In this paper, we look for normalized solutions to the following non-autonomous Schrödinger equation - Δ u = λ u + h (x) | u | q - 2 u + | u | 2 ∗ - 2 u in R N , ∫ R N | u | 2 d x = a , where N ≥ 3 , a > 0 , λ ∈ R , h ≠ c o n s t and 2 ∗ = 2 N N - 2 is the Sobolev critical exponent. In the L 2 -subcritical regime (i.e. 2 < q < 2 + 4 N ), by proposing some new conditions on h, we verify that the corresponding Pohozaev manifold is a natural constraint and establish the existence of normalized ground states. Compared to the L 2 -subcritical regime, it is necessary to apply some reverse conditions to h provided that at least L 2 -critical regime (i.e. 2 + 4 N ≤ q < 2 ∗ ) is considered. We prove the existence of minimizer on the Pohozaev manifold of the associated energy functional and determine that the minimizer is a normalized solution by using the classical deformation lemma. Olefin polymerization is one of the most important chemical reactions in industry. This work presents a strategy that emphasizes the synergistic meta/para-steric hindrance of N-aryl groups and electronic effects in newly synthesized neutral salicylaldiminato nickel catalysts. These nickel(II) catalysts exhibit exceptional thermostability, ranging from 30 °C to 130 °C, demonstrating enhanced catalytic activities and broadly regulated polyethylene molecular weights (3–341 kg·mol−1) and controlled polymer branch density (2–102 brs/1000C). The preferred catalyst Ni3 with concerted steric and electronic effects enables the production of solid-state semi-crystalline polyethylene materials at temperatures below 90 °C. Notably, Ni3 exhibits an impressive tolerance of 110 °C and can withstand even the challenging polymerization temperature of 130 °C, leading to the production of polyethylene wax and oil. While photo-cross-linking (PXL) with alkyl diazirines can provide stringent distance restraints and offer insights into protein structures, unambiguous identification of cross-linked residues hinders data interpretation to the same level that has been achieved with chemical cross-linking (CXL). We address this challenge by developing an in-line system with systematic modulation of light intensity and irradiation time, which allows for a quantitative evaluation of diazirine photolysis and photo-reaction mechanism. Our results reveal a two-step pathway with mainly sequential generation of diazo and carbene intermediates. Diazo intermediate preferentially targets buried polar residues, many of which are inaccessible with known CXL probes for their limited reactivity. Moreover, we demonstrate that tuning light intensity and duration enhances selectivity towards polar residues by biasing diazo-mediated cross-linking reactions over carbene ones. This mechanistic dissection unlocks the full potential of PXL, paving the way for accurate distance mapping against protein structures and ultimately, unveiling protein dynamic behaviors. Stimulated by the recent experimental results on the fully-charm tetraquark states, we systematically calculate the mass spectra of the fully-charm tetraquark states in 8 [ c c ¯ ] ⊗ 8 [ c c ¯ ] color configuration via QCD sum rules. By constructing nine 8 [ c c ¯ ] ⊗ 8 [ c c ¯ ] type currents with quantum numbers J PC = 0 - + , 0 - - , 1 - + , 1 + - , 1 - - and 2 + + , we perform analytic calculation up to dimension six in the operator product expansion (OPE). We find the fully-charm tetraquark states with J PC = 1 + - , 2 + + lie around 6.48 ∼ 6.62 GeV while the fully-charm tetraquark states with J PC = 0 - + , 0 - - , 1 - - , 1 - + are about 6.85 ∼ 7.02 GeV. Notably, the mass predictions for the c c ¯ c c ¯ tetraquarks, specifically those with J PC = 2 + + , align with the broad structure identified by LHCb. Moreover, the masses of fully-charm tetraquarks with J PC = 0 - + and 1 - + are anticipated to match closely with the mass of X(6900), considering the margin of error. Such findings hint at the presence of some 8 [ c c ¯ ] ⊗ 8 [ c c ¯ ] components within the di- J / ψ structures observed by LHCb. We investigate a class of the nonlinear Schrödinger equation in R N - Δ u + V (x) u = | u | 2 ∗ - 2 u + λ | u | p - 2 u , where N ≥ 3 , λ > 0 and p ∈ (2 , 2 ∗) with 2 ∗ = 2 N N - 2 . Here, V (x) = V 1 (x) for x 1 > 0 and V (x) = V 2 (x) for x 1 < 0 , where V 1 , V 2 are periodic in each coordinate direction. By providing a splitting Lemma corresponding to non-periodic external