed organic/water solutions have been carried out both by light scattering and microscopic measurements. Nanosized and microsized species have been observed. Their shape depends on the hydrophobic/hydrophilic blocks ratio as well as on the solvent composition. Attempts on stabilizing the morphology of the aggregates/micelles have been made by UV-induced cross-linking of diene entities. It has been found that in some experiments, the stabilization proceeds throughout morphological rearrangement determined by the solvent nature and by the cross-linking protocol. A swirling methane-air diffusion flame at atmospheric pressure is stabilized in a gas turbine model combustor with good optical access. The investigated flame with a thermal power of 10 kW and an overall equivalence ratio of 0.75 exhibits pronounced thermoacoustic oscillations at a frequency of 295 Hz. The main goal of the presented work is a detailed experimental characterization of the flame behavior in order to better understand the flame stabilization mechanism and the feedback loop of thermoacoustic instability. OH* chemiluminescence imaging is applied for determining the flame shape and estimating the heat release rate. Laser Raman scattering is used for simultaneous detection of the major species concentrations, mixture fractions, and temperature. The velocity fields are measured by particle image velocimetry (PIV) or stereo PIV, simultaneously with OH planar laser-induced fluorescence. The dynamic pressure in the combustion chamber is determined by microphone probes. The flow-field exhibits a conically shaped inflow of fresh gases and inner and outer recirculation zones. The instantaneous flame structures are dominated by turbulent fluctuations; however, phase-correlated measurements reveal phase-dependent changes in all measured quantities. The paper presents examples of measured results, characterizes the main features of the flame behavior, explains the feedback loop of the oscillation, and discusses the flame stabilization mechanism. The accuracy of laser-induced incandescence (LII) measurements is significantly influenced by the calibration process and the laser profile degradation due to beam steering. Additionally, the wavelength used for extinction measurements, needed for LII calibration, is critical and should be kept as high as possible in order to avoid light absorption by molecular species in the flame. The influence of beam steering on the LII measurement was studied in turbulent sooting C2H4/air flames at different pressures. While inhomogeneities in the laser profile become smoothed out in time-averaged measurements, especially at higher pressure, the corresponding single-shot beam profiles reveal an increasing effect of beam steering. In the current configuration it was observed that the resulting local laser fluence remains within certain limits (30% to 200%) of the original value. A sufficiently high incident laser fluence can thus prevent the local fluence from dropping below the LII threshold value of approximately 0.3 J/cm2 at the cost of increased soot surface vaporization. A spatial resolution in the dimension of the sheet thickness of below 1 mm cannot be guaranteed at increased pressure of 9 bars due to beam steering. A feasibility study in a combustor at technical conditions demonstrates the influence of both effects beam steering and choice of calibration wavelength and led to the conclusion that, however, a shot-to-shot calibration of LII with simultaneously measured extinction can be realized. We describe an approach of imaging the dynamic interaction of the flamefront and flowfield. Here, a diode-pumped Nd:YLF laser operating at 5 kHz is used to pump a dye laser, which is then frequency doubled to 283 nm to probe flamefront OH, while a dual cavity diode-pumped Nd:YAG system produces pulse-pairs for particle image velocimetry (PIV). CMOS digital cameras are used to detect both planar laser-induced fluorescence (PLIF) and particle scattering (in a stereo arrangement) such that a 5 kHz measurement frequency is attained. This diagnostic is demonstrated in lifted-jet and swirl-stabilized flames, wherein the dynamics of the flame stabilization processes are seen. Nonperiodic effects such as local ignition and/or extinction, lift-off and flashback events, and their histories can be captured by this technique. As such, this system has the potential to significantly extend our understanding of nonstationary combustion processes relevant to industrial and technical applications. [ABSTRACT FROM AUTHOR]

An experimental setup for the generation and investigation of periodic equivalence ratio oscillations in laminar premixed flames is presented. A special low-pressure burner was developed which generates stable flames in a wide pressure range down to 20 mbar and provides the possibility of rapid mixture fraction variations. The technical realization of the mixture fraction variations and the characteristics of the burner are described. 