Multi-layer slopes are widely found in clay residue receiving fields. A generalized horizontal slice method (GHSM) for assessing the stability of multi-layer slopes that considers the energy dissipation between adjacent horizontal slices is presented. In view of the upper-bound limit analysis theory, the energy equation is derived and the ultimate failure mode is generated by comparing the sliding surface passing through the slope toe (mode A) with that below (mode B). In addition, the influence of the number of slices on the stability coefficients in the GHSM is studied and the stable value is obtained. Compared to the original method (Chen's method), the GHSM can acquire more precise results, which takes into account the energy dissipation in the inner sliding soil mass. Moreover, the GHSM, limit equilibrium method (LEM) and numerical simulation method (NSM) are applied to analyze the stability of a multi-layer slope with different slope angles and the results of the safety factor and failure mode are very close in each case. The ultimate failure modes are shown to be mode B when the slope angle is not more than 28°. Background: Dystonia is a condition that affects the ability to control the movement and function of the body's muscles. It can cause not only physical problems, but also mental problems, resulting in impaired health-related quality of life (HRQoL). However, the effect of deep brain stimulation on quality of life in acquired dystonia remains unclear. Methods: We conducted a systematic literature review from January 2000 to October 2022，determined the eligible studies, and performed a meta-analysis of HRQoL outcomes based on the Short-Form Health Survey-36 (SF-36) after DBS to evaluate the effects of DBS on physical and mental QoL. Results: A total of 14 studies met the inclusion criteria and were systematically reviewed. A comprehensive meta-analysis was performed for 9 studies that reported physical and psychological data or physical component summary (PCS), or mental component summary (MCS) for SF-36. The mean (SD) age at DBS implantation was 34.29 (10.3) years, and the follow-up period after implantation was 2.21 (2.80) years. The random effects model meta-analysis revealed that both physical and mental domains of the SF-36 improved following DBS. There was no statistically significant difference between the physical domains (effect size=1.34; p<0.0001) and the mental domains (effect size=1.38; p<0.0001). Conclusion: This is the first meta-analysis that demonstrates significant benefits in HRQoL following DBS in patients with acquired dystonia. In the post-genome era, great progress has been made in metabolic engineering using recombinant DNA technology to enhance the production of high-value products by Streptomyces. With the development of microbial genome sequencing techniques and bioinformatic tools, a growing number of secondary metabolite (SM) biosynthetic gene clusters in Streptomyces and their biosynthetic logics have been uncovered and elucidated. In order to increase our knowledge about transcriptional regulators in SM of Streptomyces, this review firstly makes a comprehensive summary of the characterized factors involved in enhancing SM production and awakening SM biosynthesis. The isothermal oxidation behavior and oxide-scale evolution on a newly developed Ni–Fe-based superalloy were investigated. Three oxidation stages were generally observed, an initial stage of rapid mass gain, then a second stage of parabolic kinetics, followed by an equilibrium period after about 50, 75, and 100 h at 1000, 1100, and 1200 °C, respectively. Cr2O3 and NiAl2O4 play an important role in protecting the matrix from oxidation at 1000 °C. The chromium supply becomes insufficient to support the continuously growing chromia scale with the increased oxidation temperature. Ultimately, the high-temperature oxidation resistance mainly depends on the formation of the inner continuous α-Al2O3 oxide layer. The quick formation of continuous α-Al2O3 oxide layer at 1200 °C compared to that at 1100 °C leads to a significantly reduced parabolic rate constant, which indicates that the new Ni–Fe-based superalloy has excellent oxidation resistance properties at higher temperatures. CRISPR/Cas9 editing of upstream open reading frames (uORFs) in Arabidopsis thaliana and lettuce enables upregulation of translation of plant genes.Translational regulation by upstream open reading frames (uORFs) is becoming established as a general mechanism for controlling the amount of protein that is synthesized from downstream primary ORFs (pORFs)1,2,3,4,5. We found that genome editing of endogenous uORFs in plants enabled the modulation of translation of mRNAs from four pORFs that are involved in either development or antioxidant biosynthesis. A single-guide RNA that targeted the region harboring a uORF initiation codon can produce multiple mutations. Following uORF editing, we observed varying amounts of mRNA translation in four pORFs. Notably, editing the uORF of LsGGP2, which encodes a key enzyme in