Tauopathies such as Alzheimer's disease (AD), Frontotemporal dementia, and parkinsonism linked to chromosome 17 (FTDP-17), etc. are characterized by tau hyperphosphorylation and distinguished accumulation of paired helical filaments (PHFs)/or neurofibrillary tangles (NFTs) in a specific-neuronal subset of the brain. Among different reported risk factors, type 2 diabetes (T2D) has gained attention due to its correlation with tau pathogenesis. However, mechanistic details and the precise contribution of insulin pathway in tau etiology is still debatable. We demonstrate that expression of human tau causes overactivation of insulin pathway in Drosophila disease models. We subsequently noted that tissue-specific downregulation of insulin signaling or even exclusive reduction of its growth-promoting sub-branch effectively reinstates the overactivated insulin signaling pathway in human tau expressing cells, which in turn restricts pathogenic tau hyperphosphorylation and aggregate formation. It was further noted that restored tau phosphorylation was achieved due to a reestablished balance between the levels of different kinase(s) (GSK3β and ERK/P38 MAP kinase) and phosphatase (PP2A). Taken together, our study demonstrates a precise involvement of the insulin pathway and associated molecular events in the pathogenesis of human tauopathies in Drosophila, which will be immensely helpful in developing novel therapeutic options against these devastating human brain disorders. An efficient one pot synthesis of various bis-coumarinyl methane derivatives was investigated by condensing a series of aldehydes with 4-hydroxy coumarin in the presence of Amberlyst 15 catalyst under refluxing conditions in ethanol–water (1:1). This one pot domino Knoevenagel condensation/Micheal addition reaction provides excellent yield of bis-coumarinyl methane derivatives with lots of advantages like shorter reaction time, eco-friendly reaction medium, simple work up, recyclability of the catalyst and chromatography-free purification. Moreover, the synthesized compound was exploited to develop bis-coumarinyl methylene-based nanostructured materials of special interests which were investigated by UV–Vis, fluorescence spectroscopy, imaging in scanning electron microscope and atomic force microscope. Our UV–Vis and fluorescence study indicated that some of the organic compounds are organized as H-aggregate in the thin film. We also investigated I–V characteristics of the thin film of some of the selective compounds which showed write-once-read-more memory switching behaviour onto the thin film under investigations. Background: Human neuronal tauopathies are typically characterized by the accumulation of hyperphosphorylated tau in the forms of paired helical filaments and/or neurofibrillary tangles in the brain neurons. Tau-mediated heterochromatin loss and subsequent global transcriptional upsurge have been demonstrated as one of the key factors that promotes tau toxicity. We have reported earlier that expression of human tau-transgene in Drosophila induces the expression of glob1, and its restored level restricts tau etiology by regulating tau hyperphosphorylation and ROS generation via GSK-3β/p-Akt and Nrf2-keap1-ARE pathways, respectively. In view of this noted capability of glob1 in regulation of oxidative stress, and involvement of ROS in chromatin remodeling; we investigate if downregulation of glob1 restores tau-mediated heterochromatin loss in order to alleviate neurotoxicity. Methods and results: The tauV337M transgene was expressed in Drosophila eye by utilizing GAL4/UAS system. Expression of glob1 was depleted in tauV337M expressing tissues by co-expressing an UAS-glob1RNAi transgene by GMR-Gal4 driver. Immunostaining and wstern blot analysis suggested that tissue-specific downregulation of glob1 restores the cellular level of CBP and minimizes tau-mediated heterochromatin loss. It also assists in mounting an improved protective autophagic response to alleviate the human tau-induced neurotoxicity in Drosophila tauopathy models. Conclusions: Our study unfolds a novel aspect of the multitasking globin protein in restricting the pathogenesis of neuronal tauopathies. Interestingly, due to notable similarities between Drosophila glob1 and human globin gene(s), our findings may be helpful in developing novel therapeutic approaches against tauopathies. Here, we report the mixing behaviour of diacetylene monomer 10, 12-tricosadiynoic acid (TCDA) and rhodamine-800 (Rh8) using Langmuir–Blodgett (LB) technique. The presence of Rh8 affected the structures, phase behaviour as well as colorimetric properties of TCDA in the mixed films at air–water interface and onto solid support. The mixed LB films having TCDA molefraction ≥ 0.5 leads to multilayer formation, whereas multilayer formation was hardly observed for the films containing TCDA molefraction less than 0.5. It was observed that polymerization as well as phase change (blue to red) was possible only for multilayered films. Brewster angle microscopy, atomic force microscopy, and fluorescence inverted microscopy studies confirmed the formation of definite structures in the polymerized TCDA/Rh8 mixed films at air–water interface and onto solid support. Structure of TCDA polymer strands was largely influenced by the presence of Rh8 in the mixed films. Interestingly, energy transfer occurred from TCDA in red phase to Rh8 in the mixed films with TCDA molefraction greater than 0.4. Maximum energy transfer efficiency was found to be 47.3% for the mixed LB film with TCDA molefraction 0.9. [ABSTRACT FROM AUTHOR]