1D laser Raman scattering was applied to determine the temperature and concentration profiles of the major species through the flame front in correlation to the phase-angle of the periodic oscillation. OH* chemiluminescence was detected to qualitatively analyze the response of the flame to mixture fraction variations by changing shape and position. Exemplary results from a flame at p=69 mbar, forced at a frequency of 10 Hz, are shown and discussed. The experiments are part of a cooperative research project including the development of kinetic models and numerical simulation tools with the aim of a better understanding and prediction of periodic combustion instabilities in gas turbines. In highly turbulent environments, combustion is strongly influenced by the effects of turbulence chemistry interactions. Simultaneous measurement of the flow field and flame is, therefore, obligatory for a clear understanding of the underlying mechanisms. In the current studies simultaneous PIV and OH-PLIF measurements were conducted in an enclosed gas turbine model combustor for investigating the influence of turbulence on local flame characteristics. The swirling CH4/air flame that was investigated had a thermal power of 10.3 kW with an overall equivalence ratio of ϕ=0.75 and exhibited strong thermoacoustic oscillations at a frequency of approximately 295 Hz. The measurements reveal the formation of reaction zones at regions where hot burned gas from the recirculation zones mixes with the fresh fuel/air mixture at the nozzle exit. However, this does not seem to be a steady phenomenon as there always exist regions where the mixture has failed to ignite, possibly due to the high local strain rates present, resulting in small residence time available for a successful kinetic runaway to take place. The time averaged PIV images showed flow fields typical of enclosed swirl burners, namely a big inner recirculation zone and a small outer recirculation zone. However, the instantaneous images show the existence of small vortical structures close to the shear layers. These small vortical structures are seen playing a vital role in the formation and destruction of reaction zone structures. One does not see a smooth laminar flame front in the instantaneous OH-PLIF images, instead isolated regions of ignition and extinction highlighting the strong interplay between turbulence and chemical reactions. The understanding of periodic flame instabilities belongs to the major challenges in modern combustion research and technology and is of special importance for lean premixed gas turbine combustion. This paper presents experimental investigations in a gas turbine model combustor using laser diagnostic techniques. A partially premixed CH4/air flame operated at a thermal power of 10 kW at atmospheric pressure and an overall equivalence ratio of 0.75, which exhibited thermoacoustic oscillations at a frequency of 290 Hz, was investigated. Phase-locked laser Raman scattering was applied in order to determine the major species concentrations, temperature, and mixture fraction. In addition, laser Doppler velocimetry (LDV) was used separately for the measurement of the axial and radial velocity components. The measurements revealed pronounced phase-dependent variations of the velocity and the temperature, species, and mixture fraction distributions. The combined Raman and LDV results also enabled the determination of molecular species fluxes which showed that the fuel and air supply rates both varied during an oscillation cycle by ±33% but with a phase shift of 80 between them. The correlations between temperature and mixture fraction revealed strong deviations from equilibrium composition and temperature, and their phase-dependent changes reflected the transport and mixing processes near the nozzle. The emphasis of the paper lies on the demonstration of the potential of phase-locked laser Raman scattering for the study of phenomena of periodic flame instabilities. [ABSTRACT FROM AUTHOR]

Self-assembly of an amphiphilic triblock copolymer carrying polymerizable end-groups is used to prepare nanometer-sized vesicular structures in aqueous solution. The triblock copolymer shells of the vesicles can be regarded as a mimetic of biological membranes although they are 2 to 3 times thicker than a conventional lipid bilayer. Nevertheless, they can serve as a matrix for membrane-spanning proteins. Surprisingly, the proteins remain functional despite the extreme thickness of the membranes and that even after polymerization of the reactive triblock copolymers. This opens a new field to create mechanically stable protein/polymer hybrid membranes. As a representative example we functionalize (polymerized) triblock copolymer vesicles by reconstituting a channel-forming protein from the outer cell wall of Gram-negative bacteria. The protein used (OmpF) acts as a size-selective filter, which allows only for passage of molecules with a molecular weight below 400 g mol-1. Therefore substrates may still have access to enzymes encapsulated in such protein/polymer hybrid nanocontainers. We demonstrate this using the enzyme β-lactamase which is able to hydrolyze the antibiotic ampicillin. In addition, a transmembrane voltage above a given threshold causes a reversible gating transition of OmpF. This can be used to reversibly activate or deactivate the resulting nanoreactors. The nonmonotonic variation of the critical percolation temperature ( Tc) of ternary nonionic (C14E5) water-in-oil microemulsions was studied as a function of the alkyl chain length of an ionic additive ( n-alkyl sulfonate sodium salt). A thermodynamic approach shows the relationship between Tc and additive chain length, which is supplemented by a consideration of a possible molecular mechanism of the observed phenomenon. Abstract. A standard burner for confined swirling natural gas flames is presented which was developed within the German TECFLAM cooperation. The aims of the TECFLAM research program are the establishment of an extensive experimental database from selective flames and the validation and improvement of mathematical combustion models. In this paper, results from joint PDF measurements of temperature, mixture fraction, and major species concentrations in a turbulent diffusion flame with 150 kW thermal load, equivalence ratio 0.833, and swirl number 0.9 are presented. Major aspects of the investigation are the general quantitative characterization of