Heroin is an opioid agonist commonly abused for its rewarding effects. Since its synthesis at the end of the nineteenth century, its popularity as a recreational drug has ebbed and flowed. In the last three decades, heroin use has increased again, and yet the pharmacology of heroin is still poorly understood. After entering the body, heroin is rapidly deacetylated to 6-monoacetylmorphine (6-MAM), which is then deacetylated to morphine. Thus, drug addiction literature has long settled on the notion that heroin is little more than a pro-drug. In contrast to these former views, we will argue for a more complex interplay among heroin and its active metabolites: 6-MAM, morphine, and morphine-6-glucuronide (M6G). In particular, we propose that the complex temporal pattern of heroin effects results from the sequential, only partially overlapping, actions not only of 6-MAM, morphine, and M6G, but also of heroin per se, which, therefore, should not be seen as a mere brain-delivery system for its active metabolites. We will first review the literature concerning the pharmacokinetics and pharmacodynamics of heroin and its metabolites, then examine their neural and behavioral effects, and finally discuss the possible implications of these data for a better understanding of opioid reward and heroin addiction. By so doing we hope to highlight research topics to be investigated by future clinical and pre-clinical studies.

Despite the widespread use of antibiotics, deep neck infections are still a relevant clinical problem and can cause severe and potentially life-threatening complications. Imaging plays a pivotal role into the clinical management of deep neck infections, allowing identification of the source of infections, definition of the precise extension of the disease and early diagnosis of local or distant complications. The complex anatomy of deep neck spaces may hide the actual extent of disease: the knowledge of neck anatomy enables radiologists to rapidly evaluate expected routes of spread of infections and to effectively communicate crucial information to surgeons. Computed Tomography is the most used imaging modality for assessing primary site of infection, extent of disease and local complications. Magnetic resonance imaging may be used as a second level imaging modality for individuating intracranial or spinal complications. The present work aims to review the imaging of deep neck spaces infections, focusing on relevant anatomy and clinical scenarios, underlining practical teaching points for each of them. Familiarity with deep neck spaces anatomy and knowledge of most common routes of spread of infections allow the radiologist to make a prompt diagnosis and to look for early signs of potential complications.

The Large Scale Polarization Explorer (LSPE) is a cosmology program for the measurement of large scale curl-like features (B-modes) in the polarization of the Cosmic Microwave Background. Its goal is to constrain the background of inflationary gravity waves traveling through the universe at the time of matter-radiation decoupling. The two instruments of LSPE are meant to synergically operate by covering a large portion of the northern microwave sky. LSPE/STRIP is a coherent array of receivers planned to be operated from the Teide Observatory in Tenerife, for the control and characterization of the low-frequency polarized signals of galactic origin; LSPE/SWIPE is a balloon-borne bolometric polarimeter based on 330 large throughput multi-moded detectors, designed to measure the CMB polarization at 150 GHz and to monitor the polarized emission by galactic dust above 200 GHz. The combined performance and the expected level of systematics mitigation will allow LSPE to constrain primordial B-modes down to a tensor/scalar ratio of $10^{-2}$. In this study, site-specific natural background levels (NBLs) were determined for 18 elements (Al, As, Be, Cd, Co, Cu, Cr, Fe, Hg, K, Mn, Mo, Ni, Pb, Tl, U, V, and Zn) in two sediment cores collected offshore the Bagnoli-Coroglio brownfield site (Gulf of Pozzuoli, southern Italy) to accurately assess the degree of contamination and the historical trends in Heavy Metals (HMs) enrichment. This objective was pursued taking in account the high temporal and spatial variability of the geochemical properties of the area due to the local geothermal activity. Moreover, the temporal variation of Polycyclic Aromatic Hydrocarbons (PAHs) was investigated.226Ra was used as an extraordinary marker to confirm 210Pb dating. It especially allowed defining the geochronological framework of the sediment core closer the brownfield up to around 1500, providing compelling support to correlate the investigated elements’ occurrences with natural geogenic dynamic. Sediment samples were accurately dated and analyzed for chemical and particle size composition. The contamination factor (Cf) and the pollution load index (PLI) showed very high enrichment of Cd, Cu, Hg, Pb, and Zn. The contamination profiles of HMs and PAHs follow the same pattern in both sediment cores, increasing from deep to upper layers. The highest contamination levels for HMs and PAHs were observed between 10 and 30 cm, corresponding to the periods of most intense industrial activity. Decreasing trends of pollutants were observed in the surface layers (0–10 cm), probably affected by a natural attenuation process due to the cessation of industrial activities.

