We aimed to explore novel biomarker candidates and biomarker signatures of late-onset preeclampsia (LOPE) by profiling samples collected in a longitudinal discovery cohort with a high-throughput proteomics platform. Using the Somalogic 5000-plex platform, we analyzed proteins in plasma samples collected at three visits (gestational weeks (GW) 12–19, 20–26 and 28–34 in 35 women with LOPE (birth ≥ 34 GW) and 70 healthy pregnant women). To identify biomarker signatures, we combined Elastic Net with Stability Selection for stable variable selection and validated their predictive performance in a validation cohort. The biomarker signature with the highest predictive performance (AUC 0.88 (95% CI 0.85–0.97)) was identified in the last trimester of pregnancy (GW 28–34) and included the Fatty acid amid hydrolase 2 (FAAH2), HtrA serine peptidase 1 (HTRA1) and Interleukin-17 receptor C (IL17RC) together with sFLT1 and maternal age, BMI and nulliparity. Our biomarker signature showed increased or similar predictive performance to the sFLT1/PGF-ratio within our data set, and we were able to validate the biomarker signature in a validation cohort (AUC ≥ 0.90). Further validation of these candidates should be performed using another protein quantification platform in an independent cohort where the negative and positive predictive values can be validly calculated. Purpose: The mechanisms that control inflammation in scrub typhus are not fully elucidated. The Notch pathways are important regulators of inflammation and infection, but have not been investigated in scrub typhus. Methods: Plasma levels of the canonical Notch ligand Delta-like protein 1 (DLL1) were measured by enzyme immunoassay and RNA expression of the Notch receptors (NOTCH1, NOTCH2 and NOTCH4) in whole blood was analyzed by real-time PCR in patients with scrub typhus (n = 129), in patients with similar febrile illness without O. tsutsugamushi infection (n = 31) and in healthy controls (n = 31); all from the same area of South India. Results: Our main results were: (i) plasma DLL1 was markedly increased in scrub typhus patients at hospital admission with a significant decrease during recovery. (ii) RNA expression of NOTCH4 was decreased at admission in whole blood. (iii) A similar pattern for DLL1 and NOTCH4 was seen in febrile disease controls. (iv) Admission DLL1 in plasma was associated with disease severity and short-term survival. (vi) Regulation of Notch pathways in O. tsutsugamushi-infected monocytes as evaluated by public repository data revealed enhanced canonical Notch activation with upregulation of DLL1 and downregulation of NOTCH4. Conclusion: Our findings suggest that scrub typhus patients are characterized by enhanced canonical Notch activation. Background and aims: Home-field advantage (HFA) in litter decomposition occurs widely in terrestrial ecosystems, but it remains unknown that how leaf, absorptive and transport roots differ in their HFA. Here we conducted a 635-day litter transplant (leaves, absorptive roots, and transport roots) decomposition experiment, using three tree species (Cyclobalanopsis multinervis, Fagus lucida, Cryptomeria japonica), in a subtropical forest to explore their differences in HFA effects. Results: The mass loss of leaves was higher than that of absorptive and transport roots. The mass loss of fine roots differed among species. Leaves were more prone to exhibit significant HFA compared to absorptive and transport roots, and absorptive roots were more prone to exhibit significant HFA compared to transport roots. The HFA effect was not explained by the initial chemical characteristics of litter except C concentration. Conclusions: These findings suggest that HFA effect can occur in labile and recalcitrant litters and are generally not regulated by the initial chemical characteristics. Absorptive and transport roots exhibit different HFA patterns, highlighting the need to identify the effects of HFA on root decomposition by functional classification rather than diameter class to gain a better understanding of fine root decomposition. Dominant microorganisms of the Sargasso Sea are key drivers of the global carbon cycle. However, associated viruses that shape microbial community structure and function are not well characterised. Here, we combined short and long read sequencing to survey Sargasso Sea phage communities in virus- and cellular fractions at viral maximum (80 m) and mesopelagic (200 m) depths. We identified 2,301 Sargasso Sea phage populations from 186 genera. Over half of the phage populations identified here lacked representation in global ocean viral metagenomes, whilst 177 of the 186 identified genera lacked representation in genomic databases of phage isolates. Viral fraction and cell-associated viral communities were decoupled, indicating viral turnover occurred across periods longer than the sampling period of three days. Inclusion of long-read data was critical for capturing the breadth of viral diversity. Phage isolates that infect the dominant bacterial taxa Prochlorococcus and Pelagibacter, usually regarded as cosmopolitan and abundant, were poorly represented. Psoriatic arthritis (PsA) carries a severe disease burden, often leading to deterioration of health-related quality of life (HRQoL). Different comorbidities that are relatively prevalent in PsA are also responsible for compromised HRQoL. To assess real-world data of a 5-year follow-up cohort of PsA patients, focusing on changes in general HRQoL, skin HRQoL, and comorbidities. In this prospective observational study, 114 outpatients diagnosed with PsA were examined at baseline and after 5 years. Data collection included demographics, clinical disease activity measures, and patient-reported outcome measures (PROMs). General HRQoL was assessed with a 15D instrument, and skin HRQoL was assessed with the Dermatology Life Quality Index (DLQI). During the 5-year follow-up, no significant deterioration in HRQoL assessed by 15D (23.53 vs. 23.08, p = 0.85) and DLQI (3.48 vs. 2.68, p = 0.07) was observed. There was no observed decline in other PROMs. The mean total number of comorbidities increased (1.13 vs. 1.39, p < 0.01). A significant improvement in disease activity measures, including 66/68 swollen/tender joint count, Disease Activity Index for Psoriatic Arthritis (all p < 0.01), and Psoriatic Arthritis Severity Index (p = 0.04) was seen. A higher proportion of patients at 5 years were treated with b/tsDMARDs (37.7% vs. 46.5%, p = 0.03). Despite an increased number of comorbidities over 5 years, our PsA cohort showed no decline in HRQoL. This can be attributed to the widespread adoption of modern treatments, leading to improved disease control and the preservation of baseline HRQoL. [ABSTRACT FROM AUTHOR]

