The Fusarium graminearum species complex (FGSC) can produce various mycotoxins and is a major concern for food quantity and quality worldwide. In this study, we determined the effects of water activity (aw), temperature, incubation time and their interactions on mycotoxin accumulation and the expression levels of biosynthetic genes in FGSC strains from maize samples in China. The highest deoxynivalenol (DON), 3-acetyldeoxynivalenol(3ADON) and 15-acetyldeoxynivalenol (15ADON) levels of the F. boothii and F. graminearum strains were observed at 0.98 aw/30 °C or 0.99 aw/25 °C. F. asiaticum and F. meridionale reached maximum nivalenol (NIV) and 4-acetylnivalenol (4ANIV) contents at 0.99 aw and 30 °C. With the extension of the incubation time, the concentrations of DON and NIV gradually increased, while those of their derivatives decreased. F. boothii, F. meridionale and one F. asiaticum strain had the highest zearalenone (ZEN) values at 0.95 aw and 25 °C, while the optimum conditions for the other F. asiaticum strain and F. graminearum were 0.99 aw and 30 °C. Four genes associated with trichothecene and zearalenone synthesis were significantly induced under higher water stress in the early stage of production. The results indicated independence of mycotoxin production and gene expression, as maximum amounts of these toxic metabolites were observed at higher aw in most cases. Idiopathic pulmonary fibrosis (IPF) is a progressive interstitial lung disease characterized by progressive loss of pulmonary function. Drug-induced interstitial lung disease has been reported as a severe adverse effect of some drugs, such as bleomycin, amiodarone, and methotrexate. Based on good characteristics, drug-induced pulmonary fibrosis (PF) animal model has played a key role in our understanding of the molecular mechanisms of PF pathogenesis and recapitulates the specific pathology in patients and helps develop therapeutic strategies. Here, we summarize the mechanisms and characteristics of given fibrotic drug-induced animal models for PFs. Together with the key publications describing these models, this brief but detailed overview would be helpful for the pharmacological research with animal models of PFs. Deoxynivalenol (DON) is commonly found in wheat and wheat-derived foods, posing a threat to human health. Biodegradation is an efficient and eco-friendly measure for mycotoxin detoxification. Understanding the mechanism of DON biodegradation is hence of great importance. Herein, we report the application of metabolomics methods for the analysis of DON degradation by a bacterial consortium isolated from wheat leaves collected in Jiangsu Province. Metabolomics analysis combined with a nuclear magnetic resonance analysis revealed the main degradation product, 3-keto-DON, and a minor degradation product, 3-epi-DON. Further study illustrated that DON underwent a two-step epimerization through the intermediate 3-keto-DON. Sequencing analysis of the 16S rRNA metagenome of the microorganismal community suggested that the abundance of three bacterial genera, Achromobacter, Sphingopyxis, and Sphingomonas, substantially increased during the coculture of bacterial consortium and DON. Further investigation revealed that Devosia sp. might be responsible for the epimerization of 3-keto-DON. These findings shed light on the catabolic pathways of DON during biodegradation and illustrate the potential of using metabolomics approaches in biodegradation studies. β-Cyclodextrin-functionalized carbon nitride nanosheets were modified with a molecularly imprinted polymer to obtain a fluorescent probe of type MIP@β-CD/CNNS which is shown to enable fluorometric determination of sterigmatocystin (STG). The material was characterized by transmission electron microscopy, infrared spectra, powder X-ray diffraction, X-ray photoelectron spectroscopy, and by absorption and emission spectra. The modified CNNSs have a good fluorescence quantum yield (13%), high sorption capacity for STG (86 mg·g−1), fast adsorption rate (25 min), and superior adsorption selectivity (with an imprint factor 2.56). When used as an optical probe for STG, the CNNSs act as the chromophore, while β-CD and MIP act as the recognition groups. The blue fluorescence of MIP@β-CD/CNNS (with excitation/emission maxima at 368/432 nm) is quenched by STG. Fluorescence drops linearly in the 0.15 to 3.1 μM STG concentration range. The lower detection limit is 74 nM. The method was successfully applied to the determination of STG in spiked wheat extract. Dihydroxyacid dehydratase (DHAD), encoded by ILV3, catalyses the third step in the biosynthetic pathway of branched-chain amino acids (BCAAs), which include isoleucine (Ile), leucine (Leu), and valine (Val). Enzymes involved in BCAA biosynthesis exist in bacteria, plants, and fungi but not in mammals and are therefore attractive targets for antimicrobial or herbicide development. In this study, three paralogous ILV3 genes (FgILV3A, FgILV3B, and FgILV3C) were identified in the genome of Fusarium graminearum, the causal agent of Fusarium head blight (FHB). Deletion of FgILV3A alone or combined with FgILV3B or FgILV3C indicated an important role for FgILV3A in BCAA biosynthesis. FgILV3A deletion mutants lost the ability to grow on medium lacking amino acids. Exogenous supplementation of 1 mM Ile and Val rescued the auxotrophy of ΔFgIlv3A, though 5 mM was required to recover the growth defects in ΔFgIlv3AB and ΔFgIlv3AC strains, indicating that FgIlv3b and FgIlv3c exhibit redundant but accessory roles with FgIlv3a in BCAA biosynthesis. The auxotrophy of ΔFgIlv3A resulted in pleiotropic defects in aerial hyphal growth, in conidial formation and germination, and in aurofusarin accumulation. In addition, the mutants showed reduced virulence and deoxynivalenol production. In 2012, we isolated mycotoxigenic fungi from maize kernels sourced in Jiangsu and Anhui provinces, China. Fusarium spp. accounted for most isolates detected, followed by Aspergillus and Penicillium spp. Of the Fusarium species, Fusarium verticillioides was the most prevalent, as revealed by direct PCR of DNA from kernel samples, followed by F. proliferatum and F. graminearum. Most F. verticillioides strains contained the FUM1 gene, the product of which plays a key role in fumonisin biosynthesis. In terms of mating type idiomorphs, about 80 % of all strains carried the MAT-1 allele, and 20 % carried MAT-2. The genetic structures of 192 representative F. verticillioides isolates were determined via an analysis of eight simple sequence repeat markers. This analysis revealed high-level gene diversity but low linkage disequilibrium in three populations grouped by the geographical areas in which they were collected. The three populations exhibited low-level genetic differentiation and high gene flow, indicating that geographical differences minimally influenced population differentiation; all three populations were potentially components of a larger, randomly mating population. In this study we characterized FgIlv1, a homologue of the Saccharomyces cerevisiae threonine dehydratase (TD) from the important Fusarium head blight fungus Fusarium graminearum. TD catalyzes the first step in the biosynthesis pathway of isoleucine (Ile) for conversion of threonine (Thr) to 2-ketobutyrate (2-KB). The FgILV1 deletion mutant ΔFgIlv1-3 was unable to grow on minimal medium or fructose gelatin agar which lacked Ile. Exogenous supplementation of Ile or 2-KB but not Thr rescued the mycelial growth defect of ΔFgIlv1-3, indicating the involvement of FgIlv1 in the conversion of Thr to 2-KB in Ile biosynthesis. Additionally, exogenous supplementation of Methionine (Met) could also rescue the mycelial growth defect of ΔFgIlv1-3, indicating a crosstalk between Ile biosynthesis and Met catabolism in F. graminearum. Deletion of FgILV1 also caused defects in conidial formation and germination. In addition, ΔFgIlv1-3 displayed decreased virulence on wheat heads and a low level of deoxynivalenol (DON) production in wheat kernels. Taken together, results of this study indicate that FgIlv1 is an essential component in Ile biosynthesis and is required for various cellular processes including mycelial and conidial morphogenesis, DON biosynthesis, and full virulence in F. graminearum. Members of the Fusarium graminearum species complex (FGSC) cause Fusarium head blight in small cereal grains all over the world. To determine the species and trichothecene chemotype composition and population structure of FGSC in Jiangsu and Anhui provinces, an area where epidemics occur regularly, 891 isolates were collected in two consecutive years (2011 and 2012) and characterized with species- and chemotype-specific polymerase chain reaction. Of the 891 isolates typed, 83 were F. graminearum sensu stricto (s. str.) and 808 were F. asiaticum. All 83 F. graminearum s. str. isolates were of a 3ADON (26.51 %) or 15ADON (73.49 %) type, while F. asiaticum isolates included 696 3ADON producers, 46 15ADON producers, and 66 NIV producers. Eight variable number tandem repeat (VNTR) markers were tested on a representative 384 F. asiaticum isolates from 55 sampling sites. VNTR analysis showed high gene diversity and genotypic diversity but low linkage disequilibrium in both populations Fg2011 and Fg2012 grouped based on the