Aging is frequently associated with compromised cerebrovasculature and pericytes. However, we do not know how normal aging differentially impacts vascular structure and function in different brain areas. Here we utilize mesoscale microscopy methods and in vivo imaging to determine detailed changes in aged murine cerebrovascular networks. Whole-brain vascular tracing shows an overall ~10% decrease in vascular length and branching density with ~7% increase in vascular radii in aged brains. Light sheet imaging with 3D immunolabeling reveals increased arteriole tortuosity of aged brains. Notably, vasculature and pericyte densities show selective and significant reductions in the deep cortical layers, hippocampal network, and basal forebrain areas. We find increased blood extravasation, implying compromised blood-brain barrier function in aged brains. Moreover, in vivo imaging in awake mice demonstrates reduced baseline and on-demand blood oxygenation despite relatively intact neurovascular coupling. Collectively, we uncover regional vulnerabilities of cerebrovascular network and physiological changes that can mediate cognitive decline in normal aging. We have limited knowledge of how aging affects brain vascular structure and function. Continuous hemodynamic monitoring in a wearable means can play a crucial role in managing hypertension and preventing catastrophic cardiovascular events. In this study, we have described the fully wearable tonometric device, referred to as flexible adaptive sensing tonometry (FAST), which is capable of continuous and accurate monitoring of hemodynamic parameters within the medical-grade precision. In particular, the FAST system integrates a 1 × 8 unit array of highly sensitive and highly flexible iontronic sensing (FITS) with 1 mm spatial resolution and a closed-loop motion system. The flexible tonometric architecture has been used to determine the radial arterial position with high sensitivity and high conformability, which simplifies the biaxial searching process of the traditional applanation tonometry into a highly efficient uniaxial applanation while keeping the medical-precision assessments. Importantly, a self-calibration algorithm can be automatically implemented during the applanation process, from which the intra-arterial blood pressure wave can be continuously predicted within the medical-grade precision, and subsequently, multi-parametric hemodynamic analysis can be performed in real-time. Experimental validations on health volunteers have demonstrated that the FAST measurements are all within the required accuracy of the clinical standards for continuous pulse wave assessments, blood pressure monitoring as well as other key hemodynamic parameter evaluations. Therefore, the FAST system, by integrating the flexible iontronic sensing array, provides a real-time, medical-grade hemodynamic monitoring solution in a continuously wearable manner, from which remote patient-centered monitoring can be delivered with both medical precision and convenience. Heart failure continues to be a significant global health concern, causing substantial morbidity and mortality. The limited ability of the adult heart to regenerate has posed challenges in finding effective treatments for cardiac pathologies. While various medications and surgical interventions have been used to improve cardiac function, they are not able to address the extensive loss of functioning cardiomyocytes that occurs during cardiac injury. As a result, there is growing interest in understanding how the cell cycle is regulated and exploring the potential for stimulating cardiomyocyte proliferation as a means of promoting heart regeneration. This review aims to provide an overview of current knowledge on cell cycle regulation and mechanisms underlying cardiomyocyte proliferation in cases of heart failure, while also highlighting established and novel therapeutic strategies targeting this area for treatment purposes. In this work, a hot extrusion forming process for magnesium (Mg) alloy square tubes, namely, tube-extrusion-continuous expending shear (TECES), was proposed. The thin-walled hollow square tube of Mg alloy with a thickness of 3 mm was successfully prepared by hot extrusion experiment. The evolution rules of effective stress, effective strain, and flow velocity in the process of tube forming were analyzed by numerical simulation, and the influence of different expanding ratios on tube forming was compared. The results show that the effective stress distribution of the tube is not uniform during the forming process, and the maximum stress appears at the first shear corner. In the shear deformation (SD) zone, there is a high strain zone at the corner of the square tube and outside the tube wall. As the tube is extruded, the uniformity of strain distribution in the deformation zone is improved. In the whole forming process, the flow rate of the tube increases first and then decreases, and the whole remains stable. As the expansion ratio increases when the expansion ratio is greater than 1.6, the resistance of plastic deformation increases, the fluidity of the metal becomes worse, and the formed tube cracks at the corner or is difficult to fill. When the expansion ratio is less than 1.6, the damage value area appears on the inner side of the tube wall, and the distribution uniformity is poor, which may lead to cracks or uneven thickness of the tube wall. The microstructure characterization of different deformation zones of the tube was carried out, and the results show that the TECES process can effectively refine the grain, and the average grain size of the formed tube is 10.1 μm. Mulberry bacterial wilt is a devastating disease that is difficult to control and causes serious economic losses to the sericulture industry. This disease is mostly caused by a diverse group of pathogenic and opportunistic bacteria including, Ralstonia pseudosolanacearum, Pantoea ananatis, Enterobacter cloacae complex (ECC), Klebsiella pneumoniae species complex (KpSC), and K. oxytoca complex (KoC). Due to the lack of a rapid and reliable test to simultaneously detect these complex pathogens of mulberry wilt, we developed a multiplex PCR (mPCR) assay to detect five virulence-related genes carried by the pathogenic bacteria of mulberry bacterial wilt disease. The primers were designed for the virulence-related genes: pleD (GGDF structural domain-containing protein), yjfP (esterase), pelY (peripheral pectate lyase), ampD (N-acetyl-anhydromuranmyl-L-alanine amidase), and ripW (type III effector). Overall, the developed mPCR assay showed highly specific, sensitive and reproducible detection of target pathogens. Briefly, the results showed that the mPCR was highly specific in individual reactions, and the lowest detection concentration of the five pathogenic bacteria was 1.87 × 103 CFU/mL (DNA = 2.45 pg/μL). From 46 natural mulberry wilt samples, the mPCR detection rates of P. ananatis, ECC, KpSC, KoC and R. pseudosolanacearum were 8.69, 91.3, 34.7, 23.9 and 65.21%, respectively. The traditional culture media isolation methods showed comparable results. The pathogenicity test of 84 suspected pathogenic bacteria revealed that the morbidity (average morbidity level) caused by the pathogenic bacteria detected by mPCR was ≥ 65.5%, while the morbidity of the undetected pathogenic bacteria was ≤ 35.5%. Based on these results, we believe that the mPCR developed in the present study will be useful in rapid, reproducible, and sensitive detection of the pathogenic bacteria causing mulberry bacterial wilt including, R. pseudosolanacearum, P. ananatis, ECC, KpSC, and KoC. To investigate the screening and predicting functions of obesity- and lipid-related indices for type 2 diabetes (T2D) in middle-aged and elderly Chinese, as well as the ideal predicted cut-off value. This study's data comes from the 2011 China Health and Retirement Longitudinal Study (CHARLS). A cross-sectional study design was used to investigate the relationship of T2D and 13 obesity- and lipid-related indices, including body mass index (BMI), waist circumference (WC), waist–height ratio (WHtR), visceral adiposity index (VAI), a body shape index (ABSI), body roundness index (BRI), lipid accumulation product (LAP), conicity index (CI), Chinese visceral adiposity index (CVAI), triglyceride- glucose index (TyG index) and its correlation index (TyG-BMI, TyG-WC, TyG-WHtR). The unadjusted and adjusted correlations between 13 indices and T2D were assessed using binary logistic regression analysis. The receiver operating characteristic curve (ROC) was used to determine the usefulness of anthropometric indices for screening for T2D and determining their cut‑off value, sensitivity, specificity, and area under the curve (AUC). The study comprised 9488 people aged 45 years or above in total, of whom 4354 (45.89%) were males and 5134 (54.11%) were females. Among them were 716 male cases of T2D (16.44%) and 870 female cases of T2D (16.95%). A total of 13 obesity- and lipid-related indices were independently associated with T2D risk after adjusted for confounding factors (P < 0.05). According to ROC analysis, the TyG index was the best predictor of T2D among males (AUC = 0.780, 95% CI 0.761, 0.799) and females (AUC = 0.782, 95% CI 0.764, 0.799). The AUC values of the 13 indicators were higher than 0.5, indicating that they have predictive values for T2D in middle-aged and elderly Chinese. The 13 obesity- and lipid-related indices can predict the risk of T2D in middle‑aged and elderly Chinese. Among 13 indicators, the TyG index is the best predictor of T2D in both males and females. This study proposed the direct extrusion and continuous-shear deformation (DECS) process for the co-extrusion of Mg-AZ31B/Al 6063, aiming to prepare high-plasticity Mg/Al composite sheets. The effect of extrusion temperature on the microstructure and mechanical properties of the Mg/Al composite sheet was systematically studied. The results show that Mg/Al composite sheets with no voids or cracks at the interface were successfully prepared by the DECS process. The microstructure of the Mg layer at different extrusion temperatures primarily consists of equiaxed fine grains, with grain sizes of 7.29 μm, 8.98 μm, and 9.88 μm at 330℃, 360℃, and 390℃, respectively. The Mg layer exhibits a typical extrusion texture, with the c-axis of most grains parallel to the transverse direction (TD). With the increase in extrusion temperature, the c-axis of some grains deviates from the TD toward the normal direction (ND). The Al layer mainly consists of elongated deformed grains and fine dynamically recrystallized (DRXed) grains distributed along the extrusion direction (ED). The AZ31 Mg alloy with refined grains maintains the ultimate tensile strength of the Mg/Al composite sheet, while the Al matrix enhances its plasticity. At 330℃, the Mg/Al composite sheet shows an ultimate tensile strength of 244.48 MPa ± 4.35 and an elongation of 39.42% ± 0.06. The influence of the bonding layer thickness on the shear and tensile properties of the composite sheet was studied, revealing that increasing the bonding layer thickness reduces the shear strength of the Mg/Al composite sheet but positively affects its tensile strength. Objective: We aimed to evaluate the association between dietary guideline adherence and overall, outpatient, and emergency medical service utilization in Taiwanese preschoolers. Methods: We selected 614 preschoolers (2–6 years) who had one day of 24-h dietary recall data from the 2013–2016 Nutrition and Health Survey in Taiwan. The Taiwanese Children Healthy Eating Index (TCHEI) was developed on the basis of Taiwanese Food-Based Dietary Guidelines; it assesses dietary adequacy and eating behavior. Data on the participants' outpatient and emergency medical service utilization were obtained for 2013–2018 from the National Health Insurance Research Database. A multivariable generalized linear model was used to evaluate the association between the TCHEI and medical service utilization for all disease and respiratory diseases. Results: After adjustment for confounding factors, children aged 2–3 years in the Tertile (T) 2 and T3 groups of the TCHEI exhibited 25% (95% CI 0.69–0.83) and 16% (95% CI 0.77–0.92) lower overall medical visits, respectively. The same pattern was noted in the outpatient and emergency visits for all diseases and respiratory diseases. The children aged 4–6 years in the T2 group exhibited 15% (95% CI 0.80–0.91) and 11% (95% CI 0.82–0.97) lower overall visits and visits for respiratory diseases, respectively. Moreover, preschoolers in the T2 group exhibited lower overall medical expenditures than did those in the T1 group. Conclusions: TCHEI score was positively correlated with better nutritional status. Eucommia ulmoides rubber particles (EuRPs) are organelles that synthesise and store E. ulmoides rubber (EuR). In this study, the lipids dynamic change of EuRPs from leaf tissue at different growth period were investigated. The results showed that there were 1149 lipid species belonging to 39 