Despite asthma has a considerable genetic component, an important proportion of genetic risks remain unknown, especially for non-European populations. Canary Islanders have the largest African genetic ancestry observed among Southwestern Europeans and the highest asthma prevalence in Spain. Here we examined broad chromosomal regions previously associated with an excess of African genetic ancestry in Canary Islanders, with the aim of identifying novel risk variants associated with asthma susceptibility. In a two-stage cases-control study, we revealed a variant within HLA-DQB1 significantly associated with asthma risk (rs1049213, meta-analysis p = 1.30 × 10–7, OR [95% CI] = 1.74 [1.41–2.13]) previously associated with asthma and broad allergic phenotype. Subsequent fine-mapping analyses of classical HLA alleles revealed a novel allele significantly associated with asthma protection (HLA-DQA1*01:02, meta-analysis p = 3.98 × 10–4, OR [95% CI] = 0.64 [0.50–0.82]) that had been linked to infectious and autoimmune diseases, and peanut allergy. HLA haplotype analyses revealed a novel haplotype DQA1*01:02-DQB1*06:04 conferring asthma protection (meta-analysis p = 4.71 × 10–4, OR [95% CI] = 0.47 [0.29– 0.73]). [ABSTRACT FROM AUTHOR]

Plant litter decomposition is an essential ecosystem function in temperate streams. Both riparian vegetation and decomposer communities are major determinants of the decomposition efficiency and the interactions occurring within litter mixtures. However, the extent to which such litter mixture interactions are affected by combined shifts in litter traits and decomposer community is not well understood. We used leaf litter from 10 European tree species in order to study litter decomposition and litter mixture effects occurring in two-species litter mixtures in a temperate forested stream of northwestern France. The study distinguished between (1) decomposition involving microorganisms alone or together with invertebrates, and (2) decomposition involving litter mixtures of similar or dissimilar nutrient content. Increasing mean litter nutrient concentration favored both microbial activity and litter decomposition rate. Surprisingly, the highest litter mixture effects occurred in mixtures containing two nutrient-rich litters and occurred mainly in macroinvertebrate presence. Both the "mass ratio hypothesis," expressed as the community-weighted mean traits (TraitCWM), and the "niche complementarity hypothesis," expressed as the functional dissimilarity of litter traits (TraitFD), contributed to explain litter mixture effects. However, TraitCWM was found to be a better predictor than TraitFD. Finally, when evaluating the individual contributions of litter nutrients, calcium and magnesium appeared as important drivers of litter mixture effects. Our findings suggest that the mass ratio hypothesis overrules the niche complementarity hypothesis as a driver of litter diversity effects. Future climate scenarios forecast a 10-50% decline in rainfall in Eastern Amazonia. Altered precipitation patterns may change important ecosystem functions like decomposition through either changes in physical and chemical processes or shifts in the activity and/or composition of species. We experimentally manipulated hydroperiods (length of wet:dry cycles) in a tank bromeliad ecosystem to examine impacts on leaf litter decomposition. Gross loss of litter mass over 112 days was greatest in continuously submersed litter, lowest in continuously dry litter, and intermediate over a range of hydroperiods ranging from eight cycles of 7 wet:7 dry days to one cycle of 56 wet:56 dry days. The resilience of litter mass loss to hydroperiod length is due to a shift from biologically assisted decomposition (mostly microbial) at short wet:dry hydroperiods to physicochemical release of dissolved organic matter at longer wet:dry hydroperiods. Biologically assisted decomposition was maximized at wet:dry hydroperiods falling within the range of ambient conditions (12-22 consecutive dry days) but then declined under prolonged wet:dry hydroperiods (28 and 56 dry days. Fungal:bacterial ratios showed a similar pattern as biologically assisted decomposition to hydroperiod length. Our results suggest that microbial communities confer functional resilience to altered hydroperiod in tank bromeliad ecosystems. We predict a substantial decrease in biological activity relevant to decomposition under climate scenarios that increase consecutive dry days by 1.6- to 3.2-fold in our study area, whereas decreased frequency of dry periods will tend to increase the physicochemical component of decomposition. Temporary ponds, acknowledged for their conservation value, are colonized by the invasive crayfish Procambarus clarkii. We have tested the consequences of this colonization for the ecosystem under two contrasted scenarios: one single individual arrival or three individuals arrival. We recreated the temporary pond ecosystem in 1 m tanks to investigate the impact of the two