The objective of this work was to evaluate the effect of using naturally shaded pastures on scrotal thermoregulatory capacity, testicular echotexture, and sperm morphology of Nelore (Bos indicus) and Canchim (5/8 Bos taurus x 3/8 Bos indicus) bulls in a tropical climate region. Sixty-four adult Nelore and Canchim bulls were used, equally allocated in Full Sun (FS, n = 32) or Crop-Livestock-Forestry (CLF, n = 32) pasture systems. During five consecutive climate seasons, the bulls underwent monthly breeding soundness evaluations and the biometeorological variables in the systems were continuously monitored. Microclimate was significantly different between systems. CLF system had lower BGHI than FS throughout the experimental period. No triple interaction (Season x Breed x Treatment, P > 0.05) was observed for any of the variables. Animals in CLF showed lower body temperature in Summer (FS:39.41 ± 0.05 vs. CLF:39.30 ± 0.05 °C; P = 0.005) and in Autumn (FS:39.54 ± 0.05 vs. CLF:39.35 ± 0.05 °C; P = 0.005). Access to shading did not determine differences in the evolution of scrotal biometry, temperatures, and scrotal thermal gradients (P > 0.05). Regardless of breed, animals in CLF showed greater right testicular volume (FS:247.5 ± 5.7 vs. CLF:259.0 ± 5.7 cm³; P < 0.05), more suitable parenchyma echotexture, and fewer microlithiasis spots in the Spring and Summer. Testosterone concentration was higher in FS (FS:2.6 ± 0.2 vs. CLF:2.1 ± 0.2 ng/mL; P = 0.035). Canchim bulls presented higher total sperm defects during the Autumn and Winter (P = 0.010), but the total defects levels for Canchim and Nelore bulls were in normal range for adult bulls. Thus, the natural shade in CLF system was effective in improving the microclimate of pastures and minimizing adverse environmental effects on some reproductive features of interest in beef cattle. The aim of this study was to investigate the relationship between age, scrotal circumference, postweaning weight and semen quality in Nellore and Caracu bulls selected for postweaning weight. Data from the andrological evaluation of 836 bulls born between 2000 and 2019, including 583 Nellore animals (Bos indicus) and 253 Caracu animals (Bos taurus), were used. The bulls were divided into categories of age at the time of assessment: category 1 consisted of animals aged 20 to 23 months (22 ± 0.76 months, 518 ± 94.17 kg), category 2 consisted of animals aged 24 to 35 months (30 ± 4.42 months, 679 ± 137.19 kg), and category 3 consisted of animals ≥ 36 months (60 ± 14.12 months, 907 ± 161.73 kg). The statistical model included the effects of breed, age category, date of semen collection, and breed x age category interaction. Heritability estimates for scrotal circumference at 13 months of age (SC1year) and semen quality traits were obtained for the sample of Nellore animals. Most semen quality traits improved with increasing age in both Nellore and Caracu animals. High heritability was observed for SC1year (0.45), while sperm motility, vigor, turbulence, and major, minor and total sperm defects exhibited low heritability (0.11, 0.019, 0.047, 0.017, 0.017 and 0.019, respectively). Spearman correlations of breeding values for postweaning weight (W378) and SC1year with the semen quality traits were low. Nellore and Caracu bulls have similar semen quality that improves with increasing age. In the Nellore breed, the heritability of SC is high, while semen quality traits exhibit low heritability. A new anodic stripping voltammetry technique has been proposed for direct determination of zinc in seminal plasma of bulls. Two types of electrodes were used as working electrodes—a stationary glassy carbon electrode (GCE) and a thick-film-modified graphite electrode (TMGE) for comparison. The method is rapid, simple, and highly accurate, has high selectivity and low limits of detection (LODs) and limits of quantification (LOQs), and no sample pretreatment steps are also required. The data obtained in both cases agree well with each other and with the data obtained by the independent method of inductively coupled plasma optical emission spectrometry. The relative error of the zinc determination by anodic stripping voltammetry does not exceed 10%. The LOD and LOQ values obtained under optimized conditions were 3.0 and 8.0 µg L−1 for GCE and 2.0 and 5.0 µg L−1 for TMGE, respectively. In contrast to the GCE application, the TMGE application did not include the addition of toxic soluble mercury salts (II) in the electrolyte solution. The zinc content in seminal plasma of bulls is 6.1 ÷ 13.0 mg L−1. The mean values of zinc concentrations in the samples were 8.4, 11.9, and 9.0 mg L−1 for Red, Simmental, and Black-motley animals, respectively. Bovine leukemia virus (BLV) causes enzootic bovine leukosis, a persistent infection and the most important neoplastic disease in cattle. It is spread primarily by transferring infected lymphocytes through blood from carriers to healthy animals. The present study is aimed at determining the seropositivity of BLV in breeding bulls from Costa Rica and at detecting for the first time in the country BLV DNA in bull semen. Between May 2011 and August 2018, 379 blood and 133 semen samples were collected from bulls distributed in 118 farms. The serum was analyzed by an enzymatic immunoassay and the semen by polymerase chain reaction and sequencing. BLV seropositivity was 43.5% (165/379), while 64.4% (76/118) of the farms had positive reactors. Holstein (75.7%) and Jersey (73.0%) breeds showed the highest seropositivity. In addition, Bos taurus bulls (68.1%), older than seven years (50.0%), and those belonging to dairy farms (75.5%) had higher seropositivity compared to Bos indicus (17.7%), younger than seven years (42.2%), and those from beef farms (15.5%), respectively. Moreover, Bos taurus bulls had a higher risk of being seropositive than Bos indicus (OR = 3.4; 95% CI: 1.7–6.8). BLV DNA was found in one semen sample (2.5%; 1/40) from a seropositive