The study of specific glycan uptake and metabolism is an effective tool in aiding with the continued unravelling of the complexities in the human gut microbiome. To this aim fluorescent labelling of glycans may provide a powerful route towards this target. Here, we successfully used the fluorescent label 2-aminobenzamide (2-AB) to monitor and study microbial degradation of labelled glycans. Both single strain and co-cultured fermentations of microbes from the common human-gut derived Bacteroides genus, are able to grow when supplemented with 2-AB labelled glycans of different monosaccharide composition, degrees of acetylation and polymerization. Utilizing a multifaceted approach that combines chromatography, mass spectrometry, microscopy and flow cytometry techniques, it is possible to better understand the metabolism of labelled glycans in both supernatants and at a single cell level. We envisage this combination of complementary techniques will help further the understanding of substrate specificity and the role it plays within microbial communities. Sludge biosolids for agricultural application represent a valuable fertilizer but also a health risk unless pathogens are effectively reduced, and recontamination controlled. The Post Anaerobic Digestion Thermal Hydrolysis Process (Post-AD THP) is gaining interest due to improved dewaterability, reducing the volume and thus transportation costs of biosolids. However, Post-AD THP results in sterile biosolids easily exposed to recontamination by pathogens due to the lack of microbial competitors. In theory, this could be suppressed by establishing a competing community of harmless bacteria. The theory was tested by monitoring the abundance of Escherichia coli (viable counts) and gene abundance (ddPCR) in wastewater recontaminated Post-AD THP biosolids, with and without addition of compost. Respiration, total bacterial population and bacterial diversity (16S rRNA amplicon sequencing) were used to monitor the microbial community. Biosolids from the regulatory approved methods thermophilic AD (TAD) and Pre-AD THP were tested in parallel for comparison. The results demonstrated that regulatory requirements can be reached by storing the TAD and Pre-AD THP biosolids for 3 days after recontamination and the Post-AD THP biosolids for more than 13 days. However, addition of compost suppressed growth of E. coli in Post-AD THP biosolids, reducing the time to comply with regulative requirements. The charophycean green algae (CGA or basal streptophytes) are of particular evolutionary significance because their ancestors gave rise to land plants. One outstanding feature of these algae is that their cell walls exhibit remarkable similarities to those of land plants. Xyloglucan (XyG) is a major structural component of the cell walls of most land plants and was originally thought to be absent in CGA. This study presents evidence that XyG evolved in the CGA. This is based on a) the identification of orthologs of the genetic machinery to produce XyG, b) the identification of XyG in a range of CGA and, c) the structural elucidation of XyG, including uronic acid-containing XyG, in selected CGA. Most notably, XyG fucosylation, a feature considered as a late evolutionary elaboration of the basic XyG structure and orthologs to the corresponding biosynthetic enzymes are shown to be present in Mesotaenium caldariorum. Mikkelsen et al. demonstrate molecular and cellular evidence of the evolution of xyloglucan (XyG), a structural component of the cell walls of most land plants, in charophyte algae. Beneficial modulation of the gut microbiome has high-impact implications not only in humans, but also in livestock that sustain our current societal needs. In this context, we have tailored an acetylated galactoglucomannan (AcGGM) fibre to match unique enzymatic capabilities of Roseburia and Faecalibacterium species, both renowned butyrate-producing gut commensals. Here, we test the accuracy of AcGGM within the complex endogenous gut microbiome of pigs, wherein we resolve 355 metagenome-assembled genomes together with quantitative metaproteomes. In AcGGM-fed pigs, both target populations differentially express AcGGM-specific polysaccharide utilization loci, including novel, mannan-specific esterases that are critical to its deconstruction. However, AcGGM-inclusion also manifests a "butterfly effect", whereby numerous metabolic changes and interdependent cross-feeding pathways occur in neighboring non-mannanolytic populations that produce short-chain fatty acids. Our findings show how intricate structural features and acetylation patterns of dietary fibre can be customized to specific bacterial populations, with potential to create greater modulatory effects at large. Here, the authors tailor an acetylated galactoglucomannan (AcGGM) fibre from spruce wood to specifically enrich Roseburia and Faecalibacterium - beneficial species which have the enzymatic machinery to breakdown the fibre and generate butyrate. Polysaccharides from plant biomass are the most abundant renewable chemicals on Earth and can potentially be converted to a wide variety of useful glycoconjugates. Potential applications of glycoconjugates include therapeutics and drug delivery, vaccine development