1. Carrier-Phase DNS of Ignition and Combustion of Iron Particles in a Turbulent Mixing Layer.

Abstract: Three-dimensional carrier-phase direct numerical simulations (CP-DNS) of reacting iron particle dust clouds in a turbulent mixing layer are conducted. The simulation approach considers the Eulerian transport equations for the reacting gas phase and resolves all scales of turbulence, whereas the particle boundary layers are modelled employing the Lagrangian point-particle framework for the dispersed phase. The CP-DNS employs an existing sub-model for iron particle combustion that considers the oxidation of iron to FeO and that accounts for both diffusion- and kinetically-limited combustion. At first, the particle sub-model is validated against experimental results for single iron particle combustion considering various particle diameters and ambient oxygen concentrations. Subsequently, the CP-DNS approach is employed to predict iron particle cloud ignition and combustion in a turbulent mixing layer. The upper stream of the mixing layer is initialised with cold particles in air, while the lower stream consists of hot air flowing in the opposite direction. Simulation results show that turbulent mixing induces heating, ignition and combustion of the iron particles. Significant increases in gas temperature and oxygen consumption occur mainly in regions where clusters of iron particles are formed. Over the course of the oxidation, the particles are subjected to different rate-limiting processes. While initially particle oxidation is kinetically-limited it becomes diffusion-limited for higher particle temperatures and peak particle temperatures are observed near the fully-oxidised particle state. Comparing the present non-volatile iron dust flames to general trends in volatile-containing solid fuel flames, non-vanishing particles at late simulation times and a stronger limiting effect of the local oxygen concentration on particle conversion is found for the present iron dust flames in shear-driven turbulence. 3. Religious conversion, philosophy, and social science.

Abstract: I argue that empirical studies into the phenomenon of religious conversion suffer from conceptual unclarity owing to an absence of philosophical contributions. I examine the relationship between definition and empirical result in the social sciences, and I show that a wide divergence in conceptual approach threatens to undermine the possibility of useful comparative study. I stake out a distinctive role for philosophical treatments of studies into religious conversion. I conclude with the suggestion that use of the terms 'convert' and 'conversion' may not in fact be conducive to clarity in the present context, and that subsequent studies may improve their precision through replacing them. 4. Predicting Coral Reef Carbonate Chemistry Through Statistical Modeling: Constraining Nearshore Residence Time Around Guam.

Abstract: To accurately predict the impacts of ocean acidification on shallow-water ecosystems, we must account for the biogeochemical impact of local benthic communities, as well as the connectivity between offshore and onshore water masses. Estimation of residence time can help quantify this connectivity and determine the degree to which the benthos can influence the chemistry of the overlying water column. We present estimates of nearshore residence time for Guam and utilize these estimates to model the effects of benthic ecosystem metabolism on the coral reef carbonate system. Control volume and particle tracking approaches were used to estimate nearshore residence time. These estimates were paired with observed patterns in the reef carbonate system around Guam using water samples collected by NOAA's National Coral Reef Monitoring Program. Model performance results suggest that when considering the effects of benthic metabolism on the carbonate system, it is paramount to represent the contact time of the water volume with the benthos. Even coarse estimates of residence time significantly increase model skill. We observed the highest predictive skill in models including control volume derived estimates of residence time, but only when those estimates were included as an interaction with benthic composition. This work shows that not only is residence time critically important to better predict biogeochemical variability in coral reef environments, but that even coarse hydrodynamic models can provide useful residence time estimates at management