Normal pressure hydrocephalus (NPH) leads to cognitive impairment (CI) as part of its triad of symptoms. Assessment of CI before and after a tap test can be used to inform diagnosis, differentiate NPH symptoms from alternative or concomitant causes of CI, and suggest the potential benefits of valve placement. The aims of this study are: to describe cognitive performance in patients with NPH diagnosis before and after a tap test, and to compare CI between patients with NPH meeting criteria for a dementia diagnosis (D +) and those without criteria for dementia (D−) at both baseline and after the tap-test. We performed a Before-and-after study evaluating clinical features and performance on cognitive tests (CERAD, ADAS-COG, SVF, PVF, ROCF and IFS). We included 76 NPH patients, with a median age of 81 years. 65 patients (87.8%) improved cognitive performance after tap test. ROCF (p = 0.018) and IFS (p < 0.001) scores significantly change after the tap test. Dementia was concomitant in 68.4% of patients. D + group showed higher proportion of patients with altered performance in IFS, PVF, SVF, and ROCF than D- group at baseline (p < 0.05). A significant improvement in SVF and IFS was observed exclusively in the D− group after tap test. Our results suggest that executive function and praxis are the cognitive domains more susceptible to improvement after a tap test in a 24-h interval in NPH patients. This article introduces a special issue of the Hydrogeology Journal that focuses on the hydrogeology of arid environments. Arid regions, which cover a significant portion of the Earth's land surface, face challenges in accessing freshwater due to limited surface-water resources. As a result, groundwater resources play a crucial role in providing freshwater in these areas. However, overexploitation of groundwater reserves in many arid countries has become a concern, impacting health, food security, and political stability. The article emphasizes the importance of sustainable groundwater management and the need for accurate data and research to make informed decisions. The special issue includes various articles that explore topics such as groundwater extraction in Jordan, hydraulic properties of volcanic rocks in Ethiopia, groundwater recharge mechanisms in Nigeria, and shared groundwater resources in the Sahel region. The research presented in this special issue contributes to a better understanding of the hydrogeology of arid environments and can inform strategies for managing groundwater resources in these regions. Terrestrial animals are key elements in the cycling of elements in the Arctic where nutrient availability is low. Waste production by herbivores, in particular urine deposition, has a crucial role for nitrogen (N) recycling, still, it remains largely unexplored. Also, experimental evidence is biased toward short-term studies and Arctic regions under high herbivore pressure. In this study, we aimed to examine the fate of N derived from urine in a nutrient poor tundra heath in West Greenland, with historical low level of herbivory. We performed a pulse labelling with 15N-urea over the plant canopy and explored ecosystem N partition and retention in the short-term (2 weeks and 1 year) and longer-term (5 years). We found that all vascular plants, irrespective of their traits, could rapidly take up N-urea, but mosses and lichens were even more efficient. Total 15N enrichment was severely reduced for all plants 5 years after tracer addition, with the exception of cryptogams, indicating that non-vascular plants constituted a long-term sink of 15N-urea. The 15N recovery was also high in the litter suggesting high N immobilization in this layer, potentially delaying the nutrients from urine entering the soil compartment. Long-term 15N recovery in soil microbial biomass was minimal, but as much as 30% of added 15N remained in the non-microbial fraction after 5 years. Our results demonstrate that tundra plants that have evolved under low herbivory pressure are well adapted to quickly take advantage of labile urea, with urine having only a transient effect on soil nutrient availability. The physical process in which a substance moves from a location with a higher concentration to a location with a lower concentration is known as molecular diffusion. It plays a crucial role during the mixing process between different gases in porous media. Due to the petrophysical properties of the porous medium, the diffusion process occurs slower than in bulk, and the overall process is also affected by thermodynamic conditions. The complexity of measuring gas–gas diffusion in porous media at increased pressure and temperature resulted in significant gaps in data availability for modelling this process. Therefore, correlations for ambient conditions and simplified diffusivity models have been used for modelling purposes. In this study, correlations in dependency of petrophysical and thermodynamic properties were developed based on more than 30 measurements of the molecular diffusion of the binary system hydrogen–methane in gas storage rock samples at typical subsurface conditions. It allows reproducing the laboratory observations by evaluating the bulk diffusion coefficient and the tortuosity factor with relative errors of less than 50 % with minor exceptions, leading to a strong improvement compared to existing correlations. The developed correlation was implemented in the open-source simulator DuMux and the implementation was validated by reproducing the measurement results. The validated implementation in DuMux allows to model scenarios such as Underground Hydrogen Storage (UHS) on a field-scale and, as a result, can be used to estimate the temporary loss of hydrogen into the cushion gas and the purity of withdrawn gas due to the gas–gas mixing process. This study investigates chronic conditions (CC) prevalence among children in mainstream schools, their school experience and life satisfaction in Europe. Data were collected from the 2017/2018 HBSC survey, a cross-national study using self-reported questionnaires administered in classrooms. Nationally representative samples of children aged 11, 13, and 15 years in mainstream schools from 19 European countries (n = 104,812) were used. School experience was assessed using four variables: low school satisfaction, schoolwork pressure, low teacher support, and peer-victimization, which were related to life satisfaction. Latent class analysis (LCA) was conducted to identify patterns of school experience among students with CC. The prevalence of CC varied from 8.4 (Armenia) to 28.2% (Finland). Children with CC (n = 17,514) rated their school experience and life satisfaction lower than children without CC. LCA identified three school experience patterns: "negative on all items" (37%), "negative on all items, except school pressure" (40%) and "overall positive" (23%). The distribution of subgroups varied across countries—in countries with a higher proportion of children with CC in mainstream