Mixotrophy, a physiological trait combining autotrophy and heterotrophy in one organism, significantly contributes to energy and matter transfer in aquatic ecosystems. However, understanding how environmental factors influence mixoplankton success across freshwater ecosystems has been uncertain. The grand écart hypothesis (GEH) posits that light and nutrient availability are key components of mixotrophs' niche, suggesting that ecosystem properties determine opposing gradients of light and nutrients, creating environmental filtering for mixotrophs. We hypothesized that ecosystem size, a property of lake ecosystems, mediates the prevalence of patterns and control mechanisms predicted by the GEH on mixoplankton relative biomass (MRB). Using data from 98 tropical lakes, we demonstrated that lake size mediates the inverse relationship between light and nutrient availability across ecosystems. Larger lakes have more light but low nutrients, while smaller lakes have more nutrients but greater shading. Light availability better explains MRB in small lakes, and nutrients better explain MRB in large lakes, with MRB values being higher in small lakes, with secondary influence from zooplankton herbivory. Our results validate the GEH as a significant framework for explaining patterns and control mechanisms of mixoplankton across tropical lakes. This study highlights the significance of lake size as an ecosystem property that generates opposing light and nutrient gradients, further emphasizing its importance for understanding mechanisms regulating freshwater phytoplankton community structure and functioning. Mixotrophy is a combination of photosynthesis and direct access to organic carbon sources, mainly through osmotrophy or phagotrophy. This strategy is adopted by several, phylogenetically distinct, phytoplankton groups and is commonly occurring in marine, brackish and freshwater ecosystems. Traditionally, it has been put in relation to both scarcity of inorganic nutrients and poor light conditions. However, we observed blooms of the mixotrophic, toxic haptophyte Prymnesium parvum in different periods of the year and under variable resources availability. The analysis of a 6.5-year data set of phytoplankton weekly records from a Sicilian shallow lake (Biviere di Gela, south-eastern Sicily) allowed us to hypothesise that a depleted condition as regards inorganic nutrients is not the main fuel to the growth of P. parvum, neither this is due to light limitation. The results achieved show that an increased availability in suitable preys can stimulate the growth of this phagotrophic photoautotroph. Contemporarily, it was not found any clear environmental patterns to explain species dominance and growth patterns as related to inorganic nutrient availability. Moreover, it is shown that these organisms tend to monopolise resources when these become available irrespective of seasons, and under variable conditions as regards inorganic nutrient availability. We studied the phagotrophic ability of dinoflagellate strain Karenia mikimotoi KM-Lü (isolated from the South China Sea), using fluorescent microspheres, bacteria isolated from the culture of K. mikimotoi and a marine microalgae Isochrysis galbana. We found that K. mikimotoi cultured under conditions of high light intensity could ingest fluorescent microspheres (diameters 0.5 and 2.0 μm) and fluorescence-labeled bacteria and microalgae. Under a low light intensity, however, only fluorescent microspheres (diameter 0.5 μm) and fluorescence-labeled microalgae were ingested. K. mikimotoi showed better growth by ingesting living marine bacteria or microalgae I. galbana than the controls, either in nutrient-depleted or nutrient-replete conditions. In nutrient-depleted conditions, the growth of K. mikimotoi was more significant with I. galbana as the prey item. In conclusion, the harmful dinoflagellate K. mikimotoi from the South China Sea has apparent phagotrophic ability, and some marine bacteria and microalgae may promote the growth of K. mikimotoi. Lateral gene transfer – the transfer of genetic material between species – has been acknowledged as a major mechanism in prokaryotic genome evolution for some time. Recently accumulating data indicate that the process also occurs in the evolution of eukaryotic genomes. However, there are large rate variations between groups of eukaryotes; animals and fungi seem to be largely unaffected, with a few exceptions, while lateral gene transfer frequently occurs in protists with phagotrophic lifestyles, possibly with rates comparable to prokaryotic organisms. Gene transfers often facilitate the acquisition of functions encoded in prokaryotic genomes by eukaryotic organisms, which may enable them to colonize new environments. Transfers between eukaryotes also occur, mainly into larger phagotrophic eukaryotes that ingest eukaryotic cells, but also between plant lineages. Phytoplankton have traditionally been regarded as strictly phototrophic, with a well defined