Polyglutamine [poly(Q)] disorders are a class of trinucleotide repeat expansion neurodegenerative disorders which are dominantly inherited and progressively acquired with age. This group of disorders entail the characteristic formation of protein aggregates leading to widespread loss of neurons in different regions of the brain. SCA3 and HD, the two most commonly occurring types of poly(Q) disorders were examined in the present study. With the aim of elucidating novel genetic modifiers of poly(Q) disorders, the Drosophila insulin receptor (InR) was identified as a potential suppressor of poly(Q)-induced neurotoxicity and degeneration. We demonstrate for the first time that targeted upregulation of InR could effectively mitigate poly(Q)-mediated neurodegeneration in fly models. A significant reduction in poly(Q)-mediated cellular stress and apoptosis was noted upon InR overexpression in poly(Q) background. We further reveal that targeted upregulation of InR causes a substantial reduction in poly(Q) aggregate formation with the residual inclusion bodies localised to the cytoplasm. We also demonstrate that InR achieves suppression of poly(Q) toxicity by replenishing the cellular pool of CREB binding protein and improving the histone acetylation status of the cell. This leads to restoration of the cellular transcriptional machinery which is otherwise severely compromised in poly(Q) disease conditions. Interestingly, there also appeared a possibility of autophagy-mediated rescue of poly(Q) phenotype due to upregulation of InR. Tauopathies represent a group of neurodegenerative disorder which are characterized by the presence of tau positive specialized argyrophilic and insoluble intraneuronal and glial fibrillar lesions known as neurofibrillary tangles (NFTs). Tau is a neuron specific microtubule binding protein which is required for the integrity and functioning of neuronal cells, and hyperphosphorylation of tau and its subsequent aggregation and paired helical filaments (PHFs) and NFTs has emerged as one of the major pathogenic mechanisms of tauopathies in human and mammalian model systems. Modeling of human tauopathies in Drosophila results in manifestation of associated phenotypes, and a recent study has demonstrated that similar to human and mammalian models, accumulation of insoluble tau aggregates in the form of typical neurotoxic NFTs triggers the pathogenesis of tauopathies in fly models. In view of the availability of remarkable genetic tools, Drosophila tau models could be extremely useful for in-depth analysis of the role of NFTs in neurodegeneration and tau aetiology, and also for the screening of novel gene(s) and molecule(s) which suppress the toxicity of tau aggregates. Polyglutamine (poly(Q)) disorders, such as Huntington's disease (HD) and spinocerebellar ataxias, represent a group of neurological disorders which arise due to an atypically expanded poly(Q) tract in the coding region of the affected gene. Pathogenesis of these disorders inside the cells begins with the assembly of these mutant proteins in the form of insoluble inclusion bodies (IBs), which progressively sequester several vital cellular transcription factors and other essential proteins, and finally leads to neuronal dysfunction and apoptosis. We have shown earlier that targeted upregulation of Drosophila myc ( dmyc) dominantly suppresses the poly(Q) toxicity in Drosophila. The present study examines the ability of the human c-myc proto-oncogene and also identifies the specific c-Myc isoform which drives the mitigation of poly(Q)-mediated neurotoxicity, so that it could be further substantiated as a potential drug target. We report for the first time that similar to dmyc, tissue-specific induced expression of human c-myc also suppresses poly(Q)-mediated neurotoxicity by an analogous mechanism. Among the three isoforms of c-Myc, the rescue potential was maximally manifested by the full-length c-Myc2 protein, followed by c-Myc1, but not by c-MycS which lacks the transactivation domain. Human tauopathies such as Alzheimer's Disease (AD), frontotemporal dementia with parkinsonism linked to chromosome 17 (FTDP-17), Pick's disease etc., are a group of neurodegenerative diseases which are characterized by abnormal hyperphosphorylation of tau