Hydrogen peroxide is considered deleterious molecule that cause cellular damage integrity and function. Its key redox signaling molecule in oxidative stress and exerts toxicity on a wide range of organisms. Thus, to understand whether oxidative stress alters visual development, zebrafish embryos were exposed to H2O2 at concentration of 0.02 to 62.5 mM for 7 days. Eye to body length ratio (EBR) and apoptosis in retina at 48 hpf, and optomotor response (OMR) at 7 dpf were all measured. To investigate whether hydrogen peroxide-induced effects were mediated by oxidative stress, embryos were co-incubated with the antioxidant, glutathione (GSH) at 50 μM. Results revealed that concentrations of H2O2 at or above 0.1 mM induced developmental toxicity, leading to increased mortality and hatching delay. Furthermore, exposure to 0.1 mM H2O2 decreased EBR at 48 hpf and impaired OMR visual behavior at 7 dpf. Additionally, exposure increased the area of apoptotic cells in the retina at 48 hpf. The addition of GSH reversed the effects of H2O2, suggesting the involvement of oxidative stress. H2O2 decreased the expression of eye development-related genes, pax6α and pax6β. The expression of apoptosis-related genes, tp53, casp3 and bax, significantly increased, while bcl2α expression decreased. Antioxidant-related genes sod1, cat and gpx1a showed decreased expression. Expression levels of estrogen receptors (ERs) (esr1, esr2α, and esr2β) and ovarian and brain aromatase genes (cyp19a1a and cyp19a1b, respectively) were also significantly reduced. Interestingly, co-incubation of GSH effectivity reversed the impact of H2O2 on most parameters. Overall, these results demonstrate that H2O2 induces adverse effects on visual development via oxidative stress, which leads to alter apoptosis, diminished antioxidant defenses and reduced estrogen production. Infant studies have suggested that the detection of biological motion (BM) might be an innate capacity, based on newborns' spontaneous preference for BM. However, it is unclear if, like adults, infants recognize humans from BM and are able to build the representation of bodies and faces. To address this issue, we tested whether exposure to BM influences subsequent face recognition in 3- to 8-month-old infants. After familiarization with a point-light walker (PLW) of either a female or a male, the infant's preference for female and male faces was measured. If infants can build the representation of not only the body but also the face from PLWs, the familiarization effect of gender induced by the PLW might be generalized to faces. We found that infants at 7 to 8 months looked for longer at the face whose gender was opposite to that of the PLW, whereas 3- to 4- and 5- to 6-month-old infants did not. These results suggest that infants can access the representation of humans from BM and extract gender, which is shared across bodies and faces, from at least 7 to 8 months of age. Purpose: To assess the effectiveness of combined use of stereoscopic 3D video movies and part-time patching in treating older amblyopic children with poor response or compliance to traditional patching treatments and comparing this combined treatment with patching alone. Methods: Thirty-two children aged 5–12 years with amblyopia associated with anisometropia, strabismus, or both were recruited in a randomized clinical trial. Eligible participants were assigned randomly to the combined and patching groups. Here, binocular treatment refers to using the Bangerter filter to blur the fellow eye and subsequently watching a close-up 3D movie with large parallax. The primary outcome was amblyopic eye (AE) best-corrected visual acuity (BCVA) improvement at six weeks. In addition, secondary outcomes included BCVA of AE improvement at three weeks and change of stereoacuity. Results: Of 32 participants, mean (SD) age was 6.63 (1.46) years, and 19 (59%) were female. At 6 weeks, mean (SD) amblyopic eye VA improved by 0.17 ± 0.08 logMAR (2-sided 95% CI, 0.13 to 0.22; F = 57.2, p < 0.01) and 0.05 ± 0.04 logMAR (2-sided 95% CI, 0.05 to 0.09; F = 8.73, p = 0.01) in the combined and patching groups, respectively. The difference was statistically significant (mean difference, 0.13 logMAR [1.3 line]; 95% CI, 0.08–0.17 logMAR [0.8–1.7 lines]; t25 = 5.65, p < 0.01). After treatment, only the combined group had significantly improved stereoacuity, such as binocular function score (median [interquartile range], 2.30 [2.23 to 2.68] vs. 1.69 [1.60 to 2.30] log arcsec; paired, z = –3.53, p < 0.01), and mean stereoacuity gain was 0.47 log arcsec (± 0.22). Changes in other types of stereoacuity were similar. Conclusion: Our laboratory-based binocular treatment strategy engaged a high level of compliance that led to a substantial gain in visual function after a short period of treatment for older amblyopic children having poor response or compliance to traditional patching treatments. There is a need for a straightforward, accessible and accurate pediatric test for color vision deficiency (CVD). We present and evaluate ColourSpot, a self-administered, gamified and color calibrated tablet-based app, which diagnoses CVD from age 4. Children tap colored targets with saturations that are altered adaptively along the three dichromatic confusion lines. Two cohorts (Total, N = 772; Discovery, N = 236; Validation, N = 536) of 4–7-year-old boys were screened using the Ishihara test for Unlettered Persons and the Neitz Test of Color Vision. ColourSpot was evaluated by testing any child who made an error on the Ishihara Unlettered test alongside a randomly selected control group who made no errors. Psychometric functions were fit to the data and "threshold ratios" were calculated as the ratio of tritan to protan or deutan thresholds. Based on the threshold ratios derived using an optimal fitting procedure that best categorized children in the discovery cohort, ColourSpot showed a sensitivity of 1.00 and a specificity of 0.97 for classifying CVD against the Ishihara Unlettered in the independent validation cohort. ColourSpot was also able to categorize individuals with ambiguous results on the Ishihara Unlettered. Compared to the Ishihara