The policy and practice of ecological restoration and conservation in China obtained some remarkable results. For example, Sphagnum moss growing on abandoned farmland, which was peatland before agricultural use, has rapidly expanded the wetland area in SW China. Microorganisms such as testate amoebae are sensitive to environmental change and thus have been widely used as ecological indicators in various habitats. We analyzed differently aged Sphagnum growing plots on a Sphagnum growing farmland and natural Sphagnum plots in SW China to examine how Sphagnum-dwelling testate amoeba communities and corresponding protozoic silicon (Si) pools respond to ecological restoration practice. We found that abundance, taxon richness, and diversity of testate amoebae were higher in Sphagnum growing farmland plots compared to natural Sphagnum plots. Protozoic Si pools showed an increase with Sphagnum growing time representing increased Si accumulation by idiosomic testate amoeba shells. However, protozoic Si pools were negatively correlated with taxon richness and diversity of testate amoebae. Our results showed that (i) natural Sphagnum plots were not characterized by the expected higher biodiversity of testate amoebae compared to Sphagnum growing plots and (ii) consequently protozoic Si pool quantity in natural Sphagnum plots was less driven by biodiversity of testate amoebae than expected. We concluded our results to underline the value of (i) environmental restoration policy in general and (ii) testate amoeba communities and corresponding protozoic Si pools for Si cycling in restoration areas of peatlands in particular. Based on our results, we recommend a sustainable cultivation of Sphagnum moss and an additional establishment of protected areas, where no Sphagnum harvesting occurs. These protected Sphagnum areas might represent hot spots of undisturbed testate amoeba communities and corresponding protozoic Si pools and thus of microbial Si cycling. [ABSTRACT FROM AUTHOR]

We present a ~ 40–year record of environmental change in the Swan Oxbow, Yangtze River, China, inferred from testate amoeba and sedimentary pigment data, combined with remote sensing and analysis of local socio–economic growth. These data indicate there were several distinct phases of aquatic conditions linked to human activities in the region: (1) Between ca. AD 1970 and 1984, there may have been some exchange of water and organic matter between the Swan Oxbow and the main river channel, following initial hydrologic disconnection in 1972. The lake area was relatively large in the early phase after the oxbow first formed, and the trophic state generally increased during that time frame, (2) From ca. AD 1984 to 1992, the lake area was about a third smaller in size, while the human population and GDP increased about 10% and 10x, respectively, compared to values in 1975. The nutrient status (inferred from testate amoeba and pigment data) increased, owing to the greater discharge of nutrients and separation of the Swan Oxbow from the main Yangtze River, which reduced water supply and increased sedimentation in the oxbow, (3) From ca. AD 1992 to 2003, the lake continued to diminish in size, to an area < 20 km2, except in 1998, when a major flood occurred. The testate amoeba and pigment data suggest that water quality had improved by that time, which probably reflects efforts to control agricultural and industrial activities, including establishment of two national reserves in 1992 and 1993, created to protect the rare Père David's deer (Elaphurus davidianus) and the freshwater Baiji dolphin (Neophocaena phocaenoides asiaeorientalis), (4) From ca. AD 2003 to 2005, the water area remained relatively unchanged, owing to construction of a dam following the 1998 flood. Occurrence of testate amoeba species Difflugia biwae and D. tuberspinifera, however, indicates that water pollution and eutrophication had been controlled in Swan Oxbow. Testate amoebae are micro-organisms characterized by an agglutinated or autogenous shell enclosing the cytoplasm. Testate amoebae have been widely proposed as valuable bioindicators in a range of ecosystems (such as soils, peatlands and lakes). The use of testate amoebae as bioindicators of water quality in aquatic ecosystems is much less developed than for other microorganisms and previous research is geographically restricted. We investigated a large range of environmental variables and their relations to testate amoeba communities from 37 shallow lakes in the middle and lower Yangtze River Plain of China. Multiple factor analysis, redundancy analysis (RDA) and a forward-selection approach were used to explore the overall community structure and the links between the environmental variables and testate amoeba composition. Results showed that testate amoebae are widely distributed in most of the lakes despite often high levels of pollution. Our data highlight some links between water quality variables and testate amoeba communities. In particular, our results suggest that heavy metals have a role in shaping testate amoeba assemblage structure, although correlations are comparatively weak, perhaps due to lagged responses. Our results support previous suggestions that testate amoebae may be useful bioindicators in aquatic ecosystems but emphasize the need for an improved mechanistic understanding. Testate amoebae are a diverse and abundant group of protozoa that constitute a large proportion of biomass