Summary: We report the results of alendronate ingestion plus exercise in preventing the declines in bone mass and strength and elevated levels of urinary calcium and bone resorption in astronauts during 5.5 months of spaceflight. Introduction: This investigation was an international collaboration between NASA and the JAXA space agencies to investigate the potential value of antiresorptive agents to mitigate the well-established bone changes associated with long-duration spaceflight. Methods: We report the results from seven International Space Station (ISS) astronauts who spent a mean of 5.5 months on the ISS and who took an oral dose of 70 mg of alendronate weekly starting 3 weeks before flight and continuing throughout the mission. All crewmembers had available for exercise a treadmill, cycle ergometer, and a resistance exercise device. Our assessment included densitometry of multiple bone regions using X-ray absorptiometry (DXA) and quantitative computed tomography (QCT) and assays of biomarkers of bone metabolism. Results: In addition to pre- and post-flight measurements, we compared our results to 18 astronauts who flew ISS missions and who exercised using an early model resistance exercise device, called the interim resistance exercise device, and to 11 ISS astronauts who exercised using the newer advanced resistance exercise device (ARED). Our findings indicate that the ARED provided significant attenuation of bone loss compared with the older device although post-flight decreases in the femur neck and hip remained. The combination of the ARED and bisphosphonate attenuated the expected decline in essentially all indices of altered bone physiology during spaceflight including: DXA-determined losses in bone mineral density of the spine, hip, and pelvis, QCT-determined compartmental losses in trabecular and cortical bone mass in the hip, calculated measures of fall and stance computed bone strength of the hip, elevated levels of bone resorption markers, and urinary excretion of calcium. In Mediterranean climates, adoption and use of the ET-based scheduling method is limited to regions characterized by considerable contributions to evapotranspiration from fog interception, dew, and light rainfall. While the crop evapotranspiration is often accurately estimated, the water balance is frequently in error because a considerable portion of the energy expended is used to vaporize water from the plant surfaces rather from inside the leaves (i.e., transpiration). Growers in regions with considerable fog, dew, and light rainfall are hesitant to use ET-based scheduling because the cumulative crop evapotranspiration between irrigations is often considerably higher than the soil water depletion. A correction for these surface contributions is clearly needed to improve the water balance calculations and to enhance adoption of the ET-based scheduling method. In this paper, we present a simple, practical method to estimate the contribution of fog interception, dew, and light rainfall to daily crop evapotranspiration, and we show how to use the method to improve water balance calculations. Sensible and latent heat flux densities were estimated in a level vineyard, a northeast aspect vineyard and a southwest aspect vineyard in the Napa Valley of California using the eddy covariance and surface renewal methods. Surface renewal is theoretically not limited to level or extensively homogeneous terrain because it examines a more localized process of scalar exchange as compared with eddy covariance. Surface renewal estimates must be calibrated against eddy covariance data to account for unequal heating of the air parcels under a fixed measurement height. We calibrated surface renewal data against eddy covariance data in a level vineyard, and the calibration factor (α) was applied to the surface renewal measurements on the hillside vineyards. Latent heat flux density was estimated from the residual of the energy balance. In the level vineyard, the average daily actual evapotranspiration (ET) for the period of June through September was 2.4 mm per day. In the northeast aspect vineyard, the average daily ET was 2.2 mm per day, while in the southwest aspect vineyard it was 2.7 mm per day. The net radiation values for the level vineyard, the northeast aspect vineyard, and the southwest aspect vineyard were compared against the Ecosystem Water Program with good agreement. Structure functions are used to study the dissipation and inertial range scales of turbulent energy, to parametrize remote turbulence measurements, and to characterize ramp features in the turbulent field. Ramp features are associated