Unlettered, the Neitz Test generated an unacceptably high level of false positives. ColourSpot is an accurate test for CVD, which could be used by anyone to diagnose CVD in children from the start of their education. Key messages: Visual acuity is the most used method to assess visual function in children. Contrast sensitivity complements the information provided for visual acuity, but it is not commonly used in clinical practice. Digital devices are increasingly used as a method to evaluate visual function, due to multiple advantages. Testing with these devices can improve the evaluation of visual development in children from a few months of age. Visual acuity and contrast sensitivity tests, using eye tracking technology, are able to measure visual function in children across a wide range of ages, objectively, quickly and without need of an experienced examiner. Purpose: To report age-normative values for grating visual acuity and contrast sensitivity in healthy children using a digital device with eye tracking technology and to validate the grating acuity test. Methods: In the first project of the study, we examined healthy children aged between 6 months and 7 years with normal ophthalmological assessment. Grating visual acuity (VA) and contrast sensitivity (CS) were assessed using a preferential gaze paradigm with a DIVE (Device for an Integral Visual Examination) assisted with eye tracking technology to provide age norms. For the validation project, we compared LEA grating test (LGT) with DIVE VA in a group of children aged between 6 months and 4 years with normal and abnormal visual development. Results: Fifty-seven children (2.86 ± 1.55 years) were examined with DIVE VA test and 44 successfully completed DIVE CS test (3.06 ± 1.41 years). Both, VA and CS values increased with age, mainly along the first two years of life. Sixty-nine patients (1.34 ± 0.61 years) were included in the DIVE VA test validation. The mean difference between LGT and DIVE VA was − 1.05 ± 4.54 cpd with 95% limits of agreement (LoA) of − 9.95–7.84 cpd. Agreement between the two tests was higher in children younger than 1 year with a mean difference of − 0.19 ± 4.02 cpd. Conclusions: DIVE is an automatic, objective and reliable tool to assess several visual function parameters in children, and it has good agreement with classical VA tests, especially for the first stage of life. Background: The normal values of optic nerve diameter and optic nerve sheath diameter might be beneficial in defining an abnormality such as optic nerve hypoplasia, or enlarged subarachnoid space, reflecting the state of increased intracranial pressure. Objective: To study the normal optic nerve diameter and optic nerve sheath diameter on magnetic resonance imaging (MRI) in the early years of postnatal visual development from MRI of the brain. Materials and methods: MRI of the brain in patients ages 4 years and younger were gathered. Forty-two studies with normal intracranial findings and a lack of history of increased intracranial pressure were retrospectively reviewed by two reviewers using axial T2-weighted images. Measurements were performed in transverse diameter perpendicular to the optic nerve at 3 mm behind the globe. Results: The mean optic nerve diameter of the 77 optic nerves were 2.5 mm (95% confidence interval [CI] 2.4–2.6). The mean optic nerve sheath diameter values of the 79 optic nerve sheath complexes were 5.0 mm (95% CI 4.9–5.1). The mean±standard deviation optic nerve diameter and optic nerve sheath diameter stratified by each age groups were, respectively, 0 to <1 year: 2.3±0.40 and 4.81±0.37; 1 to <2 years: 2.6±0.2 and 5.0±0.4; 2 to <3 years: 2.4±0.3 and 4.9±0.6, and 3 to <4 years: 2.9±0.4 and 5.2±0.60 mm. Conclusion: Seventy-four of the 77 measurements (96%) were of the measurements were above the threshold of 2 mm for optic nerve diameter. Introduction: Amblyopia therapy appears to be most effective in children under the age of 7 years, but results from randomized control trials (RCTs) have shown that occlusion therapy and/or atropine penalization therapy may improve visual acuity in an older age group. Which of these two therapies is the most effective with fewer adverse effects in an older age group has not yet been agreed upon.Methods: We systematically searched the literature for RCTs that compared atropine penalization therapy and occlusion therapy in terms of their visual acuity outcomes and adverse events and performed a meta-analysis on the visual acuity data obtained. The adverse effects reported and their implications for clinical practice are discussed.Results: Two RCTs were identified, with the authors of both concluding that there was no detectable difference between the two therapies for the age groups they studied. The mean difference between atropine penalization and occlusion therapies was calculated to be − 0.01 logMAR (95% confidence interval − 0.07 to 0.03 logMAR) in favor of occlusion therapy, and no statistical difference between the two groups was detected (P = 0.45). Neither study detected a marked difference in terms of reported adverse effects from the two interventions.Conclusion: Based on the results of our meta-analysis we conclude that there is no difference in visual acuity outcomes between atropine penalization therapy and occlusion therapy after 17 to 24 weeks of treatment in children aged 7-12 years. Further evidence to determine the efficacy of amblyopia therapy for an older patient population is required before studies comparing atropine penalization and occlusion therapy in patients older than 12 years can be performed. We studied the postnatal refinement of feedforward (FF) projections from ferret V1 to multiple cortical targets during the period around eye opening. Our goal was to establish (a) whether the developmental refinement of FF projections parallels that of feedback (FB) cortical circuits, and (b) whether FF pathways from V1 to different target areas refine with a similar rate. We injected the tracer CTb into V1 of juvenile ferrets, and visualized the pattern of labeled axon terminals in extrastriate cortex. Bouton density of FF projections to target areas 18, 19, and 