schools, children reported more negative school experiences. Compared to the "overall positive" group, low life satisfaction was highest for students classified as "negative on all items" (relative risk (RR) = 2.9; 95% CI 2.2–3.8) with a lesser effect for "negative on all items, except school pressure" (RR) = 1.8; 95% CI 1.4–2.4). These findings provide cross-national data documenting the diversity in inclusive educational practices regarding school placement and school experiences, and suggest that efforts are still needed to allow a fully inclusive environment. Purpose: Triglycerides (TG) and their major transport lipoprotein in the circulation (VLDL) appear to be related to inflammation. Patients with common variable immunodeficiency (CVID) have inflammatory complications associated with gut microbial dysbiosis. We hypothesized that CVID patients have disturbed TG/VLDL profiles associated with these clinical characteristics. Methods: We measured plasma concentrations of TGs, inflammatory markers, and lipopolysaccharide (LPS) in 95 CVID patients and 28 healthy controls. Additionally, in 40 CVID patients, we explored plasma lipoprotein profiling, fatty acid, gut microbial dysbiosis, and diet. Results: TG levels were increased in CVID patients as compared to healthy controls (1.36 ± 0.53 mmol/l versus 1.08 ± 0.56 [mean, SD], respectively, P = 0.008), particularly in the clinical subgroup "Complications," characterized by autoimmunity and organ-specific inflammation, compared to "Infection only" (1.41 mmol/l, 0.71[median, IQR] versus [1.02 mmol/l, 0.50], P = 0.021). Lipoprotein profile analyses showed increased levels of all sizes of VLDL particles in CVID patients compared to controls. TG levels correlated positively with CRP (rho = 0.256, P = 0.015), IL-6 (rho = 0.237, P = 0.021), IL-12 (rho = 0.265, P = 0.009), LPS (r = 0.654, P = 6.59 × 10−13), CVID-specific gut dysbiosis index (r = 0.315, P = 0.048), and inversely with a favorable fatty acid profile (docosahexaenoic acid [rho = − 0.369, P = 0.021] and linoleic acid [rho = − 0.375, P = 0.019]). TGs and VLDL lipids did not appear to be associated with diet and there were no differences in body mass index (BMI) between CVID patients and controls. Conclusion: We found increased plasma levels of TGs and all sizes of VLDL particles, which were associated with systemic inflammation, LPS, and gut dysbiosis in CVID, but not diet or BMI. Aims: This study aimed at elucidating divergent effects of two dominant plant functional types (PFTs) in tundra heath, dwarf shrubs and mosses, on soil microbial processes and soil carbon (C) and nutrient availability, and thereby to enhance our understanding of the complex interactions between PFTs, soil microbes and soil functioning. Methods: Samples of organic soil were collected under three dwarf shrub species (of distinct mycorrhizal association and life form) and three moss species in early and late growing season. We analysed soil C and nutrient pools, extracellular enzyme activities and phospholipid fatty acid profiles, together with a range of plant traits, soil and abiotic site characteristics. Results: Shrub soils were characterised by high microbial biomass C and phosphorus and phosphatase activity, which was linked with a fungal-dominated microbial community, while moss soils were characterised by high soil nitrogen availability, peptidase and peroxidase activity associated with a bacterial-dominated microbial community. The variation in soil microbial community structure was explained by mycorrhizal association, root morphology, litter and soil organic matter quality and soil pH-value. Furthermore, we found that the seasonal variation in microbial biomass and enzyme activities over the growing season, likely driven by plant belowground C allocation, was most pronounced under the tallest shrub Betula nana. Conclusion: Our study demonstrates a close coupling of PFTs with soil microbial communities, microbial decomposition processes and soil nutrient availability in tundra heath, which suggests potential strong impacts of global change-induced shifts in plant community composition on carbon and nutrient cycling in high-latitude ecosystems. Background and aims: Fine-root (diameter ≤ 2 mm) decomposition contributes significantly to nutrient cycling in terrestrial ecosystems. Roots with arbuscular mycorrhizas (AM) and ectomycorrhizas (ECM) differ in root chemistry, which might affect root decomposition rate, but whether this effect differs across forest biomes is unknown. Methods: We used a compiled dataset from temperate and (sub)tropical forests (168 species from 84 studies) to investigate how root chemistry and climate influence fine-root decomposition with different mycorrhizal associations in (sub)tropical and temperate forests. Results: We show that AM trees exhibited faster fine-root decomposition rates than ECM trees did in temperate forests, but not in (sub)tropical forests. In temperate forests, root decomposition rates decreased with increasing root lignin concentrations, and ECM trees had higher root lignin concentrations than AM trees did which likely caused the difference in their decomposition rates. In (sub)tropical forests, root decomposition rates were mainly determined by root phosphorus (P) concentration. ECM and AM trees had similar root P concentrations, and thus their root decomposition rates were similar. In addition, the root decomposition rate was not affected by climate in (sub)tropical or temperate forests. However, the root decomposition rates of both AM and ECM trees were similarly affected by root chemistry and climate for all forests. Conclusions: The findings will help us better predict tree species effects on ecosystem processes by considering their mycorrhizal association, particularly in temperate forests, where AM and ECM species co-occur. Purpose: About 20–30% of patients with common variable immunodeficiency (CVID) develop granulomatous-lymphocytic interstitial lung disease (GLILD) as one of several non-infectious complications to their immunodeficiency. The purpose of this study was to identify biomarkers that could distinguish GLILD from other non-infectious complications in CVID. Methods: We analyzed serum biomarkers related to inflammation, pulmonary epithelium injury, fibrogenesis, and extracellular matrix (ECM) remodeling, and compared three subgroups of CVID: GLILD patients (n = 16), patients with other non-infectious complications (n = 37), and patients with infections only (n = 20). Results: We found that GLILD patients had higher levels of sCD25, sTIM-3, IFN-γ, and TNF, reflecting T cell activation and exhaustion, compared to both CVID patients with other inflammatory complications and CVID with infections only. GLILD patients also had higher levels of SP-D and CC16, proteins related to pulmonary epithelium injury, as well as the ECM remodeling marker