The frontal eye field (FEF) and the inferior frontal junction (IFJ) are prefrontal structures involved in mediating multiple aspects of goal-driven behavior. Despite being recognized as prominent nodes of the networks underlying spatial attention and oculomotor control, and working memory and cognitive control, respectively, the limited quantitative evidence on their precise localization has considerably impeded the detailed understanding of their structure and connectivity. In this study, we performed an activation likelihood estimation (ALE) fMRI meta-analysis by selecting studies that employed standard paradigms to accurately infer the localization of these regions in stereotaxic space. For the FEF, we found the highest spatial convergence of activations for prosaccades and antisaccades paradigms at the junction of the precentral sulcus and superior frontal sulcus. For the IFJ, we found consistent activations across oddball/attention, working memory, Stroop, and task-switching paradigms at the junction of the inferior precentral sulcus and inferior frontal sulcus. We related these clusters to previous meta-analyses, sulcal/gyral neuroanatomy, and a recent comprehensive brain parcellation, highlighting important differences compared to their results and taxonomy. Finally, we employed the ALE peak coordinates as seeds to perform a meta-analytic connectivity modeling (MACM) analysis, which revealed systematic coactivation patterns spanning the frontal, parietal and temporal cortices. We decoded the behavioral domains associated with these coactivations, suggesting that these may allow FEF and IFJ to support their specialized roles in flexible behavior. Our study provides meta-analytic groundwork for investigating the relationship between functional specialization and connectivity of two crucial control structures of the prefrontal cortex.

Background Acute kidney injury (AKI) is a severe complication of coronavirus disease-2019 (COVID-19). This study aims to evaluate incidence, risk factors and case-fatality rate of AKI in patients with COVID-19. Methods We reviewed the health medical records of 307 consecutive patients with COVID-19 hospitalized at the University Hospital of Modena, Italy. Results AKI was diagnosed in 69 out of 307 (22.4%) COVID-19 patients. Stages 1, 2, or 3 AKI accounted for 57.9%, 24.6% and 17.3%, respectively. AKI patients had a mean age of 74.7 ± 9.9 years. These patients showed higher serum levels of the main markers of inflammation and higher rate of severe pneumonia than non-AKI patients. Patients with COVID-19 may experience multiple conditions (e.g., fever, hyperventilation, anorexia, gastroenteritis, acid-base disorder) that may cause electrolyte imbalances. Hypokalemia is a concerning electrolyte disorder that may increase the susceptibility to various kinds of arrhythmia. This study aimed to estimate prevalence, risk factors and outcome of hypokalemia in a cohort of non-critically ill patients. A retrospective analysis was conducted on 290 hospitalized patients with confirmed COVID-19 infection at the tertiary teaching hospital of Modena, Italy.Hypokalemia (Hypokalemia is a frequent disorder in COVID-19 patients and urinary potassium loss may be the main cause of hypokalemia. The disorder was mild in the majority of the patients and was unrelated to poor outcomes. Nevertheless, hypokalemic patients required potassium supplements to dampen the risk of arrhythmias.

Purpose Post-neurosurgical infection caused by extensively drug resistant Pseudomonas aeruginosa (XDR-PA) are becoming a matter of great concern due to limited therapeutic options. Although not approved for these indications, the new BetaLactam-BetaLactamase Inhibitor combinations (BLBLIs) could represent a valid salvage treatment. We describe one nosocomial meningitis and two cervical osteomyelitis due to an XDR-PA who were treated with ceftazidime/avibactam (CZA) and ceftolozane/tazobactam (C/T) and review the literature. Methods The first and the third patients developed an osteomyelitis following cervical stabilization surgery due to an XDR-PA. Although the first patient started treatment with a high dose of C/T, resistance to C/T occurred, so therapy was switched to CZA plus aztreonam. The third patient switched to aztreonam plus CZA due to development of acute kidney injury during therapy with colistin. The second patient had an XDR-PA meningitis following the insertion of an external ventricular catheter and he was treated with C/T plus meropenem and amikacin. Results All three cases reported were successfully conservatively treated thanks to the use of the new BLBLIs with different combinations. Only few experiences demonstrated an equally favorable outcome: one patient treated with C/T plus fosfomycin for otogenic meningitis caused by an XDR-PA and another case of XDR-PA post-surgical meningitis with CZA in combination with colistin. Finally, the combination of CZA plus aztreonam has proven to be effective on XDR-PA only in limited mostly in vitro studies. Conclusion These recently developed antibiotics, C/T and CZA are promising and complementary therapy options against post-neurosurgical hard-to-treat P. aeruginosa infections. Further prospective real-life studies are required to validate these findings in this special setting.