crayfish densities. We studied the macrophyte community composition and abundance, chlorophyll a and total suspended solids concentrations, and the diversity and functional composition of micro-crustacean and macro-invertebrate communities. We observed a reduction of macrophyte biomass in experimental crayfish mesocosms in comparison with control tanks, nearly 80 and 40% less in 3 and 1 crayfish/m tanks, respectively. The macrophyte community shifted, followed by a filamentous algae development, an increase of bare sediment and turbidity in crayfish tanks. The macro-invertebrate community suffered a richness loss of 28 and 22%, in 3 and 1 crayfish/m tanks, respectively. Functionally, macro-invertebrate diversity reduction most strongly affected the grazer, detritivore and predator trophic groups. Microcrustaceans seemed not to be affected by the introduction of the crayfish. The introduction of the crayfish greatly altered the ecosystem structure and subsequently the ecosystem functioning. This study seeks to determine whether the relative levels of attachment to galectins 1 and 3 of cells from thyroid tissues embedded in paraffin blocks can differentiate thyroid tumors from normal tissues. A total of 48 thyroid paraffin sample blocks from 4 groups of patients were analyzed: 12 samples served as controls, 12 samples were from patients with thyroid adenoma, 12 samples were from patients with thyroid follicular carcinoma, and 12 samples were from patients with thyroid papillary carcinoma. The relative attachment of cells to galectins 1 and 3 antigens was determined using the InnoCyte™ ECM Cell Adhesion kit at different cell sample concentrations. All of the samples from thyroid tissue preparations showed attachment to galectins 1 and 3. The samples from tissues with a diagnosis of adenoma, follicular and papillary carcinoma showed an increased adherence to galectins 1 and 3 relative to the controls. Significant differences were found between the means of the adherent cells from the adenomas compared with the follicular and papillary carcinoma samples. When the outcomes from the galectins 1 and 3 cell surface binding were compared, no statistical differences were found. The cells from adenoma and carcinoma samples show more adhesion to galectins 1 and 3 than cells from the control samples. The viscoelastic and inertial properties of the arterial wall are responsible for the arterial functional role in the cardiovascular system. Cryopreservation is widely used to preserve blood vessels for vascular reconstruction but it is controversially suspected to affect the dynamic behaviour of these allografts. The aim of this work was to assess the cryopreservation's effects on human arteries mechanical properties. Common carotid artery (CCA) segments harvested from donors were divided into two groups: Fresh ( n = 18), tested for 24–48 h after harvesting, and Cryopreserved ( n = 18) for an average time of 30 days in gas-nitrogen phase, and finally defrosted. Each segment was tested in a circulation mock, and its pressure and diameter were registered at similar pump frequency, pulse and mean pressure levels, including those of normotensive and hipertensive conditions. A compliance transfer function (diameter/pressure) derived from a mathematical adaptive modelling was designed for the on line assessment of the arterial wall dynamics and its frequency response. Assessment of arterial wall dynamics was made by measuring its viscous (η), inertial ( M) and elastic ( E) properties, and creep and stress relaxation time constant (τC and τSR, respectively). The frequency response characterization allowed to evaluate the arterial wall filter or buffer function. Results showed that non-significant differences exist between wall dynamics and buffer function of fresh and cryopreserved segments of human CCA. While biologists have been aware of theexistence of native Mexican trouts for over acentury, they have received little study. Thefew early studies that did much more thanmention their existence began in the 1930s andcontinued into the early 1960s, focusingprimarily on distributional surveys andtaxonomic analyses. Starting in the 1980s theBaja California rainbow trout became thesubject of more detailed studies, but verylittle remains known of mainland trouts of theSierra Madre Occidental. We review earlierstudies and report on our own collections andobservations made between 1975 and 2000. Wepresent newly discovered historical evidencethat leads us to conclude that a 'lost'cutthroat trout, a lineage not previously knownfrom Mexico, was collected more than a centuryago from headwaters of the Río Conchos (amajor tributary of the Rio Grande (= RíoBravo)), a basin not previously considered toharbor a native trout. We review the lastcentury of regional natural resource managementand discuss our own observations of trouthabitats. Impacts of logging, road building andovergrazing are widespread and expanding. Manystreams suffer from heavy erosion, siltationand contamination, and