Sexing bird species with monomorphic plumage is generally challenging, and sexual size dimorphism (SSD) is often used to develop morphometric-based sexing tools, e.g., using discriminant functions. Within species, local selection pressures, age-related and season-related growth may, however, induce geographical and temporal variations in body size and SSD. Such variations may complicate the development of reliable morphometric-based sexing methods at a broad scale. We first investigated body size variations in a migratory shorebird species with wide breeding and wintering ranges, the Sanderling Calidris alba, within the two breeding populations (Greenland and Russia) and three staging/wintering populations (United Kingdom, Iceland and Mauritania), which belong to the same flyway. Then, for samples from each region, we tested whether site-specific (i.e., "regional") functions performed better than functions developed for birds from the other sites (i.e., "foreign" functions) or than an overall ("flyway") function that combined all sampled individuals. We found minor variations in SSD between regions, but significant differences in body size between sexes and regions. Females were larger than males and, for instance, breeders had longer wings than staging and wintering birds. Regional functions had similar sexing efficiency as any other functions applied to sample from each region, except for Western Africa where the regional function performed slightly better than some of the other functions. Furthermore, the flyway function developed after merging all subsamples had a similar efficiency than the regional functions, i.e., from 75.4% to 90% of correct sex assignment depending on the region. Given the small or lack of benefit in using regional functions, we conclude that the flyway function can be used reliably to sex Sanderlings measured at different sites, years or seasons within the East Atlantic flyway. Cancer survivors may experience long-term cardiovascular complications due to chemotherapeutic drugs such as doxorubicin (DOX). The exact mechanism of delayed DOX-induced cardiotoxicity has not been fully elucidated. Sex is an important risk factor for DOX-induced cardiotoxicity. In the current study, we identified sex differences in delayed DOX-induced cardiotoxicity and determined the underlying molecular determinants of the observed sexual dimorphism. Five-week-old male and female mice were administered intraperitoneal injections of DOX (4 mg/kg/week) or saline for 6 weeks. Echocardiography was performed 5 weeks after the last dose of DOX to evaluate cardiac function. Thereafter, mice were sacrificed and gene expression of markers of apoptosis, senescence, and inflammation was measured by PCR in hearts and livers. Proteomic profiling of the heart from both sexes was conducted to determine differentially expressed proteins (DEPs). Only DOX-treated male, but not female, mice demonstrated cardiac dysfunction, cardiac atrophy, and upregulated cardiac expression of Nppb and Myh7. No sex-related differences were observed in DOX-induced expression of most apoptotic, senescence, and pro-inflammatory markers. However, the gene expression of Trp53 was significantly reduced in hearts of DOX-treated female mice only. The anti-inflammatory marker Il-10 was significantly reduced in hearts of DOX-treated male mice only, while the pro-inflammatory marker Il-1α was significantly reduced in livers of DOX-treated female mice only. Gene expression of Tnf-α was reduced in hearts of both DOX-treated male and female mice. Proteomic analysis identified several DEPs after DOX treatment in a sex-specific manner, including anti-inflammatory acute phase proteins. This is the first study to assess sex-specific proteomic changes in a mouse model of delayed DOX-induced cardiotoxicity. Tephritid fruit fly pests pose an increasing threat to the agricultural industry due to their global dispersion and a highly invasive nature. Here we showcase the feasibility of an early-detection SEPARATOR sex sorting approach through using the non-model Tephritid pest, Ceratitis capitata. This system relies on female-only fluorescent marker expression, accomplished through the use of a sex-specific intron of the highly-conserved transformer gene from C. capitata and Anastrepha ludens. The herein characterized strains have 100% desired phenotype outcomes, allowing accurate male–female separation during early development. Overall, we describe an antibiotic and temperature-independent sex-sorting system in C. capitata, which, moving forward, may be implemented in other non-model Tephritid pest species. This strategy can facilitate the establishment of genetic sexing systems with endogenous elements exclusively, which, on a wider scale, can improve pest population control strategies like sterile insect technique. Major depressive disorder (MDD) is one of the most important causes of disability worldwide. While recent work provides insights into the molecular alterations in the brain of patients with MDD, whether these molecular signatures can be associated with the expression of specific symptom domains remains unclear. Here, we identified sex-specific gene modules associated with the expression of MDD, combining differential gene expression and co-expression network analyses in six cortical and subcortical brain regions. Our results show varying levels of network homology between males and females across brain regions, although the associations between these structures and the expression of MDD remain highly sex specific. We refined these associations to several symptom domains and identified transcriptional signatures associated with distinct functional pathways, including GABAergic and glutamatergic neurotransmission, metabolic processes and intracellular signal transduction, across brain regions associated with distinct symptomatic profiles in a sex-specific fashion. In most cases, these associations were specific to males or to females with MDD, although a subset of gene modules associated with common symptomatic features in both sexes were also identified. Together, our findings suggest that the expression of distinct MDD symptom domains associates with sex-specific transcriptional structures across brain regions. Glial Fibrillary Acidic Protein (GFAP) and Ubiquitin C-terminal hydrolase (UCH-L1) have been FDA-approved for clinical use in mild and moderate traumatic brain injury (TBI). Understanding sex differences in their diagnostic accuracy over time will help inform clinical practice. We sought to evaluate the sex differences in the temporal profile of GFAP and UCH-L1 in a large cohort of trauma patients presenting to the emergency department. To compare the biomarkers' diagnostic accuracy in male versus female patients for detecting mild TBI (MTBI), and traumatic intracranial lesions on head CT. This prospective cohort study enrolled female and male adult trauma patients presenting to a Level 1 Trauma Center. All patients underwent rigorous screening to determine whether or not they had experienced a MTBI. Of 3025 trauma patients assessed, 1030 met eligibility criteria and 446 declined. Initial blood samples were obtained in 584 patients enrolled within 4 h of injury. Repeated blood sampling was conducted at 4, 8, 12, 16, 24, 36, 48, 60, 72, 84, 96, 108, 120, 132, 144, 156, 168, and 180-h post-injury. The main outcomes included the diagnostic accuracy in detection of MTBI and traumatic intracranial lesions on head CT scan. A total of 1831 samples were drawn in 584 patients over 7 days, 362 (62%) were male and 222 (38%) were female. The pattern of elevation was similar in both sexes. Although the pattern of elevation was similar between male and female for both biomarkers, male patients had significantly higher concentrations of UCH-L1 compared to female patients at several timepoints post-injury, particularly within 24 h of injury. There were no significant differences in diagnostic accuracy for detecting MTBI or for detecting CT lesions between male and female patients