with turbulent coherent structures, which dominate energy and mass fluxes in the atmospheric surface layer. The analysis of structure functions to identify ramp characteristics is used in surface renewal methods for estimating fluxes. It is unclear how commonly observed different scales of ramp-like shapes (i.e., smaller ramps and spikes embedded in larger ramps) influence structure function analysis. Here, we examine the impact of two ramp-like scales on structure function analysis using artificially generated data. The range of time lags in structure function analysis was extended to include time lags typically associated with isotropic turbulence to those larger than the ramp durations. The Van Atta procedure (Arch Mech 29:161-171, ) has been expanded here to resolve the characteristics of two-scale ramp models. This new method accurately, and in some cases, exactly determines the amplitude and duration of both ramp scales. Spectral analysis was applied to the structure functions for a broad range of time lags to provide qualitative support for the expanded Van Atta procedure results. Ramp features in the turbulent scalar field are associated with turbulent coherent structures, which dominate energy and mass fluxes in the atmospheric surface layer. Although finer scale ramp-like shapes embedded within larger scale ramp-like shapes can readily be perceived in turbulent scalar traces, their presence has largely been overlooked in the literature. We demonstrate the signature of more than one ramp scale in structure functions of the turbulent scalar field measured from above bare ground and two types of short plant canopies, using structure-function time lags ranging in scale from isotropic to larger than the characteristic coherent structures. Spectral analysis of structure functions was used to characterize different scales of turbulent structures. By expanding structure function analysis to include two ramp scales, we characterized the intermittency, duration, and surface renewal flux contribution of the smallest (i.e., Scale One) and the dominant (i.e., Scale Two) coherent structure scales. The frequencies of the coherent structure scales increase with mean wind shear, implying that both Scale One and Scale Two are shear-driven. The embedded Scale One turbulent structure scale is ineffectual in the surface-layer energy and mass transport process. The new method reported here for obtaining surface renewal-based scalar exchange works well over bare ground and short canopies under unstable conditions, effectively eliminating the α calibration for these conditions and forming the foundation for analysis over taller and more complex surfaces. Legitimate uses of gene transfer technology can benefit from sensitive detection methods to determine vector biodistribution in pre-clinical studies and in human clinical trials, and similar methods can detect illegitimate gene transfer to provide sports-governing bodies with the ability to maintain fairness. Real-time PCR assays were developed to detect a performance-enhancing transgene (erythropoietin, EPO) and backbone sequences in the presence of endogenous cellular sequences. In addition to developing real-time PCR assays, the steps involved in DNA extraction, storage and transport were investigated. By real-time PCR, the vector transgene is distinguishable from the genomic DNA sequence because of the absence of introns, and the vector backbone can be identified by heterologous gene expression control elements. After performance of the assays was optimized, cynomolgus macaques received a single dose by intramuscular (IM) injection of plasmid DNA, a recombinant adeno-associated viral vector serotype 1 (rAAV1) or a rAAV8 vector expressing cynomolgus macaque EPO. Macaques received a high plasmid dose intended to achieve a significant, but not life-threatening, increase in hematocrit. rAAV vectors were used at low doses to achieve a small increase in hematocrit and to determine the limit of sensitivity for detecting rAAV sequences by single-step PCR. DNA extracted from white blood cells (WBCs) was tested to determine whether WBCs can be collaterally transfected by plasmid or transduced by rAAV vectors in this context, and can be used as a surrogate marker for gene doping. We demonstrate that IM injection of a conventional plasmid and rAAV vectors results in the presence of DNA that can be detected at high levels in blood before rapid elimination, and that rAAV genomes can persist for several months in WBCs. Unlabelled: GOAL OF THE WORK: Anemia is a common side effect of chemotherapy. Limited information exists about its incidence and