The aim of this study is to evaluate the predictive ability of MRI-based radiomics combined with tumor markers for TN staging in patients with rectal cancer and to develop a prediction model for TN staging. A total of 190 patients with rectal adenocarcinoma who underwent total mesorectal excision at the First Affiliated Hospital of the Air Force Medical University between January 2016 and December 2020 were included in the study. An additional 54 patients from a prospective validation cohort were included between August 2022 and August 2023. Preoperative tumor markers and MRI imaging data were collected from all enrolled patients. The 190 patients were divided into a training cohort (n = 133) and a validation cohort (n = 57). Radiomics features were extracted by outlining the region of interest (ROI) on T2WI sequence images. Feature selection and radiomics score (Rad-score) construction were performed using least absolute shrinkage and selection operator regression analysis (LASSO). The postoperative pathology TNM stage was used to differentiate locally advanced rectal cancer (T3/4 or N1/2) from locally early rectal cancer (T1/2, N0). Logistic regression was used to construct separate prediction models for T stage and N stage. The models' predictive performance was evaluated using DCA curves and calibration curves. The T staging model showed that Rad-score, based on 8 radiomics features, was an independent predictor of T staging. When combined with CEA, tumor diameter, mesoretal fascia (MRF), and extramural venous invasion (EMVI), it effectively differentiated between T1/2 and T3/4 stage rectal cancers in the training cohort (AUC 0.87 [95% CI: 0.81–0.93]). The N-staging model found that Rad-score, based on 10 radiomics features, was an independent predictor of N-staging. When combined with CA19.9, degree of differentiation, and EMVI, it effectively differentiated between N0 and N1/2 stage rectal cancers. The training cohort had an AUC of 0.84 (95% CI: 0.77–0.91). The calibration curves demonstrated good precision between the predicted and actual results. The DCA curves indicated that both sets of predictive models could provide net clinical benefits for diagnosis. MRI-based radiomics features are independent predictors of T staging and N staging. The pyrin domain-containing protein 3 (NLRP3) inflammasome drives the profound cerebral ischemia and reperfusion injury (I/R) and mediates the secretion of IL-1β (interleukin-1β), which exerts a subsequent cascade of inflammatory injury. The NLRP3-activated-microglial manipulation in adjacent neuronal and endothelial NLRP3 activation has been confirmed in our previous studies. In the present study, we extended the cognition of how microglia mediated neuronal and endothelial NLRP3-IL-1β signaling during cerebral ischemia and reperfusion injury. In vitro, Neuro-2a and bEND3 cells were cultured alone or co-cultured with BV2 cells and oxygen–glucose deprivation/reoxygenation (OGD/R) was performed. In vivo, transient middle cerebral artery occlusion (tMCAO) rat models and lentiviral silencing targeting IL-1R1 were performed. The NLRP3 inflammasome activation was evaluated by enzyme-linked immunosorbent assay, western blotting, immunoprecipitation, immunohistochemistry, and immunofluorescence. In the co-culture system after OGD/R treatment, NLRP3 inflammasomes in neurons and endothelial cells were activated by microglial IL-1β via IL-1β/IL-1R1/TRAF6 signaling pathway, with the basal protein level of NLRP3. In addition, ruptured lysosomes engulfing ASC specks which were possibly secreted from microglia triggered the enhanced NLRP3 expression. In cortices of tMCAO rats at 24 h of reperfusion, silencing IL-1R1, mainly presented in neurons and endothelial cells, was efficient to block the subsequent inflammatory damage and leukocyte brain infiltration, leading to better neurological outcome. Neuronal and endothelial NLRP3 inflammasomes were activated by microglia in cerebral ischemia and reperfusion injury mainly via IL-1β/IL-1R1/TRAF6 signaling, which might be therapeutically targetable. To provide a reference for reducing the cost of industrial wastewater treatment and alleviate the pressure on water environment governance in China, we use the non-parametric dual evaluation linear analysis framework to estimate the shadow price of China's urban industrial wastewater (IWSP) with consideration of multiple inputs based on the data of 267 cities in China from 2003 to 2016. Then, we investigate the spatiotemporal characteristics of IWSP and analyze its sources of differences. Main conclusions are as follows: (1) Mean of China's urban IWSP increased from 645.54 yuan/ton in 2003 to 5662.64 yuan/ton in 2016, implicating the significant results and increasing difficulty of emission reduction policies. In addition, the Moran's I index of IWSP decreased from 0.056 to 0.002, implicating declining spatial correlation and differentiated green production processes in various regions. (2) From stock perspective, the σ convergence result shows that the IWSP of the country and each region gradually diverges, and the β convergence results from incremental perspective show that the IWSP of a single region tends to converge in a steady state. Furthermore, regions with lower average shadow prices converge faster than regions with higher average shadow prices. (3) Using the Dagum Gini coefficient method, we find that the overall difference of IWSP dropped from 0.5758 to 0.3568. The intra-regional differences in each region continued to decline, as well as inter-regional differences. And the contribution rate of intensity of transvariation has risen from 33.71 to 60.80%, becoming the main reason for the imbalanced distribution of IWSP. [ABSTRACT FROM AUTHOR]