at any timepoint for both GFAP and UCH-L1. Although patterns of GFAP and UCH-L1 release in trauma patients over a week post-injury was similar between the sexes, there were significantly higher concentrations of UCH-L1 in males at several timepoints post-injury. Despite this, the overall diagnostic accuracies of both GFAP and UCH-L1 over time for detecting MTBI and CT lesions were not significantly different between male and female trauma patients. This review presents information on biochemical features of spermatozoa bearing X or Y chromosome, enabling production of a sperm fraction with pre-defined sex chromosome. The almost only technology currently used for such separation (called sexing) is based on the fluorescence-activated cell sorting of sperm depending on DNA content. In addition to the applied aspects, this technology made it possible to analyze properties of the isolated populations of spermatozoa bearing X or Y chromosome. In recent years, existence of the differences between these populations at the transcriptome and proteome level have been reported in a number of studies. It is noteworthy that these differences are primarily related to the energy metabolism and flagellar structural proteins. New methods of sperm enrichment with X or Y chromosome cells are based on the differences in motility between the spermatozoa with different sex chromosomes. Sperm sexing is a part of the widespread protocol of artificial insemination of cows with cryopreserved semen, it allows to increase proportion of the offspring with the required sex. Incorporating sex ratios of nestlings into population viability studies increases knowledge of overall health of endangered populations. Currently, a reliable non-invasive method to identify the sex of golden eagle nestlings is not available; however, claims are commonly made based on morphology. Ten biometric measurements from 43 Scottish golden eagles aged 2–7.5 weeks were assessed to see if sex could actually be determined using this non-invasive methodology. Sex was confirmed via molecular analysis of blood samples. Discrete and principal component analyses of the different biometrics could not correctly determine individual nestling sex. Therefore, despite being more invasive, molecular sexing remains the recommended tool of choice for accurate sex identification of Scottish golden eagle nestlings younger than 7.5 weeks of age. This has important implications for golden eagle field studies where empirical morphological measurements are frequently and typically taken, but we have shown are not reliable in determining the sex of such young nestlings. The forces which shape avian species' vocalization and coloration continue to draw scientific attention. The White-throated Kingfisher, (Halcyon smyrnensis), is a non-passerine, resident, generalist predator characterized by elaborate colors and vocalizations and is highly abundant throughout its species range. However, despite these characteristics this species has been relatively unstudied. Here we studied the White-throated Kingfisher population in the Hula Valley, Israel, a population residing at the western edge of this species' range. Using individual marking, nest surveys, acoustic recordings, digital photography, spectrometry and molecular sexing we describe this resident species' breeding ecology, morphology and vocal repertoire. We further examine whether these traits are sexually dimorphic and whether they correlate with measures of reproductive success (clutch size). We show that the breeding ecology (e.g. incubation time) differs from some of the reported studies. We further demonstrate, for the first time, that this non-passerine species has a vocal repertoire of at least 13 distinct vocalizations; both males and females seem to produce the majority of these vocalizations. A specific vocalization (long trill) is sexually dimorphic, and the rate of the trill correlates with male reproductive success. The elaborate coloration also shows significant sexual dimorphism: males have brighter blueish dorsal feathers and darker saturated brown ventral feathers. Taken together, specifically with this species being non-passerine, the unique ecology of this western population and the various traits which show sexual dimorphism define this resident species as a unique abundant model species for examining various ecological and evolutionary processes. CRISPR-based genetic engineering tools aimed to bias sex ratios, or drive effector genes into animal populations, often integrate the transgenes into autosomal chromosomes. However, in species with heterogametic sex chromsomes (e.g. XY, ZW), sex linkage of endonucleases could be beneficial to drive the expression in a sex-specific manner to produce genetic sexing systems, sex ratio distorters, or even sex-specific gene drives, for example. To explore this possibility, here we develop a transgenic line of Drosophila melanogaster expressing Cas9 from the Y chromosome. We functionally characterize the utility of this strain for both sex selection and gene drive finding it to be quite effective. To explore its utility for population control, we built mathematical models illustrating its dynamics as compared to other state-of-the-art systems designed for both population modification and suppression. Taken together, our results contribute to the development of current CRISPR genetic control tools and demonstrate the utility of using sex-linked Cas9 strains for genetic control of animals. Chub mackerel Scomber japonicus and blue mackerel S. australasicus have become important aquaculture target species in Japan. For an efficient and stable aquaculture production of mackerels, it is desirable to work with a female-biased broodstock and thus to be able to manage precisely their sex ratio. As these two mackerel species do not have any external sexual dimorphism, direct biopsy of the gonads or measurement of sex hormone plasma levels is used in research to determine their phenotypic sex. However, these methods are associated with handling stress that results in gonadal atresia. Besides, it is difficult to identify sex using these methods in immature individuals. Therefore, genetic sexing would be desirable in these species to overcome these problems. In this study, we developed molecular sex identification tools for both chub and blue mackerels using sex-specific single-nucleotide variations (SNVs) identified by pool-sequencing (pool-seq) analyses. Genomic DNA was isolated from 30 males and females, in each species, and sequenced as pooled samples. Results of these pool-seq analyses identified a clear XX/XY male heterogametic sex-determination system in blue mackerel and a ZZ/ZW female heterogametic system in chub mackerel. Using this pool-seq information, we designed primer sets for genetic sex identification in both species. Results demonstrated that these primer sets are robust and accurate in assessing the genotypic sex of both blue and chub mackerels (accuracy > 99.5% in the chub, and > 94.5% in the blue mackerel) in various geographical samples of different ages. Monitoring trends in the abundance of furbearers may be challenging, particularly at the spatio-temporal scales relevant for management. As such, wildlife managers often rely on harvest records to identify broad-scale harvest patterns and trends. Yet, the reliability of harvest records is often unknown. Analyses of harvest records to determine quotas and assess sustainability need to account for error rates when they are > 0. We evaluated the accuracy of trapper-reported sex of lynx (Lynx canadensis; n = 185) and wolverine (Gulo gulo; n = 467)—two meso-carnivores commonly targeted by fur trappers in northwestern Canada and Alaska—by comparing that to sex determined via necropsies of the same carcasses. Overall error rates differed significantly between wolverine (5%) and lynx (13%). Error rates were sex-biased for wolverine, but not lynx. Body size did not affect error rates for either species. Our data demonstrated species- and sex-specific