5. Inhibition of Wnt7b reduces the proliferation, invasion, and migration of colorectal cancer cells

Colorectal cancer is one of the most common gastrointestinal tumors. The role of Wnt7b as a ligand of the Wnt signaling pathway in colorectal cancer remains to be studied. Through bioinformatics online analysis, we found that Wnt7b is abnormally highly expressed in a variety of gastrointestinal tumors. This study mainly explored the effects of Wnt7b regulating the Wnt/β-catenin signaling pathway on the proliferation, migration, and invasion of SW480 cells in colorectal cancer.Applying the TCGA data set, Wnt7b was found to be highly expressed in most gastrointestinal tumor samples. Real-time quantitative PCR(q-PCR), Western blotting(WB) results showed that Wnt7b was significantly higher expressed in colorectal cancer cell lines compared with normal intestinal epithelial cells. SW480 cells transfected with the sh-Wnt7b showed successful knockdown of Wnt7b. MTT colorimetry showed the proliferation ability of sh-Wnt7b group decreased significantly compared with the non-transfected group. The results of double staining flow cytometry showed that the sh-Wnt7b group had more apoptosis. Cell scratch test showed that the cell migration rate of sh-wnt7b group considerably reduced. The Transwell invasion experiment demonstrated that the number of cell invasions in the sh-Wnt7b group decreased significantly. After SW480 cells was transfected with sh-Wnt7b, the protein levels of β-catenin, CCND1, and CD44 in this group of cells were detected to be reduced by WB, and the same results were obtained by q-PCR detection of mRNA.Wnt7b is highly expressed in colorectal cancer cells, which may affect the proliferation, migration, and invasion of colorectal cancer cells by activating the Wnt/β-catenin signaling pathway.

6. PLD3 affects axonal spheroids and network defects in Alzheimer’s disease

The precise mechanisms that lead to cognitive decline in Alzheimer’s disease are unknown. Here we identify amyloid-plaque-associated axonal spheroids as prominent contributors to neural network dysfunction. Using intravital calcium and voltage imaging, we show that a mouse model of Alzheimer’s disease demonstrates severe disruption in long-range axonal connectivity. This disruption is caused by action-potential conduction blockades due to enlarging spheroids acting as electric current sinks in a size-dependent manner. Spheroid growth was associated with an age-dependent accumulation of large endolysosomal vesicles and was mechanistically linked with Pld3—a potential Alzheimer’s-disease-associated risk gene1 that encodes a lysosomal protein2,3 that is highly enriched in axonal spheroids. Neuronal overexpression of Pld3 led to endolysosomal vesicle accumulation and spheroid enlargement, which worsened axonal conduction blockades. By contrast, Pld3 deletion reduced endolysosomal vesicle and spheroid size, leading to improved electrical conduction and neural network function. Thus, targeted modulation of endolysosomal biogenesis in neurons could potentially reverse axonal spheroid-induced neural circuit abnormalities in Alzheimer’s disease, independent of amyloid removal.

Investigation of the neurobiology of depression in humans depends on animal models that attempt to mimic specific features of the human disorder. However, frequently-used paradigms based on social stress cannot be easily applied to female mice which has led to a large sex bias in preclinical studies of depression. Furthermore, most studies focus on one or only a few behavioral assessments, with time and practical considerations prohibiting a comprehensive evaluation. In this study, we demonstrate that predator stress effectively induced depression-like behaviors in both male and female mice. By comparing predator stress and social defeat models, we observed that the former elicited a higher level of behavioral despair and the latter elicited more robust social avoidance. Furthermore, the use of machine learning (ML)-based spontaneous behavioral classification can distinguish mice subjected to one type of stress from another, and from non-stressed mice. We show that related patterns of spontaneous behaviors correspond to depression status as measured by canonical depression-like behaviors, which illustrates that depression-like symptoms can be predicted by ML-classified behavior patterns. Overall, our study confirms that the predator stress induced phenotype in mice is a good reflection of several important aspects of depression in humans and illustrates that ML-supported analysis can simultaneously evaluate multiple behavioral alterations in different animal models of depression, providing a more unbiased and holistic approach for the study of neuropsychiatric disorders.

Immune checkpoint blockade (ICB) is a promising treatment strategy for colorectal cancer (CRC) patients. However, most CRC patients do not response well to ICB therapy. Increasing evidence indicates that ferroptosis plays a critical role in immunotherapy. ICB efficacy may be enhanced by inducing tumor ferroptosis. Cytochrome P450 1B1 (CYP1B1) is a metabolic enzyme that participates in arachidonic acid metabolism. However, the role of CYP1B1 in ferroptosis remains unclear. In this study, we demonstrated that CYP1B1 derived 20-HETE activated the protein kinase C pathway to increase FBXO10 expression, which in turn promoted the ubiquitination and degradation of acyl-CoA synthetase long-chain family member 4 (ACSL4), ultimately inducing tumor cells resistance to ferroptosis. Furthermore, inhibiting CYP1B1 sensitized tumor cells to anti-PD-1 antibody in a mouce model. In addition, CYP1B1 expression was negatively correlated with ACSL4 expression, and high expression indicates poor prognosis in CRC. Taken together, our work identified CYP1B1 as a potential biomarker for enhancing anti-PD-1 therapy in CRC.

