Abstract

International audience; Organizations’ business processes need to be adapted in response to changing internal and external environments, which are becoming increasingly complex. We target in this research work the exploitation of a software capability profile based on requirements engineering and enterprise architecture to respond to stakeholder requirements and efficiently reuse existing technical solutions. We propose in this work an exploitation methodology based on the alignment of enterprise architecture actions with a requirement engineering process. These latter evolve together helping to investigate the highest compatibility of the desired functionalities and their related constraints. Our contribution aims to produce a ready-to-use application based on the defined requirements and the selected software capability profiles for accelerating business application development. Implementation and a case study are proposed to demonstrate the effectiveness of this approach.

Abstract

To establish national reference levels (RLs) in interventional procedures under CT guidance as required by the 2013/59/Euratom European Directive. Seventeen categories of interventional procedures in thoracic, abdominopelvic, and osteoarticular specialties (percutaneous infiltration, vertebroplasty, biopsy, drainage, tumor destruction) were analyzed. Total dose length product (DLP), number of helical acquisitions (NH), and total DLP for helical, sequential, or fluoroscopic acquisitions were recorded for 10 to 20 patients per procedure at each center. RLs were calculated as the 3rd quartiles of the distributions and target values for optimization process (TVOs) as the median. RLs and TVOs were compared with previously published studies. Results on 5001 procedures from 49 centers confirmed the great variability in patient dose for the same category of procedures. RLs were proposed for the DLPs and NHs in the seventeen categories. RLs in terms of DLP and NH were 375 mGy.cm and 2 NH for spinal or peri-spinal infiltration, 1630 mGy.cm and 3 NH for vertebroplasty, 845 mGy.cm and 4 NH for biopsy, 1950 mGy.cm and 8 NH for destruction of tumors, and 1090 mGy.cm and 5 NH for drainage. DLP and NH increased with the complexity of procedures. This study was the first nationwide multicentric survey to propose RLs for interventional procedures under CT guidance. Heterogeneity of practice in centers were found with different levels of patient doses for the same procedure. The proposed RLs will allow imaging departments to benchmark their practice with others and optimize their protocols. • National reference levels are proposed for 17 categories of interventional procedures under CT guidance. • Reference levels are useful for benchmarking practices and optimizing protocols. • Reference levels are proposed for dose length product and the number of helical acquisitions.

Abstract

8-oxoguanine (GO) is a major lesion found in DNA that arises from guanine oxidation. The hyperthermophilic and radioresistant euryarchaeon Thermococcus gammatolerans encodes an archaeal GO DNA glycosylase (Tg-AGOG). Here, we characterized biochemically Tg-AGOG and probed its GO removal mechanism by mutational studies. Tg-AGOG can remove GO from DNA at high temperature through a β-elimination reaction. The enzyme displays an optimal temperature, ca.85–95 °C, and an optimal pH, ca.7.0–8.5. In addition, Tg-AGOG activity is independent on a divalent metal ion. However, both Co2+ and Cu2+ inhibit its activity. The enzyme activity is also inhibited by NaCl. Furthermore, Tg-AGOG specifically cleaves GO-containing dsDNA in the order: GO:C, GO:T, GO:A, and GO:G. Moreover, the temperature dependence of cleavage rates of the enzyme was determined, and from this, the activation energy for GO removal from DNA was first estimated to be 16.9 ± 0.9 kcal/mol. In comparison with the wild-type Tg-AGOG, the R197A mutant has a reduced cleavage activity for GO-containing DNA, whereas both the P193A and F167A mutants exhibit similar cleavage activities for GO-containing DNA. While the mutations of P193 and F167 to Ala lead to increased binding, the mutation of R197 to Ala had no significant effect on binding. These observations suggest that residue R197 is involved in catalysis, and residues P193 and F167 are flexible for conformational change.