lipid classes that were identified; TGs, DGs and Cers were the most abundant among all lipids of EuRPs. Lipid expression analysis demonstrated that the expression intensity of TGs was almost 1 × 104-fold and 10-fold higher than those of MGs and DGs, respectively. Among sphingolipids, the expression intensity of SoP, CerG2, GM3, SM and GerG3 increased throughout the duration of investigation (August–November 2021). Further analysis found that the Cers species accounted for 9.40% of total lipid species and the number of Cers species in EuRPs isolated from leaves in August was the most, with gradual decline in the following 3 months. Meanwhile, hierarchical clustering indicated that there were seven types of highly abundant lipid species in leaf EuRPs from September to November, i.e. SQDG (18:3/18:3), SQDG (34:3), SQDG (17:5/23:2), DGDG (38:12), Co (Q9), TG (16:0/16:0/18:3) and TG (39:1). Compared with leaf EuRPs in August, there were 79, 101 and 107 lipid species which showed significant differential expression in September, October and November. Based on the high expression intensity of SQDG in EuRPs, the correlation between the genes mediated in EuR or SQDG biosynthesis was further analysed, whereby the results indicated co-expression of some genes, which implies the synergistic regulatory role in SQDG EuR biosynthesis. Social stability risk posed by energy infrastructure projects can seriously affect urban sustainable development. There is thus a need to better understand how risk strategies are designed. However, there is few considerations on risk strategy design by breaking causal relationships. This research innovatively explores social stability risk variables by content mining, develops causal relationships of the social stability risk variables via Fault Tree Analysis (FTA) method, explores core risk variables and critical causal relationships by social network analysis (SNA) method, designs and validates risk strategies. The findings show that: 1) From the overall risk network perspective, the risk network contain 8 core risk variables. 2) From individual risk network perspective, there are 75 critical causal relationships. The top 3 critical causal relationships contain: projects that destroy the cultural landscape would be regarded as threating national security, which often inspires demonstrations among the local people and the line betweenness is the largest (35.589). Traffic congestion by the project is the main reason to cause local small-scale public petition, and the line betweenness is 35.075. Projects that threaten the ecological environment often bring psychological rejection of the project by the local public, and the line betweenness is 30.837. 3) Two scenarios are evaluated in terms of basic scenario and the experiment group scenario. Compared with the basic scenario, risk strategies considering causal relationships have significant effectiveness. The overall risk network density has reduced by 35.22%. The clustering coefficients has decreased by 16.20%. Intermediate central potential has reduced by 9.49%. Cancer cachexia is characterized by weight loss and skeletal muscle wasting. Based on the up-regulation of catabolism and down-regulation of anabolism, here we showed genetic mutation-mediated metabolic reprogramming in the progression of cancer cachexia by screening for metabolites and investigating their direct effect on muscle atrophy. Treatment with 93 μM D-2-hydroxyglutarate (D2HG) resulted in reduced myotube width and increased expression of E3 ubiquitin ligases. Isocitrate Dehydrogenase 1 (IDH1) mutant patients had higher D2HG than non-mutant patients. In the in vivo murine cancer cachexia model, mutant IDH1 in CT26 cancer cells accelerated cachexia progression and worsened overall survival. Transcriptomics and metabolomics revealed a distinct D2HG-induced metabolic imbalance. Treatment with the IDH1 inhibitor ivosidenib delayed the progression of cancer cachexia in murine GL261 glioma model and CT26 colorectal carcinoma models. These data demonstrate the contribution of IDH1 mutation mediated D2HG accumulation to the progression of cancer cachexia and highlight the individualized treatment of IDH1 mutation associated cancer cachexia. IDH1 mutation mediated D2HG accumulation contributes to the progression of cancer cachexia and muscle atrophy. Falls are a serious public health problem in the aging population because of the associated clinical and socioeconomic impact. Although previous studies have investigated fall-risk-increasing drugs (FRIDs), few studies have focused on dosage among adult inpatients. This study aimed to evaluate associations between fall risk and dosage of different FRIDs classes in hospital inpatients. Inpatients who experienced falls at medical or surgical wards of Changhua Christian Hospital from January 2017 to December 2021 were identified and matched by age, sex, and hospital ward to randomly selected controls (four per case). Anonymous patient data were extracted from the hospital medical data repository, including demographic characteristics, comorbidities, fall-risk scores, and drug prescriptions. Medication dosages were computed using the anatomical therapeutic chemical classification and the defined daily dose system of the World Health Organization. A total of 852 cases and 3408 controls were identified as eligible. Reducing the use of CNS-active medications, administering lower doses of sedative-hypnotics, prescribing sufficient dopaminergic anti-Parkinson agents, and using NSAIDs instead of opioids are imperative in preventing falls among hospitalized patients according to the findings in the study. The hole fertilization is an effective means of saving fertilizer and increasing yields by applying the fertilizer needed for crop growth to a certain area below the seed during the seed fertilizing stage. In response to the current issues of low fertilizer utilization and soil non-point source pollution caused by strip fertilization, a hole-fertilizing corn planter was designed, which mainly consists of the hole-fertilizing unit, electro-driven seeding unit, and speed measuring device. A dynamic alignment control system with low cost was developed for intermittent fertilization and seeding as well as for precise alignment of the seed and fertilizer. By analyzing the movement process of the seed and fertilizer, a dynamic alignment algorithm was developed to dynamically adjust the alignment and precisely control the placement. In addition, a novel image-based fertilizer detection method