and as fine chemicals. While anomeric hydroxyl groups of carbohydrates are amenable to a variety of useful chemical modifications, selective cross-coupling to non-reducing ends has remained challenging. Several lytic polysaccharide monooxygenases (LPMOs), powerful enzymes known for their application in cellulose degradation, specifically oxidize non-reducing ends, introducing carbonyl groups that can be utilized for chemical coupling. This study provides a simple and highly specific approach to produce oxime-based glycoconjugates from LPMO-functionalized oligosaccharides. The beneficial mechanisms of the lytic polysaccharide monooxygenase CelS2 (S cLPMO10C) from Streptomyces coelicolor on interactions between cellulose and the processive cellulase cellobiohydrolase I (Cel7A) were investigated by quartz crystal microbalance with dissipation. Initial binding of CelS2 to cellulose coated SiO sensors at 40 °C was rapid, but minimal, and was followed by modest positive changes in oscillation frequencies of the quartz crystal sensors that were attributed to mass loss from the cellulose surface. The presence of oxidized cellulose on the CelS2 treated sensors was verified by X-ray photoelectron spectroscopy. Subsequent binding of purified Cel7A from Trichoderma longibrachiatum at 23 °C was significantly less in overall extent to CelS2 treated sensors than to untreated controls despite identical initial binding rates and dissipation-change: frequency-change ratios (an indication of the rigidity of the newly forming layer). Moreover, initial Cel7A binding to the treated sensors was followed by positive overall frequency changes indicating potential increased hydrolytic removal of cellulose mass by cellobiohydrolase action at 23 °C. Furthermore, secondary binding of Cel7A to the control sensors was coupled with considerable changes in dissipation (indicating greater surface viscoelasticity) not observed during the initial binding phase; the extent of this secondary binding was observed to be negligible during Cel7A interaction with the CelS2 treated sensors. This drastic difference in more viscoelastic secondary binding suggests a reduction in loose non-productive cellobiohydrolase binding following CelS2 treatment. [ABSTRACT FROM AUTHOR]

A xyloglucan-specific endo-1,4β-glucanase (XcXGHA) from Xanthomonas citri pv. mangiferaeindicae has been cloned, expressed in Escherichia coli, purified and characterised. The XcXGHA enzyme belongs to CAZy family GH74 and has catalytic site residues conserved with other xyloglucanases in this family. At its optimal reaction conditions, pH 7.0 and 40 °C, the enzyme has a k/ K value of 2.2 × 10 min M on a tamarind seed xyloglucan substrate. XcXGHA is relatively stable within a broad pH range (pH 4-9) and up to 50 °C ( t of 74 min). XcXGHA is proven to be xyloglucan-specific, and a glycan microarray study verifies that XcXGHA catalyses cleavage of xyloglucan extracted from both monocot and dicot plant species. The enzyme catalyses hydrolysis of tamarind xyloglucan in a unique way by cleaving XXXG into XX and XG (X is xylosyl-substituted glucose; G is unsubstituted glucose), is able to degrade more complex xyloglucans and notably is able to cleave near more substituted xyloglucan motifs such as L [i.e. α- l-Fuc p-(1 → 2)-β- d-Gal p-(1 → 2)-α- d-Xyl p-(1 → 6)-β- d-Glc p]. LC-MS/MS analysis of product profiles of tamarind xyloglucan which had been catalytically degraded by XcXGHA revealed that XcXGHA has specificity for X in subsite −1. The 3D model suggests that XcXGHA consists of two seven-bladed β-propeller domains with the catalytic center formed by the interface of these two domains, which is conserved in xyloglucanases in the GH74 family. However, the XcXGHA has two amino acids (D264 and R472) that differ from the conserved residues of other GH74 xyloglucanases. These two amino acids were predicted to be located on the opposite side of the active site pocket, facing each other and forming a closing surface above the active site pocket. β-Mannans are plant cell wall polysaccharides that are commonly found in human diets. However, a mechanistic understanding into the key populations that degrade this glycan is absent, especially for the dominant Firmicutes phylum. Here, we show that the prominent butyrate-producing Firmicute Roseburia intestinalis expresses two loci conferring metabolism of β-mannans. We combine multi-"omic" analyses and detailed biochemical studies to comprehensively characterize loci-encoded proteins that are involved in β-mannan capturing, importation, de-branching and degradation into monosaccharides. In mixed cultures, R. intestinalis shares the available β-mannan with Bacteroides ovatus, demonstrating that the apparatus allows coexistence in a competitive environment. In murine experiments, β-mannan selectively promotes beneficial gut bacteria, exemplified by increased R. intestinalis, and reduction of mucus-degraders. Our findings highlight that R. intestinalis is a primary degrader of this dietary fiber and that this metabolic capacity could be exploited to selectively promote key members of the healthy microbiota using β-mannan-based therapeutic interventions. How dietary β-mannans are utilized by gut Gram-positive bacteria is unclear.