Abstract

The diagnosis of atypical inflammatory demyelinating lesions can be difficult. Brain biopsy is often required to exclude neoplasms. Moreover, the relationship between these lesions and multiple sclerosis and NMOSD is not clear. Our objectives were to describe radiological and pathological characteristics of patients with acute inflammatory demyelinating lesions. We retrospectively identified patients with brain biopsy performed for diagnostic uncertainty revealing a demyelinating lesion. A complete clinical, biological, radiological and pathological analysis was performed. Twenty patients (15 with a single lesion) were included. MRI disclosed a wide range of lesions including infiltrative lesions (40%), ring-like lesion (15%) Balo-like lesion (15%) and acute haemorrhagic leukoencephalitis (20%). In spite of a marked heterogeneity, some findings were common: a peripheral B1000 hyperintense rim (70%), a slight oedema with mild mass effect (75%) and an open-rim peripheral enhancement (75%). Histopathology revealed that all cases featured macrophages distributed throughout, extensive demyelination, axonal preservation and absence of haemorrhagic changes. In the majority of cases, macrophages were the predominant inflammatory infiltrate and astrocytes were reactive and dystrophic. Aquaporin-4 staining was systematically preserved. After a mean follow-up of 5 years (1–12), 16/20 patients had a diagnosis of monophasic acute atypical inflammatory demyelinating lesion. One patient was diagnosed with MS and 3 with AQP4 negative NMOSD. Although imaging findings in patients with atypical inflammatory demyelinating lesions are heterogeneous, some common features such as peripheral DWI hyperintense rim with open-rim enhancement and absence of oedema argue in favour of a demyelinating lesion and should preclude a brain biopsy. In this context, AQP4 staining is systematically preserved and argues against an AQP4-positive NMOSD. Moreover, long-term follow-up is characterized by low recurrence rate.

Abstract

To better predict the effective energy performance of buildings in cities, this study addresses the modelling of local external radiative, thermal and aeraulic conditions. After reviewing existing modelling approaches that are suitable for estimating the building boundary conditions in energy simulation, this paper analyses external conditions derived from a building energy model (BuildSysPro) or a microclimatic model (SOLENE microclimat). Comparisons are made for the different faces of a generic building standing alone or located in an urban environment, with or without a thermally efficient envelope. When the modelling approach is adjusted, the results highlight significant deviations on the estimated radiative temperatures and wind-based quantities around the isolated building. When accounting for surrounding buildings, the results show a substantial reduction in short-wave radiative fluxes, which is explained by an imbalance between solar masks and multireflections, and a reduction in the wind-driven ventilation potential.

Abstract

BackgroundThe coronavirus disease 2019 (COVID-19) pandemic would have particularly affected acute stroke care. However, its impact is clearly inherent to the local stroke network conditions. We aimed to assess the impact of COVID-19 pandemic on acute stroke care in the Lyon comprehensive stroke center during this period.MethodsWe conducted a prospective data collection of patients with acute ischemic stroke (AIS) treated with intravenous thrombolysis (IVT) and/or mechanical thrombectomy (MT) during the COVID-19 period (from 29/02/2020 to 10/05/2020) and a control period (from 29/02/2019 to 10/05/2019). The volume of reperfusion therapies and pre and intra-hospital delays were compared during both periods.ResultsA total of 208 patients were included. The volume of IVT significantly decreased during the COVID-period (55 (54.5%) vs 74 (69.2%); p=0.03) and was mainly due to time delay among patients treated with MT. The volume of MT remains stable over the two periods (72 (71.3%) vs 65 (60.8%); p=0.14) but the door-to-groin puncture time increased in patients transferred for MT (237 [187-339] vs 210 [163-260]; pConclusionsOur study showed a decrease of the volume of IVT, whereas the volume of MT remained stable although intra-hospital delays increased for transferred patients during the COVID-19 pandemic. These results contrast in part with the national surveys and suggest that the impact of the pandemic may depend on local stroke care networks.