error rates in the sex reported in harvest records. Error rates for wolverine (5%) were likely trivial for determining harvest sustainability because sex-based bias was small, given that overall accuracy was high. While error rates were greater for lynx (13%), there was no sex-based bias in trapper-reported sex. Because accuracy was lower for lynx, managers should exercise caution when using trapper-reported sex to conduct population analyses or assess harvest sustainability. Managers should be particularly interested in error rates of harvested species that exhibit relatively little sexual size dimorphism and lack obvious genitalia, similar to lynx. We recommend assessing error in trapper-reported sex prior to analysis of harvest records, as well as ongoing education with trappers to increase their ability to reliably determine the sex of animals they harvest. Temperature-dependent sex determination, present in most turtle species, is a mechanism that uses temperature to direct the sex of the embryo. The rapid increase of global temperatures highlights the need for a clear assessment of how sex ratios of organisms with TSD are affected. In turtles with TSD, quantifying primary sex ratios is challenging because they lack external dimorphism and heteromorphic sex chromosomes. Here we describe a new technique used to identify sex in neonate turtles of two TSD species, a freshwater turtle (Trachemys scripta) and a marine turtle (Caretta caretta) via analysis of small blood samples. We used an immunoassay approach to test samples for the presence of several proteins known to play an important role in sex differentiation. Our results show that Anti-Mullerian Hormone (AMH) can be reliably detected in blood samples from neonate male turtles but not females and can be used as a sex-specific marker. Verification of sex via histology or laparoscopy revealed that this method was 100% reliable for identifying sex in both T. scripta and C. caretta 1–2 day-old hatchlings and 90% reliable for identifying sex in 83–177 day-old (120–160 g) loggerhead juveniles. The method described here is minimally invasive, and for the first time, greatly enhances our ability to measure neonate turtle sex ratios at population levels across nesting sites worldwide, a crucial step in assessing the impact of climate change on imperiled turtle species. The lack of detailed life history (LH) information (e.g. age, growth, size at maturity, sex composition etc.) for many species of conservation importance limits the implementation of appropriate conservation measures. Typically, LH information is acquired using lethal sampling techniques, which undermines the goal of conservation. This is particularly problematic for many shark species that have low fecundity and slow growth rates. Here we tested the use of non-invasive laser photogrammetry to measure body morphometry in vivo. We used random forest classification models to identify allometric relationships (ratios between body measurements) that discriminated between the sex and stage of sexual maturity of Scyliorhinus canicula. We coupled the use of allometric ratios (determined from cadavers) with parallel laser photogrammetry, in order to collect total length (TL) and finer scale morphometrics from 37 free-swimming individuals. TL measurements proved to be accurate (SE = 5.2%) and precise (CV = 1.8%), and did not differ significantly from the known TL of the respective animal ( t36 = 0.7, P = 0.5). Conditional Inference tree model predictions of free-swimming sharks correctly predicted 100% of mature males and 79% of immature males. Our results suggest that when used together, allometric ratios and parallel laser photogrammetry have the potential to be a promising alternative to collect essential life history information from free swimming animals and avoids the need for destructive sampling. D. persimilis courtship shows some flexibility and courting males sometimes perform an elaborate postural display in addition to the standard courtship behaviours shared by most Drosophila species. This postural display includes the acrobatic contortion and tremulation of their abdomen, accompanied by the generation of substrate-borne vibrations, and they proffer a nutritional droplet to the female. Here, we use courtship and choice assays to ask what triggers this display and what advantages males may gain from it during courtship. In pair assays, we found no differences in the courtship duration and copulation success between displaying and non-displaying males. In trio assays, however, the female always mated with the male who performed the display. To investigate what promotes the male display, we varied the level of receptivity of the female and studied the impact of a second male. We found that rejection by the female does not induce the male to display, contrary to what was previously suggested. We present evidence that the male display is in fact promoted by the presence of an attentive and sexually receptive female and the absence of male competition, with the greatest exhibition rate obtained if the courted female is starved. The adenohypophysis comprises six types of endocrine cells, including PIT1-lineage cells such as growth hormone (GH)-producing cells and heterogeneous non-endocrine cells, such as pituitary stem/progenitor cells as a source of endocrine cells. We determine the expression of characteristic stem cell marker genes, including sex-determining region Y-box 2 ( Sox2), in mouse pituitary-derived non-endocrine cell lines Tpit/E, Tpit/F1 and TtT/GF. We observed high expression of fibroblast growth factor (FGF) receptors in Tpit/F1 cells, which we characterised by cultivation in medium containing a basic FGF and B27 supplement as used for neural stem-cell differentiation. A 4-day cultivation of Tpit/F1 produced floating embryonic stem-cell-like clumps accompanied by a three-fold increase in Sox2 expression. Passages in these clumps maintained the proliferative activity and Sox2 expression levels. After 10 days of cultivation, Tpit/F1 cell clumps were immuno-positive for SOX2 and Ki67 (proliferation marker) and loosely attached to the well bottom. An additional 10 days of cultivation induced the emergence of GH-positive/pituitary-specific transcription factor (PIT1)-negative cells showing migration from the clumps. Pit1 overexpression in attached cells could not induce GH production. Finally, we confirmed the presence of PIT1-negative GH-producing cells (3.2-7.7 % of all GH-positive cells) in rat pituitary. In the crustacean Branchipus schaefferi cercopods are brightly coloured caudal rami at the end of the abdomen. The specific life environment of B. schaefferi (highly turbid puddles) may suggest that besides their mechanical function, cercopods may also play an ornamental role to attract a potential mate. We performed standardized major axis regression analyses of cercopod length against body length to determine the allometry. Each allometric slope of cercopod was compared with the median of slopes of non-sexual body parts: eye diameter, epipodit, and antennulae lengths. We also examined the relative differences in cercopods colouration between males and females within a population. Cercopods scaled slightly more positively when compared to both median slope of non-sexual traits as well as to typically used slope value 1. The difference in cercopods colouration between the sexes showed a strong sexual dimorphism. Sexual dimorphism in colouration and slightly positive allometry of cercopods support the hypothesis that this organ might be under the sexual selection and function as an ornament advertising fitness of an individual or a signal of sexual maturity of a male in highly turbid water. DNA extracted from faeces may be a valuable source of information about the animal itself, as well as its microflora. An isolated reindeer population from Svalbard ( Rangifer tarandus platyrhynchus) was tested for the presence of Shiga toxin encoding genes in the collected faecal samples. Even though the reindeers were