bull. The importance of serum and molecular BLV screening in semen samples and the potential role of some risk factors associated with the disease, such as the bull’s age, genotype, and type of livestock productive system, is argued in the present report. Poor semen profile reflected by suboptimum fertility statistics is a concern in bulls reared for breeding purpose. A critical review of research on candidate genes and proteins associated with semen quality traits will be useful to understand the progress of molecular marker development for bull semen quality traits. Here, we have tabulated and classified candidate genes and proteins associated with bull semen quality based on a literature survey. A total of 175 candidate genes are associated with semen quality traits in various breeds of cattle. Several studies using candidate gene approach have identified 26 genes carrying a total of 44 single nucleotide polymorphisms. Furthermore, nine genome-wide association studies (GWASes) have identified 150 candidate genes using bovine single nucleotide polymorphisms (SNP) chips. Three genes, namely membrane-associated ring-CH-type finger 1 (MARCH1), platelet-derived growth factor receptor beta, and phosphodiesterase type 1, were identified commonly in two GWASes, which, especially MARCH1, are required to explore their regulatory roles in bull semen quality in in-depth studies. With the advancement of high-throughput-omic technologies, more candidate genes associated with bull semen quality may be identified in the future. The parasite T. foetus causes trichomonosis in cattle but is generally asymptomatic in males. Thus, many bulls carrying the disease go unnoticed, making the detection of T. foetus in bulls an important aspect for its control. Due to drawbacks posed by its cultivation, PCR is a preferred option for diagnostic laboratories. Most published PCR protocols target the genomic region compring the 18S, 5.8S, and 28S rRNA genes and internal transcribed spacers 1 and 2 (rRNA-ITS region), homologous to that of other Tritrichomonas species. There is minimal information on alternative genetic targets and no comparative studies have been published. We compared a protocol based on the microsatellite TfRE (called H94) and five protocols based on the rRNA-ITS region (called M06, M15, G02, G05, and N02). We also designed and evaluated a novel PCR-based assay on the EF1-alpha-Tf1 gene (called V21). The analytical sensitivity and specificity assays for the PCR protocols were performed according to the World Organisation for Animal Health (OIE) directives and the comparative study was performed with a widely used PCR (M06) on clinical samples from 466 breeding bulls. V21 showed a high degree of agreement with our reference M06 (kappa = 0.967), as well as M15 (kappa = 0.958), G05 (kappa = 0.948), and H94 (kappa = 0.986). Protocols H94 and V21 appear to be good approaches for confirming clinical cases in preputial bull samples when genomic regions alternative to rRNA-ITS are required. In a temperature-increasing scenario, due to global warming, the individual thermic resilience of the male assumes a crucial role in the reproductive efficiency of a male since the thermic stress, such as the inability of the male to reduce body or regional temperature on a physiological level, impairs testicular function. In this study, the effect of the environmental conditions on the fresh semen quality, in terms of volume, concentration, total sperm in the ejaculate, total motility, normal morphology, membrane integrity, and discarding rate, were compared longitudinally in Belgian Blue (BB) and Brown Swiss (BS) bulls. The environmental conditions, summarized in the mean temperature-humidity index (THI), were calculated on the day of collection, as well as 7 days (epididymal maturation), 35 days (late spermatogenesis), and 70 days (early spermatogenesis) before the collection, to reflect spermatogenesis time. Our findings showed that limited seasonal effects were present in the semen quality of BS bulls. On the other hand, in BB bulls lower semen quality was found between July and November, with a different timing depending on the seminal parameter. This effect of the season on BB semen parameters appears to be related to the THI. The data presented in this study shows that the temperature and humidity, summarized in THI, could affect the semen quality of the bull on breed basis, given that volume, concentration, total sperm in the ejaculate, total motility, membrane integrity, and sperm normal morphology were significantly reduced by an increasing THI in the Belgian Blue bulls, but not in Brown Swiss bulls. The sperm-derived oocyte activating factor, phospholipase C zeta (PLC ζ), is the only PLC isoform reported in cattle. The objectives were to (1) localize PLC ζ in fresh and capacitated bovine sperm and (2) investigate the activation of PLC ζ during bull sperm capacitation and contributions of PLC activity to this process. We confirmed interaction of testis-specific isoform of Na/K-ATPase (ATP1A4) with PLC ζ (immunolocalization and immunoprecipitation) and tyrosine phosphorylation (immunoprecipitation) of PLC ζ (a post-translational protein modification commonly involved in activation of PLC in somatic cells) during capacitation. Furthermore, incubation of sperm under capacitating conditions upregulated PLC-mediated hyperactivated motility, tyrosine phosphoprotein content, acrosome reaction, and F-actin formation (flow cytometry), implying that PLC activity is enhanced during capacitation and contributing to these capacitation processes. In conclusion, we inferred that PLC ζ is activated during capacitation by tyrosine phosphorylation through a mechanism involving ATP1A4, contributing to capacitation-associated biochemical events. This study was designed to evaluate the seasonal expression of seminal plasma proteins from two bovine breeds adapted to a subtropical climate and their associations with post-thawing