Plectropomus leopardus, as known as leopard coral grouper, is a valuable marine fish that has gradually been bred artificially. To promote future conservation, molecular breeding, and comparative studies, we generated an improved high-quality chromosomal-level genome assembly of leopard coral grouper using Nanopore long-reads, Illumina short reads, and the Hi-C sequencing data. The draft genome is 849.74 Mb with 45 contigs and N50 of 35.59 Mb. Finally, a total of 846.49 Mb corresponding to 99.6% of the contig sequences was anchored to 24 pseudo-chromosomes using Hi-C technology. A final set of 25,965 genes is annotated after manual curation of the predicted gene models, and BUSCO analysis yielded a completeness score of 99.5%. This study significantly improves the utility of the grouper genome and provided a reference for the study of molecular breeding, genomics and biology in this species.

Full-color emissive carbon dots (CDs) hold a great promise for various applications, especially in light emitting diodes (LEDs). However, the existing synthetic routes for CDs are carried out in solutions, which suffer from low yields, high pressures, various byproducts, large amounts of waste solvents, and complicated photoluminescence (PL) origins. Therefore, it is necessary to explore large scale synthesis of CDs with high quantum yield (QY) across the entire visible range from a single carbon source by a solvent-free method. In this work, a series of CDs with tunable PL emission from 442 to 621 nm, QY of 23%–56%, and production yield within 34%–72%, are obtained by heating o-phenylenediamine with the catalysis of KCl. Detailed characterizations identify that, the differences between these CDs with respect to the graphitization degree, graphitic nitrogen content, and oxygen-containing functional groups, are responsible for their distinct optical properties, which can be modulated by controlling the deamination and dehydrogenation processes during reactions. Blue, green, yellow, red emissive films, and LEDs are prepared by dispersing the corresponding CDs into polyvinyl alcohol (PVA). All types of white LEDs (WLEDs) with high colorrendering- index (CRI), including warm WLEDs, standard WLEDs, and cool WLEDs, are also fabricated by mixing the red, green, and blue emissive CDs into PVA matrix by the appropriate ratios.

20. High-temperature modification and air-quenching granulation of steel slag

To solve the problem of difficult utilization of steel slag, the liquid steel slag was modified and the air-quenching granulation process was carried out to make steel slag into a value-added end product: air-quenching granulated steel slag. The granulated slag was tested to analyze the variation rule of slag properties under different modification conditions. Based on the phase diagram of CaO–Si2O–FeO–MgO–Al2O3 slag system, the feasibility of blast furnace (BF) slag as modifier was determined. When the addition of BF slag was increased from 0% to 35%, following results were obtained. The slag fluidity was improved, and the air-quenching temperature range was expanded. Then, the yield of air-quenched steel slag increased, while the granulation rate, the degree of sphericity, the compactness were decreased. Furthermore, the air-quenching granulation process could substantially improve the stability and the amorphous content of steel slag. The maximum removal rate of free CaO was above 80% and the amorphous content was up to 95%. Taking the factors of yield and properties of granulated steel slag into full consideration, the optimum proportion of BF slag is around 15%.

21. Automatic identification of benign pigmented skin lesions from clinical images using deep convolutional neural network

Objective We aimed to develop a computer-aided detection (CAD) system for accurate identification of benign pigmented skin lesions (PSLs) from images captured using a digital camera or a smart phone. Methods We collected a total of 12,836 clinical images which had been classified and location-labeled for training and validating. Four models were developed and validated; you only look once, v4 (YOLOv4), you only look once, v5 (YOLOv5), single shot multibox detector (SSD) and faster region-based convolutional neural networks (Faster R-CNN). The performance of the models was compared with three trained dermatologists, respectively. The accuracy of the best model was further tested and validated using smartphone-captured images. Results The accuracies of YOLOv4, YOLOv5, SSD and Faster R-CNN were 0.891, 0.929, 0.852 and 0.874, respectively. The precision, sensitivity and specificity of YOLOv5 (the best model) were 0.956, 0.962 and 0.952, respectively. The accuracy of YOLOv5 model for images captured using a smart-phone was 0.905. The CAD based YOLOv5 system can potentially be used in clinical identification of PSLs. Conclusion We developed and validated a CAD system for automatic identification of benign PSLs using digital images. This approach may be used by non-dermatologists to easily diagnose by taking a photo of skin lesion and guide on management of PSLs.

