Purposes: Developing the cultivation and utilization of tiger nuts on marginal land is an effective approach to tackle the shortage of arable land in northern China. However, key information on their growth is unclear. This study aims to reveal the adaption potential of tiger nuts to on marginal land to help improve and adjust management practices. Methods: Three treatments were arranged to plant tiger nut in marginal land: no fertilization (N:P = 0:0), traditional fertilization (N:P = 15:15), and additional nitrogen fertilizer (N:P = 60:15). Functional traits of tiger nut and soil nutrients over three growth periods were investigated. Results: Under no fertilization, tiger nuts extended roots and toughened leaves to maximize nutrient absorption, and then quickly transported those nutrients to tubers. However, the lack of available phosphorus in the soil prevented the expansion of tubers. With the addition of fertilizers, tiger nuts produced more individuals, and their abundant belowground tissues resulted in soil organic matter increase. Under traditional fertilization, the tuber remains the main reproductive organ for tiger nuts, and phosphorus has a significant effect on the expansion of tubers. But under additional nitrogen fertilizer, the main reproductive organ changed to the stolon, nitrogen and phosphorus acted on stolons to promote their extension. Conclusion: Tiger nuts outstandingly adapt to marginal land environments and adjust their adaptation strategies according to varying soil nutrient conditions, Best tuber production will be achieved in soils with higher phosphorus availability, and a significant nitrogen proportion increase in fertilizers is necessary for enhancing aboveground production. Intermittent hypoxia (IH) is the predominant pathophysiological disturbance in obstructive sleep apnea (OSA), characterized by neuronal cell death and neurocognitive impairment. We focus on the accumulated mitochondrial DNA (mtDNA) in the cytosol, which acts as a damage-associated molecular pattern (DAMP) and activates the cyclic GMP-AMP synthase (cGAS)—stimulator of interferon genes (STING) pathway, a known trigger for immune responses and neuronal death in degenerative diseases. However, the specific role and mechanism of the mtDNA-cGAS-STING axis in IH-induced neural damage remain largely unexplored. Here, we investigated the involvement of PANoptosis, a novel type of programmed cell death linked to cytosolic mtDNA accumulation and the cGAS-STING pathway activation, in neuronal cell death induced by IH. Our study found that PANoptosis occurred in primary cultures of hippocampal neurons and HT22 cell lines exposed to IH. In addition, we discovered that during IH, mtDNA released into the cytoplasm via the mitochondrial permeability transition pore (mPTP) activates the cGAS-STING pathway, exacerbating PANoptosis-associated neuronal death. Pharmacologically inhibiting mPTP opening or depleting mtDNA significantly reduced cGAS-STING pathway activation and PANoptosis in HT22 cells under IH. Moreover, our findings indicated that the cGAS-STING pathway primarily promotes PANoptosis by modulating endoplasmic reticulum (ER) stress. Inhibiting or silencing the cGAS-STING pathway substantially reduced ER stress-mediated neuronal death and PANoptosis, while lentivirus-mediated STING overexpression exacerbated these effects. In summary, our study elucidates that cytosolic escape of mtDNA triggers cGAS-STING pathway-dependent neuronal PANoptosis in response to IH, mainly through regulating ER stress. Initially, a model was created for image-level classification of implant cutouts using 40191 pelvic radiographs obtained from a single institution. The radiographs were partitioned into training, validation, and hold-out test datasets in a 6/2/2 ratio. Performance metrics including the area under the receiver operator characteristics curve (AUROC), sensitivity, and specificity were calculated using the test dataset. Additionally, a second object detection model was trained to localize implant cutouts within the same dataset. Bounding box visualizations were generated on images predicted as cutout-positive by the classification model in the test dataset, serving as an adjunct for assessing algorithm validity.The classification model had an accuracy of 99.7%, sensitivity of 84.6%, specificity of 99.8%, AUROC of 0.998 (95% CI: 0.996, 0.999) and AUPRC of 0.774 (95% CI: 0.646, 0.880). Inspired by an open question by Chemin, Zhang and Zhang about the regularity of the 3D Navier–Stokes equations with one initially small component, we investigate symmetry breaking and symmetry preservation. Our