sperm and environmental characteristics. Semen samples were obtained three times in summer and three times in winter from four Crioulo Lageano and four Angus bulls. Seminal plasma was obtained by centrifugation, and the other portion of the semen was cryopreserved. Seminal plasma proteins were identified by 2D-nanoUPLC-MSE. Post-thawing assessments of sperm kinetics, morphology and membrane integrity were performed. Environmental data such as air temperature, air humidity and black globe temperature (BGT) were recorded, and the temperature-humidity index (THI) was calculated in summer and winter. Results showed that the climate varied significantly between seasons. Although no statistical differences were observed in semen quality between breeds, the protein profiles varied within and between seasons. We suggest that the most critical proteins in summer affecting sperm characteristics were TIMP-2, DNase, Clusterin, CFAH and GPx6. TIMP-2 and DNase showed a higher abundance in Crioulo Lageano in comparison with Angus, while Clusterin, CFAH and GPx6 presented a lower abundance. To the best of our knowledge, this is the first report of a recently evolved type of glutathione peroxidase, GPx6, in seminal plasma of bovines. In winter, five proteins were considered to be more critical: BSP1, BSP3, CCL2, Sulfhydryl oxidase and TIMP-2. In Bostock v Clayton County (2020) Gorsuch J holds that direct discrimination because of sexual orientation is a form of direct discrimination because of sex. I argue that the same is true under the Equality Act 2010. I consider the arguments of (Finnis, in: Finnis (ed) Intention and identity: collected essays, Oxford University Press, Oxford, 2011) and (Gardner in Oxf J Leg Stud 18(1):167–187, 1998) that "because of", "on grounds of", and similar phrases in UK discrimination legislation invoke the state of mind of the discriminator. I apply this point to Bull and Bull v Hall and Preddy [2013] arguing that (i) the UK Supreme Court was wrong to find direct discrimination on grounds of sexual orientation, while, (ii), nevertheless, under the Equality Act 2010, that case and similar cases actually involve direct discrimination because of sex, not because of sexual orientation. Lethal episodes arising from interactions with cattle may be the result of a response of the animals to simple handling and herding, or from reactions to deliberate provocation or goading intended to incite aggressive behavior for public entertainment purposes. Deaths may be considered to be unprovoked and unanticipated, or provoked and predictable. Cattle cause significant numbers of deaths globally and are considered the most dangerous large animal in Britain. Behavior may be unpredictable even in apparently docile domesticated animals, and attacks may be by a single animal or a herd and result in injuries from kicking, head/butting/charging, stomping, goring, and crushing. Craniofacial injuries may involve fractures of the spine or skull with cerebral contusions and lacerations associated with subarachnoid, subdural, and extradural hemorrhages. Chest injuries are also characterized by fractures which may be multiple with flail chest, hemo- and pneumothoraces, and organ disruption. Injuries to the abdomen and perineum include intestinal perforations, splenic rupture, perineal and vaginal tears, urethral lacerations and avulsions, and bladder and rectal perforations. Neospora is one of the protozoans that can infect the male and female's reproduction system. Despite the existence of N. caninum in the genitalia, its effect on sperm characteristics was not studied yet. Therefore, the aim of this study was to investigate the influence of natural neosporosis on the sperm parameters of bulls. Using 30 bulls with neosporosis diagnosed by modified agglutination test and enzyme-linked immunosorbent assay (ELISA) and 15 healthy bulls, some sperm parameters such as sperm concentration, viability, motility, and morphology were studied and compared. Also, the activity of super oxide dismutase (SOD), glutathione peroxidase (GPX), and malondialdehyde (MDA) level as the biomarker of lipid peroxidation was investigated. Results showed that sperm concentration, viability, and motility were significantly lower in bulls with neosporosis in the present study. Capacitation comprises a series of structural and functional modifications of sperm that confer fertilizing ability. We previously reported that the testis-specific isoform of Na/K-ATPase (ATP1A4) regulated bovine sperm capacitation through signaling mechanisms involving kinases. During subsequent investigations to elucidate mechanisms by which ATP1A4 regulates sperm capacitation, we observed that ATP1A4 was localised in both raft and non-raft fractions of the sperm plasma membrane and that its total content was increased in both membrane fractions during capacitation. The objective of the present study was to investigate mechanism(s) of capacitation-associated increase in the content of ATP1A4. Despite the widely accepted dogma of transcriptional/translational quiescence, incubation of sperm with either ouabain (specific ligand for ATP1A4) or heparin increased ATP1A4 content in raft and non-raft sperm membrane fractions, total sperm protein extracts (immunoblotting) and fixed sperm (flow cytometry), with a concurrent increase in Na/K-ATPase enzyme activity. This capacitation-associated increase in ATP1A4 content was partially decreased by chloramphenicol (mitochondrial translation inhibitor) but not affected by actinomycin D (transcription inhibitor). To demonstrate de novo ATP1A4 synthesis, we evaluated incorporation of bodipy conjugated lysine in this protein during capacitation. A partial decrease in bodipy-lysine incorporation occurred in ATP1A4 from sperm capacitated in the presence of chloramphenicol. Therefore, increased ATP1A4 content during capacitation was attributed to mitochondrial translation of ATP1A4 mRNA present in ejaculated sperm, rather