relevant, whole-ecosystem scales. Abstract: Reninomas are exceedingly rare renin-secreting kidney tumours that derive from juxtaglomerular cells, specialised smooth muscle cells that reside at the vascular inlet of glomeruli. They are the central component of the juxtaglomerular apparatus which controls systemic blood pressure through the secretion of renin. We assess somatic changes in reninoma and find structural variants that generate canonical activating rearrangements of, NOTCH1 whilst removing its negative regulator, NRARP. Accordingly, in single reninoma nuclei we observe excessive renin and NOTCH1 signalling mRNAs, with a concomitant non-excess of NRARP expression. Re-analysis of previously published reninoma bulk transcriptomes further corroborates our observation of dysregulated Notch pathway signalling in reninoma. Our findings reveal NOTCH1 rearrangements in reninoma, therapeutically targetable through existing NOTCH1 inhibitors, and indicate that unscheduled Notch signalling may be a disease-defining feature of reninoma.Reninomas are very rare kidney tumours of juxtaglomerular cells. Abstract: One of the key challenges for nuclear physics today is to understand from first principles the effective interaction between hadrons with different quark content. First successes have been achieved using techniques that solve the dynamics of quarks and gluons on discrete space-time lattices1,2. Experimentally, the dynamics of the strong interaction have been studied by scattering hadrons off each other. Such scattering experiments are difficult or impossible for unstable hadrons3–6 and so high-quality measurements exist only for hadrons containing up and down quarks7. Here we demonstrate that measuring correlations in the momentum space between hadron pairs8–12 produced in ultrarelativistic proton–proton collisions at the CERN Large Hadron Collider (LHC) provides a precise method with which to obtain the missing information on the interaction dynamics between any pair of unstable hadrons. Specifically, we discuss the case of the interaction of baryons containing strange quarks (hyperons). We demonstrate how, using precision measurements of proton–omega baryon correlations, the effect of the strong interaction for this hadron–hadron pair can be studied with precision similar to, and compared with, predictions from lattice calculations13,14. The large number of hyperons identified in proton–proton collisions at the LHC, together with accurate modelling15 of the small (approximately one femtometre) inter-particle distance and exact predictions for the correlation functions, enables a detailed determination of the short-range part of the nucleon-hyperon interaction., Correlations in momentum space between hadrons created by ultrarelativistic proton–proton collisions at the CERN Large Hadron Collider provide insights into the strong interaction, particularly the short-range dynamics of hyperons—baryons that contain strange quarks.

Abstract: Germ cell tumours (GCTs) are a collection of benign and malignant neoplasms derived from primordial germ cells. They are uniquely able to recapitulate embryonic and extraembryonic tissues, which carries prognostic and therapeutic significance. The developmental pathways underpinning GCT initiation and histogenesis are incompletely understood. Here, we study the relationship of histogenesis and clonal diversification in GCTs by analysing the genomes and transcriptomes of 547 microdissected histological units. We find no correlation between genomic and histological heterogeneity. However, we identify unifying features including the retention of fetal developmental transcripts across tissues, expression changes on chromosome 12p, and a conserved somatic evolutionary sequence of whole genome duplication followed by clonal diversification. While this pattern is preserved across all GCTs, the developmental timing of the duplication varies between prepubertal and postpubertal cases. In addition, tumours of younger children exhibit distinct substitution signatures which may lend themselves as potential biomarkers for risk stratification. Our findings portray the extensive diversification of GCT tissues and genetic subclones as randomly distributed, while identifying overarching transcriptional and genomic features. The molecular characterisation of germ cell tumours (GCT) is necessary to understand their development and histological diversification. 9. Logical form and logical space in Wittgenstein’s Tractatus.