21 declined steadily from 4 to 8 weeks postnatal. However, in area Ssy this decline was delayed somewhat, not occurring until after 6 weeks. During this postnatal period, mean interbouton intervals along individual FF axons to all visual areas increased, and we observed a concomitant moderate decrease in axon density in areas 18, 21, and Ssy. These data suggest that FF circuits linking V1 to its main extrastriate targets remodel largely synchronously in the weeks following eye opening, that FF and FB cortical circuits share a broadly similar developmental timecourse, and that postnatal visual experience is critical for the refinement of both FF and FB cortical circuits. Purpose: To analyze long-term visual development in children with Down syndrome who received early ophthalmological intervention. Methods: A total of 125 children with Down syndrome who were examined before 6 years of age and followed up for more than 5 years were selected. Visual development, refraction, visual acuity testing, and the prescription of spectacles were examined retrospectively. Results: Mean visual acuity by age was as follows: 2 years, 1.13 ± 0.23 logarithm of the minimum angle of resolution (logMAR); 5 years, 0.55 ± 0.25 logMAR; 8 years, 0.27 ± 0.19 logMAR; 11 years, 0.17 ± 0.16 logMAR; and 14 years, 0.10 ± 0.15 logMAR. In 32 children (25.6%), visual acuity reached 0.0 logMAR or better. Hyperopia of +2D or more was observed in 132 eyes (52.8%), and astigmatism of 2D or more was observed in 153 eyes (61.2%). Subjective testing was difficult in many children prior to 4.5 years of age, and grating acuity testing was necessary. Spectacles were prescribed at a mean age of 3.5 ± 1.6 years in 120 children (96.0%). The average duration until the spectacles were worn constantly was 9.0 ± 9.3 months. Visual motion perception can arise from non-directional visual stimuli, such as still images (implied motion, cf. Kourtzi, Trends Cogn Sci 8:47-49, ). We tested 5- to 8-month-old infants' implied motion perception with two experiments using the forced-choice preferential looking method. Our results indicated that a still image of a person running toward either the left or right side significantly enhanced infants' visual preference for a visual target that consistently appeared on the same side as the running direction (the run condition in Experiment 1). Such enhanced visual preference disappeared in response to an image of the same person standing and facing the left/right side (the stand condition in Experiment 1), an image of the running figure covered with a set of opaque rectangles (the block condition in Experiment 2) (Gervais et al. in Atten Percept Psychophys 72:1437-1443, ), and an image of the inverted running figure (the inversion condition in Experiment 3). These results suggest that only the figure that implied dynamic body motion shifted the infants' visual preference to the same direction as the implied running action. Electrophysiological studies indicate that cat visual cortical critical period neuronal plasticity peaks around 5 weeks and largely disappears by 20 weeks. Dark rearing slows this time course. Normal cats are more plastic than dark-reared cats at 5 weeks, but the opposite is true at 20 weeks. Thus, a stringent criterion for identifying genes controlling neuronal plasticity is that normal and dark rearing produce opposite direction differences in expression between young and older animals. Differential display polymerase chain reaction identified Abelson interacting protein 2 (Abi-2) as a candidate plasticity gene regulated according to this criterion. Western blotting showed bidirectional regulation of Abi-2 protein levels in cats and mice that was specific to visual cortex and did not occur in frontal cortex. Immunohistochemistry indicated developmental changes in Abi-2 laminar expression in cat visual cortex. Dark rearing altered laminar expression such that at 5 weeks, dark-reared cats were similar to 1-week normally reared cats, and at 20 weeks, dark-reared cats were similar to 5-10-week normally reared animals. The effect of dark rearing on both Abi-2 expression levels and laminar expression patterns was to slow the normal developmental process, the same effect seen on physiologically assessed plasticity in visual cortex. A critical question in brain development is whether different brain circuits mature concurrently or with different timescales. To characterize the anatomical and functional development of different visual cortical areas, one must be able to distinguish these areas. Here, we show that zinc histochemistry, which reveals a subset of glutamatergic processes, can be used to reliably distinguish visual areas in juvenile and adult ferret cerebral cortex, and that the postnatal decline in levels of synaptic zinc follows a broadly similar developmental trajectory in multiple areas of ferret visual cortex. Zinc staining in all areas examined (17, 18, 19, 21, and Suprasylvian) is greater in the 5-week-old than in the adult. Furthermore, there is less laminar variation in zinc staining in the 5-week-old visual cortex than in the adult. Despite differences in staining intensity, areal boundaries can be discerned in the juvenile as in the adult. By 6 weeks of age, we observe a significant decline in visual cortical synaptic zinc; this decline was most pronounced in layer IV of areas 17 and 18, with much less change in higher-order extrastriate areas during the important period in visual cortical development following eye opening. By 10 weeks of age, the laminar pattern of zinc staining in all visual areas is essentially adultlike. The decline in synaptic zinc in the supra- and infragranular layers in all areas proceeds at the same rate, though the decline in layer IV does not. These results suggest that the timecourse of synaptic zinc decline is lamina specific, and further confirm and extend the notion that at least some aspects of cortical maturation follow a similar developmental timecourse in multiple areas. The postnatal decline in synaptic zinc we observe during the second postnatal month begins after eye opening, consistent with evidence that synaptic zinc is regulated by