Obesity is a major risk factor underlying the development of metabolic disease and a growing public health concern globally. Strategies to promote skeletal muscle metabolism can be effective to limit the progression of metabolic disease. Here, we demonstrate that the levels of the Hippo pathway transcriptional co-activator YAP are decreased in muscle biopsies from obese, insulin-resistant humans and mice. Targeted disruption of Yap in adult skeletal muscle resulted in incomplete oxidation of fatty acids and lipotoxicity. Integrated 'omics analysis from isolated adult muscle nuclei revealed that Yap regulates a transcriptional profile associated with metabolic substrate utilisation. In line with these findings, increasing Yap abundance in the striated muscle of obese (db/db) mice enhanced energy expenditure and attenuated adiposity. Our results demonstrate a vital role for Yap as a mediator of skeletal muscle metabolism. Strategies to enhance Yap activity in skeletal muscle warrant consideration as part of comprehensive approaches to treat metabolic disease. Endoluminal surgery for the treatment of colorectal neoplasia is typically carried out using electrocautery tools which imply limited precision and the risk of harm through collateral thermal damage to the adjacent healthy tissue. As a potential alternative, we present the successful colonic epithelial laser ablation by means of picosecond laser pulses. Laser ablation studies performed in ex-vivo colon tissue result in cavities with comparable thickness to early stage colorectal cancers. The corresponding histology sections exhibit only minimal collateral damage to the surrounding tissue and the depth of the ablation can be controlled precisely by means of the pulse energy. High-speed imaging has been used for the first time to visualize picosecond laser ablation of cancerous tissue in a clinically relevant model. This information was correlated with histopathology and optical surface profilometry revealing the dynamic nature of the laser tissue interaction and the need for temporal or spatial separation of pulses for optimum efficacy with regards to tissue removal. Overall, the application of picosecond laser pulses to ablate endoluminal bowel lesions demonstrates significantly improved precision and reduced thermal damage to the adjacent tissue in comparison to conventional procedures and hence will enable more precise surgical treatment of cancers. Purpose: To date, very few studies have attempted to quantify the environmental impacts of a wave energy converter, and almost all of these focus solely on the potential climate change impacts and embodied energy. This paper presents a full life cycle assessment (LCA) of the first-generation Pelamis wave energy converter, aiming to contribute to the body of published studies and examine any potential trade-offs or co-benefits across a broad range of environmental impacts.Methods: The process-based attributional LCA was carried out on the full cradle-to-grave life cycle of the Pelamis P1 wave energy converter, including the device, its moorings and sub-sea connecting cable up to the point of connection with the grid. The case study was for a typical wave farm located off the north-west coast of Scotland. Foreground data was mostly sourced from the manufacturer. Background inventory data was mostly sourced from the ecoinvent database (v3.3), and the ReCiPe and CED impact assessment methods were applied.Results and discussion: The Pelamis was found to have significantly lower environmental impacts than conventional fossil generation in 6 impact categories, but performed worse than most other types of generation in 8 of the remaining 13 categories studied. The greatest impacts were from steel manufacture and sea vessel operations. The device performs quite well in the two most frequently assessed impacts for renewable energy converters: climate change and cumulative energy demand. The carbon payback period is estimated to be around 24 months (depending on the emissions intensity of the displaced generation mix), and the energy return on investment is 7.5. The contrast between this and the poor performance in other impact categories demonstrates the limitations of focussing only on carbon and energy.Conclusions: The Pelamis was found to generally have relatively high environmental impacts across many impact categories when compared to other types of power generation; however, these are mostly attributable to the current reliance on fossil fuels in the global economy and the early development stage of the technology. Opportunities to reduce this also lie in reducing requirements for steel in the device structure, and decreasing the requirements for sea vessel operations during installation, maintenance and decommissioning. The microstructure in the heat-affected zone (HAZ) of multipass welds typical of those used in power plants and made from 9 wt pct chromium martensitic Grade 92 steel is complex. Therefore, there is a need for systematic microstructural investigations to define the different regions of the microstructure across the HAZ of Grade 92 steel welds manufactured using the traditional arc welding processes in order to understand possible failure mechanisms after long-term service. In this study, the microstructure in the HAZ of an as-fabricated two-pass bead-on-plate weld on a parent metal of Grade 92 steel has been systematically investigated and compared to a complex, multipass thick section weldment