vitamin C biosynthesis in lettuce, not only increased oxidation stress tolerance, but also increased ascorbate content by ∼150%. These data indicate that editing plant uORFs provides a generalizable, efficient method for manipulating translation of mRNA that could be applied to dissect biological mechanisms and improve crops. This study investigated a dynamic stability analytical model for bedding rock slope with soil interlayer, considering various rain patterns. The critical rain intensities for anhydrous and water-filled fissure were derived, respectively. Rain coefficients (i.e., concentration and peak position coefficients) were subsequently introduced to investigate rain patterns. The evolving water head under different infiltration scenarios was derived based on Darcy's law and mass conversation law. Additionally, the time-varying water pressure along soil interlayer was obtained based on triangular distribution assumption. An analytical model incorporating time-varying water head was established to predict slope stability based on limit equilibrium method. Numerical simulations further validated the analytical solutions for time-varying water head and pressure. The evolving characteristic of slope stability was analyzed, and a failure threshold was proposed. Results demonstrated that rain patterns and rain coefficients significantly impact on the rainfall threshold corresponding to slope failure. For uniform rain-induced slope failure, rainfall threshold tends to increase as rain intensity decreases. For unimodal rain-induced slope failure, the rainfall threshold is the largest for post-peak rain corresponding to slope failure, followed by the medium-peak rain, and the smallest for forward-peak rain. These results and findings enhance our understanding of the dynamic stability of bedding rock slope with inclined soil interlayer and provide valuable insights into assessing the evolving slope stability under different rain patterns. Rhamnolipid congeners have been widely used in agriculture and biomedicine as potent surfactants. They have recently attracted attention due to their diverse and versatile biological functions, which include an important bacterial virulence factor that makes them attractive targets for research into biosynthetic pathways and gene regulation. The intricate gene expression and regulation network controlling their biosynthesis remain to be completely understood. This article summarizes current knowledge about the biosynthesis pathways and regulatory mechanisms of rhamnolipid congeners, that meet the pharmacological needs of human health and agriculture. Magnetization transfer imaging (MTI) provides a quantitative measure of the macromolecular structural integrity of brain tissue, as represented by magnetization transfer ratio (MTR). In this study, we utilized MTI to identify biophysical alterations in MDD patients with a history of suicide attempts relative to MDD patients without such history. The participants were 36 medication-free MDD patients, with (N = 17) and without (N = 19) a history of a suicide attempt, and 28 healthy controls matched for age and gender. Whole brain voxel-based analysis was used to compare MTR across three groups and to analyze correlations with symptom severity and illness duration. We identified decreased MTR in left inferior parietal lobule and right superior parietal lobule in suicide attempters relative to both non-attempters and controls. Non-attempters also showed significantly reduced MTR in left inferior parietal lobule relative to controls, as well as an MTR reduction in left cerebellum. These abnormalities were not correlated with symptom severity or illness duration. Depressed patients with a history of suicide attempt showed bilateral abnormalities in parietal cortex compared to nonsuicidal depressed patients and healthy controls. A sulfated galactofucan designated as UpG was obtained from the brown algae Undaria pinnatifida by calcium chloride extraction. Chemical analyses showed that UpG is composed of galactose and fucose at a high sulfation level. Low-molecular weight UpGP-0.5 was prepared from UpG through acid hydrolysis for structure characterization. The backbones of UpG are determined to be α-(1,3)-Fuc, α-(1,4)-Gal, α-(1,3)-Gal, and α-(1,6)-Gal by GC-MS, FT-IR, NMR, and LC-MS analyses. Sulfate groups are modified at C2 and/or C4 of fucose and C3 and/or C4 of galactose. UpG could partially lower blood sugar and serum lipid levels in type 2 diabetic mice. Moreover, UpG treatment regulates the abundance of some specific gut microbiota, such as enriching the abundance of Muribaculum and Christensenellaceae, and reducing that of Bilophila, Tannerellaceae, Candidatus Saccharimonas and Anaerotruncus. A single-mold Gasar technique was developed to produce lotus-type porous micro-channel copper with uniform porous structure. In this paper the effect of withdrawal rate on the solid/liquid interface morphology and the corresponding porous structure was systematically investigated, especially the pore