not interacting with any other ruminants, which are considered to be a major reservoir of Shiga toxin containing bacteria, the stx1 gene was detected in 9 out of 10 tested samples, and the stx2 subtype c was found in five tested samples. This is an exceptionally high proportion, especially in the case of stx1, when compared to those observed in semi-domesticated or wild cervid populations in less remote locations. Distribution of the investigated genes in a small, local population indicates a different pattern of transmission, which seems to favour bacteria carrying the stx1 genes over those carrying the stx2 genes. The overall high percentage of animals with microbiota containing the stx genes suggests an important role of these genes in such an extreme environment for either hosts or their gut bacteria. Additionally, male-specific DNA found in the faeces was isolated in order to establish a given animal's sex. PCR based on two pairs of primers, DBY7 and DBY8 gave the expected length of product characteristic for Y chromosome. The results of molecular sexing were consistent with field observations. In sex-role-reversed species, females compete for resources (e.g., mates) more intensively than do males. However, it remains unclear whether these species exhibit sex differences in the intensity of aggressive behavior in the context of within-sex contests. Cichlid fish in the genus Julidochromis exhibit intraspecific variation in mating systems, ranging from monogamy to cooperative polyandry with sex-role reversal. In the study reported here, we observed aggressive interactions among three same-sex individuals in Julidochromis regani in the laboratory and tested whether inter-female aggression was more intense than inter-male aggression. Although difference in body size strongly determined the direction of aggression in fish, aggression by a smaller-sized individuals toward larger ones was occasionally observed. This type of aggression was common between individuals of a similar body size (≤5 mm) and occurred more frequently among females than males. In contrast, differences in body size and sex did not affect the frequency of aggression by larger-sized individuals against smaller ones. Bidirectional aggression (i.e., mouth fighting) occurred frequently when two individuals had similar body size, and there was no difference in its frequency between sexes. However, temporal analysis showed that females performed bidirectional aggression more persistently than males. Microsatellite loci are widely used in ecological and evolutionary studies to assess inbreeding, genetic parentage and population structure. Such loci are often optimised in multiplexes to allow for economical and efficient use. Here, we tested 11 microsatellite loci designed for use in European bee-eaters ( Merops apiaster), along with 31 loci isolated in other species, for their utility in European bee-eaters sampled on Susak Island, Croatia. Of these 42 loci, 20 were polymorphic in 38 individuals. These polymorphic loci were further assessed in a sub-set of 23 adults, excluding close relatives, and exhibited between three and 13 alleles each. All loci were autosomal, as indicated by the presence of heterozygotes in both males and females. One of the polymorphic loci exhibited low heterozygosity, three loci deviated from Hardy-Weinberg equilibrium and three pairs of loci displayed linkage disequilibrium. The remaining selected eight cross-species loci and seven loci isolated in European bee-eaters were combined with two sex-typing markers and optimised in five multiplexes. A combination of 15 autosomal loci of varying degrees of polymorphism makes this multiplex set particularly suitable for both parentage and spatial genetic analyses. Understanding inter- and intraspecific variation in parental care has been an important focus in studies of avian behaviour and evolution. Unfortunately, typical methods for quantifying parental care, such as field observation and video recordings, can be extremely time-consuming. Here, we demonstrate that utilizing behavioural analysis software, such as EthoVision XT, can reduce time required for video data extraction by 37−69 %. This method is highly accurate; results and error rates did not differ from those of manual observation. Male sexually selected traits can evolve through different mechanisms: conspicuous and colorful ornaments usually evolve through intersexual selection, while weapons usually evolve through intra-sexual selection. Male ornaments are rare among mammals in comparison to birds, leading to the notion that female mate choice generally plays little role in trait evolution in this taxon. Supporting this view, when ornaments are present in mammals, they typically indicate social status and are products of male-male competition. This general mammalian pattern, however, may not apply to rhesus macaques ( Macaca mulatta). Males of this species display conspicuous skin coloration, but this expression is not correlated to dominance rank and is therefore unlikely to have evolved due to male-male competition. Here, we investigate whether male color expression influences female proceptivity toward males in the Cayo Santiago free-ranging rhesus macaque population. We collected face images of 24 adult males varying in dominance rank and age at the peak of the mating season and modeled these to rhesus macaque visual perception. We also recorded female sociosexual behaviors toward these males. Results show that dark red males received more sexual solicitations, by more females, than pale pink ones. Together with previous results, our study suggests that male color ornaments are more likely to be a product of inter- rather than intra-sexual selection. In order to avoid the invasiveness of sexing associated with biopsy in beluga ( Huso huso), we explored the possibilities of using sex steroid levels to determine the sex of farmed immature fish. Belugas were selected randomly from 2-, 4- and 5-year-old stocks, cultured under the same environmental and feeding regimes. Conventional methods of sexing by biopsy and histological observation were used to determine the sex and gonadal stage, while enzyme-linked immunosorbent assay was employed to measure the levels of testosterone (T) and 17β-estradiol (E) in serum. All fish had differentiated gonads, and males and females could be distinguished microscopically in all age groups. T levels differed significantly between males and females but made for poor predictors of sex due to the substantial overlap in ranges of T levels in both sexes. There was no significant difference in E levels between males and females. When calculating the T/E ratios (TER), a clear separation was obtained for 5-year-old fish, ratios over 40 predicting male and those below 40 predicting female phenotypes. Y chromosomes underlie sex determination in mammals, but their repeat-rich nature has hampered sequencing and associated evolutionary studies. Here we trace Y evolution across 15 representative mammals on the basis of high-throughput genome and transcriptome sequencing. We uncover three independent sex chromosome originations in mammals and birds (the outgroup). The original placental and marsupial (therian) Y, containing the sex-determining gene SRY, emerged in the therian ancestor approximately 180 million years ago, in parallel with the first of five monotreme Y chromosomes, carrying the probable sex-determining gene AMH. The avian W chromosome arose approximately 140 million years ago in the bird ancestor. The small Y/W gene repertoires, enriched in regulatory functions, were rapidly defined following stratification (recombination arrest) and erosion events and have remained considerably stable. Despite expression decreases in therians, Y/W genes show notable conservation of proto-sex chromosome expression patterns, although various Y genes evolved testis-specificities through differential regulatory decay. Ohno's hypothesis states that dosage compensation in mammals evolved in two steps: a twofold hyperactivation