Spaceflight presents a multifaceted environment for plants, combining the effects on growth of many stressors and factors including altered gravity, the influence of experiment hardware, and increased radiation exposure. To help understand the plant response to this complex suite of factors this study compared transcriptomic analysis of 15 Arabidopsis thaliana spaceflight experiments deposited in the National Aeronautics and Space Administration’s GeneLab data repository. These data were reanalyzed for genes showing significant differential expression in spaceflight versus ground controls using a single common computational pipeline for either the microarray or the RNA-seq datasets. Such a standardized approach to analysis should greatly increase the robustness of comparisons made between datasets. This analysis was coupled with extensive cross-referencing to a curated matrix of metadata associated with these experiments. Our study reveals that factors such as analysis type (i.e., microarray versus RNA-seq) or environmental and hardware conditions have important confounding effects on comparisons seeking to define plant reactions to spaceflight. The metadata matrix allows selection of studies with high similarity scores, i.e., that share multiple elements of experimental design, such as plant age or flight hardware. Comparisons between these studies then helps reduce the complexity in drawing conclusions arising from comparisons made between experiments with very different designs.

Plant biology is an important area for the future of space exploration, but biological spaceflight experiments have been always constrained by the hardware capabilities. The European Modular Cultivation System (EMCS) unit was an incubator for small organisms, such as Arabidopsis thaliana, built by the European Space Agency (ESA) and was decommissioned in 2018. Here, we describe the FixBox system as add-on hardware to provide fixation capabilities to the plant growth cassettes, which, initially, were not designed to be used for that purpose. Tests were performed to ensure the successful use of this device in the EMCS facility. We also evaluate the required adaptations to the hardware, e.g., to guarantee that the reduced fluid motion in microgravity does not cause any bubbles that could impair the quality of fixation. Arabidopsis thaliana seedlings grown during spaceflight were fixed in the FixBox either in glutaraldehyde or formaldehyde. Electron microscopical images and confocal microscopy immunofluorescent localizations showed an excellent preservation of both cell ultrastructure and antigen conformation. Thus, it is possible to modify existing hardware to comply with the scientific requirements to augment the existing capabilities on board the ISS. In addition, it is also possible to reuse culture chambers from predesigned experimental containers into new modular subunits as FixBox. Similarly, we can design new hardware compatible with a novel cultivation chamber on board, such as is available in BIOLAB, to be used later with FixBox. Lessons learned for future space plant biology researchers include how to manage the number of hardware requirements and constraints on how to preserve the biological samples.

[Purpose of Review] This paper reviews the recent literature on numerical modelling of the dynamics of the Greenland ice sheet with the goal of providing an overview of advancements and to highlight important directions of future research. In particular, the review is focused on large-scale modelling of the ice sheet, including future projections, model parameterisations, paleo applications and coupling with models of other components of the Earth system. [Recent Findings] Data assimilation techniques have been used to improve the reliability of model simulations of the Greenland ice sheet dynamics, including more accurate initial states, more comprehensive use of remote sensing as well as paleo observations and inclusion of additional physical processes. [Summary] Modellers now leverage the increasing number of high-resolution satellite and air-borne data products to initialise ice sheet models for centennial time-scale simulations, needed for policy relevant sea-level projections. Modelling long-term past and future ice sheet evolution, which requires simplified but adequate representations of the interactions with the other components of the Earth system, has seen a steady improvement. Important developments are underway to include ice sheets in climate models that may lead to routine simulation of the fully coupled Greenland ice sheet–climate system in the coming years.

Due to their sessile nature, plants utilize environmental cues to grow and respond to their surroundings. Two of these cues, light and gravity, play a substantial role in plant orientation and directed growth movements (tropisms). However, very little is currently known about the interaction between light- (phototropic) and gravity (gravitropic)-mediated growth responses. Utilizing the European Modular Cultivation System on board the International Space Station, we investigated the interaction between phototropic and gravitropic responses in three Arabidopsis thaliana genotypes, Landsberg wild type, as well as mutants of phytochrome A and phytochrome B. Onboard centrifuges were used to create a fractional gravity gradient ranging from reduced gravity up to 1g. A novel positive blue-light phototropic response of roots was observed during conditions of microgravity, and this response was attenuated at 0.1g. In addition, a red-light pretreatment of plants enhanced the magnitude of positive phototropic curvature of roots in response to blue illumination. In addition, a positive phototropic response of roots was observed when exposed to red light, and a decrease in response was gradual and correlated with the increase in gravity. The positive red-light phototropic curvature of hypocotyls when exposed to red light was also confirmed. Both red-light and blue-light phototropic responses were also shown to be affected by directional light intensity. To our knowledge, this is the first characterization of a positive blue-light phototropic response in Arabidopsis roots, as well as the first description of the relationship between these phototropic responses in fractional or reduced gravities.

The recent discovery of liquid water-related sulphates on Mars is of great astrobiological interest. UV radiation experiments, using natural Ca and Fe sulphates (gypsum, jarosite), coming from two selected areas of SE Spain (Jaroso Hydrothermal System and the Sorbas evaporitic basin), were performed using a Xe Lamp with an integrated output from 220 nm to 500 nm of 1.2 Wm-2. The results obtained demonstrate a large difference in the UV protection capabilities of both minerals and also confirm that the mineralogical composition of the Martian regolith is a crucial shielding factor. Whereas gypsum showed a much higher transmission percentage, jarosite samples, with a thickness of only 500 microns, prevented transmission. This result is extremely important for the search for life on Mars as: a) jarosite typically occurs on Earth as alteration crusts and patinas, and b) a very thin crust of jarosite on the surface of Mars would be sufficient to shield microorganisms from UV radiation.

Integral field optical spectroscopy with the INTEGRAL fiber-fed system is used to investigate the properties of the tidal-induced star formation in the Ultraluminous Infrared Galaxy IRAS 12112+0305. Implications for the study of the SCUBA sources are briefly mentioned.