was used to study the fertilizer distribution in the soil and evaluate the operating performance of the prototype in the field. Field trials were conducted to investigate the seeding quality, hole-fertilizing effect, and alignment precision of the seed and fertilizer at different operating speeds. Results indicate that (1) the feed index ( I qf ), miss index ( I miss ), and multiple index ( I mult ) are 91.9%, 6.8%, and 1.3%, respectively. (2) 85.4% of the fertilizer distances are greater than 60 mm in the soil, and the amount of fertilizer applied increases progressively with increasing soil depth. (3) The mean value of the offset distances is 28.1 mm, and 94% of the offset distances are less than 50 mm. Moreover, the offset distance increases as the operating speed rises. Metabolic dysfunction-associated fatty liver disease (MAFLD) is a new diagnostic criterion based on hepatic steatosis and metabolic dysfunction. However, a comprehensive evaluation of the association of MAFLD dynamic transitions with arterial stiffness progression has yet to be conducted. This cohort study included 8807 Chinese health check-up participants (median follow-up = 50.2 months). Participants were categorized into four groups according to MAFLD status at baseline and follow-up (none, persistent, developed and regressed). Arterial stiffness progression was assessed by the annual brachial-ankle pulse wave velocity (ba-PWV) increase and arterial stiffness incidence. Compared with the non-MAFLD group, the annual increase in ba-PWV was highest in the persistent-MAFLD group [6.75 cm/s/year, (95% CI 4.03–9.33)], followed by the developed—[6.35 cm/s/year, (95% CI 3.80–8.91)] and the regressed—[1.27 cm/s/year, (95% CI − 2.18 to 4.72)] MAFLD groups. Similarly, compared with the non-MAFLD group, the persistent-MAFLD group had a 1.31-fold increased arterial stiffness risk [OR 1.31; 95% CI 1.03–1.66]. The associations of MAFLD transition patterns with arterial stiffness incidence did not differ across any clinically specific subgroups evaluated. Furthermore, the potential effect of dynamic changes in cardiometabolic risk factors on arterial stiffness incidence among persistent-MAFLD participants was mostly driven by annual fasting glucose and triglyceride increases. In conclusion, persistent MAFLD was associated with an increased risk of arterial stiffness development. Endophytes have a wide range of potential in maintaining plant health and sustainable agricultural environmental conditions. In this study, we analysed the diversity of endophytic bacteria in four mulberry cultivars with different resistance capacity against bacterial wilt using metagenomic sequencing and culture-dependent approaches. We further assessed the role of 11 shared genera in the control of bacterial wilt of mulberry. The results of the present study showed that Actinobacteria, Firmicutes, and Proteobacteria were the three dominant phyla in all communities, with the representative genera Acinetobacter and Pseudomonas. The diversity analysis showed that the communities of the highly and moderately resistant varieties were more diverse compared to those of the weakly resistant and susceptible varieties. The control tests of mulberry bacterial wilt showed that Pantoea, Atlantibacter, Stenotrophomonas, and Acinetobacter were effective, with a control rate of over 80%. Microbacterium and Kosakonia were moderately effective, with a control rate between 50 and 80%. At the same time, Escherichia, Lysinibacillus, Pseudomonas, and Rhizobium were found to be less effective, with a control rate of less than 40%. Cardiovascular diseases (CVDs) are a leading cause of death worldwide. For early diagnosis, intervention and management of CVDs, it is highly desirable to frequently monitor blood pressure (BP), a vital sign closely related to CVDs, during people's daily life, including sleep time. Towards this end, wearable and cuffless BP extraction methods have been extensively researched in recent years as part of the mobile healthcare initiative. This review focuses on the enabling technologies for wearable and cuffless BP monitoring platforms, covering both the emerging flexible sensor designs and BP extraction algorithms. Based on the signal type, the sensing devices are classified into electrical, optical, and mechanical sensors, and the state-of-the-art material choices, fabrication methods, and performances of each type of sensor are briefly reviewed. In the model part of the review, contemporary algorithmic BP estimation methods for beat-to-beat BP measurements and continuous BP waveform extraction are introduced. Mainstream approaches, such as pulse transit time-based analytical models and machine learning methods, are compared in terms of their input modalities, features, implementation algorithms, and performances. The review sheds light on the interdisciplinary research opportunities to combine the latest innovations in the sensor and signal processing research fields to achieve a new generation of cuffless BP measurement devices with improved wearability, reliability, and accuracy. We aimed to investigate whether exosomes (Exo) affected chronic obstructive pulmonary disease (COPD) by influencing ferroptosis of bronchial epithelial cells (BECs) and the mechanisms involved. Here we took the peripheral blood samples of normal subjects and COPD patients, extracted and identified endothelial progenitor cells (EPCs) and EPC-Exo. An animal model of COPD was established. Then human BECs were taken and treated with cigarette smoke extract (CSE) for 24 h to construct a COPD cell model. Next, we screened differentially expressed ferroptosis-related genes in COPD patients by bioinformatics. Bioinformatics predicted the miRNA targeting PTGS2. Then, the mechanism of action of miR-26a-5p and Exo-miR-26a-5p was investigated in vitro. We successfully isolated and identified EPC and Exo. In vitro, EPC alleviated CSE-induced ferroptosis in BECs by transporting Exo. In vivo, Exo alleviated cigarette smoke-induced ferroptosis and airway remodeling in mice. Through further validation, we found that CSE-induced ferroptosis promoted the epithelial-mesenchymal transition (EMT) of BECs. Bioinformatics analysis and validation showed that PTGS2/PGE2 pathway affected CSE-induced ferroptosis in BECs. Meanwhile, miR-26a-5p targeting PTGS2 affected CSE-induced ferroptosis in BECs. Additionally, we found that miR-26a-5p affected CSE-induced BECs EMT. Exo-miR-26a-5p alleviated CSE-induced ferroptosis and EMT. In conclusion, EPC-exosomal miR-26a-5p improved airway remodeling in COPD by inhibiting ferroptosis of BECs via the PTGS2/PGE2 pathway. [ABSTRACT FROM AUTHOR]

