Opinion statement: Esophageal cancer is a global health problem, which is 7th most common and 6th most deadly cancer. It has been the era of immuno-oncology for esophageal cancer management. Radiation therapy has been one of the key local therapeutic approaches for esophageal cancer treatment, while its role in advanced disease is challenging and debatable. There have been emerging clinical and translational studies of radiation therapy in recurrent or metastatic esophageal cancer. Immunotherapy has been established the standard care of 1st and 2nd line systemic therapies of advanced esophageal cancer, and the development of tumor immunity has opened a new chapter for the esophageal cancer radiation therapy. The current review will summarize the classic radiation therapy research in advanced esophageal cancer, as well as the most recent key findings. The subtitles will cover palliative radiotherapy for dysphagia, re-radiation for recurrent disease, oligo-focal disease management and stereotactic radiation therapy, and radiotherapy with immunotherapy. Radiotherapy plays vital role in multidisciplinary management of advanced EC. External or intratumoral irradiation has been used for palliation of dysphagia and improving QOL in esophageal cancer patients traditionally, while recent clinical and technical advance enables radiotherapy to be considered in recurrent or metastatic disease for curation attention. Immunity to previously encountered viruses can alter response to unrelated pathogens. We reasoned that similar mechanism may also involve SARS-CoV-2 and thereby affect the specificity and the quality of the immune response against the virus. Here, we employed high-throughput next generation phage display method to explore the link between antibody immune response to previously encountered antigens and spike (S) glycoprotein. By profiling the antibody response in COVID-19 naïve individuals with a diverse clinical history (including cardiovascular, neurological, or oncological diseases), we identified 15 highly antigenic epitopes on spike protein that showed cross-reactivity with antigens of seasonal, persistent, latent or chronic infections from common human viruses. We observed varying degrees of cross-reactivity of different viral antigens with S in an epitope-specific manner. The data show that pre-existing SARS-CoV-2 S1 and S2 cross-reactive serum antibody is readily detectable in pre-pandemic cohort. In the severe COVID-19 cases, we found differential antibody response to the 15 defined antigenic and cross-reactive epitopes on spike. We also noted that despite the high mutation rates of Omicron (B.1.1.529) variants of SARS-CoV-2, some of the epitopes overlapped with the described mutations. Finally, we propose that the resolved epitopes on spike if targeted by re-called antibody response from SARS-CoV-2 infections or vaccinations can function in chronically ill COVID-19 naïve/unvaccinated individuals as immunogenic targets to boost antibodies augmenting the chronic conditions. Understanding the relationships between prior antigen exposure at the antibody epitope level and the immune response to subsequent infections with viruses from a different strain is paramount to guiding strategies to exit the COVID-19 pandemic. Cumulative evidence over the last decades have supported the role of gum infections as a risk for future major cardiovascular events. The precise mechanism connecting coronary artery disease (CAD) with periodontal findings has remained elusive. Here, we employ next generation phage display mimotope-variation analysis (MVA) to identify the features of dysfunctional immune system that associate CAD with periodontitis. We identify a fine molecular description of the antigenic epitope repertoires of CAD and its most severe form - acute coronary syndrome (ACS) by profiling the antibody reactivity in a patient cohort with invasive heart examination and complete clinical oral assessment. Specifically, we identify a strong immune response to an EBV VP26 epitope mimicking multiple antigens of oral biofilm as a biomarker for the no-CAD group. With a 2-step biomarker test, we stratify subjects with periodontitis from healthy controls (balanced accuracy 84%), and then assess the risk for ACS with sensitivity 71–89% and specificity 67–100%, depending on the oral health status. Our findings highlight the importance of resolving the immune mechanisms related to severe heart conditions such as ACS in the background of oral health. Prospective validation of these findings will support incorporation of these non-invasive biomarkers into clinical practice. Regular physical activity (PA) has been associated with decreased risk of many chronic diseases and increased longevity among older adults. In addition, ageing has been associated with higher levels of different inflammatory biomarkers while the association between inflammatory biomarkers and PA has remained controversial. The aim of the current investigation was to examine the association between blood biomarkers and objectively assessed PA among a sample of healthy older women with different levels of PA engagement. A total of 81 healthy women were recruited. Study participants were allocated to three groups according to accelerometer-obtained PA data. Body composition was assessed with dual-energy X-ray absorptiometry. Fasting blood samples were collected for the measurement of resistin, leptin, tumour necrosis factor alpha (TNFα) and C-reactive protein (CRP) concentrations. There were no significant differences between groups for resistin, leptin, TNFα and for CRP concentrations; however, higher moderate-to-vigorous physical activity (MVPA) groups tended to have lower level of blood biomarker concentrations. There was a significant negative relationship between resistin and steps per day. Inverse association between leptin and MVPA was significant after controlling for age. In multivariate stepwise linear regression analysis, steps per day were the strongest independent predictor for resistin, whereas for leptin, TNFα and CRP the strongest independent predictor was whole body fat mass. In conclusion, this study demonstrated negative association between resistin concentration and steps per day. Sedentary time and light PA had no relationship with resistin, leptin, TNFα or CRP concentrations. [ABSTRACT FROM AUTHOR]