in many ecosystems and probably fill important roles in ecosystem function. These microorganisms have attracted the interest of paleoecologists because the preserved shells of testate amoebae and their known hydrological preferences enable reconstruction of past hydrological change. In ombrotrophic peatlands, surface wetness reflects hydroclimate, so testate amoebae can play an important role in reconstruction of Holocene climate change. Previous studies, however, have been geographically restricted, mostly to North America and Europe. We studied the ecology of testate amoebae in peatlands from central China in relation to hydrology, pH and metal concentrations. We found that testate amoeba community structure was correlated with depth to water table (DWT) and that the hydrological preferences of species generally matched those of previous studies. We developed a weighted average DWT transfer function that enables prediction of water table depth with a cross-validated mean error of <5 cm. Our results demonstrate the potential for using testate amoebae to reconstruct paleohydrology in China. The middle Yangtze Reach is one of the most developed regions of China. As a result, most lakes in this area have suffered from eutrophication and serious environmental pollution during recent decades. The aquatic biodiversity in the lakes of the area is thus currently under significant threat from continuous human activities. Testate amoebae (TA) are benthic (rarely planktonic) microorganisms characterized by an agglutinated or autogenous shell. Owing to their high abundance, preservation potential in lacustrine sediments, and distinct response to environmental stress, they are increasingly used as indicators for monitoring water quality and reconstructing palaeoenvironmental changes. However this approach has not yet been developed in China. This study presents an initial assessment of benthic TA assemblages in eight lakes of Lake Donghu in the region of Wuhan, China. Testate amoeba community structure was most strongly correlated to water pH. In more alkaline conditions, communities were dominated by Centropyxis aculeata, Difflugia oblonga, Pontigulasia compressa, Pon. elisa and Lesquereusia modesta. These results are consistent with previous studies and show that TA could be useful for reconstructing past water pH fluctuations in China. This study investigates the testate amoeba communities of a large peatland in Central China. The ecology and seasonal variability of testate amoeba communities were studied during 2009-2010. Investigation of environmental controls using ordination showed that the relationship between testate amoeba communities and depth to water table (DWT) and pH are extremely weak. The small proportion of variance explained by water table depth here (only 1.9% in the full data) shows that the hydrological control is weaker than we expected in this peatland, and weaker than any study we are aware of using a similar methodology. Attempts to develop species-environment (transfer function) models or identify indicator species for future palaeoecological studies were unsuccessful. Previous large-scale studies of peatland testate amoeba ecology have been largely restricted to Europe and North America and results have been relatively consistent among studies. Our results contrast with this consensus and suggest that at least in minerotrophic peatlands in China testate amoeba communities may be primarily controlled by different environmental variables. In China, testate amoebae have been relatively little studied but may prove to be valuable for a variety of applications in palaeoecology and biomonitoring and much further work is required. The article focuses on a study which examined the biodiversity and hydro-ecological selection of testate amoebae from Dajiuhu peatland in Shennongjia, Central China. Samples were collected from microsites within the peatland. Results of the study showed high variations in the morphology of testate amoebae in 136 Sphagnum samples. The study observed that variations in shell morphology can be attributed to different moisture conditions in their habitat.

The distributions of microbial tetraether lipids, i.e. glycerol dialkyl glycerol tetraethers (GDGTs), were investigated in a variety of Chinese soils featured by a wide range of pH values in different climate zones. The bacterial branched GDGTs and archaeal isoprenoid GDGTs were found to be widely distributed in Chinese soils and their relative abundance was controlled primarily by soil pH. The bacterial bGDGTs usually dominate over archaeal iGDGTs in abundance when the soil pH is below 8.0. However, archaeal iGDGTs become dominant and the abundance of halophilic archaea characterized by GDGT V without cyclopentyl ring increases dramatically in the alkaline soil with pH greater than 8.0. The relative abundance of archaeal iGDGTs to bacterial bGDGTs was defined here as RAI index. The RAI index decreases with elevated soil pH and a linear correlation is found between them, indicating that the relative abundance of archaeal iGDGTs to bacterial bGDGTs increases with increased pH. The RAI index thus could be a novel proxy of the pH values of paleosol. The terrestrial input index, BIT (Branched and Isoprenoid Tetraethers Index), decreases with increased soil pH, suggesting that soil pH should be taken into consideration when the terrestrial contribution to marine sediments is estimated by BIT index.