Artemisia caerulescens L. is a rare aromatic perennial shrub growing in saline or brackish environments of the central and western part of the Mediterranean region. On the other hand, despite its distribution in unstable and highly selective environments such as salt marshes, no data are available on its ecological adaptability or on its growth strategy. In an effort to fill these gaps, we performed three experiments in order to evaluate the effects of selected environmental abiotic factors on the seed germination of A. caerulescens. In the first experiment, we determined the effect of different temperatures, photoperiod regimes and salinity on the germination of 120 days old seeds. Secondly, we tested the germination behaviour of the seeds in relation to seed after-ripening time and salinity. The results showed that a temperature regime of 10/20°C and a photothermoperiod of 12/12 h were optimal conditions for seed germination. Salinity affected the final germination rate and mean time to germination. Until 60 days of after-ripening, the seeds exhibited a primary dormancy state. Then, their germination performance was affected by seed after-ripening time and salinity. The data obtained in this study allow us to outline a fairly accurate picture of the ecological requirements of A. caerulescens during germination and of the degree of viability of its seeds at least in the first year of their life. The significant different susceptibility of the seeds to salinity leads to the conclusion that the spread of this species by seeds strongly depends on soil salinity fluctuations. Knowledge of optimal germination requirements could also be useful for the possible use of plant seeds as propagation material in restoration and biodiversity conservation programmes.

The first thousand days of life from conception have a significant impact on the health status with short, and long-term effects. Among several anthropometric and maternal lifestyle parameters birth weight plays a crucial role on the growth and neurological development of infants. Recent genome wide association studies (GWAS) have demonstrated a robust foetal and maternal genetic background of birth weight, however only a small proportion of the genetic hereditability has been already identified. Considering the extensive number of phenotypes on which they are involved, we focused on identifying the possible effect of genetic variants belonging to taste receptor genes and birthweight. In the human genome there are two taste receptors family the bitter receptors (TAS2Rs) and the sweet and umami receptors (TAS1Rs). In particular sweet perception is due to a heterodimeric receptor encoded by the TAS1R2 and the TAS1R3 gene, while the umami taste receptor is encoded by the TAS1R1 and the TAS1R3 genes. We observed that carriers of the T allele of the TAS1R1-rs4908932 SNPs showed an increase in birthweight compared to GG homozygotes Coeff: 87.40 (35.13–139.68) p-value = 0.001. The association remained significant after correction for multiple testing. TAS1R1-rs4908932 is a potentially functional SNP and is in linkage disequilibrium with another polymorphism that has been associated with BMI in adults showing the importance of this variant from the early stages of conception through all the adult life.

An increased awareness on neonatal pain-associated complications has led to the development of pain scales adequate to assess the level of pain experienced by newborns such as the ABC score. A commonly used analgesic procedure is to administer a 33% oral dextrose solution to newborns prior to the painful intervention. Although this procedure is very successful, not in all subjects it reaches complete efficacy. A possible explanation for the different response to the treatment could be genetic variability. We have investigated the genetic variability of the OPRM1 gene in 1077 newborns in relation to non-pharmacologic pain relief treatment. We observed that the procedure was successful in 966 individuals and there was no association between the genotypes and the analgesic efficacy when comparing individuals that had an ABC score = 0 and ABC score >0. However, considering only the individuals with ABC score>0, we found that the homozygous carriers of the G allele of the missense variant SNP rs1799971 (A118G) showed an interesting association with higher ABC score. We also observed that individuals fed with formula milk were more likely to not respond to the analgesic treatment compared to those that had been breastfed.