risk factors. The objective of this study was to evaluate the incidence of anemia and risk factors for anemia occurrence in patients with early breast cancer who received adjuvant chemotherapy.Materials and Methods: We evaluated risk factors for anemia in pre- and post/perimenopausal patients with lymph node-positive early breast cancer treated with adjuvant chemotherapy in two randomized trials. All patients received four cycles of doxorubicin and cyclophosphamide (AC) followed by three cycles of cyclophosphamide, methotrexate, fluorouracil (CMF). Anemia incidence was related to baseline risk factors. Multivariable analysis used logistic and Cox regression.Main Results: Among the 2,215 available patients, anemia was recorded in 11% during adjuvant chemotherapy. Grade 2 and 3 anemia occurred in 4 and 1% of patients, respectively. Pretreatment hemoglobin and white blood cells (WBC) were significant predictors of anemia. Adjusted odds ratios (logistic regression) comparing highest versus lowest quartiles were 0.18 (P < 0.0001) for hemoglobin and 0.52 (P = 0.0045) for WBC. Age, surgery type, platelets, body mass index, and length of time from surgery to chemotherapy were not significant predictors. Cox regression results looking at time to anemia were similar.Conclusions: Moderate or severe anemia is rare among patients treated with AC followed by CMF. We evaluated quality of life (QL) and quality-adjusted survival in International Breast Cancer Study Group Trial IX, a randomised trial including 1669 eligible patients receiving tamoxifen for 5 years or three prior cycles of cyclophosphamide, methotrexate and 5-fluorouracil (CMF) followed by 57 months tamoxifen. During the time with CMF toxicity (Tox), without symptoms and toxicity (TWiST), and following relapse (Rel), patients scored their QL indicators and a utility indicator for subjective health estimation between 'perfect' and 'worst' health. Scores were averaged within Tox, TWiST and Rel and transformed to utilities. Mean durations for the three transition times were weighted with utilities to obtain mean quality-adjusted TWiST (Q-TWiST). Patients receiving CMF reported significantly worse scores for most QL domains at month 3, but less hot flushes. After completing chemotherapy, there were no differences by treatment groups. Benefits evaluated by Q-TWiST favoured the additional chemotherapy. CMF provided 3 more months of Q-TWiST for patients with ER-negative tumours, but CMF provided no benefit in Q-TWiST for patients with ER-positive tumours. Q-TWiST analysis based on patient ratings is feasible in large-scale cross-cultural clinical trials. [ABSTRACT FROM AUTHOR]

Rapidly developing techniques for manipulating the pathways of polyketide biosynthesis at the genomic level have created the demand for new pathways with novel biosynthetic capability. Polyketides derived from dinoflagellates are among the most complex and unique structures identified thus far, yet no studies of the biosynthesis of dinoflagellate-derived polyketides at the genomic level have been reported. Nine strains representing 7 different species of dinoflagellates were screened for the presence of type I and type II polyketide synthases (PKSs) by polymerase chain reaction (PCR) and reverse transcriptase PCR. Seven of the 9 strains yielded products that were homologous with known and putative type I PKSs. Unexpectedly, a PKS gene was amplified from cultures of the dinoflagellate Gymnodinium catenatum, a saxitoxin producer, which is not known to produce a polyketide. In each case the presence of a PKS gene was correlated with the presence of bacteria in the cultures as identified by amplification of the bacterial 16S ribosomal RNA gene. However, amplification from polyadenylated RNA, the lack of PKS expression in light-deprived cultures, residual phylogenetic signals, resistance to methylation-sensitive restriction enzymes, and the lack of hybridization to bacterial isolates support a dinoflagellate origin for most of these genes. In an arid environment, the effect of evaporation on energy balance can affect air temperature recordings and greatly impact on degree-day calculations. This is an important consideration when choosing a site or climate data for phenological models. To our knowledge, there is no literature showing the effect of the underlying surface and its fetch around a weather station on degree-day accumulations. In this paper, we present data to show that this is a serious consideration, and it can lead to dubious models. Microscale measurements of temperature and energy balance are presented to explain why the differences occur. For example, the