using an extensive range of electron and ion-microscopy-based techniques. A dilatometer has been used to apply controlled thermal cycles to simulate the microstructures in distinctly different regions in a multipass HAZ using sequential thermal cycles. A wide range of microstructural properties in the simulated materials were characterized and compared with the experimental observations from the weld HAZ. It has been found that the microstructure in the HAZ can be categorized by a combination of sequential thermal cycles experienced by the different zones within the complex weld metal, using the terminology developed for these regions based on a simpler, single-pass bead-on-plate weld, categorized as complete transformation, partial transformation, and overtempered. It was found that the microstructure in the HAZ of a single-pass Grade 92 steel weld can be categorized as a function of a decreasing peak temperature reached as (1) the completely transformed (CT) region, in which the original matrix is completely reaustenitized with complete dissolution of the pre-existing secondary precipitate particles; (2) the partially transformed (PT) region, where the original matrix is partially reaustenitized along with a partial dissolution of the secondary precipitate particles from the original matrix; and (3) the overtempered (OT) region, where the pre-existing precipitate particles coarsen. Functionally graded, multi-layered coatings are designed to provide corrosion protection over a range of operating conditions typically found in industrial gas turbines. A model incorporating diffusion, equilibrium thermodynamics and oxidation has been developed to simulate the microstructural evolution within a multi-layered coating system. The phase and concentration profiles predicted by the model have been compared with an experimental multi-layered system containing an Al-rich outer layer, a Cr-enriched middle layer and an MCrAlY-type inner layer deposited on a superalloy substrate. The concentration distribution and many microstructural features observed experimentally can be predicted by the model. The model is expected to be useful for assessing the microstructural evolution of multilayer coated systems which can be potentially used on industrial gas turbine aerofoils. [ABSTRACT FROM AUTHOR]

The coefficient of thermal expansion (CTE) of nickel-based superalloys and bond coat layers was modelled by considering contributions from their constituent phases. The equilibrium phase composition of the examined materials was determined using thermodynamic equilibrium software with an appropriate database for Ni-based alloys, whereas the CTE and elastic properties of the principal phases were modelled using published data. The CTEs of individual phases were combined using a number of approaches to determine the CTE of the phase aggregate. As part of this work, the expansion coefficients of the superalloy IN-738LC and bond coat Amdry-995 were measured as a function of temperature and compared with the model predictions. The predicted values were also validated with the published data for the single-crystal superalloy CMSX-4 and a number of other Ni-based alloy compositions at 1000 K. A very good agreement between experiment and model output was found, especially up to 800 $$^\circ $$ C. Nest predation is a key source of selection for birds that has attracted increasing attention from ornithologists. The inclusion of new concepts applicable to nest predation that stem from social information, eavesdropping or physiology has expanded our knowledge considerably. Recent methodological advancements now allow focus on all three players within nest predation interactions: adults, offspring and predators. Indeed, the study of nest predation now forms a vital part of avian research in several fields, including animal behaviour, population ecology, evolution and conservation biology. However, within nest predation research there are important aspects that require further development, such as the comparison between ecological and evolutionary antipredator responses, and the role of anthropogenic change. We hope this review of recent findings and the presentation of new research avenues will encourage researchers to study this important and interesting selective pressure, and ultimately will help us to better understand the biology of birds. General cognitive function is substantially heritable across the human life course from adolescence to old age. We investigated the genetic contribution to variation in this important, health- and well-being-related trait in middle-aged and older adults. We conducted a meta-analysis of genome-wide association studies of 31 cohorts (N = 53 949) in which the participants had undertaken multiple, diverse cognitive tests. A general cognitive function phenotype was tested for, and created in each cohort by principal component analysis. We report 13 genome-wide significant single-nucleotide polymorphism (SNP) associations in three genomic regions, 6q16.1, 14q12 and 19q13.32 (best SNP and closest gene, respectively: rs10457441, P = 3.93× 10-9, MIR2113; rs l7522122, P = 2.55 × 10-8, AKAP6; rsl 0119, P=5.67 × 10-9, APOE/TOMM40).