Deciphering the physical origin of transient crustal deformation is a difficult challenge in the field of continuous borehole observations of crustal movement because the relevant influencing factors of the phenomenon are extremely multifarious. In the Shanxi Rift, squall lines are a frequently occurring mesoscale convective system. Consequently, strain transients are always observed by the borehole dilatometer network within the Shanxi Rift; however, a fundamental understanding of the transient phenomena induced by squall lines remains elusive. To address this challenge, we adopted a second-order Butterworth band-pass filters (0.5–2 h) to retrieve the borehole volumetric deformations excited by a "dry" squall line from September 21, 2017, that occurred in the Shanxi Rift. The elastic loading of Ricker-wavelet-shaped atmospheric waves induced by squall line, which can cause the main signature of transient deformation to adopt a highly similar shape; this pressure-induced deformation process can last approximately 148–164 min and has a maximum magnitude of approximately 19 nstrain. This work of signal processing clearly identifies the crustal transient induced by a squall line and provides new insights into the origin of some short-lasting crustal deformations in the Shanxi Rift and elsewhere. IL-6 plays an important role in oogenesis in humans. However, at the preimplantation stage, IL-6 production and the role in embryo development remain unclear. In this study, IL-6 concentrations in single-embryo media were analyzed. In addition, the association between IL-6 production and blastocyst formation was investigated. Single-embryo culture media from 194 embryos were collected on day 6 after fertilization and divided into four groups according to the developmental stage of the corresponding embryo, as follows: cleavage stage group, morula-early blastocyst group, unavailable full blastocyst group, and available full blastocyst group. IL-6 concentrations were significantly lower in the cleavage stage group than in the morula-early blastocyst group (p = 0.009), in the unavailable full blastocyst group (p = 0.003), and in the available full blastocyst group (p < 0.001). Logistic regression analysis showed that IL-6 concentration in single-embryo medium was significantly associated with blastocyst formation (odds ratios β1 = 1.876, 95% CI 1.433 to 2.644, p < 0.0001). Controlled blasting technology is an effective way to enhance coalbed methane recovery, but the theoretical research has fallen behind engineering practice seriously. Therefore, in this work, based on the damage and fracture mechanics theory, the coal damage process and mechanism under the multiple actions of the blasting stress wave (BSW) were analyzed firstly. Then, the simulation experiment of coal damage accumulation was designed and carried out; during the experiment, the ultrasonic velocity of the samples was measured and the damage values were calculated. Under the multiple actions of BSW, the initial radial cracks around the blast-hole wall or in the area near the explosion source are formed under the action of tangential tension of BSW and the internal pressure of the blast-hole, and under the combined actions of stress wave, gas, and in situ stress, the primary cracks in the middle and far area of the explosion source expand and form the mesoscopic cracks because of the damage accumulation at the crack tip. And then the mesoscopic cracks grow and intersect to form a macroscopic crack. Finally, the macroscopic cracks expand and intersect with the initial radial cracks, the samples break suddenly under the load far below their fracture toughness because of the damage accumulation effects. A very challenging but meaningful task in geodynamic research is determining how to clearly disentangle and quantify the frequency-dependent barometric responses of borehole dilatometers. However, we still lack a fundamental understanding of how the Chinese TJ-2 borehole dilatometer network has responded to barometric pressure at different frequencies since the late 1990s. For this reason, we systematically analyze the transfer functions of volumetric strain changes associated with barometric pressure at 21 stations. The results demonstrate that the barometric responses for most sites are generally flat and near-flat in the low- (0.1–0.5 cpd) and high-frequency (8–30 cpd) bands, respectively, while drastically fluctuating in the intermediate-frequency band (0.5–8 cpd) due to the tidal influences. Overall, many sites exhibit strongly frequency-dependent behaviors over the whole frequency range (0.1–30 cpd), which are likely controlled mainly by the complex properties of