position at the base of pelagic food webs. However, recently we have learned that the nutritional demands of a growing number of phytoplankton species can be met, at least partially, or under specific environmental conditions, through heterotrophy. Mixotrophy is the ability of an organism to be both phototrophic and heterotrophic, in the latter case utilizing either organic particles (phagotrophy) or dissolved organic substances (osmotrophy). This finding has direct implications for our view on algal survival strategies, particularly for harmful species, and energy- and nutrient flow in pelagic food webs. Mixotrophic species may outcompete strict autotrophs, e.g. in waters poor in inorganic nutrients or under low light. In the traditional view of the ‘microbial loop’ DOC is thought to be channeled from algal photosynthesis to bacteria and then up the food chain through heterotrophic flagellates, ciliates and mesozooplankton. Are mixotrophic phytoplankton that feed on bacteria also significantly contributing to this transport of photosynthetic carbon up the food chain? How can we estimate the fluxes of carbon and nutrients between different trophic levels in the plankton food web involving phagotrophic algae? These questions largely remain unanswered. Bacterivory was detected by incorporation of 0.57 μm diameter, fluorescent polystyrene beads and fluorescently labeled bacteria (FLB) in two cultured species of Cryptomonas (C. ovata and C. erosa), and a population of Cryptomonas sp in a humic, mesotrophic lake. Rates of ingestion and clearance were very low, and similar for the cultures and the in situ population. The in situ population incorporated 0.7-1.7 bacteria cell h, thereby ingesting 0.3%-2.0% of the total bacterial numbers present in the water per day, and receiving less than 2% of its carbon content per day through bacterivory. Thus, bacterivory by Cryptomonas was quantitatively important neither as a sink for bacterial biomass, nor as a carbon source for the algal cells. The cytoskeleton of flagellate chromophyte algae, zoospores and gametes is active during swimming, phototaxis, several types of phagotrophic feeding, the formation, secretion and deployment of silica-scales, and the abrupt movement of spine-scales. The flagellar basal bodies are anchored by microtubular roots and/or fibrous roots. The kinds, numbers, and paths of these roots are characteristic of different taxonomic groups within the chromophytes. There are more differences in flagellar apparatuses for taxonomic classes dominated by flagellates as compared to classes dominated by coccoid, filamentous, or parenchymatous forms. Swimming cells that exhibit phototaxis often contain an autofluorescent substance that is located at the base of one flagellum. Phagotrophy occurs in flagellates of several distantly related taxonomic classes, suggesting that phagotrophy evolved independently several times. The most complex phagotrophic process occurs in the Chrysophyceae where one microtubule of a flagellar root forms a feeding basket or pouch into which food particles are moved. The silica-scales of the Synurophyceae are formed, secreted and finally moved into position outside the cell by cytoskeletal components. The six spinescales of Apedinella (Pedinellophyceae) lie outside the plasma membrane, but they are attached by microligaments and are repositioned almost instantly by a cytoskeletal complex of actin, centrin, and microtubules. A phylogenetic classification based upon a cladistic analysis suggests that aquatic fungi are natural members of the chromophyte group. [ABSTRACT FROM AUTHOR]

Mixotrophy seems to be widespread among phytoplankton, but whether this strategy is more relevant in oligotrophic lakes remains unclear. Here, we tested the hypothesis that the relative abundance of mixotrophic flagellates in lakes increases along an elevational gradient paralleling increasingly oligotrophic conditions. For this purpose, 12 lakes located between 575 and 2796 m above sea level were sampled in summer and fall to include two different seasonal windows in phytoplankton dynamics and environmental conditions. The degree of mixotrophy in phytoplankton was estimated in tracer experiments using fluorescently-labeled bacteria and done with composite samples collected in the euphotic zone and in samples obtained from the chlorophyll-a maximum. The results indicated the existence of a positive trend particularly in summer in the relative abundance of mixotrophic flagellates with elevation, however, this trend was not linear, and exceptions along the elevational gradient were found. Changes in the relative abundance of mixotrophic flagellates were related with significant changes in water transparency, DOC and phosphorus concentrations, as well as in bacterial and flagellate abundance. Overall, our results reveal that the harsh growth conditions found in oligotrophic high mountain lakes favor a mixotrophic trophic strategy among phytoplankton. [ABSTRACT FROM AUTHOR]