year of collection. Low genetic differentiation ( F = 0.026) and high gene flow ( N = 15.13) was observed between the two populations and among subpopulations within the same population ( N = 3.53 to 48.37), indicating that few influence of temporal and spatial variations on population differentiation in this area. Similar result was obtained from 3ADON, 15ADON and NIV populations or carbendazim resistant and sensitive populations, indicating that chemotype of Fusarium isolates and carbendazim application had minor influence on population subdivision. In order to get a broader view of the s-process nucleosynthesis we study the abundance distribution of heavy elements of 35 barium stars and 24 CEMP-stars, including nine CEMP-s stars and 15 CEMP-r/s stars. The similar distribution of [Pb/hs] between CEMP-s and CEMP-r/s stars indicate that the s-process material of both CEMP-s and CEMP-r/s stars should have a uniform origin, i.e. mass transfer from their predominant AGB companions. For the CEMP-r/s stars, we found that the r-process should provide similar proportional contributes to the second s-peak and the third s-peak elements, and also be responsible for the higher overabundance of heavy elements than those in CEMP-s stars. Which hints that the r-process origin of CEMP-r/s stars should be closely linked to the main r-process. The fact that some small r values exist for both barium and CEMP-s stars, implies that the single exposure event of the s-process nucleosynthesis should be general in a wide metallicity range of our Galaxy. Based on the relation between C and C, we suggest that the origin of r-elements for CEMP-r/s stars have more sources. A common scenario is that the formation of the binary system was triggered by only one or a few supernova. In addition, accretion-induced collapse(AIC) or SN 1.5 should be the supplementary scenario, especially for these whose pre-AGB companion with higher mass and smaller orbit radius, which support the higher values of both C and C. The heavy elements in the Universe are formed during the s- and r-processes mainly in AGB stars and supernovae, respectively. Simulation of s- and r-nucleosynthesis critically depends on the neutron capture and weak decay rates for all the nuclei on the reaction chain. The present work analyzes systematically the neutron capture rates (cross sections) for the s-process nuclei, including ∼3000 rates on ∼200 nuclei. The network calculations for the constant temperature s-process have been performed using the different data sets selected as the nuclear inputs to investigate the uncertainties in the predicted s-abundances. We show that the available cross sections of neutron capture on many s-process nuclei still carry large uncertainties, which lead to low accuracy in the determination of s-process isotope abundances. We analyze the neutron capture cross section data for the same unique isobar nucleus accorded by year from previous work. Such an analysis indicates that the s-process has been studied for more than fifty years and there exist two research stages around 1976 and 2002, respectively. Fusarium head blight (FHB) is a destructive wheat disease of global importance. Resistance breeding depends heavily on the Fhb1 gene. The CIMMYT line Shanghai-3/Catbird (SHA3/CBRD) is a promising source without this gene. A recombinant inbred line (RIL) population from the cross of SHA3/CBRD with the German spring wheat cv. Naxos was evaluated for FHB resistance and related traits in field trials using spray and spawn inoculation in Norway and point inoculation in China. After spray and spawn inoculation, FHB severities were negatively correlated with both anther extrusion (AE) and plant height (PH). The QTL analysis showed that the Rht- B1b dwarfing allele co-localized with a QTL for low AE and increased susceptibility after spawn and spray inoculation. In general, SHA3/CBRD contributed most of the favorable alleles for resistance to severity after spray and spawn inoculation, while Naxos contributed more favorable alleles for reduction in FDK and DON content and resistance to severity after point inoculation. SHA3/CBRD contributed a major resistance QTL close to the centromere on 2DLc affecting FHB severity and DON after all inoculation methods. This QTL was also associated with AE and PH, with high AE and tall alleles contributed by SHA3/CBRD. Several QTL for AE and PH were detected, and low AE or reduced PH was always associated with increased susceptibility after spawn and spray inoculation. Most of the other minor FHB resistance QTL from SHA3/CBRD were associated with AE or PH, while the QTL from Naxos were mostly not. After point inoculation, no other QTL for FHB traits was associated with AE or PH, except the 2DLc QTL which was common across all inoculation methods. Marker-assisted selection based on the 2DLc QTL from SHA3/CBRD combined with phenotypic selection for AE is recommended for resistance breeding based on this valuable source of resistance. Powdery mildew is one of the most important wheat diseases in temperate regions of the world. Resistance breeding is considered to be an economical and environmentally benign way to control this disease. The German spring wheat cv. 