Abstract

Described is a hybrid metal additive manufacturing (AM) method that integrates in situ laser shock peen (LSP) forming with laser powder bed fusion (PBF) to mitigate vertical distortions during part builds. LSP has recently been proposed to reduce tensile residual stresses during selective laser melting (SLM). The effects of LSP on part distortion, however, have not been rigorously examined. It is proposed here that SLM can be integrated with in situ LSP forming to reduce distortion of the upper surface of parts during or after printing. To study the distortion correction capability, a 2-stage computational framework is created, which includes physics-based models of the SLM process and LSP treatment. Stage 1 includes thermomechanical SLM simulation to predict surface geometry and is applied to model four 50-μm layers of a 316L part having a 4 mm × 4 mm footprint. Stage 2 of the framework includes an elastic-plastic thermomechanical shock-wave simulation to predict LSP surface treatment forming effects. Surface distortion is examined for varying laser spot size, overlap, and part temperatures from 300 to 500 K, using a nanosecond-pulsed infrared laser. For the 316L SLM sample, the upper surface is predicted to have $\sim $ 9-μm vertical distortion on the 200-μm 4-layer build. With a 2-μm allowable distortion, only 44.13% of the surface initially conforms. After one LSP forming treatment at 300 K, conformance improves to 84.75%. After a third LSP forming, with 50% laser power-density increase, surface conformance increases to 91%, demonstrating potential of the hybrid AM-LSP process in reducing finish-machining.

Abstract

A multiscale simulation method is established to study the microstructural responses of near-surface grain boundary structures of copper subjected to ultrashort femtosecond laser pulse. By integrating a two-temperature model with molecular dynamics, the presented approach allows for incorporation of both laser processing parameters and microstructures, enabling systematic simulation studies on the process-properties link. Following a brief introduction of the simulation methodology, a detailed modeling study on the ultrashort laser-material interaction is presented. In particular, we highlight the effects of laser process parameters on the near-surface response and corresponding phase change, formation of voids and their growth, and mechanism of dislocation nucleating and propagating from grain boundary.

Abstract

Accumulating evidence indicates that the neuropeptide substance P (SP) is predominantly involved in neurogenic inflammation and pain perception via its high-affinity neurokinin 1 receptor (NK-1R). Intriguingly, decreased pain sensitivity is found to be associated with high plasma progesterone levels. We hypothesize that progesterone may attenuate nociception and associated inflammatory response via NK-1R-dependent pathways. To address our hypothesis, we incubated splenic lymphocytes from CBA/J female mice with different concentrations of the progesterone derivative dydrogesterone. Subsequently, the expressions of NK-1R and T helper (Th1)-type cytokines were analyzed by flow cytometry. Next, we subcutaneously injected CBA/J mice with 1.25 mg of dydrogesterone in 200-microl sesame oil; control mice were sham-injected. Tail flick test to detect the nociceptive threshold was performed in 30-min intervals upon injection. Lymphocytes were isolated from blood and uterus and analyzed for NK-1R surface expression. Immunohistochemical analyses were performed to investigate the uterine tissue distribution of NK-1R. Dydrogesterone induced a decrease in the percentage of NK-1R+ lymphocytes in vitro and in vivo. Additionally, an increase in Th2-type and a decrease in Th1-type cytokines could be detected in vitro after incubation with dydrogesterone. An increased tail flick latency following dydrogesterone injection supported the concept that decreased expression of the NK-1R on lymphocytes is associated with an increased pain threshold. Taken together, these results clearly reveal a pathway by which dydrogesterone or progesterone respectively modulates the cross talk of the nervous, endocrine and immune systems in inflammation and pain.

Abstract

Twelve diseases, most with neuropsychiatric features, arise from trinucleotide repeat expansion mutations. Expansion mutations may also cause a number of other disorders, including several additional forms of spinocerebellar ataxia, bipolar affective disorder, schizophrenia, and autism. To obtain candiate genes for these disorders, cDNA libraries from adult and fetal human brain were screened at high stringency for clones containing CAG repeats. Nineteen cDNAs were isolated and mapped to chromosomes 1, 2, 4, 6, 7, 8, 9, 12, 16, 19, 20, and X. The clones contain between 4 and 17 consecutive CAG, CTG, TCG, or GCA triplets. Clone H44 encodes 40 consecutive glutamines, more than any other entry in the nonredundant GenBank protein database and well within the range that causes neuronal degeneration in several of the glutamine expansion diseases. Eight cDNAs encode 15 or more consecutive glutamine residues, suggesting that the gene products may function as transcription factors, with a potential role in the regulation of neurodevelopment or neuroplasticity. In particular, the conceptual translation of clone CTG3a contains 18 consecutive glutamines and is 45% identical to the C-terminal 306 residues of the mouse numb gene product. These genes are therefore candidates for diseases featuring anticipation, neurodegeneration, or abnormalities of neurodevelopment.