22. Effects of geometrical characteristics on defect distributions in alloy components produced by selective laser melting

Selective laser melting (SLM) has been applied to manufacture various alloy components with excellent properties, but its further application is restricted by the intrinsic defects. In this work, the internal defect distributions in samples of three alloys (316L stainless steel, AlSi10Mg and Inconel 718) were investigated respectively, considering the effects of geometrical characteristics of the samples. The defects in the 316L stainless steel sample tend to be formed densely in the central part with large wall thickness, indicating a strong sensitivity to heat accumulation. Contrarily, the Inconel 718 sample shows a higher relative density with homogeneous defect distribution, indicating better formability for the SLM process. For the AlSi10Mg sample, the defect density keeps increasing as the deposition goes on. Typically, the defect density located at sample edges shows an abnormally high level comparing with the inner part, especially in the top sections of AlSi10Mg and Inconel 718 samples. The results are helpful for the geometrical design, the adjustment of building orientation and the further optimization of process parameters in the SLM process.

23. Laser ultrasonic testing for near-surface defects inspection of 316L stainless steel fabricated by laser powder bed fusion

The laser powder bed fusion (L-PBF) method of additive manufacturing (AM) is increasingly used in various industrial manufacturing fields due to its high material utilization and design freedom of parts. However, the parts produced by L-PBF usually contain such defects as crack and porosity because of the technological characteristics of L-PBF, which affect the quality of the product. Laser ultrasonic testing (LUT) is a potential technology for on-line testing of the L-PBF process. It is a non-contact and non-destructive approach based on signals from abundant waveforms with a wide frequency-band. In this study, a method of LUT for on-line inspection of L-PBF process was proposed, and a system of LUT was established approaching the actual environment of on-line detection to evaluate the method applicability for defects detection of L-PBF parts. The detection results of near-surface defects in L-PBF 316L stainless steel parts show that the crack-type defects with a sub-millimeter level within 0.5 mm depth can be identified, and accordingly, the positions and dimensions information can be acquired. The results were verified by X-ray computed tomography, which indicates that the present method exhibits great potential for on-line inspection of AM processes.

Transforaminal endoscopic surgery (TES) is effective for treatment of intervertebral disc-related diseases. To avoid injury to the critical structures, preoperative planning is required to find a safe working channel. Therefore, accurate patient-specific vertebral segmentation is important. The purpose of this work is to develop a convenient, stable and feasible lumbar vertebrae segmentation method for TES planning. Based on the chain structure of the spine, an interactive dual-output vertebrae instance segmentation network was designed to segment the specific vertebrae in CT images. First, an initialization locator module was set up to provide an initial locating box. Then the dual-output network was designed to segment two adjacent vertebrae inside the locating box. Finally, iteration was performed until all the expected vertebrae were segmented. Verification on reconstructed public dataset showed that the vertebral segmentation Dice coefficient was 96.8 ± 1.2% and average surface distance (ASD) was 0.25 ± 0.10 mm. For intervertebral foramen (IVF) region, the Dice coefficient was 96.1 ± 1.5% and ASD was 0.29 ± 0.10 mm. For IVF forming region, the Dice coefficient was 93.4 ± 3.1% and ASD was 0.28 ± 0.13 mm. The evaluation on private dataset showed that more than 90% of the segmentation were suitable for TES planning. For IVF region, the Dice coefficient was 94.4 ± 1.8% and ASD was 0.71 ± 0.49 mm. This work provides a convenient, stable and feasible segmentation method for lumbar vertebrae, IVF region, and IVF forming region. The segmentation can meet the requirement for TES planning.

The preparation of atomically dispersed catalysts with high metal loading remains a formidable challenge due to the high surface energy of single atoms. Here we prepared PtPd/TiO2 catalysts possessing metal loading as high as 8.17 wt% by a new versatile method which based on metal oxide carriers with abundant oxygen defects. PtPd/TiO2 catalysts consist of PtPd nanoparticles and atomically dispersed Pt and Pd atoms, and the content of PtPd nanoparticles is little. Pt3Pd1/TiO2-400 catalyst exhibited the highest catalytic activity in toluene oxidation, and with a 94.7% conversion at 110 °C. Kinetic investigation reveals that the toluene oxidation follows a typical Langmuir-Hinshelwood mechanism. Experimental research indicates that the superior catalytic activity could be attributed to a large number of metal atoms atomically dispersed on the surface of the catalyst. Pt and Pd atoms are close to each other, which produces the synergetic effect and thereby promotes toluene oxidation. This work provides a promising pathway to fabricate single atom catalysts with high loading.