Abstract

Background During spontaneous breathing trial, low-pressure support is thought to compensate for endotracheal tube resistance, but it actually should provide overassistance. Automatic tube compensation is an option available in the ventilator to compensate for flow-resistance of endotracheal tube. Its effects on patient effort have been poorly investigated. We aimed to compare the effects of low-pressure support and automatic tube compensation during spontaneous breathing trial on breathing power and lung ventilation distribution. Results We performed a randomized crossover study in 20 patients ready to wean. Each patient received both methods for 30 min separated by baseline ventilation: pressure support 0 cmH2O and automatic tube compensation 100% in one period and pressure support 7 cmH2O without automatic tube compensation in the other period, a 4 cmH2O positive end-expiratory pressure being applied in each. Same ventilator brand (Evita XL, Draeger, Germany) was used. Breathing power was assessed from Campbell diagram with esophageal pressure, airway pressure, flow and volume recorded by a data logger. Lung ventilation distribution was assessed by using electrical impedance tomography (Pulmovista, Draeger, Germany). During the last 2 min of low-pressure support and automatic compensation period breathing power and lung ventilation distribution were measured on each breath. Breathing power generated by the patient’s respiratory muscles was 7.2 (4.4–9.6) and 9.7 (5.7–21.9) J/min in low-pressure support and automatic tube compensation periods, respectively (P = 0.011). Lung ventilation distribution was not different between the two methods. Conclusions We found that ATC was associated with higher breathing power than low PS during SBT without altering the distribution of lung ventilation.

Abstract

The use of constitutive equations calibrated from data has been implemented into standard numerical solvers for successfully addressing a variety problems encountered in simulation-based engineering sciences (SBES). However, the complexity remains constantly increasing due to the need of increasingly detailed models as well as the use of engineered materials. Data-Driven simulation constitutes a potential change of paradigm in SBES. Standard simulation in computational mechanics is based on the use of two very different types of equations. The first one, of axiomatic character, is related to balance laws (momentum, mass, energy, $$\ldots $$ ), whereas the second one consists of models that scientists have extracted from collected, either natural or synthetic, data. Data-driven (or data-intensive) simulation consists of directly linking experimental data to computers in order to perform numerical simulations. These simulations will employ laws, universally recognized as epistemic, while minimizing the need of explicit, often phenomenological, models. The main drawback of such an approach is the large amount of required data, some of them inaccessible from the nowadays testing facilities. Such difficulty can be circumvented in many cases, and in any case alleviated, by considering complex tests, collecting as many data as possible and then using a data-driven inverse approach in order to generate the whole constitutive manifold from few complex experimental tests, as discussed in the present work.

Abstract

The control over the crystallinity of chitosan and chitosan/ovalbumin films can be achieved via an appropriate balance of the hydrophilic/hydrophobic interactions during the film formation process, which then controls the release kinetics of ovalbumin. Chitosan films were prepared by solvent casting. The presence of the anhydrous allomorph can be viewed as a probe of the hydrophobic conditions at the neutralization step. The semicrystalline structure, the swelling behavior of the films, the protein/chitosan interactions, and the release behavior of the films were impacted by the DA and the film processing parameters. At low DAs, the chitosan films neutralized in the solid state corresponded to the most hydrophobic environment, inducing the crystallization of the anhydrous allomorph with and without protein. The most hydrophilic conditions, leading to the hydrated allomorph, corresponded to non-neutralized films for the highest DAs. For the non-neutralized chitosan acetate (amorphous) films, the swelling increased when the DA decreased, whereas for the neutralized chitosan films, the swelling decreased. The in vitro release of ovalbumin (model protein) from chitosan films was controlled by their swelling behavior. For fast swelling films (DA = 45%), a burst effect was observed. On the contrary, a lag time was evidenced for DA = 2.5% with a limited release of the protein. Furthermore, by blending chitosans (DA = 2.5% and 45%), the release behavior was improved by reducing the burst effect and the lag time. The secondary structure of ovalbumin was partially maintained in the solid state, and the ovalbumin was released under its native form.