\Ne report one gene-based significant association with the HMGN1 gene located on chromosome 21 (P= 1 × 10-6). These genes have previously been associated with neuropsychiatric phenotypes. Meta-analysis results are consistent with a polygenic model of inheritance. To estimate SNP-based heritability, the genome-wide complex trait analysis procedure was applied to two large cohorts, the Atherosclerosis Risk in Communities Study (N= 6617) and the Health and Retirement Study (N= 5976). The proportion of phenotypic variation accounted for by all genotyped common SNPs was 29% (s.e. = 5%) and 28% (s.e. = 7%), respectively. Using polygenic prediction analysis, -1.2% of the variance in general cognitive function was predicted in the Generation Scotland cohort (N = 5487; P= 1.5 × 10 17). In hypothesis-driven tests, there was significant association between general cognitive function and four genes previously associated with Alzheimer's disease: TOMM40, APOE, ABCG1 and MEF2C. This paper presents the results of a study into a novel application of the “stacked-shell” laminate modelling approach to dynamically loaded bolted composite joints using the explicit finite element code PAM-CRASH. The stacked-shell approach provides medium-high fidelity resolution of the key joint failure modes, but is computationally much more efficient than full 3D modelling. For this work, a countersunk bolt in a composite laminate under in-plane bearing loading was considered. The models were able to predict the onset of damage, failure modes and the ultimate load of the joint. It was determined that improved debris models are required in order to accurately capture the progressive bearing damage after the onset of joint failure. [ABSTRACT FROM AUTHOR]

The nickel based alloys IN617 and IN625 that have been selected for their candidacy in the construction of the hottest regions of the supercritical steam cycle have been oxidised under isothermal conditions at 750 °C and atmospheric pressure in atmospheres of 100 % steam, 50/50 % steam/argon and air for up to 4,200 h. Both alloys developed a thin protective oxide under each condition. Scale thickness measurements using SEM micrographs were performed and showed that exposures in steam exhibited a higher rate of scale formation than exposures in air in both alloys. IN617 developed an extensive internal network of alumina which resulted in the formation of alloy protrusions into the scale altering scale growth kinetics, IN625 also formed alumina to a lesser extent. Voids formed in the matrix below the scale in both alloys in each environment. The extent of alumina formation alters the void morphology which eventually impacts the scale growth rate as inward scale growth occurred into the voids in IN625 but not in IN617. Empirical evidence shows that educational experiences in the context of the outdoors lead to elevated self-esteem. Although elevated self-esteem is widely assumed to promote beneficial outcomes, recent evidence suggests that elevated self-esteem may also facilitate a variety of negative outcomes (i.e., increased prejudice, aggression, drug and alcohol abuse). The current research was conducted in order to examine whether one type of outdoor educational experience-a 10-day developmental voyage-could elevate adolescents' self-esteem without also elevating negative outcomes. The data from three separate studies are reported. Study one revealed that adolescents who undertook the voyage manifested elevated self-esteem and decreased gender prejudice (i.e., less negative ratings of opposite sex outgroup members). Study two replicated these findings and further demonstrated that these effects were maintained 4-5 months following the voyage. In study three, in addition to self-esteem, risky attitudes (i.e., towards drug and alcohol use), physical aggression, verbal aggression, racial and gender bias were also examined. Adolescents again showed elevated self-esteem that was maintained 4-5 months following the voyage. There were no changes in risky attitudes, aggression, or racial and gender bias. Taken together these results indicate that taking part in a 10-day developmental voyage-can lead to an elevation in self-esteem, that is maintained over time and which does not facilitate a variety of negative outcomes. The principles of shared decision making are well documented but there is a lack of guidance about how to accomplish the approach in routine clinical practice. Our aim here is to translate existing conceptual descriptions into a three-step model that is practical, easy to remember, and can act as a guide to skill development. Achieving shared decision making depends on building a good relationship in the clinical encounter so that information is shared and patients are supported to deliberate and express their preferences and views during the decision making process. To accomplish these tasks, we propose a model of how to do shared decision making that is based on choice, option and decision talk. The model has three steps: a) introducing choice, b) describing options, often by integrating the use of patient decision support, and c) helping patients explore preferences and make decisions. This model rests on supporting a process of deliberation, and on understanding that decisions should be influenced by exploring and respecting "what matters most" to patients as individuals, and that this exploration in turn depends on them developing informed preferences. A multicomponent, one-dimensional diffusion model that was developed for simulating microstructure evolution in coated gas turbine blade systems has been used to compare the phase structures of three MCrAlY coated superalloy systems. The model is based on finite differences and incorporates oxidation and