than gene transcription. In the present case, jugular vein engorgement and oedema of the brisket region which mimic the signs of pseudopericarditis due to theileriosis in a bull was reported. An adult Ongole crossbred bull with history of weakness, anorexia and oedema at brisket region was referred to Teaching Veterinary Clinical Complex, College of Veterinary Science, Proddatur. Physical examination revealed the presence of ticks over the body, right and left prescapular lymphadenopathy and oedema at brisket region. Blood examination revealed the presence of piroplasms in erythrocytes, schizont in lymphocytes, anaemia and lymphopenia. Bull was treated with injection buparvaquone along with supportive medications. By the 3rd week of therapy improvement was noticed by disappearance of brisket oedema and recurrence was not noticed during the 8 months of observation. [ABSTRACT FROM AUTHOR]

A 9 years old bull was presented to the hospital with a history of scrotal swelling for a period of 12 days and progressive emaciation with intermittent fever. Clinical examination revealed the swollen and painful scrotum with fluid thrill, mild abduction and disorientation of hindlimbs were noticed. Ultrasonography of the testes revealed the anechoic fluid in between the visceral and parietal layers of tunica vaginalis of testis with variation in the echogenicity of the testes. Stained peripheral blood smear examination revealed the presence of Trypanosoma evansi organisms. The bull was administered with diminazene aceturate, enrofloxacin, flunixin meglumine, multivitamin injections and dextrose normal saline. Uneventful recovery was noticed following the therapy. Very limited information is available on the trans-scrotal ultrasonography of cross-bred Ongole bulls with scrotal swelling in India. The present study was carried out on the bulls presented to the hospital with the history of scrotal swelling. Haematology revealed increased leucocyte count, neutrophilia and mild reduction in haemoglobin levels. Microscopic evaluation of the semen revealed abnormal morphology and the presence of the dead spermatozoa. Trans-scrotal ultrasonography helps in the differential diagnosis of scrotal disorders and to identify the appropriate aetiology for developement of the condition. On ultrasonography, all the bulls showed abnormal scrotal morphology. Among these, one had the presence of normal echogenicity testes which are surrounded by the anechoic fluid which will cover by a hyper echoic scrotal skin. The second one showed the anechoic fluid was mixed with the hyper echoic fibrinous threads and degenerative changes in one testis. Third bull showed the diffused hyper echoic threads scattered within the anechoic fluid and different echogenicity of the two testes. Contagious Bovine Pleuropneumonia (CBPP) is a highly infectious cattle disease, which is widespread in pastoral areas of Africa, and it imposes a major problem on Ethiopian livestock export market. Cross-sectional study was conducted in 2011 on bulls originated from Borena pastoral area to determine seroprevalence of CBPP. Forty batches of bulls containing 38,187 Borana bulls were tested using c-ELISA. Of the total 40 batches tested for the presence of antibodies, 25 (62.5 %) of them contained at least one seropositive bull. From the total of 38,187 bulls tested, 150 (0.4 %) bulls were positive. The number of seropositive animals increases as the herd size increases ( P < 0.05). Both at herd and individual level, the highest CBPP prevalence was recorded in herd size >1000, and the difference was found statistically significant ( P < 0.05). There was statistically significant ( χ = 23.73, df = 9, P = 0.005) difference of CBPP prevalence between months of the year. The present low prevalence of CBPP in the cattle feedlots indicates that the disease is decreasing progressively in Borena pastoral area, this might be associated with the ongoing mass vaccination campaign against economically important livestock diseases in pastoral areas. The decrease in the prevalence of CBPP offered a great opportunity to livestock producers and live animal and meat exporters by improving the demand of Ethiopian livestock on international market. Regular reintroduction of infected cattle from neighboring countries or herds where the disease remains endemic may change the disease dynamics again. Therefore, mass blanket vaccinations coupled with prompt diagnosis, isolation and stamping out of the outbreaks, intensive surveillance, followed by strict cattle movement control should be implemented by concerned parties. The aim of this study was to assess the seasonal effects of the environment on semen quality in bulls, using infrared thermography. Sperm motility (M), mass motion (MM), and vigor (VIG) were evaluated in sperm samples from 17 Bradford bulls aged approximately 24 months at the beginning of the study. Infrared thermography images and data were collected using an infrared FLIR T 300 camera and Quick Report 1.2 SP2 software to determine the temperature of the proximal and distal poles of the testis and to assess the testicular temperature gradient. The seasonal effects on physiological, seminal, and climatic variables were analyzed by the GLM ANOVA and CORR procedures using SAS®. The microclimatic factors were recorded in hourly intervals, and the daily mean temperature and mean relative humidity were calculated to determine the daily temperature-humidity index (THI) every day for 1 year. The temperature gradient (TG) variations of the testes were significantly higher in the autumn (4.5 °C), winter (4.0 °C), and spring (2.9 °C) compared to summer (0.9 °C) ( P < 0.05). Ocular globe temperatures were lower in the winter (27.6 °C) and autumn (26.8 °C) compared to summer (33.9 °C) and spring (31.1 °C) ( P < 0.05). The average MM (2.58), M (52.64), and VIG (2.70) of the semen decreased in the summer compared to other seasons ( P < 0.01). The TG was negatively