Abstract

Background Myelin oligodendrocyte glycoprotein antibodies (MOG-Ab) are related to several acquired demyelinating syndromes in adults, but the therapeutic approach is currently unclear. We aimed to describe the response to different therapeutic strategies in adult patients with relapsing MOG-Ab-associated disease. Methods This is a retrospective study conducted in France and Spain including 125 relapsing MOG-Ab patients aged ≥ 18 years. First, we performed a survival analysis to investigate the relapse risk between treated and non-treated patients, performing a propensity score method based on the inverse probability of treatment weighting. Second, we assessed the annualised relapse rates (ARR), Expanded Disability Status Scale (EDSS) and visual acuity pre-treatment and on/end-treatment. Results Median age at onset was 34.1 years (range 18.0–67.1), the female to male ratio was 1.2:1, and 96% were Caucasian. At 5 years, 84% (95% confidence interval [CI], 77.1–89.8) patients relapsed. At the last follow-up, 66 (52.8%) received maintenance therapy. Patients initiating immunosuppressants (azathioprine, mycophenolate mophetil [MMF], rituximab) were at lower risk of new relapse in comparison to non-treated patients (HR, 0.41; 95CI%, 0.20–0.82; p = 0.011). Mean ARR (standard deviation) was reduced from 1.05(1.20) to 0.43(0.79) with azathioprine (n = 11; p = 0.041), from 1.20(1.11) to 0.23(0.60) with MMF (n = 11; p = 0.033), and from 1.08(0.98) to 0.43(0.89) with rituximab (n = 26; p = 0.012). Other immunosuppressants (methotrexate/mitoxantrone/cyclophosphamide; n = 5), or multiple sclerosis disease-modifying drugs (MS-DMD; n = 9), were not associated with significantly reduced ARR. Higher rates of freedom of EDSS progression were observed with azathioprine, MMF or rituximab. Conclusion In adults with relapsing MOG-Ab-associated disease, immunosuppressant therapy (azathioprine, MMF and rituximab) is associated with reduced risk of relapse and better disability outcomes. Such an effect was not found in the few patients treated with MS-DMD. Abstract

Background Islets of Langerhans transplantation is a promising therapy for type 1 diabetes mellitus, but this technique is compromised by transplantation stresses including inflammation. In other tissues, co-transplantation with mesenchymal stem cells has been shown to reduce damage by improving anti-inflammatory and anti-oxidant defences. Therefore, we probed the protection afforded by bone marrow mesenchymal stem cells to islets under pro-inflammatory cytokine stress. Methods In order to evaluate the cytoprotective potential of mesenchymal stem cells on rat islets, co-cultures were exposed to the interleukin-1, tumour necrosis factor α and interferon γ cocktail for 24 h. Islet viability and functionality tests were performed. Reactive oxygen species and malondialdehyde were measured. Expression of stress-inducible genes acting as anti-oxidants and detoxifiers, such as superoxide dismutases 1 and 2, NAD(P)H quinone oxidoreductase 1, heme oxygenase-1 and ferritin H, was compared to non-stressed cells, and the corresponding proteins were measured. Data were analysed by a two-way ANOVA followed by a Holm-Sidak post hoc analysis. Results Exposure of rat islets to cytokines induces a reduction in islet viability and functionality concomitant with an oxidative status shift with an increase of cytosolic ROS production. Mesenchymal stem cells did not significantly increase rat islet viability under exposure to cytokines but protected islets from the loss of insulin secretion. A drastic reduction of the antioxidant factors heme oxygenase-1 and ferritin H protein levels was observed in islets exposed to the cytokine cocktail with a prevention of this effect by the presence of mesenchymal stem cells. Conclusions Our data evidenced that MSCs are able to preserve islet insulin secretion through a modulation of the oxidative imbalance mediated by heme and iron via heme oxygenase-1 and ferritin in a context of cytokine exposure. Abstract

International audience; Peroxisomal biogenesis disorders (PBDs) consist of a heterogeneous group of autosomal recessive diseases, in which peroxisome assembly and proliferation are impaired leading to severe multisystem disease and early death. PBDs include Zellweger spectrum disorders (ZSDs) with a relatively mild clinical phenotype caused by PEX1, (MIM# 602136), PEX2 (MIM# 170993), PEX6 (MIM# 601498), PEX10 (MIM# 602859), PEX12 (MIM# 601758), and PEX16 (MIM# 603360) mutations. Three adult patients are reported belonging to a non-consanguineous French family affected with slowly progressive cerebellar ataxia, axonal neuropathy, and pyramidal signs. Mental retardation and diabetes mellitus were optional. The age at onset was in childhood or in adolescence (3-15 years). Brain MRI showed marked cerebellar atrophy. Biochemical blood analyses suggested a mild peroxisomal defect. With whole exome sequencing, two mutations in PEX10 were found in the three patients: c.827G\textgreaterT (novel) causing the missense change p.Cys276Phe and c.932G\textgreaterA causing the missense change p.Arg311Gln. The phenotypic spectrum related to PEX10 mutations includes slowly progressive, syndromic recessive ataxia.