MMP-7, than patients with other non-infectious complications. Conclusion: GLILD patients have elevated serum markers of T cell activation and exhaustion, pulmonary epithelium injury, and ECM remodeling, pointing to potentially important pathways in GLILD pathogenesis, novel targets for therapy, and promising biomarkers for clinical evaluation of these patients. Introduction: We aimed to compare demographics and clinical characteristics between patients with inflammatory arthritis (IA) with vs. without neutralizing anti-drug antibodies (nADAb) against tumor necrosis factor inhibitors (TNFi). A secondary aim of the study was to explore if current smokers were more frequently nADAb-positive. Methods: TNFi-treated outpatients with IA were recruited and a broad range of disease activity measures were assessed. nADAb were assessed using a reporter gene assay. Comparisons between nADAb-positive and -negative patients were done in unadjusted analyses as well as in adjusted logistic regression and general linear models. Results: A total of 282 patients with IA currently under treatment with TNFi were included. nADAb were identified in 11 patients (nine treated with infliximab, one with etanercept and one with certolizumab pegol). Patients with nADAb reported significantly worse joint pain, patient's global assessment, Health Assessment Questionnaire, Bath Ankylosing Spondylitis Disease Activity/Functional Index and Short-Form-36 physical functioning scale score than patients without nADAb (p < 0.04, adjusted analyses). 28-joint Disease Activity Score, Simplified Disease Activity Index and Maastricht Ankylosing Spondylitis Enthesitis score were also significantly worse in the nADAb-positive patients (p < 0.04, adjusted analyses), as were serum calprotectin, C-reactive protein and numbers of circulating peripheral leukocytes (p ≤ 0.001). A significantly higher proportion of nADAb-positive patients were current smokers (46 vs. 15%), in unadjusted as well as adjusted analyses (p ≤ 0.008). Conclusions: nADAb-positive patients were more frequently smokers and had significantly worse disease activity, physical function, and inflammatory markers, than patients without nADAb. The soil nitrogen (N) cycle in cold terrestrial ecosystems is slow and organically bound N is an important source of N for plants in these ecosystems. Many plant species can take up free amino acids from these infertile soils, either directly or indirectly via their mycorrhizal fungi. We hypothesized that plant community changes and local plant community differences will alter the soil free amino acid pool and composition; and that long-term warming could enhance this effect. To test this, we studied the composition of extractable free amino acids at five separate heath, meadow, and bog locations in subarctic and alpine Scandinavia, with long-term (13 to 24 years) warming manipulations. The plant communities all included a mixture of ecto-, ericoid-, and arbuscular mycorrhizal plant species. Vegetation dominated by grasses and forbs with arbuscular and non-mycorrhizal associations showed highest soil free amino acid content, distinguishing them from the sites dominated by shrubs with ecto- and ericoid-mycorrhizal associations. Warming increased shrub and decreased moss cover at two sites, and by using redundancy analysis, we found that altered soil free amino acid composition was related to this plant cover change. From this, we conclude that the mycorrhizal type is important in controlling soil N cycling and that expansion of shrubs with ectomycorrhiza (and to some extent ericoid mycorrhiza) can help retain N within the ecosystems by tightening the N cycle. Nitrogen (N) cycling in organic tundra soil is characterised by pronounced seasonal dynamics and strong influence of the dominant plant functional types. Such patterns in soil N-cycling have mostly been investigated by the analysis of soil N-pools and net N mineralisation rates, which, however, yield little information on soil N-fluxes. In this study we investigated microbial gross N-transformations, as well as concentrations of plant available N-forms in soils under two dominant plant functional types in tundra heath, dwarf shrubs and mosses, in subarctic Northern Sweden. We collected organic soil under three dwarf shrub species of distinct growth form and three moss species in early and late growing season. Our results showed that moss sites were characterised by significantly higher microbial N-cycling rates and soil N-availability than shrub sites. Protein depolymerisation, the greatest soil N-flux, as well as gross nitrification rates generally did not vary significantly between early and late growing season, whereas gross N mineralisation rates and inorganic N availability markedly dropped in late summer at most sites. The magnitude of the seasonal changes in N-cycling, however, clearly differed among plant functional types, indicating interactive effects of seasonality and plant species on soil N-cycling. Our study highlights that the spatial variation and seasonal dynamics of microbial N transformations and soil N availability in tundra heath are intimately linked with the distinct influence of plant functional types on soil microbial activity and the plant species-specific patterns of nutrient uptake and carbon assimilation. Purpose: In many northern ecosystems, mosses account for an important component of the plant community. Further, mosses host nitrogen (N2)-fixing bacteria that have been shown to contribute a large part of 'new' N in pristine ecosystems. This study aimed to assess direct and indirect effects of warming on moss-associated N2 fixation. Methods: We assessed how N2 fixation activity at ecosystem- and at moss species-level is affected by direct (warming) and indirect (vegetation shift) climate change using two long-term field manipulation experiments in a subarctic birch forest combined with mesocosm studies from a close-by heath. We linked the abundance of ground covering mosses to N2 fixation activity and assessed the temperature sensitivity of N2 fixation in two common feather mosses, Hylocomium splendens and Pleurozium schreberi. Results: Our study shows a steep decline in moss ground cover as well as in associated N2 fixation in response to 10 years of manipulated field-warming, whereas a shift in dominant shrub species did not impact moss cover or N2 fixation activity. Interestingly, the moss H. splendens was more sensitive than P. schreberi to warming both in terms of cover and N2 fixation activity. Conclusion: Our study shows that warming affects moss-associated N2 fixation activity negatively - both indirectly via a decline in moss cover and directly by reducing N2 fixation activity. We compared the odds of smoking cessation at 2-months post-myocardial infarction (MI), before and after implementing routines optimizing use of evidence-based smoking