sensor-grout-formation systems. In light of both the low- and high-frequency barometric responses of dilatometers, whether the surrounding formations are linear elastic medium can be verified. The mechanical properties of three stations, with surrounding formations that are thought to be linear elastic structures, can therefore be roughly but quantitatively estimated by utilizing barometric responses, which are shown to be consistent with the parameters tested by rock mechanics. Altogether, this work not only provides a useful method for frequency-dependent pressure correction, but also offers profound insights to advance sensor-grout-formation modeling. Oleanane-type ginsenosides are a class of compounds with remarkable pharmacological activities. However, the lack of effective preparation methods for specific rare ginsenosides has hindered the exploration of their pharmacological properties. In this study, a novel glycoside hydrolase PlGH3 was cloned from Paenibacillus lactis 154 and heterologous expressed in Escherichia coli. Sequence analysis revealed that PlGH3 consists of 749 amino acids with a molecular weight of 89.5 kDa, exhibiting the characteristic features of the glycoside hydrolase 3 family. The enzymatic characterization results of PlGH3 showed that the optimal reaction pH and temperature was 8 and 50 °C by using p-nitrophenyl-β-d-glucopyranoside as a substrate, respectively. The Km and kcat values towards ginsenoside Ro were 79.59 ± 3.42 µM and 18.52 s−1, respectively. PlGH3 exhibits a highly specific activity on hydrolyzing the 28-O-β-d-glucopyranosyl ester bond of oleanane-type saponins. The mechanism of hydrolysis specificity was then presumably elucidated through molecular docking. Eventually, four kinds of rare oleanane-type ginsenosides (calenduloside E, pseudoginsenoside RP1, zingibroside R1, and tarasaponin VI) were successfully prepared by biotransforming total saponins extracted from Panax japonicus. This study contributes to understanding the mechanism of enzymatic hydrolysis of the GH3 family and provides a practical route for the preparation of rare oleanane-type ginsenosides through biotransformation. An electrochemiluminescence (ECL) bioassay with high sensitivity and anti-fouling ability was developed for determination of matrix metalloproteinase 9 (MMP-9) secreted from living cells under external stimulation. A peptide with sequence of CLGRMGLPGK and a new cyclometalated iridium(III) complex bearing carboxyl group, (pq)2Ir(dcbpy) (pq = 2-phenylquinoline, dcbpy = 2,2′-bipyridyl-4,4′-dicarboxyli acid, abbreviated as Ir) were employed as molecular recognition substrate and ECL emitter, respectively. The peptide was labelled with the Ir to form Ir-peptide as ECL probe. Ir-peptide was self-assembled onto Nafion and gold nanoparticles (AuNPs) modified glassy carbon electrode (AuNPs/Nafion/GCE) and then both of 6-mercapto-1-hexanol (MCH) and zwitterionic peptide as blocking reagents were co-assembled on Ir-peptide/AuNPs/Nafion/GCE to form an anti-fouling ECL peptide-based biosensor. MMP-9 can be quantified in the range 1.0–50 ng·mL−1 with a detection limit of 0.50 ng·mL−1 based on the decreased ECL intensity. Relative standard derivation was 2.3% for six fabricated anti-fouling ECL peptide-based biosensors after reaction with 50 ng·mL−1 MMP-9. The anti-fouling ECL peptide-based biosensor can be used to monitor MMP-9 secreted from living cells under external stimulation. 96.0%–108.0% of recoveries were obtained in 60-diluted cell culture media. This study demonstrates that the ECL biosensor by the combination of iridium(III) complex-based sensitive ECL method and the anti-fouling interface provides a promising way for the determination of MMP-9 in biological sample, which is viable in clinical diagnosis and point-of-care test of protease. Based on CT and MRI images acquired from normal pressure hydrocephalus (NPH) patients, using machine learning methods, we aim to establish a multimodal and high-performance automatic ventricle segmentation method to achieve an efficient and accurate automatic measurement of the ventricular volume. First, we extract the brain CT and MRI images of 143 definite NPH patients. Second, we manually label the ventricular volume (VV) and intracranial volume (ICV). Then, we use the machine learning method to extract features and establish automatic ventricle segmentation model. Finally, we verify the reliability of the model and achieved automatic measurement of VV and ICV. In CT images, the Dice similarity coefficient (DSC), intraclass correlation coefficient (ICC), Pearson correlation, and Bland–Altman analysis of the automatic and manual segmentation result of the VV were 0.95, 