potential, we obtain the existence of ground state solution for the above problem. We study an initial-boundary value problem of two-dimensional nonhomogeneous asymmetric fluids with magnetic field and density-dependent viscosity μ (ρ) . Applying Desjardins' interpolation inequality and delicate energy estimates, we show the global-in-time existence of a unique strong solution when ‖ ∇ μ (ρ 0) ‖ L q is properly small. In this paper, we investigate the following nonlinear Choquard equation with prescribed L 2 -norm constraint - Δ u = λ u + (| x | - 1 ∗ | u | 2) u in Ω , u = 0 on ∂ Ω , ∫ Ω | u | 2 d x = a 2 , where a > 0 , λ ∈ R appears as an unknown Lagrange multiplier and Ω ⊂ R 3 is an exterior domain with smooth boundary ∂ Ω ≠ ∅ such that R 3 \ Ω is bounded. By using the splitting lemma for the unconstrained problem in exterior domains, we prove the compactness of Palais–Smale sequences corresponding to the above problem at higher energy levels. Then combining the barycentric function and Brouwer degree theory, we establish the existence of positive normalized bound states for any a > 0 provided that R 3 \ Ω is contained in a small ball and explain that the restriction on domain Ω can be equivalently transferred to a. In addition, under the radial setting of domain Ω , we use genus theory to obtain the existence and multiplicity of radial normalized solutions for any a > 0 . Objectives: To evaluate the prognostic value of radiomics features based on late gadolinium enhancement (LGE) cardiac magnetic resonance (CMR) images in patients with cardiac amyloidosis (CA). Methods: This retrospective study included 120 CA patients undergoing CMR at three institutions. Radiomics features were extracted from global and three different segments (base, mid-ventricular, and apex) of left ventricular (LV) on short-axis LGE images. Primary endpoint was all-cause mortality. The predictive performance of the radiomics features and semi-quantitative and quantitative LGE parameters were compared by ROC. The AUC was used to observe whether Rad-score had an incremental value for clinical stage. The Kaplan–Meier curve was used to further stratify the risk of CA patients. Results: During a median follow-up of 12.9 months, 30% (40/120) patients died. There was no significant difference in the predictive performance of the radiomics model in different LV sections in the validation set (AUCs of the global, basal, middle, and apical radiomics model were 0.75, 0.77, 0.76, and 0.77, respectively; all p > 0.05). The predictive performance of the Rad-score of the base-LV was better than that of the LGE total enhancement mass (AUC:0.77 vs. 0.54, p < 0.001) and LGE extent (AUC: 0.77 vs. 0.53, p = 0.004). Rad-score combined with Mayo stage had better predictive performance than Mayo stage alone (AUC: 0.86 vs. 0.81, p = 0.03). Rad-score (≥ 0.66) contributed to the risk stratification of all-cause mortality in CA. Conclusions: Compared to quantitative LGE parameters, radiomics can better predict all-cause mortality in CA, while the combination of radiomics and Mayo stage could provide higher predictive accuracy. Clinical relevance statement: Radiomics analysis provides incremental value and improved risk stratification for all-cause mortality in patients with cardiac amyloidosis. In this paper, we consider the following fractional Schrödinger equations with prescribed L 2 -norm constraint: (- Δ) s u = λ u + h (ε x) f (u) in R N , ∫ R N | u | 2 d x = a 2 , where 0 < s < 1 , N ≥ 3 , a , ε > 0 , h ∈ C (R N , R +) and f ∈ C (R , R) . In the mass subcritical case but under general assumptions on f, we prove the multiplicity of normalized solutions to this problem. Specifically, we show that the number of normalized solutions is at least the number of global maximum points of h when ε is small enough. Before that, without any restrictions on ε and the number of global maximum points, the existence of normalized ground states can be determined. In this sense, by studying the relationship between h 0 : = inf x ∈ R N h (x) and h ∞ : = lim | x | → ∞ h (x) , we establish new results on the existence of normalized ground states for nonautonomous elliptic equations. This paper is concerned with the existence and concentration of positive solutions for the following quasilinear Schrödinger equation QS - Δ u + λ V (x) u + κ 2 [ Δ (u 2) ] u = μ f (u) , x ∈ R N , u ∈ H 1 (R N) , where N ≥ 3 and κ , λ > 0 , μ ≥ 1 