cessation methods, with start during admission. The following routines were implemented at six Swedish hospitals: cardiac rehabilitation nurses offering smokers consultation during admission, optimizing nicotine replacement therapy and varenicline prescription, and contacting patients by telephone during the 1st week post-discharge. Using logistic regression, odds for smoking cessation at 2-months before (n smokers/n admitted = 188/601) and after (n = 195/632) routine implementation were compared. Secondary outcomes included adherence to implemented routines and assessing the prognostic value of each routine on smoking cessation. After implementation, a larger proportion of smokers (65% vs. 54%) were abstinent at 2-months (OR 1.60 [1.04–2.48]). Including only those counselled during admission (n = 98), 74% were abstinent (2.50 [1.42–4.41]). After implementation, patients were more often counselled during admission (50% vs. 6%, p < 0.001), prescribed varenicline (23% vs. 7%, p < 0.001), and contacted by telephone post-discharge (18% vs. 2%, p < 0.001). Being contacted by telephone post-discharge (adjusted OR 2.74 [1.02–7.35]) and prescribed varenicline (adjusted OR 0.39 [0.19–0.83]) predicted smoking cessation at 2-months. Understanding N budgets of tundra ecosystems is crucial for projecting future changes in plant community composition, greenhouse gas balances and soil N stocks. Winter warming can lead to higher tundra winter nitrogen (N) mineralization rates, while summer warming may increase both growing season N mineralization and plant N demand. The undulating tundra landscape is inter-connected through water and solute movement on top of and within near-surface soil, but the importance of lateral N fluxes for tundra N budgets is not well known. We studied the size of lateral N fluxes and the fate of lateral N input in the snowmelt period with a shallow thaw layer, and in the late growing season with a deeper thaw layer. We used 15N to trace inorganic lateral N movement in a Low-arctic mesic tundra heath slope in West Greenland and to quantify the fate of N in the receiving area. We found that half of the early-season lateral N input was retained by the receiving ecosystem, whereas half was transported downslope. Plants appear as poor utilizers of early-season N, indicating that higher winter N mineralization may influence plant growth and carbon (C) sequestration less than expected. Still, evergreen plants were better at utilizing early-season N, highlighting how changes in N availability may impact plant community composition. In contrast, later growing season lateral N input was deeper and offered an advantage to deeper-rooted deciduous plants. The measurements suggest that N input driven by future warming at the study site will have no significant impact on the overall N2O emissions. Our work underlines how tundra ecosystem N allocation, C budgets and plant community composition vary in their response to lateral N inputs, which may help us understand future responses in a warmer Arctic. There is generally a mismatch in the land use classification of life cycle inventory (LCI) databases and life cycle impact assessment (LCIA) methods. This mismatch can hinder the proper assessment of land use impacts on biodiversity. To facilitate such assessments, we matched the land use classes of two global LCIA methods to five widely used LCI databases, one LCI nomenclature, and one multi-regional input–output database. In unclear cases, we assumed the worst case. Assumptions were especially necessary for unspecified land use intensity classes. Angina and no obstructive coronary artery disease (CAD) have an unfavorable prognosis, possibly due to diffuse myocardial fibrosis (DMF). In DMF the proteoglycans biglycan and versican are actively remodeled by matrix metalloproteinase. We investigated biglycan and versican in females with angina and possible DMF assessed by cardiac magnetic resonance (CMR). Seventy-one females with angina and no obstructive CAD were included. Asymptomatic females served as controls. Versican and biglycan were measured and CMR was performed measuring extracellular volume. Biglycan and versican levels were higher in symptomatic females compared with controls; 31.4 ng/mL vs. 16.4 ng/mL (p < 0.001) and 2.1 ng/mL vs. 1.8 ng/mL (p < 0.001) and moderately correlated to extracellular volume (r2 = 0.38, p<0.001 and r2 = 0.26, p = 0.015). Turnover of biglycan and versican was increased in angina females compared with controls and associated with extracellular volume, supporting a link between angina with no obstructive CAD and fibrotic remodeling. The construction sector is progressively becoming more circular by reducing waste, re-using building materials and adopting regenerative solutions for energy production and biodiversity protection. The implications of circularity on construction activities are complex and require the careful evaluation of impacts to select the appropriate path forward. Evaluations of circular solutions and their environmental effectiveness are often performed based on various types of life cycle-based impact assessments. This paper uses systemic thinking to map and evaluate different impact assessment methodologies and their implications for a shift to more circular solutions. The following systemic levels are used to group the methodologies: product (material life cycle declarations and building assessments), organisation (certification and management schemes) and system (policies, standards and regulations). The results confirm that circular economy is integrated at all levels. However, development and structure are not coordinated or governed unidirectionally, but rather occur simultaneously at different levels. This recursive structure is positive if the methods are applied in the correct context, thus providing both autonomy and cohesion in decision making. Methods at lower systemic levels may then improve production processes and stimulate the market to create circular and innovative building solutions, whereas methods at higher systemic levels can be used, for example, by real estate builders, trade organisations and governments to create incentives for circular development and innovation in a broader perspective. Use of the performance methods correctly within an actor network is therefore crucial for successful and effective implementation of circular economy in the construction sector. Objective: The associations between fatigue and disease activity in patients with rheumatoid arthritis (RA) have not been defined. The present objectives were to explore in RA patients the cross-sectional and longitudinal relation of fatigue with subjective as well as objective assessments of disease activity. Methods: RA patients were consecutively included when initiating biologic disease-modifying anti-rheumatic drugs (DMARDs) and assessed at baseline, 