0.99, 0.99, and 4.2 ± 2.6, respectively. The results of ICV were 0.96, 0.99, 0.99, and 6.0 ± 3.8, respectively. The whole process takes 3.4 ± 0.3 s. In MRI images, the DSC, ICC, Pearson correlation, and Bland–Altman analysis of the automatic and manual segmentation result of the VV were 0.94, 0.99, 0.99, and 2.0 ± 0.6, respectively. The results of ICV were 0.93, 0.99, 0.99, and 7.9 ± 3.8, respectively. The whole process took 1.9 ± 0.1 s. We have established a multimodal and high-performance automatic ventricle segmentation method to achieve efficient and accurate automatic measurement of the ventricular volume of NPH patients. Energy sorghum tolerates adverse climatic and edaphic conditions and has great potential as biofuel feedstock in marginal land. This study investigates the potential energy sorghum biomass production and uptake of nitrogen (N), phosphorus (P), and potassium (K) on a sandy loam marginal land in a semi-arid region, in order to define optimum N fertilizer rate to produce the highest biomass yield with minimal nutrient elimination. Five N rate treatments (0, 60, 120, 180, and 240 kg ha) and two sorghum varieties (sweet type Guotian-8 (GT-8) and biomass type Guoneng-11 (GN-11)) were used. Yield increment was observed as N level increased, but the standout treatment appeared to be N rate of 60 kg ha which significantly increased biomass yield vs. controls by 68.8% in 2014 and 64.1% in 2015. Biomass yield exhibited non-significant differences between N rate treatments from 60 to 240 kg ha, although the highest biomass yield (9.2-11.9 t ha) was observed in the 120 kg N ha treatment. Nutrient analysis showed that N, P, and K accumulation in aboveground plants increased with N rate increase, ranging between 32.2 and 119.1, 7.9 and 19.2, and 22.1 and 94.0 kg ha, respectively, for the highest N rate of 240 kg ha. Substantial amounts of N were extracted from the soil in control and 60 kg N ha treatments, despite the low fertility and organic matter content of the soil. Moreover, nitrogen (N) use efficiency (NUE) was maximized at lower N rates. A decline in physiological N use efficiency (PNUE) resulted in decreased agronomic N use efficiency (ANUE) at higher N rates. Hence, it is concluded that N fertilizer rate between 60 and 120 kg ha would be the optimal N requirement to facilitate sustainable production of energy sorghum on a sandy wasteland. Purpose: We examined associations between childhood temperamental activity, physical activity (PA), and television (TV) viewing over a 30-year period. Method: The participants (1220 boys and 1237 girls) were aged 3, 6, 9, and 12 years in 1980 and were followed until 2011. Temperamental activity was evaluated by participants' mothers at baseline. The PA was assessed based on maternal ratings of the child from ages 3 to 6 and via self-report age from the age of 9 across all measurements. TV viewing was assessed using self-reports taken from 2001 to 2011. The associations between temperamental activity and the level and change of PA and TV viewing were determined using linear growth modeling stratified by gender and age group. Results: High temperamental activity assessed from ages 9 to 12 was associated with high levels of childhood PA in both genders, but with a steeper decline in PA levels during the first 9 years of follow-up in boys. High temperamental activity assessed from ages 3 to 6 was associated with the decline of PA from childhood to youth in girls. High childhood temperamental activity was associated with decreased levels of PA in adulthood in men, but not in women. The associations between childhood temperamental activity and TV viewing during adulthood seemed to be positive but not consistently significant in all age and gender groups. Conclusion: High temperamental activity may contribute to the development of a physically inactive lifestyle. This study examined whether active commuting to school in childhood and adolescence predicted active commuting to work and overall physical activity (PA) in adulthood. Participants from the Young Finns Study (N = 2436) were aged 9–18 years in 1980 and followed up until 2018/2020. Their commuting modes to school were assessed with a self-reported questionnaire in 1980. Adulthood PA was assessed through self-reports regarding commuting modes to work (2001–2018), leisure-time physical activity (LTPA) (2001–2018), and objectively measured daily steps (2007–2018/2020). Associations between childhood commuting and adulthood PA were evaluated using regression analyses and multilevel models. Demographic, socioeconomic and environmental covariates were adjusted for in the analyses. Active commuting to school in childhood contributed