Abstract

[DOI:\hrefhttps://dx.doi.org/10.1007/s00330-015-4090-810.1007/s00330-015-4090-8] [PubMed:\hrefhttps://www.ncbi.nlm.nih.gov/pubmed/1496318114963181]; To evaluate the value of the diameter of the draining vein of pulmonary arteriovenous malformation (PAVM) on unenhanced chest MDCT in diagnosing reperfusion after percutaneous vaso-occlusion therapy.\ We retrospectively reviewed our long-term experience of patients with hereditary haemorrhagic telangiectasia and selected cases on the following criteria: an initial pulmonary angiogram with embolotherapy of at least one PAVM, a follow-up MDCT examination in the following year followed by a second pulmonary angiogram with embolotherapy if needed. Follow-up unenhanced chest MDCT examinations were analyzed blindly from results of pulmonary artery angiogram and clinical data, the diameter of the efferent vein close to the PAVM sac was measured, then compared to those of pulmonary artery angiogram as a gold standard.\ Eighty-eight of 100 patients met inclusion criteria, in whom 62 of 176 PAVMs were reperfused at angiogram. The mean diameter of the efferent vein on MDCT was 4.3 ± 2.1 mm in patent PAVMs and 1.8 ± 0.9 mm in non-patent PAVMs (p < 0.0001). The optimal cutoff diameter based on ROC analysis was 2.5 mm (sensitivity = 98.4 %; specificity = 87.7 %).\ A diameter of the draining vein of PAVM of 2.5 mm or greater on unenhanced MDCT is a strong predictor of reperfusion.\ • Diameter of draining vein of 2.5 mm or greater is associated with reperfusion. • Unenhanced chest MDCT predicts reperfusion of PAVMs with good sensitivity and specificity. • Unenhanced MDCT can guide a decision of repeat pulmonary angiogram and embolotherapy. • The mean vein diameter change of PAVMs occluded at follow-up is 3.8 mm. • Overall success rate after a median of 6 months embolotherapy was 64.7 %.

Abstract

International audience; Single stage injection blow molding process, without preform storage and reheat, could be run on a standard injection molding machine, with the aim of producing short series of specific hollow parts. The polypropylene bottles are blow molded right after being injected. This implies that the preform has to remain sufficiently malleable to be blown while being viscous enough to avoid being pierced during the blow molding stage. These constraints lead to a small processing window, and so the process takes place between the melting temperature and the crystallization temperature, where the polypropylene is in its molten state but cool enough to enhance its viscosity without crystallizing. This single stage process introduces temperature gradients, high stretch rate and high cooling rate. Melt rheometry tests were performed to characterize the polymer behavior in the temperature range of the process, as well as Differential Scanning Calorimetry. A viscous Cross model is used with the thermal dependence assumed by an Arrhenius law. The process is simulated through a finite element code (POLYFLOW) in the ANSYS Workbench framework. The geometry allows an axisymmetric approach. The transient simulation is run under anisothermal conditions and viscous heating is taken into account. Sensitivity studies are carried out and reveal the influence of process parameters such as the material behavior, the blowing pressure and the initial temperature field. Thickness measurements using image analysis are performed and the simulation results are compared to the experimental ones. The simulation shows broad agreements with the experimental results. An optimization loop is run to determine the optimal initial thickness repartition. Design points are defined along the preform and the optimization modifies the thickness at these locations.

Abstract

This paper presents a study of the effects of microscopic particles of talc on the crystallization kinetics of PLA. Our study covers the quiescent conditions and the case where a shear flow is applied “short term shearing”. The incorporation of talc increases the crystallization rate; it enhances the nucleation mechanism through additional heterogeneous nuclei. The microstructure is highly affected by the addition of talc due to the increase of the number of nuclei (i.e. reduced crystalline size). The application of a shear flow increases the ability of PLA to crystallize even in the presence of talc particles. The impact of shear rate becomes dramatically important just after a critical shear rate of 0.1 s−1. It turns out that the shear rate enhances more the crystallization of PLA with talc than the pure PLA under quiescent conditions. Consequently, a supplementary contribution “synergistic effects” is responsible of the relative enhancement of the crystallization of PLA in the presence of shear flow and talc. With combining different experimental analysis techniques and modeling of the crystallization kinetics, the synergistically effects were quantified in terms of the nucleation density induced by the mutual interaction between shear flow and particles.

