Peach is a popular and important tree fruit widely produced in the world, and the production of high-quality peach fruit does require management of pests and diseases. In this review, major peach diseases from China, Spain, and USA are described in detail for the benefit of producers, consultants, researchers, and other interested parties. Minor diseases of concern in these countries are also described. Current progress on pathogen resistance to major chemical classes of fungicides as well as current resistance management practices are discussed. Specific cultural practices applied in China, Spain, and USA are also described to provide an overview of peach disease management. A ‘Future Outlook’ section is included at the end of this review to highlight the challenges and opportunities for disease management in the future.

Variation in susceptibility of 28 Prunus rootstock genotypes to the causal agents of Armillaria root rot, Armillaria mellea, A. solidipes, and Desarmillaria tabescens, were studied by conducting in vitro root screening assays. Root segments with wounded and intact periderm were placed next to and on the top of the fungal cultures. At day 21, the percent success of fungal penetration and the circumferential and longitudinal lengths of fungal growth were measured. A parallel investigation using the inoculated root segments was carried out to characterize the active host defense mechanisms involved, including anatomical responses in bark and wood. Overall, the success of penetration and the longitudinal and circumferential spread of Armillaria spp. and D. tabescens were significantly different among various Prunus rootstock genotypes. None of the tested rootstock genotypes were completely resistant to infection. However, plum and plum derived rootstocks, and some of the cherry genotypes were less susceptible to infection compared to the peach genotypes. The host’s ability to limit infection by Armillaria spp. and D. tabescens was not limited to a single mechanism but appeared to be regulated by several collective nonspecific host responses acting together. Differential levels of a series of non-specific coordinated events were triggered in Prunus genotypes such as the formation of new callus tissue on the root surface, a colored reaction zone, necrophylactic periderm, new cells, and new vascular cambium to compartmentalize the pathogen. The host responses were elevated in the genotypes with low level of infection as compared to highly infected genotypes.

Penja pepper (Piper nigrum) produced in Cameroon has long been recognized for its exceptional organoleptic quality. The pepper vine is grown using a support tree (Spondias mombin) in Cameroon. A root disease is associated with plant deaths in both plants. The disease symptoms are characterized by collar cracking and gummosis and the disease was tentatively identified as Armillaria root rot. In this work the extent of the problem was characterized by surveying 35 farms in Cameroon. Samples were taken from diseased support trees and pepper vines. Support trees exhibiting typical symptoms were found in approximately one third of the surveyed farms. In these farms, disease incidence and Pepper vine mortality ranged from 1.3 to 50% and 1.2 to 87.7%, respectively. Analysis of the ribosomal internal transcribed spacer region (ITS) locus suggested that the isolates collected from S. mombin and P. nigrum are most likely, which is a first for both hosts, represented by Armillaria camerunensis (Henn.) Volk & Burdsall, a fungus previously associated with declining cacao trees in Cameroon. Given the increasing demand for Penja pepper and the great damaging potential of this root rot, research on management strategies should be prioritized.

Species of Armillaria are distributed globally and include some of the most important pathogens of forest and ornamental trees. Some of them form large long-living clones that are considered as one of the largest organisms on earth and are capable of long-range spore-mediated transfer as well as vegetative spread by drought-resistant hyphal cords called rhizomorphs. However, the virus community infecting these species has remained unknown. In this study we used dsRNA screening and high-throughput sequencing to search for possible virus infections in a collection of Armillaria isolates representing three different species: Armillaria mellea from South Africa, A. borealis from Finland and Russia (Siberia) and A. cepistipes from Finland. Our analysis revealed the presence of both negative-sense RNA viruses and positive-sense RNA viruses, while no dsRNA viruses were detected. The viruses included putative new members of virus families Mymonaviridae, Botourmiaviridae and Virgaviridae and members of a recently discovered virus group tentatively named “ambiviruses” with ambisense bicistronic genomic organization. We demonstrated that Armillaria isolates can be cured of viruses by thermal treatment, which enables the examination of virus effects on host growth and phenotype using isogenic virus-infected and virus-free strains.

Draft genomes of Penicillium roqueforti, Fusarium sororula, Chalaropsis populi, and Chrysoporthe puriensis are presented. Penicillium roqueforti is a model fungus for genetics, physiological and metabolic studies, as well as for biotechnological applications. Fusarium sororula and Chrysoporthe puriensis are important tree pathogens, and Chalaropsis populi is a soil-borne root-pathogen. The genome sequences presented here thus contribute towards a better understanding of both the pathogenicity and biotechnological potential of these species.

The genomes of Armillaria fuscipes, Ceratocystiopsis minuta, Ceratocystis adiposa, Endoconidiophora laricicola, E. polonica, and Penicillium freii DAOMC 242723 are presented in this genome announcement. These six genomes are from plant pathogens and otherwise economically important fungal species. The genome sizes range from 21 Mb in the case of Ceratocystiopsis minuta to 58 Mb for the basidiomycete Armillaria fuscipes. These genomes include the first reports of genomes for the genus Endoconidiophora. The availability of these genome data will provide opportunities to resolve longstanding questions regarding the taxonomy of species in these genera. In addition these genome sequences through comparative studies with closely related organisms will increase our understanding of how these pathogens cause disease.