favourably to LTPA in 2001 (B =.38, p <.001), in 2007 (B =.35, p <.001), and in 2018 (B =.28, p <.01). Active commuting in childhood was associated with higher number of daily aerobic steps (B = 299.00, p =.03) and daily aerobic steps during weekdays in 2011 (B = 312.15, p =.03). In 2018, active commuting associated favourably with daily aerobic steps (B = 370.42, p <.01), daily aerobic steps during weekdays (B = 347.65, p =.01), daily steps during weekends (B = 628.49, p =.02), and daily aerobic steps during weekends (B = 402.69, p =.03). Covariate adjustments attenuated the associations excluding the one between active commuting and LTPA in 2007 (B =.36, p =.01) and daily steps during weekends in 2018 (B = 782.25, p =.04). Background: Although previous studies have associated physical activity (PA) with lower depressive symptoms, the combined effects of the (1) frequency, (2) intensity, and (3) duration of long-term PA have not been examined in detail. Purpose: We examined the dose-response association between changes in frequency, intensity, and duration of PA and depressive symptoms in men and women over 6 years. Methods: Participants comprised 1,959 healthy adults (833 men and 1,126 women), aged 24-39 years in 2001, drawn from the ongoing Young Finns Study. PA was assessed using a self-report questionnaire completed in connection with a medical examination in 2001 and 2007. Depressive symptoms were simultaneously assessed using a modified version of Beck's Depression Inventory in both phases. Results: High doses of PA at baseline were prospectively associated with fewer depressive symptoms in men, while moderate doses of PA at baseline were inversely associated with the prevalence of depressive symptoms in women. Associations between baseline PA and depressive symptom changes were mediated by social and health-related factors which differed between men and women. Long-term participation in regular PA in all dimensions remained remarkably stable (all p < 0.001). Compared to those who remained inactive, the persistently active participants in all dimensions, with the exception of women's intensity group, were more likely to show decreases in depressive symptoms independent of the included confounders. An increase in PA in certain groups was also independently associated with fewer depressive symptoms, particularly in women. Conclusions: Regular and persistent participation in different doses of PA may provide short-term and long-term beneficial effects on depressive symptom changes. Background: Research on the long-term effects of Type A behavior and its components in the prediction of physical activity in adulthood is scarce and there is a lack of prospective data that are able to show such an association. Purpose: We examined the relations between components of Type A behavior and physical activity from youth to early midlife. Method: The sample included 2,031 participants (43.8% of males) aged 9 to 24 years in 1986 from the Young Finns Study. Type A behavior was measured by the Hunter-Wolf A-B Rating Scale at three phases in 1986, 1989, and 2001. Physical activity was assessed using a short self-report questionnaire at five phases between 1986 and 2007. Results: High Type A leadership was associated with high physical activity in 1986 ( r = 0.37, P < 0.01), 1989 ( r = 0.36, P < 0.01) and 2001 ( r = 0.31, P < 0.01), and youth leadership also predicted high adult physical activity ( P < 0.001). After adjustment for age, education, occupation, smoking, body mass index, and baseline physical activity, the association remained significant. There was also a bidirectional association between Type A leadership and physical activity. Persistent physical activity during the adult years was associated with a higher Type A leadership than persistent physical inactivity (Cohen's d = 0.34, P < 0.001), even after controlling for potential confounders. The associations of other components of Type A behavior, i.e., hard-driving, eagerness-energy, and aggression with physical activity were marginal. Elevation of plasma glucose concentration may induce generation of oxygen-free radicals, which can play an important role in the progression of diabetes and/or development of its complications. Various glutathione transferases utilize the availability of reduced glutathione for the cellular defense against oxygen-free radicals. One such enzyme is microsomal glutathione S-transferase 3 encoded by MGST3, which maps to chromosome 1q23, a region linked to Type 2 diabetes mellitus (T2DM) in Pima Indians, Caucasian, and Chinese populations. We investigated the MGST3 gene as a potential susceptibility gene for T2DM by screening this locus for