The fig tree weevil Aclees sp. cf. foveatus (Coleoptera: Curculionidae), introduced in Italy in 2005, is currently causing significant economic and environmental losses to fig tree nurseries and orchards in Central Italy. Fig damages are due to the adults feeding on leaves and fruits, and to the galleries dug by the xylophagous larvae in the trunk, which lead the plants to death. To date, no chemical or biological control methods resulted to be effective against this invasive pest. In order to gain information about possible semiochemicals involved in mate recognition and choice, both the volatile organic compounds (VOCs) and the epicuticular lipids of male and female specimens were analysed. VOCs emissions of specimens were characterized essentially by monoterpenes, while epicuticular lipids contained long chained 2-ketones, alkanes, alkenes, including some methyl alkenes, and several fatty acid propyl esters. The attractiveness of reconstituted VOCs blends of the two sexes was tested in electrophysiological and behavioural assays in laboratory conditions. Both the male and the female reconstituted VOCs drove a significant response towards individuals of the opposite sex, thus demonstrating features of sexual attractants. Our results suggest a possible application of VOCs blends as pheromonic attractants in field monitoring and mass trapping of Aclees sp. cf. foveatus.

The Sarfartoq carbonatite complex occurs in the transition zone between Archaean and Paleoproterozoic gneisses in southern West Greenland. The Sarfartoq carbonatite complex hosts ore deposits and occurrences of rare earth elements and niobium. In this study, airborne HyMap hyperspectral imagery and coincident airborne gamma-ray spectroscopy data were analysed for mineral exploration of Sarfartoq carbonatite complex. The study area included the carbonatite core zone and a hydrothermally altered shear zone prospective for rare earth elements mineralization in the southern margin of the carbonatite complex. The analysis of the hyperspectral imagery was focused on the mapping of the spatial distribution of carbonatite, fenite and limonite. Significant anomalies of uranium, thorium and potassium were outlined from the gamma-ray spectroscopy data. Decision trees were used to combine the results from the hyperspectral image analysis and airborne gamma-ray spectroscopy. Significant anomalies of thorium at the hydrothermally altered shear zone coinciding with the presence of carbonatite rocks are generally indicative of outcropping rare earth element mineralization. The study indicates the benefits of coincident airborne hyperspectral and gamma-ray spectroscopy surveys for the mineral exploration of carbonatite complexes.

Orbicules are sub- or pauci-micronic sporopollenin-coated particles associated with pollen grains. Recent studies have highlighted their importance in terms of systematic value or as vectors of allergens that may trigger pollinosis. While orbicules have indeed been reported for most families and genera of allergological importance, their usefulness in systematics remains little explored. Our study aims: (a) to discover the presence/absence of orbicules in species of allergological interest hitherto overlooked regarding these particles; (b) to propose standard descriptors for orbicule morphology; and (c) to link the features of orbicules to a systematic framework. We observed or measured selected orbicule traits in SEM micrographs of pollen sacs and pollen grains of 23 species of 17 genera in Asteraceae, Betulaceae, Fagaceae, Oleaceae, Poaceae, Plantaginaceae and Polygonaceae families. Categorical and continuous traits were included in a matrix and then subjected to NMDS as ordination method to assess the usefulness of orbicule traits as systematic characters. We detected orbicules both on the locule wall and on the pollen exine in almost all investigated species and described unknown nanoparticles smaller than orbicules. Based on orbicules traits, NMDS separated the samples at species level. We detected and described orbicules on the tapetum and on the pollen exine in 19 target species and observed unknown nanoparticles that might explain (a) the process of deposition of sporopollenin during orbicule development and (b) the onset of pollinosis before and after the dispersion of pollen grains into the air. Finally, within the restricted sample examined, selected orbicule traits allowed to resolve the data set at species level.

Lipid A is the lipophilic region of lipopolysaccharides and lipooligosaccharides, the major components of the outer leaflet of most part of Gram-negative bacteria. Some lipid As are very promising immunoadjuvants. They are obtained by extraction from bacterial cells or through total chemical synthesis. A novel, semisynthetic approach to lipid As is ongoing in our laboratories, relying upon the chemical modification of a natural lipid A scaffold for the fast obtainment of several other lipid As and derivatives thereof. The first requisite for this strategy is to have this scaffold available in large quantities through a scalable process. Here, we present an optimized fed-batch fermentation procedure for the gram-scale production of lipid A from Escherichia coli K4 and a suitable phenol-free protocol for its purification. A study for regioselective de-O-phosphorylation reaction was then performed to afford pure monophosphoryl lipid A with an attenuated endotoxic activity, as evaluated by cytokine production in human monocytic cell line THP-1 in vitro. The reported method for the large-scale obtainment of monophoshoryl lipid A from the fed-batch fermentation broth of a recombinant strain of E. coli may permit the access to novel semisynthetic lipid A immunoadjuvant candidates.