results fall in three classes. First we prove strong symmetry breaking. Specifically, we demonstrate third component norm inflation (3rdNI) and Isotropic Norm Inflation (INI) starting from zero third component. Second we prove symmetry breaking for initially zero third component, even in the presence of a favorable initial pressure gradient. Third we study certain symmetry preserving solutions with a shear flow structure. Silent information regulator factor 2-related enzyme 1 (sirtuin 1, Sirt1) is a nicotinamide adenine dinucleotide-dependent deacetylase, which can deacetylate histone and non-histone proteins and other transcription factors, and is involved in the regulation of many physiological functions, including cell senescence, gene transcription, energy balance, and oxidative stress. Ischemia/hypoxia injury remains an unresolved and complicated situation in the diseases of ischemia stroke, heart failure, and coronary heart disease, especially among the old folks. Studies have demonstrated that aging could enhance the vulnerability of brain, heart, lung, liver, and kidney to ischemia/hypoxia injury and the susceptibility in old folks to ischemia/hypoxia injury might be associated with Sirt1. In this review, we mainly summarize the role of Sirt1 in modulating pathways against energy depletion and its involvement in oxidative stress, apoptosis, and inflammation under the condition of ischemia/hypoxia. Purpose: Elevated carboxypeptidase E (CPE) levels play crucial roles in tumorigenesis and metastasis. This study investigated the expression and clinicopathological significance of CPE in early-stage cervical cancer.Methods: Elevated carboxypeptidase E expression was analyzed using quantitative polymerase chain reaction and western blotting in normal cervical tissue, cervical cancer cell lines, and in cervical cancer tissues and adjacent noncancerous tissues (ANTs) from the same patient. Immunohistochemistry (IHC) was used to examine CPE expression in tissue samples from 112 patients with early-stage cervical cancer (FIGO stages Ia2-IIa2), 60 patients with cervical intraepithelial neoplasia, and 19 patients with normal cervical tissues (NCTs). Associations between CPE expression and prognostic and diagnostic factors were evaluated statistically.Results: CPE expression was significantly higher in cervical cancer cell lines and tissues than in normal tissues and ANTs. Semi-quantitative analysis of IHC indicated that CPE gradually increased from CIN I to cervical cancer, but was absent in NCTs. CPE expression was seen in 40.2 % (45/112) of the cervical cancer samples. CPE expression was significantly associated with FIGO stage (P = 0.003), tumor size (P = 0.012), stromal invasion (P < 0.001), lymphovascular space invasion (P = 0.016), parametrial infiltration (P = 0.027), vaginal involvement (P = 0.007), postoperative adjuvant therapy (P = 0.024), recurrence (P < 0.001), survival (P < 0.001), and pelvic lymph node metastasis (PLNM) (P < 0.001), and it was significantly higher in tissues from patients with PLNM than without PLNM. Logistic regression analysis identified high-level CPE expression as an independent risk factor for PLNM (P = 0.001). Patients with higher CPE expression had shorter overall survival duration than patients with lower CPE expression. Univariate and multivariate Cox-regression analyses suggested that high-level CPE expression is an independent prognostic factor for overall survival in early-stage cervical cancer.Conclusions: High-level CPE expression was associated with a poor prognosis in early-stage cervical cancer. Background: Laparoscopic liver resection (LLR) has now been established as a safe and minimally invasive technique that is deemed feasible for treating hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (ICC). However, the role of LLR in treating combined hepatocellular-cholangiocarcinoma (cHCC-CC) patients has been rarely reported. This study aimed to assess the efficacy of LLR when compared with open liver resection (OLR) procedure for patients with cHCC-CC. Methods: A total of 229 cHCC-CC patients who underwent hepatic resection (34 LLR and 195 OLR patients) from January 2014 to December 2018 in Zhongshan Hospital, Fudan University were enrolled and underwent a 1:2 propensity score matching (PSM) analysis between the LLR and OLR groups to compare perioperative and oncologic outcomes. Overall survival (OS) and recurrence-free survival (RFS) parameters were assessed by the log-rank test and the sensitivity analysis. Results: A total of 34 LLR and 68 OLR patients were included after PSM analysis. The LLR group displayed a shorter postoperative