sensory experience. It has been shown that infants over the age of 6 months will reach for an object in complete darkness. This experiment measured the reaching movements of 9- to 16-month-old infants and adults under several different conditions of illumination to investigate the role of vision and stored visual representations in reach control. In one condition, participants reached for an object with the lights on. In a second condition, participants reached for an object glowing in the dark (glowing condition). This allowed us to measure the effects of vision of the arm and vision of the reach space. We also looked at the effect of removing vision of the object on reach control: in the final two conditions, participants reached for an object in complete darkness (0-s dark) and in complete darkness after a 4-s delay (4-s dark). We compared the kinematics of a reach (e.g. average speed, reach straightness) between the four illumination conditions. The results showed that infants reached faster and decelerated for a shorter period of time in the dark (0- and 4-s dark) than in the light. By comparison, adults reached slower and decelerated for a longer period of time in the dark (0- and 4-s dark) than in the light. We did not find any effect of the glowing condition compared to full vision on infant reaching movements. These results suggest that infant reaching movements only become compromised when the target is not visible, whereas vision of the hand and the reach space are less significant. Without online visual feedback, an infant reach in the dark appears to be driven by feedforward mechanisms and control may be affected by an immature ability to form and/or retain visual spatial memory. Increased prevalence of visual impairments has been reported in preterm populations. However, it remains unclear about the long-term visual electrophysiological outcomes and their association with visual cognitive functions in premature infants. We investigated visual electrophysiological outcome of 2-year-old infants of different birth weights by flash visual evoked potentials (FVEPs) in order to explore the correlation between visual cognitive functions and FVEPs and to assess the application of FVEPs in evaluating the visual capability of an infant. The FVEPs of 77 infants, including 25 very low birth weight (VLBW) premature infants, 16 low birth weight (LBW) premature infants and 36 full-term infants, were tested with a visual electrophysiological testing device. Neuromotor development was assessed with the Bayley Scales of Infant Development, Second Edition (BSID-II). The visual cognitive functions were evaluated by scoring the proportion passed of 12 items chosen from the BSID-II for infants at 23 to 25 months of age. The second prominent positive wave (P2) was the major component presented in all three groups. The mean latency of P2 in the VLBW, LBW and full-term groups was 149.65±23.79 ms, 129.39±8.70 ms, and 126.14±7.73 ms respectively. There was no significant difference in mean latency of P2 wave between the LBW and full-term groups; the mean latency of the P2 wave in the VLBW group was delayed more significantly than those of the other two groups. The difference in amplitude of the P2 peak to the preceding N2 peak (N2P2) between the three groups was not statistically significant. The latency of the P2 main wave was negatively correlated with mental developmental index (MDI) (r’MDI = −0.35) and visual cognitive capability (r’visual capability = −0.21). The latency of the P2 main wave on FVEPs was delayed more significantly in premature infants than in full-term infants at 2 years of corrected age. The visual functional development was delayed in premature infants, especially in VLBW infants (gestational age 〈32 weeks). The FVEPs were reported low but there were statistically significant correlations between measures of visual cognition and P2 peak latency. The development of dynamic vision was investigated in 400 healthy subjects (200 females and 200 males) aged between 4 and 24 years. The test consisted of a computer-generated random-dot kinematogram in which a Landolt ring was briefly presented as a form-from-motion stimulus. Motion contrast between the ring and background was varied in terms of the percentage of dots moving coherently within the ring in four levels (100%, 50%, 30%, and 20%). The subject’s task was to indicate the position of a gap in the ring (left, right, top, bottom). Results show a clear increase in performance with age for all motion contrast levels, with the greatest changes for the lowest levels. Adult performance was reached at the age of 15 years. Different features of visual function mature along unique timescales through infancy and early childhood. It is not clear which functions continue to mature in school age children. Functions believed to be mediated by the Magnocellular (M) and Parvocellular (P) pathways were compared in five- ( n=25), eight- ( n=21) and eleven-year-old children ( n=21) and young adult controls ( n=20). Steady-state visual evoked potentials were recorded from occipital electrodes in response to very low spatial frequency gratings, at a series of contrasts (M), and to high contrast gratings at a series of spatial frequencies (P). No evidence was found to indicate M pathway development across these age groups. However, the youngest children demonstrated elevated VEP thresholds to the high contrast gratings compared with either the adults or eleven-year-olds. A recent model for the development of the pattern of eye-dominance domains in primary visual cortex predicts that stimulus conditions during early visual life determine the spacing (or periodicity) of ocular dominance columns (ODC). The model predicts that normal binocular visual experience consists of highly correlated binocular stimulation and leads to relatively narrow ODC spacing, while abnormal binocular visual stimulation attendant with strabismus consists of non-correlated, incoherent, and asynchronous stimulation and leads to wider than normal ODC spacing. Evidence in support of the model has been presented for strabismus in the kitten. We tested the predictions of the model in