The precise use of specific types of herbicides according to the weed species and density in fields can effectively reduce chemical contamination. A weed species and density evaluation method based on an image semantic segmentation neural network was proposed in this paper. A combination of pre-training and fine-tuning training methods was used to train the network. The pre-training data were images that only contain one species of weeds in one image. The weeds were automatically labeled by an image segmentation method based on the Excess Green (ExG) and the minimum error threshold. The fine-tuning dataset was real images containing multiple weeds and crops and manually labeled. Due to the limitation of computational resources, larger images were difficult to segment at one time. Therefore, this paper proposed a method of cutting images into sub-images. The relationship between sub-image size and segment accuracy was studied. The results showed that the training method reduced the workload of labeling training data while effectively avoiding overfitting. The accuracy of image segmentation decreases as the sub-image size decreases. Considering the limitation of computational resources, a subgraph of 256 × 256 size was selected. The semantic segmentation network achieved 97% overall accuracy. The coefficient of determination ( R 2 ) of weed density calculated by the algorithm and manually assessed was 0.90, and the root means square error (σ ) was 0.05. To quantitatively evaluate the impact of domestic aviation control measures on the spread of COVID-19 in China. The number of international flights from March to September 2019 simulated the number of flights from March to September 2020 without implementing aviation control measures. In addition, the proportion of asymptomatic persons and the delay in case reporting were adjusted to estimate the prevalence of each country during the same period and calculate the estimated imported cases. The estimated imported cases were assigned each day with weight, and the estimated daily reported cases were obtained based on the actual daily number of domestic cases in China. Effective Reproduction Number ( R t ) was calculated based on delayed distribution, Basic Reproductive Number ( R 0 ) distribution, and generation time distribution were reported in previous studies. Gaussian Process was used to estimate the effect of time-varying on R t , and the estimated R t was compared with the actual R t . The estimated imported cases increased significantly compared with the actual number of imported cases. The estimated imported cases were mainly concentrated in North America and Europe from March to April and gradually increased in many East Asian countries from May to September. The difference between predicted R t and actual R t was statistically significant. The estimated imported cases and the estimated R t have increased compared to the actual situation. This paper quantitatively proves that Chinese aviation control measures significantly suppress the COVID-19 epidemic, which is conducive to promoting and applying this measure. In this paper, we present an efficient numerical algorithm for evaluating the determinants of general bordered k-tridiagonal matrices in linear time. The algorithm is based on a novel incomplete block-diagonalization (IBD) approach which preserves the low-rank structure and sparsity of the original matrix, and a reliable algorithm for the determinants of general bordered tridiagonal matrices. An advantage of the proposed algorithm is that there is no division operation involved and hence the algorithm will never suffer from breakdown. Moreover, the algorithm theoretically produces exact values of the determinants when all entries of the bordered k-tridiagonal matrix are given in integer. Some numerical results with simulations in Matlab implementation are provided in order to illustrate the accuracy and effectiveness of the proposed algorithm, and its competitiveness with Gaussian elimination algorithm and MATLAB built-in function. Background: Extended curettage has increasingly become the preferred treatment for giant cell tumour of bone (GCTB), but the high recurrence rate after curettage poses a major challenge for orthopaedic surgeons. Computed tomography (CT) is valuable in the evaluation of GCTB. Our aim was to identify specific features of GCTB around the knee in pre-operative CT images that might have prognostic value for local recurrence. Methods: We retrospectively analyzed data from 124 patients with primary GCTB around the knee who underwent extended curettage from 2010 through 2019. We collected demographic, clinical, and therapeutic data along with several CT-derived tumour characteristics. CT-derived tumor characteristics included tumour size, the distance between the tumour edge and articular surface (DTA), and destruction of posterior cortical bone (DPC). Akaike information criterion (AIC) was used to select which variables to enter into multivariate logistic regression models and to determine significant factors affecting recurrence. Results: The total recurrence rate was 21.0% (26/124), and the average follow-up time was 69.5 ± 31.2 months (24–127 months). Age, DTA (< 2 mm), and DPC were significantly related to recurrence, as determined by multivariate logistic regression. The C-index of the final model was 0.79 (95% CI: 0.71 to 0.88), representing a good model for predicting recurrence. Conclusion: Identifying certain features of GCTB around the knee on CT has prognostic value for patients treated with extended curettage. Adsorption of secreted protein on contact lenses is a dynamic and complex behavior. To understand this behavior, we used three-dimensional (3D) printing technology to create an eye model that simulated the anterior segment of the actual human eyeball. In this model, the fluid inlet was connected to a syringe pump to mimic the rate of human tear secretion and the outlet was connected to an ultraviolet-visible (UV-Vis) spectrophotometer. The experimental results revealed that the symmetrical eye model with a 180° inlet-outlet angle was suitable for dynamic analysis of protein adsorption. In this model, protein adsorption was slow and desorption was rapid. The contact lens was soaked in poly(2-methacryloyloxyethyl phosphorylcholine-co-butyl methacrylate) (PMB) to confirm the anti-protein adsorption property of this polymer