hospital stay (6.61 vs. 8.26 days; p value < 0.001) when compared with the OLR group. No significant differences were observed in the postoperative complications' incidence or a negative surgical margin rate between the two groups (p value = 0.409 and p value = 1.000, respectively). The aspartate aminotransferase (AST), alanine aminotransferase (ALT), and inflammatory indicators in the LLR group were significantly lower than those in the OLR group on the first and third postoperative days. Additionally, OS and RFS were comparable in both the LLR and OLR groups (p value = 0.700 and p value = 0.780, respectively), and similar results were obtained by conducting a sensitivity analysis. Conclusion: LLR can impart less liver function damage, better inflammatory response attenuation contributing to a faster recovery, and parallel oncologic outcomes when compared with OLR. Therefore, LLR can be recommended as a safe and effective therapeutic modality for treating selected cHCC-CC patients, especially for those with small tumors in favorable location. Reactive oxygen species (ROS) and antioxidant ingredients are a series of crucial signaling molecules in oxidative stress response. Under some pathological conditions such as traumatic brain injury, ischemia/reperfusion, and hypoxia in tumor, the relative excessive accumulation of ROS could break cellular homeostasis, resulting in oxidative stress and mitochondrial dysfunction. Meanwhile, autophagy is also induced. In this process, oxidative stress could promote the formation of autophagy. Autophagy, in turn, may contribute to reduce oxidative damages by engulfing and degradating oxidized substance. This short review summarizes these interactions between ROS and autophagy in related pathological conditions referred to as above with a focus on discussing internal regulatory mechanisms. The tight interactions between ROS and autophagy reflected in two aspects: the induction of autophagy by oxidative stress and the reduction of ROS by autophagy. The internal regulatory mechanisms of autophagy by ROS can be summarized as transcriptional and post-transcriptional regulation, which includes various molecular signal pathways such as ROS-FOXO3-LC3/BNIP3-autophagy, ROS-NRF2-P62-autophagy, ROS-HIF1-BNIP3/NIX-autophagy, and ROS-TIGAR-autophagy. Autophagy also may regulate ROS levels through several pathways such as chaperone-mediated autophagy pathway, mitophagy pathway, and P62 delivery pathway, which might provide a further theoretical basis for the pathogenesis of the related diseases and still need further research. Flat-top transmission bands are found in a plasmonic structure constructed by periodic ring resonators. Fabry-Perot effects can be suppressed in a set of parameters determined by an effective medium method. Two kinds of transmission bands with flat-top profiles can emerge due to either ring resonance or Bragg scattering. These bands are both red shifted when the distance of rings increases. Both transfer matrix method and finite difference time domain method are utilized to investigate the flat-top bands. Introduction: Interactions of different hemoglobin variants with thalassemia alleles can result in various clinical phenotypes. HbE-beta-thalassemia generally manifests with severe anemia where individuals exhibit beta-thalassemia major with regular blood transfusions or beta-thalassemia intermedia with periodic blood transfusions. This study presents a unique Malay family with three beta-globin gene defects-HbE, Hb South Florida, and IVS1-1 (G-->A).Materials and Methods: HbE activates a cryptic splice site that produces non-functional mRNAs. Hb South Florida is a rare beta-hemoglobin variant, and its interactions with other beta-thalassemia alleles have not been reported. IVS1-1 is a Mediterranean mutation that affects mRNA processing giving rise to beta(o)-thalassemia.Results and Discussion: Fifteen mutations along the beta-globin gene complex were analyzed using the amplification refractory mutation system. Hb South Florida was identified by direct sequencing using genomic DNA.Conclusion: The affected child with HbE/IVS1-1 produced a beta-thalassemia major phenotype. Introduction: Real-world data on the comparative effectiveness of endothelin receptor antagonists (ERAs; macitentan, bosentan, ambrisentan) for pulmonary arterial hypertension (PAH), particularly in Asian countries, are scarce. We evaluated the persistence of these ERAs before and after macitentan approval in Japan (2015). Methods: We used real-world data from the Japanese Medical Data Vision administrative claims database between April 2008 and November 2020. Patients