Abstract: In this article I offer a novel explanation of Wittgenstein’s claim, in his Tractatus, that to represent the logical form of a proposition would require our being positioned outside of logic. The account here presented aims to exploit a connection, widely noticed, between the logical forms of objects and those of the propositions in which the names of those objects figure. I show that the logical forms of propositions may, on Wittgenstein’s view, be identified with places in logical space, and that places in logical space are reducible to the forms of both objects and their names. I argue, though, that according to Wittgenstein the representation of a proposition’s logical form would require the existence of a place in logical space not so reducible. I conclude that on Wittgenstein’s position, the attempt to represent logical forms cannot, therefore, succeed. [ABSTRACT FROM AUTHOR]

Abstract: Over the course of an individual’s lifetime, normal human cells accumulate mutations1. Here we compare the mutational landscape in 29 cell types from the soma and germline using multiple samples from the same individuals. Two ubiquitous mutational signatures, SBS1 and SBS5/40, accounted for the majority of acquired mutations in most cell types, but their absolute and relative contributions varied substantially. SBS18, which potentially reflects oxidative damage2, and several additional signatures attributed to exogenous and endogenous exposures contributed mutations to subsets of cell types. The rate of mutation was lowest in spermatogonia, the stem cells from which sperm are generated and from which most genetic variation in the human population is thought to originate. This was due to low rates of ubiquitous mutational processes and may be partially attributable to a low rate of cell division in basal spermatogonia. Abstract: Starting from the zygote, all cells in the human body continuously acquire mutations. Mutations shared between different cells imply a common progenitor and are thus naturally occurring markers for lineage tracing1,2. Here we reconstruct extensive phylogenies of normal tissues from three adult individuals using whole-genome sequencing of 511 laser capture microdissections. Reconstructed embryonic progenitors in the same generation of a phylogeny often contribute to different extents to the adult body. The degree of this asymmetry varies between individuals, with ratios between the two reconstructed daughter cells of the zygote ranging from 60:40 to 93:7. Asymmetries pervade subsequent generations and can differ between tissues in the same individual. The phylogenies resolve the spatial embryonic patterning of tissues, revealing contiguous patches of, on average, 301 crypts in the adult colonic epithelium derived from a most recent embryonic cell and also a spatial effect in brain development. Using data from ten additional men, we investigated the developmental split between soma and germline, with results suggesting an extraembryonic contribution to primordial germ cells. This research demonstrates that, despite reaching the same ultimate tissue patterns, early bottlenecks and lineage commitments lead to substantial variation in embryonic patterns both within and between individuals.Somatic mutations obtained from laser microdissected biopsies of human tissues are used to reconstruct the developmental phylogenies of these tissues back to the zygote. Abstract: The cellular landscape of the human intestinal tract is dynamic throughout life, developing in utero and changing in response to functional requirements and environmental exposures. Here, to comprehensively map cell lineages, we use single-cell RNA sequencing and antigen receptor analysis of almost half a million cells from up to 5 anatomical regions in the developing and up to 11 distinct anatomical regions in the healthy paediatric and adult human gut. This reveals the existence of transcriptionally distinct BEST4 epithelial cells throughout the human intestinal tract. Furthermore, we implicate IgG sensing as a function of intestinal tuft cells. We describe neural cell populations in the developing enteric nervous system, and predict cell-type-specific expression of genes associated with Hirschsprung’s disease. Finally, using a systems approach, we identify key cell players that drive the formation of secondary lymphoid tissue in early human development. We show that these programs are adopted in inflammatory bowel disease to recruit and retain immune cells at the site of inflammation. 13. Agrochemicals interact synergistically to increase bee mortality.

Abstract: Global concern over widely documented declines in pollinators1–3 has led to the identification of anthropogenic stressors that, individually, are detrimental to bee populations4–7. Synergistic interactions between these stressors could substantially amplify the environmental effect of these stressors and could therefore have important implications for policy decisions that aim to improve the health of pollinators3,8,9. Here, to quantitatively assess the scale of this threat, we conducted a meta-analysis of 356 interaction effect sizes from 90 studies in which bees were exposed to combinations of agrochemicals, nutritional stressors and/or parasites. We found an overall synergistic effect between multiple stressors on bee mortality. Subgroup analysis of bee mortality revealed strong evidence for synergy when bees were exposed to multiple agrochemicals at field-realistic levels, but interactions were not greater than additive expectations when bees were exposed to parasites and/or nutritional stressors. All interactive effects on proxies of fitness, behaviour, parasite load and immune responses were either additive or antagonistic; therefore, the potential mechanisms that drive the observed synergistic interactions for bee mortality remain unclear. Environmental risk assessment schemes that assume additive effects of the risk of agrochemical exposure may underestimate the interactive effect of anthropogenic stressors on bee mortality and will fail to protect the pollinators that provide a key ecosystem service that underpins sustainable agriculture.A meta-analysis of studies in which bees were exposed to combinations of agrochemicals, nutritional stressors and/or parasites revealed evidence for synergistic effects on mortality when bees were exposed to multiple agrochemicals at field-realistic levels. Abstract: Tumor cells may share some patterns of gene expression with their cell of origin, providing clues into the differentiation state and origin of cancer. Here, we study the differentiation state and cellular origin of 1300 childhood and adult kidney tumors. Using single cell mRNA reference maps of normal tissues, we quantify reference "cellular signals" in each tumor. Quantifying global differentiation, we find that childhood tumors exhibit fetal cellular signals, replacing the presumption of "fetalness" with a quantitative measure of immaturity. By contrast, in adult cancers our assessment refutes the suggestion of dedifferentiation towards a fetal state in most cases. We find an intimate connection between developmental mesenchymal populations and childhood renal tumors. We demonstrate the diagnostic potential of our approach with a case study of a cryptic renal tumor. Our findings provide a cellular definition of human renal tumors through an approach that is broadly applicable to human cancer. Transcriptomic analysis may provide information about the differentiation state and cell of origin of a cancer. 15. Inherent mosaicism and extensive mutation of human placentas.