correlated with THI (−0.44; P < 0.05). For the seminal variables, MaD (−0.45; P < 0.05) and TD (−0.50; P < 0.01) presented a negative correlation with TG. The TG had a positive correlation between M and VIG, which had values of 0.36 and 0.35, respectively ( P < 0.05). Bovine besnoitiosis is a chronic and debilitating disease observed in many European countries that may cause important economic losses in cattle. The recent widespread of the parasite in Europe had led the European Food Safety Authority to declare bovine besnoitiosis as a re-emerging disease in Europe. Many aspects of the epidemiology of bovine besnoitiosis such as the main routes of transmission are still unclear and need to be further studied. Among the different hypotheses, a sexual transmission has not yet been investigated. Therefore, the aim of this study was to evaluate the presence of Besnoitia besnoiti DNA in the semen of naturally infected bulls by using a highly sensitive method (real-time qPCR). Both pre-sperm and sperm fractions of 40 bulls, including seronegative ( n = 11), seropositive subclinically ( n = 17), and seropositive clinically ( n = 12) infected animals, were collected by electroejaculation and analyzed by real-time qPCR. No B. besnoiti DNA was detected in 27 pre-sperm and 28 sperm fractions of the 40 examined bulls, suggesting that the transmission of B. besnoiti infection by the semen of chronically infected bulls is very unlikely. With the premium bull plays a growing important role in cattle industry, semen detection technology based on individual identification and phylogenetic relationship is paid more and more attention. In order to lay the foundation for the establishment of the China Holstein bull identification method, this research takes 20 Chinese Holstein dairy bull's blood and their corresponding semen, and then extracts the DNA both from the blood and semen, analysis the genetic polymorphisms of 10 microsatellite loci (TGLA227, INRA23, TGLA122, BM2113, SPS115, ETH3, ETH225, MCM158, MAF45 and UMN0108) by microsatellite marker, discuss the feasibility of this method used to individual identification. The results showed that Chinese Holstein dairy bull genetic diversity in the ten microsatellite loci were both high, and the average polymorphic information content of TGLA227, which highest, is 0.8162, ETH225 has the lowest, which is 0.6224. Use STR loci to identify the bull's semen, the cumulative individual identification capacity is 99.99 %, which indication that 10 STR loci can be used to the frozen semen quality test and cows individual identification. Purpose: This study was designed to determine the combined effects of adding source of n-3 fatty acids (FA) and α-tocopherol (vitamin E, VE) to semen extender on freezability and FA composition of Brown Swiss bull sperm. Methods: Semen samples were collected from 6 Brown Swiss bulls and pooled. In the first trial, semen was divided into 12 groups including 4 levels of n-3 FA (0, 1, 10, 100 ng ml) and 3 levels of VE (0. 0.2, 0.4 mM). Motility, viability and fatty acid composition of sperm were measured. Results: The treatment of 10 ng ml n-3 FA and 0.4 mM VE had the best post-thaw sperm characteristics ( P < 0.01). In the second trial, sperm lipid composition of this treatment and control (without FA and VE) was determined. Supplementing n-3 fatty acids during cryopreservation increased docosahexaenoic acid (DHA) and the ratio of n-3 to n-6 FA in sperm before freezing and after thawing. Conclusions: The results suggest that combining the optimal level of n-3 FA (10 ng ml) with the highest level of VE tested (0.4 mM) in a semen extender changed the membrane lipid composition and improved freezablity of Brown Swiss bull sperm. This study evaluated the protective effect of butylated hydroxytoluene (BHT), a lipid-soluble antioxidant against cryopreservation damage on bull spermatozoa. Four BHT concentrations (0.5, 1, 2 and 4 mM) were evaluated. Sperm characteristics were evaluated when BHT was added to a post-thaw-freezing extender by measuring the degree of sperm lipid peroxidation (using malondialdehyde, MDA) and by measuring parameters such as motility and viability of spermatozoa. Production of MDA as an indicator of lipid peroxidation was obtained when BHT ranged from 0.5 to 1 mM in both extenders ( P < 0.0001). Sperm motility and viability evaluated immediately after thawing was higher in BHT-treated spermatozoa, this was significant ( P < 0.001) when the freezing egg yolk-citrate extender was supplemented with 0.5 and 1 mM BHT and when egg yolk-Tris extender was supplemented with 0.5 mM BHT. The addition of 0.5 and 1 mM BHT to semen egg yolk-citrate extender increased the viability of frozen semen after thawing ( P < 0.007). The addition of 0.5 and 1 mM BHT to both extenders resulted in a significant ( P < 0.0001) decrease in MDA production. In conclusion, the addition of BHT to freezing egg yolk-citrate extender improved the overall efficiency of thawed bull spermatozoa, but the addition of BHT to the freezing egg yolk-Tris extender did not. The effect of castration on carcass quality was investigated by ten Chinese Simmental calves. Five calves were castrated randomly at 2 months old and the others were retained as normal intact bulls. All animals were slaughtered at 22 months old. The results showed that bulls carcass had higher weight ( P < 0.05), dressing percentages and bigger longissimus muscle areas ( P < 0.05) than steers. But steer meat had lower shear force values and was fatter ( P < 0.05) than bull. Furthermore, in order to discover genes that were involved in determining steer meat quality, we compared related candidate gene expression in longissimus muscle between steer (tester) and bull (driver) using suppressive subtractive hybridization. Ten genes were identified as preferentially expressed in longissimus muscle of steer. The expression of four selected differentially expressed genes was confirmed by quantitative