single nucleotide polymorphisms (SNPs) in diabetic and non-diabetic Pima Indians. We also measured the skeletal muscle MGST3 mRNA level by Real-Time (RT) PCR and its relationship with insulin action in non-diabetic individuals. We identified 25 diallelic variants, most of which, based on their genotypic concordance, could be divided into three distinct linkage disequilibrium (LD) groups. We genotyped unique representative SNPs in selected diabetic and non-diabetic Pima Indians and found no evidence for association with T2DM. Furthermore, inter-individual variation of skeletal muscle MGST3 mRNA was not correlated with differences in insulin action in non-diabetic subjects. Purpose: The survival benefits of neoadjuvant chemotherapy (NAC) compared with those of concurrent chemoradiotherapy (CRT) for locally advanced cervical cancer (LACC) patients remain uncertain. Meta-analysis was used to compare NAC and CRT. Methods: A systematic search was performed up to 9 September 2020. Survival outcomes were analyzed based on event frequency or hazard ratios (HRs). Multilevel mixed-effects logistic regression was applied to analyze the effect of regimen variables on survival outcomes. Results: Analysis based on Cox regression showed that CRT was better than NAC + radical hysterectomy (RT) (HR 1.25; 95% confidence interval (CI)) 1.02–1.54; p = 0.034) in terms of overall survival (OS). According to multilevel mixed-effects model analysis comparing NAC + RT and CRT, LACC patients who used cisplatin instead of carboplatin had a better Progression-free survival (PFS) (odds ratio (OR) 1.54; 95% CI 1.08–2.20; p = 0.016). When NAC + CRT and CRT were compared, gemcitabine administration was associated with a decrease in PFS (OR 0.47; 95% CI 0.22–0.99; p = 0.047). Increased doses of cisplatin and paclitaxel were associated with survival improvement. Conclusion: Based on traditional meta-analysis, CRT was better than NAC + RT in terms of OS. Carboplatin instead of cisplatin as part of the NAC + RT strategy or gemcitabine use in NAC + CRT may not be a good choice. An increased total dosage of paclitaxel and/or cisplatin as part of NAC + CRT and CRT strategies may improve the survival outcome of LACC patients. [ABSTRACT FROM AUTHOR]

China is a major producer of wheat, maize and rice, which generates massive greenhouse gas emissions due to the consumption of elevated amounts of energy, land, and water, yet there is actually few knowledge on the spatial heterogeneity of greenhouse gas emissions. Here, we introduce a polyphasic approach to quantify the emissions from the land, energy, and water in Chinese cereal production. Results show that total greenhouse gas emissions from wheat, maize and rice crops reached 505.5 Tg CO2eq, including 247.5 Tg CO2eq for the land, 222.1 Tg CO2eq for energy, and 35.9 Tg CO2eq for water. The major greenhouse gas contributors are the land, of 55.8%, and energy to produce nitrogen fertilizers, of 30.5%. Moreover, greenhouse gas emissions from cereal production show large-scale spatial heterogeneity at the provincial scale due to differences in crop type, natural resources, and managerial practices. The collective adoption of green technologies and reasonable food trading practices could reduce greenhouse gas emissions from the cereal production supply chain, further promoting the carbon neutrality of agriculture. Objectives: To explore the value of structural neuroimaging in predicting the prognosis of shunt surgery for idiopathic normal-pressure hydrocephalus (iNPH) using two different standard semi-quantitative imaging scales. Methods: A total of 47 patients with iNPH who underwent shunt surgery at our hospital between 2018 and 2020 were included in this study. The modified Rankin Scale (mRS) and iNPH grading scale (iNPHGS) were used to evaluate and quantify the clinical symptoms before and after shunt surgery. The disproportionately enlarged subarachnoid space hydrocephalus (DESH) and iNPH Radscale scores were used to evaluate the preoperative MR images. The primary endpoint was improvement in the mRS score a year after surgery, and the secondary endpoint was the iNPHGS after 1 year. The preoperative imaging features of the improved and non-improved groups were compared. Results: The rates of the primary and secondary outcomes were 59.6% and 61.7%, respectively, 1 year after surgery. There were no significant differences in preoperative DESH score, iNPH Radscale, Evans' index (EI), or callosal angle (CA) between the improved and non-improved groups. Significant correlations were observed between the severity of gait disorder and EI and the CA. Conclusions: The value of structural neuroimaging in predicting the prognosis