morphology, pore growth direction, porosity, and pore diameter of porous copper ingots. In addition, a temperature field simulation was carried out based on ProCast software to investigate the shape and movement velocity of the solidifying solid/liquid interface. The experimental results show that the solidification interface changes from convex to planar, then to concave shape with an increase in withdrawal rate. The enzymatic resolution of ( R, S)-1-phenylethanol produced by hydrolysis of ( R, S)-1-phenylethyl acetate catalyzed by immobilized Candida antarctica lipase B (CALB) was successfully carried out in different solvent systems. A systematic screening and optimization of the reaction parameters such as enzyme amount, the nature and the content of organic solvent, pressure and temperature in supercritical carbon dioxide (SC-CO) and phosphate buffer, with respect to the conversion rate, were performed. CALB exhibits high enantioselectivity in both tert-butanol with 0.025 mol/l phosphate buffer (pH 7.5) and SC-CO with 0.025 mol/l phosphate buffer (pH 7.5) systems. The conversion rate was 41.2% higher in SC-CO with 0.025 mol/l phosphate buffer (pH 7.5) then in tert-butanol with 0.025 mol/l phosphate buffer (pH 7.5) and the reaction time decreased from 8 h to 90 min. The foam stability of dodecyl diphenyl ether disulfonate solution with liquid paraffin droplets in the presence and absence of electrolytes was evaluated, and the stabilization mechanisms were deduced. The foam film is stabilized when the monovalent and divalent counterion concentration is lower than a critical value. However, the foam stability declined with the addition of trivalent counterions. There are two mechanisms we have speculated. Firstly, the counterions reduce the repulsive interaction between the polar groups of surfactants adsorbed at the air/water and oil/water interfaces in the pseudo-emulsion film. Secondly, comparing with the monovalent counterions, the multivalent counterions are not only able to interconnect head groups of adjacent ionic surfactant molecules which are adsorbed at the air/water or oil/water interface, but also to attract the couples of neighboring surfactant head groups adsorbed at the air/water and oil/water interfaces. Short insertions and deletions can be produced in plant genomes using CRISPR–Cas editors, but reliable production of larger deletions in specific target sites has proven difficult to achieve. We report the development of a series of APOBEC–Cas9 fusion-induced deletion systems (AFIDs) that combine Cas9 with human APOBEC3A (A3A), uracil DNA-glucosidase and apurinic or apyrimidinic site lyase. In rice and wheat, AFID-3 generated deletions from 5′-deaminated C bases to the Cas9-cleavage site. Approximately one-third of deletions produced using AFID-3 in rice and wheat protoplasts (30.2%) and regenerated plants (34.8%) were predictable. We show that eAFID-3, in which the A3A in AFID-3 is replaced with truncated APOBEC3B (A3Bctd), produced more uniform deletions from the preferred TC motif to the double-strand break. Ectoine is widely produced by various bacteria as a natural cell protectant against environment stress, e.g., osmotic and temperature stress. Its protective properties therefore exhibit high commercial value, especially in agriculture, medicine, cosmetics, and biotechnology. Here, we successfully constructed an engineered Escherichia coli for the heterologous production of ectoine. Firstly, the ectABC genes from Halomonas elongata were introduced into E. coli MG1655 to produce ectoine without high osmolarity. Subsequently, lysA gene was deleted to weaken the competitive l-lysine biosynthesis pathway and ectoine bioconversion was further optimized, leading to an increase of ectoine titer by 16.85-fold. Finally, at the low cell density of 5 OD600/mL in Erlenmeyer flask, the concentration of extracellular ectoine was increased to 3.05 mg/mL. At the high cell density of 15 OD600/mL, 12.7 g/L of ectoine was achieved in 24 h and the overall yield is 1.27 g/g glycerol and sodium aspartate. Zigui Basin is a major landslide-prone region in the Three Gorges Reservoir region of China, and the stabilizing pile is an effective and widely employed countermeasure to reinforce landslides in this region. However, stabilizing piles are mostly designed using deterministic and stability-oriented methods, which generally ignore the system performance and cost-effectiveness. Using the Majiagou landslide reinforced with stabilizing piles as a case study, a probabilistic multi-objective optimization framework for the design of stabilizing piles is proposed and illustrated. Specifically, performance objectives related to failure probability, system robustness and life-cycle cost of the landslide-stabilizing pile system with feasible designs are evaluated, then the best compromised design is obtained by means of Pareto optimality. Expert knowledge and professional judgment are required to set necessary restrictions and finally determine the optimal design. The results show that there