are parameters. f is a continuous function satisfying some local assumptions near the origin. Under some appropriate assumptions on the potential V, the existence of positive solutions is obtained for λ > 0 large enough by using a change of variables arguments and L ∞ -estimates. In this article, we study the mass spectrum of the low-lying triply heavy hybrid baryon, which consists of three valence heavy quarks in a color octet and one valence gluon, with spin-parity J P = (1 2) + via QCD sum rules. This is the first study on the triply heavy hybrid baryons in the framework of QCD sum rules. After performing the QCD sum rule analysis, we find that the mass of cccg hybrid baryon lies in M cccg = 5.91–6.13 GeV. As a byproduct, the mass of the triply bottom hybrid baryon state is extracted to be around M bbbg = 14.62–14.82 GeV. The contributions up to dimension eight at the leading order of α s (LO) in the operator product expansion are taken into account in the calculation. The triply charmed hybrid baryon predicted in this work can decay into one doubly charmed baryon and one charmed meson. Especially, we propose to search for cccg hybrid baryon with J P = (1 / 2) + in the P-wave decay channels Ξ cc + + D 0 , Ξ cc + D + , and Ξ ccs + D s + , which may be accessible in future BelleII, Super-B, PANDA, and LHCb experiments. Keto-enol tautomerism, describing an equilibrium involving two tautomers with distinctive structures, provides a promising platform for modulating nanoscale charge transport. However, such equilibria are generally dominated by the keto form, while a high isomerization barrier limits the transformation to the enol form, suggesting a considerable challenge to control the tautomerism. Here, we achieve single-molecule control of a keto-enol equilibrium at room temperature by using a strategy that combines redox control and electric field modulation. Based on the control of charge injection in the single-molecule junction, we could access charged potential energy surfaces with opposite thermodynamic driving forces, i.e., exhibiting a preference for the conducting enol form, while the isomerization barrier is also significantly reduced. Thus, we could selectively obtain desired and stable tautomers, which leads to significant modulation of the single-molecule conductance. This work highlights the concept of single-molecule control of chemical reactions on more than one potential energy surface. Keto-enol tautomerism offers a promising platform for modulating charge transport at the nanoscale. In this paper, we investigate the existence of a least energy sign-changing solution for the following Schrödinger-Kirchhoff equation SK - (a + b ∫ R 3 | ∇ u | 2 d x) Δ u + V (x) u = f (x , u) , x ∈ R 3 , u ∈ H 1 (R 3) where a , b > 0 are parameters, V ∈ C (R 3 , R +) and f ∈ C (R 3 × R , R) . The potential function V satisfies some suitable conditions and the nonlinearity f satisfies mild assumptions. By using sign-changing Nehari manifold, we prove that this problem possesses a ground state sign-changing solution with precisely two nodal domains, and its energy is strictly larger than twice that of ground state solutions. Objectives: To investigate the predictive value of CT-derived fractional flow reserve (FFRCT) in anastomosis occlusion after coronary artery bypass graft (CABG) surgery. Methods: Patients undergoing CABG with both pre- and post-operative coronary computed tomographic angiography (CCTA) were retrospectively included. Preoperative CCTA studies were used to evaluate anatomical and FFRCT information of target vessels. A diameter stenosis (DS) ≥ 70% or left main > 50% was considered to be anatomically severe, while FFRCT value ≤ 0.80 be functionally significant. The primary endpoint was anastomosis occlusion evaluated on post-operative CCTA during follow-up. Predictors of anastomosis occlusion were assessed by the multivariate binary logistic regression with generalized estimating equations. Results: A total of 270 anastomoses were identified in 88 enrolled patients. Forty-one anastomoses from 30 patients exhibited occlusion during a follow-up of 15.3 months after CABG. The occluded group had significantly increased prevalence of non-severe DS (58.5% vs. 40.2%; p = 0.023) and non-significant FFRCT (48.8% vs. 10.0%; p < 0.001). Multivariable analysis indicated FFRCT ≤ 0.80 (odds ratio [OR]: 0.10, 95% CI: 0.03–0.33; p < 0.001) and older age (OR: 0.92, 