real-time PCR. Overall, a 1.96, 2.41, 2.89, 2.41-fold increase in expression level was observed in steer compared with bull for actin, gamma 2, smooth muscle, tropomyosin-2, insulin like growth factor 1 and hormone-sensitive lipase, respectively. Reproductive effort (RE) is defined as the investment in any current reproductive act. In male ungulates, variation in RE can be explained by differences in age, dominance, phase in the breeding season and number of oestrus females simultaneously present. We investigated the relation between these factors and RE in American bison males in semi-natural conditions. We looked for differences between males in RE, measured by tending behaviour, and maintenance activities (feeding/resting) during the rut. We found no effect of age on RE. During early rut there was no difference in RE between males of different rank. Dominant males had a higher RE during peak rut, and in late rut both the first and second ranking males showed more RE than lower ranking bulls. For all males, there was a clear increase in RE towards peak rut and when more females were attractive. Maintenance behaviours decreased when more females were synchronously in oestrus. The most dominant bulls spent less time grazing than lower ranking males, and all males decreased feeding during peak rut. Resting decreased with age. We conclude that dominance, phase of rut and the number of females synchronously in oestrus are the factors that best explain variation in RE. [ABSTRACT FROM AUTHOR]

The dynamics of the intrapopulation parameters of the northern fur seal on Tyuleniy Island are considered for the period when their population was recovering after a depression. Almost all its characteristics, except for those dependent on density, showed a positive tendency. Dynamic processes in the northern fur seal populations in the post-depression years are found to be based on the demographic transformation of the community, which closely correlates with its qualitative changes, predetermined by the dynamics of its age structure. The process of complete reversal in the state of the fur seal population, which is distinguished by peak values at the depression and in the years of prosperity, is approximately equal to the life span of one generation of these animals. Leptin, a 167-amino acid hormone, is secreted mainly by fat tissue. It has some powerful effects on the regulation of metabolism and reproductive function through endocrine and probably paracrine mechanisms. The contribution rate of leptin function on the male reproductive system is not still clear. Characterization of leptin expression in reproductive organs will suggest that in addition to its endocrine action, leptin has also paracrine/autocrine effects on reproduction. The expression of functional leptin receptor mRNA has been already recognized in testis of rodents, human and cattle. Thus, the aim of the present study was to investigate the presence of leptin mRNA in the bovine testis, because it will be the first step for understanding of its paracrine/autocrine effects on the male reproductive organs in cattle. The present study was the first to showed leptin mRNA expression in the testis of Holstein cattle using reverse transcription and polymerase chain reaction (RT-PCR) analysis. RT-PCR products were amplified with nested PCR using inner leptin primer pairs to emphasis the first results. Besides, bovine beta actin gene was acted as an internal positive control as well as RNA purification marker. Our findings suggest that in addition to its endocrine actions at the hypothalamic-pituitary axis, leptin can has an autocrine and/or paracrine role in bull testicular function. Purpose The objectives were to test how the source of oocytes and semen impacted vitrification of large numbers of bovine oocytes and subsequent IVF and early embryo development to test procedures that may assist with assisted reproductive technologies in humans. Methods Bovine oocytes were vitrified from follicles of different diameters, small (≤4 mm) and medium (4 to 10 mm), using nylon mesh. Oocytes were exposed to the cryoprotectant composed of 40% (v/v) ethylene glycol, 18% (w/v) Ficoll-70, and 0.3 M sucrose in three stepwise dilutions. Thawing was conducted with a series of 0.5, 0.25 and 0.125 M sucrose dilutions in 20% fetal bovine serum. Results The cleavage (39.1% vs. 58.5%) and blastocyst rates (5.1% vs. 22.9%) were significantly lower for the vitrified oocytes. Follicle size had a significant impact on the development of embryos. Sires had significant effects on embryonic developmental rates. Conclusions We conclude that differences in development exist due to follicle source and sire used for IVF after vitrification. To determine the presence of constitutive nitric oxide synthase (NOS-3) mRNA in the caput, corpus, cauda epididymis, and vas deferens of adult bull, semiquantitative reverse transcription-polymerase chain reaction was performed on total RNAs isolated from those tissues. Expression of NOS-3 gene was exhibited in the samples of vas deferens and three parts of epididymis (caput, corpus, and cauda). Of course, the NOS-3 gene was detected in the part of the corpus weakly and was significant in comparison to other parts. NOS-3 mRNA in the part of cauda was nonsignificantly more than in other parts of the epididymis. Expression of this gene was significantly elevated in the two parts of the epididymis (caput and cauda) when compared with the vas deferens samples. The fine structure of bovine Sertoli cells was studied from the 4th to the 40th week post natum in order to correlate the progressive acquisition of normal adult morphology with functional development. The considerable increase in tubular size during the first 20 weeks is due to the proliferation of both presumptive Sertoli and germ cells. Aside from this, the presumptive Sertoli cells are seen to expand radially and lengthen considerably. From then on however, the observed increase