with PAH were identified from the dataset. Persistence to ERA treatment before and after approval of macitentan in Japan was defined as the time between start of the index ERA and treatment discontinuation or death. Propensity score adjustment was applied to minimize confounding effects among treatment groups. Results: In the pre-macitentan approval cohort, 153 and 51 patients received bosentan and ambrisentan, respectively. In the post-macitentan approval cohort, 331, 284, and 91 patients received macitentan, bosentan, and ambrisentan, respectively. Unadjusted median persistence for ambrisentan- and bosentan-treated patients was 19 and 10 months, respectively (adjusted HR 0.87 [95% CI 0.61–1.24]; P = 0.434 [bosentan as reference]). In the post-macitentan approval cohort, unadjusted median persistence was 18 months for macitentan-treated patients versus 6 and 8 months for ambrisentan- and bosentan-treated patients, respectively. Adjusted HRs for ambrisentan and bosentan were 1.48 (95% CI 1.12–1.95; P = 0.006) and 1.63 (95% CI 1.30–2.04; P < 0.001 [macitentan as reference]), respectively. Conclusions: Real-world data for Japanese patients with PAH showed that persistence was significantly higher for macitentan, versus ambrisentan and bosentan, since its approval. An urban subway network system is a complex public transportation system. To compare rail transit braking digital command control strategies based on neural network, this article analyzes and studies the characteristics of subway vehicle driver controllers and their design methods from three aspects: mechanical, electrical and software-assisted design. The learning rule of the BP neural network is called the mentor system learning rule, which is a kind of error-correcting algorithm. In the learning and training process, the expected output value needs to be given. The weights and thresholds of the BP neural network are optimized by selecting the parameters of the SA algorithm. The search method of SA is heuristic, and it has the following advantages: The selection of the initial solution does not affect the optimal solution. The simplified model extracts the core data processing individual analysis. In this paper, the physical data are extracted from the physical entity operation process for analysis, and the twin model is established to extract the twin data for analysis. This paper uses the characteristics of physical data to test the modeling effect and utilizes the twin data to carry out algorithm experiments on physical data. The ultimate goal is to use twin data to predict the state information of physical entities. The network error in the scheme designed by the article is 6%. The smooth implementation of this research constitutes an important reference for the design of subway train network control systems in other cities in China. Understanding of the complex mechanical behavior of metal-organic frameworks (MOF) beyond their elastic limit will allow the design of real-world applications in chemical engineering, optoelectronics, energy conversion apparatus, and sensing devices. Through in situ compression of micropillars, the uniaxial stress-strain curves of a copper paddlewheel MOF (HKUST-1) were determined along two unique crystallographic directions, namely the (100) and (111) facets. We show strongly anisotropic elastic response where the ratio of the Young's moduli are E(111) ≈ 3.6 × E(100), followed by extensive plastic flows. Likewise, the yield strengths are considerably different, in which Y(111) ≈ 2 × Y(100) because of the underlying framework anisotropy. We measure the fracture toughness using micropillar splitting. While in situ tests revealed differential cracking behavior, the resultant toughness values of the two facets are comparable, yielding Kc ~ 0.5 MPa m . This work provides insights of porous framework ductility at the micron scale under compression and failure by bonds breakage. Understanding the complex mechanical behaviour of metal–organic frameworks is of importance for their real-world application. In this work, the distinctions in the synergistic effects between carbon nanotubes (CNTs) and different metal oxide (MeO) nanoparticles on enhancing the thermal oxidative stability of silicone rubber (SR) were studied in depth. Specifically, three kinds of MeO nanoparticles were attached onto the surface of CNTs, obtaining FeO-CNTs, TiO-CNTs, and SnO-CNTs, respectively. These MeO nanoparticles-attached CNTs were then separately embedded into SR matrix to investigate their effect on the thermal oxidative stability of SR. The results indicated that different synergies between CNTs and