The Potamotrygon from the Potamotrygonidae family is a freshwater stingray that is native to and distributed throughout South American watersheds. The degradation suffered by the environment in recent years, particularly in aquatic habitats, directly affects native fish species. Due to anthropic actions, some species of the genus Potamotrygon are threatened with extinction, encouraging a study on captive breeding procedures. Six Potamotrygon falkneri adult males, kept under the care of the Sao Paulo Aquarium, Brazil, were divided into groups to compare two distinct semen collection protocols: physical restraint (PR) and chemical restraint (CR – 3 mg / kg propofol, directly into the gills). The protocols were alternated between the two groups in a 50-day interval. For both restraining protocols, after positioning the animal on a moistened foam in dorsal recumbency, semen collection was performed by placing a collecting tube at the cloaca opening while performing a gentle massage of the vas deferens. Sperm evaluation was performed at the moment of collection (time 0) by means of conventional semen analysis. There was no significant difference between the restraining methods examined for any parameter evaluated. The results suggest that physical restraint followed by manual massage is an effective approach to harvest semen in stingrays and should be considered the first choice of method, as it not only demonstrated to be a faster procedure but more importantly eliminated anaesthetic-associated risks for the animal. To our knowledge, this is the first study that validates the semen collection technique in live Potamotrygon falkneri rays and is extremely relevant for the development of future reproductive biotechniques in Potamotrygon.

Trace elements are widespread contaminants that can potentially threaten ecosystems and human health. Considering their distribution and toxicity, monitoring their presence in animals represents a priority in environmental risk assessment. Migratory birds have been suggested to be useful biomonitors for trace elements because they can provide information on contaminants even from remote areas that they may exploit during their life cycle. The aim of this study was to analyse the contamination fingerprint of trace elements of African non-breeding staging grounds and European breeding areas in a long-distance migratory passerine bird, the barn swallow (Hirundo rustica). We collected feathers grown in the African non-breeding grounds and those grown in the breeding areas of Northern Italy and measured the levels of 12 trace elements (Al, As, Cd, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se and Zn) by DRC-ICP-MS. Multivariate analysis showed that elemental profiles of feathers grown in African non-breeding areas and in the Italian breeding ones clearly differed, with feathers grown in Africa showing higher concentrations of Al, Cu, Fe, Mn and Ni, but lower concentrations of As, Se and Zn, compared to those grown in Italy. In addition, levels of trace elements were age-dependent, with higher levels in older individuals than in younger ones. Our results add to the growing evidence that feathers of long-distance migratory birds are useful tools to monitor trace elements contamination profiles across continents. Gentiana lutea roots have been widely used in the traditional medicine, especially for their stomachic properties. In this study, the effect of G. lutea root methanol extract on SH-SY5Y cell survival and on anti-proliferative activity of pro-apoptotic agents was evaluated. In neuroblastoma cell line SH-SY5Y, the extract did not exhibit any cytotoxic activity, but protected cells from vinblastine-induced apoptosis. In the combined treatment with 200 µg/ml extract plus 0.1 µM vinblastine, extract significantly increased cell survival, reduced the percentage of apoptotic cells and decreased caspase-3 activity if compared with the vinblastine alone treatment. The anti-apoptotic activity of the extract was associated to the down-regulation of Sirt-1 protein and to the increase of anti-apoptotic Bcl-2 protein expression and intracellular GSH levels. Since the apoptosis modulation was evidenced on a neuroblastoma cell line, G. lutea extract could have a promising potential as a neuroprotective agent. Moreover, G. lutea root extract exhibited anti-inflammatory activity, since it inhibited LPS-induced expression of TNF-α in macrophage RAW264.7 cells.

Adequate oxygen delivery to the heart during stress is essential for sustaining cardiac function. Acute increases in myocardial oxygen demand evoke coronary vasodilation and enhance perfusion via functional upregulation of smooth muscle voltage-gated K+ (Kv) channels. Because this response is controlled by Kv1 accessory subunits (i.e., Kvβ), which are NAD(P)(H)-dependent aldo-keto reductases, we tested the hypothesis that oxygen demand modifies arterial [NAD(H)]i, and that resultant cytosolic pyridine nucleotide redox state influences Kv1 activity. High-resolution imaging mass spectrometry and live-cell imaging reveal cardiac workload-dependent increases in NADH:NAD+ in intramyocardial arterial myocytes. Intracellular NAD(P)(H) redox ratios reflecting elevated oxygen demand potentiate native coronary Kv1 activity in a Kvβ2-dependent manner. Ablation of Kvβ2 catalysis suppresses redox-dependent increases in Kv1 activity, vasodilation, and the relationship between cardiac workload and myocardial blood flow. Collectively, this work suggests that the pyridine nucleotide sensitivity and enzymatic activity of Kvβ2 controls coronary vasoreactivity and myocardial blood flow during metabolic stress.