in tubular diameter during the later period of postnatal development is solely due to the great increase in the number of germ cells. Presumptive Sertoli cells undergo morphological differentiation to mature Sertoli cells during the first 28 weeks of proliferative development. The maturation process includes distinct changes in cell shape, nucleus and cellular organelles, as well as an increase in and differentiation of Sertoli cell surface specializations. At 24 weeks the development of inter-Sertoli cell junctions has reached a point of differentiation where, in our opinion, a functional blood-testis barrier can be expected. During the first 8 weeks an extensive development of rough endoplasmic reticulum and a well-developed Golgi apparatus can be observed, which suggests a high secretory activity in the presumptive Sertoli cells at this time. We speculate that these secretory activities may play a role in the formation of the basal lamina which is extremely well developed during early postnatal life. The present lectin histochemical comparison of cauda epididymal and ampullary bovine sperm was conducted to investigate whether ampullary secretions are involved in altering the plasma-membrane glycoconjugates of sperm. A marked redistribution of glycoconjugates between sperm from these two regions was indeed revealed on the basis of changes in binding patterns of the following fluoroscein-isothiocyanate (FITC)-labelled lectins: wheat-germ agglutinin (WGA), Maclura pomifera agglutinin (MPA), Griffonia simplicifolia I agglutinin (GS I) and Bauhinea purpurea agglutinin (BPA). This was evidenced in the first three cases by a relative reversal of staining intensity between the acrosomal and post-acrosomal regions, and by a pronounced increase in the staining of the midpiece. Changes in the distribution of BPA binding sites were limited to the latter phenomenon. The results are compared with previous findings, discussed in the context of the hypermotility characteristic of ampullary sperm and related to previously reported differences in the lectin-binding patterns of the luminal and glandular epithelia. The morphology of the bovine Sertoli cell was studied during 6 different phases of the spermatogenetic cycle. Tubular dimensions do not vary significantly during the phases. Sertoli cells occupy 27.0% (phase 4) to 38.4% (phase 8) of the tubular epithelium. Sertoli cells of phase 1 are approximately 20% larger than during the other phases. 30-35% of Sertoli cell volume consists of organelles. Mitochondrial (about 5.0%) and nuclear (about 5.7%) volume densities remain remarkably stable during the cycle, irrespective of changes in Sertoli cell size. Phagocytic capacity of bovine Sertoli cells is only moderate. Elimination of excess spermatid cytoplasm occurs to a large extent prior to spermiation. The majority of spermatid residual bodies undergoes autolytic decay while attached to the Sertoli cell apical surface. Aggregates of densely packed cisternae of the smooth endoplasmic reticulum (ER) located in a basal position and associated with the acrosome-phase and maturation-phase spermatids contribute between 14 and 17% to Sertoli cell volume. During phase 3 the ER pinches off a large number of small, smooth-walled vesicles filled with flocculent content. The contact area between Sertoli cells and other tubular constituents changes considerably during the different phases. The postnatal development of intertubular cells and vessels and of the tubular lamina propria was studied in three locations of perfusion-fixed bovine testes from 31 animals ranging from 4 to 78 weeks. The postnatal morphological differentiation of the testis is not uniform, regional differences have to be considered. The intertubular cell population is composed of mesenchyme-like cells, fibrocytes, Leydig cells, peritubular cells and mononuclear cells. In 4 and 8-week-old testes mesenchyme-like cells are the dominating element. These pluripotent cells proliferate by frequent mitoses and are the precursors of Leydig cells, contractile peritubular cells and fibrocytes. Morphologically differentiated Leydig cells are encountered throughout the entire period of postnatal development. In 4-week-old testes degenerating fetal and newly formed postnatal Leydig cells are seen in juxtaposition to each other. From the 8th week on, only postnatal Leydig cells are present. Between 16 and 30 weeks large-scale degeneration of prepuberal Leydig cells is observed. The Leydig cells that survive this degenerative phase constitute the long-lasting adult population. 20-30% (numerically) of all intertubular cells at all ages are free mononuclear cells. These are found as lymphocytes, plasma cells, monocytes, macrophages and light intercalated cells (LIC). The latter are monocyte-derived, Leydig cell-associated typical cells of the bovine testis. The differentiation of the two main components of the tubular lamina propria, (i) basal lamina and (ii) peritubular cell sheath, seems to be effected rather independent from each other and also from hormonal signals important for the development of the germinal cells. The laminated basal lamina reaches nearly 3 μm at 16 weeks and is later on continuously reduced. At 25 weeks the peritubular cells have transformed into contractile myofibroblasts. The seminal vesicles synthesize in an androgen-dependent manner a neutral protein of 13.5 kDa molecular weight that makes up about 40% of their secretion ('major protein'). An antiserum against this protein raised in rabbits was used to localize the antigen within the seminal vesicles. In addition to intraluminal secretion of the seminal vesicles and the ampulla of the vas deferens, ejaculated and ampullary spermatozoa revealed an intense immunoreaction, which was restricted to the neck region of the sperm head and the middle piece, while the principal piece of the tail as well as the sperm head were devoid of immunoreactive material. Comparison of spermatozoa