Background: ER positive breast cancer is currently targeted using various endocrine therapies. Despite the proven therapeutic efficacy, resistance to the drug and reoccurrence of tumor appears to be a complication that many patients deal with. Molecular pathways underlying the development of resistance are being widely studied. Methods and results: In this study, using four established endocrine resistant breast cancer (ERBC) cell lines, we characterized CXCL1 as a secreted factor in crosstalk between ERBC cells and fibroblasts. Protein array revealed upregulation of CXCL1 and we confirmed the CXCL1 expression by real-time qRT-PCR and U-Plex assay. Co-culturing ERBC cells with fibroblasts enhanced the cell growth and migration compared to monoculture. The crosstalk of ERBC cells with fibroblasts significantly activates ERK/MAPK signaling pathway while reparixin, CXCR1/2 receptor inhibitor, attenuates the activity. Reparixin displayed the ERBC cell growth inhibition and the combination treatment with reparixin and CDK4/6 inhibitor (palbociclib and ribociclib) increased these inhibitory effect. Conclusions: Taken together, our study implicates CXCL1 as a critical role in ERBC growth and metastasis via crosstalk with fibroblast and cotargeting CXCR1/2 and CDK4/6 could potentially overcome endocrine resistant breast cancer. Primitive myxoid mesenchymal tumour of infancy (PMMTI) is a rare mesenchymal tumour that typically appears in those under 6 months of age and preferentially affects the deep soft tissues of the trunk and paravertebral spinal regions. PMMTI has only recently been described, and there is scarce literature reporting cases regarding the management paradigm of the tumour. We report the case of an 11-week-old male who presented with bilaterally reduced movement and brisk reflexes in his lower limbs, and irritability. Despite numerous radiological investigations, including MRI, PMMTI was only diagnosed upon biopsy and histopathology. Although PMMTI is known to be relatively unresponsive to chemotherapy, we observed a notable decrease in tumour size after a series of chemotherapy sessions. After two-staged surgical resection of the tumour, the patient is currently stable and under close follow-up. In this article, we aim to report on the patient's clinical presentation, investigations, diagnosis, and treatment, while also discussing the findings from a review of the literature pertaining to future approaches in managing PMMTI. Overall, this case highlights the importance of considering PMMTI in the differential diagnosis of deep soft tissue tumours in young infants and the potential for a combination of chemotherapy and surgical resection to be effective in treating this rare tumour. Thermoplastic parts manufactured via fused filament fabrication (FFF) have limited strength and toughness compared to other types of polymer additive and subtractive manufacturing. Low strength results from poor interlayer adhesion, making FFF parts not suitable for most engineering applications. Post processing solutions, such as annealing, enable healing of these interlayers, thus approaching injection molded parts. Prior work demonstrated a core–shell polycarbonate (PC)—acrylonitrile butadiene styrene (ABS) structured dual material filament to provide thermo-structural stability during annealing of the ABS component; however, annealing was limited to relatively low temperatures (135 °C) and required long annealing times (72 h). In the current work, a PC copolymer with a higher glass transition temperature (173 °C) than conventional PC is processed along with an extrusion-grade ABS into a PC-ABS core–shell filament. This improved dual material filament was printed, annealed, and evaluated via Izod impact testing, ultimately yielding 83% of bulk annealed ABS z-direction strength at an accelerated annealing time (8 h) and higher annealing temperature (155—175 °C). A demonstration part is printed with the dual material filament and annealed at 155 °C for 8 h, resulting in excellent dimensional accuracy, and a ductile failure at 73% higher ultimate load compared to the brittle failure of an as-printed part. This work highlights that material selection and design of a bicomponent filament geometry can lead to parts printed with FFF, with increased strength compared to other post-processing techniques at reduced processing times. We describe a case of a young male patient with KRIT1-driven familial cavernous malformation syndrome who developed multiple brain cavernomas, intracranial bleeding, and persistent seizures. Due to the relentless growth of cavernous malformations and recurrent intracranial bleeds, it was decided to enrol the patient in the "Propranolol for Intracranial Cavernoma" (PICC) pilot trial at our institution. Over the 5-year treatment period with 20 to 40-mg oral propranolol three times daily (TDS), we noted the near-complete arrest of the growth of cavernous malformations with no further evidence of intracranial bleeding or any further seizures. The observed outcome is consistent with the extremely limited published literature on the topic; thus, this case provides important evidence that supports the use of propranolol as a prophylactic treatment for paediatric intracranial cavernomas. We strongly encourage and recommend future prospective randomised controlled trials to definitively assess and characterize the therapeutic utility of propranolol in this patient population. Collaboration and successful teamworking are important