effect of fetch of irrigated grass and wetting of bare soil around a weather station on diurnal temperature are reported. A 43-day experiment showed that temperature measured on the upwind edge of an irrigated grass area averaged 4% higher than temperatures recorded 200 m inside the grass field. When the single-triangle method was used with a 10°C threshold and starting on May 19, the station on the upwind edge recorded 900 degree-days on June 28, whereas the interior station recorded 900 degree-days on July 1. Clearly, a difference in fetch can lead to big errors for large degree-day accumulations. Immediately after wetting, the temperature over a wet soil surface was similar to that measured over grass. However, the temperature over the soil increased more than that over the grass as the soil surface dried. Therefore, the observed difference between temperatures measured over bare soil and those over grass increases with longer periods between wettings. In most arid locations, measuring temperature over irrigated grass gives a lower mean annual temperature, resulting in lower annual cumulative degree-day values. This was verified by comparing measurements over grass with those over bare soil at several weather stations in a range of climates. To eliminate the effect of rainfall frequency, using temperature data collected only over irrigated grass, is recommended for long-term assessment of climate change effects on degree-day accumulation. In high evaporative conditions, a fetch of at least 100 m of grass is recommended. Our results clearly indicate that weather stations sited over bare soil have consistently higher degree-day accumulations. Therefore, especially in arid environments, phenology models based on temperature collected over bare soil are not transferable to those based on temperature recorded over irrigated grass. Although using hourly weather data offers the greatest accuracy for estimating growing degree-day values, daily maximum and minimum temperature data are often used to estimate these values by approximating the diurnal temperature trends. This paper presents a new empirical model for estimating the hourly mean temperature. The model describes the diurnal variation using a sine function from the minimum temperature at sunrise until the maximum temperature is reached, another sine function from the maximum temperature until sunset, and a square-root function from then until sunrise the next morning. The model was developed and calibrated using several years of hourly data obtained from five automated weather stations located in California and representing a wide range of climate conditions. The model was tested against an additional data-set at each location. The temperature model gave good results, the root-mean-square error being less than 2.0 °C for most years and locations. The comparison with published models from the literature showed that the model was superior to the other methods. Hourly temperatures from the model were used to calculate degree-day values. A comparison between degree-day estimates determined from the model and those obtained other selected methods is presented. Bacterivorous protists have been recovered from pristine and contaminated aquifer environments, but the ecological role of these organisms in bioremediation strategies has not been well defined. Burkholderia cepacia G4 PR1 constitutively expresses a toluene ortho-monooxygenase (tom) due to a secondary transposition of a Tn5 transposable element in a trichloroethylene (TCE) degradative plasmid (TOM). Groundwater and sediment from a potential site for a TCE bioremediation field demonstration were used in laboratory microcosms to test the survival of this organism. In nonsterile aquifer sediment slurries, the bacterium was eliminated in a logrithmic decay concomitant with an increase in bacterivorous protists. A half-life for the organism calculated from extinction coefficients increased logarithmically with increasing inoculation density above 1 × 106 PR1 ml-1. For inoculation densities below this level, the half-life of PR1 increased exponentially with decreasing inoculation density. The lowest half-lives corresponded to densities of bacteria that stimulate response of bacterivores. In a column system designed to incorporate aquifer flow, repeated addition of PR1 resulted in a buildup of bacterivore populations and reduced half-life of the bacterium. Addition of TCE and growth substrate in the eluent resulted in prolonged survival of PR1 and apparent mineralization of TCE. The results indicate significant but predictable losses due to native bacterivores would occur