of the X chromosome in both sexes to compensate for gene losses on the Y chromosome, and silencing of one X (X-chromosome inactivation, XCI) in females to restore optimal dosage. Recent tests of this hypothesis have returned contradictory results. In this review, we explain this ongoing controversy and argue that a novel view on dosage compensation evolution in mammals is starting to emerge. Ohno's hypothesis may be true for a few, dosage-sensitive genes only. If so few genes are compensated, then why has XCI evolved as a chromosome-wide mechanism? Abstract: African endemic pygmy mice (Genus Mus; sub-genus Nannomys) have considerable economic and public health significance, and some species exhibit novel sex determination systems, making accurate knowledge of their phylogenetics and distribution limits important. This phylogenetic study was based on the mitochondrial control region and cytochrome b gene, for which a substantial body of published data was available. Study specimens were sourced from eight previously unsampled or poorly sampled countries, and include samples morphologically identified as Mus bufo, M. indutus, M. callewaerti, M. triton and M. neavei. These analyses increase the known genetic diversity of Nannomys from 65 to 102 haplotypes; at least 5 unassigned haplotypes are distinguished by potentially species-level cytochrome b genetic distances. The monophyly of Nannomys is supported. Mus musculoides, M. callewaerti, M. indutus, M. bufo, M. haussa, M. mattheyi, M. baoulei and M. sorella are supported as discrete species. The range of M. indutus is extended to include Botswana. M. setulosus and M. minutoides appear to be species complexes. A south and east African M. minutoides clade was defined and includes 8 new haplotypes out of 15. M. setulosus sensu lato includes M. setulosus sensu stricto and a strongly-supported M. bufo clade. In this study, the SoxB2 gene was cloned from a mollusk, the Zhikong scallop ( Chlamys farreri), and characterized with respect to phylogeny and tissue distribution. The full-length cDNA and genomic DNA sequences of C. farreri SoxB2 ( Cf SoxB2) were obtained by rapid amplification of cDNA ends and genome walking, respectively, using a partial cDNA fragment from the highly conserved DNA-binding domain, i.e., the High Mobility Group (HMG) box. The full-length cDNA sequence of Cf SoxB2 was 2 048 bp and encoded 268 amino acids protein. The genomic sequence was 5 551 bp in length with only one exon. Several conserved elements, such as the TATA-box, GC-box, CAAT-box, GATA-box, and Sox/sry-sex/testis-determining and related HMG box factors, were found in the promoter region. Furthermore, real-time quantitative reverse transcription PCR assays were carried out to assess the mRNA expression of Cf SoxB 2 in different tissues. Much is still to be learned about the spatial ecology of foraging marine turtles, especially for juveniles and adult males which have received comparatively little attention. Additionally, there is a paucity of ecological information on growth rates, size and age at maturity, and sex ratios at different life stages; data vital for successful population modelling. Here, we present results of a long-term (2002-2011) study on the movements, residency, growth and sex ratio of loggerhead turtles ( Caretta caretta) in Amvrakikos Gulf (39°0′N 21°0′E), Greece, using satellite telemetry ( N = 8) and ongoing capture-mark-recapture (CMR; N = 300 individuals). Individuals encountered at sea ranged from large juvenile to adult (46.2-91.5 cm straight carapace length) and demonstrated growth rates within published norms (<2.7 cm yr) that slowed with increasing body size. We revealed that an unexpectedly high proportion of animals were male (>44 % of captures above 65 cm straight carapace length), compared to region-wide female-biased hatchling production, indicating sex-biased survival or possible behavioural drivers for likelihood of capture in the region. Satellite tracking confirmed that some turtles establish discrete, protracted periods of residency spanning more than 1 year, whilst others migrated away from the site. The effects of climate change on populations are complex and difficult to predict, and can result in mismatches between interdependent organisms or between organisms and their environment. Reptiles with temperature-dependent sex determination may be able to compensate for potential skews in offspring sex ratio caused by climate change by selecting cooler (i.e., shadier) nest sites. Although changing nest location may prevent sex ratio skews, it may also affect thermally sensitive performance traits in offspring. I tested righting, sprinting, and swimming performance in hatchling painted turtles ( Chrysemys picta), produced by female turtles from five populations across the species' geographic range, nesting in a common-garden environment. I found that speed of hatchling performance was faster in hatchlings whose mothers originated from warmer climates, and that nests with higher mean daily variation in incubation temperature produced faster hatchlings. These results suggest that the increased temperatures predicted by climate change models could result in hatchling turtles that are faster at sprinting and swimming; however, it is not yet known how these performance measures translate into fitness. Sexual competition is highly prevalent within multi-male multi-female primate groups and may lead to copulations in absence of potentially interfering bystanders. Such avoidance of bystanders may result from tactical deception or from simpler mechanisms such as taking advantage of encountered situations without bystanders, operant conditioning or a peripheral positioning of non-alpha males. We investigated which individuals are avoided as bystanders, how individuals react to the presence of bystanders and whether copulation partners separate themselves from the group in a tactical way. Our observations of a group of 15 female and seven male long-tailed macaques housed in three interconnected, but visually separated compartments revealed that both males and females can interrupt sexual behaviour and that bystanders of both sexes were avoided during copulations ( n = 256). The strength of the effect of bystanders tended to decrease with the dominance rank of male bystanders, but did not depend on the dominance rank of female bystanders. The audience effects of non-alpha individuals did not depend on the strong audience effect of the alpha male in combination with proximity with the alpha male. The effects that we found for separate bystanders suggest that sexual competition concerns rank dependent male-male competition and rank independent female-female competition. Additionally, both male and female copulation partners paid attention to the presence of bystanders and conducted fewer copulation solicitations in their presence. The timing of a male and female's separation from the group suggests that exploitation of the peripheral position of non-alpha males, and not tactical deception, may cause these audience effects. One of the persistent problems in biology is understanding how genetic variation contributes to phenotypic variation. Associations at many levels have been reported, and yet causal inference has remained elusive. We propose to rely on the knowledge of causal relationships established by molecular biology approaches. The existing molecular knowledge forms a firm backbone upon which hypotheses connecting genetic variation, transcriptional variation and phenotypic variation can be built. The sex determination pathway is a well-established molecular network, with the Yolk protein 1-3 (Yp) genes as the most downstream target. Our analyses reveal that genetic variation in expression for genes known to be upstream in the pathway explains variation in downstream targets. Relationships differ between the two sexes, and each Yp has a distinct transcriptional pattern. In social insects, the reproductive strategies adopted by colonies emerge as a complex property of