is a better design of stabilizing piles than the existing one, with which acceptable reinforcement effectiveness, compromised life-cycle cost and robust system performance can be realized. The optimal design will also vary with the concerned performance objectives and knowledge-based judgment. Crop improvement by inbreeding often results in fitness penalties and loss of genetic diversity. We introduced desirable traits into four stress-tolerant wild-tomato accessions by using multiplex CRISPR–Cas9 editing of coding sequences, cis-regulatory regions or upstream open reading frames of genes associated with morphology, flower and fruit production, and ascorbic acid synthesis. The natural products discovery program in our group utilizes deep-sea sediment-derived microorganisms and employs a bio-active guided isolation procedure and one strain many compounds (OSMAC) approach to screen bio-active natural products for practical applications in the medicinal and agricultural industry. OSMAC strategy is employed to stimulate secondary metabolite production through changing culture conditions. In this paper, we applied cultivation-dependent procedure, changing media type, leading to the discovery of a bio-active compound named inthomycin B (1) from a marine-derived Streptomyces sp. YB104. The compound was characterized based on extensive spectroscopic analyses and comparison to that in the reported literature. The quantification of inthomycin B demonstrated that Streptomyces sp. YB104 produced moderate yield of inthomycin B with a yield around 25 mg/l after 14 days. Thus, Streptomyces sp. Rhamnolipids have recently emerged as promising bioactive molecules due to their novel structures, diverse and versatile biological functions, lower toxicity, higher biodegradability, as well as production from renewable resources. The advantages of rhamnolipids make them attractive targets for research in a wide variety of applications. Especially rhamnolipids are likely to possess potential applications of the future in areas such as biomedicine, therapeutics, and agriculture. The purpose of this mini review is to provide a comprehensive prospective of biosurfactant rhamnolipids as potential antimicrobials, immune modulators, and virulence factors, and anticancer agents in the field of biomedicine and agriculture that may meet the ever-increasing future pharmacological treatment and food safety needs in human health. Protectin DX (10S,17S-dihydroxydocosa-4Z,7Z,11E,13Z,15E,19Z-hexaenoic acid) (PDX), generated from Ω-3 fatty docosahexaenoic acids, is believed to exert anti-inflammatory and proresolution bioactions. To date, few studies have been performed regarding its effect on pulmonary fibrosis. Herein we show that PDX exerts a potential therapeutic effect which is distinct from its anti-inflammation and pro-resolution activity on mice with pulmonary fibrosis. In the present study, we showed that bleomycin (BLM) increased inflammatory infiltration, collagen deposition, and lung dysfunction on day7 after challenged in mice. Posttreatment with PDX ameliorated BLM-induced inflammatory responses, extracellular matrix (ECM) deposition and the level of cytokines related to fibrosis as evaluated by histology analysis, transformation electron microscope (TEM), lung hydroxyproline content and cytokines test. Moreover, PDX improved lung respiratory function, remedied BLM-induced hypoxemia and prolonged life span. In addition, we found that PDX reversed epithelial-mesenchymal transition (EMT) phenotypic transformation in vivo and in vitro, reinforcing a potential mechanism of promoting fibrosis resolution. In summary, our findings showed that posttreatment with PDX could ameliorate BLM-induced pulmonary fibrosis and lung dysfunction in mice and PDX may be considered as a promising therapeutic approached to fibrotic lung diseases. Multi-structured Ag/MnO2-cordierite molded catalysts were prepared by hydrothermal method and applied to the catalytic oxidation of VOCs. Catalytic activities of Ag/MnO2-cordierite were evaluated by 1000 ppm of toluene, ethyl acetate and chlorobenzene degradation respectively at the air atmosphere, and their physicochemical properties were characterized through multiple techniques containing XRD, SEM, TEM, H2-TPR and XPS. It is found that nanorod Ag/MnO2-cordierite molded catalyst showed prominent catalytic activity for VOCs decomposition and the T90 for toluene, ethyl acetate and chlorobenzene are 275 °C, 217 °C and 385 °C respectively under the space velocity of 10,000 h−1. High valence manganese oxide, more active lattice oxygen proportion and superior low-temperature reducibility were the great contributors to the high activity of the catalyst with nanorod morphology. Studies of space velocity and catalytic stability over nanorod Ag/MnO2-cordierite molded catalyst have confirmed the good catalytic performance, excellent mechanical strength and satisfied anti-toxicity to Cl at higher space velocity, which indicates that this molded catalyst have promise for industrial application.