within and beyond a treatment zone where PR1 would be added to the aquifer, and mineralization of TCE in contaminated groundwater might be possible with repeated inoculation and addition of nutrients. This paper compares several methods for determining degree-day (° D) threshold temperatures from field observations. Three of the methods use the mean developmental period temperature and simple equations to estimate: (1) the smallest standard deviation in ° D, (2) the least standard deviation in days, and (3) a linear regression intercept. Two additional methods use iterations of cumulative ° D and threshold temperatures to determine the smallest root mean square error (RMSE). One of the iteration methods uses a linear model and the other uses a single triangle ° D calculation method. The method giving the best results was verified by comparing observed and predicted phenological periods using 7 years of kiwifruit data and 10 years of cherry tree data. In general, the iteration method using the single triangle method to calculate ° D provided threshold temperatures with the smallest RMSE values. However, the iteration method using a linear ° D model also worked well. Simply using a threshold of zero gave predictions that were nearly as good as those obtained using the other two methods. The smallest standard deviation in ° D performed the worst. The least standard deviation in days and the regression methods did well sometimes; however, the threshold temperatures were sometimes negative, which does not support the idea that development rates are related to heat units. A comparison is made between the Pruitt and Doorenbos version of an hourly Penman-type equation, the Food and Agriculture Organization (FAO) hourly Penman-Monteith equation, and an independent measure of reference evapotranspiration (ET0) from lysimeter data. Reducing the canopy resistance improved the hourly FAO Penman-Monteith estimates. Daytime soil heat flux density is estimated as 10% of net radiation in the FAO hourly Penman-Monteith equation; however, the measured soil heat flux density under grass that was never shorter than 0.10 m in this study was between 3% and 5% of net radiation. The daytime totals of hourly ET0 from the hourly Penman-Monteith and Pruitt-Doorenbos equations and ET0 from the 24-h FAO Penman-Monteith equation were computed using data from five Italian and five Californian stations. A comparison showed that all of the equations gave acceptable results. The Pruitt-Doorenbos equation may slightly over-estimate ET0 in conditions of summertime cold air advection. In this article, all of the known superconducting systems are surveyed, and the structural conditions required for high-temperature superconductivity in the cuprate system are reviewed. This article does not focus on the details of the individual crystal structures of the known superconducting cuprates, but on the commonalties among all the different structures to understand the structural requirements for superconductivity in these ceramic materials. Rapid axonal transport is generally viewed as being exactly analogous to the secretory process in nonneuronal cells. The cell biology of rapid axonal transport is reviewed, the central concern being to explore those aspects that do not fit into the general secretory model and which may thus represent specific neuronal adaptations. Particular attention is paid to the relationship between the transport of newly synthesized proteins and of the membranous organelles that act as carriers. Sites in the transport sequence at which the behavior of axonal transport may differ from the secretory model are at the initiation of axonal transport at the trans-side of the Golgi apparatus, within the axon where molecules are deposited from the moving phase to a stationary phase, and at nerve terminals or axonal lesions where transport reversal takes place. The relation between the effect of silver on the amount of residual carbon and the rate of carbonate build-up on the surface of the particles of Tl1212 and Bi2212 was studied by infrared spectroscopy using a diffuse reflectance infrared–Fourier transform (DRIFT) cell. The result showed that appreciable amounts of carbonate still remained after calcination at 940°C when carbonates were used as starting materials. Samples prepared using oxides also showed the presence of carbonate that originated from the CO2 adsorption at the surface during processing. The small amounts of surface carbonates were below the detection limit of X-ray diffraction. However, the DRIFT technique has been found to be a convenient and effective method for qualitative and