Abstract: Placentas can exhibit chromosomal aberrations that are absent from the fetus1. The basis of this genetic segregation, which is known as confined placental mosaicism, remains unknown. Here we investigated the phylogeny of human placental cells as reconstructed from somatic mutations, using whole-genome sequencing of 86 bulk placental samples (with a median weight of 28 mg) and of 106 microdissections of placental tissue. We found that every bulk placental sample represents a clonal expansion that is genetically distinct, and exhibits a genomic landscape akin to that of childhood cancer in terms of mutation burden and mutational imprints. To our knowledge, unlike any other healthy human tissue studied so far, the placental genomes often contained changes in copy number. We reconstructed phylogenetic relationships between tissues from the same pregnancy, which revealed that developmental bottlenecks genetically isolate placental tissues by separating trophectodermal lineages from lineages derived from the inner cell mass. Notably, there were some cases with full segregation—within a few cell divisions of the zygote—of placental lineages and lineages derived from the inner cell mass. Such early embryonic bottlenecks may enable the normalization of zygotic aneuploidy. We observed direct evidence for this in a case of mosaic trisomic rescue. Our findings reveal extensive mutagenesis in placental tissues and suggest that mosaicism is a typical feature of placental development. Abstract: Malignant rhabdoid tumour (MRT) is an often lethal childhood cancer that, like many paediatric tumours, is thought to arise from aberrant fetal development. The embryonic root and differentiation pathways underpinning MRT are not firmly established. Here, we study the origin of MRT by combining phylogenetic analyses and single-cell mRNA studies in patientderived organoids. Comparison of somatic mutations shared between cancer and surrounding normal tissues places MRT in a lineage with neural crest-derived Schwann cells. Single-cell mRNA readouts of MRT differentiation, which we examine by reverting the genetic driver mutation underpinning MRT, SMARCB1 loss, suggest that cells are blocked en route to differentiating into mesenchyme. Quantitative transcriptional predictions indicate that combined HDAC and mTOR inhibition mimic MRT differentiation, which we confirm experimentally. Abstract: Some researchers have suggested that corals living in deeper reefs may escape heat stress experienced by shallow corals. We evaluated the potential of deep coral reef refugia from bleaching stress by leveraging a long record of satellite-derived sea surface temperature data with a temporal, spatial, and depth precision of in situ temperature records. We calculated an in situ stress metric using a depth bias-adjusted threshold for 457 coral reef sites among 49 islands in the western and central Pacific Ocean over the period 2001–2017. Analysis of 1,453 heating events found no meaningful depth refuge from heat stress down to 38 m, and no significant association between depth and subsurface heat stress. Further, the surface metric underestimated subsurface stress by an average of 39.3%, across all depths. Combining satellite and in situ temperature data can provide bleaching-relevant heat stress results to avoid misrepresentation of heat stress exposure at shallow reefs. [ABSTRACT FROM AUTHOR]

Abstract: The 2014–2017 Global Coral Bleaching Event is the longest, most widespread, and impactful on record. Rapid ecological assessment surveys by NOAA's Pacific Reef Assessment and Monitoring Program reported widespread coral mortality at Jarvis Island in the aftermath of the 2015–2016 super-El Niño warming event; hard coral cover declined from 18.7% in April 2015 (pre-bleaching) to 0.4% in May 2016 (post-bleaching), representing a catastrophic > 98% decline. Between 2015 and 2016, corals at Jarvis experienced maximum heat stress of 22.25 °C-weeks exceeding the bleaching threshold (28.72 °C) for 66 consecutive weeks. Mass coral bleaching was observed in November 2015, which resulted in mass mortality across all coral taxa, depths, and island sectors. The bleaching event altered the benthic community composition including the coral assemblage. In the 2 yrs post-bleaching, the benthic community has transitioned from a short-lived increase of encrusting macroalgae to a more recent near-recovery of crustose coralline algae. Coral cover had not recovered by 2017 and could be potentially delayed by fast-growing turf algae. Within the coral community, the pre-bleaching dominant genus Montipora exhibited extreme mortality and only a handful of colonies of this taxon were enumerated