normal monkeys and others subjected to various forms of abnormal visual experience during infancy. We identified and measured the inter-column spacing (or periodicity) in the V1 cortex of 19 adult monkeys ( M. mulatta) using the cytochrome-oxidase (CO) histochemical method. There were no significant differences in the V1 inter-column spacing between normal adult monkeys ( n=5) and other adult monkeys having had monocular-form deprivation ( n=5), experimental anisometropia ( n=5), or experimental strabismus ( n=4) early in life. The quality of early binocular visual experience is not a significant determinant of the inter-column spacing in primate V1 cortex. We studied the development of spatial contrast-sensitivity and temporal-frequency selectivity for neurons in the monkey lateral geniculate nucleus. During postnatal week 1, the spatial properties of P-cells and M-cells are hardly distinguishable, with low contrast-sensitivity, sluggish responses, and poor spatial resolution. The acuity of P-cells improves progressively until at least 8 months, but there is no obvious increase in their maximum contrast-sensitivity with age. The contrast sensitivity of M-cells is already clearly higher than that of P-cells by 2 months, and at 8 months of age this characteristic difference between M- and P-cells approaches the adult pattern. There is a major increase in responsiveness during the first 2 postnatal months, especially for M-cells, the peak firing rate of which rises fivefold, on average, between birth and 2 months. Many P-cells in the neonatal and 2-month-old animals did not give statistically reliable responses to achromatic gratings, even at the highest contrasts: this unresponsiveness of P-cells might result from low gain and/or chromatic opponency. The upper limit of temporal resolution in the neonate is low – about one-third of that in the adult. Among M-cells, the improvement in temporal resolution, like that in contrast sensitivity, is rapid over the first 2 months, followed by a slower change approaching the adult value by 8 months of age. The development of contrast sensitivity, responsiveness and temporal tuning are little affected, if at all, by binocular deprivation of pattern vision from birth for even a prolonged period. Purpose: Our aim was to answer three questions 1) Do adult rhesus monkeys have binocular luminance interactions (BLIs) similar to those found in adult humans? 2) Is BLI in very young rhesus monkeys functionally mature? 3) If not, how does it change with age? Methods: We recorded visually evoked potentials (VEPs) in response to sinusoidally modulated uniform fields. The fields were presented dichoptically by varying the relative temporal phase between the two eyes. Monkeys varied in age from 5.6 weeks to 5.25 years. Results: VEPs were Fourier analyzed and the relative second harmonic amplitudes were taken as the response measure. The second harmonic amplitudes in adult monkeys had an asymmetrical ‘V-shaped’ function as interocular phase difference (IPD) varied from 0° to 180°, as had been observed previously in adult humans [1]. The youngest monkeys exhibited a symmetrical pattern which became more asymmetrical at older ages and was adult like by about 19 wee ks. Asymmetry magnitude and log age correlated 0.97 (p < 0.05) in the monkeys younger than 19 weeks. Conclusions: The adult rhesus data are consistent with a model derived from humans which involves two types binocular luminance processing. One combines monocular responses nonlinearly (MNL) and a second combines monocular responses linearly followed by a binocular nonlinearity (MLBNL). These two processes have been associated with the parvocellular (P-) and magnocellular (M-) streams. Within this framework, the data from the youngest monkeys indicate that BLI in the P-stream is relatively less mature at birth than that in the M-stream and develops reaching functional maturity on these measures by around 19 weeks. We strictly limited small injection of horseradish peroxidase (HRP) to lamina A of the lateral geniculate nucleus of cats. This was done to label retrogradely only the alpha (Y) and beta (X) classes of retinal ganglion cell. Eighty-six such injections at a range of matched eccentricities were made bilaterally in 9 normal adult cats, 7 cats reared from birth to adulthood with monocular lid suture, and 9 normal kittens at 4 weeks of age; 5348 alpha and beta cells were retrogradely labeled from these injections. Quantitative measurements were made from these labeled cells and compared among 4 experimental conditions, these being normal adult retinas, the nondeprived and deprived retinas of lid sutured cats, and the retinas of kittens. Each injection led to a similar relative ratio of labeled alpha and beta cells (typically 5-15% alpha cells) that did not differ significantly among the experimental conditions, but further analysis suggested a slight dimunition of labeled alpha cells in deprived retinas. Because the larger arbors of retinogeniculate Y axons are more likely to penetrate small geniculate HRP injection sites from eccentric locations than would be the case for the more restricted arbors of X axons, a normal tendency resulted for the peripheral halo of zones of retrograde labeling to be dominated by alpha cells. Thus a more accurate reflection of the relative numbers of labeled alpha and beta cells would result from considering only the core of zones of retrograde labeling. When this is done, deprived retinas exhibited relatively fewer labeled alpha cells than did normal, nondeprived, or kitten retinas. This may relate to prior observations (Sur et al. 1982) that abnormally few Y axons from the derpived retina innervate lamina A. No statistically significant differences in alpha or beta cell size were seen among normal, nondeprived, and deprived retinas, although both of these cell types in the kittens were equally smaller than their normal adult counterparts. This is particularly interesting in view of the postnatal growth of retinogeniculate axon arbors (Sur et al. 1984). The results are not surprising for alpha cells, since retinogeniculate Y axon arbors grow considerably after 4 weeks of