taken from the tail of the epididymis with ampullary spermatozoa showed that about 90% of the latter, but only 10-20% of the former presented this distributional pattern of immunoreactive sites. Epididymal epithelium as well as calf seminal vesicle epithelium showed no immunoreactivity with major protein antiserum. Using a pre-embedding staining technique with gold-labeled primary or secondary antibodies, respectively, no immunostaining could be achieved at the ultrastructural level. Incubation experiments of epididymal spermatozoa in EGTA-containing solutions in the absence of calcium resulted in a gradual labilization and eventual loss of the plasma membrane of the sperm middle piece. After removal of (at least part of) the plasma membrane, bound major protein could be visualized immunohistochemically close to the mitochondria of the middle piece using a gold-labeled primary or secondary antibody. The acceptor site for major protein therefore seems to reside inside the plasma membrane of the sperm middle piece. Incubation of epididymal spermatozoa in phospholipase-containing solutions removed the acceptor site from the spermatozoa. Separation by polyacrylamide treatment of proteins from epididymal sperm cells extracted by sodium hydroxide or phospholipase treatment, subsequently transblotted on nitrocellulose sheets and directly labeled with gold-tagged major protein, demonstrated a protein duplet with a molecular weight of 65 and 67 kDa, respectively, which appears to represent the specific binder of major protein underneath the sperm surface. A rapid screening assay for Campylobacter fetus in bull semen was developed using the polymerase chain reaction (PCR) and restriction endonuclease analysis (REA) to complement isolation by culture. An oligonucleotide primer pair (C/C) from the hypervariable region of 16S rRNA of C. fetus was used to amplify a 362 base pair fragment by PCR. The PCR/REA assay, which is completed in 10 hours, detected as few as three C. fetus subsp. venerealis cells in experimentally infected raw bull semen and in semen diluted with milk or egg yolk Tris (EYT). All the strains tested, of both subspecies of C. fetus, were amplified, as were some other Campylobacter species. Restricting the amplified products by AluI differentiated C. fetus from the other organisms. There was no visible product generated by PCR from C. sputorum subsp. bubulus, a saprophytic organism found in the prepuce of bulls, or from seven other species of bacteria found in semen. A modification of the PCR assay, using another primer pair (C/C) and two temperature PCR cycling conditions, increased the probability of detecting C. fetus subsp. venerealis. PCR amplification followed by REA could be used to screen bovine semen rapidly for C. fetus. Consecutively collected semen samples from a breeding bull were found to be contaminated with Pseudomonas aeruginosa. Palpation through the bull's scrotum revealed inflammatory changes suggestive of chronic orchiepididymitis in one testicle. For 10 months, all the bull's 13 ejaculates were discarded because the post-thaw viability was <20%. Norfloxacin nicotinate was injected intramuscularly into the bull at 5 mg/kg daily for 7 days. Serum and semen samples were collected at 24-h intervals during the course of treatment and afterwards and were assayed for NFN concentrations. Drug concentrations in the semen, by microbiological assay, during treatment and up to 120 h after the last treatment ranged from 2.6 to 5.1 µg/ml, 14.2 to 43.2 times the corresponding serum drug levels. P. aeruginosa was not isolated from the semen 4 or 15 days after the last injection but was re-isolated after 32 and 64 days. A second similar course of NFN was administered and P. aeruginosa was not isolated from semen samples collected on four occasions, 6, 22, 44 and 94 days after the last treatment. The ultrastructure of vacuolar formation in the nucleus of the Sertoli cells in normal adult bulls is described. It occurs regularly in all the Sertoli cells instead of the typically arranged nucleolus. It consists of membrane-limited vesicles bearing 150 Å granules on their outer surface and a smaller, fibrillar component composed of coiled filaments 50 Å in diameter. Free and membrane attached ribosomes, and polyribosomes found in the numerous folds of the nuclear membrane indicate RNA synthesis. The possible role of the vacuolar formation, named multivesicular nuclear body, in the ribosomal RNA synthesis is briefly discussed. [ABSTRACT FROM AUTHOR]

The aim of this study was investigation of polymorphism in somatotropin ( GH), prolactin ( PRL), leptin ( LEP), and thyreoglobulin ( TG) genes in thoroughbred and crossbred with Holstein breed bulls. Our results are in agreement with previously published literature data, and we found allele frequencies in bulls including 0.86 (L) and 0.14 (V) for the GH gene, 0.87 (A) and 0.13 (B) for the PRL gene, 0.59 (C) and 0.41 (T) for the LEP gene, and 0.81 (C) and 0.19 (T) for the TG gene. Fibropapilloma of the penis is caused by the bovine papillomavirus. Two 12- and 18-month-old bulls were presented with a history of gradually enlarging mass on the glans of penis. Grossly, the masses were multinodular proliferations, of varied sizes mainly between 0.5 and 5 cm in diameter, well circumscribed, and nonpigmented and were attached to the glans by a relatively broad pedicle. Histopathologically, the masses were papillomatous features with irregular hyperplasia and epidermal rete ridges. The dermis consisted of densely packed irregular collagenous connective tissue with many fibroblasts. In the epidermis, orthokeratotic hyperkeratosis, many koilocytes with variably sized keratohyalin granules, and rare presence of intranuclear inclusion bodies were present. The morphologic patterns were typical of fibropapillomas of papillomavirus etiology. In one case, recurrence of lesion occurred about 2 months after surgical treatment. [ABSTRACT FROM AUTHOR]