95% CI: 0.87–0.97; p = 0.001) were predictors for bypass patency during follow-up, while myocardial infarction history and anastomosis to a local lesion or bifurcation (all p value < 0.05) were predictors of occlusion. Adding FFRCT into the model based on the clinical and anatomical predictors had an improved AUC of 0.848 (p = 0.005). Conclusions: FFRCT ≤ 0.80 was associated with a significant risk reduction of anastomosis occlusion after CABG. Preoperative judgment of the hemodynamic significance may improve the CABG surgery strategy and reduce graft failure. Developing an eco-friendly, efficient, and highly selective gold-recovery technology is urgently needed in order to maintain sustainable environments and improve the utilization of resources. Here we report an additive-induced gold recovery paradigm based on precisely controlling the reciprocal transformation and instantaneous assembly of the second-sphere coordinated adducts formed between β-cyclodextrin and tetrabromoaurate anions. The additives initiate a rapid assembly process by co-occupying the binding cavity of β-cyclodextrin along with the tetrabromoaurate anions, leading to the formation of supramolecular polymers that precipitate from aqueous solutions as cocrystals. The efficiency of gold recovery reaches 99.8% when dibutyl carbitol is deployed as the additive. This cocrystallization is highly selective for square-planar tetrabromoaurate anions. In a laboratory-scale gold-recovery protocol, over 94% of gold in electronic waste was recovered at gold concentrations as low as 9.3 ppm. This simple protocol constitutes a promising paradigm for the sustainable recovery of gold, featuring reduced energy consumption, low cost inputs, and the avoidance of environmental pollution. Eco-friendly, efficient, and selective gold recovery technologies are urgently desired to satisfy the increasing demand for gold. In this paper, we study the Schrödinger–Bopp–Podolsky system - Δ u + V (x) u + ϕ u = μ f (u) + u 5 in R 3 , - Δ ϕ + a 2 Δ 2 ϕ = 4 π u 2 in R 3 , thereinto, we request that a , μ > 0 , the function V(x) and f(u) satisfies some specified conditions. By using constraint variational method and quantitative deformation lemma, we derive two results. If μ is large enough, the system has a least-energy sign-changing solution u μ . Van der Waals heterostructures composed of low-dimensional atomic layers host rich physics for new device applications, such as magic-angle twisted bilayer graphene and coaxial multi-walled hetero-nanotubes. Aside from exploring their abnormal physical behavior, fabrication of such structures also presents a great challenge to this area, owing to the subtle and sensitive interactions among neighboring layers. Here we show by molecular dynamics simulations that narrow blue phosphorene nanoribbons can be encapsulated into carbon nanotubes driven by van der Waals interactions and form one-dimensional heterostructures. It shows that by varying carbon nanotube diameters and nanoribbon width, the nanoribbons can either retain their original straight structures or twist into tubular structures. Wrapping phases are also observed for large-sized blue phosphorus. It is found that the underlying mechanism originates from the competition between van der Waals energy and bending energy induced by tube curvature. A phase diagram of the resultant 1D structure is thus obtained based on a simple analysis of energetics. Besides, V is a positive continuous potential satisfying 0 < V 0 ≤ V (x) ≤ lim inf | x | → ∞ V (x) = V ∞ . Under the conditions of f and V, it is not easy to obtain bounded Palais–Smale sequences, so there we use a cut-off technique to overcome. Moreover, in order to conquer the difficulties of the lack of compactness due to the unboundedness of the domain R 3 and V is a non-constant function, we use a new global compactness lemma to show the convergence of Palais–Smale sequences. In this paper, we study the existence of ground-state solution for the coupled nonlinear Schrödinger–Korteweg–de Vries system - Δ u + λ 1 u = u 3 + β u v in R N , - Δ v + λ 2 v = 1 2 v 2 + 1 2 β u 2 in R N , under the constrains ∫ R N | u | 2 d x = a , ∫ R N | v | 2 d x = b , where N = 1 , 2 , β > 0 and a , b > 0 are prescribed. In the system, the parameters λ 1 and λ 2 are unknown and will correspond to the Lagrange multipliers. Based on the variational method and rearrangement techniques, we prove that