individual behaviours, but as yet we are often unable to fully explain them in evolutionary terms. In bumblebees, colonies adopt either a short-lived strategy specializing in male production, or a longer-lived strategy in which mainly new queens are produced, but this results in males emerging long before mates are available; this strategy can only easily be explained if older males are at a significant reproductive advantage. Here we examine how age and morphological characters influence mating success of male bumblebees. In two separate experiments in which single virgin males and females were confined together, we found that young males and heavy males mated more swiftly and copulated for less time compared to old males or lighter males, respectively. However, in competitive situations age proved to be unimportant and the only factors to influence mating success were the lengths of the fore and hind tibiae, with strong directional selection for long leg length. Fore and hind legs are both used in courtship, so both traits are associated with plausible mechanisms under selection. It has previously been argued that, in times of food stress, bumblebee colonies should produce males as male size is less likely to be strongly correlated with fitness than female size. Our results suggest that this may not be so, since aspects of male size directly impact on their mating success. Our results leave unexplained the emergence of males many days before new queens. We investigated the reliability of buccal swabs for molecular sex determination in very young nestlings (0-14 days old) and adults of the Common Swift ( Apus apus) compared to the results from blood samples of the same individuals. Sex determination based on buccal swabs matched the result of sex determination based on blood samples in 46 out of 47 nestlings (98 %) and in all 10 adults (100 %). Therefore, we consider that buccal swab sampling is a reliable noninvasive method to obtain DNA for sex determination in swifts. Human-induced alteration of natural habitats has the potential to impact on the genetic structuring of remnant populations at multiple spatial scales. Species from higher trophic levels, such as snakes, are expected to be particularly susceptible to land-use changes. We examined fine-scale population structure and looked for evidence of sex-biased dispersal in smooth snakes (Coronella austriaca), sampled from 10 heathland localities situated within a managed coniferous forest in Dorset, United Kingdom. Despite the limited distances between heathland areas (maximum <6 km), there was a small but significant structuring of populations based on eight microsatellite loci. This followed an isolation-by-distance model using both straight line and 'biological' distances between sampling sites, suggesting C. austriaca's low vagility as the causal factor, rather than closed canopy conifer forest exerting an effect as a barrier to dispersal. Within population comparisons of male and female snakes showed evidence for sex-biased dispersal, with three of four analyses finding significantly higher dispersal in males than in females. We suggest that the fine-scale spatial genetic structuring and sex-biased dispersal have important implications for the conservation of C. austriaca, and highlight the value of heathland areas within commercial conifer plantations with regards to their future management. Sex in mammals is determined in the fetal gonad by the presence or absence of the Y chromosome gene Sry, which controls whether bipotential precursor cells differentiate into testicular Sertoli cells or ovarian granulosa cells. This pivotal decision in a single gonadal cell type ultimately controls sexual differentiation throughout the body. Sex determination can be viewed as a battle for primacy in the fetal gonad between a male regulatory gene network in which Sry activates Sox9 and a female network involving WNT/?-catenin signalling. In females the primary sex-determining decision is not final: loss of the FOXL2 transcription factor in adult granulosa cells can reprogram granulosa cells into Sertoli cells. Here we show that sexual fate is also surprisingly labile in the testis: loss of the DMRT1 transcription factor in mouse Sertoli cells, even in adults, activates Foxl2 and reprograms Sertoli cells into granulosa cells. In this environment, theca cells form, oestrogen is produced and germ cells appear feminized. Thus Dmrt1 is essential to maintain mammalian testis determination, and competing regulatory networks maintain gonadal sex long after the fetal choice between male and female. Dmrt1 and Foxl2 are conserved throughout vertebrates and Dmrt1-related sexual regulators are conserved throughout metazoans. Antagonism between Dmrt1 and Foxl2 for control of gonadal sex may therefore extend beyond mammals. At the turn of the twentieth century, biologists such as Oscar Riddle, Thomas Hunt Morgan, Frank Lillie, and Richard Goldschmidt all puzzled over the question of sexual difference, the distinction between male and female. They all offered competing explanations for the biological cause of this difference, and engaged in a fierce debate over the primacy of their respective theories. Riddle propounded a metabolic theory of sex dating from the late-nineteenth century suggesting that metabolism lay at the heart of sexual difference. Thomas Hunt Morgan insisted on the priority of chromosomes, Frank Lillie emphasized the importance of hormones, while Richard Goldschmidt supported a mixed model involving both chromosomes and hormones. In this paper, I will illustrate how the older metabolic theory of sex was displaced when those who argued for the relatively newer theories of chromosomes and hormones gradually formed an alliance that accommodated each other and excluded the metabolic theory of sex. By doing so, proponents of chromosomes and hormones established their authority over the question of sexual difference as they laid the foundations for the new disciplines of genetics and endocrinology. Their debate raised urgent questions about what constituted sexual difference, and how scientists envisioned the plasticity and controllability of this difference. These theories also had immediate political and cultural consequences at the turn of the twentieth century, especially for the eugenic and feminist movements, both of which were heavily invested in knowledge of sex and its determination, ascertainment, and command. Daphnia are often used as test organisms to determine toxicity of chemicals found in the environment. Fecundity and mortality are the classic endpoints in such tests; however, there is increased interest in other sub-lethal endpoints. Most Daphnia reproduce by cyclic parthenogenesis, typically reproducing asexually to produce genetically identical females. Environmental cues trigger the production of males allowing for sexual reproduction. This ability to switch reproductive strategies has been increasingly used as a bioassay endpoint in screens for sub-lethal effects of contaminants. In order to optimize such test, it is necessary to know when sex is determined during Daphnia development. A previous study using the insect juvenile hormone methyl farnesoate has shown that sex determination occurred during the egg maturation period in Daphnia. Our study exposed female Daphnia magna carrying eggs and embryos at different stages of maturation, to the insecticide toxaphene to determine if the timing of sex determination is similarly influenced by a pesticide. Our results suggest that in response to toxaphene exposure sex is determined in the immature oocyte, before the final maturation cleavage, and before the developing egg is extruded into the brood chamber. Provisioning of nutrients to the young during parental care is one of the mechanisms by which parents can affect growth and survival of their young and thus their reproductive success. We examined the hypothesis that food quality, i.e., ratio of macronutrients, provided to the young via maternal care affects their