1, 2, 3, 6, and 12 months with investigation of fatigue, patient-reported outcome measures (PROMs; joint pain and patient's global disease activity, MHAQ, pain catastrophizing, Mental Health score), clinical examinations (examiner's global disease activity, 28 tender and swollen joint counts), and laboratory variables (ESR, CRP, calprotectin). Ultrasound examinations (semi-quantitative scoring (0–3)) with grey scale and power Doppler were performed of 36 joints and 4 tendons. Statistics included one-way analysis of variance, Pearson's correlations, and multiple linear and logistic regression analysis. Results: A total of 208 RA patients (mean (SD) age 53.2 (13.2) years, disease duration 9.8 (8.5) years) were included. Fatigue levels diminished during follow-up (mean (SD) baseline/12 months; 4.8 (2.8)/3.0 (2.5) (p < 0.001)). Substantial correlations were cross-sectionally found between fatigue and PROMs (median (IQR) r=0.61 (0.52-0.71)) but not with the objective inflammatory assessments. During follow-up, baseline fatigue was associated with PROMs (p < 0.001) but not with objective inflammatory assessments. However, change of fatigue was associated with change in all variables. Higher baseline fatigue levels were associated with lower clinical composite score remission rates. Conclusion: Fatigue was cross-sectionally associated to subjective but not to objective disease assessments. However, change of fatigue during treatment was associated to all assessments of disease activity. Arctic and subarctic ecosystems are experiencing substantial changes in hydrology, vegetation, permafrost conditions, and carbon cycling, in response to climatic change and other anthropogenic drivers, and these changes are likely to continue over this century. The total magnitude of these changes results from multiple interactions among these drivers. Field measurements can address the overall responses to different changing drivers, but are less capable of quantifying the interactions among them. Currently, a comprehensive assessment of the drivers of ecosystem changes, and the magnitude of their direct and indirect impacts on subarctic ecosystems, is missing. The Torneträsk area, in the Swedish subarctic, has an unrivalled history of environmental observation over 100 years, and is one of the most studied sites in the Arctic. In this study, we summarize and rank the drivers of ecosystem change in the Torneträsk area, and propose research priorities identified, by expert assessment, to improve predictions of ecosystem changes. The research priorities identified include understanding impacts on ecosystems brought on by altered frequency and intensity of winter warming events, evapotranspiration rates, rainfall, duration of snow cover and lake-ice, changed soil moisture, and droughts. This case study can help us understand the ongoing ecosystem changes occurring in the Torneträsk area, and contribute to improve predictions of future ecosystem changes at a larger scale. Purpose: Do-not-resuscitate (DNR) decision-making in severely ill patients presents many difficult medical, ethical, and legal challenges. The primary aim of this study was to explore cancer patients' and health care professionals' attitudes regarding DNR decision-making authority and timing of the decision. Methods: This study was a questionnaire survey among Danish cancer patients and their attending physicians and nurses in an oncology outpatient setting. Potential differences between patients', physicians', and nurses' answers to the questionnaire were analyzed using Fisher's exact test. Results: Responses from 904 patients, 59 physicians, and 160 nurses were analyzed. The majority in all three groups agreed that DNR decisions should be made in collaboration between physician and patient. However, one-third of the patients answered that the patient alone should make the decision regarding DNR, which contrasts with the physicians' and nurses' attitudes, 0% and 6% pointing to the patient as sole decision-maker, respectively. In case of disagreement between patient and physician, a majority of both patients (66%) and physicians (86%) suggested themselves as the ultimate decision-maker. Additionally, 43% of patients but only 19% of physicians preferred the DNR discussion being brought up early in the course of the disease. Conclusions: With regard to the decisional role of patient vs. physician and the timing of the DNR discussion, we found a substantial discrepancy between the attitudes of cancer patients and physicians. Aims: Plant growth in the Arctic is often nutrient limited due to temperature constraints on decomposition and low atmospheric input of nitrogen (N). Local hotspots of nutrient enrichment found in up to 4000-year-old archaeological deposits can be used to explore the recycling and long-term retention of nutrients in arctic ecosystems. Methods: We investigated old Inuit and Norse deposits (known as middens) and adjacent tundra ecosystems along a wet-dry fjord gradient in western Greenland to explore the isotopic fingerprinting of plant and soil carbon and nitrogen (13C/12C and 15N/14N) derived from human presence. Results: At all locations we observed a significant isotopic fingerprint in soil and plant N related to human deposits. This demonstrates a century-long legacy of past human habitation on plant and soil characteristics and indicates a surprisingly high N retention in these ecosystems. This is consistent with the significantly higher plant biomass in areas with archaeological deposits. Conclusion: Vegetation composition and N in plants and soils displayed marked differences along the wet-dry fjord gradient. Furthermore, the profound nutrient enrichment and organic matter accumulation in archaeological deposits compared to surrounding tundra demonstrates a century-long legacy of past habitation on plant and soil characteristics as well as efficient N cycling with surprisingly limited N loss. Women with angina and no obstructive coronary artery disease (CAD) have worse cardiovascular prognosis than asymptomatic women. Limitation in myocardial perfusion caused by coronary microvascular dysfunction (CMD) is one of the proposed mechanisms contributing to the adverse prognosis. The aim of this study was to assess myocardial perfusion in symptomatic women with no obstructive CAD suspected for CMD compared with asymptomatic sex-matched controls using static CT perfusion (CTP). We performed a semi-quantitative assessment of the left ventricular myocardial perfusion and myocardial perfusion reserve (MPR), using static CTP with adenosine provocation, in 105 female patients with angina and no obstructive CAD (< 50% stenosis) and 33 sex-matched controls without a history of angina or ischemic heart disease. Patients were on average 4 years older (p = 0.04) and had a higher burden of cardiovascular risk factors. While global perfusion