Abstract

International audience; The dielectric properties of composite materials consisting of a host matrix filled with spherical particles are investigated as a function of frequency by means of numerical calculations. Two different cases are analyzed: (a) composites with a conductive matrix and insulating fillers and (b) composites with an insulating matrix and conductive fillers. In both situations, dielectric dispersions due to interfacial polarization effects are observed in the dielectric spectra. In the present contribution, the characteristic frequencies of interfacial polarization effects are systematically analyzed in dependence on the volume fraction of the spherical fillers and on the conductivity values of the composite phases. The resulting scaling laws are discussed in detail.

Abstract

International audience; The electrical and rheological properties of an ethylene vinyl acetate (EVA) copolymer filled with bamboo charcoal were investigated. The composites were prepared by melt process in an internal batch mixer. Size distribution analysis showed that d(50) and d(90) values of the bamboo charcoal particles are 12.7 and 40 μm, respectively, with a mean diameter of 22 μm. Scanning electron microscopy proved that the particles of bamboo charcoal present a rectangular shape. The electrical percolation threshold was observed at 0.18 volume fraction (35 wt%) of bamboo. Beyond the percolation threshold, a considerable increase in electrical properties was observed up to a limit value of 10-2 S/m. The rheological percolation was studied from different rheological models. As a result, the rheological percolation threshold was observed at 0.3 volume fraction (50 wt%) of bamboo charcoal contents. So, the electrical percolation occurs before the rheological percolation. This is principally due to the filler's characteristics such as the specific surface area, the aspect ratio, and the surface properties. Finally, the bamboo charcoal confers high electrical properties to the EVA composite without inducing strong changes in its viscoelastic properties.

Abstract

International audience; Hydridosilazane compounds containing Si-N and Si-H bonds can be used as precursors of SiOx materials. The hydrolysis-condensation reactions of tetramethyldisilazane, as a polyhydridosilazane model compound, were investigated by H-1 and Si-29 liquid NMR spectroscopy. These reactions were carried out at room temperature for up to 120 min in presence of water. The identified products are short linear siloxane species (hydride terminated polydimethylsiloxanes (MDxMH)-D-H) and cyclosiloxanes. Silicon hydride persistence in the reactional mixture suggested that silazane group is more sensitive to hydrolysis reaction than silicon hydride group. Moreover, additional experiments evidenced that the low steric hindrance of the silicon hydride influences the silazane hydrolysis kinetic. Hence the presence of ammonia released during silazane hydrolysis reaction was demonstrated to be a catalyst of the silicon hydride hydrolysis reaction.

Abstract

International audience; The dispersion of Mg(OH)2 agglomerates at low concentration in a polymer melt was investigated in a transparent counter-rotating shear cell. The influence of the viscosity of the matrix, the initial agglomerate size and the infiltration of the matrix was evaluated. Mg(OH)2 agglomerates have a low cohesion and a fractal structure. Two dispersive mechanisms already mentioned in the literature were identified: erosion and rupture. Critical conditions for rupture were measured and particle size analysis was performed in order to determine the kinetics of erosion. The infiltration of the matrix, which depends on the viscosity, was found to play a key role on dispersion mechanisms. In contrast to previous works, infiltration is more important with the high viscosity matrix. In infiltrated matrix, rupture was found to occur firstly through plastic deformation of the infiltrated agglomerate, and then the agglomerates split into small fragments. In the low viscosity matrix, fragments produced either by rupture or erosion are small aggregates