performance and the female's reproductive success. The subsocial spider Stegodyphus lineatus exhibits intensive maternal care behaviors, including feeding the young and matriphagy (consumption of the mother). Our results showed that a protein-enriched diet resulted in larger females at maturation and higher survival of young, relative to intermediate or lipid-enriched diets. However, fecundity was not affected by female diet. We suggest that most of the nutrients are provided to the young during maternal care rather than deposited in the eggs, allowing females to economize on limited nutritional reserves. Females before maternal care showed a low protein and high lipid content relative to females before maturation and oviposition, suggesting a change in the nutritional requirement of females before maternal care. This change in macronutrient composition may be adaptive for the success of the young in the wild and shows a novel approach to animals' ability to increase their reproductive success. Field-collected females showed a similar macronutrient composition as that of protein-enriched females in the lab, suggesting a high reproductive success of females in the wild. Color polymorphisms have provided classical examples of how frequency-dependent selection maintains genetic variation in natural populations. Here we tested for the first time, the hypothesized adaptive function of a female-specific color polymorphism in odonates to lower male harassment towards females generally. Under conditions controlling for sex ratio, population density and morph frequency, we also tested two major frequency-dependent selection hypotheses for the maintenance of the polymorphism. Using groups of captive Enallagma hageni, whose females are either green or a male-like blue, we varied morph frequency at two sex ratios. We quantified sexual harassment towards females by visual observations, and by the presence of dust on females that was transferred from dusted males. Per capita harassment rate for the female-monomorphic treatments did not differ from that of the female-polymorphic treatments. At a male-biased sex ratio, per capita harassment rate towards blue, but not green females increased with morph frequency, providing partial support for frequency-dependent selection resulting from male learning of female morphs. Even at high frequency, green females were not harassed more than blue, contrary to the prediction that males should always recognize green females as mates. Moreover, frequency-dependent harassment towards blue females was not detectable using harassment measured with dust evidence, which greatly underestimated the incidence of sexual harassment. Our findings identified problems with the use of insectaries and the dusting technique to quantify male sexual harassment towards females, as well as with a past insectary experiment on Ischnura elegans that failed to demonstrate frequency-dependent harassment. Noninvasive sampling approaches are becoming increasingly important for enabling genetic studies of wildlife populations. While a number of methods have been described to noninvasively sample hair from carnivores and medium-sized mammals, they have largely remained untested in elusive small mammals. Here we describe a novel and inexpensive noninvasive hair snare targeted at an elusive small mammal, the American pika ( Ochotona princeps). We explore the quality of the sample by assessing PCR amplification success of mitochondrial and nuclear DNA fragments across four commercially available DNA isolation kits and two different quantities of hair in a factorial design. Additionally, we determined the sex of the individual samples using PCR-RFLP of ZFX/ZFY loci. We found that our snare is effective in obtaining hair that yield DNA of sufficient quality and quantity to successfully amplify a range of mitochondrial and nuclear fragment sizes. Specifically, we found the greatest success in amplifying mitochondrial DNA, nuclear microsatellites and ZFX/ZFY loci using at least 25 hairs as starting material and the DNA IQ™ system. The hair snares thus provide a cost-effective and minimally intrusive approach to sample elusive or rare small mammals. Environmental effects on sex allocation are common, yet the evolutionary significance of these effects remains poorly understood. Environmental effects might influence parents, such that their condition directly influences sex allocation by altering the relative benefits of producing sons versus daughters. Alternatively, the environment might influence the offspring themselves, such that the conditions they find themselves in influence their contribution to parental fitness. In both cases, parents might be selected to bias their sex ratio according to the prevailing environmental conditions. Here, we consider sex allocation in the citrus mealybug Planococcus citri, a species with an unusual genetic system in which paternal genes are lost from the germline in males. We test environmental factors that may influence either female condition directly (rearing temperature and food restriction) or that may be used as cues of the future environment (age at mating). Using cytological techniques to obtain primary sex ratios, we show that high temperature, older age at mating and starvation all affect sex allocation, resulting in female-biased sex ratios. However, the effect of temperature is rather weak, and food restriction appears to be strongly associated with reduced longevity and a truncation of the usual schedule of male and offspring production across a female's reproductive lifetime. Instead, facultative sex allocation seems most convincingly affected by age at mating, supporting previous work that suggests that social interactions experienced by adult P. citri females are used when allocating sex. However, the behavioral and emotional alterations in male rats exposed to isolation after sexual experience remain poorly understood. Therefore, the present study examined anxiety- and depressive-like parameters, as well as USV emissions in male rats isolated for 1, 2, or 7 d following 1-week sexual (male-female paired housing) or social (male-male paired housing) interaction. Isolation for 1 d resulted in increased depressive-like symptoms, and 2-d isolation induced elevated anxiety-like behavior in sexually inexperienced rats, while rats with sexual experience exhibited comparable levels of emotionality at each isolation time. Moreover, isolated rats without sexual experience emitted less 22- and 50-kHz USVs, but sexually experienced rats exhibited similar affective states compared with control ones. Purpose: We analyzed the sex chromosome-encoding ZFX-ZFY genes and tested molecular sexing using the amplification patterns of intron 9 of ZFX-ZFY in the horse. Methods and results: The amplification of the ZFX-ZFY produced two distinct patterns, reflecting sexual dimorphism based on a length difference between the X and Y chromosomes. The amplification products from foals showed two distinct bands: one was common to all foals and mares, indicating that this band was amplified from ZFX, while the other was specific to some foals, indicating that it was from ZFY. The result based on the PCR assay was identical to the results of amplification of the Y chromosome-specific SRY gene and those of investigations of the phenotypic gender in three different horse populations. Conclusion: We suggest that this PCR strategy for determining sexes by comparing the amplification patterns of ZFX-ZFY genes is a convenient and precise method for discriminating sexes in horses. Sex determination mechanisms (SDMs) show striking diversity and appear to evolve rapidly. Although interspecific comparisons and studies of ongoing major transitions in sex determination (such as the establishment of new sex chromosomes) have shed light on how SDMs evolve, comparatively little attention has been paid to intraspecific variation with less drastic effects. In this study, I used mutant strains carrying a temperature-sensitive sex determination