26. Combined inhibition of BADSer99 phosphorylation and PARP ablates models of recurrent ovarian carcinoma

- Abstract
Background Poly (ADP-ribose) polymerase inhibitors (PARPis) have been approved for the treatment of recurrent epithelial ovarian cancer (EOC), regardless of BRCA status or homologous recombination repair deficiency. However, the low response of platinum-resistant EOC, the emergence of resistance in BRCA-deficient cancer, and therapy-associated toxicities in patients limit the clinical utility of PARPis in recurrent EOC. Methods The association of phosphorylated (p) BADS99 with clinicopathological parameters and survival outcomes in an EOC cohort was assessed by immunohistochemistry. The therapeutic synergy, and mechanisms thereof, between a pBADS99 inhibitor and PARPis in EOC was determined in vitro and in vivo using cell line and patient-derived models. Results A positive correlation between pBADS99 in EOC with higher disease stage and poorer survival is observed. Increased pBADS99 in EOC cells is significantly associated with BRCA-deficiency and decreased Cisplatin or Olaparib sensitivity. Pharmacological inhibition of pBADS99 synergizes with PARPis to enhance PARPi IC50 and decreases survival, foci formation, and growth in ex vivo culture of EOC cells and patient-derived organoids (PDOs). Combined inhibition of pBADS99 and PARP in EOC cells or PDOs enhances DNA damage but impairs PARPi stimulated DNA repair with a consequent increase in apoptosis. Inhibition of BADS99 phosphorylation synergizes with Olaparib to suppress the xenograft growth of platinum-sensitive and resistant EOC. Combined pBADS99-PARP inhibition produces a complete response in a PDX derived from a patient with metastatic and chemoresistant EOC. Conclusions A rational and efficacious combination strategy involving combined inhibition of pBADS99 and PARP for the treatment of recurrent EOC is presented.

Face-based biometric recognition is widely used nowadays, where substantial face images are commonly stored on third-party servers. Since the sensitive information of an individual is contained in facial image such as the age and health condition, it is necessary to protect its privacy and security. This paper investigates a cancelable color face template protection algorithm. To make full use of quaternion representation, the structural information including local variance and gradient is respectively served as the real part. To achieve revocability and ability to redistribute, the strategy of random permutation with binary matrix is adopted. Afterwards, the quaternion-based two-dimensional principal component analysis is employed to extract features. With them, the extreme learning machine can be trained and used for recognition. Experimental results performed on four different color face datasets have demonstrated that the fusion of structural information can greatly improve the accuracy. More importantly, the random permutation not only does not reduce the recognition accuracy, but also guarantees the security and revocation of face template.

Autophagy, the self-renewal process of cells, is dependent on lysosomes to degrade damaged organelles and proteins. The increased or damaged level of autophagy is proven to relate to a number of disorders, including metabolic disorders, malignant tumors, pulmonary diseases, and neurodegenerative disorders. This review aims to examine the effects of autophagy on the pathogenic mechanism of obstructive sleep apnea (OSA) in order to guide relevant disease treatment. We conducted a search of the literature using the electronic database, focusing on articles that explored the association between OSA and autophagy. OSA can induced autophagy through hypoxia, oxidative stress, endoplasmic reticulum stress, endothelial dysfunction, miRNA, etc. We propose that the mechanism of the autophagy in patients with OSA should be eclucidated in further studies.

Lung cancer is a leading cause of cancer-associated deaths worldwide. Lung cancer may lead to circadian disruption, which could contribute to the development of lung cancer. Recently, several studies using animal models indicated that tumors influence systemic circadian homeostasis in remote tissues. However, it is unclear whether carcinoma of the lungs influences remote circadian rhythm, whether this effect exists in humans, and whether signals from the tumor travel through the blood. In this study, we used a cell-based assay to determine whether serum from patients with lung adenocarcinoma could modulate the molecular clock. We found that the daily oscillation period of Bmal1 was significantly lengthened following treatment with serum from untreated lung adenocarcinoma patients. In addition, heat inactivation of this serum abolished the effect, suggesting that a heat-sensitive circulating factor(s) is present in the serum of untreated lung adenocarcinoma patients. Using real-time PCR, we also examined the mRNA abundance of Bmal1, Cry1, and Per1 in human osteosarcoma u2os cell line, HUVECs and A549 cell lines. The expression of Bmal1 was changed in A549 cells in the presence of sera from lung adenocarcinoma patients. Our study revealed a direct effect of serum from lung adenocarcinoma patients on the molecular clock.