Probiotics can ameliorate diseases of humans and wildlife, but the mechanisms remain unclear. Host responses to interventions that change their microbiota are largely uncharacterized. We applied a consortium of four natural antifungal bacteria to the skin of endangered Sierra Nevada yellow-legged frogs, Rana sierrae, before experimental exposure to the pathogenic fungus Batrachochytrium dendrobatidis (Bd). The probiotic microbes did not persist, nor did they protect hosts, and skin peptide sampling indicated immune modulation. We characterized a novel skin defense peptide brevinin-1Ma (FLPILAGLAANLVPKLICSITKKC) that was downregulated by the probiotic treatment. Brevinin-1Ma was tested against a range of amphibian skin cultures and found to inhibit growth of fungal pathogens Bd and B. salamandrivorans, but enhanced the growth of probiotic bacteria including Janthinobacterium lividum, Chryseobacterium ureilyticum, Serratia grimesii, and Pseudomonas sp. While commonly thought of as antimicrobial peptides, here brevinin-1Ma showed promicrobial function, facilitating microbial growth. Thus, skin exposure to probiotic bacterial cultures induced a shift in skin defense peptide profiles that appeared to act as an immune response functioning to regulate the microbiome. In addition to direct microbial antagonism, probiotic-host interactions may be a critical mechanism affecting disease resistance.

Pseudomonas aeruginosa is a ubiquitous opportunistic pathogen that forms robust biofilms in the different niches it occupies. Numerous physiological adaptations are required as this organism shifts from soil or aquatic environments to a host-associated lifestyle. While many conditions differ between these niches, temperature shifts are a factor that can contribute to physiological stress during this transition. To understand how temperature impacts biofilm formation in this pathogen, we used proteomic and transcriptomic tools to elucidate physiological responses in environment-relevant vs. host-relevant temperatures. These studies uncovered differential expression of various proteins including a phage protein that is associated with the EPS matrix in P. aeruginosa. This filamentous phage was induced at host temperatures and was required for full biofilm-forming capacity specifically at human body temperature. These data highlight the importance of temperature shift in biofilm formation and suggest bacteriophage proteins could be a possible therapeutic target in biofilm-associated infections.

Volumetric absorptive microsampling (VAMS) is a novel approach that allows single-drop (10 μL) blood collection. Integration of VAMS with mass spectrometry (MS)-based untargeted metabolomics is an attractive solution for both human and animal studies. However, to boost the use of VAMS in metabolomics, key pre-analytical questions need to be addressed. Therefore, in this work, we integrated VAMS in a MS-based untargeted metabolomics workflow and investigated pre-analytical strategies such as sample extraction procedures and metabolome stability at different storage conditions. We first evaluated the best extraction procedure for the polar metabolome and found that the highest number and amount of metabolites were recovered upon extraction with acetonitrile/water (70:30). In contrast, basic conditions (pH 9) resulted in divergent metabolite profiles mainly resulting from the extraction of intracellular metabolites originating from red blood cells. In addition, the prolonged storage of blood samples at room temperature caused significant changes in metabolome composition, but once the VAMS devices were stored at - 80 °C, the metabolome remained stable for up to 6 months. The time used for drying the sample did also affect the metabolome. In fact, some metabolites were rapidly degraded or accumulated in the sample during the first 48 h at room temperature, indicating that a longer drying step will significantly change the concentration in the sample. Rbfox proteins regulate alternative splicing, mRNA stability and translation. These proteins are involved in neurogenesis and have been associated with various neurological conditions. Here, we analyzed Rbfox2 expression in adult and developing mouse retinas and the effect of its downregulation on visual function and retinal transcriptome. In adult rodents, Rbfox2 is expressed in all retinal ganglion cell (RGC) subtypes, horizontal cells, as well as GABAergic amacrine cells (ACs). Among GABAergic AC subtypes, Rbfox2 was colocalized with cholinergic starburst ACs, NPY (neuropeptide Y)- and EBF1 (early B-cell factor 1)-positive ACs. In differentiating retinal cells, Rbfox2 expression was observed as early as E12 and, unlike Rbfox1, which changes its subcellular localization from cytoplasmic to predominantly nuclear at around P0, Rbfox2 remains nuclear throughout retinal development. Rbfox2 knockout in adult animals had no detectable effect on retinal gross morphology. However, the visual cliff test revealed a significant abnormality in the depth perception of Rbfox2-deficient animals. Gene set enrichment analysis identified genes regulating the RNA metabolic process as a top enriched class of genes in Rbfox2-deficient retinas. Pathway analysis of the top 100 differentially expressed genes has identified Rbfox2-regulated genes associated with circadian rhythm and entrainment, glutamatergic/cholinergic/dopaminergic synaptic function, calcium and PI3K-AKT signaling.