in the 2016 surveys and none in 2017. Some coral taxa have persisted in low densities, including the ESA-threatened Acropora retusa and colonies of encrusting Pavona, Psammocora, and the free living Fungia. As the frequency and intensity of these high-temperature events is projected to increase in coming years, it is essential to track how remote ecosystems normally undisturbed by human influence, such as Jarvis, respond to a climate change. Abstract: Behavioral activation (BA) effectively treats depression in adults, and shows promise in treating anxiety. Research into its application to children and adolescents is emerging. This review aimed to explore the scope of studies, current evidence of effectiveness and how the intervention has been delivered and adapted, to inform future research. A systematic review was undertaken searching PsycInfo, PubMed including Medline, EMBASE, and Scopus for terms relating to BA and children and adolescents. Two researchers scored abstracts for inclusion. Data extraction was completed by one researcher and checked by another. 19 studies were identified, across 21 published articles. 12 were case studies, with three pre–post pilot designs and four randomized-controlled trials. Case studies found early support for the feasibility and potential effectiveness of BA to address both anxiety and depression. The RCTs reported largely positive outcomes. Meta-analysis of depression scores indicated that BA may be effective; however, high heterogeneity was observed. Sample sizes to date have been small. BA has been delivered by trained therapists, doctoral trainee psychologists, social workers, or psychology graduates. Studies are uniquely in high-income settings. Adaptations include flexibility in content delivery, youth friendly materials, and parental involvement. There is some limited evidence to support BA as effective for young people. Feasibility and acceptability are supported. Fully powered trials are now required, with expansion to delivery in low- and middle-income settings, and detailed consideration of implementation issues that consider culture and environment. Abstract: Our ability to understand natural constraints on coral reef benthic communities requires quantitative assessment of the relative strengths of abiotic and biotic processes across large spatial scales. Here, we combine underwater images, visual censuses and remote sensing data for 1566 sites across 34 islands spanning the central-western Pacific Ocean, to empirically assess the relative roles of abiotic and grazing processes in determining the prevalence of calcifying organisms and fleshy algae on coral reefs. We used regression trees to identify the major predictors of benthic composition and to test whether anthropogenic stress at inhabited islands decouples natural relationships. We show that sea surface temperature, wave energy, oceanic productivity and aragonite saturation strongly influence benthic community composition; overlooking these factors may bias expectations of calcified reef states. Maintenance of grazing biomass above a relatively low threshold (~ 10-20 kg ha−1) may also prevent transitions to algal-dominated states, providing a tangible management target for rebuilding overexploited herbivore populations. Biophysical relationships did not decouple at inhabited islands, indicating that abiotic influences remain important macroscale processes, even at chronically disturbed reefs. However, spatial autocorrelation among inhabited reefs was substantial and exceeded abiotic and grazing influences, suggesting that natural constraints on reef benthos were superseded by unmeasured anthropogenic impacts. Evidence of strong abiotic influences on reef benthic communities underscores their importance in specifying quantitative targets for coral reef management and restoration that are realistic within the context of local conditions. Abstract: A 32-year-old male developed neuropsychiatric symptoms 2 weeks after starting mefloquine 250 mg/week for malaria prophylaxis. He continued to take the medication for the next 4 months. Initial symptoms included vivid dreams and anxiety, as well as balance problems. These symptoms persisted and progressed over the next 4 years to include vertigo, emotional lability, and poor short-term memory, which have greatly affected his personal and professional life. An extensive evaluation revealed objective evidence supporting a central vestibulopathy. These symptoms have been unresponsive to pharmacologic therapy and psychotherapy. A Naranjo assessment score of 6 was obtained for his initial symptoms, indicating a probable adverse drug reaction to mefloquine given the relationship between