age, but they are surprising for beta cells, since retinogeniculate arbors of X axons decrease after 4 weeks of age. This suggests no clear, general relationship between soma size and the extent of a cell's axonal arbor. Overall, these results suggest that no dramatic abnormalities due to rearing with monocular suture are evident at the level of the retina, although subtle effects can be demonstrated there (see also Leventhal and Hirsch 1983). In order to isolate some of the factors responsible for strabismic amblyopia as well as to ascertain the time course of its development, frequent measurements were made of the visual acuity of the two eyes of kittens following imposition of surgically induced strabismus. Following behavioural training on a jumping stand, strabismus was induced in all but one animal by simple section of either the lateral (esotropia) or medial (exotropia) rectus muscle of one eye. The one exception was a kitten on which esotropia was induced by another common but more radical surgical procedure that involved removal of the body of both the lateral rectus and superior oblique muscles of one eye. There was a surprising difference between the immediate consequences for vision of section of the lateral and medial rectus muscles that were reflected by equally large differences in the magnitude of the amblyopia that developed eventually in the two situations. Following section of the lateral rectus muscle, there was an immediate reduction in the visual acuity of the operated eye of as much as 2 octaves after which vision returned to normal levels over 4 to 8 days. The acuity of the two eyes remained comparable for a few days after which the vision of the operated eye began to decline once more, signalling the onset of amblyopia some 10 to 12 days following imposition of strabismus. In contrast to the severity of these effects, the effects observed following section of the medial rectus were both mild and transitory. Furthermore, whereas all kittens that were rendered esotropic early developed amblyopia, none of the kittens that were made exotropic at the same age did so. Together, these results suggest that factors associated with the immediate consequences of the surgical procedure employed to produce a misalignment of the visual axes may contribute to the severity of the effects of surgically induced strabismus. The retina of the California ground squirrel ( Spermophilus beecheyi) contains three classes of photopigments (λ = 440, 500, 525 nm). From optic nerve recordings it was previously discovered that the effectiveness of signals from the 440 nm cone increase gradually over the weeks following lid opening. In this experiment several features of the electroretinogram (ERG) were examined to assess developmental changes in signals originating in the outer retina. As judged by threshold sensitivity, suprathreshold responsivity, or adaptability, the contribution of the 525 nm mechanism reaches its adult level two to three weeks after lid opening. The 500 nm mechanism appears to have a similar developental time course. The development of the 440 nm mechanism was tracked using a chromatic adaptation measure. So indexed, the time required for this mechanism to reach its adult status was much greater than that for the other two mechanisms (70-80 days vs 40-45 days of age). Visual callosal connections were examined using autoradiographic (ARG) and horse-radish peroxidase (HRP) techniques in normal adult hamsters, and in adults subjected to ablation of the superficial tectal laminae at birth. Additional ARG and HRP experiments were carried out in hamsters 1-27 days of age in order to describe the normal development of this pathway. Neonatal collicular lesions, which deprived visual cortical neurons of a major terminal zone in the midbrain, substantially altered the visual callosal pathway. In the lesioned animals, the numbers of supragranular callosal cells in the 17-18a border region and lamina VI callosal neurons in medial area 17 were significantly greater than normal. The ARG experiments demonstrated additional clearcut abnormalities in the visual callosal pathway of the lesioned hamsters. First, the mediolateral extent of the supragranular callosal zone around the 17-18a border was increased. Secondly, dense label was visible over lower layer V and lamina VI throughout area 17. Finally, labelling in lamina I could also be observed across the entire mediolateral extent of area 17. Experiments in the developing hamsters suggested that some of the abnormalities observed in the lesioned animals may have resulted from the maintenance of normally transient developmental states. During the first postnatal week, both callosal cells and anterograde labelling were evenly distributed throughout the dorsal posterior neocortex, but only in the subplate region. During the second postnatal week, supragranular callosal cells were also labelled in both medial and lateral area 17, but from their first appearance, they were always most numerous in the 17-18a border region. At the same time callosal axons invaded the supragranular laminae, but only near the 17-18a border. The California ground squirrel ( Spermophilus beecheyi) has two classes of cone photopigments (λ = 440 and 525 nm). Under photopic conditions about 30% of all optic nerve fibers receive inputs from both cone classes, the remainder are driven solely by the 525 nm cone. Recordings were made from optic nerves in young ground squirrels to trace the development of their spectral responsivity. Animals were reared from birth in one of three photic environments: white light, darkness, or red light. In the youngest ground squirrels examined (ca. 50 days old) many units receiving inputs from the 525 nm cone were found, but the proportion of units receiving inputs from both cone classes was significantly lower than that of adults. From these initial low levels the proportion of such units increases gradually. Adult proportions were achieved at varying rates which depended on the photic environment in which the animal was reared: animals reared in white light achieved the adult standard earliest, those reared in darkness somewhat later, and