This study aims to improve an analytical method to quantify phenolic substances in olive oil. In order to minimize time required and quantity of solvents, sample extract preparation performed for a previously developed high-performance liquid chromatography–diode array detector to quantify olive oil polyphenols has been ten times downscaled and then validated. The new method performs the extraction of phenolic substances from 0.5 g of oil and allows to quantify the phenolic acids vanillic acid, p-coumaric acid, and ferulic acid; the phenolic alcohols tyrosol and hydroxytyrosol; secoiridoid derivatives; the lignans pinoresinol and acetoxypinoresinol; and the flavonoids luteolin and apigenin. Recoveries obtained were 66–89% for phenolic alcohols, 64–90% for phenolic acids, 93–96% for oleuropein (used as a reference for secoiridoid derivatives), 71–95% for flavonoids, and 97–100% for lignans. The total quantity of organic solvents used in the sample preparation is decreased from 30 to 3 mL with an important abatement of waste, costs, and working time requested.

Addiction treatment has not been appreciably improved by neuroscientific research. One problem is that mechanistic studies using rodent models do not incorporate volitional social factors, which play a critical role in human addiction. Here, using rats, we introduce an operant model of choice between drugs and social interaction. Independent of sex, drug class, drug dose, training conditions, abstinence duration, social housing, or addiction score in Diagnostic & Statistical Manual IV-based and intermittent access models, operant social reward prevented drug self-administration. This protection was lessened by delay or punishment of the social reward but neither measure was correlated with the addiction score. Social-choice-induced abstinence also prevented incubation of methamphetamine craving. This protective effect was associated with activation of central amygdala PKCδ-expressing inhibitory neurons and inhibition of anterior insular cortex activity. These findings highlight the need for incorporating social factors into neuroscience-based addiction research and support the wider implantation of socially based addiction treatments.

Rbpms (RNA-binding protein with multiple splicing) is a member of the RRM (RNA Recognition Motif) family of RNA-binding proteins, which is expressed as multiple alternatively spliced transcripts encoding different protein isoforms. We have shown earlier that Rbpms expression in the retina is restricted to retinal ganglion cells (RGCs), and have characterized this gene as a marker for RGCs. The aim of this study was to identify isoforms representing Rbpms in human retinas and to analyze its transcriptional regulation. We found that Rbpms is expressed as transcription variants 1 and 3 encoding isoforms A and C, respectively. These isoforms are encoded by the same first 6 exons but have different C-terminal ends encoded by exon 8 in variant 1 and exon 7 in variant 3. Computational analysis of the Rbpms 5′ untranslated and flanking regions reveals the presence of three CpG islands and four predicted promoter regions (PPRs). The effect of PPR 1 (− 1672/− 1420) and PPR2 (− 330/− 79) on transcriptional activation was minimal, whereas PPR 3 (− 73/+ 177) and PPR4 (+ 274/+ 524) induced the expression by ~ 7 and ninefold compared to control, respectively. The maximum activity, a 30-fold increase above the control level, was obtained from the construct containing both PPRs 3 and 4. Site-directed mutagenesis of several cis-elements within PPR3 and PPR4 including five for Sp1, one for AP1, and two for NF-kB showed that mutation of the first three and especially the first GC box resulted in a threefold downregulation of gene expression. AP1, NF-kB, and two downstream Sp1 sites had no significant effect on expression level. The possible involvement of the GC box 1 at position − 54 in transcriptional regulation of Rbpms was corroborated by EMSA, which showed formation of a DNA–protein complex in the presence of the oligonucleotide corresponding to this Sp1-binding site.