the clinical picture and drug exposure. Abstract: Purpose: This study aims to provide recommendations on the optimal strategies and interventions for the prevention, screening, assessment, and management of cancer-related sleep disturbance (insomnia and insomnia syndrome) in adult cancer populations. Methods: A systematic search of the published health literature was conducted to identify randomized controlled trials, clinical practice guidelines, systematic reviews, and other guidance documents. The Sleep Disturbance Expert Panel [comprised of nurses, psychologists, primary care physicians, oncologists, physicians specialized in sleep disturbances, researchers, and guideline methodologists] reviewed, discussed, and approved the final version of the guideline. Health care professionals across Canada were asked to provide feedback through an external review process. Results: Three clinical practice guidelines and 12 randomized controlled trials were identified as the evidence base. Overall, despite the paucity of evidence, the evidence and expert consensus suggest that it is important to screen and assess adult cancer patients for sleep disturbances using standardized screening tools on a routine basis. While prevention of sleep disturbance is the desired objective, cognitive behavioral therapies are effective in improving sleep outcomes. As part of the external review with 16 health care providers, 81 % indicated that they agreed with the recommendations as written. Conclusions: Sleep difficulty is a prevalent problem in cancer populations that needs greater recognition by health professionals. Prevention, screening, assessment, and treatment strategies supported by the best available evidence are critical. Recommendations and care path algorithms for practice are offered. 29. Radiographically occult femoral and pelvic fractures are not mutually exclusive: a review of fractures detected by MRI following low-energy trauma.

Abstract: Objectives: The purpose of this study was to review the MRI examinations of a large group of low-energy trauma patients in whom pelvic MRI had detected radiographically occult fractures, in order to characterize prevailing fracture patterns and determine how often co-existing proximal femoral and pelvic fractures were observed.Methods: All patients having pelvic MRI over 5 years were identified. Word-search software selected 269 MRI reports containing the term ‘fracture’. Further scrutiny identified 168 with diagnosis of fracture. MRI request and imaging record review identified 102 low-energy trauma cases that had MRI for clinical suspicion of fracture despite normal radiographs. Sixty-six cases were excluded for the following reasons: no expressed clinical suspicion of occult fracture; history suggesting high-energy trauma;skeletal co-morbidity hindering acute fracture identification;interval more than 2 weeks between radiographs and MRI. The 102 study MRI examinations, which employed a limited two-sequence protocol, were reviewed. Any fracture that had not been appreciated on radiographs was recorded and characterized as femoral, pelvic, or co-existing femoral and pelvic fractures.Results: The 102 study cases had a median age of 82 years.The median interval between pelvic radiographs and MRI was 3 days. MRI showed undiagnosed femoral fracture in 48/102 cases (47.1%), sacral fracture in 41/102 (40.2%),and pubic fracture in 55/102 (53.9%). In 11/102 cases(10.8%), MRI showed undiagnosed fractures of both proximal femur and pelvic ring (seven sacral, six pubic bone, two other site fractures). In 10/11 cases with coexisting femoral and pelvic fractures, the femoral fracture was incomplete.Conclusions: Limited pelvic MRI found a high prevalence of radiographically occult femoral and pelvic fractures in low-energy trauma patients, with clinical suspicion of fracture despite normal radiographs. Co-existing occult femoral and pelvic ring fractures were commonly observed, and in such cases, the femoral fracture was likely to be incomplete and multiple pelvic fractures were typically present. Abstract: Mangrove and saltmarsh ecosystems appear particularly vulnerable to the impacts of climate change, and their effective management will require forecasts of how these wetland habitats are likely to respond to sea-level rise through the twenty-first century. We describe a preliminary study of a small stand of mangrove and saltmarsh that involves measuring of elevation change and accretion, mapping of wetland communities, and modelling of their potential response to sea-level rise. The wetland occurs on the banks of the Minnamurra River estuary in southern New South Wales and has been the focus of several studies over