through dynamic adsorption and desorption eye model analysis. The persistence or recurrence of minimal residual disease (MRD) after chemotherapy predicts relapse of B-cell acute lymphoblastic leukemia (B-ALL). CD19-directed chimeric antigen receptor T (CD19 CAR-T) cells have shown promising responses in B-ALL. However, their role in chemotherapy-refractory MRD-positive B-ALL remains unclear. Here we aimed to assess the effectiveness and safety of CD19 CAR-T cells in MRD-positive B-ALL patients. From January 2018, a total of 14 MRD-positive B-ALL patients received one or more infusions of autogenous CD19 CAR-T cells. Among them, 12 patients achieved MRD-negative remission after one cycle of CAR-T infusion. At a median follow-up time of 647 days (range 172–945 days), the 2-year event-free survival rate in MRD-positive patients was 61.2% ± 14.0% and the 2-year overall survival was 78.6 ± 11.0%, which were significantly higher than patients with active disease (blasts ≥ 5% or with extramedullary disease). Moreover, patients with MRD had a lower grade of cytokine release syndrome (CRS) than patients with active disease. However, the peak expansion of CAR-T cells in MRD positive patients showed no statistical difference compared to patients with active disease. Five patients received two or more CAR-T cell infusions and these patients showed a decreased peak expansion of CAR-T cell in subsequent infusions. In conclusion, pre-emptive CD19 CAR-T cell treatment is an effective and safe approach and may confer sustained remission in B-ALL patients with chemotherapy-refractory MRD. Diabetes results from a decline in functional pancreatic β-cells, but the molecular mechanisms underlying the pathological β-cell failure are poorly understood. Here we report that large-tumor suppressor 2 (LATS2), a core component of the Hippo signaling pathway, is activated under diabetic conditions and induces β-cell apoptosis and impaired function. LATS2 deficiency in β-cells and primary isolated human islets as well as β-cell specific LATS2 ablation in mice improves β-cell viability, insulin secretion and β-cell mass and ameliorates diabetes development. LATS2 activates mechanistic target of rapamycin complex 1 (mTORC1), a physiological suppressor of autophagy, in β-cells and genetic and pharmacological inhibition of mTORC1 counteracts the pro-apoptotic action of activated LATS2. We further show a direct interplay between Hippo and autophagy, in which LATS2 is an autophagy substrate. On the other hand, LATS2 regulates β-cell apoptosis triggered by impaired autophagy suggesting an existence of a stress-sensitive multicomponent cellular loop coordinating β-cell compensation and survival. Our data reveal an important role for LATS2 in pancreatic β-cell turnover and suggest LATS2 as a potential therapeutic target to improve pancreatic β-cell survival and function in diabetes. Aim: This study identified factors associated with interstitial lung disease (ILD) in patients with idiopathic inflammatory myopathy (IIM) based on the latest classification and recent advances in autoantibody serology. Methods: We retrospectively analyzed data of 173 patients who underwent complete myositis autoantibody serology examination in a medical center in Taiwan from July 2018 to February 2020. After exclusion of patients who did not receive a final diagnosis of IIM, clinical features, serology data, concomitant diseases, treatment, presence of respiratory failure, and mortality rate of the remaining 97 patients were analyzed. Results: Of IIM patients in our cohort, 47.4% had ILD. ILD was significantly associated with subtypes of IIM, older age of onset, presence of mechanic's hand, and presence of anti-Jo-1 and anti-Ro52 antibodies. Among five IIM subtypes, overlap myositis (OM) and dermatomyositis (DM) were significantly associated with a higher prevalence rate of ILD (67.5% in OM and 53.3% in DM). Among patients with OM, the presence of anti-Jo-1 (100%), anti-PL-7 (100%), and anti-EJ antibodies (77.8%) was most significantly associated with ILD. Conclusion: The latest classification of IIM, older age of onset, presence of mechanic's hand, and presence of anti-Jo-1 and anti-Ro52 antibodies were significantly associated with ILD. Among five IIM subtypes, OM and DM had higher prevalence rate of ILD. Among OM patients, the presence of anti-Jo-1, anti-EJ, and anti-PL-7 antibodies was significantly associated with ILD. The combination of edge and cloud computing is going to make the Internet of Things (IoT) rapid, light, and more reliable. IoT and cloud-edge computing are distinct disciples that have evolved separately over time. However, they are increasingly becoming interdependent, and are what the future holds. A crucial aspect is how to design a compound of cloud and edge computing architectures, and implement IoT effectively. In this paper, we proposed a combination of Cloud and Edge Computing architecture and built a set of an intelligent air-quality monitoring system in Tunghai University as a case study. In this case, we implemented container-based virtualization which constructs Kubernetes Minion (Nodes) in the Docker container service independently for each service on the Edge side. Finally, to monitor the high-performance computing systems, clusters, and networks, we used Ganglia Monitoring System. The local electrochemical measurement and corrosion mechanism of the multi-layer aluminium sheets (AA4343/AA3xxx/AA4343 and AA7072/AA3xxx/AA4343) after brazing are determined by a combination of mechanical polishing and work function calculation based on density functional theory. The work functions of (100), (110), and (111) crystal planes of Si and α-Al(Fe,Mn)Si particles which are common in multi-layer aluminium sheets are calculated. The results demonstrate Si and α-Al(Fe,Mn)Si particles are considered to be cathodic with respect to the Al matrix, thus promoting cathodic reactions. The potential differences between the Si and α-Al(Fe,Mn)Si particles with respect to the Al matrix may lead to localized dissolution of the Al matrix. Besides, the microstructure and composition of multi-layered aluminium sheets are studied by optical microscopy, scanning electron microscopy, and glow discharge optical emission spectrometry and compared with those obtained for conventional multi-layer aluminium sheets (AA4343/AA3xxx/AA4343). The presence of the AA7072 layer reduces the outward diffusion of copper. The purpose of this pictorial essay is to review the imaging appearances of the spectrum of thrombophlebitis