mutation, along with a second null mutation, in different wild genetic backgrounds to uncover hidden variation in the SDM of the model nematode Caenorhabditis elegans. I then used quantitative trait locus (QTL) mapping to begin to investigate its genetic basis. I identified several QTLs, and although this variation apparently involved genotype-by-temperature interactions, QTL effects were generally consistent across temperatures. These QTLs collectively and individually explained a relatively large fraction of the variance in tail morphology (a sexually dimorphic trait), and two QTLs contained no genes known to be involved in somatic sex determination. These results show the existence of within-species variation in sex determination in this species, and underscore the potential for microevolutionary change in this important developmental pathway. Illegal trade of wildlife is growing internationally and is worth more than USD$20 billion per year. DNA technologies are well suited to detect and provide evidence for cases of illicit wildlife trade yet many of the methods have not been verified for forensic applications and the diverse range of methods employed can be confusing for forensic practitioners. In this review, we describe the various genetic techniques used to provide evidence for wildlife cases and thereby exhibit the diversity of forensic questions that can be addressed using currently available genetic technologies. We emphasise that the genetic technologies to provide evidence for wildlife cases are already available, but that the research underpinning their use in forensics is lacking. Finally we advocate and encourage greater collaboration of forensic scientists with conservation geneticists to develop research programs for phylogenetic, phylogeography and population genetics studies to jointly benefit conservation and management of traded species and to provide a scientific basis for the development of forensic methods for the regulation and policing of wildlife trade. We report the development of a fast and reliable PCR-based method for sex identification of African Rhinoceros ( Ceratotherium simum and Diceros bicornis) that could easily be incorporated into fluorescent short tandem repeat (STR) profiling. A single primer pair, consisting of a fluorescently labelled forward primer and an unlabelled reverse primer, is used to co-amplify homologous fragments of a zinc finger (ZF) protein intron which exhibits size polymorphism between the X and Y chromosomes. In both species, the amplified ZFX and ZFY amplicons differ in size by 7 bp and can thus be differentiated by capillary electrophoresis. Blood, tissue, horn, and faecal samples were correctly sexed using this method. Cross species testing also demonstrated that this method could be used to sex Indian rhinoceros ( Rhinoceros unicornis) samples. [ABSTRACT FROM AUTHOR]

Sex determination of birds is important for many ecological studies but is often difficult in species with monomorphic plumage. Morphology often provides a possibility for sex determination, but the characters need to be verified. We tested whether five passerine species can be sexed according to standard morphological measurements applying a forward logistic regression with sex determined by molecular analysis as the dependent variable. Furthermore, we tested whether the results can be used on a larger geographic scale by applying morphological sexing methods gained by similar studies from other regions to our data set. Of the five species of this study only Garden Warblers Sylvia borin could not be sexed morphologically. In the Robin Erithacus rubecula, 87.2% of all individuals were sexed correctly. For Reed Warblers Acrocephalus scirpaceus, Willow Warblers Phylloscopus trochilus and Reed Buntings Emberiza schoeniclus, the respective values were 77.6, 89.4 and 86.4%. When the logistic regression functions from similar studies on Robins and Reed Buntings in Denmark and Scotland were applied to the birds from south-western Germany, they performed less well compared to the original dataset of these studies and compared to the logistic regression function of our own study. The same was the case for Willow Warblers when a wing length criterion used in Great Britain was applied to the birds of our study. These discrepancies may have several explanations: (1) the models are optimised for the dataset from which they were extracted, (2) inter-ringer variation in measurements, (3) the use of different age cohorts, (4) different morphology due to different habitat availability around the study site, or, most likely, (5) different morphology due to different migratory behaviour. We recommend that morphological sex differentiation methods similar to this study (1) be only used population specific, (2) only with one age cohort and (3) to adjust the extracted equations from time to time. Adaptive radiations often follow the evolution of key traits, such as the origin of the amniotic egg and the subsequent radiation of terrestrial vertebrates. The mechanism by which a species determines the sex of its offspring has been linked to critical ecological and life-history traits but not to major adaptive radiations, in part because sex-determining mechanisms do not fossilize. Here we establish a previously unknown coevolutionary relationship in 94 amniote species between sex-determining mechanism and whether a species bears live young or lays eggs. We use that relationship to predict the sex-determining mechanism in three independent lineages of extinct Mesozoic marine reptiles (mosasaurs, sauropterygians and ichthyosaurs), each of which is known from fossils to have evolved live birth. Our results indicate that each lineage evolved genotypic sex determination before acquiring live birth. This enabled their pelagic radiations, where the relatively stable temperatures of the open ocean constrain temperature-dependent sex determination in amniote species. Freed from the need to move and nest on land, extreme physical adaptations to a pelagic lifestyle evolved in each group, such as the fluked tails, dorsal fins and wing-shaped limbs of ichthyosaurs. With the inclusion of ichthyosaurs, mosasaurs and sauropterygians, genotypic sex determination is present in all known fully pelagic amniote groups (sea snakes, sirenians and cetaceans), suggesting that this mode of sex determination and the subsequent evolution of live birth are key traits required for marine adaptive radiations in amniote lineages. A vast literature suggests that maternal factors and egg incubation conditions have substantial effects on offspring phenotypes in oviparous species. However, many studies that evaluate these effects have relied on experimental conditions that are rarely, if ever, encountered under natural conditions. To address this issue, we evaluated relationships among maternal factors, natural nest conditions, egg development in the field, and the resultant offspring phenotypes in a lizard with temperature-dependent sex determination, the jacky dragon ( Amphibolurus muricatus, Agamidae). Many, but not all, of the relationships shown in our field-based study corroborate results from laboratory-based experiments. Offspring body size was affected primarily by egg size at oviposition, as well as by water uptake by eggs, rather than by environmental variables measured within the nest. Date of oviposition was related to offspring growth rate and body size prior to the onset of winter; this relationship is likely mediated through an influence on the timing of hatching. Nest temperature generated substantial variation in egg survival; nests that experienced higher temperatures and higher thermal fluctuations suffered relatively high egg mortality. Contrary to results from laboratory incubation, however, nest temperature did not predict offspring sex ratios. Hence, although many results from this field study corroborate those from the laboratory, caution is needed when extrapolating laboratory-incubation results to field conditions. [ABSTRACT FROM AUTHOR]