equilibrium thermodynamic computations. The superalloy substrates considered were the nickel-based superalloy CMSX-4, a high-Cr single-crystal superalloy, and a cobalt-based MAR-M509, and these were all coated with an MCrAlY bond coat of similar composition. The results predicted by the model have been compared with similar experimental systems. The model can predict many features observed experimentally and therefore can be expected to be a useful tool in lifetime prediction and microstructural assessment of turbine blade systems based on superalloys. Introduction: The association between hormonal contraceptive use and bone mineral density remains controversial. Hypothesis: Hormonal contraceptive use is positively associated with bone mass in young premenopausal women. Methods: Cross-sectional analysis of data collected from women aged 26-36 years ( n = 687) in the Childhood Determinants of Adult Health study-a longitudinal study investigating childhood determinants of cardiovascular disease, diabetes, and other chronic diseases in adulthood. Participants were not currently pregnant or breast-feeding. Contraceptive use was obtained by self-administered questionnaire. Women were categorized as combined oral contraceptive users ( n = 219), progestogen-only contraceptive users ( n = 43), and non-users of hormonal contraceptives ( n = 425). Bone mass was measured by quantitative ultrasound. Results: Compared with women who were not using any hormonal contraceptives, women using combined oral contraceptives had significantly higher values of broadband ultrasound attenuation (BUA), speed of sound, and quantitative ultrasound index. These associations remained after adjustment for confounders. Progestogen-only contraceptive users had higher BUA than non-users, but the differences were not statistically significant in this small group. Conclusion: Combined oral contraceptive use was associated with higher bone mass measured by quantitative ultrasound in this population-based sample of premenopausal women aged 26-36 while progestogen-only contraceptives appeared to have no deleterious effect on bone mass. Major earthquakes occurred in the region of the Central Kuril Islands on November 15, 2006 ( M w = 8.3) and January 13, 2007 ( M w = 8.1). These earthquakes generated strong tsunamis recorded throughout the entire Pacific Ocean. The first was the strongest trans-Pacific tsunami of the past 42 years (since the Alaska tsunami in 1964). The high probability of a strong earthquake ( M w ≥ 8.5) and associated destructive tsunami occurring in this region was predicted earlier. The most probable earthquake source region was investigated and possible scenarios for the tsunami generation were modeled. Investigations of the events that occurred on November 15, 2006, and January 13, 2007, enabled us to estimate the validity of the forecast and compare the parameters of the forecasted and observed earthquakes and tsunamis. In this paper, we discuss the concept of “seismic gaps,” which formed the basis for the forecast of these events, and put forward further assumptions about the expected seismic activity in the region. We investigate the efficiency of the tsunami warning services and estimate the statistical parameters for the observed tsunami waves that struck the Far Eastern coast of Russia and Northern Japan. The propagation and transformation of the 2006 and 2007 tsunamis are studied using numerical hydrodynamic modeling. The spatial characteristics of the two events are compared. [ABSTRACT FROM AUTHOR]

The paired-pulse transcranial magnetic stimulation (TMS) paradigm is increasingly employed to examine intracortical inhibitory processes in different motor tasks. Short-interval intracortical inhibition (SICI) has been shown to vary with the size of the MEP elicited by the test TMS pulse. This suggests that factors that alter MEP size, such as changes in cortical excitability, may confound the interpretation of SICI. However, the effect of excitability on SICI has not been systematically investigated. The present study examined SICI in 11 volunteers. The effect of test TMS intensities ranging from 90 to 150% resting motor threshold (RMT) on SICI was examined in three excitability states in the right first dorsal interosseous muscle: rest, isometric abduction of the left index finger (Contra) and isometric abduction of the right index finger (Active). For all excitability states SICI was not observed when test TMS intensity was less than 110% resting motor threshold. This was true even for the Active condition in which 90 and 100% test TMS intensities elicited large and consistent MEPs. For all conditions moderately suprathreshold test TMS intensities (110–120% RMT) yielded the greatest measure of SICI; increasing test TMS intensities resulted in a progressive reduction in the estimate of SICI. These results suggest that estimates of SICI are systematically affected by the intensity of the test TMS pulse, regardless of excitability state. Analysing the collapse of skin-stiffened structures requires capturing the critical phenomenon of skin-stiffener separation, which can be considered analogous to interlaminar cracking. This paper presents the development of a numerical approach for simulating the propagation of interlaminar cracks in composite structures. A degradation methodology was introduced in MSC.Marc, which involved the modelling of a structure with shell layers connected by user-defined multiple-point constraints (MPCs). User subroutines were written that employ the virtual crack closure technique (VCCT) to determine the onset of crack growth and modify the properties of the user-defined MPCs to simulate crack propagation. Methodologies for the release of failing MPCs are presented and are discussed with reference to the VCCT assumption of self-similar crack growth. The numerical results obtained by using the release methodologies are then compared with experimental data for a double-cantilever beam specimen. Based on this comparison, recommendations for the future development of the degradation model are made, especially with reference to developing an approach for the collapse analysis of fuselage-representative structures. Widespread health service goals to improve consistency and safety in patient care have prompted considerable investment in the development of evidence-based clinical guidelines. Computerised decision support (CDS) systems have been proposed as a means to implement guidelines in practice. This paper discusses the general concept in oncology and presents an evaluation of a CDS system to support triple assessment (TA) in breast cancer care. Balanced-block crossover experiment and questionnaire study. One stop clinic for symptomatic breast patients. Twenty-four practising breast clinicians from United Kingdom National Health Service hospitals. A web-based CDS system. Clinicians made significantly more deviations from guideline recommendations without decision support (60 out of 120 errors without CDS; 16 out of 120 errors with CDS, P < 0.001). Ignoring minor deviations, 16 potentially critical errors arose in the no-decision-support arm of the trial compared with just one (P = 0.001) when decision support was available. Opinions of participating clinicians towards the CDS tool became more positive after they had used it (P < 0.025). The use of decision support capabilities in TA may yield significant measurable benefits for quality and safety of patient care. Recently revised UK and US hypertension guidelines have reduced thresholds for both diagnosis and treatment and differ in their recommendations. We have used data from a random, stratified community-based sample of 4784 people aged 65 years and over to compare the prevalence of treatable hypertension and the potential impact on patients and primary care from using current guidelines. BHS, NICE and JNC7 guidelines were applied to blood pressures obtained from primary care medical records (94%) or measured at a screening clinic (6%). Risk factors were obtained by questionnaire and from medical records, supplemented by epidemiological data. Workload was estimated for a representative practice population of 10 000 patients. Blood pressures were obtained on 4514 patients (94%). Prevalence of treatable hypertension was over 67%. Compared to BHS4, prevalence estimates using NICE guidelines were comparable for men but significantly lower for women (P<0.05). They were significantly higher using JNC7 compared with BHS4 and NICE guidance (P<0.05). A general practice of 10 000 patients could expect 1287 older hypertensive patients using BHS4 guidelines and 1231 patients using NICE guidelines. Under BHS4, an extra 94 patients will require annual, rather than 5-yearly review compared with that using the previous guideline. In conclusion, implementation of BHS4 guidelines, with their revised thresholds for diagnosis, will not add materially to the prevalence of treatable hypertension compared to previous BHS3 guidelines but will have a major impact on practice workload. The severity of disease caused by infection with Leishmania major depends critically on the genetics of the host. Early induction of T helper (Th)1-type immune responses in the resistant C57BL/6 mice and Th2-type responses in the susceptible BALB/c mice are thought to determine cure or disease, respectively. We have previously mapped three host response loci in a genetic cross between C57BL/6 and BALB/c mice, and here we show definitively the involvement of these loci in disease severity using animals congenic for each of the loci. Surprisingly, in the late stage of infection when the difference in disease severity between congenic and parental mice was most pronounced, their cytokine profile correlated with the genetic background of the mice and not with the severity of disease. We have previously reported pilot data regarding the safety of saving partially used syringes of a glutaraldehyde cross-linked collagen for use in subsequent treatment sessions with the same individual. That single institution study involved 56 partially used syringes cultured for aerobic bacteria. Only one weakly positive culture was detected among these 56 samples, which prompted us to carry out this expanded study involving multiple centers and different injection techniques. Samples were collected from four centers. Following periurethral injection in an office setting, 166 partially used syringes of glutaraldehyde cross-linked collagen were refrigerated for between 1 and 104 weeks (average 58). Material from all 166 syringes was then cultured qualitatively and quantitatively for both aerobic and anaerobic organisms. Collagen from one syringe grew >100 000 colonies of Escherichia coli. All other cultures were negative. In the pilot study, one culture of 56 syringes was weakly positive for coagulase-negative staphylococcus. When the results from both studies were considered together, only two of 222 partially used