of abdominal veins on computed tomography (CT) scans. Thrombophlebitis of abdominal veins is rare but mimics other more common conditions presenting with acute abdomen. Objective: To propose a novel classification and scoring system called the posterior ligament-bone injury classification and severity score (PLICS) that offers a quantitative score to guide the need for posterior stabilization in addition to anterior reconstruction for subaxial cervical fracture dislocations (SCFDs). Methods: A total of 456 patients with SCFDs were prospectively included. Patients with PLICS ≥ 7 together with extremely unstable lateral mass fracture (EULMF) were classified as high-risk group, and the other patients were classified as low-risk group. For patients in the low-risk group, anterior-only reconstruction was performed; for patients in the high-risk group, additional posterior lateral mass fixation and fusion was performed after anterior reconstruction. Clinical outcome evaluation included using the visual analogue score (VAS), the Neck Disability Index (NDI), and the American Spinal Injury Association (ASIA) impairment scale. The change in the local sagittal alignment kyphosis Cobb angle was also recorded. Results: A total of 370 patients (81.1%) completed the minimal 12-month follow-ups, including 321 patients of low-risk group and 49 patients of high-risk group. Compared with the average VAS score preoperatively, the score at 12-month follow-up was significantly improved (from 6.1 + 0.3 to 1.1 + 0.2 in the low-risk group, P < 0.001; from 6.4 + 0.2 to 1.4 + 0.2 in the high-risk group, P < 0.001). The average NDI score at the 12-month follow-up was statistically low in the low-risk group (8.8 + 2.5 vs 13.8 + 3.4, P = 0.034). At least more than one grade improvement in the ASIA scale was observed in 80.5% of all patients. The local kyphosis Cobb angle at the injured segment averaged improved in both groups. Osteosarcoma is the most frequent primary bone tumor with poor prognosis. Through RNA-sequencing of 100,987 individual cells from 7 primary, 2 recurrent, and 2 lung metastatic osteosarcoma lesions, 11 major cell clusters are identified based on unbiased clustering of gene expression profiles and canonical markers. The transcriptomic properties, regulators and dynamics of osteosarcoma malignant cells together with their tumor microenvironment particularly stromal and immune cells are characterized. The transdifferentiation of malignant osteoblastic cells from malignant chondroblastic cells is revealed by analyses of inferred copy-number variation and trajectory. A proinflammatory FABP4+ macrophages infiltration is noticed in lung metastatic osteosarcoma lesions. Lower osteoclasts infiltration is observed in chondroblastic, recurrent and lung metastatic osteosarcoma lesions compared to primary osteoblastic osteosarcoma lesions. Importantly, TIGIT blockade enhances the cytotoxicity effects of the primary CD3+ T cells with high proportion of TIGIT+ cells against osteosarcoma. These results present a single-cell atlas, explore intratumor heterogeneity, and provide potential therapeutic targets for osteosarcoma. Biocompatible 2-methacryloyloxyethyl phosphorylcholine (MPC) can enhance the adsorption of water molecules and is therefore used for manufacturing contact lenses. This study investigated the mechanical strength, anti-protein deposition, and anti-lipid adsorption effects of MPC addition to contact lenses. Experimental contact lenses produced by copolymerizing multiple ratios of MPC to 2-hydroxyethyl methacrylate (HEMA) were analyzed. Atomic force microscopy revealed that MPC addition increased surface roughness. The anti-protein deposition and anti-lipid adsorption effects on poly(HEMA-MPC) polymers of various phosphorylcholine quantities were experimentally confirmed. The water content of the contact lenses was proportional to the MPC content in the polymer. The hydrated PC moiety of MPC drastically altered the network of the poly-HEMA polymer by inserting water molecules, which were trapped in the concave region of the surface. MPC addition had negative effects on all examined strength factors because of structural destabilization of the copolymer through water insertion. The anti-deposition effects of MPC were verified by examining the lysozyme and lipid adsorption abilities of the prepared contact lenses. Our results revealed that MPC enhanced interactions of the poly(HEMA-MPC) copolymer with water molecules; these interactions weakened the mechanical strength of the copolymer but markedly improved the anti-adsorption property of the biomolecules. A log management system is essential for the networks administrator. With a log management tool, we can collect, store, analyze, archive, and finally dispose of the log information. In this paper, we propose the architecture model of a log management system using ELK Stack with Ceph to provide a safe network, good Wi-Fi signal strength, and adequate backup data mechanism. In this case, we use our campus data of Wi-Fi log and NetFlow log. First, we collect and store data of our Wi-Fi log using Filebeats tool, and then, we use Elasticsearch, Logstash, and Kibana Stack to visualize the Wi-Fi log data. Second, we collect and store our NetFlow log using NFDUMP, and then, we also use ELK Stack to visualize the NetFlow log data. Third, we integrate the Wi-Fi log and NetFlow log data in one architecture using a distributed storage Ceph file system (CephFS). Focal axon swelling refers to localized swelling in axons that may occur because of trauma (e.g., traumatic brain injury) or neurodegenerative diseases (e.g., Alzheimer's disease). Since the swelling region can be many times larger than its original axon size, many researchers hypothesize that the swelling can alter the action potential (AP) signal. This article discusses the results of a series of newly developed computational studies to elucidate the possible intervention or blockage of AP signals due to swelling in the brain. We argue that the spherical geometry of the swelling site with its enlarged conducting interior causes the entering electric currents to spread evenly over the entire swelled membrane. As such, when the swelled surface becomes larger than the threshold size, the electric current will spread too thin to trigger the AP to spike. In this study, we have used a hybrid membrane model to simulate AP propagation across axons of different radii and swelling radii. We used an integrated model where a cylindrical symmetric 2D model is used