'Naxos' exhibits high levels of partial and race non-specific resistance to powdery mildew in the field and is a valuable source in resistance breeding. The main objective of the present study was to map the genetic factors behind the resistance in Naxos, based on a population of recombinant inbred lines (RIL) from a cross with the susceptible CIMMYT breeding line SHA3/CBRD. Powdery mildew severity was evaluated in six field trials in Norway and four field trials in China. The major quantitative trait locus (QTL) with resistance from Naxos was detected close to the Pm3 locus on 1AS in all environments, and explained up to 35% of the phenotypic variation. Naxos was shown to carry another major QTL on 2DL and minor ones on 2BL and 7DS. QTL with resistance from SHA3/CBRD were detected on 1RS, 2DLc, 6BL and 7AL. The QTL on the 1B/1R translocation showed highly variable effects across environments corresponding to known virulence differences against Pm8. SHA3/CBRD was shown to possess the Pm3 haplotype on 1AS, but none of the known Pm3a- g alleles. Lithium isotopes have attracted an intense interest because the abundance of both Li and Li from big bang nucleosynthesis (BBN) is one of the puzzles in nuclear astrophysics. Many investigations of both astrophysical observation and nucleosynthesis calculation have been carried out to solve the puzzle, but it is not solved yet. Several nuclear reactions involving lithium have been indirectly measured at China Institute of Atomic Energy, Beijing. The Standard BBN (SBBN) network calculations are then performed to investigate the primordial Lithium abundance. The aim of the study was to evaluate retrospectively the usefulness of polymerase chain reaction (PCR)-microarray technology, which can simultaneously detect seven human herpes viruses for rapid and accurate diagnosis of herpetic encephalitis in children. We simultaneously amplified herpes simplex virus type 1 and type 2 (HSV-1 and HSV-2); varicella-zoster virus; Epstein-Barr virus (EBV); cytomegalovirus (CMV); and human herpes virus 6 (HHV-6A and HHV-6B) by multiplex PCR, and genotyped by DNA microarray technology. The multiplex primers and oligonucleotide probes were designed and synthesized based on the highly conserved regions of the DNA polymerase gene in human herpes viruses. Two hundred ninety cerebrospinal fluid (CSF) specimens from children with clinical suspicion of viral encephalitis were screened by PCR-microarray technology. The results were compared with those of TaqMan PCR kits of common herpes virus. The PCR-microarray technology could detect as few as 10 copies of viral loads. There was no nonspecific hybridizing signal between probes and no cross-reaction to DNA extracted from the pathogens we used. Of 290 cases, 11 were tested positive by PCR-microarray technology. Among them, three were positive for HSV-1, two were positive for HSV-2, one was positive for EBV, two were positive for CMV, two were positive for HHV-6A, one was positive for HHV-6B, and one showed mixed infection of HSV-2 and CMV, and the positive rate was 3.8%. Compared with the results of TaqMan PCR, the sensitivity of PCR-microarray technology was 91.7%, the specificity was 100%, and the index of accurate diagnosis was 0.917. None of the 30 control CSF specimens was tested positive in both methods. Our study suggests that the simultaneous detection of seven human herpes viruses by PCR-microarray technology is the method of choice for rapid, accurate, and specific etiological diagnosis of herpetic encephalitis in children. Abstract  Methylglyoxal is a kind of poisonous metabolite that can react with RNA, DNA and protein, which generally results in a number of side advert effects to cell. Glyoxalase I is a member of glyoxalase system that can detoxify methylglyoxal. An EST encoding a glyoxalase I was isolated from a SSH (suppression subtractive hybridization)-cDNA library of wheat spike inoculated by Fusarium graminearum. The corresponding full length gene, named TaGly I, was cloned, sequenced and characterized. Its genomic sequence consists of 2,719 bp, including