Glioblastoma multiforme (GBM) is a highly aggressive brain tumor with an extremely low survival rate. New and effective approaches for treatment are therefore urgently needed. Here, we successfully developed M1-like macrophage-derived extracellular vesicles (M1EVs) that overcome multiple challenges via guidance from two macrophage-related observations in clinical specimens from GBM patients: enrichment of M2 macrophages in GBM; and origination of a majority of infiltrating macrophage from peripheral blood. To maximize the synergistic effect, we further functionalized the membranes of M1EVs with two hydrophobic agents (the chemical excitation source CPPO (C) and the photosensitizer Ce6 (C)) and loaded the hydrophilic hypoxia-activated prodrug AQ4N (A) into the inner core of the M1EVs. After intravenous injection, the inherent nature of M1-derived extracellular vesicles CCA-M1EVs allowed for blood-brain barrier penetration, and modulated the immunosuppressive tumor microenvironment via M2-to-M1 polarization, which increased hydrogen peroxide (H2O2) levels. Furthermore, the reaction between H2O2 and CPPO produced chemical energy, which could be used for Ce6 activation to generate large amounts of reactive oxygen species to achieve chemiexcited photodynamic therapy (CDT). As this reaction consumed oxygen, the aggravation of tumor hypoxia also led to the conversion of non-toxic AQ4N into toxic AQ4 for chemotherapy. Therefore, CCA-M1EVs achieved synergistic immunomodulation, CDT, and hypoxia-activated chemotherapy in GBM to exert a potent therapeutic effect. Finally, we demonstrated the excellent effect of CCA-M1EVs against GBM in cell-derived xenograft and patient-derived xenograft models, underscoring the strong potential of our highly flexible M1EVs system to support multi-modal therapies for difficult-to-treat GBM.

Background This study aims to investigate thrombospondin 2 (TSP2) expression levels in gastric cancer (GC) and determine the relationship between TSP2 and clinical characteristics and prognosis. Methods The online database Gene Expression Profile Interactive Analysis (GEPIA) was used to analyse TSP2 mRNA expression levels in GC. The Kaplan–Meier plotter prognostic analysis tool was used to evaluate the influence of TSP2 expression on clinical prognosis in GC patients. TSP2 expression levels were analysed in paraffin-embedded GC samples and adjacent normal tissues by immunohistochemistry. The relationship between the clinicopathological characteristics and prognosis of GC patients was assessed. Transwell experiments were used to evaluate the effect of TSP2 on HGC27 and AGS cell invasion and migration. The EdU experiment was used to detect the effect of transfection of TSP2 on cell proliferation, and the flow cytometry experiment was used to detect the effect of TSP2 on cell apoptosis and the cell growth cycle. Western blotting (Wb) technology was used to detect MMP, E-cadherin, N-cadherin, Vimentin, Snail, AKT, PI3K, and VEGF protein expression in HGC27 cells. Results Compared with normal tissues, TSP2 mRNA expression in GC was significantly upregulated and was closely related to the clinical stage of GC. High TSP2 expression significantly affected the OS, FP and PPS of patients with GC. Among these patients, TSP2 expression levels did not affect the prognosis of patients with GC in the N0 subgroup but significantly affected the prognosis of patients with GC in the N (1 + 2 + 3) subgroup. TSP2 protein expression levels were significantly higher in GC tissue compared with normal tissues (P < 0.01). The overall survival (OS) and relapse-free survival (RFS) of patients with high TSP2 expression were lower than those of patients with low TSP2 expression. Cells transfected with the TSP2-silencing sequence exhibited increased apoptosis and inhibition of proliferation, migration and invasion. AKT and PI3K expression in cells was significantly downregulated (P < 0.01). AKT, PI3K and VEGF expression in cells transfected with the TSP2 silencing sequence was significantly reduced. Proliferation, migration, invasion ability, and TSP2 expression levels significantly correlated with mismatch repair genes, such as PMS2, MSH6, MSH2, and MLH1 (P < 0.05). Conclusion TSP2 expression is significantly increased in GC. TSP2 expression is closely related to metastasis and the mismatch repair process in GC patients and affects GC patient prognosis. The mechanism may involve regulating gastric cancer cell proliferation and migration by modulating the VEGF/PI3K/AKT signalling pathway. TSP2 is a potential marker and therapeutic target for the prognosis of GC patients.

Nanozymes are nanomaterials with enzyme-like properties that have attracted significant interest owing to their capability to address the limitations of traditional enzymes such as fragility, high cost, and impossible mass production. Over the past decade, a broad variety of nanomaterials have been found to mimic the enzyme-like activity by engineering the active centers of natural enzymes or developing multivalent elements within nanostructures. Carbon nanomaterials with well-defined electronic and geometric structures have served as favorable surrogates of traditional enzymes by mimicking the highly evolved catalytic center of natural enzymes. In particular, by combining the unique electronic, optical, thermal, and mechanical properties, carbon nanomaterials-based nanozymes can offer a variety of multifunctional platforms for biomedical applications. In this review, we will introduce the enzymatic characteristics and recent advances of carbon nanozymes, and summarize their significant applications in biomedicine.