semi-quantitative analysis of carbonate formation on the powder surface and atmospheric degradation of thallium- and bismuth-compound superconductors. The presence of silver decreased the rate of the carbonate formation on the Tl1212 and Bi2212 particle surface influencing the degradation process. Carbonate formation was more sensitive to the presence of silver in Tl1212 than in Bi2212. A heptavalent lipopolysaccharide vaccine of Pseudomonas aeruginosa (Pseudogen) was administered at four dose levels (0.1, 0.25, 0.5, and 0.75 mg/m) to patients with advanced metastatic cancer that had proved refractory to chemotherapy. The vaccine was administered subcutaneously twice weekly. Local toxicity was seen in erythema, edema, pain and tenderness at the site of injection, and painful regional lymphadenopathy; manifestations of systemic toxicity included fever, chills, myalgias, nausea, and vomiting. Toxicity showed a clear-cut dose dependence. The maximally tolerated dose by this route and schedule was 0.5 mg/m. A significant rise of antibody titers was observed at all four dose levels. Evaluation of the delayed cutaneous hypersensitivity response to recall antigens and to the pseudomonas vaccine, and quantification of peripheral blood T and B cell levels and of in vitro lymphocyte blastogenic responses to commonly used mitogens and pseudomonas vaccine failed to demonstrate significant change from pretreatment values. Clinical trials to evaluate the therapeutic efficacy of pseudomonas vaccine with or without chemotherapy can be undertaken safely with this route and schedule. [ABSTRACT FROM AUTHOR]

The applicability of some existing droplet sedimentation theories to erythrocyte suspensions was investigated using glutaraldehydefixed erthrocytes from horse, canine, pig, chicken, and human. The Svensson criteria were shown to underestimate the load supportable by a density gradient. The available theories on droplet formation times could not predict each other, and the data on erythrocyte suspensions, especially at high suspension concentrations. It was finally argued that, since the behavior of erythrocytes was not controlled by the diffusion process, the erythrocyte suspensions were not expected to exhibit lasting or absolute stability even when the particle load was small. Changes in the average linear attenuation coefficient (LAC) within a fixed measurement volume in the proximal end of the dog tibia, which contains trabecular bone and associated soft tissues (the trabecular bone "space"), were monitored continuously using gamma-ray computed tomography (gamma-CT) prior to, during, and following intravenous infusion of strontium (Sr) lactate. An infusion of 1.3-4.7 g of Sr over a period of 110-160 minutes into 20-kg dogs resulted, within 6-8 hours, in an increase of 0.019-0.045 cm-1 (P less than 0.002) in the LAC. Calibration of the gamma-CT system showed that 0.44 mg/cm3 of Sr produced a change of 0.01 cm-1 in the LAC. Using this conversion factor, the Sr concentration in the trabecular bone space resulting from infusion, as measured by flame atomic absorption spectroscopy, agreed with that predicted by the change observed in the LAC. Sr present in the serum and urine was consistent with the changes observed in the LAC over the study period. Control dogs infused with mineral-free solutions showed no change in LAC. Calcium equivalents required to give the changes observed in the LAC using Sr indicate that variations in skeletal turnover in man can be monitored in the peripheral skeleton using gamma-CT. High temperature x-ray diffraction was used to determine the effect of Ag on the peritectic melting sequence of low carbon content BiSrCaCuO. Only slight changes in the solid solution compositions were found with 20 wt.% Ag added to the system, with the phases and stability regions remaining the same. Micro-structure, sintering behavior, lattice parameters, and superconducting property changes in TlPbSrCaCuO were investigated with different amounts of Ag added to investigate the effect of Ag. From the results in both systems, the presence of Ag does not seem to change the intrinsic properties of these superconductors, but affects the processing variables, like reaction temperatures, which change the optimum temperatures for obtaining best properties. The effects of a variable water supply on the water use, growth and yield of two crisphead and one romaine (i.e., Cos) lettuce cultivar were examined in a field experiment using a line source sprinkler system that produced a range of water regimes that occur in growers fields. Four locations at increasing distances from the main line were