during rest was comparable between the groups (age-adjusted p = 0.12), global perfusion during hyperemia was significantly reduced in patients compared with controls (163 ± 23 HU vs. 171 ± 25 HU; age-adjusted p = 0.023). The ability to increase myocardial perfusion during adenosine-induced vasodilation was significantly diminished in patients (MPR 148% vs. 158%; age-adjusted p < 0.001). This remained unchanged after adjustment for cardiovascular risk factors (p = 0.008). Women with angina and no obstructive CAD have reduced hyperemic myocardial perfusion and MPR compared with sex-matched controls. Aims: Soils of northern latitude tundra ecosystems have accumulated large amounts of carbon that might be released as CO2 when temperature rises and the tree-line moves north. We aim to investigate the potential CO2 flux changes at a subarctic tundra heath under changing climate. Methods: We measured daytime ecosystem respiration and photosynthesis at a subarctic heath over a full year under ambient conditions and in factorial long-term (13 years) increased summer temperature and leaf litter addition plots, and in additional short-term (2 years) summer warming plots. Results: Under ambient conditions the ecosystem was a daytime sink of CO2 in the five warmest months, but a net daytime source in the cold season. Thirteen years of summer warming by 1 °C at soil surface increased CO2 emissions, as daytime respiration increased by 37% and photosynthesis by 29% over the year. Short-term warming likewise increased fluxes. Litter addition also increased the emission of CO2 as ecosystem respiration rose by 21% but photosynthesis remained unchanged. Both warming and litter addition significantly enhanced the amount of green biomass. Conclusions: This study suggests that in a changed climate subarctic ecosystems will act as a positive feedback source of atmospheric CO2. It shows the significance of CO2 fluxes outside the growing season and demonstrates a cold-season long- but not short-term legacy effect of increased summer warming on CO2 emission. Herbivores are key drivers of vegetation dynamics in most ecosystems. However, the effect of high arctic herbivores on vegetation dynamics throughout a growing season is not well understood. In this study, we examine the impacts of a large dominant herbivore in the tundra ecosystem, the muskox, Ovibos moschatus, using exclosures established in a fen in high Arctic Greenland. Using weekly sampling throughout the growing season, we quantified the effects of muskox exclusion on aboveground plant biomass and on concentrations and pools of carbon and nitrogen, and explored the timing of peak biomass and nutrient pools. Excluding muskoxen profoundly changed plant abundances and dynamics of the fen, with more than a doubling of aboveground plant biomass and carbon and nitrogen pool sizes. Specifically, large increases in mosses and litter were observed, while graminoid biomass did not change significantly with exclusion of muskoxen. Excluding muskoxen advanced the peak of plant biomass, but also led to a more rapid decline, resulting in an earlier and shorter period of maximal plant biomass inside the exclosures. The largest impact of muskoxen on the arctic fen ecosystem seems to be through their trampling effects on the moss layer, which likely mediated most of the observed changes. This study demonstrates how quickly an ecosystem may respond to changes in the abundance of large herbivores and highlights their pivotal role in modifying vegetation dynamics and nutrient cycling in tundra ecosystems. Soils in northern latitudes store more than twice the amount of carbon (C) currently in the atmosphere and are warming faster than the rest of the globe. Warming has been linked to an expansion of woody vegetation across tundra, raising questions about how these two phenomena interact to modulate C stocks and turnover. We investigated how long-term warming and litter addition have modified microbial processes, soil characteristics, and C and nitrogen (N) stocks. We hypothesized that warming and litter would interact to amplify soil C losses and would be accompanied by increases in microbial activity. Using soil samples from a 16-year warming and litter addition field manipulation, we measured soil C and N stocks, heterotrophic respiration, extracellular enzyme activity, and microbial stoichiometry. We found that warming decreased C and N stocks across the entire soil profile. Depth-specific analyses illustrated that these changes are driven by increasing microbial activity at 5-10 and 10-15 cm depth, and trends toward higher dissolved organic C and N at 5-10 cm depth. This emphasizes the potential for increased leaching losses with warming and additional litter. While litter addition did not change overall C and N stocks, it appears to modify the ecosystem by adding nutrients and C to the soil. Collectively, these findings highlight the vulnerability of northern soils to continued warming with respect to nutrient and C turnover and provide insights into the mechanistic responses of tundra soil to prolonged global change. In the light of the unconventional monetary policies implemented by most large central banks around the world, there is an intense debate about the potential impact on the prices of capital assets. Particularly in Germany, skepticism about the sustainability of the current policy by the European Central Bank is wide spread and concerns about the emergence of a speculative price bubble in the housing market are increasing. The present study analyzes a comprehensive data set covering 127 large German cities from 1990 through 2013, using tests for speculative bubbles, both at a national and city level. Furthermore, we apply two new testing approaches: panel-data and principal components versions of explosive root tests. We find evidence for explosive price increases in many cities. However, it is only in select urban housing markets that prices decouple from their fundamental values. There is no evidence for speculative price movements nationally. [ABSTRACT FROM AUTHOR]