there exists a positive ground state solution for the system by the precompactness of the minimizing sequences, up to translation. We investigate the global well-posedness of strong solutions to the three-dimensional (3D) nonhomogeneous magnetohydrodynamic equations with density-dependent viscosity and vacuum. Combined energy method with the structure of the system under consideration, we obtain global existence and uniqueness of strong solutions provided that ‖ ρ 0 ‖ L 1 + ‖ b 0 ‖ L 2 is suitably small. In particular, the initial velocity can be arbitrarily large. Moreover, we also derive exponential decay rates of the solution. As a direct application, we show global strong solutions for the 3D nonhomogeneous Navier–Stokes equations with density-dependent viscosity as long as the initial mass is properly small. This work improves Liu’s result (Z Angew Math Phys 70:Paper No. 107, 2019), where the author requires the smallness condition on ‖ ρ 0 ‖ L ∞ + ‖ b 0 ‖ L 3 . While NK cell dysfunction has been observed in numerous malignancies, the underlying mechanisms in gastric cancer remain unclear.Flow cytometry was used to identify the phenotypic distribution and expression of activated receptors on NK cells. ELISA was used to determine the expression of cytokines. We examined the expression of NK cell-related genes and explored their association with survival and prognosis. Additionally, we conducted PCR detection of miR-552-5p expression levels in plasma exosomes of patients and investigated its correlation with phenotypic distribution and activated receptors. We used flow cytometry and ELISA to verify the role of miR-552-5p in NK cell dysfunction. Furthermore, we investigated the potential role of PD-1/PD-L1 in regulating NK cell dysfunction in patients’ cells.We observed a significant decrease in the percentage of NKG2D and NKp30 and IFN-γ and TNF-α in patients than in healthy volunteers. Patients with low levels of CD56, CD16, NKG2D, and NKP46 exhibited poorer survival prognoses. By using variational methods with a more general global compactness lemma, we obtain a positive least energy solution and a least energy sign-changing solution with exactly two nodal domains, and we also prove that the energy of least energy sign-changing solution is strictly larger than twice that of least energy solutions. Objectives: To propose a novel functional Coronary Artery Disease–Reporting and Data System (CAD-RADS) category system integrated with coronary CT angiography (CCTA)–derived fractional flow reserve (FFRCT) and to validate its effect on therapeutic decision and prognosis in patients with coronary artery disease (CAD). Methods: Firstly, we proposed a novel functional CAD-RADS and evaluated the performance of functional CAD-RADS for guiding treatment strategies with actual clinical treatment as a reference standard in a retrospective multicenter cohort with CCTA and invasive FFR performed in all patients (n = 466). Net reclassification improvement (NRI) of functional CAD-RADS over anatomical CAD-RADS was calculated. Secondly, the prognostic value of functional CAD-RADS in a prospective two-arm cohort (566 [FFRCT arm] vs. 567 [CCTA arm]) was calculated, after a 1-year follow-up, functional CAD-RADS in FFRCT arm (n = 513) and anatomical CAD-RADS in CCTA arm (n = 511) to determine patients at risk of adverse outcomes were compared with a Cox hazard proportional model. Results: Functional CAD-RADS demonstrated superior value over anatomical CAD-RADS (AUC: 0.828 vs. 0.681, p < 0.001) and comparable performance to FFR (AUC: 0.828 vs. 0.848, p = 0.253) in guiding therapeutic decisions. Functional CAD-RADS resulted in the revision of management plan as determined by anatomical CAD-RADS in 30.0% of patients (n = 140) (NRI = 0.369, p < 0.001). Functional CAD-RADS was an independent predictor for 1-year outcomes with indexes of concordance of 0.795 and the corresponding value was 0.751 in anatomical CAD-RADS. Conclusion: The novel functional CAD-RADS gained incremental value in guiding therapeutic decision-making compared with anatomical CAD-RADS and comparable power in 1-year prognosis with anatomical CAD-RADS in a real-world scenario. Objectives: To explore downstream management and outcomes of machine learning (ML)–based CT derived fractional flow reserve (FFRCT) strategy compared with an anatomical coronary computed tomography angiography (CCTA) alone assessment in participants