syringes (0.9%) were contaminated. The background risk of local infection associated with periurethral collagen injection is approximately 0.29%. Using the statistical equation ‘number needed to harm’, we found that a clinician would have to reuse 111 syringes at a saving of $34,965 before he or she would cause a single local injection by so doing. Therefore, we feel that it may be cose-effective and safe to reinject material from a partially used syringe of glutaraldehyde cross-linked collagen during a subsequent treatment session on an individual. Dendritic segregation in conventionally cast, commercially heat treated and exposed MarM002 turbine blades has been investigated using scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS). During casting Cr, Co and W segregate to the dendrites and Hf, Ti and Ta segregate to the eutectics. As a result of the dissolution of γ′ precipitates in the dendrites and coarsening of γ′ precipitates in the eutectics this segregation aggravates during exposure at 1000°C. At this high exposure temperature homogenisation of dendritic segregation does not occur because it involves long diffusion distances and a low driving force compared with dissolution and coarsening of γ′ precipitates. A method has been developed to predict the effect of delaminations in a postbuckling stiffened structure manufactured from laminated composite materials. The emphasis of the technique, driven by aircraft certification requirements, was towards establishing whether delamination growth would initiate under given loading conditions. A geometric nonlinear finite element analysis was used to calculate the strain energy release rate around the circumference of a circular delamination using the virtual crack closure technique. In order to deal with the complex structural response in a computationally efficient manner, the structure was modelled using plate elements with two layers of plate elements used in the delaminated region. The effect of delamination size on the strength of postbuckling panels was shown to be a complex phenomenon in which trends were difficult to predict. Large delaminations could significantly affect the global and sub-laminate buckling modes and therefore be less critical than smaller delaminations. It was concluded that the method could accurately predict the load and location at which delamination growth would initiate, given suitable critical strain energy release rate data. A process for the treatment of animal wastes and the production of protein is under development at the University of Queensland. Animal wastes are fermented to produce organic acids which are then utilised by a pellicle-forming yeast, Candida ingens. This is harvested by skimming. The process removes carbon and minerals, particularly calcium and phosphorus, from the medium and Salmonella and Escherichia coli in the wastes are killed. The dry matter yield of C. ingens and its utilisation of volatile fatty acids (VFA) and oxygen were studied in order to improve the understanding of the process. When C. ingens was grown on a synthetic medium containing seven VFA and salts there was a significant increase in yield when the medium was reduced with sodium sulphide or cysteine hydrochloride. The VFA were utilised continuously over 24 h. After inoculation there was a shorter lag phase for the straight chain acids than for propionic and the branched chain acids, isobutyric, and isovaleric, but all acids showed a similar rate of utilisation of carbon during the period of most rapid utilisation. Candida ingens is an aerobe which, when growing as a pellicle, requires a reduced medium analogous to that used by anaerobes. It was concluded that pellicle growth on a medium containing several VFA takes place by utilising oxygen from the gaseous phase above, while all other nutrients are obtained from the reduced liquid phase below. The chemistry and other characteristics of carbide precipitates in 12CrlMoV steel of the type used in the power generation industry were studied using energy dispersive X-ray spectroscopy and electron and X-ray diffraction techniques, and the results have been compared against thermodynamic calculations. As a result of the much larger substitutional solute concentrations present in the alloy, unlike the carbides that occur in steels containing smaller concentrations of chromium and molybdenum, it is found that the equilibrium MC carbide precipitates very rapidly during heat treatments of the kind used routinely for stress-relief purposes. 5,8-Dihydroxyflavone (primetin) has been shown to be the sensitizer in Primula mistassinica Michaux and probably the source of allergic contact dermatitis in four milkers. Its sensitizing properties as determined in guinea-pigs, are strong. As far as is known this is the first experimental demonstration of the sensitizing potency of a flavone. Presumably the flavone, with its uncommon 5,8-arrangement of hydroxy groups, is oxidized in the skin to the corresponding quinone (primetinquinone). Quinone was prepared from primetin and used for experimental sensitization of guinea-pigs. It also revealed strong sensitizing properties. Cross-reactions were obtained not only with the synthetic quinone in the flavone-sensitive animals but also with primetin in primetinquinone-sensitive guineapigs. Preliminary sensitization tests with other flavones have demonstrated that the whole group of flavonoid components should be taken into consideration as potential sensitizers. [ABSTRACT FROM AUTHOR]