During May 2010, sporocarps of what appeared to be an Armillaria sp. were found in large clumps in historic Kirstenbosch Botanical Gardens on the foot of Table Mountain, Cape Town, South Africa. These sporocarps could be physically linked to the roots of unidentified dead trees and Protea spp. The aim of this study was to identify the Armillaria sp. found fruiting in Kirstenbosch. To achieve this goal isolates were made from the mycelium under the bark of dead roots linked to sporocarps. The ITS and IGS-1 regions were sequenced and compared to sequences of Armillaria spp. available on GenBank. Cladograms were generated using ITS sequences to determine the phylogenetic relationship of the isolates with other Armillaria spp. Sequence comparisons and phylogenetic analyses showed that the isolates represented A. mellea. They were also identical to isolates of this species previously discovered in the Company Gardens in South Africa and introduced from Europe apparently by the early Dutch Settlers. Armillaria mellea is alien and apparently invasive in Cape Town, fruits profusely and has the potential to spread to sensitive native forests on the foothills of the City.

Trap bags were used to assess the presence of anArmillaria species in aPinus koraiensis plantation in Heilongjiang, China. 216 trap bags containing bark were placed in three plots at 2 × 2m spacing. After 10 weeks, the presence ofArmillaria was determined from the growth of rhizomorphs in the trap bags. The trap bag method proved successful in determining the presence of viableArmillaria rhizomorphs where only a few dead trees showed signs of the disease. The distribution of positive traps corresponded generally with the presence of rhizomorphs in the soil taken from the trap holes. However, the trap bags were superior to soil samples for evaluating the presence of viable rhizomorphs. The fresh, abundant rhizomorphs were easily identified by persons inexperienced in the assessmentof Armillaria root rot. Several practical problems need to be solved before the method is applicable in general forest management in China.

ASEXUALLY reproducing organisms occur in a variety of taxa in all biological kingdoms1 and distinguishing asexually propagated genotypes is essential for the understanding of their population biology. Among the higher fungi, however, the clonal 'individual' is especially difficult to define2 because most of the fungal thallus consists of a network of anastamosing hyphae embedded in the substratum. Whether fruit-bodies, the most recognizable part of a fungus, are produced by a single supporting mycelium can only be determined by establishing direct physiological continuity or genetic identity. We report a means by which individual fungi can be unambiguously identified within local populations and identify an individual of Armillaria bulbosa that occupies a minimum of 15 hectares, weighs in excess of 10,000kg, and has remained genetically stable for more than 1,500 years.

Armillaria species are important root rot pathogens with a wide host range and a worldwide distribution. The taxonomy of these fungi has been problematic for many years but the understanding of the relationships between them has been substantially improved through the application of DNA sequence comparisons. In this study, relationships between different Armillaria species were determined using elongation factor 1-alpha DNA sequence data for the first time. A total of 42 isolates, representing the majority of Armillaria species, with diverse geographic distributions and hosts, were included in this study. PCR amplification yielded products of 600 bp for all the isolates. Phylogenetic trees resulting from parsimony analysis showed that this gene region is useful for studying relationships between species. Generally, results were similar to those emerging from previous comparisons using ITS and IGS-1 sequence data. Phylogenetic trees generated from the dataset grouped the African taxa in a strongly supported clade, basal to the rest of the Armillaria species included in the study. The Armillaria species originating from the Northern Hemisphere formed a monophyletic group. Within this group, isolates of A. mellea constituted four subclades, representing their geographical origin. The phylogenetic relationships among species from the Southern Hemisphere were not entirely resolved. However, A. pallidula, A. fumosa and A. hinnulea grouped in a strongly supported clade and isolates of A. limonea formed a sister clade with those of A. luteobubalina. This is the first time a single-copy protein coding gene has been used to study phylogenetic relationships in Armillaria, and overall the data support previously held views regarding the relationships between species.

A specific molecular diagnostic test was developed to aid in the identification of Australian Armillaria species. The test involves polymerase chain reaction (PCR) amplification using the primer pair A1F and A2R followed by restriction cleavage by Taq 1. Sample populations of the four predominant Australian species, A. luteobubalina, A. hinnulea, A. fumosa and A. novae-zelandiae were collected in the form of basidiome tissue, diploid vegetative culture or infected root material. All samples were correctly identified as Armillaria using the test and subsequently identified to species level by comparing the Taq 1 restriction patterns. Variation was observed within A. luteobubalina Taq 1 restriction patterns, which suggested the presence of sexual recombination within the population. This procedure will assist in the diagnosis of Armillaria root rot and enable an accurate species identification to be made from infected root material or basidiome tissue.

Following South African reports of the effectiveness of phosphonic acid for control of non-Oomycete diseases, preliminary testing of phosphonic acid was made for control of root and foliar diseases of stonefruit, apples and celery. Phosphonic acid showed potential for control of stonefruit Armillaria root rot (caused by Armillaria luteobuballna), apple white root rot (caused by Dematophora necatrix), and apple black spot (caused by Venturia inaequalis) but was ineffective for peach brown rot (caused by Monilinia fructicola) and celery leaf spot (caused by Septoria apiicola).