Improving the performance of forward osmosis (FO) membranes has always been moving forward. Here we fabricate the novel Turing structures in the active layer (AL) of thin film composite (TFC) membrane through effectively introducing the polyvinylpyrrolidone (PVP) macromolecule with hydrophilicity both in the support layer (SL) and AL. The addition of PVP facilitates the formation of sponge-like pores in the SL and stripe-like Turing structures in the AL. The influence of PVP on the morphology and performance of the membranes was investigated by SEM, AFM, XPS, NMR, FTIR. Compared with the water flux (Jw) of the original composite membrane (approximately 25 L m−2 h−1), TFC-PVP (3:1) membrane with Turing structure achieved better performance (approximately 75 L m−2 h−1), and the reverse solute flux/water flux (Js/Jw) value was as low as 0.02 g/L. This study demonstrated that the Turing structures have a favorable effect on the performance of FO membrane and a promisingly practical application in water treatment.

Clinical cases have reported pulmonary arterial structural and functional abnormalities in patients with Kawasaki disease (KD); however, the underlying mechanisms are unclear. In this study, a KD rat model was established via the intraperitoneal injection of Lactobacillus casei cell wall extract (LCWE). The results showed that pulmonary arterial functional and structural abnormalities were observed in KD rats. Furthermore, proliferative endoplasmic reticulum stress (ER stress) was observed in the pulmonary arteries of KD rats. Notably, the level of lipocalin-2 (Lcn 2), a trigger factor of inflammation, was remarkably elevated in the plasma and lung tissues of KD rats; increased Lcn 2 levels following LCWE stimulation may result from polymorphonuclear neutrophils (PMNs). Correspondingly, in cultured pulmonary artery smooth muscle cells (PASMCs), Lcn 2 markedly augmented the cleavage and nuclear localization of activating transcription factor-6 (ATF6), upregulated the transcription of glucose regulated protein 78 (GRP78) and neurite outgrowth inhibitor (NOGO), and promoted PASMCs proliferation. However, proapoptotic C/EBP homologous protein (CHOP) and caspase 12 levels were not elevated. Treatment with 4-phenyl butyric acid (4-PBA, a specific inhibitor of ER stress) inhibited PASMCs proliferation induced by Lcn 2 and attenuated pulmonary arterial abnormalities and right ventricular hypertrophy and reduced right ventricular systolic pressure in KD rats. In conclusion, Lcn 2 remarkably facilitates proliferative ER stress in PASMCs, which probably accounts for KD-related pulmonary arterial abnormalities.

Preoperative treatment planning is key to ensure successful thermal ablation of liver tumors. The planning aims to minimize the number of electrodes required for complete ablation and the damage to the surrounding tissues while satisfying multiple clinical constraints. This is a challenging multiple objective planning problem, in which the trade-off between different objectives must be considered. We propose a novel method to solve the multiple objective planning problem, which combines the set cover-based model and Pareto optimization. The set cover-based model considers multiple clinical constraints and generates several clinically feasible treatment plans, among which the Pareto optimization is performed to find the trade-off between different objectives. We evaluated the proposed method on 20 tumors of 11 patients in two different situations used in common thermal ablation approaches: with and without the pull-back technique. Pareto optimal plans were found and verified to be clinically acceptable in all cases, which can find the trade-off between the number of electrodes and the damage to the surrounding tissues. The proposed method performs well in the two different situations we considered: with or without the pull-back technique. It can generate Pareto optimal plans satisfying multiple clinical constraints. These plans consider the trade-off between different planning objectives.

Deficiency of caspase recruitment domain-containing protein 9 (CARD9) is an autosomal recessive primary immunodeficiency disorder, which typically predisposes immunocompetent individuals to single fungal infections and multiple fungal infections are very rare. We study an otherwise healthy 48-year-old man, who had been admitted to our hospital diagnosed with deep dermatophytosis caused by Trichophyton rubrum for three times at 29, 33 and 48 years old, respectively. At the age of 39 years, he suffered from cutaneous mucormycosis due to Mucor irregularis. Moreover, he had a long history of superficial fungal diseases and occasional oral candidiasis. Whole-exome sequencing revealed two compound heterozygous splicing variants in CARD9 gene, c. 184 + 5 G > T and c. 951G > A, confirmed by Sanger sequencing. Patients with recurrent fungal infections especially invasive fungal infections in the absence of known immunodeficiencies should be tested for CARD9 mutations.