Purpose: Guidance is needed on best-suited indicators to quantify and monitor the man-made impacts on human health, biodiversity and resources. Therefore, the UNEP-SETAC Life Cycle Initiative initiated a global consensus process to agree on an updated overall life cycle impact assessment (LCIA) framework and to recommend a non-comprehensive list of environmental indicators and LCIA characterization factors for (1) climate change, (2) fine particulate matter impacts on human health, (3) water consumption impacts (both scarcity and human health) and 4) land use impacts on biodiversity.Methods: The consensus building process involved more than 100 world-leading scientists in task forces via multiple workshops. Results were consolidated during a 1-week Pellston Workshop™ in January 2016 leading to the following recommendations.Results and discussion: LCIA framework: The updated LCIA framework now distinguishes between intrinsic, instrumental and cultural values, with disability-adjusted life years (DALY) to characterize damages on human health and with measures of vulnerability included to assess biodiversity loss. Climate change impacts: Two complementary climate change impact categories are recommended: (a) The global warming potential 100 years (GWP 100) represents shorter term impacts associated with rate of change and adaptation capacity, and (b) the global temperature change potential 100 years (GTP 100) characterizes the century-scale long term impacts, both including climate-carbon cycle feedbacks for all climate forcers. Fine particulate matter (PM2.5) health impacts: Recommended characterization factors (CFs) for primary and secondary (interim) PM2.5 are established, distinguishing between indoor, urban and rural archetypes. Water consumption impacts: CFs are recommended, preferably on monthly and watershed levels, for two categories: (a) The water scarcity indicator “AWARE” characterizes the potential to deprive human and ecosystems users and quantifies the relative Available WAter REmaining per area once the demand of humans and aquatic ecosystems has been met, and (b) the impact of water consumption on human health assesses the DALYs from malnutrition caused by lack of water for irrigated food production. Land use impacts: CFs representing global potential species loss from land use are proposed as interim recommendation suitable to assess biodiversity loss due to land use and land use change in LCA hotspot analyses.Conclusions: The recommended environmental indicators may be used to support the UN Sustainable Development Goals in order to quantify and monitor progress towards sustainable production and consumption. Taurine is regarded as an essential amino acid in utero, and fetal taurine supply is believed to rely solely on placental transfer from maternal plasma. Despite its potential role in intrauterine growth restriction and other developmental disturbances, human in vivo studies of taurine transfer between the maternal, placental, and fetal compartments are scarce. We studied placental transfer of taurine in uncomplicated human term pregnancies in vivo in a cross-sectional study of 179 mother-fetus pairs. During cesarean section, we obtained placental tissue and plasma from incoming and outgoing vessels on the maternal and fetal sides of the placenta. Taurine was measured by liquid chromatography-tandem mass spectrometry. We calculated paired arteriovenous differences, and measured placental expression of the taurine biosynthetic enzyme cysteine sulfinic acid decarboxylase (CSAD) with quantitative real-time polymerase chain reaction and western blot. We observed a fetal uptake (p < 0.001), an uteroplacental release (p < 0.001), and a negative placental consumption of taurine (p = 0.001), demonstrating a bilateral placental release to the maternal and fetal compartments. Increasing umbilical vein concentrations and fetal uptake was associated with the uteroplacental release to the maternal circulation (rs = − 0.19, p = 0.01/rs = − 0.24, p = 0.003), but not with taurine concentrations in placental tissue. CSAD-mRNA was expressed in placental tissue, suggesting a potential for placental taurine synthesis. Our observations show that the placenta has the capacity to a bilateral taurine release, indicating a fundamental role of taurine in the human placental homeostasis beyond the supply to the fetus. Snow may prevent Arctic herbivores from accessing their forage in winter, forcing them to aggregate in the few patches with limited snow. In High Arctic Greenland, Arctic hare and rock ptarmigan often forage in muskox feeding craters. We therefore hypothesized that due to limited availability of forage, the dietary niches of these resident herbivores overlap considerably, and that the overlap increases as winter progresses. To test this, we analyzed fecal samples collected in early and late winter. We used molecular analysis to identify the plant taxa consumed, and stable isotope ratios of carbon and nitrogen to quantify the dietary niche breadth and dietary overlap. The plant taxa found indicated only limited dietary differentiation between the herbivores. As expected, dietary niches exhibited a strong contraction from early to late winter, especially for rock ptarmigan. This may indicate increasing reliance on particular plant resources as winter progresses. In early winter, the diet of rock ptarmigan overlapped slightly with that of muskox and Arctic hare. Contrary to our expectations, no inter-specific dietary niche overlap was observed in late winter. This overall pattern was specifically revealed by combined analysis of molecular data and stable isotope contents. Hence, despite foraging in the same areas and generally feeding on the same plant taxa, the quantitative dietary overlap between the three herbivores was limited. This may be attributable to species-specific consumption rates of plant taxa. Net methane (CH4) flux across the ecosystem-atmosphere boundary is governed by two counteracting processes, CH4 oxidation and production. Recent research on CH4 cycling has focused on net CH4 fluxes, however, the separate processes of CH4 oxidation and production may vary at local scales and respond differently to environmental change. Here, we separate CH4 oxidation and production, measured as emission, in situ using CH4 oxidation inhibition combined with a novel in situ 13CH4 labeling experiment to determine the rate of soil oxidation of atmospheric CH4. The study was conducted in a subarctic heath ecosystem with three characteristic plant community types: moist mixed species heath, dry Carex-dominated heath, and wet Eriophorum-dominated fen. We further explored the projected climate change effects of increased temperature and enhanced leaf litter input. The CH4 oxidation inhibition experiment revealed significant potential CH4 emission despite net CH4 uptake. Total CH4 oxidation and potential CH4 emission rates differed significantly between plant communities, demonstrating high local-scale variation in CH4 fluxes. Climate treatments did not affect CH4 oxidation rates, however, warming tended to increase potential CH4 emission, indicating that climate change may affect oxidation and production rates asymmetrically. Near-surface soil oxidation of atmospheric CH4 was successfully traced using 13C stable isotope labeling in situ. CH4 oxidation rates ranged widely, yet preliminarily suggested some degree of substrate limitation. Accounting for the local-scale variation in CH4 fluxes and the relative importance of the separate processes of CH4 oxidation and production will contribute importantly to predicting changes in landscape-scale CH4 budgets and climate feedbacks.