with intermediate coronary artery stenosis. Methods: In this prospective study conducted from April 2018 to March 2019, participants were assigned to either the CCTA or FFRCT group. The primary endpoint was the rate of invasive coronary angiography (ICA) that demonstrated non-obstructive disease at 90 days. Secondary endpoints included coronary revascularization and major adverse cardiovascular events (MACE) at 1-year follow-up. Results: In total, 567 participants were allocated to the CCTA group and 566 to the FFRCT group. At 90 days, the rate of ICA without obstructive disease was higher in the CCTA group (33.3%, 39/117) than that (19.8%, 19/96) in the FFRCT group (risk difference [RD] = 13.5%, 95% confidence interval [CI]: 8.4%, 18.6%; p = 0.03). The ICA referral rate was higher in the CCTA group (27.5%, 156/567) than in the FFRCT group (20.3%, 115/566) (RD = 7.2%, 95% CI: 2.3%, 12.1%; p = 0.003). The revascularization-to-ICA ratio was lower in the CCTA group than that in the FFRCT group (RD = 19.8%, 95% CI: 14.1%, 25.5%, p = 0.002). MACE was more common in the CCTA group than that in the FFRCT group at 1 year (HR: 1.73; 95% CI: 1.01, 2.95; p = 0.04). Conclusion: In patients with intermediate stenosis, the FFRCT strategy appears to be associated with a lower rate of referral for ICA, ICA without obstructive disease, and 1-year MACE when compared to the anatomical CCTA alone strategy. Objectives: To examine the diagnostic accuracy of machine learning–based coronary CT angiography–derived fractional flow reserve (FFRCT) in diabetes mellitus (DM) patients. Methods: In total, 484 patients with suspected or known coronary artery disease from 11 Chinese medical centers were retrospectively analyzed. All patients underwent CCTA, FFRCT, and invasive FFR. The patients were further grouped into mild (25~49 %), moderate (50~69 %), and severe (≥ 70 %) according to CCTA stenosis degree and Agatston score < 400 and Agatston score ≥ 400 groups according to coronary artery calcium severity. Propensity score matching (PSM) was used to match DM (n = 112) and non-DM (n = 214) groups. Sensitivity, specificity, accuracy, and area under the curve (AUC) with 95 % confidence interval (CI) were calculated and compared. Results: Sensitivity, specificity, accuracy, and AUC of FFRCT were 0.79, 0.96, 0.87, and 0.91 in DM patients and 0.82, 0.93, 0.89, and 0.89 in non-DM patients without significant difference (all p > 0.05) on a per-patient level. The accuracies of FFRCT had no significant difference among different coronary stenosis subgroups and between two coronary calcium subgroups (all p > 0.05) in the DM and non-DM groups. After PSM grouping, the accuracies of FFRCT were 0.88 in the DM group and 0.87 in the non-DM group without a statistical difference (p > 0.05). Conclusions: DM has no negative impact on the diagnostic accuracy of machine learning–based FFRCT. Objective: To study the effect and mechanism of Schistosoma japonicum soluble egg antigen (SEA) on protecting against type 1 diabetes, 4-week-old female BALB/c and NOD mice were divided randomly into four groups: BALB/c control, BALB/c treated with SEA, NOD control, and NOD treated with SEA. Methods: Treated mice were injected intraperitoneally with 50 μg of SEA twice a week for 6 weeks, and control mice received the same volume of phosphate-buffered saline. Blood glucose in all mice was determined weekly from 8 weeks of age. Flow cytometry was used to detect the percentages of regulatory T cells of splenocytes in each group. Enzyme-linked immunosorbent assays were used to detect the levels of interferon-γ, interleukin (IL)-2, IL-4, and IL-5 in splenic cell culture supernatants. Results: Compared with those of the NOD group, the blood glucose level and percentage incidence of diabetes in NOD mice treated with SEA decreased significantly. This indicated that SEA treatment prevented spontaneous type 1 diabetes. After SEA administration, the frequency of splenic regulatory T cells increased significantly, and the secretion of IL-4 and IL-5 by splenic cells increased. Conclusions: These results demonstrated that SEA can prevent type 1 diabetes by enhancing regulatory T cells and the T helper 2 cell immune response in NOD mice. [ABSTRACT FROM AUTHOR]