different metal oxides existed: both FeO and SnO had positive synergistic effects with CNTs, while a negative synergic effect existed between TiO and CNTs. This phenomenon may be attributed to the fact that the existence of CNTs could promote the radical-capture reactions to some extent for Fe and Sn element, while a reverse trend was found in TiO-CNTs/SR. Meanwhile, all the performances of SnO-CNTs/SR exhibited a much higher positive synergy than those of FeO-CNTs/SR: the synergy percentages of the tear strength and elongation at break for SnO-CNTs/SR were even above 100 %. This result may be ascribed to the combined effect of the multi-electron transfer process in SnO-CNTs/SR and the restriction of crystalline form of FeO in FeO-CNTs/SR during thermal aging. A key component in this process is 2,5-bis(hydroxymethyl)furan (BHMF), a significant biomass-derived platform chemical. However, producing BHMF with high selectivity through the selective hydrogenation of 5-hydroxymethylfurfural, while avoiding the creation of unwanted byproducts, is a complex challenge. In our research, we have developed a novel catalyst featuring a magnetic core. This catalyst, which consists of platinum loaded onto a graphene-like shell, has been applied to the catalytic hydrogenation of HMF in an aqueous environment. Under the optimized conditions of 373 K, 3 MPa H2, and 1 h reaction time, a BHMF yield exceeding 99% was achieved. The injurious effect of hydrogen peroxide (HO) on renal epithelial cells of the African green monkey (Vero cells) and the difference in the modulation of Vero cells on crystal growth of calcium oxalate (CaOxa) before and after injury were investigated. The degree of injury of Vero cells was proportional to the concentration and action time of HO. After the cells had been injured, the released amount of malonaldehyde in the culture medium increased, the superoxide dismutase activity decreased, the expression quantity of osteopontin on the surface of Vero cells increased significantly, the zeta potential became more negative, and the amount of CaOxa crystals adhering to cells increased. The CaOxa crystals induced by the cells in the control group were round and blunt; however, those induced by the injured cells had irregular shapes with sharp edges and corners. As the crystallization time increased from 6 to 24 h, the size of the crystals induced by the injured cells increased accordingly, whereas that of crystals induced by the control cells did not increase significantly. The injured cells could promote the growth of CaOxa crystals and their adhesion to the cells; thus, the formation of CaOxa stones was promoted. The cells in the control group could also be injured after being incubated with supersaturated CaOxa solution for a long time, which promoted the crystallization of CaOxa. The results suggest that the retention of supersaturated CaOxa solution or CaOxa crystals in the urinary tract for a long time is a risk factor for the formation of kidney stones. Coilia nasus is influenced by various external pressures during spawning migration and these anadromous transitions can lead to specific gut microbiome characteristics that affecting the host biological process. Therefore, the purpose of this study was to determine the variations of components and functions in the gut prokaryotic microbiome during spawning migration as well as the key factors that triggered the changes. The gut microbiome in C. nasus was mainly consisted of Proteobacteria, Bacteroidetes, Firmicutes, Deinococcus-Thermus and Fusobacteria via 16S rRNA Gene Amplicon Sequencing. The relative abundance of Acinetobacter and Clostridium increased, while Corynebacterium, Actinomyces, Bacillus, Klebsiella and Ochrobactrum decreased after entering freshwater, indicated the preference of C. nasus gut microbial members transferred from seawater to freshwater. Additionally, the proportion of Firmicutes significantly decreased and then increased, as well as the arise of some soil bacteria in gut, corresponding to the phenomenon that C. nasus are fasting during the upstream process and refeeding after entering the spawning grounds. The function prediction of gut microbiome was also consistent with the above results. The present study generally demonstrated the gut microbiome dynamics and the significant correlation between the gut microbiome and salinity and feeding behavior in the spawning migration of C. nasus. How to screen and evaluate the radical scavenging activity of each compound in the mixture remains a challenging project. In the present study, a sensitive and selective method of offline HPLC coupled with chemical