With more and more color images are spread and shared in the untrusted cloud server remotely over the open Internet, some private or confidential information may be contained, it is necessary to develop cryptosystem for these images to free from serious threats. This paper investigates a secure and robust cryptosystem for color image based on discrete trinion Fourier transform and random-multiresolution singular value decomposition. Firstly, a color image is represented into a trinion matrix by precoding and followed by block-wise discrete trinion Fourier transform. Then a mapping from trinion number domain to real number domain is implemented to facilitate random-multiresolution singular value decomposition. To further enhance the randomness of the transformed data, the scrambling based on chaotic sequence is applied to obtain the cipherimage. Experimental results have demonstrated that the proposed color image encryption algorithm is feasible. The adoption of logistic map ensures that the proposed cryptosystem attains high level security. Furthermore, the quality of the retrieved image is better than that of the algorithm based on single color channel processing separately and quaternion matrix based algorithms when it is decrypted correctly or under Gaussian noise and shear attacks. Moreover, employing a trinion matrix to represent a color image is more compact than using quaternion matrix. It would have potential applications in digital image protection.

Crush injury/crush syndrome (CI/CS) is the second most common cause of death during earthquakes. Most studies of CI/CS have mainly focused on kidney injury after decompression. Few studies have focused on myocardial injury caused by crush injury and its potential mechanisms. We first verified cardiomyocyte injury during compression in rats with a crush injury. The survival rate, electrocardiographic results, histological results, catecholamine changes and cardiac β1-AR expression were evaluated. Next, we explored the effects of pretreatment with a selective β1-blocker (bisoprolol) with or without fluid resuscitation on rats with a crush injury. In addition to evaluating the survival rates, biochemical and histological analyses and echocardiographic measurements were also performed. Reduced heart rates, elevated ST segments, and tall-peaked T waves were observed in the rats with a crush injury. The changes in the myocardial enzymes and pathological results demonstrated that myocardial damage occurred during compression in rats with a crush injury. The levels of the catecholamine norepinephrine in both the serum and myocardial tissue were elevated during compression. Pretreatment with a selective β1-blocker combined with fluid resuscitation significantly improved the survival rates of the rats with lethal crush injury. The myocardial enzymes and pathological results showed that the combined therapy decreased myocardial damage. The echocardiography measurements showed that the rats that received the combined therapy exhibited decreased left ventricular mass (LVM), left ventricular volume at end-systole (LVVs) and left ventricular internal diameter (LVID) compared with the rats with a crush injury. Our findings demonstrated the presence of myocardial injury in the early stage of compression in rats with a crush injury. Pretreatment with a β1-blocker (bisoprolol) with fluid resuscitation significantly reduced mortality, decreased myocardial tissue damage, and improved ventricular remodelling in rats with a lethal crush injury.

Sulforhodamine B (SRB) is widely utilized for cell staining and laser field. But its application is limited by aggregation-caused quenching (ACQ). In this work, we evaluated the use of UiO-66 and UiO-67 of Zr-based metal organic frameworks (Zr-MOFs) as the host to adsorb SRB molecules due to the high stabily and good loading capacity of Zr-MOFs. The fluorescence properties of the compounds were then discussed respectively. Due to the aperture difference between UiO-66 and UiO-67, they showed distinct fluorescence properties after loading SRB. When the concentration reaches 5 ppm, fluorescence quenching begins to occur in SRB@UiO-66, while it occurs in SRB@UiO-67 at 2 ppm. The solution of quenching phenomenon could open new avenues for the extensive use of SRB.

Background DAXX is a transcription repressor that has been implicated in several types of cancers, but its role in the development of gastric cancer remains unknown. Methods We analysed the expression of DAXX in 83 pairs of gastric cancer samples, including neoplastic and adjacent tissues, and correlated the expression levels with clinical stages. We also investigated the molecular mechanisms by which DAXX downregulation promotes cancer growth using both in vitro and in vivo models. Results DAXX was downregulated in advanced gastric cancer samples. The expression of DAXX inversely correlates with that of cancer stem cell markers CD44 and Oct4 in gastric cancer lines. DAXX overexpression in gastric cancer cells inhibited migration, invasion and epithelial– mesenchymal transition (EMT). The inhibition of EMT was achieved through the repression of SNAI3, a key inducer of EMT, by recruiting HDAC-1 into the nucleus. Using a xenograft mouse model, we demonstrated that the MKN45 cells formed smaller tumours when DAXX was overexpressed. Wild-type AGS cells were not able to form tumours in nude mice, but in contrast, formed visible tumours when DAXX was silenced in the cells. Conclusion We for the first time demonstrated that DAXX functions as a tumour suppressor in gastric cancer by inhibiting stem cell growth and EMT.

Soccer training assisted robot system is a combination of robotics and artificial intelligence. Motion path planning is an important part of soccer training assisted robot decision system. Path planning aims to find an optimal path, complete dynamic and static obstacle avoidance, and thus can timely and quickly carry out the path planning of the soccer training assisted robot cooperation strategy. Aiming at the problem of soccer training auxiliary robot motion path planning, this paper proposes a global path planning method based on genetic algorithm, which avoids the problem of unsmooth path and large computation. Under the premise of ensuring the completeness of search, this method transforms the soccer training assisted robot path planning problem into the path search problem of the soccer training assisted robot system center. Under the premise of using the fixed-point rotation method on a large scale, the genetic algorithm is adopted for the case of some special narrow-channel environments, and the action area is determined by simple search. Finally, the planning path of each soccer training-assisted robot in the system is obtained. Finally, the simulation results verify the effectiveness of the algorithm.