Lake Ülemiste, the drinking water reservoir of Estonia’s capital city Tallinn, was biomanipulated by manual removal of cyprinids in 2004–2006 and its impact on water quality in the vegetation period was studied. A total biomass of 156 tonnes corresponding to 160 kg ha−1 of fish, predominantly cyprinids, were removed. A decline in the unit catches of fishing was observed. The removed fish biomass versus phosphorus concentration of the lake was considered sufficient to reduce the impact of cyprinids on water quality. The phosphorus removed within fish biomass corresponded to 38 μg l−1 and 21% of the external phosphorus load of the fishing period. The mean total phosphorus concentration dropped from >50 to ≤36 μg l−1. However, the densities of planktivorous young-of-the-year percids remained high and the role of zooplankton grazing in improving water quality was found non-significant or transient. The cladocerans biomass decreased and the small-sized Daphnia cucullata remained almost the only daphnid in Lake Ülemiste during and after the manipulation. Predomination of filamentous cyanobacteria was replaced by a more diverse phytoplankton composition and co-domination of micro- and pico-sized colonial cyanobacteria during summer. Mean phytoplankton biomass decreased from 15 to 6 mg l−1 primarily as a result of decreased in-lake TP availability. The Secchi disc transparency increased only in May 2005–2007. The effects of coincidental events, a decline of external loading of phosphorus and a simultaneous flushing induced by heavy rainfall, on lake water quality are discussed with some implications to the future management of the reservoir. The increasing availability of high-quality reference genomic sequences has created a demand for ways to survey the sequence differences present in individual genomes. Here we describe a DNA sequencing method based on hybridization of a universal panel of tiling probes. Millions of shotgun fragments are amplified in situ and subjected to sequential hybridization with short fluorescent probes. Long fragments of 200 bp facilitate unique placement even in large genomes. The sequencing chemistry is simple, enzyme-free and consumes only dilute solutions of the probes, resulting in reduced sequencing cost and substantially increased speed. A prototype instrument based on commonly available equipment was used to resequence the Bacteriophage λ and Escherichia coli genomes to better than 99.93% accuracy with a raw throughput of 320 Mbp/day, albeit with a significant number of small gaps attributed to losses in sample preparation. Cell cycle progression is dependent on the sequential activity of cyclin-dependent kinases (CDKs). For full activity, CDKs require an activating phosphorylation of a conserved residue (corresponding to Thr160 in human CDK2) carried out by the CDK-activating kinase (CAK). Two distinct CAK kinases have been described: in budding yeast Saccharomyces cerevisiae, the Cak1/Civ1 kinase is responsible for CAK activity. In several other species including human, Xenopus, Drosophila and fission yeast Schizosaccharomyces pombe, CAK has been identified as a complex homologous to CDK7—cyclin H (Mcs6—Mcs2 in fission yeast). Here we identify the fission yeast Csk1 kinase as an in vivo activating kinase of the Mcs6—Mcs2 CAK defining Csk1 as a CAK-activating kinase (CAKAK). [ABSTRACT FROM AUTHOR]

The content and isotopic composition of uranium extracted under various leaching and extraction conditions from the dyctionemic shale from Estonian deposits are determined by alpha-spectrometry. The interaction of uranium with organic matter (OM) was confirmed by a correlation between the specific radioactivity of uranium and contents of OM in the fractions obtained by electrostatic separation of the ground shale powder. Correlation between the results of two different analytical schemes is shown. The thickness of uranium phase layer deposited on organic matter in two parts of the seam is compared. Activating phosphorylation of cyclin-dependent kinases (Cdks) is mediated by at least two structurally distinct types of Cdk-activating kinases (Caks): the trimeric Cdk7-cyclin H-Mat1 complex in metazoans and the single-subunit Cak1 in budding yeast. Fission yeast has both Cak types: Mcs6 is a Cdk7 ortholog and Csk1 a single-subunit kinase. Both phosphorylate Cdks in vitro and rescue a thermosensitive budding yeast CAK1 strain. However, this apparent redundancy is not observed in fission yeast in vivo. We have identified mutants that exhibit phenotypes attributable to defects in either Mcs6-activating phosphorylation or in Cdc2-activating phosphorylation. Mcs6, human Cdk7 and budding yeast Cak1 were all active as Caks for Cdc2 when expressed in fission yeast. Although Csk1 could activate Mcs6, it was unable to activate Cdc2. Biochemical experiments supported these genetic results: budding yeast Cak1 could bind and phosphorylate Cdc2 from fission yeast lysates, whereas fission yeast Csk1 could not. These results indicate that Mcs6 is the direct activator of Cdc2, and Csk1 only activates Mcs6. This demonstrates in vivo specificity in Cdk activation by Caks. [ABSTRACT FROM AUTHOR]

The trimeric Cdk7-cyclin H-Mat1 complex comprises the kinase subunit of basal transcription factor TFIIH and has been shown to function as a cyclin-dependent kinase (Cdk)-activating kinase. Herein we report that disruption of the murine Mat1 gene leads to periimplantation lethality coincident with depletion of maternal Mat1 protein. In culture, Mat1-/- blastocysts gave rise to viable post-mitotic trophoblast giant cells while mitotic lineages failed to proliferate and survive. In contrast to wild-type trophoblast giant cells, Mat1-/- cells exhibited a rapid arrest in endoreduplication, which was characterized by an inability to enter S phase. Additionally, Mat1-/- cells exhibited defects in phosphorylation of the C-terminal domain (CTD) of RNA polymerase II on both Ser5 and Ser2 of the heptapeptide repeat. Despite this, Mat1-/- cells demonstrated apparent transcriptional and translational integrity. These data indicate an essential role for Mat1 in progression through the endocycle and suggest that while Mat1 modulates CTD phosphorylation, it does not appear to be essential for RNA polymerase II-mediated transcription.