the animals reared in red light required much longer to achieve the adult organization. We conclude that: (a) the neural substrates for normal color vision in this species develop to some extent postnatally, and (b) the normal sequence of development can be significantly extended by spectral environments which provide a highly biased stimulation of the two cone mechanisms. Thirty-four kittens reared by allowing each eye patterned visual input, but on alternate days and for unequal periods of time (unequal alternating monocular exposure (AME)), were tested for their ability to orient to targets at different positions in visual space. In all unequal AME cats, the visual field of the more experienced eye (MEE) was normal, while that of the less experienced eye (LEE) was restricted. In contrast, in 14 cats reared with equal AME, the fields of both eyes were equal and of normal size. The field deficits observed in the unequal AME cats must therefore be due to the imbalance in stimulation and thus result from a competitive interaction between the afferents from the MEE and the LEE. The field deficits observed in the unequal AME cats differed from those observed in two monocularly deprived (MD) cats. Neither of the MD cats ever responded to targets presented in the region of normal binocular overlap when tested with the deprived eye (DE). The unequal AME cats all showed a nasal field loss in the LEE, but responded normally to targets throughout the temporal portion of the binocular visual field. When the imbalance in stimulation was large (8 to 1) or moderate (8 to 4), there was an abrupt drop in responsiveness (from 100% to zero) as the position of the target was changed from temporal to nasal. When the imbalance was slight (8 to 7), the drop in responsiveness was more gradual and occurred within the nasal field. Our results demonstrate that (1) the paradigm of unequal AME is a useful one for studying binocular competition quantitatively, (2) even the slightest imbalance in stimulation of the two eyes can affect the outcome of the competitive interaction, and (3) the pathways serving binocular vision are not uniformly affected by binocular competition: the ipsilateral pathway is more sensitive than the contralateral pathway. Visual pattern and brightness discriminations were tested in cats which had undergone surgical cyclodeviations of one or both eyes. Half of the pattern discriminations were learned with both eyes open and performance was tested with each eye separately; the other half were learned monocularly with the normal eye (in cats with monocular rotations, MR) or with the less amblyopic eye (in cats with binocular rotations, BR) and then the rotated (or amblyopic) eye was tested alone. No deficits were found in brightness discriminations. With monocularly learned pattern discriminations all but one cat showed positive savings when tested with the rotated eye. However, on binocularly learned discriminations half of the animals performed poorly with the rotated eye and required extensive retraining; they showed negative savings when compared to original learning. These animals seem to be suppressing the strabismic eye during binocular vision. For MR cats, there was a positive relationship between visual acuity and percent savings after binocular learning. In adult cats that had previously undergone surgical section of the posterior corpus callosum at 13-18 days after birth, the striate cortex was examined using extracellular single unit recordings. The receptive fields of the cells examined were located from the vertical meridian to 39 ° peripherally, and ranged from above to below the horizontal meridian. Cells were classified according to type (simple, complex), ocular dominance, receptive field size and location. Callosum sectioned cats had 53% of striate cells activated monocularly as compared to 25% for control cats. This increase in monocularly activated units primarily occurred for receptive fields in the paracentral region of the visual field, from 4-39 °. The age at which the neonatal surgery had occurred was correlated with the individual cat's proportion of monocularly activated cells. Therefore, the increase in monocular activation of striate units occurred within a large portion of the normal binocular visual field. This physiological change was partially predicted by a previous behavioral study showing a substantial loss in the extent of the binocular visual field following neonatal corpus callosum section (Elberger 1979). In young cats, the posterior portion of the corpus callosum was sectioned 13-29 days after birth. The animal's eyes were photographed at weekly intervals for six months using the pupil-reflex method. From the corneal reflection evident in the photographs the degree of alignment for the optical axes of each cat was estimated (Sherman, 1972). The 17 experimental cats all showed a significant tendency toward permanent divergent strabismus, as compared to six normal cats. The limits of the visual field were determined for both groups of cats using a perimetry technique similar to that of Sprague and Meikle (1965) and Sherman (1973). With one eye open normal cats responded from 90 ° ipsilateral to 45 ° past the vertical midline into the contralateral visual field. With either eye the experimental cats responded from 90 ° ipsilateral to approximately the vertical midline. The loss of visual responsiveness is within the contralateral region of the normally binocular zone. Three cats received the same operation at 9, 13, or 20 months old. Eye alignment and visual field perimetry were unaffected by the surgery. It is known that kittens reared in ways that restrict movement while visual stimulation is received exhibit deficits in visually guided behavior. Presumably, the behavioral dysfunction is due to a lack of sensorimotor coordination during visual exposure. The current investigation was undertaken to study this effect both physiologically and behaviorally. Two groups of kittens were normally reared until they were nearly 4 weeks old. They were then placed in a darkroom and exposed daily for 1-2 hours while one eye was occluded. On alternate days, alternate eyes