to examine the electric current inside a spherical swelling site. In addition, two 1D models are used to capture the current flows along the upstream and downstream stretch before and after the swelling site. The parameters for this model are obtained from literature dedicated to modeling the experimental outcomes of mammal neurons. We observed two factors, which simultaneously affect AP transmission across a swelled axon: a) the axon radius and b) the ratio of the swelled and unswelled axon radii. In general, a thicker axon needs a smaller swelling size and axon ratio to block AP transmission. On the other hand, a thinner axon will reach the threshold at a larger swelling size and axon ratio. When only swelling size is considered, then thinner axons will block AP transmission at a smaller swelling radius. The AP transmission delay inside the swelled region determines whether the AP transmits forward or not. Notably, the blockage is worse if the AP fires at a high frequency. The ongoing outbreak of viral pneumonia in China and across the world is associated with a new coronavirus, SARS-CoV-21. This outbreak has been tentatively associated with a seafood market in Wuhan, China, where the sale of wild animals may be the source of zoonotic infection2. Although bats are probable reservoir hosts for SARS-CoV-2, the identity of any intermediate host that may have facilitated transfer to humans is unknown. Here we report the identification of SARS-CoV-2-related coronaviruses in Malayan pangolins (Manis javanica) seized in anti-smuggling operations in southern China. Metagenomic sequencing identified pangolin-associated coronaviruses that belong to two sub-lineages of SARS-CoV-2-related coronaviruses, including one that exhibits strong similarity in the receptor-binding domain to SARS-CoV-2. The discovery of multiple lineages of pangolin coronavirus and their similarity to SARS-CoV-2 suggests that pangolins should be considered as possible hosts in the emergence of new coronaviruses and should be removed from wet markets to prevent zoonotic transmission. Mg alloys containing long period stacking ordered (LPSO) structures possess excellent mechanical properties and corrosion resistance, exhibiting great application potential in biodegradable implants. The corrosion process of different LPSO phases in the Mg alloys need to be furtherly clarified in different solution environments. In this paper, we mainly reviewed the influencing factors on the corrosion behavior of LPSO-containing Mg alloys, including the alloying elements, processing technologies, and the types, volume fractions and distributions of LPSO phases. Recent researches are well summarized with an emphasis on their corrosion mechanism. Special attention is given to the key issues for LPSO-containing Mg alloys as biomedical implants, with their biodegradation behavior comprehensively discussed. A new portable molecular imprinting polymer (MIP)-SERS nanoprobe is fabricated by a convenient electrochemical method. Single-layered MoS2 is electrochemically reduced on a screen-printed electrode as the scaffold. Functional monomers o-phenylenediamine (oPD), template theophylline (THP), and SERS-active Au nanoparticles (AuNPs) are then one-step electropolymerized on the scaffold. The morphology of the nanoprobe is found to be a three-dimensional and porous structure. The abundant AuNPs with the size of 45~50 nm are trapped within the growing MIP instead of being confined to the surface. The thickness of MIP film is calculated to 25.1 nm. The nanoprobe displays a strong SERS effect for THP using 532 nm as excitation wavelength with a detection limit (LOD) of 0.01 nM. The SERS peak intensity at 1487 cm−1 increases linearly with the concentration of THP in the range 0.1 nM to 0.1 mM. After the template is removed, the imprint-removed nanoprobe is generated for selective binding of THP. The re-binding kinetics study implies the portable MIP-SERS nanoprobe can reach the adsorption equilibrium within 8 min. This nanoprobe exhibits low SERS interference for structural analogues theobromine (THB) and caffeine (CAF). Respiratory rate (RR) is an important vital sign which can be difficult to measure accurately and unobtrusively in routine clinical practice. Pulse transit time (PTT), on the other hand, is unobtrusive to collect from electrocardiogram (ECG) and photoplethysmogram (PPG) signals. Using PTT is a novel method to estimate and monitor blood pressure (BP) and RR. This study aimed to estimate continuous RR using PTT with singular spectrum analysis to extract respiratory components. The performance of this method was validated on 17 subjects who carried out spontaneous breathing and controlled deep breathing conditions. Three types of estimated RR parameters (average RR by power spectral density (PSD) (RRPSD), number of breaths (RR#), and instantaneous RR (RRinst)) were compared with the corresponding reference RR. The reference RR was collected using a respiratory belt. Our findings demonstrate that the PTT signal reliably tracked respiratory variation with a root mean square error of 0.84, 1.11, and 0.74 breaths/min for RRPSD, RR#, and RRinst estimations, respectively. Overall, RR estimated by PTT was more accurate than heart/pulse rate interval, QRS area, and PPG amplitude, which were also extracted from ECG and PPG. The results suggest that it may be feasible to use PTT as an estimation of RR and that ECG and PPG may be relied upon for monitoring not only RR but also BP and heart rate. Novel bio-based and biodegradable block copolymers were synthesized by "click" reaction between poly(L-lactide) (PLLA) and polyamide 4 (PA4). Upon tuning the molar mass of PLLA block, the properties of copolymers and electrospun ultrafine fibers were investigated and compared with those of PLLA and PA4 blends. PLLA and PA4 were found incompatible and formed individual crystalline regions, along with reciprocal inhibition in crystallization. Electrospun fibers were highly hydrophobic, even if hydrophilic PA4 was the rich component. The crystallinity of either PLLA or PA4 decreased after electrospinning and PLLA-rich as-spun fibers were almost amorphous. Immersion tests proved that fibers of block copolymers were relatively homogeneous with micro-phase separation between PLLA and PA4. The fibrous structures of copolymers were different from those of the fibers electrospun from blends, for which sheath-core structure induced by macro-phase separation between homopolymers of PLLA and PA4 was confirmed by TEM, EDS, and XPS.