Characterizing the three-dimensional (3D) morphological alterations of microvessels under both normal and seizure conditions is crucial for a better understanding of epilepsy. However, conventional imaging techniques cannot detect microvessels on micron/sub-micron scales without angiography. In this study, synchrotron radiation (SR)-based X-ray in-line phase-contrast imaging (ILPCI) and quantitative 3D characterization were used to acquire high-resolution, high-contrast images of rat brain tissue under both normal and seizure conditions. The number of blood microvessels was markedly increased on days 1 and 14, but decreased on day 60 after seizures. The surface area, diameter distribution, mean tortuosity, and number of bifurcations and network segments also showed similar trends. These pathological changes were confirmed by histological tests. Thus, SR-based ILPCI provides systematic and detailed views of cerebrovascular anatomy at the micron level without using contrast-enhancing agents. This holds considerable promise for better diagnosis and understanding of the pathogenesis and development of epilepsy.

47. Association of long-term sleep habits and hypertension: a cross-sectional study in Chinese adults

Poor sleep has been associated with risk of hypertension, but previous studies were limited by treating one or two sleep factors as predictor. Our previous study has developed a sleep factor questionnaire (SFQ) to comprehensively assess wide range of sleep characteristics including sleep duration, sleep quality, light at night (LAN) exposure, night/shift work, daytime napping, and frequency of nighttime waking, insomnia, and snoring. In this cross-sectional study we used the SFQ to evaluated the associations between these sleep domains and hypertension. Comparing with the subjects who slept 7–8 h, subjects with sleep duration 6–7 h (OR = 1.42, 95% CI: 1.01, 2.00), 8–9 h (OR = 1.60, 95% CI: 1.07, 2.40), and over 9 h (OR = 2.39, 95% CI: 1.60, 3.58) had an increased prevalence of hypertension. The associations were significant among individuals under 45 years sleeping 6–7 h and those over 45 years sleeping over 8 h. The OR of prevalent hypertension was 0.80 (95% CI: 0.62, 0.97) for habitual daytime napping compared with never napping, and the association was also significant among individuals over 45 years old. Moreover, among subjects with sleep duration 7–8 h, habitual daytime napping may decrease prevalent hypertension (OR = 0.635, 95% CI: 0.437, 0.924). In conclusion, our data suggested a positive association between both short (6–7 h) and long sleep duration and hypertension, and a preventive pattern for habitual daytime napping among over 45 years old and hypertension. Moreover, interaction analysis indicated that habitual midday nap may decrease prevalent hypertension among subjects with sleep duration 7–8 h.

Current stereotactic surgical robots system relies on cumbersome operations such as calibration, tracking and registration to establish the accurate intraoperative coordinate transformation chain, which makes the system not easy to use. To overcome this problem, a novel stereotactic surgical robot system has been proposed and validated. First, a hand–eye integrated scheme is proposed to avoid the intraoperative calibration between robot arm and motion tracking system. Second, a special reference-tool-based patient registration and tracking method is developed to avoid intraoperative registration. Third, a model-free visual servo method is used to reduce the accuracy requirement of hand–eye relationship and robot kinematic model. Finally, a prototype of the system is constructed and performance tests and a pedicle screw drilling experiment are performed. The results show that the proposed system has acceptable accuracy. The target positioning error in the plane was − 0.68 ± 0.52 mm and 0.06 ± 0.41 mm. The orientation error was 0.43 ± 0.25°. The pedicle screw drilling experiment shows that the system can complete accurate stereotactic surgery. The stereotactic surgical robot system described in this paper can perform stereotactic surgery without the intraoperative hand–eye calibration and nor manual registration and can achieve an acceptable position and orientation accuracy while tolerating the errors in the hand–eye coordinate transformation error and the robot kinematics model error.

To develop and validate a radiomics-based nomogram for preoperatively predicting grade 1 and grade 2/3 tumors in patients with pancreatic neuroendocrine tumors (PNETs). One hundred thirty-eight patients derived from two institutions with pathologically confirmed PNETs (104 in the training cohort and 34 in the validation cohort) were included in this retrospective study. A total of 853 radiomic features were extracted from arterial and portal venous phase CT images respectively. Minimum redundancy maximum relevance and random forest methods were adopted for the significant radiomic feature selection and radiomic signature construction. A fusion radiomic signature was generated by combining both the single-phase signatures.