were covered. One group was restrained in a body cast while a given eye was exposed, but the kittens were free to move about while the other eye received visual experience. A second control group was alternately occluded, but freely mobile during all exposures. An additional cat was also unrestrained during daily rearing periods and neither eye was ever occluded. Behavioral tests showed clear deficits when the 'passive' eye of the restrained-unrestrained group was used. Deficiencies were found in visually guided paw placing, pursuit eye movements, and jumping behavior to a platform. Responses of single cells were studied in area 17 of the visual cortex. Ocular dominance distributions showed marked reductions in binocularity for alternate occluder cats. The eye that had received exposure while animals were active in the restrained-unrestrained group also tended to predominate but the difference was not statistically significant. In twelve kittens, convergent squint was surgically produced at either 3, 6, 8, 10, 13, or 16 weeks. When these kittens reached the age of 4-8 months, the spatial resolution of 'sustained' cells which received inputs from the area centralis in layers A and A1 of both the left and right lateral geniculate nucleus (LGN) was measured under nitrous oxide/halothane anaesthesia, using the highest spatial frequency of a sinusoidal grating resolved by cells as a measure of cellular visual acuity. Spatial resolution of cells fed by the squinting eye's area centralis was poorest in the kittens in which the squint was produced at 3 weeks. The resolution of the cells driven by the area centralis of the squinting eye gradually improved for the kittens in which the squint was produced at progressively later stages. There was no effect of convergent squint on the resolving power of cells in the kittens in which squint was produced at 13 weeks and 16 weeks. The developmental curve of spatial resolution of LGN cells obtained from normal kittens of different ages was found to fit very closely with a plot of the spatial resolution of cells driven by the squinting eye against age at squint production. These results suggest that the loss of spatial resolution (amblyopia) in the eye with convergent squint is due to the arrest of development of spatial resolution during the sensitive period in early postnatal life. The time course of development of visual acuity for square wave gratings was measured behaviourally in a number of cats that were reared in total darkness until they were either 4 (5 cats) or 6 (1 cat) months of age. Less extensive measurements were also made on animals reared in a similar manner until they were either 1 1/2 or 10 months old. Initially, all the animals appeared to be blind, but signs of vision became evident after periods of time in an illuminated environment that ranged from a few days, in the case of animals dark-reared for only 1 1/2 to 4 months, to 1 to 2 months for those animals that were deprived for 6 months or more. Thereafter, visual acuity as measured on a jumping stand progressively improved, reaching, in the case of animals deprived for 4 months, values that were comparable to those of normal animals (6.9 cycles/deg) after 4 months of exposure to light. The animal deprived for 6 months remained apparently blind for a month and eventually attained an acuity (5.7 cycles/deg) that was somewhat less than that of normal animals. The fact that such high acuities can be achieved after periods of binocular deprivation that extend throughout the classically defined 'critical period' suggest that the effect of dark-rearing is somehow to impede the natural decline with age in the degree to which cortical neurones are susceptible to environmental modification. One optic nerve was cut in a series of larval Xenopus laevis; in approximately half of these the corresponding eye was also rotated. The resultant primary visual projections were mapped in the adult. Although in most cases the projections were retinotopically normal, some of them were not. These abnormal projections were consistently associated with a very thin optic nerve which reached the tectum via the oculomotor route. Animals with rotated eyes which developed retinotopically organised primary projections had a normal intertectal ipsilateral projection from the rotated eye and a rotated projection from the normal eye, thus confirming earlier findings that a process of binocular interaction determines the formation of intertectal connections. In all frogs with retinotopically abnormal primary projections the intertectal connections from the normal eye were of the type found after enucleation and those from the abnormal side were fragmentary or absent. Since the development of abnormal retinotopic patterns preceded that of intertectal connections the mechanisms of binocular interaction must have broken down in the presence of the abnormal primary projections. The development of intertectal neuronal connections has been investigated in Xenopus laevis. Contralateral eye grafts and enucleations were performed in embryos and the resultant visual projections to the optic tecta were mapped electrophysiologically after metamorphosis. In enucleated animals the ipsilateral projections were found to be normally organised retinotopically but consisted of visual units with abnormally large multi-unit receptive fields. In 10 animals with contralaterally grafted eyes a normal ipsilateral projection had developed from the abnormal eye and an abnormal projection from the normal eye, to produce congruent maps via the two eyes to one tectum. All the maps in these animals were retinotopically organised. In another 11 animals the ipsilateral projection from the operated eye was fragmentary or absent, while that from the unoperated eye resembled the pattern found after enucleation. Retinotopically abnormal contralateral projections had developed in 5 animals of this group. These results suggest that prefunctional specification determines the initial development of diffuse intertectal visual connections but these may be modified by a process of binocular interaction in the presence of a normal primary contralateral input.