though long-termhydrologic data are generally not available,there is evidence of decreased discharge inmany streams. These problems appear related toregion-wide land management practices as wellas recent regional drought. Trout cultureoperations using exotic rainbow trout haverapidly proliferated throughout the region,threatening genetic introgression and/orcompetition with native forms and predation onthem. Knowledge of distribution, abundance,relationships and taxonomy, not to mentionecology and population biology, of nativetrouts of the Sierra Madre Occidental remainsinadequate. Vast areas of most mainlanddrainages are still unexplored by fishcollectors, and even rudimentary informationregarding basic biology, ecology and populationstructure of stocks remains lacking.Concentrated exploration, research andmanagement of this long overlooked andundervalued resource are all urgently needed.The history of natural resources exploitationthat placed so many native trouts of thewestern United States on threatened andendangered species lists is repeating itself inthe Sierra Madre Occidental. Without concertedaction and development of region-widesocio-economic solutions for current, largelynon-sustainable resource management practices,native Mexican trout gene pools will soon be ingrave danger of extinction. The distribution of cichlids in Lake Bacalar was assessed from January to September 1996, in eight shallow-water (<2 m) stations, using a 20 m long seine net. Temperature, salinity/conductivity and dissolved oxygen were sensor-obtained; depth, submerged vegetation and bottom type were determined by visual inspection. Affinities between stations, based on fish abundance and environmental parameters, were analyzed by multivariate methods. Littoral of Bacalar town was the site with highest richness, diversity and abundance, probably promoted by the additional urban waste of organic matter. PCA defined two station-groups, one associated with muddy substrates, the other one with rocky/sandy bottoms. Cichlids are more abundant over muddy bottoms with abundant submerged vegetation, probably because of the abundance of prey and shelter. [ABSTRACT FROM AUTHOR]

The present report describes an initial characterization of actin from non-infected roots and symbiotic nodules of Phaseolus vulgaris L. cv. Negro damapa. Using anti-actin monoclonal antibodies, a 42-kDa polypeptide was identified in plant extracts. After two-dimensional SDS-PAGE analysis and Western blotting of actin fractions enriched using diethylaminoethyl-resin, the presence of one major isoform of actin in symbiotic nodules and two main isoforms in non-infected roots was revealed. Bacteria mineralization is a promising biotechnological approach to apply in biomaterials development. In this investigation, we demonstrate that Bacillus subtilis 168 induces and influences CaCO3 composites precipitation. Crystals were formed in calcium-carbon non-coupled (glycerol + CaCl2, GLY; or glucose + CaCl2, GLC) and coupled (calcium lactate, LAC; or calcium acetate, ACE) agar-sources, only maintaining the same Ca2+ concentration. The mineralized colonies showed variations in morphology, size, and crystallinity form properties. The crystals presented spherulitic growth in all conditions, and botryoidal shapes in GLC one. Birefringence and diffraction patterns confirmed that all biogenic carbonate crystals (BCC) were organized as calcite. The CaCO3 in BCC was organized as calcite, amorphous calcium carbon (ACC) and organic matter (OM) of biofilm; all of them with relative abundance related to bacteria growth condition. BCC-GLY presented greatest OM composition, while BCC-ACE highest CaCO3 content. Nucleation mechanism and OM content impacted in BCC crystallinity. [ABSTRACT FROM AUTHOR]

The aim of this study was to evaluate the effectiveness of antiretroviral treatment (ART) on the proportion and functions of Th17 and Treg cells in peripheral blood and female genital tract (FGT) respectively. To this aim, samples from 41 HIV-neg, 33 HIV+ ART-naïve and 32 HIV+ ART+ subjects were obtained. In peripheral blood, altered Th17 and Th17/Treg proportions were normalized in HIV+ ART+, but certain abnormal Treg and activated T-cell proportions were still observed. In FGT, abnormal patterns of secretion for Th17-related cytokines were observed in cervical mononuclear cells (CMCs) from HIV+ women, even in those from HIV+ ART+, compared to the HIV-neg group. Moreover, these altered patterns of secretion were associated with diminished levels of CXCL5 and CXCL1 chemokines and with an immunoregulatory skew in the CCL17/CCL20 ratio in ectocervix samples of these women. Finally, ART did not restore proportions of Th17-precursor cells with gut-homing potential in PBMCs, and positive correlations between these cells and the levels of IL-17F and IL-21 production by CMCs may suggest that a better homing of these cells to the intestine could also imply a better restoration of these cells in the female genital tract.