Members of Cupressaceae and Taxaceae are known to release large amounts of highly allergenic pollen grains into the atmosphere, which are responsible for the onset of pollinosis in many countries throughout the world. In addition to pollen grains, their pollen sacs produce orbicules, which are submicron particles reported to carry allergens and which are potentially able to reach much further down the respiratory tract than pollen grains. Previous research has postulated the presence of orbicules in the atmosphere; however, direct observations have not yet been reported. The aim of this research was to provide the first direct evidence that Cupressaceae orbicules are released into the atmosphere by detecting them in daily aerobiological samples. We observed pollen sacs, pollen grains, and orbicules of nine species of Cupressaceae using scanning electron microscope (SEM). We then used a light and confocal microscope, to examine daily aerobiological samples. Under SEM, we measured the orbicule size (0.494–0.777 µm) and detected unknown nanometric particles (130–200 nm). Under the light microscope, aerobiological samples showed clusters of stained dots surrounding the pollen grains of Cupressaceae. Under the confocal microscope, the same clusters were resolved into submicron particles with the same autofluorescence as the pollen grains. These features enabled us to identify them as orbicules. We believe that our findings help to explain the onset of pollinosis and allergic asthma related to Cupressaceae pollen grains in many countries, even before pollen grains are actually detected or after they are no longer observed in aerobiological monitoring samples.

Ultrasound (US) induced transient membrane permeabilisation has emerged as a hugely promising tool for the delivery of exogenous vectors through the cytoplasmic membrane, paving the way to the design of novel anticancer strategies by targeting functional nanomaterials to specific biological sites. An essential step towards this end is the detailed recognition of suitably marked nanoparticles in sonoporated cells and the investigation of the potential related biological effects. By taking advantage of Synchrotron Radiation Fourier Transform Infrared micro-spectroscopy (SR-microFTIR) in providing highly sensitive analysis at the single cell level, we studied the internalisation of a nanoprobe within fibroblasts (NIH-3T3) promoted by low-intensity US. To this aim we employed 20 nm gold nanoparticles conjugated with the IR marker 4-aminothiophenol. The significant Surface Enhanced Infrared Absorption provided by the nanoprobes, with an absorbance increase up to two orders of magnitude, allowed us to efficiently recognise their inclusion within cells. Notably, the selective and stable SR-microFTIR detection from single cells that have internalised the nanoprobe exhibited clear changes in both shape and intensity of the spectral profile, highlighting the occurrence of biological effects. Flow cytometry, immunofluorescence and murine cytokinesis-block micronucleus assays confirmed the presence of slight but significant cytotoxic and genotoxic events associated with the US-nanoprobe combined treatments. Our results can provide novel hints towards US and nanomedicine combined strategies for cell spectral imaging as well as drug delivery-based therapies.

Repellents are a main tool to prevent the outbreak of mosquito-borne diseases that represents a threat for millions of people worldwide. Plant-based products are very promising, low-toxic and eco-friendly alternative to synthetic repellents. Here, we performed an olfactory screening of the essential oils (EOs) of Artemisia verlotiorum Lamotte (Asteraceae), Lavandula dentata L. (Lamiaceae), and Ruta chalepensis L. (Rutaceae) for their possible use as ingredients in topical repellents. The EOs smell profiles were then matched with their repellence against the mosquito Aedes albopictus (Skuse) (Diptera Culicidae). To obtain a more complete bioactivity description, we also tested the EOs oviposition deterrence and the larvicidal activity. The best smell profile was associated with A. verlotiorum EO, while R. chalepensis EO showed the lowest overall pleasantness. All the EOs had a significant activity as skin repellent against Ae. albopictus, deterred the oviposition in the field, and exerted a clear larvicidal activity. Beside the best smell profile, A. verlotiorum EO showed also the longest lasting repellent effect, assuring the complete protection of the treated skin against Ae. albopictus for a time 60% longer than the synthetic repellent DEET.