Chemical cross-linking of proteins coupled with mass spectrometry is widely used in protein structural analysis. In this study we develop a class of non-hydrolyzable amine-selective di-ortho-phthalaldehyde (DOPA) cross-linkers, one of which is called DOPA2. Cross-linking of proteins with DOPA2 is 60–120 times faster than that with the N-hydroxysuccinimide ester cross-linker DSS. Compared with DSS cross-links, DOPA2 cross-links show better agreement with the crystal structures of tested proteins. More importantly, DOPA2 has unique advantages when working at low pH, low temperature, or in the presence of denaturants. Using staphylococcal nuclease, bovine serum albumin, and bovine pancreatic ribonuclease A, we demonstrate that DOPA2 cross-linking provides abundant spatial information about the conformations of progressively denatured forms of these proteins. Furthermore, DOPA2 cross-linking allows time-course analysis of protein conformational changes during denaturant-induced unfolding. Conformations sampled by a protein while it unfolds are difficult to visualize. Here, the authors develop di-ortho-phthalaldehyde cross-linkers for rapid chemical cross-linking mass spectrometry analysis and demonstrate that this method captures the conformations of protein unfolding intermediates. SOX2 is related to drug resistance in many types of cancer, including lung cancer. Herein, we investigated the role of SOX2 and its regulatory signaling in cisplatin-treated non-small-cell lung cancer (NSCLC). The effects of SOX2 on cell viability, proliferation, and apoptosis were evaluated in vitro. Western blotting, real-time quantitative PCR, immunohistochemistry, and luciferase reporter assays were used to investigate the underlying mechanism. Kaplan-Meier survival analysis and the log-rank test were used to assess the relationship between SOX2 expression and patient survival. A549/CDDP cells had marked resistance to cisplatin and stronger colony formation ability than A549 cells. The expression of SOX2 protein or mRNA in A549/CDDP was higher than that in A549. Knockdown of SOX2 in A549/CDDP-induced apoptosis by inhibiting colony formation and decreasing viability, but overexpression of SOX2 reversed these effects. Interestingly, Genomatix software predicted that the APE1 promoter has some SOX2 binding sites, while the SOX2 promoter has no APE1 binding sites. Furthermore, luciferase reporter assays proved that SOX2 could bind the promoter of APE1 in 293T cells. We further verified that SOX2 expression was not affected by shAPE1 in A549/CDDP. As expected, colony formation was obviously inhibited and apoptosis was strongly enhanced in A549/CDDP treated with SOX2 siSOX2 alone or combined with CDDP compared with control cells. Meaningfully, patients with low expression of SOX2, and even including its regulating APE1, survived longer than those with high expression of SOX2, and APE1. siSOX2 overcomes cisplatin resistance by regulating APE1 signaling, providing a new target for overcoming cisplatin resistance in NSCLC. Purpose: To explore the diagnostic value of American College of Radiology Contrast-Enhanced Ultrasound-Liver Imaging Reporting and Data System (ACR-CEUS-LI-RADS) for hepatocellular carcinoma (HCC) in patients with cirrhosis and chronic hepatitis B. Methods: A total of 205 patients at high risk of HCC with solitary hepatic nodule were enrolled and retrospectively analyzed. All patients were over 18 years old and had a single lesion with a diameter < 50 mm. Lesions were categorized according to size and contrast enhancement patterns in the arterial, portal venous and late phases. Diagnostic efficacy of CEUS LI-RADS for HCC, and the rate of non-HCC malignancies in the LR-M class were compared between patients with cirrhosis and chronic hepatitis B. Results: Of all 205 nodules (median nodule size was 34 mm), 142 (69.3%) were HCC. Of the 127 (61.9%) LR-5 category nodules, 95.8% (92/96) nodules were corresponded to HCC in cirrhosis, while 61.3% (19/31) nodules were corresponded to HCC in chronic hepatitis B (P = 0.000). Positive predictive value (PPV) of LR-5 category for HCC was 95.8% in cirrhosis and 61.3% in chronic hepatitis B (P = 0.000). More category of LR-4 nodules were proved to be HCC in patients with cirrhosis than chronic hepatitis B (80.0% vs 8.3%, P = 0.000). Of 41 LR-M category nodules, more non-HCC malignancies were found in chronic hepatitis B (76.0%) than that in cirrhosis (25.0%, P = 0.001). Conclusions: The LR-5 category is highly specific for the diagnosis of HCC in patients with cirrhosis. The instability of slope has already