The toxicity of silver nanoparticles (AgNPs) with a single morphology to aquatic organisms has been well demonstrated in the past decade, but few studies have been carried out to evaluate the differences in toxicity among AgNPs with various microstructural morphologies. In this work, C. vulgaris was used as the tested organism to examine the differences in toxic effects among AgNSs, AgNCs, and AgPLs at concentrations of 0.5, 1.0, 2.0, and 5.0 mg/L. The results showed that the cell density and chlorophyll a content of C. vulgaris decreased when the dose of AgNPs was increased, while the inhibiting effects that were caused by AgPLs were stronger than those that were caused by AgNCs and AgNSs. Under short-term exposure to AgPLs, the ROS content was significantly higher than those under exposure to AgNCs and AgNSs, while the MDA content fluctuated without obvious regularity. The dose of AgPLs affected the antioxidative enzyme activity and lipid peroxidation more obviously than those of AgNSs and AgNCs. The superoxide dismutase and catalase contents in the former case were distinctly higher than those in the latter cases. Consequently, the cell apoptosis rate under exposure to AgPLs reached 83%, which was higher than those under exposure to AgNSs (50%) and AgNCs (71%). This work shows that the level of toxicity to C. vulgaris was in the order of AgPLs > AgNCs > AgNSs. Alcohol use–associated disorders are highly comorbid with anxiety disorders; however, their mechanism remains unknown. The amygdala plays a central role in anxiety. We recently found that 7,8-dihydroxyflavone (7,8-DHF) significantly reduces withdrawal symptoms in a rat model of chronic intermittent alcohol (ethanol) exposure. This study aimed to determine the role of 7,8-DHF in regulating anxiety induced by chronic alcohol exposure and its associated underlying mechanism. Male C57BL/6J mice were exposed to chronic intermittent alcohol for 3 weeks followed by alcohol withdrawal for 12 h with or without 7,8-DHF administered intraperitoneally. All mice were tested using an open field test and elevated plus maze to assess anxiety-like behaviors. Synaptic activity and intrinsic excitability in basal and lateral amygdala (BLA) neurons were assessed using electrophysiological recordings. 7,8-DHF alleviated alcohol-induced anxiety-like behavior and attenuated alcohol-induced enhancement of activities in BLA pyramidal neurons. Furthermore, 7,8-DHF prevented alcohol withdrawal–evoked augmentation of glutamatergic transmission in the amygdala and had no effect on GABAergic transmission in the amygdala, as demonstrated by unaltered frequency and amplitude of spontaneous inhibitory postsynaptic currents. Microinjection of K252a, a tropomyosin-related kinase B (TrkB) antagonist, into the BLA blocked the effects of 7,8-DHF on anxiety-like behavior and neuronal activity in the BLA. Our findings suggest that 7,8-DHF alleviates alcohol-induced anxiety-like behavior induced by chronic alcohol exposure through regulation of glutamate transmission involving TrKB in the BLA. In this study, 0.2 wt% zirconium (Zr) added Sn–1.0Ag–0.5Cu (SAC105) lead-free solder alloy was prepared and its microstructure, tensile property, and aging stability were investigated and compared with the original SAC105 solder alloy. The SAC105 and SAC105-0.2Zr solder alloys were fabricated by vacuum induction melting method. The results showed that the Zr-added SAC105 solder exhibited a significantly refined microstructure. The primary β-Sn phases, Ag3Sn particle size, and spacing of Ag3Sn had been obviously reduced with the addition of trace amounts of Zr. The tensile test demonstrated that both strength and elongation of the SAC105-0.2Zr solder improved because of the finer microstructure. Isothermal aging was performed at 180 °C for 36 h, 84 h, and 156 h. The existence of Zr could significantly inhibit the coarsening of the Ag3Sn phases, thus enhancing the microstructure stability under severe aging conditions. Moreover, the Zr-added SAC105 solder showed significant decrease in supercooling, but the melting range was almost unchanged. [ABSTRACT FROM AUTHOR]

Mesenchymal stromal cells (MSCs) can differentiate into multiple tissues. Preclinical studies have shown that MSC-based therapy is a potential new treatment approach for ischemic stroke. These results support the urgent need for further studies of MSC transplantation in the treatment of ischemic stroke in humans. Here, we develop a prospective, randomized, controlled, observer-blinded phase II trial to assess the clinical safety, feasibility, and therapeutic mechanisms of allogenic bone marrow-derived MSCs (BM-MSCs) by intrathecal infusion in the treatment of patients with cerebral infarction within the middle cerebral artery and with a National Institutes of Health Stroke Scale (NIHSS) score from 15 to 25. Sample size calculation has determined that a patient population of 118, with ischemic stroke between 30 and 90 days following onset, will be randomly divided into experimental (n = 59) and control (n = 59) groups. Then eligible patients will receive four intrathecal infusions of allogenic BM-MSCs (1 × 106 cells/kg body weight) once a week. All patients have detailed functional assessments and magnetic resonance imaging prior to cell infusion and at intervals up to 1 year after. The primary outcome is the score on the modified Rankin Scale at 90 days after treatment, and the second outcomes include multiple indicators of safety and feasibility.