of shunt surgery is limited, and screening for shunt surgery candidates should not rely only on preoperative imaging findings. The hard-shelled character of ordinary cultivated Tartary buckwheat has become a factor influencing its taste and nutritional efficacy. However, the local variety, Rice-Tartary, can dehull easily. The genetic mechanism regulating easily-shelled Fagopyrum tataricum is unknown. In this study, the F2 generation segregating population was constructed by crossing Yunqiao No.1 (hard-shelled) and Rice-Tartary (easily-shelled) as parents, and the bulked segregant analysis sequencing (BSA-seq) strategy was used to initial mapping. The gene locus controlling the easily-shelled trait of Tartary buckwheat was preliminarily located in the 4.07 Mb region of the first chromosome. To further narrow the range, the Kompetitive allele-specific PCR primers based on the single nucleotide polymorphisms in the initial location range were designed and tested in 335 individual plants in the hybrid F2 population. The candidate gene Ftes1 was located between Ft6,705,225 and Ft7,041,921, according to the genetic linkage map constructed based on typing data. Combined with the location of candidate genes, the RNA-Seq data, and qRT-PCR results, the results indicate that 3 genes, FtPinG0001427400.01, FtPinG0001428600.01, and FtPinG0002492200.01 may regulate the easily-shelled Tartary buckwheat. The results of this study provide vital candidate genes for the cloning and functional analysis of genes related to easily-shelled traits, as well as prominent molecular markers for breeding of new varieties of Tartary buckwheat. Research on drugs against SARS-CoV-2 (cause of COVID-19) has been one of the major world concerns at present. There have been abundant research data and findings in this field. The interference of drugs on gene expression in cell lines, drug-target, protein-virus receptor networks, and immune cell infiltration of the host may provide useful information for anti-SARS-CoV-2 drug research. To simplify the complex bioinformatics analysis and facilitate the evaluation of the latest research data, we developed OmiczViz (http://medcode.link/omicsviz), a web tool that has integrated drug-cell line interference data, virus-host protein–protein interactions, and drug-target interactions. To demonstrate the usages of OmiczViz, we analyzed the gene expression data from cell lines treated with chloroquine and ruxolitinib, the drug-target protein networks of 48 anti-coronavirus drugs and drugs bound with ACE2, and the profiles of immune cell infiltration between different COVID-19 patient groups. Our research shows that chloroquine had a regulatory role of the immune response in renal cell line but not in lung cell line. The anti-coronavirus drug-target network analysis suggested that antihistamine of promethaziney and dietary supplement of Zinc might be beneficial when used jointly with antiviral drugs. The immune infiltration analysis indicated that both the COVID-19 patients admitted to the ICU and the elderly with infection showed immune exhaustion status, yet with different molecular mechanisms. The interactive graphic interface of OmiczViz also makes it easier to analyze newly discovered and user-uploaded data, leading to an in-depth understanding of existing findings and an expansion of existing knowledge of SARS-CoV-2. Lateral flow immunostrips were newly designed and a sensitive and rapid fluorometric method for the determination of 8-hydroxy-2′-deoxyguanosine (8-OHdG) as a model target of small biomarker molecules was developed. The upconversion nanoparticles (UCNPs, NaYF4:Yb/Er core, and polyacrylic acid (PAA)-modified shell, size ~ 39 nm, excitation wavelength = 980 nm; emission wavelength = 540 nm) were employed as fluorescence signal material. The 8-OHdG antibody (Ab) was taken as the recognition probe while UCNP-labeled Ab was taken as the signal probe. Bovine serum albumin (BSA) was designed as carrier protein for 8-OHdG to form 8-OHdG-BSA conjugate as the capture probe. The lateral flow immunostrips were prepared by laminating a sample pad (glass fiber membrane), a test pad (nitrocellulose membrane), and adsorption pad (filter paper) on PVP backing. The capture probe was immobilized on the test zone while an IgG antibody taken as the control probe was immobilized on the control zone. When the signal probe and the sample were in sequence loaded on the sample pad, 8-OHdG analyte bound with the signal probe, and then the excess of the signal probe move along the strip and is collected by the capture probe on the test zone while the remnant signal probe is collected by the control probe on the control zone. The