monitored through the season (i.e., from thinning to harvest, 28-63 days after planting (DAP)). These locations at the end of the season corresponded to: (1) rewatering to field capacity (FC); (2) watering with a volume 13% below that required in the field capacity treatment (0.87*FC); (3) 30% below FC (0.70*FC); and (4) 55% below FC (0.45*FC). A linear production function for dry matter accumulation and fresh weight vs. crop evapotranspiration (ET) was determined for lettuce during this period, giving a water use efficiency for dry matter of 1.86 g m mm and for fresh weight of 48 g m mm . For lettuce irrigated to field capacity, ET between thinning and harvest was 146 mm; maximum crop coefficients of 0.81-1.02 were obtained at maturity (55-63 DAP). For the three irrigation treatments receiving the largest water application, ET was higher in the Cos culivar than in the two crisphead lettuce cultivars which had similar ET. Plant fresh weight was more sensitive than dry weight to reduction in water supply. In the FC treatment, root length density and soil water extraction were greatest in the top 0-45 cm, and decreased rapidly below 45 cm depth. Soil water extraction by roots increased at lower depths when irrigation was reduced. Instantaneous rates of leaf photosynthesis and leaf water potential showed no response to the irrigation treatments in this study, despite differences in biomass production. Evaporation was determined to be the major component of ET for 45 of the 63 days of the growing season. The large loss of water by evaporation during mid-season and the apparent insensitivity of lettuce to the volume of irrigation during this period may provide an opportunity for reducing irrigation applications. Reactive metabolites of benzene (BZ) play important roles in BZ-induced hematotoxicity. Although reactive metabolites of BZ covalently bind to DNA, the significance of DNA adduct formation in the mechanism of BZ toxicity is not clear. These studies investigated the covalent binding of the BZ metabolites hydroquinone (HQ) and 1,2,4-benzenetriol(BT) using the DNA [P]postlabeling method and explored the potential relationship between DNA adduct formation and cell differentiation in human promyelocytic leukemia (HL-60) cells, a model system for studying hematopoiesis. Maturation of HL-60 cells to granulocytes, as assessed by light and electron microscopy, was significantly inhibited in cells that were pretreated with HQ or BT prior to inducing differentiation with retionic acid (RA). The capacity of RA-induced cells to phagocytose sheep red blood cells (RBC) and to reduce nitroblue tetrazolium (NBT), two functional parameters characteristic of mature, differentiated neutrophils, was also inhibited in cells pretreated with HQ or BT. These BZ metabolite treatments induced DNA adduct formation in HQ- but not in BT-treated cells. These results indicate that whereas HQ and BT each block granulocytic differentiation in HL-60 cells, DNA adducts were observed only following HQ treatment. Rat liver mitoplasts were incubated first with []dGTP, to form DNA labeled in G, and then with [C]benzene. The DNA was isolated and upon isopycnic density gradient centrifugation in CsCl yielded a single fraction of DNA labeled with both [] and [C]. These data are consistent with the covalent binding of one or more metabolites of benzene to DNA. The DNA was enzymatically hydrolyzed to deoxynucleosides and chromatographed to reveal at least seven deoxyguanosine adducts. Further studies with labeled deoxyadenine revealed one adduct on deoxyadenine. []Deoxyguanosine was reacted with [C]hydroquinone or benzoquinone. CT in childhood bacterial meningitis has indicated that bacterial meningitis is more than an inflammation of the subarachnoid space; it is a disease process which often exerts a profound effect on the brain parenchyma. In some cases ventricular widening may be a reflection of loss of brain substance rather than increased intracranial pressure. The trophic link between bacteria and bacterivorous protists is a complex interaction that involves feedback of inorganic nutrients and growth substrates that are immeadiately available for prey growth. These interactions were examined in the laboratory and in incubations ofconcentrated natural assemblages of bacterioplankton. Growth dynamics of estuarine and marine bacterivorous protists were determined in laboratory culture using Vibrio natriegens as prey and were compared to growth of protists on bacterioplankton assemblages concentrated by tangential flow filtration from four northwest Florida Estuaries. Biomass transfers from bacteria to protists were