Laboratory techniques are described for evaluating the lethal and behavioral impact of pesticides on a range of stream macroinvertebrates under continuous through-flow and simulated stream conditions. The same basic test unit has been used, with slight modifications, to study the reactions of both Simulium larvae and non-target stream invertebrates. On the basis of a standard 1-hr exposure period to different concentrations followed by a 24-hr holding period in a continuous flow of clean water, different test organisms showed wide and consistent differences in tolerance to each of the two insecticides tested. The widest difference between two organisms occurred in the case of the Amphipod, Gammarus pulex (LC90-95, >> 1 ppm) which was found to be about 5000 x more tolerant to temephos than are nymphs of the mayfly, Baetis rhodani. (LC 90-95, 0.001- 0.002 ppm) The widest difference in the reactions of any one species is shown on the part of Gammarus which is about 100 times more susceptible to chlorpyrifos (LC 90-95, 0.05-0.1 ppm) than to temephos. The susceptibility levels of other indicator species such as Agrion, Hydropsyche, Brachycentras, Ephemera, etc. are discussed in relation to susceptibility levels of Simulium larvae under the same test conditions, and also in relation to current field dosages of the two insecticides in practical and experimental Simulium control. The behavioral reactions of a range of stream macroinvertebrates to short exposures to the organophosphorus insecticides temephos and chlorpyrifos methyl in running water have been studied in a laboratory experimental channel or simulated stream. The tests showed widely different types of response not only in accordance with susceptibility levels previously established, but also with regard to activation responses leading to detachment and downstream drift. The amphipod Gammarus pulex was the most readily activated by both insecticides, even at sub-lethal levels. Nymphs of the mayfly Baetis rhodani were also readily activated, but to a less extent than would be expected in view of their established high susceptibility to both compounds. In the case of Simulium larvae, detachment leading to drift during the 30-min exposure period was negligible with late instars exposed to each of the two insecticides, even at lethal levels. Both types of caddis larvae tested, the web-spinning Hydropsyche pellucidula and the mobile case-bearing Brachycentrus subnubilis, showed little sign of activation to either insecticide, even at concentrations which subsequently produced high mortalities. Observations extended for up to 24 hr after the initial 30-min exposure, showed continued complete absence of activation and detachment, the majority of exposed larvae dying in situ. These differences in reaction are discussed in relation to the interpretation of downstream drift following field application of these chemicals in practical and experimental Simulium control. Brasses containing 20 and 30 pct Zn and up to 1.15 pct Be have been examined to provide tensile test data on the age-hardening response of these ternary alloys. It was found that yield strengths of 1100 Mpa were attainable with cold working prior to aging at 300°C. The mode of precipitation, as determined by electron micros copy and electron diffraction, agrees substantially with recent work on the binary Cu-Be system. Precipitation commences as GP zones on the (001) planes. The observed precipitate phase, after aging times of up to seven days at 400°C, is CuBe with an orientation relationship of the type (301) ∥(113) [010] ∥[110]. [ABSTRACT FROM AUTHOR]

The cellular mechanisms responsible for rectal acidification in the desert locust, Schistocerca gregaria, were investigated in isolated recta mounted as flat sheets in modified Ussing chambers. Previous studies conducted in the nominal absence of exogenous CO2 and HCO3- suggested that the acidification was due to a proton-secretory rather than bicarbonate-reabsorptive mechanism (Thomson, R.B., Speight, J.D., Phillips, J.E. 1988. J. Insect Physiol. 34:829-837). This conclusion was confirmed in the present study by demonstrating that metabolic CO2 could not contribute sufficient HCO3- to the lumen to account for the rates of rectal acidification observed under the nominally CO2/HCO3(-)-free conditions used in these investigations. Rates of luminal acidification (JH+) were completely unaffected by changes in contraluminal pH, but could be progressively reduced (and eventually abolished) by imposition of either transepithelial pH gradients (lumen acid) or transepithelial electrical gradients (lumen positive). Under short-circuit current conditions, the bulk of JH+ was not dependent on Na+, K+, Cl-, Mg2+, or Ca2+ and was due to a primary electrogenic proton translocating mechanism located on the apical membrane. A small component (10-16%) of JH+ measured under these conditions could be attributed to an apical amiloride-inhibitable Na+/H+ exchange mechanism. [ABSTRACT FROM AUTHOR]

Three possible methods for determining, separately, thermal and load stresses in a structure from the results of strain measurements are considered. It is shown theoretically that only one of these is feasible. An experimental verification of the proposed method, for temperatures up to 350° F, was obtained by means of thermal and load tests of a flat plate, and through analysis of data obtained by other investigators in experiments on a box beam.