Glycosaminoglycans, such as hyaluronic acid and chondroitin sulphate, are not only more and more required as main ingredients in cosmeceutical and nutraceutical preparations, but also as active principles in medical devices and pharmaceutical products. However, while biotechnological production of hyaluronic acid is industrially established through fermentation of Streptococcus spp. and recently Bacillus subtilis, biotechnological chondroitin is not yet on the market. A non-hemolytic and hyaluronidase negative S. equi subsp. zooepidemicus mutant strain was engineered in this work by the addition of two E. coli K4 genes, namely kfoA and kfoC, involved in the biosynthesis of chondroitin-like polysaccharide. Chondroitin is the precursor of chondroitin sulphate, a nutraceutical present on the market as anti-arthritic drug, that is lately being studied for its intrinsic bioactivity. In small scale bioreactor batch experiments the production of about 1.46 ± 0.38 g/L hyaluronic acid and 300 ± 28 mg/L of chondroitin with an average molecular weight of 1750 and 25 kDa, respectively, was demonstrated, providing an approach to the concurrent production of both biopolymers in a single fermentation. Spent hops is a waste produced in large amount by the brewing industry. Rhyzopertha dominica and Sitophilus granarius are insects that cause important eco- nomic losses of stored foods. In this study, for the first time, spent hops has been evaluated as a source of essential oil (EO) and chemicals with repellent activity against R. dominica and S. granarius. Spent hops EO yield was 0.11 %. The terpenes myrcene, a-humulene, and b-caryophyllene were its main components (47 %). Spent hops EO RD50 values were 0.01 and 0.19 l Lc m -2 for R. dominica and S. granarius, respectively. Among the chemicals, myrcene was able to exert the highest repel- lency against R. dominica (RD50 = 0.27 l Mc m -2 ), while limonene was the most effective compound against S. granarius (RD50 = 0.89 l Mc m -2 ). These results indi- cate spent hops as an excellent source of EO and chemicals to be utilized as low-cost, eco-friendly insect pests repel- lents in the protection of stored food.

Neem seed oil (NSO) of Azadirachta indica (Meliaceae) contains more than 100 determined biologically active compounds, and many formulations deriving from them showed toxicity, antifeedancy and repellence against a number of arthropod pests. However, it is widely known that botanical products can differ in their chemical composition and bioactivity, as function of the production site and production process. We used high-performance thin layer chromatography (HPTLC) to investigate differences in chemical constituents of NSOs from three production sites. HPTLC analyses showed several differences in chemical abundance and diversity among NSOs, with special reference to limonoids. Furthermore, the three NSOs and their fractions of increasing polarities [i.e. ethyl acetate (EA) fraction and butanol (BU) fraction] were evaluated for larvicidal toxicity and field oviposition deterrence against the Asian tiger mosquito, Aedes albopictus, currently the most invasive mosquito worldwide. Results from bioactivity experiments showed good toxicity of NSOs and EA fractions against A. albopictus fourth instar larvae (with LC50 values ranging from 142.28 to 209.73 ppm), while little toxicity was exerted by BU fractions. A significant effect of the production site and dosage was also found and is probably linked to differences in abundance of constituents among samples, as highlighted by HPTLC analyses. NSOs and EAs were also able to deter A. albopictus oviposition in the field (effective repellence values ranging from 98.55 to 70.10 %), while little effectiveness of BU fractions was found. Concerning ovideterrent activity, no difference due to the production site was found. This is the first report concerning larvicidal toxicity of NSO against A. albopictus and ovideterrence against Culicidae in the field. The chance to use chemicals from the NSO EA fraction seems promising, since they are effective at lower doses, if compared to synthetic products currently marketed, and could be an advantageous alternative to build newer and safer mosquito control tools.

Local gene transfer of the human LIM Mineralization Protein (LMP), a novel intracellular positive regulator of the osteoblast differentiation program, can induce efficient bone formation in rodents. In order to develop a clinically relevant gene therapy approach to facilitate bone healing, we have used primary dermal fibroblasts transduced ex vivo with Ad.LMP3 and seeded on an hydroxyapatite/collagen matrix prior to autologous implantation. Here we demonstrate that genetically modified autologous dermal fibroblasts expressing Ad.LMP-3 are able to induce ectopic bone formation following implantation of the matrix into the mouse triceps and paravertebral muscles. Moreover, implantation of the Ad.LMP-3-modified dermal fibroblasts into a rat mandibular bone critical size defect model results in efficient healing as determined by X-ray, histology and three dimensional micro computed tomography (3DμCT). These results demonstrate the effectiveness of the non-secreted intracellular osteogenic factor LMP-3, in inducing bone formation in vivo. Moreover, the utilization of autologous dermal fibroblasts implanted on a biomaterial represents a promising approach for possible future clinical applications aimed at inducing new bone formation.