seven exons and six introns, and its coding sequence is 929 bp with an open reading frame encoding 291 amino acids. Sequence alignment showed that there were two glyoxalase I domains in the deduced protein sequence. By using specific primers, TaGly I was mapped to chromosome 7D of wheat via a set of durum wheat ‘Langdon’ D-genome disomic-substitution lines. The result of Real-time quantitative polymerase chain reaction demonstrated that TaGly I was induced by the inoculation of Fusarium graminearum in wheat spikes. Additionally, it was also induced by high concentration of NaCl and ZnCl2. When TaGly I was overexpressed in tobacco leaves via Agrobacterium tumefaciens infection, the transgenic tobacco showed stronger tolerance to ZnCl2 stress relative to transgenic control with GFP. Using the high precision data of the proper motions and parallaxes from Hipparcos catalogue, we obtained the orbital parameters of 1302 stars in the Galaxy based on the mass distribution model provided by Allen and Santillón. Fe abundances of 1295 stars among our samples were analyzed. With the correlation analyses between [Fe/H] and orbital parameters, we obtained that the Fe gradient is - 0.057 ± 0.007 dex/kpc along the direction of the maximum galactocentric distance (hereafter DGmax) in the range of 8.5 kpc < DGmax < 17 kpc. We also got the result that the vertical gradient is steeper than the radial gradient. Furthermore, we divided the samples into two subgroups: giants and dwarfs; F, G and K stars; and then analyzed them respectively. Our results show that the gradient becomes flatter and flatter from giants to dwarfs, from F type to G and K type stars. We also divided the samples into disk and halo stars using maximum vertical distance Zmax = 1 kpc as the criterion and got the result that the abundances of the disk stars are much higher than that of the halo stars. Our work suggests the existence of the galactic gradient and supports those chemlcal evolution medels which show that the halo was formed before the disk at the early stage of the Galaxy. X-ray stars have been studied since the beginning of X-ray astronomy. Investigating and studying the chromospheric activity from X-ray stellar optical spectra is highly significant in providing insights into stellar magnetic activity. The big data of LAMOST survey provides an opportunity for researching stellar optical spectroscopic properties of X-ray stars. We inferred the physical properties of X-ray stellar sources from the analysis of LAMOST spectra. First, we cross-matched the X-ray stellar catalogue (12254 X-ray stars) from ARXA with LAMOST data release 3 (DR3), and obtained 984 good spectra from 713 X-ray sources. We then visually inspected and assigned spectral type to each spectrum and calculated the equivalent width (EW) of Hα line using the Hammer spectral typing facility. Based on the EW of Hα line, we found 203 spectra of 145 X-ray sources with Hα emission above the continuum. For these spectra we also measured the EWs of Hβ, Hγ, Hδ and Ca II IRT lines of these spectra. After removing novae, planetary nebulae and OB-type stars, we found there are 127 X-ray late-type stars with Hα line emission. By using our spectra and results from the literature, we found 53 X-ray stars showing Hα variability; these objects are Classical T Tauri stars (CTTs), cataclysmic variables (CVs) or chromospheric activity stars. We also found 18 X-ray stars showing obvious emissions in the Ca II IRT lines. Of the 18 X-ray stars, 16 are CTTs and 2 are CVs. In this study, we characterized FgIlv2 and FgIlv6, the catalytic and regulatory subunits of acetohydroxyacid synthase (AHAS) from the important wheat head scab fungus Fusarium graminearum. AHAS catalyzes the first common step in the parallel pathways toward branched-chain amino acids (BCAAs: isoleucine, leucine, valine) and is the inhibitory target of several commercialized herbicides. Both FgILV2 and FgILV6 deletion mutants were BCAA-auxotrophic and showed reduced aerial hyphal growth and red pigmentation when cultured on PDA plates. Conidial formation was completely blocked in the FgILV2 deletion mutant ΔFgIlv2-4 and significantly reduced in the FgILV6 deletion mutant ΔFgIlv6-12. The auxotrophs of ΔFgIlv2-4 and ΔFgIlv6-12 could be restored by exogenous addition of BCAAs but relied on the designated nitrogen source the medium contained. Deletion of FgILV2 or FgILV6 also leads to hypersensitivity to various cellular stresses and reduced deoxynivalenol production. ΔFgIlv2-4 lost virulence completely on flowering wheat heads, whereas ΔFgIlv6-12 could cause scab symptoms in the inoculated spikelet but lost its aggressiveness.