methodology was developed for the evaluation of the radical scavenging activity of each compound in a common vegetable, crowndaisy (Chrysanthemum coronarium L.) seeds alcohol extract (CDAE) without the isolation of monomers. The results of LC–ESI–MS/MS showed that the main components of CDAE were chlorogenic acid (3-CQA) (1693.0 μg/g, dry weight), 1,3-dicaffeoylquinic acid (1,3-DCQA, 1378.5 μg/g), 3,5-DCQA (1027.0 μg/g), and 1,4-DCQA (1031.5 μg/g). The results of scavenging free radical activity analysis showed that, among all components, 1,3-DCQA, 3,5-DCQA, 1,4-DCQA, and 3-CQA were mainly responsible for the radical scavenging activity of the extract. Their scavenging rates for 2,2-diphenyl-1-picrylhydrazyl (DPPH) were 86.72%, 83.50%, 83.50%, and 83.43%, respectively. Laboratory-scale column experiments were conducted to investigate the influence of water table fluctuation on light non-aqueous phase liquid (LNAPL) migration and redistribution in porous media. A simplified image analysis method (SIAM) was used to evaluate the saturation distributions of the LNAPL and water in the entire domain under dynamic conditions. Two LNAPL volumes differentiated as low and high volumes were exposed to two patterns of water table fluctuations. High-resolution SIAM images of the soil column during LNAPL migration and subsequent water table fluctuations were collected and analyzed. Results show that when the water table was lowered in the experiment of low LNAPL volume by 10 cm (from 28 to 18 cm) and then raised back to its initial location (28 cm), the water saturation dropped from 100% initially to around 30% while the LNAPL saturation increased and remained at 7% to 25% at 36 cm. By doubling the LNAPL volume to 50 ml, the initial LNAPL saturation above the water table increased and the capillary fringe height increased from 36 to 38 cm. Comparison between the two LNAPL volumes showed that the LNAPL covered a larger area for the case of high LNAPL. The LNAPL saturation extended up to around 53 cm while for the case of low LNAPL, the LNAPL extended up to height of 50 cm. Moreover, water saturation below was lesser for the case of high LNAPL. This was due to the increasing volume of LNAPL which caused the water to be pushed downward. However, similar downward migration of water was observed in both sets of experiments. This indicates that doubling the volume of LNAPL while maintaining the same groundwater fluctuation pattern did not affect the general LNAPL behavior. Overall, this study demonstrates that the SIAM technique is an accurate and cost-effective tool for the visualization of the time-dependent NAPL/water movement in laboratory-scale experiments and dynamic changes in fluid saturation in porous media. In our recent study, we observed consistent increases in miR-124-3p levels in exosomes derived from cultured BV2 microglia which was treated with repetitive traumatic brain injury (rTBI) mouse model brain extracts. To clarify the mechanisms underlying increases in microglia-derived exosomal miR-124-3p and their role in regulating neuronal autophagy after TBI, we investigated the impact of exosomal miR-124-3p on neuronal autophagy in scratch-injured HT22 neurons and rTBI mice. We harvested injured brain extracts from rTBI mice at 3 to 21 days post injury (DPI) for the treatment of cultured BV2 microglia in vitro. We observed significant induction of autophagy following TBI in vitro, and that inhibition of activated neuronal autophagy could protect against trauma-induced injury. Our results indicated that co-culture of injured HT22 neurons with miR-124-3p overexpressing BV2 microglia exerted a protective effect by inhibiting neuronal autophagy in scratch-injured neurons. Further research revealed that these effects were achieved mainly via upregulation of exosomal miR-124-3p, and that Focal adhesion kinase family-interacting protein of 200 kDa (FIP200) plays a key role in trauma-induced autophagy. Injection of exosomes into the vena caudalis in in vivo experiments revealed that exosomal miR-124-3p was associated with decreases in the modified neurological severity score (mNSS) and improvements in Morris water maze (MWM) test results in rTBI mice. Altogether, our results indicate that increased miR-124-3p in microglial exosomes following TBI may inhibit neuronal autophagy and protect against nerve injury via their transfer into neurons. Bacterial cellulose (BC) is a polysaccharide with tubular structure and can be produced by various species of bacteria particularly Acetobacter xylinum. It is a promising matrix for fabricating electrochemical devices as a mechanically strong, flexible, and