signal probe and capture probe were synthesized and characterized. The fluorescence intensity on the test zone was inversely proportional to the concentration of 8-OHdG for the quantitative determination while the fluorescence emission on the control zone was observed to validate the assay. The developed method showed a wide linear range from 0.10 to 10 nM, a quite low detection limit of 0.05 nM, small sample volume requirement (100 μL), short assay time (15 min), and good method reproducibility (RSD = 4.4%, nine immunostrips). A homogeneous electrogenerated chemiluminescence (ECL) immunoassay for highly sensitive quantification of specific biomarkers based on immunomagnetic beads and homogeneous detection on a magnetic electrode was developed, for the first time. The magnetic electrode is made of a glassy carbon electrode and a series of ring permanent magnets. D-Dimer antigen was taken as a model analyte while biotinylated D-dimer antibody bound on the streptavidin-coated magnetic beads was utilized as a magnetic capture probe and ruthenium complex-labeled D-dimer antibody was employed as an ECL probe. After a fixed amount of magnetic capture probe and the ECL probe was introduced into analyte D-dimer solution, the "sandwich" immunoconjugates on the magnetic beads were formed in tested solution and then magnetically concentrated on the surface of the magnetic electrode. The homogeneous ECL immunoassay for quantification of specific biomarker was directly carried out in the presence of co-reactant tripropylamine. The low detection limit of 1 ng/mL in magnetic enrich time of 2 min and the good magnetic regeneration for the detection of D-dimer were achieved. The magnetic bead shield ECL emission was extensively discussed. This work demonstrates that the homogeneous (separation-free) ECL immunoassay using magnetic beads and magnetic electrode is a promising approach to quantify the biomarkers with high sensitivity and selectivity and in a short time. While searching for natural anti-hepatocellular carcinoma (HCC) components in Ailanthus altissima, we discovered that ailanthone had potent antineoplastic activity against HCC. However, the molecular mechanisms underlying the antitumor effect of ailanthone on HCC have not been examined. In this study, the antitumor activity and the underlying mechanisms of ailanthone were evaluated in vitro and in vivo. Mechanistic studies showed that ailanthone induced G0/G1-phase cell cycle arrest, as indicated by decreased expression of cyclins and CDKs and increased expression of p21 and p27. Our results demonstrated that ailanthone triggered DNA damage characterized by activation of the ATM/ATR pathway. Moreover, ailanthone-induced cell death was associated with apoptosis, as evidenced by an increased ratio of cells in the subG1 phase and by PARP cleavage and caspase activation. Ailanthone-induced apoptosis was mitochondrion-mediated and involved the PI3K/AKT signaling pathway in Huh7 cells. In vivo studies demonstrated that ailanthone inhibited the growth and angiogenesis of tumor xenografts without significant secondary adverse effects, indicating its safety for treating HCC. In conclusion, our study is the first to report the efficacy of ailanthone against Huh7 cells and to elucidate its underlying molecular mechanisms. Lymph node metastasis is of major prognostic significance for breast cancer. Lymph node metastasis arises at a very early stage in some patients. Using the data downloaded from the TCGA database, we studied the differences between primary tumors with and without lymph node metastasis at the multi-omics level using bioinformatics approaches. Our study found that low mutation and neoantigen burdens correlated with lymph node metastazation of breast cancer. All three conserved domains in TP53 were mutated in lymph node-negative breast cancers, whereas only one domain was mutated in lymph node-positive samples. Mutations in microtubule-related proteins appear to help immune cells recognize tumors and inhibit their lymph node metastasis. Destroying microtubule-related proteins is a potential therapeutic strategy to inhibit lymph node metastasis of breast cancer. As the neoantigens specifically present in lymph node-positive breast cancers, MAPK10, BC9L, TRIM65, CD93, KITLG, CNPPD1, CPED1, CCDC146, TMEM185A, INO80D, and PSMD11 are potential targets for vaccine design. In the tumor microenvironment, reduced numbers of effector immune cells, especially activated memory CD4+ T cells and activated mast cells, facilitate breast cancer metastasis to the lymph nodes. According to transcriptome data, lymph node metastasis was mostly driven by gene mutation rather than by gene expression.