recent decades. The research includes empirical measurements of sedimentation at sites in both mangrove and saltmarsh vegetation using the surface elevation table-marker horizon technique. This is a site at which mapping has been undertaken to delineate the extent of each vegetation community from a time-series of aerial photographs using geographical information systems; the gradual incursion of the mangrove, Avicennia marina, into more landward saltmarsh communities, observed over past decades for many systems in southeastern Australia, has continued into the twenty-first century. The observed patterns of change are compared with simulations of how this wetland system might respond to future sea-level rise, adopting several different approaches and the upper and lower bounds of Intergovernmental Panel on Climate Change sea-level rise projections. The model results show considerable variability in response depending on the parameters adopted. We advocate the need for the integration of these three approaches, measuring, mapping and modelling, as a basis for future management and adaptation. Our study demonstrates the considerable opportunities to refine the data input and model outputs as part of adaptive management, as more sophisticated technologies and data become available. Abstract: Coral stress tolerance is intricately tied to the animal's association with microbial symbionts. The most well-known of these symbioses is that between corals and their dinoflagellate photobionts (Symbiodinium spp.), whose genotype indirectly affects whether a coral can survive cyclical and anthropogenic warming events. Fungi comprise a lesser-known coral symbiotic community whose taxonomy, stability and function is largely un-examined. To assess how fungal communities inside a coral host correlate with water temperature and the genotype of co-occurring Symbiodinium, we sampled Acropora hyacinthus coral colonies from adjacent natural pools with different water temperatures and Symbiodinium identities. Phylogenetic analysis of coral-associated fungal ribosomal DNA amplicons showed a high diversity of Basidiomycetes and Ascomycetes, including several clades separated from known fungal taxa by long and well-supported branches. Community similarity did not correlate with any measured variables, and total fungal community composition was highly variable among A. hyacinthus coral colonies. Colonies in the warmer pool contained more phylogenetically diverse fungal communities than the colder pool and contained statistically significant 'indicator' species. Four taxa were present in all coral colonies sampled, and may represent obligate associates. Messenger RNA sequenced from a subset of these same colonies contained an abundance of transcripts involved in metabolism of complex biological molecules. Coincidence between the taxonomic diversity found in the DNA and RNA analysis indicates a metabolically active and diverse resident marine fungal community. [ABSTRACT FROM AUTHOR]

Abstract: The oceans are warming and coral reefs are bleaching with increased frequency and severity, fueling concerns for their survival through this century. Yet in the central equatorial Pacific, some of the world's most productive reefs regularly experience extreme heat associated with El Niño. Here we use skeletal signatures preserved in long-lived corals on Jarvis Island to evaluate the coral community response to multiple successive heatwaves since 1960. By tracking skeletal stress band formation through the 2015-16 El Nino, which killed 95% of Jarvis corals, we validate their utility as proxies of bleaching severity and show that 2015-16 was not the first catastrophic bleaching event on Jarvis. Since 1960, eight severe (>30% bleaching) and two moderate (<30% bleaching) events occurred, each coinciding with El Niño. While the frequency and severity of bleaching on Jarvis did not increase over this time period, 2015–16 was unprecedented in magnitude. The trajectory of recovery of this historically resilient ecosystem will provide critical insights into the potential for coral reef resilience in a warming world. Abstract: Sampling and laboratory analysis are not well adapted to wastewater quality monitoring in a process control or hazards prevention context, for which on-line/on-site measurement is preferable. Before considering the implementation and constraints of on-line systems, the reasons for and ways of monitoring are discussed. The main existing and up-and-coming solutions are then presented, showing that with respect to the number of parameters and substances to be monitored, for regulation purposes for example, only a few of them are measurable with on-line devices.