Abstract

The purpose of the study was to show the feasibility of a positive contrast technique GRadient echo Acquisition for Superparamagnetic particles with Positive contrast (GRASP), for a specific type of magnetic particles, designed for tumor treatment under MRI monitoring.A simulation study was performed to estimate field inhomogeneity intensities induced by increasing concentrations of particles at different static fields. The GRASP sequence was setup on a 4.7 T Bruker system during an in vitro study. Six mice, included in the in vivo study received particles in the left calf muscle and contrast enhancement values, were measured over three time points, for both negative and positive contrast images.Comparing values obtained by simulation at 1.5, 3, and 4.7 T, the strongest susceptibility effect was obtained at 4.7 T. Based on simulation and in vitro data, gradient settings were chosen for in vivo imaging. GRASP resulted in bright regions at and around the injection site, and higher enhancement values, compared to standard GRE imaging. Both contrasts were useful for longitudinal follow-up, with a faster decay over time for GRASP.The magnetic nanoparticles for drug delivery can be detected using positive contrast. Combining imaging sequences, i.e., negative contrast and susceptibility methods, increased imaging specificity of large magnetic particles and enabled their follow-up for theranostic applications.

Abstract

International audience; Superinsulating materials are currently of much interest because of the price of energy on the one hand and CO 2 emissions attributed to offices and houses cooling and heating on the other hand. In this work, we aim at understanding and modeling the radiative transfer within silica nanoporous matrices that are the principal components of nanoporous superinsulating materials. We first elaborate samples of various thicknesses from a pyrogenic silica powder. These samples are characterized using two spectrophotometers on the whole wavelength range [250 nm; 20 µm]. Using a parameter identification technique, we compute the radiative properties of the various samples. Then, our samples being made of packed quasi-spherical particles, we use the Mie theory to model the radiative properties of these materials. Due to the observed discrepancies between the experimental radiative properties and those computed from the Mie theory with a uniform value of 10 nm for the scatterer diameter (value derived from TEM images), we determine an effective scatterer diameter that allows a good agreement between the experimental radiative properties and the Mie results. Nevertheless, in the short wavelength range, the Mie theory gives results that significantly differ from the experimental radiative properties. This behavior is attributed to structure effects as the wavelength is of the same order of magnitude as the diameter of the scatterer that is now regarded as an aggregate of nanoparticles. Hence, to take into account these effects, we use the discrete dipole approximation (DDA). The DDA extinction coefficient spectra appear to be much closer to the experimental results than the Mie spectra, and these first results are quite encouraging.

Abstract

International audience; The main difficulty in the overall process of optimal assembly plan generation is the great number of different ways to assemble a product (typically thousands of solutions). This problem confines the application of most existing automated planning methods to products composed of only a limited number of components. The presented method of assembly plan generation belongs to the approach called “disassembly” and is founded on a new representation of the assembly process, with introduction of a new concept, the equivalence of binary trees. This representation allows to generate the minimal list of all non-redundant (really different) assembly plans. Plan generation is directed by assembly operation constraints and plan-level performance criteria. The method was tested for various assembly applications and compared to other generation approaches. Results show a great reduction in the combinatorial explosion of the number of plans. Therefore, this simplifying approach of assembly sequence modeling allows to handle more complex products with a large number of parts.

Abstract

A performant reagentless electrochemiluminescent (ECL) detection system for H(2)O(2) is presented, based on an electropolymerized polyluminol film prepared under near-neutral conditions. Such an original polyluminol electrodeposition is reported for the first time and on a screen-printed electrode (SPE) surface. Electropolymerized luminol acts as an active luminophore of the electrochemiluminescent reaction, as the monomer does. Polymerization conditions have been optimized in order to obtain the best ECL responses to H(2)O(2). By performing electrodeposition in a potentiostatic mode, at 425 mV vs. Ag|AgCl, in 0.1 mol L(-1) phosphate/0.1 mol L(-1) KCl pH 6 and 1 mmol L(-1) luminol, with a total charge of 0.5 mC, the linear range for H(2)O(2) detection extends from 7.9 x 10(-8) mol L(-1) to 1.3 x 10(-3) mol L(-1). Such performant disposable reagentless easy-to-use miniaturized systems based on SPEs should be applicable to the electrochemiluminescent detection of many oxidase-substrate compounds.