threatened life and property safety of the people, and improving the monitoring method of slope stability has important theoretical and practical significance for disaster prevention and reduction. According to the idea of "Newton force sudden drop and catastrophic occurrence" proposed by M.C. He in the landslide monitoring, a numerical model with constant resistance bolt has been established. Through numerical simulation research, it is found that the maximum principal stress, minimum principal stress and shear stress of the intersection point P of landslide surface and constant resistance bolt are sudden growth and sudden decrease, the vertical displacement and lateral displacement of this point P appear rise and fall before three kinds of stress. When loading to the next step of the step where three stress have reduced to a minimum value the slope is unstable and destroyed. At this time, the constant resistance bolt has undergone larger plastic deformation and damaged. Finally, comparing the stress curves and the acoustic emission (AE) curves, it can be seen that stress decreases from the maximum value and the AE curves begin to show a significant rise, the two curves display opposite law. It can be seen from the AE diagram that the failure mode of the slope is a combined tension and shear failure. This study aimed to explore the effect and mechanism underlying the role of the Schistosoma japonicum antigen of fatty acid-binding protein (SjFABP) on the growth of the schistosomula. SjFABP levels were evaluated by quantitative real-time polymerase chain reaction of samples of mice infected with S. japonicum; SjFABP was expressed and its levels gradually increased during all stages of S. japonicum schistosomula, including on 3, 10, 14, and 21 days of the growth process. Immunohistochemistry results demonstrated that SjFABP was distributed in the parenchyma, especially in the digestive tract of the S. japonicum schistosomula. RNA interference resulted in more than 60% knockdown of SjFABP leading to a reduction in length, volume, width, and area of the schistosomula as compared to control samples, as determined by light microscopy. Terminal deoxynucleotidyl transferase dUTP nick-end labeling detection further suggested that SjFABP knockdown resulted in increased apoptosis of schistosomes. Taken together, these results suggest that SjFABP may be related to the growth and survival of S. japonicum schistosomula, thereby representing a potential target for the treatment of schistosomiasis. Objectives: To investigate the feasibility and prognostic implications of coronary CT angiography (CCTA) derived fractional flow reserve (FFRCT) in patients who have undergone stents implantation. Methods: Firstly, the feasibility of FFRCT in stented vessels was validated. The diagnostic performance of FFRCT in identifying hemodynamically in-stent restenosis (ISR) in 33 patients with invasive FFR ≤ 0.88 as reference standard, intra-group correlation coefficient (ICC) between FFRCT and FFR was calculated. Secondly, prognostic value was assessed with 115 patients with serial CCTA scans after PCI. Stent characteristics (location, diameter, length, etc.), CCTA measurements (minimum lumen diameter [MLD], minimum lumen area [MLA], ISR), and FFRCT measurements (FFRCT, ΔFFRCT, ΔFFRCT/stent length) both at baseline and follow-up were recorded. Longitudinal analysis included changes of MLD, MLA, ISR, and FFRCT. The primary endpoint was major adverse cardiovascular events (MACE). Results: Per-patient accuracy of FFRCT was 0.85 in identifying hemodynamically ISR. FFRCT had a good correlation with FFR (ICC = 0.84). 15.7% (18/115) developed MACE during 25 months since follow-up CCTA. Lasso regression identified age and follow-up ΔFFRCT/length as candidate variables. In the Cox proportional hazards model, age (hazard ratio [HR], 1.102 [95% CI, 1.032–1.177]; p = 0.004) and follow-up ΔFFRCT/length (HR, 1.014 [95% CI, 1.006–1.023]; p = 0.001) were independently associated with MACE (c-index = 0.856). Time-dependent ROC analysis showed AUC was 0.787 (95% CI, 0.594–0.980) at 25 months to predict adverse outcome. After bootstrap validation with 1000 resamplings, the bias-corrected c-index was 0.846. Conclusions: Noninvasive ML-based FFRCT is feasible in patients following stents implantation and shows prognostic value in predicting adverse events after stents implantation in low-moderate risk patients.