the flame and the study of the thermochemical state, e.g. effects of turbulence-chemistry interactions. Scatterplots of temperature, CH[sub 4], and CO mole fractions as well as mean mixture fraction and temperature fields are presented and discussed. Furthermore, CFD calculations have been performed using the code Fluent 5 as an example of a commercially available code that is frequently used for technical applications. The comparison between the calculated and measured results reveals some significant deviations which are discussed with respect to the applicability of this code to swirling turbulent flames. Abstract The oscillating drop surfactometer (ODS) measures surface tension (7) and energy dissipation (damping constant b) of surfactant on a 1 micro l sample. gamma is obtained from the period of oscillation and b from its free decay or from the phase shift slope in resonance. After calibration with substances with different gamma, corrections were made for capillary fixation and loss of mass by evaporation. Surface active substances are delivered from liposomes in the interior (subphase) or injected from outside, with microdrops (180 pl each) of solution. As an application example, we have investigated surfactant extract and pure phospholipid. In minutes after formation of a drop containing a diluted Survanta suspension, gamma decreases by 20 mN/m, while b increases three-fold. This effect, assigned to spontaneous adsorption from liposomes to the surface, is not seen with pure dipalmitoylphosphatidylcholine (DPPC) under our conditions. However, microdrop injection of DPPC triggers a rapid decrease of gamma and a delayed strong increase in b. The effect is modulated by DPPC in the subphase and by cholesterol. Investigations with L-alpha-lysophosphatidylcholine show the high sensitivity of the ODS technique in the determination of the energy dissipation at air-liquid boundary surfaces. Although the ODS is limited to applications with gamma > 15 mN m[sup -1], it offers the advantage to give, with small samples and within seconds, a simultaneous readout of both surface tension gamma and the parameter b, as a measure of surface viscosity. Abstract. A single-pulse spontaneous Raman scattering apparatus was used to determine the temperature and major species concentrations in a turbulent CH[sub 4]/H[sub 2]/N[sub 2] diffusion flame. Important aspects of the measuring technique, such as accuracy, cross talk between different Raman bands, and the correction procedure for background from laser-induced fluorescence are discussed. In addition, a 2D LIF and Rayleigh imaging system were used to study the structures of OH, CH, NO, and temperature distributions in the flame. A comparison between two different CH detection schemes is presented. A main goal of the investigations was a detailed and accurate characterization of the investigated flame as well as the study of experimental techniques. Joint pdfs of the temperature and major species concentrations were determined at nearly 100 measuring locations covering the complete flame. Parts of the results are presented in the paper in order to discuss effects of differential diffusion, flame extinction, and interaction between flow field and chemistry. Further investigations about the role of the mitochondria-rich cell (MR cell) in hormone-mediated transport regulation in the epithelium of frog skin brought the following results: Unlike toad bladder, in frog skin the spontaneous potential difference cannot be reversed when Na transport is blocked. A similar situation is obtained when in addition to transport-blockade, one applies a chemical gradient for chloride to the epithelium. Under these conditions we found that in the intact preparation as well as in the separated epithelium: (i) the reversed current (RC) is linearly related to the number of MR cells; (ii) RC is mainly carried by a passive, transcellular chloride flux inwards and (iii) RC is sensitive to nor-adrenaline (10 m). The beta-blocker propranolol abolishes this effect. We propose that the MR cells are the sites of transepithelial shunt-path and that this chloride flux is transcellular. As it is hormone sensitive, it could be an important regulatory instrument for the regulation of overall salt transport (internal shorting). [ABSTRACT FROM AUTHOR]

Unlabelled: Monitoring of therapy-related late effects after acute lymphoblastic leukaemia (ALL) therapy in childhood has become an increasingly important field in posttherapeutic patient surveillance. The usefulness of neurophysiological investigations (e.g. EEG, evoked potentials (EP)) as part of these attempts is controversial. The present report focuses on this problem and the question whether and to what extent routinely performed EEG recordings and visual evoked potentials (VEP) were correlated with further measures of CNS integrity. EEGs and VEPs were recorded in 163 asymptomatic long-term survivors of ALL in childhood during a large retrospective multicentre study evaluating CNS late sequelae following antileukaemic therapy. Fifty-two ALL long-term survivors (4.5-10.6 years after end of therapy, median: 8.8 years), who had been treated according to BFM-81 SR-A (n = 30) or SR-B (n = 22) were selected for this analysis focusing on therapy-related CNS late effects. Therapy protocols differed with regard to the mode for CNS prophylaxis: SR-A, cranial irradiation with intrathecal methotrexate; SR-B, intrathecal and iv methotrexate. Neurophysiological findings were correlated with illness- and treatment-related parameters, as well as with data on the morphological, neurological and psychological status of the CNS. At the time of follow-up neurophysiological measures were abnormal in 28/52 cases (53.8%). Neither illness- nor therapy-specific differences in CNS prophylaxis showed any relationship to EEG/VEP outcome any relationship to EEG/VEP outcome in this reduced group of the whole study population. Children with EEG/VEP abnormalities showed a significantly higher incidence of structural CNS disturbances compared to those with inconspicuous neurophysiological recordings (60.9% vs 31.8%). However, in this special subject group there was no specific neurophysiological finding for a specific morphological substrate, neurological or psychological deficiency and vice versa.Conclusion: Routinely performed EEG/VEP investigations are not very helpful measures to predict the presence or degree of behavioural deficiencies, neurological disturbances, or morphological CNS abnormalities. Patients who received cranial irradiation or systemic methotrexate applications showed the same incidence of neurophysiological disturbances without evidence for specific neurotoxic correlates. The detection of vitellogenin, a yolk precursor protein, may serve as a biomarker for exposure to environmental oestrogens as its induction by xenobiotic oestrogens in the immature and male fish has been reported repeatedly. In the present work, juvenile brown trout were injected with oestradiol (5 μg g−1 body weight oestradiol benzoate) in order to assess the induction and organ distribution of vitellogenin by means of immunohistochemistry. In addition, brown trout collected from Swiss rivers were analysed. Vitellogenin was detected in the oestradiol-injected juvenile trout but not in uninjected controls. The presence of vitellogenin was also demonstrated in a male and an immature feral brown trout from one of two locations downstream of three sewage treatment plants. In contrast, no positive staining was found in livers of trout upstream of the respective plants. The results demonstrate the suitability of immunohistochemistry for monitoring feral fish fo r the presence of vitellogenin production. [ABSTRACT FROM AUTHOR]

The droplet phase of a water-in-oil microemulsion is used as a matrix to create amphiphilic polymer network structures from a poly(oxyethylene)-poly(isoprene)-poly(oxyethylene) triblock copolymer bearing polymerizable methacrylate end groups. Conductivity measurements and freeze fracture electron microscopy are used to obtain information about the structure of these gels. The droplet structure of the underlying microemulsion is only preserved for low droplet and polymer concentration. ABA-triblock copolymers bearing polymerizable methacrylate end-groups are covalently crosslinked in the droplet phase of W/O- and O/W-microemulsions. The resulting microemulsion elastomers combine the structure and the phase behavior of microemulsions with solid state properties such as elasticity or stability of shape. These new materials are characterized with the help of conductivity, dynamic-mechanical and dynamic light-scattering investigations. N Q-branch CARS spectra have been recorded and evaluated for temperature determination in a turbulent, premixed CH/air stagnation flame with a burner of 40 mm diameter and 22 kW thermal load. Temperature histograms on the flame axis at different distances from the stagnation plate have been measured. Problems of practical applicability are addressed, including those arising from the limited spatial resolution of the BOXCARS geometry, from an insufficient dynamic range of the diode array detector, and from a memory effect of the detector in the case of measurements in highly turbulent flame areas with strong intermittency. Some information is given on the computerized acquisition and on the evaluation of the large amounts of data that are necessary for extensive investigations in large combustion systems. This is the first report on a primary culture of dispersed skin epidermal cells of rainbow trout Oncorhynchus mykiss Walbaum. These primary cells revealed a low seeding efficiency after 3 days (11.6 ± 4.6%), whereas subcultured cells had a higher seeding efficiency at the same time point (75.5 ± 34.0%) and increased in cell number (150 – 200% of seeded cells after 20 to 30 days). The cells were characterized applying histological, immunocytochemical and ultrastructural methods. The culture consisted of undifferentiated keratinocytes. Mucous cells as well as differentiated epithelial cells were absent. To date the cells were cultured for maximally 9 passages and 402 days and therefore provide the possibility for long-term studies. The time dependence of small elastic extensional RBC deformation by micropipette aspiration has been analyzed. This process shows two-phases which are characterized by time constants of the order of some tenths of seconds and about ten seconds, respectively. The equilibrium tongue length is reached after about 30 s. For the first, fast step we assume that the membrane model of immobilized boundaries holds, i.e., the skeleton is tightly associated with the lipid bilayer and no redistribution of the skeleton with respect to the lipid bilayer is allowed. This lipid-spectrin interaction or anchorage is characterized by some association force density. It has been shown that at a given tongue length the force generated owing to the membrane deformation and acting to redistribute the spectrin, overcomes (in some membrane area) the association force density and results in an additional increase of the sucked membrane length. Equations have been derived to describe this process. From the experimental conditions of an RBC aspiration and the determined tongue length corresponding to the second slow aspiration step, the association force density between the lipid bilayer and the spectrin network may be determined.