biocompatible carbon-based material. In the present work, BC and graphene oxide (GO) composite was obtained simply via a mixing method. The scanning electron microscopy (SEM) and transmission electron microscopy (TEM) images showed that the BC nanofibers were imbedded in the GO wrinkled sheets. Raman spectra showed the D and G bans of BC-GO shifted towards higher frequency. Fourier transform infrared (FT-IR) spectroscopy spectra confirmed a hybrid structure was successfully obtained. Cyclic voltammetry (CV) results showed the BC-GO modified electrode had the best electrochemical activity. Nitrite could be oxidized using the BC-GO modified electrode in a wide range of pHs. The amperometric response result indicated the BC-GO modified GCE can be used to determine nitrite concentration in a wide linear range of 0.5 to 4590 μM with detection limit and sensitivity of 0.2 μM and 527.35 μA μM cm, respectively. Beta-thalassemia is one of the most prevalent inherited diseases and a public health problem in Malaysia. Malaysia is geographically divided into West and East Malaysia. In Sabah, a state in East Malaysia, there are over 1000 estimated cases of β-thalassemia major patients. Accurate population frequency data of the molecular basis of β-thalassemia major are needed for planning its control in the high-risk population of Sabah. Characterization of β-globin gene defects was done in 252 transfusion dependent β-thalassemia patients incorporating few PCR techniques. The study demonstrates that β-thalassemia mutations inherited are ethnically dependent. It is important to note that 86.9% of transfusion-dependent β-thalassemia major patients in Sabah were of the indigenous population and homozygous for a single mutation. The Filipino β0-deletion was a unique mutation found in the indigenous population of Sabah. Mutations common in West Malaysia were found in 11 (4.3%) patients. Four rare mutations (Hb Monroe, CD 8/9, CD 123/124/125 and IVS I-2) were also found. This study is informative on the population genetics of β-thalassemia major in Sabah. [ABSTRACT FROM AUTHOR]

The goal of this study was to analyze the patterns, therapeutic modalities, and short-term outcomes of patients with chest injuries in the aftermath of the Wen-Chuan earthquake, which occurred on May 12, 2008 and registered 8.0 on the Richter scale. Of the 1522 patients who were referred to the West China Hospital of Sichuan University from May 12 to May 27, 169 patients (11.1%) had suffered major chest injuries. The type of injury, the presence of infection, Abbreviated Injury Score (AIS 2005), New Injury Severity Score (NISS), treatment, and short-term outcome were all documented for each case. Isolated chest injuries were diagnosed in 129 patients (76.3%), while multiple injuries with a major chest trauma were diagnosed in 40 patients (23.7%). The mean AIS and the median NISS of the hospitalized patients with chest injuries were 2.5 and 13, respectively. The mortality rate was 3.0% (5 patients). Most of the chest injuries were classified as minor to moderate trauma; however, coexistent multiple injuries and subsequent infection should be carefully considered in medical response strategies. Coordinated efforts among emergency medical support groups and prior training in earthquake preparedness and rescue in earthquake-prone areas are therefore necessary for efficient evacuation and treatment of catastrophic casualties. Homozygosity for the α-thalassaemia Southeast Asian (α-SEA) and Filipino β0-thalassaemia (β-FIL) deletions can cause serious complications leading to foetal death or life-long blood transfusions. A rapid and accurate molecular detection assay is essential in populations where the deletions are common. In this study, gap-polymerase chain reaction (PCR) with high resolution melting (HRM) analysis was developed to detect both the large deletions. Melting curves at 86.9 ± 0.1 °C were generated by normal individuals without the α-SEA deletion, 84.7 ± 0.1 °C by homozygous α-SEA deletion individuals and two melting curves at 84.7 ± 0.1 °C and 86.9 ± 0.1 °C by α-SEA deletion carriers. Normal individuals without the β-FIL deletion produce amplicons with a melting temperature (Tm) at 74.6 ± 0.1 °C, homozygous β-FIL individuals produce amplicons with Tm at 73.6 ± 0.1 °C and heterozygous β-FIL individuals generate two amplicons with Tm at 73.6 ± 0.1 °C and 74.6 ± 0.1 °C. Evaluation using blinded tests on 220 DNA samples showed 100% sensitivity and specificity. The developed assays are sensitive and specific for rapid molecular and prenatal diagnosis for the α-SEA and β-FIL deletions. [ABSTRACT FROM AUTHOR]