components of clinical practise, and these skills should be cultivated early in medical school. The breadth of current medical school curricula means that students often have limited exposure to clinical neurosciences. Since its inception in 2009, the Neurology and Neurosurgery Interest Group (NANSIG) has become a national (UK and Republic of Ireland) example of student and junior doctor synergistic collaboration to deliver educational materials, research, conferences, seminars and workshops, as well as advocating for diversity in this field. Recently, it has expanded to incorporate an international audience and cater for a larger group of young medical professionals. The organisation has overcome numerous challenges and is constantly innovating new approaches to harness the necessary knowledge, skills and network to succeed in a career in neurosciences, neurology and neurosurgery. This article summarises the initiatives undertaken by the group over its first 10 years of existence and its organisational structure, as well as its future plans. [ABSTRACT FROM AUTHOR]

Stiff, strong and tough ultrafine polyethylene fibers that rival the best high performance fibers, but with diameters less than one micron, are fabricated for the first time by 'gel-electrospinning.' In this process, solution concentration and process temperatures are chosen to induce the formation of gel filaments 'in flight,' which are subsequently drawn at high rates as a consequence of the whipping instability. The resulting submicron-diameter fibers exhibited Young's moduli of 73 ± 13 GPa, yield strengths of 3.5 ± 0.6 GPa, and toughnesses of 1.8 ± 0.3 GPa, on average. Among the smallest fibers examined, one with a diameter of 490 ± 50 nm showed a Young's modulus of 110 ± 16 GPa, ultimate tensile strength of 6.3 ± 0.9 GPa, and toughness of 2.1 ± 0.3 GPa, a combination of mechanical properties that is unparalleled among polymer fibers to date. The correlation of stiffness, strength and toughness with fiber diameter is attributed to high crystallinity and crystallite orientation, combined with fewer defects and enhanced chain slip associated with small diameter and high specific surface area. Polyacrylonitrile (PAN) and ambient temperature-curable organopolysilazane were combined to successfully fabricate PAN/polymer-derived ceramic (PDC) hybrid nanofiber separator using a single-step electrospinning process. The amount of added precursor was varied from 10 to 30% to characterize the effects of various loadings on the mechanical, thermal properties, and electrochemical performance on the hybrid membranes. TEM images reveal that all composite fibers have a thin (~5 nm) ceramic-rich sheath layer surrounding each fiber. In addition, there is also ceramic present inside the fiber with the ceramic forming continuous network within the nanofiber at high concentrations. The interconnected ceramic network within the fiber disrupts the polymers ability to properly crystallize, leading to an increase in amorphous regions with an increase in ceramic inclusion. The presence of the ceramic on the surface of the membrane in addition to the increased amorphous regions leads to excellent ionic conductivity and cycling performance. The 30 wt% PDC sample has an ionic conductivity of 1.05 mS cm compared to 0.29 mS cm of pristine PAN separator. All separators with additional PDC content showed increased initial capacity and capacity retention at 0.2C charging and discharging rate, with the 90:10, 80:20, and 70:30 wt% of PAN:PDC showing 89, 90 and 93% capacity retention of graphite/LiCoO full cells over 100 cycles, respectively. In rate capability testing, PAN/PDC fibers demonstrated increased capacity retention even at increased charge rates. The results suggest that the increased ionic conductivity and wetting behavior by both the ceramics within the membrane and on the surface of the membrane are more correlated to an increase in capacity retention and rate capability than the porosity. We report a facile fabrication of porous silicon nanofibers by a simple three-stage procedure. Polymer/silicon precursor composite nanofibers are first fabricated by electrospinning, a water-based spinning dope, which undergoes subsequent heat treatment and then reduction using magnesium to be converted into porous silicon nanofibers. The porous silicon nanofibers are coated with a graphene by using a plasma-enhanced chemical vapor deposition for use as an anode material of lithium ion batteries. The porous silicon nanofibers can be mass-produced by a simple and solvent-free method, which uses an environmental-friendly polymer solution. The graphene-coated silicon nanofibers show an improved cycling performance of a capacity retention than the pure silicon nanofibers due to the suppression of the volume change and the increase of electric conductivity by the graphene. A survey of Human Resource practitioners' perceptions of pre-employment screening procedures was conducted. The results indicated differences between tests in terms of perceived accuracy and offensiveness, as well as other attributes. Regression analyses suggested that beliefs about validity, the number of other companies using these procedures, perceived offensiveness of the test, and ease in passing the test were important factors in the willingness to use a particular test. Conversely, perceptions regarding the discriminatory impact of the test and the cost of using the test generally were not related to willingness to use the test.