that leads to formation of neurofibrillary tangles. Recapitulating several features of human neurodegenerative disorders, the Drosophila tauopathy model displays compromised lifespan, locomotor function impairment, and brain vacuolization in adult brain which is progressive and age dependent. Here, we demonstrate that tissue-specific downregulation of the Drosophila homolog of human c-myc proto-oncogene (dMyc) suppresses tau-mediated morphological and functional deficits by reducing abnormal tau hyperphosphorylation and restoring the heterochromatin loss. Our studies show for the first time that the inherent chromatin remodeling ability of myc proto-oncogenes could be exploited to limit the pathogenesis of human neuronal tauopathies in the Drosophila disease model. Interestingly, recent reports on successful uses of some anti-cancer drugs against Alzheimer's and Parkinson's diseases in clinical trials and animal models strongly support our findings and proposed possibility. Lactic acid bacteria are known to produce antimicrobial peptides (AMPs) such as bacteriocins which can be employed to control pathogens and food spoilage microorganisms. However, their possible role as toxic agents against a eukaryotic system still remains unexplored. The present study deals with the in vivo evaluation of acute toxic effect of AMPs LR14, a mixture of AMPs isolated from Lactobacillus plantarum LR/14 on Drosophila melanogaster. The fly was used as a model system to measure the extent of toxicity of these peptides. The results showed that concentrations below 10 mg/ml are not significantly effective. When exposed to 10 mg/ml of AMPs LR14, acute toxic effect and a significant delay in the developmental cycle of the fly could be observed. Also, the weight and size of the flies were significantly reduced upon ingestion of these peptides. Higher concentrations (beyond 15 mg/ml) exerted a strong larvicidal effect. Detailed analysis on larval tissues and adult germ cells of the insect revealed deformity in cellular architecture, DNA fragmentation, and premature apoptosis, confirming that the peptides have a dose-dependent toxic property. Heat shock proteins (Hsps) or molecular chaperones, are highly conserved protein families present in all studied organisms. Following cellular stress, the intracellular concentration of Hsps generally increases several folds. Hsps undergo ATP-driven conformational changes to stabilize unfolded proteins or unfold them for translocation across membranes or mark them for degradation. They are broadly classified in several families according to their molecular weights and functional properties. Extensive studies during the past few decades suggest that Hsps play a vital role in both normal cellular homeostasis and stress response. Hsps have been reported to interact with numerous substrates and are involved in many biological functions such as cellular communication, immune response, protein transport, apoptosis, cell cycle regulation, gametogenesis and aging. The Jaduguda U (-Cu-Fe) deposit in the Singhbhum shear zone has been the most productive uranium deposit in India. Pyrite occurs as disseminated grains or in sulphide stringers and veins in the ore zone. Veins, both concordant and discordant to the pervasive foliation, are mineralogically either simple comprising pyrite ± chalcopyrite or complex comprising pyrite + chalcopyrite + pentlandite + millerite. Nickel-sulphide minerals, though fairly common in concordant veins, are very rare in the discordant veins. Pyrite in Ni-sulphide association is commonly replaced by pentlandite at the grain boundary or along micro-cracks. Based on concentrations of Co and Ni, pyrite is classified as: type-A - high Co (up to 30800 ppm), no/low Ni; type-B - moderate Co (up to 16500 ppm) and moderate to high Ni (up to 32700 ppm); type-C - no/low Co and high Ni (up to 43000 ppm); type-D - neither Co nor Ni. Textural and compositional data of pyrites suggest that the hydrothermal fluid responsible for pre-/early-shearing mineralization evolved from Co-rich to Ni-rich and the late-/post-shearing fluid was largely depleted in minor elements. Sulphur isotope compositions of pyrite mostly furnish positive values ranging between −0.33 and 12.06‰. Composite samples of pyrites with only type-A compositions and mixed samples of type-A and type-B are consistently positive. However, pyrite with mixed type-A and type-C and pyrite with type-D compositions have negative values but close to 0‰. By integrating minor element and sulphur isotope compositions of pyrite in conjunction with other published data on the Jaduguda deposit, it is proposed that reduced sulphur for the precipitation of most pyrites (type-A, type-B) was likely derived from isotopically heavy modified seawater. However, some later sulphur might be magmatic in origin remobilized from existing sulphides in the mafic volcanic rocks in the shear zone. [ABSTRACT FROM AUTHOR]