Abstract

Plasma membrane ion channels contribute to virtually all basic cellular processes, including such crucial ones for maintaining tissue homeostasis as proliferation, differentiation, and apoptosis. Enhanced proliferation, aberrant differentiation, and impaired ability to die are the prime reasons for abnormal tissue growth, which can eventually turn into uncontrolled expansion and invasion, characteristic of cancer. Prostate cancer (PCa) cells express a variety of plasma membrane ion channels. By providing the influx of essential signaling ions, perturbing intracellular ion concentrations, regulating cell volume, and maintaining membrane potential, PCa cells are critically involved in proliferation, differentiation, and apoptosis. PCa cells of varying metastatic ability can be distinguished by their ion channel characteristics. Increased malignancy and invasiveness of androgen-independent PCa cells is generally associated with the shift to a 'more excitable' phenotype of their plasma membrane. This shift is manifested by the appearance of voltage-gated Na(+) and Ca(2+) channels which contribute to their enhanced apoptotic resistance together with downregulated store-operated Ca(2+) influx, altered expression of different K(+) channels and members of the Transient Receptor Potential (TRP) channel family, and strengthened capability for maintaining volume constancy. Abstract

The physical modification of glass transition temperature (Tg) and properties of materials via blending is a common practice in industry and academia and has a large economic advantage. In this context, simple production of hitherto unattainable new inorganic glass blends from already existing glass compositions via blending raises much hope with the potential to provide new glasses with new and improved properties, that cannot be achieved with classical glass synthesis, for a plethora of applications such as computers screens, glass-to-metal seals and storage materials for nuclear wastes. Here, we demonstrate that blends of the specific glass compositions studied are miscible in all proportions, an unreported phenomenon in hard condensed matter like glass. Interestingly, excellent agreement was found between the obtained data and calculated Tgs from theoretical equations (Supplementary information) for predicting the composition dependence of Tg for miscible blends with weak but significant specific interactions between the blend components. That this blending method is at present not applied to inorganic glasses reflects the fact that water and chemically resistant phosphate glasses with relatively low Tgs have become available only recently.

Abstract

International audience; Neuronal activity regulates the phosphorylation states at multiple sites on MeCP2 in postmitotic neurons. The precise control of the phosphorylation status of MeCP2 in neurons is critical for the normal development and function of the mammalian brain. However, it is unknown whether phosphorylation at any of the previously identified sites on MeCP2 can be induced by signals other than neuronal activity in other cell types, and what functions MeCP2 phosphorylation may have in those contexts. Here we show that in neural progenitor cells isolated from the adult mouse hippocampus, cell cycle-linked phosphorylation at serine 421 on MeCP2 is directly regulated by aurora kinase B and modulates the balance between proliferation and neural differentiation through the Notch signalling pathway. Our findings suggest MeCP2 S421 phosphorylation may function as a general epigenetic switch accessible by different extracellular stimuli through different signalling pathways for regulating diverse biological functions in different cell types.

Abstract

The structural evolution of a maize flour was followed by means of oscillatory shear measurements during heating at a rate of 10 °C/min in presence of water. These measurements were performed in a special plate and plate vessel designed to prevent moisture loss. The phenomena of gelatinization and gelation were clearly identified and their connection with the moisture content also demonstrated. Moreover, the complex mechanisms involved in gelatinization and gelation in native starch were separated. Softening of the amylose zones, exchange of water and amylose within the starch granules followed by amylopectin melting leads to the gelation of starch.

Abstract

International audience; Nanoparticles became an important and wide-used tool for cell imaging because of their unique optical properties. Although the potential of nanoparticles (NPs) in biology is promising, a number of questions concerning the safety of nanomaterials and the risk/benefit ratio of their usage are open. Here, we have shown that nanoparticles produced from silicon carbide (NPs) dispersed in colloidal suspensions are able to penetrate into surrounding air environment during the natural evaporation of the colloids and label biological cells via vapor phase. Natural gradual size-tuning of NPs in dependence to the distance from the NP liquid source allows progressive shift of the fluorescence color of labeled cells in the blue region according to the increase of the distance from the NP suspension. This effect may be used for the soft vapor labeling of biological cells with the possibility of controlling the color of fluorescence. However, scientists dealing with the colloidal NPs have to seriously consider such a NP's natural transfer in order to protect their own health as well as to avoid any contamination of the control samples.