monitored by tracing elemental carbon and nitrogen in particulate fractions of protist addedand grazer free controls. Gross growth efficiencies of the protists on naturally occurring bacteria were within the range determined in lab estimates of growth efficiency on cultured bacteria (~50%). However, bacterial response to protist excretion products was different in the lab and field incubations, and bacterial growth contributed to the biomass available to protists in the field incubations. As determined by radioisotope-labeled substrate incorporation, a time lag in bacterial response to protist excretion products was observed for laboratory batch cultures, allowing accurate estimation of growth efficiency. In incubations with concentrated natural bacterial assemblages, bacterial growth response coincided with protist growth and excretion. The additional bacterial production on protist excretion products reached a maximum of 2--3-fold higher than protist-free controls. In addition, ammonium concentrations increased with protist grazing and growth in lab cultures, but ammonium excreted by protists in concentrates did not accumulate. The C:N values for the bacterial concentrates suggests that these bacteria were nitrogen limited. The relevance of KRAS mutation alleles to clinical outcome remains inconclusive in pancreatic adenocarcinoma (PDAC). We conducted a retrospective study of 803 patients with PDAC (42% with metastatic disease) at MD Anderson Cancer Center. Overall survival (OS) analysis demonstrated that KRAS mutation status and subtypes were prognostic (p < 0.001). Relative to patients with KRAS wildtype tumors (median OS 38 months), patients with KRAS G12R had a similar OS (median 34 months), while patients with KRAS Q61 and KRAS G12D mutated tumors had shorter OS (median 20 months [HR: 1.9, 95% CI 1.2-3.0, p = 0.006] and 22 months [HR: 1.7, 95% CI 1.3-2.3, p < 0.001], respectively). There was enrichment of KRAS G12D mutation in metastatic tumors (34% vs 24%, OR: 1.7, 95% CI 1.2-2.4, p = 0.001) and enrichment of KRAS G12R in well and moderately differentiated tumors (14% vs 9%, OR: 1.7, 95% CI 1.05-2.99, p = 0.04). Historically, International Space Station (ISS) exercise countermeasures have not fully protected astronauts' musculoskeletal and cardiorespiratory fitness. Although these losses have been reduced on more recent missions, decreasing the time required to perform in-flight exercise would permit reallocation of that time to other tasks. To evaluate the effectiveness of a new training prescription, ISS crewmembers performed either the high intensity/lower volume integrated Sprint resistance (3 d wk -1 ) and aerobic (interval and continuous workouts, each 3 d wk -1 in alternating fashion) exercise program ( n  = 9: 8M/1F, 48 ± 7 y, 178 ± 5 cm, 77.7 ± 12.0 kg) or the standard ISS countermeasure consisting of daily resistance and aerobic exercise ( n  = 17: 14M/3F, 46 ± 6 y, 176 ± 6 cm, 80.6 ± 10.5 kg) during long-duration spaceflight. Bone mineral density (dual energy X-ray absorptiometry (DXA)), muscle strength (isokinetic dynamometry), muscle function (cone agility test), and cardiorespiratory fitness (VO 2peak ) were assessed pre- and postflight. Mixed-effects modeling was used to analyze dependent measures with alpha set at P  < 0.05. After spaceflight, femoral neck bone mineral density (-1.7%), knee extensor peak torque (-5.8%), cone agility test time (+7.4%), and VO 2peak (-6.1%) were decreased in both groups (simple main effects of time, all P  < 0.05) with a few group × time interaction effects detected for which Sprint experienced either attenuated or no loss compared to control. Despite major initiatives in public and professional education about foetal alcohol syndrome (FAS) in Saskatchewan in the last 20 years, its incidence rate has not fallen. The rate was 0.515 per 1,000 live births in 1973-1977 and 0.589 in 1988-1992. Two hundred and seven (207) cases were ascertained, the majority being patients of the Alvin Buckwold Child Development Program in Saskatoon. These individuals were severely handicapped: 72% had at least one malformation, the mean intelligence quotient was 67.8 (range 35-106) and 45.9% had a behaviour problem. Only 25.6% still lived with their biological parents when last seen, and only 27 of 108 cases were in a regular class at school without additional support being necessary. New approaches are needed to reduce the incidence of FAS. Emphasis should be placed on individual case-finding, counselling for high-risk women, and community development programs. We are currently attempting this through a provincial coordinating committee.
